For studying the interactions of the cholinergic and noradrenergic systems in the central nervous system rats were given muscarine or nicotine and the peripheral receptors were blocked with atropine or pempidine. After killing the animals the level of biogenic amines was determined in various parts of the brain. Evident changes were found in the levels of biogenic amines after stimulation or blockade of muscarinic or nicotinic receptors.  相似文献

Investigations were carried out on the role of biogenic amines in the development of glomerulonephritis in rabbits using the experimental model of acute and chronic serum sickness. The animals immunized with foreign-species albumin received additionally 5HT and some of them were given reserpine or PCPA for reducing the level of biogenic amines. The results of histological examinations demonstrated a clear cut inhibitory effect of PCPA and reserpine on the development of inflammatory changes in the glomeruli, and their intensification with simultaneous 5HT administration. The obtained results seem to suggest a significant role of biogenic amines in the pathogenesis of inflammatory lesions developing in renal glomeruli in acute or chronic serum sickness in rabbits.  相似文献

The effects of mutations that change the level of biogenic amines (octopamine and dopamine) on stress-reactivity and fitness of Drosophila imagoes are considered. It has been shown that (1) the biogenic amines represent an important but not triggering factor of the development of stress reaction; (2) under normal conditions, reproduction is regulated by genes that control dopamine metabolic pathways (indirectly via regulation of the juvenile hormone level). Under unfavorable conditions, reproduction is regulated by genes that control octopamine metabolism; (3) heat-stress adaptation depends on expression of genes controlling the background level of dopamine.  相似文献

The peculiarities of oriental research behavior and correlation between activities of catecholaminergic and serotoninergic brain systems dependently on their stress resistance degree were investigated in Wistar rats. The experiments showed that resistant (R) to a sound stimulus rats differed from nonresistant (NR) ones by increased research activity under moderate stress in the open field test and by decreased research behavior level in the cell chamber test. The biochemical analysis of biogenic amines in different brain structures revealed increased norepinephrine levels in stress R rats, and increased dopamine and serotonin levels in NR ones. The findings suggest that inborn behavior regulation in animals with different stress resistance is determined by different correlations between serotonin-dopamine and norepinephrine brain systems activities.  相似文献

By means of fluorescent method biogenic amines in the thymus tissue basophils have been investigated in intact and immunized rats and, simultaneously, in the peripheral blood of the immunized animals the hemolysins titer has been defined as an index of the immune response of the organism. In the immunized rats the number of the tissue basophils is increasing, there are many degranulated forms among them. Hence, the antigenic stimulation facilitates to the tissue basophils secretory activation, which is morphologically manifested as their degranulation. The hemolysins titer level in the peripheral blood is gradually decreasing, as the time of immunization increases. The content of biogenic amines in the tissue basophils does not undergo similar changes.  相似文献

The processes of serotonin and histamine absorption and release by the lungs were studied in dogs during 1 to 3.5 hour hypovolemic hypotension and during 24 hours after blood retains fusion. Absorption of biogenic amines by the lungs tended to increase in all the animals under hypovolemic hypotension. In the group of non-survivors the serotonin absorption by the lungs in the post-terminal period remained increased, while in the group of survivors it came down to normal soon, though the histamine release was increased. The above processes were aggravated in the group of animals whose lungs were affected by oleic acid. It resulted in the absorption of histamine instead of its release. The intensified absorption of biogenic amines by the lungs was accompanied by a quick fall in cardiac output, by the increase in resistance of systemic and pulmonary circulation.  相似文献

The changes of photo- and peroxide resistance of erythrocytic and lymphocytic cells of human under influence of UV and some active oxygen forms in presence of biogenic amines such as serotonin, dopamine, adrenalin, histamine had been investigated. While investigating the degree of photohemolysis of erythrocytes it was discovered that biogenic compounds raise UV stability of erythrocytic membranes. By using the method of chemiluminescence it was established that biogenic amines increased the degree of peroxide resistance of human erythrocytic and lymphocytic cells. The decrease of the level of erythrocytic diene conjugates under influence of UV by serotonin and histamine was also discovered.  相似文献

The content of some biogenic monoamines and their metabolites in rat brain and heart in different periods of oxygen epilepsia was studied using high performance liquid chromatography with electrochemical detection. It was shown that already at the 5th minute of exposure to oxygen adrenaline, DOPA and some noradrenaline metabolites disappeared in the brain and noradrenaline level reduced. At this period in rat heart the reduction of catecholamine content was the most distinct and serotonin level was unchanged. At the beginning of convulsive period the modifications of biogenic amines content were nonparallel in brain regions: in the heart the reduction of catecholamine level went on, especially in right ventricle. In the terminal phase of oxygen epilepsia brain biogenic amines increased, however, not up to normal meaning, heart catecholamines at this period were at the same level as at the beginning of the convulsive period.  相似文献

In vivo microdialysis was used to determine biogenic amines in medial prefrontal cortex of rats exposed to eight minutes of swim stress on two consecutive days. On the first day of stress, norepinephrine (NE) efflux increased by 183% over baseline after stress, while dopamine (DA) and serotonin (5-HT) remained stable throughout. On the second day of stress, a robust increase was observed in all 3 neurotransmitters measured, with (NE), (DA), and (5-HT) increasing by 310%, 441% and 496% respectively, and remaining elevated for an hour or more after stress. This suggests that the first exposure to swim stress, while not causing dramatic changes in biogenic amine release, may sensitize biogenic amines in medial prefrontal cortex to subsequent swim stress. Our results also serve as preliminary data concerning the neurochemical changes which might underlie the forced swimming model of behavioral despair.  相似文献

Levels of brain neurotransmitters and their metabolites, as well as concentrations of enzymes associated with their synthesis and metabolism, fluctuate during the day in patterns defined as circadian. The present study examined these rhythms in albino rats exposed to 60-Hz electric fields. Thirty-six animals were exposed to a 39 kV/m field for 4 weeks, 20 h/day, in a parallel-plate electrode system. A group of 36 sham animals was similarly handled and housed in a nonenergized exposure system. On the sampling day, animals were sacrificed at 4-h intervals throughout the 24-h day. Brains were removed, dissected, and kept frozen until chemically analyzed. The levels of biogenic amines and their acidic metabolites in the striatum, hypothalamus, and hippocampus were determined by high-performance liquid chromatography with electrochemical detection (HPLC-ECD) methods. Repeated exposure to 60-Hz electric fields produced significant alterations in the diurnal rhythms of several biogenic amines: dihydroxyphenylacetic acid (DOPAC, the primary metabolite of dopamine in the rat) in the striatum, and norepinephrine, dopamine, and 5-hydroxyindoleacetic acid (5-HIAA; serotonin metabolite) in the hypothalamus. Levels of serotonin in the striatum and hypothalamus showed clear circadian patterns that was not affected by the field. No diurnal or field-related changes were observed in the hippocampal amines.  相似文献

棘头梅童鱼(Collichthys lucidus)和龙头鱼(Harpodon nehereus)是我国沿海常见的两种小型海水鱼, 常被作为水产动物饵料, 也可被人类食用。研究检测了这两种鱼在30℃下贮存48h 每隔6h 的挥发性盐基氮(T-VBN)和9 种生物胺(尸胺、腐胺、组胺、酪胺、5-羟色胺、亚精胺、精胺、多巴胺、章鱼胺)的含量变化, 并对这两种鱼的T-VBN 和生物胺含量与时间的相关性进行分析, 为水产品类饵料安全投喂和人类食品安全提供基础资料。结果表明: 两种鱼在相同贮存条件中T-VBN 和生物胺含量均存在一定差异。T-VBN 含量随着贮存时间的延长而逐渐增加, 棘头梅童鱼T-VBN 含量从0h 的8.19 mg/100 g 增加到48h 的568.05 mg/100 g,龙头鱼从0h 的13.16 mg/100 g 增加到48h 的361.34 mg/100 g, 棘头梅童鱼增长值显著高于龙头鱼(PPPP>0.05);多巴胺在两种鱼体内均未检测到。这两种鱼体内T-VBN、腐胺、尸胺、组胺、酪胺含量与时间的相关性均极其显著(P<0.01)。    相似文献

In vivo microdialysis was used to determine the effect of diazepam, flumazenil and FG-7142 upon the biogenic amine response to acute and repeated swim stress in the medial prefrontal cortex of the rat. Acute swim stress increased norepinephrine levels, although dopamine and serotonin levels remained stable. Upon re-exposure to swim stress twenty-four hours later, sustained increases (200–300% of baseline) in all three biogenic amines were detected. This enhanced response to re-stress was not seen in rats pretreated with either a benzodiazepine agonist (diazepam, 2 mg/kg), an antagonist (flumazenil, 10 mg/kg), or an inverse agonist (FG-7142, 10 mg/kg) given prior to the first swim stress. Therefore, the sensitization of biogenic amine response to re-stress may be prevented by compounds which differ in their activity at the benzodiazepine receptor.  相似文献

具有脱羧酶活性的乳酸菌可通过氨基酸的脱羧反应产生具有潜在安全风险的生物胺。本文利用脱羧酶培养基初步筛查61株乳酸菌产生物胺情况,再通过RT-HPLC法测定其在发酵液和发酵乳中的生物胺含量。用10%的三氯乙酸提取样品中的生物胺,采用苯甲酰氯衍生处理后,以甲醇/水为流动相,进行梯度洗脱,流速0.8mL/min,紫外检测器波长为254nm。结果显示,组胺和酪胺得到良好的分离,在给定的浓度范围内呈现良好的线性关系(R20.995)。在发酵液和发酵乳中添加生物胺混合标准溶液,平均回收率为97.92%-101.14%,相对偏差RSD5%。结果表明,发酵液与发酵乳中生物胺的RT-HPLC法,是一种快捷、稳定、灵敏度高的检测方法,其与脱羧酶培养基法结合可以准确地实现对乳酸菌产生物胺的评价。  相似文献

Earlier we have shown that some non-hormonal activators of adenylyl cyclase (AC) and hormones of higher vertebrate animals are able to affect functional activity of the AC system in the infusorian Dileptus anser. In the present work, sensitivity of this infusorian AC to Ca2+ was studied and it was found that calcium cations at concentrations of 0.5-10 microM stimulated significantly the enzyme activity in D. anser partially purified membranes. An increase of Ca2+ concentrations to 100 microM and higher led to the complete block of their stimulatory effect. In the EDTA-treated membranes the enzyme activity was reduced markedly, but it was restored significantly by addition of Ca2+. Calmodulin antagonists--chlorpromazine, W-7, and W-5--caused a dose-dependent decrease of the enzyme activity stimulated by 5 microM Ca2+ with IC50 values of 35, 137, and 174 microM, respectively. The AC-stimulating effects of biogenic amines (serotonin and octopamine) were completely retained in the presence of 2.5 and 100 microM Ca2+, whereas effects of peptide hormones (relaxine and EGF) were hardly changed in the presence of 2.5 microM calcium ions, but were markedly inhibited by 100 microM Ca2+. In the EDTA-treated membranes, the AC effects of biogenic amines were reduced, while the effects of peptide hormones were not revealed. On addition of Ca2+, the AC effects of biogenic amines were completely restored, whereas the effects of peptide hormones were not detected or were restored to a non-significant degree. Calmodulin antagonists slightly affected the AC effects of peptide hormones at concentrations efficient in the case of vertebrate AC, but decreased them markedly at higher concentrations. The AC effects of biogenic amines were little sensitive even to high antagonist concentrations. The obtained data show that targets of action of peptide hormones in the infusorian D. anser cell culture are the AC forms whose activity does not D. depends on calcium cations and possibly is regulated by Ca2+/calmodulin, whereas targets of action of biogenic amines are calcium-independent enzyme forms.  相似文献

Summary The sulphur-containing radioprotectors mercaptoethylamine (MEA), aminoethylisothiourea (AET), 2-aminothiazoline, 4-oxo-2-aminothiazoline, and S-S-3-oxapentane-1,5-diisothiourea, and the radioprotective biogenic amines serotonin, histamine, and dopamine, caused the elevation of cAMP content and intensified the rate of cAMP-dependent protein phosphorylation in tissues of animals following intraperitoneal injection at radioprotective doses. Biogenic amines stimulated the adenylate cyclase activity in membrane preparations from liver, spleen, and small-intestine mucosa; sulphur-containing radioprotectors caused no such effects. None of the radioprotectors affected cAMP and cGMP phosphodiesterases in vitro. AET and MEA inhibited guanylate cyclase in vitro, whereas serotonin and dopamine stimulated the enzyme. A biphasic change in the level of cGMP was observed in tissues after the administration of MEA and AET (more than 2-fold fall by 1–3 min after the administration of drug and 1.4-fold rise after 15–20 min); serotonin and dopamine caused a slow rise in the cGMP level; the cAMP/cGMP ratio in liver showed biphasic changes in level during the 20 min following injection of serotonin.The data obtained support the conclusion that the action of radioprotectors on cellular metabolism in animals may be mediated by the cAMP system. The reciprocal regulation of radioresistance by cAMP and cGMP is unlikely to exist.  相似文献

Biogenic amine production by lactic acid bacteria isolated from cider   总被引:1,自引:0,他引:1  
AIMS: To study the occurrence of histidine, tyrosine and ornithine decarboxylase activity in lactic acid bacteria (LAB) isolated from natural ciders and to examine their potential to produce detrimental levels of biogenic amines. METHODS AND RESULTS: The presence of biogenic amines in a decarboxylase synthetic broth and in cider was determined by reversed-phase high-performance liquid chromatography (RP-HPLC). Among the 54 LAB strains tested, six (five lactobacilli and one oenococci) were biogenic amine producers in both media. Histamine and tyramine were the amines formed by the LAB strains investigated. Lactobacillus diolivorans were the most intensive histamine producers. This species together with Lactobacillus collinoides and Oenococcus oeni also seemed to produce tyramine. No ability to form histamine, tyramine or putrescine by Pediococus parvulus was observed, although it is a known biogenic amine producer in wines and beers. CONCLUSIONS: This study demonstrated that LAB microbiota growing in ciders had the ability to produce biogenic amines, particularly histamine and tyramine, and suggests that this capability might be strain-dependent rather than being related to a particular bacterial species. SIGNIFICANCE AND IMPACT OF THE STUDY: Production of biogenic amines by food micro-organisms has continued to be the focus of intensive study because of their potential toxicity. The main goal was to identify the microbial species capable of producing these compounds in order to control their presence and metabolic activity in foods.  相似文献

A study of the action of iproniaside on alimentary conditioning has shown that even its small doses (25 mg/kg) disturb the formation of the conditioned reaction, while large doses (200-250 mg/kg) do not disturb the reproduction of the conditioned reaction elaborated and stabilized before the administration of the drug. Hence, dissociated learning with the use of iproniaside is impossible. The applied doses of iproniaside result in an increased level of biogenic amines in the dopaminergic nigro-neostriate and reticulo-septal brain systems. It is therefore assumed that the effects of iproniaside on learning are due to its influence on the level of the CNS biogenic amines.  相似文献

EDAC fixation, which polymerizes biogenic amines, and clorgyline, which blocks monoamine oxydase, were used to demonstrate the perinuclear and nuclear localization of histamine and serotonin (5-HT) of immune cells in rat peritoneal fluid and thymus. For detection, an immunocytochemical method combined with confocal microscopy was used. In peritoneal cells (lymphocytes) 5-HT formed spots around the nucleus, while histamine was localized diffusely. In thymocytes, 5-HT was distributed in patches, while histamine formed a garland around the nuclear envelope. Clorgyline enhanced the fluorescence and increased the number of fluorescent positive cells only for histamine, and then histamine-positive nuclei were also present. The results show that: (1) EDAC fixation demonstrates the presence of biogenic amines in immune cells, (2) using this fixation, the perinuclear localization of biogenic amines can be demonstrated, (3) the localization of the two amines is different and (4) the nuclear localization of histamine in thymocytes can be surmised and in mast cells (described previously) is proven again. The potential importance of the nuclear and near-nuclear localization of biogenic amines is discussed.  相似文献

Aims: To develop rapid methods allowing enumeration of lactic acid bacteria producing biogenic amines in wines and to analyse wine samples by the methods. Methods and Results: Methods based on quantitative PCR targeting bacterial genes involved in histamine, tyramine and putrescine production were developed and applied to detect and quantify the bacteria producing these biogenic amines in wine. Analysis of 102 samples revealed low populations of the targeted bacteria in grape must samples, an increased bacteria biomass in wine samples after alcoholic fermentation, reaching the highest population levels (above 106 cells ml?1) during spontaneous malolactic fermentation. A minimum of 103 ml?1 producing cells was required for production of more than 1 mg l?1 of biogenic amines. Accumulation of putrescine in wine was correlated with the presence of bacteria carrying an ornithine decarboxylation pathway. Trials of winemaking showed that the use of selected bacteria for inducing malolactic fermentation was efficient to limit the proliferation of undesirable bacteria and the production of biogenic amines. Conclusion: Methods using quantitative PCR are efficient to enumerate biogenic amines‐producing cells in wine. Significance and Impact of the Study: The methods can help to better control and to improve winemaking conditions in order to avoid biogenic amine production.  相似文献
