The C. parapsilosis sensu lato group involves three closely related species, C. parapsilosis sensu stricto, C . orthopsilosis and C . metapsilosis . Although their overall clinical importance is dramatically increasing, there are few studies regarding the virulence properties of the species of the psilosis complex. In this study, we tested 63 C. parapsilosis sensu stricto, 12 C . metapsilosis and 18 C . orthopsilosis isolates for the ability to produce extracellular proteases, secrete lipases and form pseudohyphae. Significant differences were noted between species, with the C . metapsilosis strains failing to secrete lipase or to produce pseudohyphae. Nine different clinical isolates each of C. parapsilosis sensu stricto, C . orthopsilosis and C . metapsilosis were co-cultured with immortalized murine or primary human macrophages. C. parapsilosis sensu stricto isolates showed a significantly higher resistance to killing by primary human macrophages compared to C . orthopsilosis and C . metapsilosis isolates. In contrast, the killing of isolates by J774.2 mouse macrophages did not differ significantly between species. However, C. parapsilosis sensu stricto isolates induced the most damage to murine and human macrophages, and C . metapsilosis strains were the least toxic. Furthermore, strains that produced lipase or pseudohyphae were most resistant to macrophage-mediated killing and produced the most cellular damage. Finally, we used 9 isolates of each of the C. parapsilosis sensus lato species to examine their impact on the survival of Galleria mellonella larvae. The mortality rate of G . mellonella larvae infected with C . metapsilosis isolates was significantly lower than those infected with C. parapsilosis sensu stricto or C . orthopsilosis strains. Taken together, our findings demonstrate that C . metapsilosis is indeed the least virulent member of the psilosis group, and also highlight the importance of pseudohyphae and secreted lipases during fungal-host interactions.  相似文献

Males of certain species belonging to the Bactrocera dorsalis complex are strongly attracted to, and readily feed on methyl eugenol (ME), a plant secondary compound that is found in over 480 plant species worldwide. Amongst those species is one of the world’s most severe fruit pests the Oriental fruit fly, Bactrocera dorsalis s.s., and the former taxonomic species Bactrocera invadens, Bactrocera papayae and Bactrocera philippinensis. The latter species have been recently synonymised with Bactrocera dorsalis based on their very similar morphology, mating compatibility, molecular genetics and identical sex pheromones following consumption of ME. Previous studies have shown that male fruit fly responsiveness to lures is a unique phenomenon that is dose species-specific, besides showing a close correlation to sexual maturity attainment. This led us to use ME sensitivity as a behavioural parameter to test if Bactrocera dorsalis and the three former taxonomic species had similar sensitivity towards odours of ME. Using Probit analysis, we estimated the median dose of ME required to elicit species’ positive response in 50% of each population tested (ED50). ED50 values were compared between Bactrocera dorsalis and the former species. Our results showed no significant differences between Bactrocera dorsalis s.s., and the former Bactrocera invadens, Bactrocera papayae and Bactrocera philippinensis in their response to ME. We consider that the Bactrocera males’ sensitivity to ME may be a useful behavioural parameter for species delimitation and, in addition to other integrative taxonomic tools used, provides further supportive evidence that the four taxa belong to one and the same biological species, Bactrocera dorsalis.  相似文献

A new species of Ogdoconta Butler (Lepidoptera, Noctuidae, Condicinae, Condicini) is described from the Patagonia Mountains, Santa Cruz County, Arizona, USA. Ogdoconta margareta sp. n., is related closely to Ogdoconta tacna (Barnes) from Texas. This paper launches an open access DNA barcoding project “AUSBS” under the Barcoding of Life Datasystems (BOLD). The aims of the project are to help scientists who lack the necessary morphological knowledge to identify known species using molecular markers, to aid native bee specialists with the recognition of species groups that morphologically are difficult to define, and, eventually, to assist with the recognition of new species among known species. Using integrative taxonomy, i.e. morphological comparison to type specimens in Australian museum collections combined with phylogenetic analysis of a fragment of the mitochondrial DNA cytochrome c oxidase subunit I (mtCOI) gene sequences led to the recognition of four new species of Euhesma Michener (Hymenoptera: Colletidae: Euryglossini) collected during intensive surveys in remote Australian conservation areas, which are described. The new species are Euhesma micans, Euhesma lyngouriae, and Euhesma aulaca in a species group associated with Eremophila flowers, and Euhesma albamala in the walkeriana species group.  相似文献

The taxonomic position of two antarctic dorylaimid species Amblydorylaimus isokaryon (Loof, 1975) Andrássy, 1998 and Pararhyssocolpus paradoxus (Loof, 1975), gen. n., comb. n. are discussed on the basis of morphological, including SEM study, morphometric, postembryonic and sequence data of 18S rDNA and the D2-D3 expansion fragments of large subunit rDNA. The evolutionary trees inferred from 18S sequences show insufficient resolution to determine the assignment of the two species to particular families, moreover Pararhyssocolpus paradoxus gen. n., comb. n. (=Rhyssocolpus paradoxus) previously regarded as a member of Nordiidae or Qudsianematidae, showed distant relationship both to Rhyssocolpus vinciguerrae and Eudorylaimus spp. The phylogram inferred from 28S sequences revealed that Amblydorylaimus isokaryon is a member of a well-supported group comprised of several Aporcelaimellus spp., while, no close relationships could be revealed for the Pararhyssocolpus paradoxus gen. n., comb. n. to any nematode genus. On the basis of molecular data and morphological characteristics, some taxonomic changes are proposed. Amblydorylaimus isokaryon is transferred from family Qudsianematidae to family Aporcelaimidae, and a new monotypic genus Pararhyssocolpus gen. n. is proposed, attributed to Pararhyssocolpidae fam. n. The diagnosis of the new family is provided together with emended diagnosis of the genera Amblydorylaimus and Pararhyssocolpus gen. n. Data concerning distribution of these endemic genera in the Antarctic region are also given.  相似文献

The formely monotypic Neotropical genus Megalocraerus Lewis is revised to include five species, known from southeastern Brazil to Costa Rica: Megalocraerus rubricatus Lewis, Megalocraerus mandibularis sp. n., Megalocraerus chico sp. n., Megalocraerus madrededios sp. n., and Megalocraerus tiputini sp. n. We describe the species, map their distributions, and provide a key for their identification. Their subcylindrical body form and emarginate mesosternum have previously hindered placement to tribe, although their curent assignment to Exosternini now appears well supported by morphological evidence. Ilex, the largest genus of dioecious woody plants, is a good study system to assess the role of hybridization in speciation and evolution. Ilex dabieshanensis, a tree endemic to Dabieshan Mountains region, was initially described as a new species. Based on morphological intermediacy and sympatric distribution with its putative parental species, I. cornuta and I. latifolia, we proposed it as a natural hybrid between them. In this study, we sequenced one chloroplast intergenic spacer (trnH-psbA) and two nuclear genes (gapC and nepGS) in I. dabieshanensis and its putative parental species to test the hybrid origin hypothesis. Our results showed that there were one to two differentially fixed sequence differences between I. cornuta and I. latifolia at the two nuclear genes. Twelve of the 14 individuals of I. dabieshanensis exhibited additivity in chromatograms on these differentially fixed sites at both nuclear genes, and the remaining two exhibited additivity in chromatograms on the fixed site at only the nepGS gene. Except one haplotype of I. cornuta at the nepGS gene, all of the haplotypes of I. cornuta at the two nuclear genes were well separated from those of I. latifolia, and most haplotypes of I. dabieshanensis were shared with those of I. cornuta and I. latifolia. Phylogenetic analysis of these haplotypes was largely consistent with haplotype network analysis. I. cornuta and I. latifolia differed by two nucleotide substitutions in the chloroplast intergenic spacer, and 12 individuals of I. dabieshanensis had the same sequences as I. latifolia, while the remaining two were identical with I. cornuta. The molecular data provide convincing evidence for the hybrid origin of I. dabieshanensis and asymmetrical direction of hybridization. One haplotype of I. cornuta at the nepGS gene was nested with those of I. latifolia, indicating introgression to I. cornuta.  相似文献

The population phenology of the cassidines, Coptocycla arcuata and Omaspides trichroa, and the chrysomeline, Platyphora axillaris, was studied at Serra dos Órgãos National Park, State of Rio de Janeiro, southeast Brazil. Monthly surveys of larvae and adults were conducted between 2008 and 2011 at approximately 1000 m altitude on their respective host plants, Cordia polycephala (Boraginaceae), Ipomoea philomega (Convolvulaceae) and Solanum scuticum (Solanaceae). This is the first observation of larviparity and host record for Platyphora axillaris. Although having different life history traits, all species showed similar phenologies. They were abundant from October to March, months of high temperatures and intense rainfall, with two distinct reproductive peaks in the same season. Abundance dropped abruptly during the coldest and driest months, from May to August. Frequently none of these species were recorded during June and July. This phenological pattern is similar to other Chrysomelidae living in subtropical areas of Brazil. Temperature and rainfall appear to be the major factors influencing the fluctuation of these three species.  相似文献

Plasmodium vivax is responsible for the majority of malaria cases outside Africa. Unlike P. falciparum, the P. vivax life-cycle includes a dormant liver stage, the hypnozoite, which can cause infection in the absence of mosquito transmission. An effective vaccine against P. vivax blood stages would limit symptoms and pathology from such recurrent infections, and therefore could play a critical role in the control of this species. Vaccine development in P. vivax, however, lags considerably behind P. falciparum, which has many identified targets with several having transitioned to Phase II testing. By contrast only one P. vivax blood-stage vaccine candidate based on the Duffy Binding Protein (PvDBP), has reached Phase Ia, in large part because the lack of a continuous in vitro culture system for P. vivax limits systematic screening of new candidates. We used the close phylogenetic relationship between P. vivax and P. knowlesi, for which an in vitro culture system in human erythrocytes exists, to test the scalability of systematic reverse vaccinology to identify and prioritise P. vivax blood-stage targets. A panel of P. vivax proteins predicted to function in erythrocyte invasion were expressed as full-length recombinant ectodomains in a mammalian expression system. Eight of these antigens were used to generate polyclonal antibodies, which were screened for their ability to recognize orthologous proteins in P. knowlesi. These antibodies were then tested for inhibition of growth and invasion of both wild type P. knowlesi and chimeric P. knowlesi lines modified using CRISPR/Cas9 to exchange P. knowlesi genes with their P. vivax orthologues. Candidates that induced antibodies that inhibited invasion to a similar level as PvDBP were identified, confirming the utility of P. knowlesi as a model for P. vivax vaccine development and prioritizing antigens for further follow up.  相似文献

Two divergent reports have been published on the genetic complementation of rhizobial nod mutants using Frankia DNA. In 1991 putative Frankia cosmid library clones were reported to restore normal nodulation properties to Rhizobium leguminosarum biovar viciaenodD::Tn5, but no supporting sequence data were published. In 1992 a second group reported a failure to find any evidence of functional complementation of various rhizobial nod mutants by Frankia DNA (nodA, nodB and nodC). Complementation tests of nine Nod? R. leguminosarum bv. viciae or Sinorhizobium meliloti Tn5 mutants (nodA ? , nodB ? , nodC ? , nodD ? , nodF? ? , nodL ? , nodH ? ) were thus performed using a Frankia gene library in pLAFR3 to clarify this situation. Rhizobial transconjugants obtained by tri-parental matings were screened for restoration of the nodulation phenotype on their host plants, Vicia sativa subsp. nigra or Medicago sativa. Nodulation was observed on plants inoculated with transconjugants of the R. leguminosarum bv. viciaenodC::Tn5 mutant. The Nod+ rhizobial transconjugants were isolated and analysed. The Nod+ phenotype of these transconjugants was found to be due to Tn5 excision/transposition. No functional complementation was found with any of the mutants used, suggesting that rhizobial complementation of nod mutants with Frankia DNA is unlikely to occur.  相似文献

Two new species of oribatid mites of the genus Scapheremaeus (Oribatida, Cymbaeremaeidae), Scapheremaeus gibbus sp. n. and Scapheremaeus luxtoni sp. n., are described from New Zealand. Scapheremaeus gibbus sp. n. is morphologically most similar to Scapheremaeus humeratus Balogh & Mahunka, 1967, but differs from the latter by the number of notogastral, genital and adanal setae, morphology of bothridial setae, position of adanal lyrifissures and absence of humeral processes. Scapheremaeus luxtoni sp. n. is morphologically most similar to Scapheremaeus yamashitai Aoki, 1970, but differs from the latter by the morphology of notogastral and rostral setae, morphology of leg solenidia φ2 and development of humeral processes. The species Scapheremaeus zephyrus Colloff, 2010 is recorded for the first time in New Zealand. An identification key to the known New Zealand species of Scapheremaeus is provided.  相似文献

We investigated the role of Drosophila larva olfactory system in identification of congeners and aliens. We discuss the importance of these activities in larva navigation across substrates, and the implications for allocation of space and food among species of similar ecologies. Wild type larvae of cosmopolitan D. melanogaster and endemic D. pavani, which cohabit the same breeding sites, used species-specific volatiles to identify conspecifics and aliens moving toward larvae of their species. D. gaucha larvae, a sibling species of D. pavani that is ecologically isolated from D. melanogaster, did not respond to melanogaster odor cues. Similar to D. pavani larvae, the navigation of pavani female x gaucha male hybrids was influenced by conspecific and alien odors, whereas gaucha female x pavani male hybrid larvae exhibited behavior similar to the D. gaucha parent. The two sibling species exhibited substantial evolutionary divergence in processing the odor inputs necessary to identify conspecifics. Orco (Or83b) mutant larvae of D. melanogaster, which exhibit a loss of sense of smell, did not distinguish conspecific from alien larvae, instead moving across the substrate. Syn 97CS and rut larvae of D. melanogaster, which are unable to learn but can smell, moved across the substrate as well. The Orco (Or83b), Syn 97CS and rut loci are necessary to orient navigation by D. melanogaster larvae. Individuals of the Trana strain of D. melanogaster did not respond to conspecific and alien larval volatiles and therefore navigated randomly across the substrate. By contrast, larvae of the Til-Til strain used larval volatiles to orient their movement. Natural populations of D. melanogaster may exhibit differences in identification of conspecific and alien larvae. Larval locomotion was not affected by the volatiles.  相似文献

Five species of Ooctonus Haliday (Hymenoptera, Mymaridae) from Heilongjiang Province, China, are reviewed. One species, Ooctonus huberi sp. n., is described as new, and four species, Ooctonus orientalis Doutt, Ooctonus saturn Triapitsyn, Ooctonus sublaevis Förster and Ooctonus vulgatus Haliday are reported as new to China. A key to the females of the 10 described Chinese species is given. All the specimens are deposited in the insect collections of Northeast Forestry University, China.  相似文献

Coexistence of closely related species may be promoted by niche differentiation or result from interspecific trade-offs in life history and ecological traits that influence relative fitness differences and contribute to competitive inequalities. Although insufficient to prove coexistence, trait comparisons provide a first step to identify functional differences between co-occurring congeneric species in relation to mechanisms of coexistence. Here, a comparative review on life history and ecological traits is presented for two pairs of co-occurring species of spiny lobsters in the genus Panulirus: Panulirus gracilis and Panulirus inflatus from the Eastern Central Pacific region, and Panulirus argus and Panulirus guttatus from the Caribbean region. Panulirus gracilis and Panulirus inflatus have similar larval, postlarval, and adult sizes and a similar diet, but differ in degree of habitat specialization, fecundity, and growth rate. However, little is known on behavioral traits of these two species that may influence their competitive abilities and susceptibility to predators. The more abundant information on Panulirus argus and Panulirus guttatus shows that these two species differ more broadly in degree of habitat specialization, larval, postlarval and adult sizes, diet, fecundity, growth rate, degree of sociality, defense mechanisms, susceptibility to predators, and chemical ecology, suggesting a greater degree of niche differentiation between Panulirus argus and Panulirus guttatus than between Panulirus gracilis and Panulirus inflatus. Whether the substantial niche differentiation and apparent interspecific trade-offs between Panulirus argus and Panulirus guttatus relative to Panulirus gracilis and Panulirus inflatus reflect an earlier divergence of the former pair of species in the evolution of the genus constitutes an intriguing hypothesis. However, whether or not post-divergence evolution of each species pair occurred in sympatry remains uncertain.  相似文献

Five bee-fly species (Bombyliidae, Diptera) have been listed in this paper as new to the Kingdom of Saudi Arabia. Four of the recorded species have been identified to the level of species, namely: Bombomyia discoidea (Fabricius, 1794), Spogostylum candidum (Sack, 1909), Exoprosopa linearis Bezzi, 1924, and Exoprosopa minos (Meigen, 1804), while the fifth one only to genus, Desmatoneura sp. The species have been collected from Al-Baha and Asir Provinces in the south-western part of the Kingdom. One of the four identified species, Exoprosopa linearis, has an Afrotropical affinity, and another two, Spogostylum candidum and Bombomyia discoidea, have considerable Afrotropical distributions, and this result agrees to some extent with studies considering these parts of the Arabian Peninsula, including Al-Baha and Asir Provinces, having Afrotropical influences and may be included in the Afrotropical Region rather than in the Palaearctic Region or the Eremic zone.  相似文献

Heteranassa Smith (Erebidae, Omopterini), native to the southwestern United States and Mexico, includes two recognized species, namely Heteranassa mima (Harvey) and Heteranassa fraterna Smith. These are separated mainly by subtle differences in wing color and pattern, leading to speculation about the validity of the described species. This study examines variation in external and internal morphology across the geographic range of the genus, aiming to clarify species limits, describe morphology, and provide a comprehensive assessment of variation within the genus. Results indicate that Heteranassa fraterna syn. n., is a junior synonym of Heteranassa mima.  相似文献

The Liolaemus nigroviridis group is a clade of highland lizards endemic to Chile. These species are distributed from northern to central Chile, and currently there are no cases of sympatric distribution. This study describes a new species, Liolaemus uniformis sp. n., from this group, and provides a detailed morphological characterization and mitochondrial phylogeny using cytochrome-b. Liolaemus uniformis was found in sympatry with Liolaemus nigroviridis but noticeably differed in size, scalation, and markedly in the color pattern, without sexual dichromatism. This new species has probably been confused with Liolaemus monticola and Liolaemus bellii, both of which do not belong to the nigroviridis group. The taxonomic issues of this group that remain uncertain are also discussed.  相似文献
