High-quality NMR structures of the C-terminal domain comprising residues 484–537 of the 537-residue protein Bacterial chlorophyll subunit B (BchB) from Chlorobium tepidum and residues 9–61 of 61-residue Asr4154 from Nostoc sp. (strain PCC 7120) exhibit a mixed α/β fold comprised of three α-helices and a small β-sheet packed against second α-helix. These two proteins share 29 % sequence similarity and their structures are globally quite similar. The structures of BchB(484–537) and Asr4154(9–61) are the first representative structures for the large protein family (Pfam) PF08369, a family of unknown function currently containing 610 members in bacteria and eukaryotes. Furthermore, BchB(484–537) complements the structural coverage of the dark-operating protochlorophyllide oxidoreductase.  相似文献

Vector pMPM‐A4Ω and vectors pQE‐30 and pET‐45b(+) containing the 6x His‐tag sequence were used for expression of Potato leafroll virus (PLRV) structural and non‐structural proteins in Escherichia coli. Coat protein (CP) and RNA‐dependent RNA polymerase (RdRp)–fragments RdRp43‐616 and RdRp304‐537 were chosen for expression. A high level of CP and RdRp304‐537 was obtained only in an expression system using pET‐45b(+) vector and E. coli Rosetta‐gami 2(DE3) cells. After purification, the His‐tagged PLRV proteins were used for immunization of rabbits.  相似文献

Summary A review of 569 male patients with X-chromosome polysomies (544 Klinefelter and 25 patients with other types of X-chromosome polysomy) is presented here. These patients were detected among the 77000 persons karyotyped in the Leuven cytogenetic center between the years 1966 and 1987. In the group of 544 Klinefelter patients special attention was paid to (1) the age at diagnosis, (2) social and marital status of the postpubertal males, (3) physical and intellectual abilities of the prepubertal boys, (4) delineation of the concurrence of Klinefelter syndrome and fragile X syndrome, and (5) the frequency of malignancies. In 25 patients with other X-chromosome polysomies (2 n48 chromosomes) genotype/phenotype correlation is reviewed, especially for the patients with 48,XXYY and 49,XXXXY karyotypes. Finally, double aneuploidy and rare structural X-chromosome aberrations are briefly discussed.  相似文献

Brucella as intracellular pathogen requires a coordinate interaction between Th1 subset of gamma interferon-secreting CD4 T cells and CD8 T cells for optimal protective immunity. It was previously recognized that L7/L12 as T cell-reactive antigen from the pathogen. On other hand, Omp25 was found as another antigen to provide protection against the Brucella infection by eliciting both Th1 and Th2 type of immune responses in mice. Here, we analyzed the prophylactic and therapeutic efficacy of a divalent fusion protein (rL7/L12-Omp25) comprising these two promising immunogens of Brucella in the presence of murine IFN-gamma in mice against B. abortus 544 challenge. rIFN-gamma with rL7/L12-Omp25 resulted in superior immune response when compared to the animal vaccine strain B. abortus S19. The vaccine candidate caused dominance of IgG1 over IgG2a and upregulated cytokine secretion (IFN-gamma, TNF-α, and IL-10) among immunized mice. Moreover, the antigen in combination with murine IFN-gamma elicited stronger cell-mediated immune response among the immunized animals when compared to standard vaccine (S19). The registered log protection unit among challenged mice with B. abortus 544 pathogen was 2.16, p = 0.0001 when rL7/L12-Omp25 was administered alone and 2.4, p = 0.0001 when it was administered along with rIFN-gamma. However, the molecule upon administration with murine IFN-gamma imparted very minimal or no therapeutic effect against brucellosis. To conclude, our study demonstrates the potential of rL7/L12-Omp25 as an immunogen of prospective and efficient prophylaxis as it is capable of eliciting both cell-mediated and humoral immune responses against brucellosis.

The human ether‐à‐go‐go related gene (hERG) encodes a protein that forms a voltage‐gated potassium channel and plays an important role in the heart by controlling the rapid delayed rectifier K+ current (IKr). The S4–S5 linker was shown to be important for the gating of the hERG channel. Nuclear magnetic resonance study showed that a peptide derived from the S4–S5 linker had no well‐ordered structure in aqueous solution and adopted a 310‐helix (E544‐Y545‐G546) structure in detergent micelles. The existence of an amphipathic helix was confirmed, which may be important for interaction with cell membrane. Close contact between side chains of residues R541 and E544 was observed, which may be important for its regulation of channel gating. BACKGROUND: ESR1 mutation in circulating cell-free DNA (cfDNA) is emerging as a noninvasive biomarker of acquired resistance to endocrine therapy, but there is a paucity of data comparing the status of ESR1 gene in cfDNA with that in its corresponding tumor tissue. The objective of this study is to validate the degree of concordance of ESR1 mutations between plasma and tumor tissue. METHODS: ESR1 ligand-binding domain mutations Y537S, Y537N, Y537C, and D538G were analyzed using droplet digital PCR in 35 patients with metastatic breast cancer (MBC) (35 tumor tissue samples and 67 plasma samples). RESULTS: Of the 35 paired samples, 26 (74.3%) were concordant: one patient had detectable ESR1 mutations both plasma (ESR1 Y537S/Y537N) and tumor tissue (ESR1 Y537S/Y537C), and 25 had WT ESR1 alleles in both. Nine (25.7%) had discordance between the plasma and tissue results: five had mutations detected only in their tumor tissue (two Y537S, one Y537C, one D538G, and one Y537S/Y537N/D538G), and four had mutations detected only in their plasma (one Y537S, one Y537N, and two Y537S/Y537N/D538G). Furthermore, longitudinal plasma samples from 19 patients were used to assess changes in the presence of ESR1 mutations during treatment. Eleven patients had cfDNA ESR1 mutations over the course of treatment. A total of eight of 11 patients with MBC with cfDNA ESR1 mutations (72.7%) had the polyclonal mutations. CONCLUSION: We have shown the independent distribution of ESR1 mutations between plasma and tumor tissue in 35 patients with MBC.  相似文献

Novel chiral diisopropyl spiro bis(isoxazoline) ligands, anti‐i‐Pr‐SPRIX and syn‐i‐Pr‐SPRIX, were designed and synthesized. Their catalytic utility, X‐ray crystallographic analyses, and complexation studies demonstrated the structural features of tetraisopropyl spiro bis(isoxazoline) ligand, i‐Pr‐SPRIX, which is a prominent ligand in various enantioselective Pd catalytic processes: All i‐Pr groups work in collaboration to create an effective asymmetric environment. Food utilization by larvae of the fall armyworm (Spodoptera frugiperda [J. E. Smith]) (Lepidoptera: Noctuidae), showed greater consumption of corn (Zea mays L.) than pinto bean diet, Tifton 10, or Coastal bermudagrass (Cynodon dactylon [L.] Pers.). Transfer of larvae from diet to susceptible grasses such as corn, Tifton 10 or Coastal produced differences in growth rates as a result of food consumption rates. Transfer of larvae from diet to resistant grasses such as common centipedegrass (Eremochola ophiuroides [Munro] Hack) Tifton 292 bermudagrass, and zoysiagrass (Zoysia sp.) reduced larval growth as a result of low consumption rates and/or greater metabolic expenditures. Larvae initially fed Tifton 10, Coastal, or centipedegrass before feeding on corn grew significantly faster than when they fed continuously on corn.
Résumé Des chenilles de S. frugiperda ont consommé plus de maïs (Zea mays), que d'un régime à base de Phaseolus vulgaris de la variété Pinto, ou de Tifton 10, ou de Cynodon dactylon de la variété Coastal. Des chenilles, transférées du régime artificiel sur des plantes sensibles comme le maïs, le Tifton 10 ou le Coastal, ont présenté des taux de croissance différents provenant de modifications de leur consommation. Le transfert du régime à des plantes résistantes, telles que Eremochola ophiuroides, Tifton 292, C. dactylon et Zoysia sp., a entraîné une diminution de la croissance larvaire par suite d'une plus faible consommation et/ou de dépenses métaboliques plus élevées. Des chenilles ayant consommé du Tifton 10, du Coastal ou de l'E. ophiuroides avant de consommer du maïs, se sont développées significativement plus vite que celles qui s'étaient alimentées continuellement sur maïs.

Structures of mitochondrial bc 1 complex have been reported based on four different crystalforms by three different groups. In these structures, the extrinsic domain of the Rieske [2Fe–2S]protein, surprisingly, appeared at three different positions: the c 1 position, where the [2Fe–2S]cluster exists in close proximity to the heme c 1; the b position, where the [2Fe–2S] clusterexist in close proximity to the cytochrome b; and the intermediate position where the[2Fe–2S] cluster exists in between c 1 and b positions. The conformational changes betweenthese three positions can be explained by a combination of two rotations; (1) a rotation of theentire extrinsic domain and (2) a relative rotation between the cluster-binding fold and thebase fold within the extrinsic domain. The hydroquinone oxidation and the electron bifurcationmechanism at the QP binding pocket of the bc 1 complex is well explained using theseconformational changes of the Rieske [2Fe–2S] protein.  相似文献

The genetic properties of 45 pseudomonad strains isolated from cereal cultures exhibiting symptoms of basal bacteriosis have been investigated. Considerable genetic diversity has been demonstrated using DNA fingerprints obtained by amplification with REP, ERIC, and BOX primers. Restriction analysis of the 16S–23S internal transcribed spacer (ITS1) allowed the strains to be subdivided into two major groups. In a phylogenetic tree, the ITS1s of these groups fell into two clusters, which also included the ITS1 of Pseudomonas syringae (“Syringae” cluster) and the ITS1 of P. fluorescens, P. tolaasii, P. reactans, P. gingeri, and P. agarici (“Fluorescens” cluster) from the GenBank database. Comparison of the ITS1 divergence levels within the “Fluorescens” cluster suggests expediency of treating P. tolaasii, P. reactans, various P. fluorescens groups, and, possibly, P. gingeri and P. agarici as subspecies of one genospecies. The intragenomic heterogeneity of ITS1s was observed in some of the pseudomonad strains studied. Summary Pleurotus ostreatus (444) and P. sajorcaju (537) were grown on a pasteurized mixture of chopped wheat straw (70%) and milled corncobs (30%) nonsupplemented and supplemented with two levels of delayed-release nutrient. Yields for both lines increased 2.3 and 3.2 fold on substrate containing 16% and 32% (dry weight basis) delayed-release nutrient additions, respectively. For mushroom size, a differential response was observed for genotype and delayed-release nutrient. Line 444, a wild isolate from Missouri, produced larger mushrooms on substrate containing higher levels of delayed-release nutrient. Line 537, a commercial isolate used in Italy, produced smaller mushrooms with higher levels of nutrient. The ionophores A23187 and X537A have markedly different actions on the MEPP frequency recorded at the frog neuromuscular junction. A23187 has no significant effect at 9–17°C, but causes a small increase in MEPP frequency at 6°C. At 25°C, on the other hand, A23187 causes a marked and progressive rise in MEPP rate. It is suggested that, in spite of increased Ca2+ influx associated with application of the ionophore, the presynaptic terminals can maintain [Ca2+]i constant at 9–17°C, although [Ca2+]i rises at higher and lower temperatures, causing an increase in frequency of MEPPs. As previously reported by Kita and Van der Kloot (5) X537A causes a dramatic increase in MEPP frequency, but it is suggested that its action is more complex and probably involves an increase in Na+ permeability.  相似文献

Investigations were conducted to study the effects of browse availability and quality on black rhinoceros (Diceros bicornis michaeliGroves 1967) diet in Nairobi National Park, Kenya, between September 1994 and February 1995. Line intercept method was used to assess availability of browse. The ratios of browsed crowns to the total crown were applied in the estimate of browse use. Proximate analysis and extraction methods were used, respectively, to appraise browse nutritional quality and phytochemical status of twelve species. Thirty‐four plant species were identified as potential black rhino browse, whose availability differed significantly (P < 0.001). Lippia javanica (20.581 ± 3.101, n = 544) and Lannea cornuta (0.963 ± 0.529, n = 544) were the most and least available species, respectively. Only 32 species, however, were utilized, as L. cornuta and Rhynchosia hirta did not show any level of rhino feeding activity. Browse use was found to vary significantly (P < 0.001) during the study period. Grewia similis was most (20.564 ± 2.764, n = 544) and Commelina africana least (0.974 ± 0.974, n = 544) exploited, respectively. Differences in total alkaloids and phenols, crude fibre, crude protein and total ash contents among species investigated were highly significant (P < 0.001). Ether extract values were similar (P= 0.0933) across plant taxa. The study shows that the black rhino is a selective browser feeding on plants with low phenol and alkaloid contents and high fibre values. Of the nutritionally similar species, browsing was driven by availability. These findings suggest that a key habitat factor important in black rhinoceros conservation include diverse plant species with low levels of phenols and alkaloids.  相似文献

Disulfide bonds in soybean trypsin inhibitor (Kunitz) were simultaneously reduced and alkylated using tri-n-butylphosphine and 2-vinylquinoline at pH 7.6 in 0.11 M Tris-4.4 M urea, 41% ethanol. The resulting S--2-quinolylethylated protein (2-QE-STI) has a new absorption peak at 315–318 nm. Its quinoline fluorescence can be excited above 310 nm independently of intrinsic protein fluorescence. Free 2-quinolylethylcysteine (2-QEC) shows unexpectedly weak fluorescence. Quinoline absorption in 2-QEC and 2-QE-STI changes with pH. The apparentpK values determined spectrophotometrically are near 5 for 2-QEC and 3 for 2-QE-STI. Fluorescence decreased with increasing pH and in the presence of chloride ions. Both structural and charge effects thus appear to influence the absorption and fluorescence of the quinoline group. Corrected fluorescence emission (excited at 316 nm) of neutral 2-QE-STI diluted in 0.1 N H2SO4 was directly proportional to concentration in the range 0.4–8 m 2-QEC. The 2-QEC content of the protein derivative determined by UV absorption at pH 1.5 was in agreement with the expected value of four residues per mole. A unique residue W544 in the β18–β19 loop of the Bacillus thuringiensis Cry1Ac toxin has been implicated in its toxicity. In this study, the effects of mutations at this residue on protein stability during protease treatment, UV irradiation, and preservation were examined. Residue 544 of Cry1Ac was involved in maintaining structural stability, and substitution of a polar group at this position was unfavorable to protein stability. One mutant, W544F, produced larger crystals and was more stable. This mutant showed greater resistance to UV radiation than the wild type Cry1Ac but retained equal toxicity. This is the first report showing that residue 544 in the Cry1Ac domain III plays a significant role in toxin structural stability. Our W544F mutant is a significant development in terms of field applications of Cry1Ac toxin.  相似文献

Objective: Obesity, especially abdominal, is a risk factor for many diseases. This study explored trends in the prevalence of general and abdominal obesity, 1986–2004, in northern Sweden. Methods and Procedures: Cross‐sectional population surveys were performed in 1986, 1990, 1994, 1999, and 2004; 250 men and 250 women aged 25–34, 35–44, 45–54, and 55–64 years (from 1994, also 65–74 years) were randomly selected; the overall participation rate was 77%. Anthropometric data were used. Results: Weight and BMI increased in all men, most significantly in men aged 25–64 years (P < 0.0005). Weight increased in women aged 25–64 years (P < 0.005) and BMI in women aged 25–44 years (P < 0.005). Prevalence of obesity (BMI ≥ 30) increased significantly in men aged 25–44 and 55–74 years (P < 0.005; for men 65–74 years old, P < 0.05) and in women aged 25–44 years (P < 0.005). Waist circumference decreased significantly between 1986 and 1990 in all women (P < 0.005) and in men aged 55–64 years (P < 0.05). After 1990 waist circumference increased, most markedly so in women; by 2004 circumference measurements for women, and for men aged 55–64 years, were equal to those of 1986, while for men aged 25–54 years they were higher. Prevalence of abdominal obesity has increased since 1990, most markedly so in women aged 45–64 years (P < 0.0005). Discussion: The rapid increase in both general and central obesity raises concern for the future; increasing abdominal obesity in women is particularly alarming.  相似文献

An -N-acetylgalactosaminidase IV able to remove blood type specificity of human A(II)-erythrocytes and not effecting B(III)-erythrocytes was isolated from the marine bacterium Arenibacter latericius KMM 426T. The -N-acetylgalactosaminidase IV preparation exhibits high activity during inhibition of hemagglutination with blood group substance A containing determinants analogous to A-erythrocytes. The enzyme has a pH optimum from 7.0 to 8.0 and completely retains its activity during 30-min heating at 50°C and for a week at 20°C. The enzyme can be stored under the sterile conditions for any length of time at 4°C, but it does not withstand freezing. The -N-acetylgalactosaminidase is resistant to NaCl; for p-nitrophenyl--N-acetyl-D-galactosaminide, the K m is 0.38 mM. The molecular mass of the enzyme determined by gel filtration is 84 kD.  相似文献

The degree of modal specificity of neurons of the tectum, dorsal thalamus, and primordial hippocampus relative to visual (photic) and somatic (tactile) stimulation was investigated in experiments on frogs (Rana temporaria) immobilized with tubocurarine. The specificity of sensory information arriving at the successive levels of the frog's thalamo-telencephalic afferent system diminishes in an ascending direction. Evoked potentials arising in the telencephalon in response to electrical stimulation of the sciatic nerve and to flashes are found not only in the primordial hippocampus, but also bilaterally in the septum and striatum. It is concluded on the basis of discharges recorded in the fibers of the thalamo-telencephalic tracts during somatic and visual stimulation that fibers carrying nonolfactory information into the telencephalon run in the medial and lateral forebrain bundles.I. Abstract

To assess the tolerance, the rye-grass L. grown on soil amended with petroleum wastewater (PWW) containing four metals lead, zinc, nickel and mercury. The PWW (25 to 50%) showed remarkable increase in length and biomass. Chlorophyll ‘a and b’ increased with an increase of PWW from 25–50% while such contents decreased on increasing the 75–100% compared to control. The mass balance performed on the system showed the removal of 90–97.6% lead, 85.5–92.9% zinc, 78.9–85.5% nickle and 47.6–27.5% mercury. The model for the maximum metal reduction rate (Rmax) was much better for Pb (89.5) and Zn (72.1) with respect to Ni (57.3) and Hg (32.4). Survival of rye-grass (30-days, statics, and renewal exposures) was increased by 50% as compared to control. The toxicity index Y of PWW showed 0–25% deficiency level, 25–50% tolerance level, 50–90% toxic level and 90–100% lethal level.
