The metabolic fate of the bile add analogs, 3α,7α-dihydroxy-7β-methyl-5β-cholanoic acid and 3α,7β-dihydroxy-7α-methyl-5β-cholanoic acid, was investigated and compared with that of chenodeoxycholic acid in hamsters. Both bile acid analogs were absorbed rapidly from the intestine and excreted into bile at similar to that of chenodeoxycholic acid. In the strain of hamster studied, the biliary bile were conjugated with both glycine and taurine. After continuous intravenous infusion, chenodeoxycholic acid the analogs became the major bile acid constituents in bile. After oral administration of a single dose of these compounds, fecal analysis revealed the existence of unchanged material (25–35%) as well as considerable amounts of metabolites (65–75%). The major metabolites excreted into feces were more polar than the starting material and were tentatively identified as trifaydroxy-7-methyl compounds by radioactive thin-layer chromatography. However, monohydroxy compounds were also found in the fecal extracts. These results show that chenodeoxycholic acid and ursodeoxycholic acid with a methyl group at the 7-position are resistant to bacterial 7-dehydroxylation than the normally occurring bile acids and that a certain proportion of these analogs is hydroxylated to give the corespondiag trihydroxy compound(s), In a control experiment, about 5% of administered chenodeoxychoulic acid was metabolized to a trihydroxy feile acid, but most of the compound (95%) was transformed into lithocholic acid.  相似文献

Treatment of 3β-benzoyloxy-14α, 15α-epoxy-5α-cholest-7-ene with boron trifluoride-etherate gave, in 43% yield, 3β-benzoyloxy-5α, 14β-cholest-7-en-15-one with the unnatural C ring juncture. Reduction of the latter compound with lithium aluminum hydride gave 15α, 14β-cholest-7-en-3β, 15α-diol and 5α, 14β-cholest-7-en-3β, 15β-diol in 9% and 81% yields, respectively.  相似文献

From incubations of testosterone with rat testicular homogenates in the presence of a NADPH-generating system, the following 7α-hydroxylated metabolites could be isolated and identified: 7α,17β-dihydroxy-4-androsten-3-one (7α-hydroxy-testosterone), 7α-17β-dihydroxy-5α-androstan-3-one (7α-hydroxy-Dht), 5α-androstan-3α,7α,17β-triol (7α-hydroxy-3α-A'DIOL) and 5α-androstane-3β,7α,l7β-triol (7α-hydroxy-3β-A'DIOL). To our knowledge this is the first demonstration of the formation of 5α-reduced-7α-hydroxylated metabolites of testosterone in the male gonad. These 5α-reduced-7α-hydroxylated metabolites could also be isolated after incubations of 5α-androstane-3α,17β-diol (3α-A'D10L) with testicular homogenates in the presence of a NADPH-generating system.Measured as the sum of 7α-hydroxy-testosterone, 7α-hydroxy-Dht. 7α-hydroxy-3α-A'DIOL and 7α-hydroxy-3β-A'DIOL formed using testosterone as substrate, total 7α-hydroxylase activity was six times higher in testes of mature rats than in testes from animals 23 days old. With 3α-A'DIOL as substrate total 7α-hydroxylase in the mature testis was about three times greater than in the sexually immature testis.  相似文献

Both enantiomers of 3α,6β-dibenzoyloxytropane (1) have been prepared from optical active 6β-hydroxyhyoscyamines establishing their absolute configurations as (?)-(3R,6R) and (+)-(3S,6S)-dibenzoyloxytropane. Independent stereochemical confirmation was obtained by vibrational circular dichroism measurements, since bands characteristic of (3R,6R) and (3S,6S) configurations of tropanediols derivatives were observed. In addition, a chiral HPLC method was developed for determining absolute configurations of tropane-related natural substances at the microgram (μg) level. The complete 1H NMR characterization of the scaffold of 1 is also reported.  相似文献

The 5β-pregnane-3α,6α,20α-iriol and 5β-pregnane-3α,6α,20β-triol obtained by reduction of 3α,6α-dihydroxy-5β-pregnan-20-one were converted to trimethylsilyl ether derivatives and analysed by gas-liquid chromatography (GLC) and gas chromatography-mass spectrometry (GC-MS).After extraction, solvolysis and enzymatic hydrolysis of urinary steroid conjugates (from 19 normal pregnant women), the liberated steroids were separated by liquid-gel chromatography and were analysed by GLC and GLC-MS. The 5β-pregnane-3α,6α,20α-triol isolated and identified in the Sephadex LH-20 fractions 8 and 9, was present in urine from 15 pregnant women after the 16th week of gestation. After this time, this metabolite was found in a quantity between 0.20 to 2.90 mg/24 h, with a significant increase between the 26th to 30th week of the gestation.With the present in vivo data, it is not possible to establish the exact enzymatic pathway involved in the biosynthesis of the 5β-pregnane-3α,6α,20α-triol. However, it is probable that the immediate precursor of this compound was the 3α,6α-dihydroxy-5β-pregnan-20-one, and that urinary excretion of 5β-pregnane-3α,6α,20α-triol reflected one part of hepatomaternal metabolism of 6-hydroxyprogesterone formed in the foeto-placental unit.  相似文献

Metal-tolerant Datura innoxia cells synthesize large amounts of cadystin, [poly(-glutamylcysteinyl) glycines, (-EC)nG, n=2–5], a class of metal-binding polypeptides, when exposed to Cd. These polypeptides have a high affinity for Cd (II) and certain other metal ions and are thought to play a role in metal tolerance in higher plants. Cells rapidly synthesize these metal-binding polypeptides when exposed to Cd and cellular concentrations of glutathione and cysteine, precursors for the synthesis of these compounds, are initially depleted then rapidly replenished. The time-frame of de novo polypeptide, glutathione and cysteine biosynthesis suggests that this pathway is, at least initially, regulated at the enzyme level. Significant amounts of Fe are associated with Cd: polypeptide complexes isolated from D. innoxia. Exposure of cultures to Cd results in an increased Fe accumulation by the cells. All the additional Fe found in the soluble portion of cell extracts is associated with the Cd: polypeptide complexes. The physiological significance of the synthesis of these polypeptides and their precursors and its relevance to Cd tolerance and metal homeostasis are discussed.  相似文献

Treatment of 3β-p-bromobenzoyloxy-14α, 15α-epoxy-5α-cholest-7-ene with gaseous HCI in chloroform at ?25°C gave 3β-p-bromobenzoyloxy-7α, 15β-dichloro-5α-cholest-8(14)-ene in 93% yield. Measurement of isotope ratios in 1α,2α,3β-trihydroxy-p-menthane, which has been biosynthesized in Fusicoccum amygdali from 3H- and 14C-labelled mevalonate and in its degradation product diosphenol indicates that: (a) four tritium atoms arising from [5-3H2, 2-14C]MVA are retained, one more than suggested from the hydroxylation pattern, (b) menth-2-ene-1-ol is generated from an α-terpinyl cation through a 1,3-hydride shift and (c) trans-cleavage of an α-epoxide by hydrolysis gives 1α,2α,3β-trihydroxy-p-menthane.  相似文献

This study has identified the polar metabolites of 5α-androstane-3β, 17β-diol(3β-diol) produced by the canine prostate. The major metabolite is 5α-androstane-3β, 7α, 17β-triol (7α-triol) accounting for approximately 80% of the total polar metabolites of 3β-diol. The remaining 20% is accounted for exclusively by another triol, 5α-androstane-3β, 6α, 17β-triol(6α-triol). This study has also characterized two enzymatic hydroxylases responsible for respective triol formation: 5α-androstane-3β, 17β-diol 6α-hydroxylase (6α-hydroxylase) and 5α-androstane-3β, 17β-diol 7α-hydroxylase (7α-hydroxylase). Both of these irreversible hydroxylases are located in the particulate fraction of the prostate and can utilize either NADH or NADPH as cofactor. Several in vitro steroid inhibitors of these hydroxylases were identified including cholesterol, estradiol and diethylstilbestrol. Neither of the hydroxylases were found to be decreased by castration (3 months) when expressed as activity/DNA. Using a variety of C19 androstane substrates, 6α- and 7α-triol were found to be major components of the total 3β-hydroxy-5α-androstane metabolites produced by the canine prostate.  相似文献

A short and efficient method for the stereospecific synthesis of 3α,7α-dihydroxy-5β-androstan-17-one was accomplished from the readily available 4-androstene-3,17-dione. Key steps are the stereospecific and selective epoxidation of 4,6-androstadiene-3,17-dione, followed by hydrogenations with carefully selected reagents, solvents and reaction conditions.  相似文献

The effect of estradiol, hydrocortisone and progesterone on 3,20-and 3,17-hydroxysteroid dehydrogenase (HSD) in mutants of Streptomyces hydrogenans was compared to the steroid response of the wild type. Mutants were defective in arginine biosynthesis and/or aerial mycelial formation and lacked both enzymes or only 17-HSD. Some 17-HSD mutants had lost the ability to be induced by estradiol, by progesterone or by both. Some 20-HSD mutants had lost the ability to be induced by hydrocortisone, by progesterone or by both. Non-inducibility of 17-and 20-HSD by progesterone was not co-ordinate. The reduction of 3-ethylenedioxy-7-oximino-5-androsten-17β-yl acetate and of its 17β-tetrahydropyranyl ether analog with sodium in ethanol, followed by thin-layer chromatography, allowed the isolation of the corresponding 17β-hydroxy- and 17β-tetrahydropyranyioxy-5-en-7β- and 7α-amines which were also characte-rized as 7-acetamides. The acylation of the two epimeric 17β-hydroxy-5-en-7-amines with succinic anhydride followed by selective saponification of the 17β-hemisuccinate group and diazomethane esterification, gave the corresponding 17β-hydroxy-5-en-7β- and 7α-hemisuccinamido methyl esters characterized also as 17β-acetates. On the other hand, the acylation of the two 17β-tetrahydropyranyl-oxy-5-en-7-amines with the acid chloride of terephthalic acid monomethyi ester led to the more rigid 7β- and 7α-terephthalamido methyl ester side-chains. The acidolysis of the 3-ethyleneketal protecting group of the preceding 5-en-7-N-acyl derivatives regenerated the 4-en-3-oxo function while the 17β-tetrahydropyranyl ether group was cleaved simultaneously into the 17β-alcohol. The four desired 7β- and 7α-hemisuccinamido- and terephthalamido carboxylic side-chain derivatives of 17β-hydroxy-4-androsten-3-one (testosterone) were finally obtained by saponification of the corresponding methyl esters.  相似文献

A single thin layer chromatography and three antibodies were used for the specific radioimmunoassay of four androgens in pooled rat plasma (Sprague-Dawley adult males). At radioimmunoassay (RIA) for 5α-androst-16-en-3α-ol in human and porcine plasma has been developed. Antibodies were produced in rabbits immunized against 5α-androst-16-en-3-(O-car☐ymethyl) oxime conjugated to BSA. The antiserum cross-reacted with other 16-androstenes as follows: 5α-androst-16-en-3α-ol, 42%; 4,16-androstadien-3-one, 23.8% 5α-androst-16-en-3β-ol, 16.1% and 5,16-androstadien-3β-ol, 1.19%.Although the method gave an acceptable standard curve for 5α-androst-16-en-3-one (10–1000 pg), it was not possible to separate by t.l.c. an unknown contaminant in plasma which interfered with the RIA of the steroid. However, by exploiting the high (42%) cross-reaction with 5α-androst-16-en-3α-ol, a RIA has been developed involving a thin layer chromatographic step. Regression analysis of the data in an accuracy study gave the equation y = 0.986 x + 151.5, withr = 0.999, while the coefficients of variation of replicate assays of plasma 5α-androst-16-en-3α-ol were in the range 9.5–15.6%.The mean values for plasma 5α-androst-16-en-3α-ol in 31 healthy men and 16 healthy women were 3.08 (range 0.3–14.9) and 0.66 (range 0–2.4) ng/ml, respectively. In boars, sows and castrated male pigs, the corresponding mean values were 30.7 (range 10.9–58.9), 0.24 (range 0.08–0.77) and 0.27 (range 0.06–0.51) ng/ml, respectively.  相似文献

Both the 5α, 6α- and 5β, 6β-dichloromethylene adducts (2a and 2b) of 3β-acetoxy-5-androsten-17-one (1) are produced when the latter is exposed to dichlorocarbene generated from chloroform and base by Phase Transfer Catalysis using ultrasound as a means of agitation. The 1H NMR substituent effects of 5α, 6α- and 5β, 6β-dichloromethylene on the angular methyl groups (Zürcher values) are given.
