Background and aims
Soil nutrient dynamics are affected by root-microbe interactions and plant development. We investigated the influence of plant growth stage and arbuscular mycorrhiza fungi (AMF) on carbon (C) and nitrogen (N) rhizodeposition and the transfer into the microbial biomass (MB).Methods
Pea varieties (Pisum sativum L.) with (Frisson) and without mycorrhiza (P2) were 13C-15N-labelled and harvested at 45, 63, 71, and 95 days after sowing. Mycorrhization, MB, total C, N, 13C, 15N were determined in plant and soil compartments to calculate C and N derived from rhizodeposition (CdfR, NdfR).Results
Total CdfR increased until pea maturity, NdfR until end of flowering. Their relative contribution steadily decreased over time, accounting for 4–10% of total plant C and N at harvest. Rhizodeposition contributed between 1 and 6% to MB C and N, although 20% of the rhizodeposits were discovered in the MB. Frisson released more NdfR than P2 but it was not possible to accurately estimate AMF effects on C and N due to differences in biomass partitioning.Conclusions
CdfR followed an even flow from early growth until senescence. NdfR flow ceased after flowering possibly due to N relocation within the plant. Rhizodeposits contribute very little to MB in our study.2.

Background
Most studies focus on macronutrient of C, N and P and ignore other elements, which restrict our understanding on the strategy of plant nutrient adaption and nutrient cycling.Methods
We investigated 14 element (C, N, P, S, K, Ca, Mg, Fe, Mn, Zn, Cu, Na, Al, and Ba) concentrations of green and senesced leaves in Quercus variabilis along the altitude in the Baotianman Mountains, China, and assessed their relationships with climate, soil, and plant functional traits.Results
Leaf N,S and K increased with, C, Ca, Na, Fe, Mn, Cu and Ba decreased with, and P, Mg, Al, Zn and N:P did not change significantly with altitude. NRE and SRE increased with, and CRE decreased with altitude (p < 0.05). Among the 14 elements, nucleic acid-protein elements (N, K, S and P) were resorbed preferentially, compare to structural (Ca, Mn, and B) and enzymatic (C, Cu, Mg and Zn) that were discriminated against, and toxic (Al and Fe) elements that were totally excluded.Conclusions
Q. variabilis can synergetically regulate green leaf multielement stoichiometry and nutrient resorption in responses to environment change. Deciduous plants may have a trade-off mechanism at the end of growing season to rebalance somatic nutrients.3.

Aims
Visual guidance through echocardiography and fluoroscopy is crucial for a successful transseptal puncture (TSP) in a prespecified region of the fossa ovalis. The novel EchoNavigator system Release II (EchoNav II, Philips Healthcare, Andover, Massachusetts, USA) enables the real-time fusion of fluoroscopic and echocardiographic images. We evaluated this new imaging method in respect to safety and efficacy of TSP during MitraClip implantation and left atrial appendage closure.Methods
Forty-four patients before (?EchoNav) and 44 patients after (+EchoNav) the introduction of real-time fusion were included in our retrospective, single-centre study. The primary endpoint was the occurrence of adverse events due to TSP. Secondary endpoints were successful puncture at the prespecified region and time until TSP (min).Results
In both groups TSP was performed successfully in the prespecified region and no adverse events occurred during or due to the accomplishment of TSP. Time until TSP was significantly reduced in the +EchoNav group in comparison with the EchoNav group (18.48 ± 5.62?min vs. 23.20 ± 9.61?min, p = 0.006).Conclusions
Real-time fusion of echocardiography and fluoroscopy proved to be as safe and successful as standard best practice for TSP. Moreover, efficacy was improved through significant reduction of time until TSP.4.

Aims
The aim was to study effects of slurry acidification, separation technology and thermal processing on the availability of P in soil amended with the solid fraction of pig slurry.Methods
Acidified and non-acidified slurry were separated using different technologies: screw press (SCR), decanting centrifuge (DEC) and drainage after chemical pretreatment (CHE). Solids and pyrolysed (400 °C or 600 °C, char) or combusted (625 °C, ash) solids were applied to two soils, with triple superphosphate (TSP) as a reference. Soil P availability was determined over 12 weeks using diffusive gradients in thin films (DGT).Results
The initial availability of P from DEC and CHE solids was similar to TSP in both soils. After 6 weeks no significant difference was observed between DEC solids and DEC chars. Acidification did not significantly affect P availability in the solids-amended soils over time, but it did affect P availability with the thermally processed materials.Conclusions
Application of separated pig slurry solids generally increased soil P availability initially, but declining with time, indicating soil P fixing reactions. Chars and ashes on the other hand showed lower initial P availability, but remained constant or increased slightly with chars yielding P availability similar to solids after 12 weeks.5.
Aims
Our goals were (1) to determine whether tree species diversity affects nutrient (N, P and K) cycling, and (2) to assess whether there is competition for these nutrients between microbial biomass and trees.Methods
We measured nutrient resorption efficiency by trees, nutrient contents in leaf litterfall, decomposition rates of leaf litter, nutrient turnover in decomposing leaf litter, and plant-available nutrients in the soil in mono-species stands of beech, oak, hornbeam and lime and in mixed-species stands, each consisting of three of these species.Results
Cycling of nutrients through leaf litter input and decomposition were influenced by the types of tree species and not simply by tree species diversity. Trees and microbial biomass were competing strongly for P, less for K and only marginally for N. Such competition was most pronounced in mono-species stands of beech and oak, which had low nutrient turnover in their slow decomposing leaf litter, and less in mono-species stands of hornbeam and lime, which had high nutrient turnover in their fast decomposing leaf litter.Conclusions
The low soil P and K availability in beech stands, which limit the growth of beech at Hainich, Germany, were alleviated by mixing beech with hornbeam and lime. These species-specific effects on nutrient cycling and soil nutrient availability can aid forest management in improving productivity and soil fertility.6.

Objective
To use a transient expression system to express a truncated human tissue plasminogen activator (K2S) gene in cucurbit plants.Results
The recombinant tissue plasminogen activator protein (K2S form) was expressed in active form in cucurbit plants. Its molecular weight was 43 kDa. The plant-derived rt-PA was determined using goat anti-rabbit antibody by western blotting. Among the infected lines, the highest expression of rt-PA was 62 ng/100 mg per leaf tissue as measured by ELISA. The enzymatic activity of the plant-derived rt-PA was 0.8 IU/ml.Conclusions
The K25 form of rt-PA was expressed for the first time using the viral expression system. Plant-derived rt-PA showed similar potency to commercially-available PA.7.

Objective
To compare methods for erythroid differentiation of K562 cells that will be promising in the treatment of beta-thalassemia by inducing γ-globin synthesis.Results
Cells were treated separately with: RPMI 1640 medium without glutamine, RPMI 1640 medium without glutamine supplemented with 1 mM sodium butyrate, RPMI 1640 medium supplemented with 1 mM sodium butyrate, 25 µg cisplatin/ml, 0.1 µg cytosine arabinoside/ml. The highest differentiation (84 %) with minimum toxicity was obtained with cisplatin at 15 µg /ml. Real-time RT-PCR showed that expression of the γ-globin gene was significantly higher in the cells differentiated with cisplatin compared to undifferentiated cells (P < 0.001).Conclusions
Cisplatin is useful in the experimental therapy of ß-globin gene defects and can be considered for examining the basic mechanism of γ-reactivation.8.

Introduction
Human seminal fluid (hSF) has been suggested as a biofluid suitable to characterise male reproductive organ pathology with metabolomics. However, various enzymatic processes, including phosphorylcholine hydrolysis mediated by prostatic acid phosphatase (PAP), cause unwanted metabolite variation that may complicate metabolomic analysis of fresh hSF samples.Objectives
To investigate the effects of PAP inhibition with tartrate.Methods
Using NMR spectroscopy, the kinetics of phosphorylcholine to choline hydrolysis was characterized in hSF samples from three subjects at different temperatures and tartrate concentrations. Principal components analysis was used to characterise the effects of tartrate and temperature on personal differences in metabolite profiles. Potential effects of tartrate on RNA quantification were also determined.Results
Metabolite profiles and the kinetics of phosphorylcholine degradation are reproducible in independent samples from three ostensibly normal subjects. Increasing concentrations of tartrate and refrigerated sample storage (279 K) resulted in greatly reduced reaction rates as judged by apparent rate constants. Multivariate statistical analysis showed that personal differences in metabolite profiles are not overshadowed by tartrate addition, which stabilises phosphorylcholine and choline concentrations. The tartrate signal also served as an internal concentration standard in the samples, allowing the determination of absolute metabolite concentrations in hSF. Furthermore, the presence of tartrate did not affect RNA expression analysis by qPCR.Conclusion
Based on these results we recommend as standard protocol for the collection of hSF samples, that 10 mM tartrate are added immediately to samples, followed by sample storage/handling at 277 K until clinical processing within 6 h to remove/inactivate enzymes and isolate metabolite supernatant and other cellular fractions.9.
Objective
To re-engineer the active site of proteins for non-natural substrates using a position-based prediction method (PBPM).Results
The approach has been applied to re-engineer the E. coli glutamate dehydrogenase to alter its substrate from glutamate to homoserine for a de novo 1,3-propanediol biosynthetic pathway. After identification of key residues that determine the substrate specificity, residue K92 was selected as a candidate site for mutation. Among the three mutations (K92V, K92C, and K92M) suggested by PBPM, the specific activity of the best mutant (K92 V) was increased from 171 ± 35 to 1328 ± 71 μU mg?1.Conclusion
The PBPM approach has a high efficiency for re-engineering the substrate specificity of natural enzymes for new substrates.10.
Background and aims
Aluminum (Al) accumulator plants are occasionally found in certain genera or families of woody plant species that are broadly dispersed in the angiosperm phylogeny. However, spatial and seasonal patterns in Al accumulation within the closely related species of each group remain poorly understood.Methods
We quantitatively monitored the internal Al levels of eight Theaceae and Ternstroemiaceae species growing on acidic soils at multiple sites.Results
Among the eight species, seven other than Ternstroemia gymnanthera shared a rapid Al accumulation in the developing leaves. Species comparison revealed that Al accumulation in mature leaves saturates within a flushing year, regardless of differences in leaf structure, seasonality, and acidic soil pH (4.5–5.5) at multiple sites. In tall trees of Stewartia monadelpha, the Al contents of the leaves were constantly high irrespective of their height positions up to 12 m. Moreover, the Al content of the leaves was only slightly decreased in the last 2 weeks of autumn senescence, in which nitrogen (N) or phosphate (P) retranslocation had been completed.Conclusion
These results suggest that most of the Theaceae and Ternstroemiaceae species possess an effective metal-transport mechanism that rapidly loads Al into the young leaves until each level reaches a species-specific threshold.11.

Background and Aims
Rock fragments within topsoil have important effects on soil properties and plant growth. This study mainly aimed to investigate the relationships between rock fragments, soil carbon (C) and nitrogen (N) densities and vegetation biomass in an alpine steppe.Methods
Rock fragments, plant and soil samples were collected from four topographic positions (top, upper, lower, and bottom) on a hillslope.Results
Volumetric rock fragment content within the 0–30 cm soil profile varied from 17.8 to 30.5%, the upper position value was significantly greater (P < 0.05) than those at other positions. The highest aboveground biomass was observed at the lower position (921 kg ha?1), while the highest belowground biomass within the 0–30 cm profile was found at the upper position (4460 kg ha?1). More fine earth and plant litter input accompanied by lower C and N losses induced by rainfall erosion resulted in higher soil organic C and total N densities (28.6 Mg C ha?1 and 2.87 Mg N ha?1) at the lower position.Conclusions
Rock fragments may promote root growth but limit aboveground biomass production, and can therefore change the biomass distribution pattern. Our findings provide more evidence for scientifically assessing alpine steppe productivity.13.

Purpose
To evaluate the efficiency of corneal collagen cross-linking (CXL) in addition to topical voriconazole in cases with mycotic keratitis.Design
Retrospective case series in a tertiary university hospital.Participants
CXL was performed on 13 patients with mycotic keratitis who presented poor or no response to topical voriconazole treatment.Methods
The clinical features, symptoms, treatment results and complications were recorded retrospectively. The corneal infection was graded according to the depth of infection into the stroma (from grade 1 to grade 3). The visual analogue scale was used to calculate the pain score before and 2 days after surgery.Main Outcome Measures
Grade of the corneal infection.Results
Mean age of 13 patients (6 female and 7 male) was 42.4 ± 17.7 years (20–74 years). Fungus was demonstrated in culture (eight patients) or cytological examination (five patients). Seven of the 13 patients (54%) were healed with topical voriconazole and CXL adjuvant treatment in 26 ± 10 days (15–40 days). The remaining six patients did not respond to CXL treatment; they initially presented with higher grade ulcers. Pre- and post-operative pain score values were 8 ± 0.8 and 3.5 ± 1, respectively (p < 0.05).Conclusions
The current study suggests that adjunctive CXL treatment is effective in patients with small and superficial mycotic ulcers. These observations require further research by large randomized clinical trials.14.
Background
An influenza H3N2 epidemic occurred throughout Southern China in 2012.Methods
We analyzed the hemagglutinin (HA) and neuraminidase (NA) genes of influenza H3N2 strains isolated between 2011–2012 from Guangdong. Mutation sites, evolutionary selection, antigenic sites, and N-glycosylation within these strains were analyzed.Results
The 2011–2012 Guangdong strains contained the HA-A214S, HA-V239I, HA-N328S, NA-L81P, and NA-D93G mutations, similar to those seen in the A/ Perth/16/2009 influenza strain. The HA-NSS061–063 and NNS160–162 glycosylation sites were prevalent among the 2011–2012 Guangdong strains but the NA-NRS402–404 site was deleted. Antigenically, there was a four-fold difference between A/Perth/16/2009 -like strains and the 2011–2012 Guangdong strains.Conclusion
Antigenic drift of the H3N2 subtype contributed to the occurrence of the Southern China influenza epidemic of 2012.15.

Background
In this report the phytochemical profile of Nitraria. Retusa (N. Retusa) leaf extracts were identified and their ability to induce apoptosis in human chronic myelogenous erythroleukaemia (K562) was evaluated.Methods
Apoptosis of the human chronic myelogenous erythroleukaemia (K562) was evidenced by investigating DNA fragmentation, PARP cleavage and caspases 3 and 8 inducing activities, in the presence of N. retusa extracts.Results
Our study revealed that the tested extracts from N. Retusa contain many useful bioactive compounds. They induced in a time-dependent manner the apoptosis the tested cancerous our cell line. This result was confirmed by ladder DNA fragmentation profile and PARP cleavage, as well as a release in caspase-3 and caspase-8 level.Conclusion
Our results indicate that the tested compounds have a significant antiproliferative effect which may be due to their involvement in the induction of the extrinsic apoptosic pathway.16.

Background and aims
The utilization of phosphorus (P) in cattle slurry as a starter fertilizer in maize cropping is poor. To improve this and to obviate the use of additional mineral starter-P fertilization, we examined if slurry acidification, placement strategy and application time could increase maize yield and phosphorus uptake (PU) in the early growth stages.Methods
In a climate-controlled pot experiment, untreated (pH 6.5) and acidified (pH 5.5 or pH 3.8) cattle slurry was injected in narrow or broad bands two or 30 days before sowing of maize on a coarse sandy and a sandy loam soil and compared with mineral P fertilizer.Results
After broad band slurry injection, the P concentration in maize tissues at the five-leaf stage and the dry matter yield at the seven-leaf stage were equal to or higher than the mineral P fertilizer treatment. Treatments with strongly acidified slurry (pH 3.8) had 49% higher PU at the seven-leaf stage compared to untreated slurry, but only on the sandy soil, suggesting an indirect pH effect on PU. Application time had no effect.Conclusion
Broad band slurry injection or strong acidification can improve early-stage growth of maize and potentially obviate the use of mineral P.17.

Aims
Intercropping can improve plant yields and soil phosphorus (P) use efficiency. This study compares inter- and intra-species intercropping, and determines whether P uptake and shoot biomass accumulation in intercrops are affected by soil P availability.Methods
Four barley cultivars (Hordeum vulgare L.) and three legume species (Trifolium subterreneum, Ornithopus sativus and Medicago truncatula) were selected on the basis of their contrasting root exudation and morphological responses to P deficiency. Monocultures and barley-barley and barley-legume intercrops were grown for 6 weeks in a pot trial at very limiting, slightly limiting and excess available soil P. Above-ground biomass and shoot P were measured.Results
Barley-legume intercrops had 10–70% greater P accumulation and 0–40% greater biomass than monocultures, with the greatest gains occurring at or below the sub-critical P requirement for barley. No benefit of barley-barley intercropping was observed. The plant combination had no significant effect on biomass and P uptake observed in intercropped treatments.Conclusions
Barley-legume intercropping shows promise for sustainable production systems, especially at low soil P. Gains in biomass and P uptake come from inter- rather than intra-species intercropping, indicating that plant diversity resulted in decreased competition between plants for P.18.

Aims
Little is known about how plant leaf litter decomposing on the soil surface is affecting microbial communities in the underlying soil. Here we examined the effects of decomposing leaf litter of different initial chemistry on biomass, stoichiometry, community structure and activity of microorganisms in the soil underneath the decaying litter layer.Methods
Leaf litter from six different neotropical tree species with contrasted quality decomposed on top of a common tropical soil in a laboratory microcosm experiment over 98 days. At the end of the experiment we determined microbial biomass C, N, and P, microbial community structure (PLFA), and community level physiological profiles (CLPP) from the top soil.Results
Despite growing in a common soil substrate, soil microorganisms were strongly affected by litter species, especially by the soluble litter fraction. While litters with low soluble C content did not affect the soil microbial community, litters with high soluble C content led to an increase of microbial biomass and to a structural shift to relatively more Gram-negative bacteria. Changing community structure resulted in changes of catabolic capacity of microorganisms to metabolize a range of different C substrates. The large differences in leachate N and P among litter species, in contrast, had no effect on soil microbial parameters.Conclusions
Our data suggest that plant litter decomposing on the soil surface exhibit a strong and predictable leachate C-control over microbial community biomass, structure and function in the underlying soil.19.

Introduction
Few studies have investigated the influence of storage conditions on urine samples and none of them used targeted mass spectrometry (MS).Objectives
We investigated the stability of metabolite profiles in urine samples under different storage conditions using targeted metabolomics.Methods
Pooled, fasting urine samples were collected and stored at ?80 °C (biobank standard), ?20 °C (freezer), 4 °C (fridge), ~9 °C (cool pack), and ~20 °C (room temperature) for 0, 2, 8 and 24 h. Metabolite concentrations were quantified with MS using the AbsoluteIDQ? p150 assay. We used the Welch-Satterthwaite-test to compare the concentrations of each metabolite. Mixed effects linear regression was used to assess the influence of the interaction of storage time and temperature.Results
The concentrations of 63 investigated metabolites were stable at ?20 and 4 °C for up to 24 h when compared to samples immediately stored at ?80 °C. When stored at ~9 °C for 24 h, few amino acids (Arg, Val and Leu/Ile) significantly decreased by 40% in concentration (P < 7.9E?04); for an additional three metabolites (Ser, Met, Hexose H1) when stored at ~20 °C reduced up to 60% in concentrations. The concentrations of four more metabolites (Glu, Phe, Pro, and Thr) were found to be significantly influenced when considering the interaction between exposure time and temperature.Conclusion
Our findings indicate that 78% of quantified metabolites were stable for all examined storage conditions. Particularly, some amino acid concentrations were sensitive to changes after prolonged storage at room temperature. Shipping or storing urine samples on cool packs or at room temperature for more than 8 h and multiple numbers of freeze and thaw cycles should be avoided.20.
