Vertical polyacrylamide gel electrophoresis was used to separate enzyme proteins at 73 putative loci in natural house fly populations sampled in central Iowa. Thirty-nine of the loci were polymorphic (53%). The mean effective number of alleles per polymorphic locus was 1.93 and 1.47 alleles among 68 scored loci. Observed and expected heterozygosities at 34 house fly loci were 0.1628 and 0.1834, respectively. No statistically significant differentiation was detected among nine central Iowa fly populations in 1989 or among nine Iowa and three Minnesota populations in 1990. The LDH isozymes were surveyed in bacterized cultures of syngens 1, 3, 12, and 13 of Paramecium caudatum by polyacrylamide gel electrophoresis. All the examined stocks of different syngens except one stock in syngen 3 had a single common LDH isozyme, and intra- and intersyngenic variation was not observed except for the one stock in syngen 3. Breeding data using the exceptional stock indicated that the LDH isozymes of P. caudatum are controlled by two codominant alleles at a single locus whose products aggregate randomly, forming a dimer.  相似文献

Adult house flies, Musca domestica L., of four ages, <1, 3, 7, and 14 day post-eclosion, were exposed to three strains of Beauveria bassiana (P89, L90 and 447). Flies were exposed to moistened filter paper treated with either a low (1.57×104 conidia/cm2) or high (1.57×105 conidia/cm2) concentration of each fungal strain for 6 h. Strain 447 was superior to the two house fly-derived B. bassiana strains in inducing host infection and mortality. Significant spikes in infection and mortality occurred as early as 5 days post-exposure with higher concentration exposures acting more quickly. Few differences were observed in either infection or mortality among the four fly age classes. On Day 10 post-exposure, 77% of the high-concentration, 447-exposed flies were infected, compared with only 24% of the flies from the P89 low-concentration exposure. Potential applications of these results in integrated house fly management programs are discussed.  相似文献

Abstract. The genetic basis of variation in local searching behaviour in the house fly, Musca domestica L., was examined by estimating the heritability of locomotory parameters using parent—offspring regressions. Pathways of adult flies were videotaped for 3 min after ingestion of a 1 μl drop of 0.25 M sucrose in a 75 cm diameter circular arena with minimal external cues. Locomotory variables computed from digitized walking pathways, including locomotory rate, turning rates per unit of distance and time, turn bias, number of stops, and stop duration, were significantly heritable, but heritabilities (additive genetic variance ÷ total phenotypic variance) was generally low (<0.25). Phenotypic variance in search parameters increased with time since ingestion of the sucrose drop, but heritabilities were highest when only the first 30 s of the searching bout were examined, and declined as longer periods were considered. Significant genetic correlations among search parameters suggest that many of these characters are influenced pleiotropically by the same genes or closely linked genes. However, some aspects of local search were inherited independently (e.g. stop duration and locomotory rate). The independence of controlling mechanisms was supported by observed differences between genetic and environmental correlations. This genetic independence could enhance the likelihood of appropriate population responses to temporally or spatially changing selective forces.  相似文献

拟除虫菊酯对家蝇Ca—ATPase和Ca—Mg—ATPase的抑制作用   总被引:2,自引:4,他引:2  
通过对家蝇神经系统的Ca-ATPase、Ca-Mg-ATPase性质的研究,表明Ca-ATPase、Ca-Mg-AT-Pase反应的适宜pH值分别为7.0-8.5和6.5。适温皆为35-40℃;底物(ATP)最适浓度均为0.5mmol/L。比较测定了家蝇三个品系中两种ATPase的活性及拟除虫菊酯对该酶的抑制作用,实验证明,敏感与Del-R、2Cl-R品系间Ca-ATPase、Ca-Mg-ATPase活力无明显的差异。溴氰菊酯、氯菊酯可部分地抑制敏感品系家蝇Ca-ATPase活性,而对拟除虫菊酯抗性品系Ca-ATPase无抑制作用,从而证明,Del-R、2Cl-B品系Ca-ATPase对拟除虫菊酯的敏感性已明显降低,这可能能是击倒抗性机制之一。实验还表明,拟除虫菊酯对Ca-Mg-ATPase基本上无抑制作用,这说明在家蝇中,Ca-Mg-ATPase并不是拟除虫菊酯的一个靶标位点。  相似文献

ABSTRACT. Potentiation in joint action was demonstrated between solutions of L-leucine and sodium phosphate buffer (pH 6.3) as feeding stimulants for protein-deprived females of the house fly, Musca domestica L. Both components alone elicited feeding. In two-choice feeding tests, mixtures consisting of equi-stimulating concentrations of the two components were taken in greater quantities than either component alone at twice the concentration in the mixture.
The presence of 1×10-1 M phosphate buffer markedly lowered the threshold for detection of L-leucine. The presence of phosphate buffer strengthened the preferences shown by flies given choices of concentrations of L-leucine differing by a factor of 2 and enabled them to display preferences at lower concentrations.
The presence of 1×10-3 M L-leucine increased, somewhat, the ability of flies to detect low concentrations of phosphate buffer. Its presence had relatively little effect on the strength of preference shown between two-fold differences in concentration of phosphate buffer when the higher concentration was 6.3×10-3 M or less, but markedly strengthened the preferences when the higher concentration was 2.5×10-2M or greater. Leucine increased the optimal concentration of phosphate buffer by a factor of more than 2 and converted 2×10-1 M phosphate buffer from a mild feeding deterrent to a powerful feeding stimulant.  相似文献

Methods are described for the resolution of house fly, Musca domestica L., enzymes by vertical polyacrylamide gel electrophoresis. An electrophoretic survey in Ames, Iowa, of 51 loci distributed among 26 enzyme systems revealed that 40% of the loci are polymorphic. Observed and expected heterozygosities measured at 33 loci were 0.0981 and 0.1148, respectively. Epicuticular lipids serve two major roles in insects. Their waterproofing properties are crucial to survival in terrestrial environments, and they serve as contact pheromones in a wide array of taxa. Both functions may be affected by the physical properties of the surface lipids. This provides the opportunity for natural selection on water conservation, mediated by lipid phase behavior, to interact with and perhaps conflict with sexual selection on communication and mate recognition. We used the common house fly, Musca domestica, as a model for these interacting selective forces. Male house flies preferred female models treated with a high melting-point lipid mixture, suggesting that sexual and natural selection may both act to favor longer-chain, more saturated hydrocarbons. However, higher melting points did not result in lower rates of water loss. We propose a working model in which phase separation between the unsaturated female pheromone and saturated hydrocarbons results in areas of melted, pheromone-rich lipids and regional variation in cuticular permeability.  相似文献

Parasitoid development, parasitoid-induced host mortality and parasitoid progeny emergence were determined at five constant temperatures for Muscidifurax raptor Girault and Sanders, Muscidifurax zaraptor Kogan and Legner, Spalangia cameroni Perkins and Spalangia endius Walker using pupae of the house fly, Musca domestica L., as hosts. At temperatures of 20, 25, 30 and 35 degrees C the median development times (days from oviposition to adult emergence), respectively, were M. raptor (28.4, 20.7, 14.3, 14.5), M. zaraptor (30.6, 22.8, 14.1, 14.2), S. cameroni (55.6, 35.2, 21.8, 25.0) and S. endius (52.4, 31.5, 16.3, 14.6). All species failed to emerge at 15 degrees C. Using densities of five parasitoids and 100 hosts and a 24 h exposure period, Muscidifurax species oviposited at a greater rate over a wider range of temperatures than Spalangia species. At 15, 20, 25, 30 and 35 degrees C the mean number of pupae killed per parasitoid were, respectively, M. raptor (1.4, 7.4, 10.5, 13.7, 14.1), M. zaraptor (0.0, 3.3, 8.9, 14.4, 15.0), S.cameroni (0.0, 7.8, 11.0, 11.9, 7.4), S.endius (0.6, 4.0, 7.5, 12.0, 11.7), and means of the number of parasitoid progeny per parasitoid were, respectively, M.raptor (0.2, 5.2, 7.9, 11.8, 11.6), M.zaraptor (1.3, 4.4, 8.2, 13.0, 13.7), S.cameroni (0.0, 2.4, 4.7, 5.1, 1.0), S.endius (0.0, 0.9, 3.4, 7.5, 4.9). Development and ovipositional activity in S.cameroni was strongly inhibited at 35 degrees C. The model by Sharpe & DeMichele (1977) was used to describe temperature-dependent development and the number of parasitoid progeny produced per parasitoid at temperatures of 15-30 degrees C in all species.  相似文献

Resistance to the organophosphate insecticide tetrachlorvinphos was examined in a house fly (Musca domestica L.) strain with an altered acetylcholinesterase (AChE) of decreased sensitivity to inhibition by the insecticide. Genetic tests showed that both resistance and the altered AChE were controlled by semidominant gene(s) on chromosome II. The gene for resistance was five crossover units from the mutant marker stubby wing (stw). A house fly strain was prepared in which resistance was introduced in to a susceptible stw strain by recombination. Biochemical assays revealed that the altered AChE was introduced along with resistance. Assays of the AChE of resistant and susceptible stw strains by two independent methods showed that the enzyme from resistant flies was 30 times more slowly inhibited by tetrachlorvinphos than the enzyme from susceptible flies.This work was supported in part by NIH Grant ES 00901.Technical Article 13340, Texas Agricultural Experiment Station.  相似文献

Parasitoid-induced mortality of house fly, Musca domestica L., pupae and parasitoid progeny emergence by four species of pteromalid parasitoids, Muscidifurax raptor Girault & Sanders, M.zaraptor Kogan & Legner, Spalangia cameroni Perkins and S.endius Walker, were determined for a 24 h exposure period using parasitoid: host ratios ranging from 1:2 to 1:50. When the number of parasitoids was held constant (n = 5) and the numbers of hosts varied, and when the number of hosts was held constant (n = 100) and the number of parasitoids varied, both the number of pupae killed per parasitoid and the number of parasitoid progeny per parasitoid increased with increasing parasitoid:host ratios to reach an upper limit asymptotically. Maximum values were, respectively: M.raptor (14.7, 11.1), M.zaraptor (12.3, 9.3), S.cameroni (16.9, 5.5), S.endius (14.8, 9.7) with no consistent effects attributed to parasitoid interference. For M.raptor and S.cameroni at parasitoid:host ratios of 1:10, the pupal mortality and progeny emergence were determined for a 24 h exposure period when hosts were distributed in poultry manure at four levels of aggregation ranging from clumped to uniform. Pupal mortality was least in clumped distributions, while parasitoid progeny emergence was not significantly different.  相似文献

Summary The terminal organs of the cephalic lobes of the house fly larva, Musca domestica L., were studied by scanning and transmission electron microscopy. Six different types of sensilla were found: (1) papilla sensillum, (2) pit sensillum, (3) spot sensillum, (4) modified papilla sensillum, (5) knob sensillum, and (6) scolopidium. The papilla, pit, spot, and modified papilla sensilla have the essential structure of contact chemoreceptors, i.e., the unbranched dendritic tips are exposed externally through a single opening. However, a tubular body, which is a characteristic structure of tactile setae, is also present in some of the dendritic tips. We assume these sensilla serve a dual function—contact chemo- and mechanoreception. The role of the knob sensilla is obscure. The scolopidia present in the dorsal and the terminal organ are probably stress detectors. Two basal bodies occur in the dendritic ciliary region of all sensilla. Summary The ventral organs of the cephalic lobes of the house fly larvae, Musca domestica L., were studied by scanning and transmission electron microscopy. Four sensilla were found. Three of them are each innervated by a single dendrite whose ending possesses a tubular body and communicates to the exterior through an opening. These sensilla are assumed to be mechanoreceptors. The 4th sensillum is supplied by 2 bipolar neurons with the unbranched dendritic tips (without tubular bodies) exposed to the exterior through a single opening and is probably a contact chemoreceptor.This research was supported in part by the Office of Naval Research and NIH Training Grant ES-00069.
