The Adelaide River in Australia's Northern Territory is a popular recreational fishing area, as well as habitat for threatened and protected river sharks (Glyphis species). Both the Critically Endangered Speartooth Shark (Glyphis glyphis) and Endangered Northern River Shark (Glyphis garricki) are identified here in illegal catches from recreational angling. The identification of a decayed shark specimen using a DNA barcoding‐like approach is the first such application to the identification of protected sharks in a recreational fishery. While the extent of catches by recreational anglers is unknown, the threatened status of these sharks, their suspected low population sizes, restricted distributions and importance of the Adelaide River as a nursery area call for the consideration of this as a potential conservation issue. As such, appropriate measures should be taken to reduce interactions with recreational anglers. The primary target species in the river is the iconic sportfish, Barramundi, which is predominantly caught by unbaited lure. Sharks are rarely caught on lure, allowing an opportunity for mitigation to focus on a fishing activity (baited hooks) which would limit any regulatory impact on popular lure fishing. Potential mitigation measures range from increased angler education and compliance checks, to the implementation of a spatial closure to baited hook fishing (a lure‐only zone). Such measures may assist in meeting a stated objective of the Australian Government's river shark Recovery Plan to ‘reduce and, where possible, eliminate adverse impacts of recreational fishing'.  相似文献

Productivity measurements from carbon uptake have been suggested as good indicators of the physiological health of seagrasses. As seagrasses acquire carbon from the surrounding water, the rate of uptake often provide a good measure of the efficiency at which seagrasses meet their resource demands for growth. This rate is often used to assess the photosynthetic efficiency of the plants, a proxy for the physiological status of seagrass. This has special relevance to the Adelaide region as over 5000 ha of seagrasses have been lost from Adelaide coastal waters over the last 70 years, with much of this loss attributed to nutrient inputs from wastewater, industrial and stormwater discharges. This study used an in-situ inorganic carbon isotope-labelling and spike approach to obtain ecologically relevant estimates of seasonal variability in carbon uptake and its allocation in two species of temperate seagrass common to this coast (Amphibolis antarctica and Posidonia angustifolia). Uptake of carbon by the seagrass complex (leaves, roots, phytoplankton and epiphytes) was affected by both season and species. Carbon uptake rates of phytoplankton were generally higher than other components of the system. Uptake rates ranged from 0.01 mg C g− 1 DW h− 1 (summer) to 0.61 mg C g− 1 DW h− 1 (spring) in Posidonia and 0.02 mg C g− 1 DW h− 1 (summer) to 0.93 mg C g− 1 DW h− 1 (winter) in Amphibolis. Carbon uptake by the Amphibolis complex was higher than in the Posidonia complex. The Amphibolis complex had higher uptake rates in summer whereas the Posidonia complex was higher in spring. Fine sediments probably from a nearby dredging operation, are likely to have resulted in lower carbon uptake and a reduction in the above-ground and below-ground biomass in summer.  相似文献

Evolution of the triplet code is reconstructed on the basis of consensus temporal order of appearance of amino acids. Several important predictions are confirmed by computational sequence analyses. The earliest amino acids, alanine and glycine, have been encoded by GCC and GGC codons, as today. They were succeeded, respectively, by A- and G-series of amino acids, encoded by pyrimidine-central and purine-central codons. The length of the earliest proteins is estimated to be 6–7 residues. The earliest mRNAs were short G+C-rich molecules. These short sequences could have formed hairpins. This is confirmed by analysis of modern prokaryotic mRNA sequences. Predominant size of detected ancient hairpins also corresponds to 6–7 amino acids, as above. Vestiges of last common ancestor can be found in extant proteins in form of entirely conserved short sequences of size six to nine residues present in all or almost all sequenced prokaryotic proteomes (omnipresent motifs). The functions of the topmost conserved octamers are not involved in the basic elementary syntheses. This suggests an initial abiotic supply of amino acids, bases and sugars. This article outlines the life and career of H.A. (Bert) Hawkes, who has been referred to as one of the founders of field ecology, due to the work he conducted in the late 1940s and 1950s, investigating the fly nuisance emanating from biological filters. Relaxation kinetics measurements on two types of ribosome preparations were parformed by the pressure-jump and temperature-Jump techniques, using light scattered at 90° as detector. For freshly prepared tibosomes isolated as 70S tight coupled from 26 000 RPM sucrose gradint sedimentation in 10 mM Mg2+, surprisingly large reaction amplitudes were found in 10 mM Mg2+ wilh both techniques, leading to an overall formation constant for 70S couples approximately three orders of magnitude smaller than that reported fot tight couples. For pelleted, two-tunes salt-washed ribosomes, amplitude titration versus Mg2+ in the pressure-jump apparatus showed an amplitude maximum near 10 mM Mg2+ with a relaxation time near 20 ms, and a second amplitude maximum near 2.5 mM Mg2+ with a relaxation time near 25 s. Both types of preparation on reanalysis on sucrose gradients at 5 mM Mg2+ showed approximately 15% of subunits, with a distinct zone in the 50S region. 70S light couples recovered from a sucrose density gradient separation at 5 mM Mg2+ on pelleted two-times salt-washed ribosomes behaved in the same way as the original sample in pressure-jump experiments at 10 mM Mg2+. These findings have been interpreted as follows (I) the processes observed at 10 mM Mg2+ are due entirety to the relatively small loose couple content of the samples, even in the case of material isolated as 70S tight couples, (2) the processes observed at 2.5 mM Mg2+ are due almost entirely to the preponderant tight couple population of the material, and (3) samples isolated as 70S tight couples from sucrose gradients at 5 mM Mg2+ spontaneously revert within hours into micro-heterogeneous material containing about 15% loose couples, for both types of ribosomes.  相似文献

ABSTRACT Sarcocystis dubeyella n. sp. and S. phacochoeri n. sp. from muscle fibers of the skeletal musculature of two warthogs in South Africa are described by light and and electron microscopy. Sarcocystis dubeyella sarcocysts are macroscopic (up to 12 mm long and 1 mm wide), with a parasite-induced encapsulation of the host muscle fiber in which the plasma membrane of the latter remained unaltered. The sarcocyst wall is characterized by evenly arranged, irregularly semicircular or rectangular villar protrusions (5.0 T. 2.8-11.0 μm) with indented margins and no specific content. Sarcocystis phacochoeri formed filiform microcysts (up to 4 mm long and 0.13 mm wide). Its cyst wall is provided with tightly packed, molarlike villar protrusions (1.6-3.3 T. 1.7-3.3 μm), with smooth margins, hollow on one side, and with longitudinal condensations of the fine granular matrix at various locations in the interior.  相似文献

The rotational brownian diffusion of Escherichia coli ribosomes has been studied by following the transient dichroism generated by optical excitation of a covalent probe into its triplet state. The induced absorption anisotropy decays exponentially with characteristic correlation times: 2.5 microseconds, 1.6 microseconds and 1.1 microseconds for the 70S ribosome and the 50S and 30S subparticles respectively. The corresponding Stokes radii are in the same order, 133 A, 115 A and 103 A. The hydrodynamic properties are discussed in terms of an ellipsoidal shape of the ribosome particles.  相似文献

Background

Genome signatures of artificial selection in U.S. Jersey cattle were identified by examining changes in haplotype homozygosity for a resource population of animals born between 1953 and 2007. Genetic merit of this population changed dramatically during this period for a number of traits, especially milk yield. The intense selection underlying these changes was achieved through extensive use of artificial insemination (AI), which also increased consanguinity of the population to a few superior Jersey bulls. As a result, allele frequencies are shifted for many contemporary animals, and in numerous cases to a homozygous state for specific genomic regions. The goal of this study was to identify those selection signatures that occurred after extensive use of AI since the 1960, using analyses of shared haplotype segments or Runs of Homozygosity. When combined with animal birth year information, signatures of selection associated with economically important traits were identified and compared to results from an extended haplotype homozygosity analysis.

Results

Overall, our results reveal that more recent selection increased autozygosity across the entire genome, but some specific regions increased more than others. A genome-wide scan identified more than 15 regions with a substantial change in autozygosity. Haplotypes found to be associated with increased milk, fat and protein yield in U.S. Jersey cattle also consistently increased in frequency.

Conclusions

The analyses used in this study was able to detect directional selection over the last few decades when individual production records for Jersey animals were available.
