Mesophyll resistance to CO2 uptake was calculated from gas exchange data on intact leaves of 12 species of woody plants. Plants studied were native to habitats ranging from streamsides to deserts. Gas exchange measurements were made at light saturation and constant temperature to eliminate possible effects of light and temperature on estimates of mesophyll resistance. Cuticular transpiration was measured and used in calculation of stomatal resistances from whole leaf transpiration rates. In all species examined, an increase in mesophyll resistance was observed as leaves dried. The increase in mesophyll resistance in all cases occurred at the same water potential as the initial decline in net photosynthesis, and was accompanied by an increase in stomatal resistance.

Rates of photosynthesis, respiration, and transpiration of Monterey pine (Pinus radiata D. Don) were measured under controlled conditions of soil water suction and soil temperature. Air temperature, relative humidity, light intensity, and air movement were maintained constant. Rates of net photosynthesis, respiration, and transpiration decreased with increasing soil water suction. The decrease in the rates of net photosynthesis and transpiration as a function of the soil temperature at low soil water suctions may be attributed to changes in the viscosity of water. At soil water suctions larger than 0.70 bars rates of transpiration and net photosynthesis may be affected in the same proportion by changes in stomatal apertures.

Measurements of carbon dioxide exchange and transpiration were made, at various air temperatures, on wheat and barley using a field enclosure system. From these were derived the stomatal and mesophyll resistances to carbon dioxide transfer. Optimum temperatures for net CO2 uptake were about 24°C for wheat and barley. Above these optima, as temperature increased so net CO2 uptake rates decreased, because of increasing stomatal and mesophyll resistances; transpiration rates decreased in wheat but were constant in barley. In laboratory growth cabinets, wheat plants were subjected to different regimes of temperature and humidity. Optimum temperature for net CO2 uptake of individual leaves was 25°C. At constant humidity, a decline in net uptake rates above 25°C was associated with large increases in mesophyll resistance. At a constant 25°C, as the vapour pressure deficit (v.p.d. was increased above 1 k Pa (10 mb) v.p.d. the net uptake declined, with an increase in mesophyll resistance and a small increase in stomatal resistance. When the v.p.d. exceeded 1 k Pa at a temperature of 30°C, conditions that are experienced by field plants, then there were large increases in both mesophyll and stomatal resistances and the net uptake rates declined. Photo-respiration, as measured by CO2 uptake in oxygen-free air, was independent of temperature, but both dark respiration and CO2 compensation concentration increased with temperature.

Net photosynthesis, transpiration, dark respiration rates and stomatal and mesophyll resistances were studied in young potted seedlings of Pinus halepensis Mill. under gradually decreasing soil and leaf water potentials. Stomatal resistance under non-limiting xylem water potentials was 6–7 times higher than mesophyll resistance. Stomata started to close at threshold xylem water potentials of −0.8 MPa, whereas mesophyll resistance started to increase at about −1.4 MPa. Decreasing xylem water potentials increased the CO2 compensation point and decreased the water use efficiency (expressed by the photosynthesis to transpiration ratio) and dark respiration rate. It is concluded that at least part of the drought resistance characteristics of P. halepensis are associated with a sensitive stomatal mechanism which enables an efficient control of water loss.

Summary Evidence is presented which suggests that the mesophyll cell walls of cotton leaves may influence observed rates of transpiration. The net diffusive flux of water vapour, from the upper and lower surfaces of a leaf, was compared with the flux of nitrous oxide through a leaf and evidence obtained of an extra resistance in the water-vapour pathway associated with water transport in the mesophyll cell walls. This extra resistance appeared to be insignificant at low transpiration rates and in turgid leaves, but increased with transpiration rate and dehydration. The most likely explanation for its origin appeared to be a reduction in hydraulic conductivity across the internal cuticle which lines the outer surfaces of the mesophyll cell walls. In turn this served to reduce the relative vapour pressure at the sites of evaporation. The experiments were conducted under conditions where stomatal opening was induced by CO2-free air. Under normal conditions stomatal closure would tend to reduce the development of this extra resistance. Even so, the results throw doubt on the validity of the long-standing assumption that the water-vapour pressure at the evaporation sites is equal to the saturation vapour pressure under all conditions.

Rate of net CO2 assimilation by soil-grown soybean plants were studied over a range of relative leaf water contents at each of four levels of irradiance. There was a large interaction between light level and leaf water deficit on the rate of CO2 assimilation. The effect of leaf water deficit on assimilation became larger as irradiance increased. Both stomatal resistance to CO2 transport and mesophyll resistance to CO2 assimilation increased as leaf-water deficit increased. The increase in both resistance with changing leaf-water content was largest at high irradiance and became smaller as irradiance decreased. Relief of soil-moisture stress by watering induced large oscillations of CO2 assimilation, stomatal resistance, and mesophyll resistance. The oscillation of the mesophyll resistance occurred in the absence of changes in relative water content and appeared to be related to oscillations in leaf temperature. The observed increase in mesophyll resistance with decreasing leaf-water content under nonoscillative conditions may be caused by changes in leaf temperature rather than leaf water content.

Jojoba [ Simmondsia chinensis (Link) Schneider] cuttings were grown in pots under constant light intensity and vapour pressure deficit at wir temperatures of 18 and 27°C in climate-controlled cabinets. Leaf conductance and transpiration rate decreased exponentially as the xylen water potential (Ψx) decreased concurrently with the drying out of the soil. At high Ψx'leaf conductance and transpiration rate were much higher at the higher air temperature, and as Ψx declined both parameters decreased more rapidly at 27°C than at 18°C. When soil temperatures were decreased from 27 to 13°C, leaf water potential was not affected at either air temperatures, but transpiration rate was reduced. A linear negative correlation was found between transpiration rates and soil temperatures. It is suggested that the low soil temperature may restrict reducion of water flux in turn reduces stomatal conductance and transpiration without affecting the water potential in the shoot. The releavance of the response to changes in soil or air temperature to the performance of the plant in its semi-arid habitat is discussed.

The efficiency with which plants transport water is related to the water potential differences required to drive water fluxes from the soil to the leaf. A comparative study of two woody and three herbaceous species (Citrus sinensis L. cv. Koethen, Pyrus kawakami L., Helianthus annuus L. cv. Mammoth Russian, Capsicum frutescens L. cv. Yolo Wonder, and Sesamum indicum L. cv. Glauca) indicated contrasts in water transport efficiency. Depression of leaf water potential in response to transpiration increases was found in the woody species; the herbaceous species, however, had more efficient water transport systems and presented no measurable response of leaf water potential to transpiration changes. Different maximum transpiration rates under the same climatic conditions were observed with different species and may be accounted for by stomatal response to humidity gradients between leaf and air. Leaf diffusion resistance in sesame increased markedly as the humidity gradient was increased, while leaf resistance of sunflower responded less to humidity. Stomata appeared to respond directly to the humidity gradient because changes in leaf water potential were not detected when leaf resistance increased or decreased.

The rates of transpiration from the upper and lower surfaces of leaves of Gossypium hirsutum, Xanthium strumarium, and Zea mays were compared with the rates at which helium diffused across those leaves. There was no evidence for effects of CO2 concentration or rate of evaporation on the resistance to water loss from the evaporating surface (“resistance of the mesophyll wall to transpiration”) and no evidence for any significant wall resistance in turgid tissues. The possible existence of a wall resistance was also tested in leaves of Commelina communis and Tulipa gesneriana whose epidermis could be easily peeled. Only when an epidermis was removed from a leaf, evaporation from the mesophyll tissue declined. We conclude that under conditions relevant to studies of stomatal behavior, the water vapor pressure at the sites of evaporation is equal to the saturation vapor pressure.

Photosynthetic Response to Water Stress in Phaseolus vulgaris
Water stressed Phaseolus vulgaris L. plants were monitored to detect the relationships between net photosynthesis, transpiration, boundary layer plus stomatal resistance, mesophyll resistance, CO2 compensation point, ribulose, 1,5-diphosphate carboxylase activity and leaf water potential. At full expansion, the first trifoliate leaves of greenhouse grown bean plants were subjected to water stress by withholding irrigation. Gas exchange and enzyme activity of the central trifoliolate leaflets were monitored as leaf water potential decreased. Although increased stomatal resistance appeared to be the primary causal factor of reduced net photosynthesis, increased mesophyll resistance and decreased ribulose 1,5-diphosphate carboxylase activity further documented the role of non-stomatal factors.

Continuous and simultaneous measurements of CO2 exchange andtranspiration rates of whole soybean plants were made undercontrasting, controlled environmental conditions when waterstress was imposed by withholding water. Daytime temperaturesand vapour pressure deficits were 27.5 ° C/12 mbar; 27.5° C/5 mbar; 22.5 ° C/12 mbar, and 22.5 ° C/5 mbar. The experimental conditions were virtually the same as the conditionsunder which the plants had been grown. Under all four treatments photosynthesis and transpiration rateswere closely correlated as water stress increased, but in viewof the evidence for a significant mesophyll resistance to photosynthesisin- both stressed and unstressed plants it is not consideredthat this is due to total stomatal control. At — 0.4 bar soil water potential (soll) the rates oftranspiration and photosynthesis became independent of the atmosphericconditions and were very similar under all treatments. Thisis attributed to slow movement of water into the root zone fromthe surrounding soil with associated stomatal closure limitingthe rates of water uptake and transpiration. With decreasing soll, relative water content of the leaf (RWC)fell more rapidly and to lower levels under 27.5 ° C/12mbar conditions than under the other treatments. The least reductionin RWC was under the 22.5 ° C/5 mbar treatment. Increasingsoil water stress had the greatest relative effect on ratesof gaseous exchange under 27.5 ° C/12mbar conditions andleast under 22.5 ° C/5mbar conditions.

Simultaneous cyclic variation in rates of both net photosynthesis and transpiration were induced in attached leaves of cotton and pepper plants under constant environmental conditions. The cyclic variations in photosynthesis and transpiration were found to be in phase, and the ratio net photosynthetic rate/transpiration rate remained constant over a wide range of gas exchange rates. A similar constancy of this ratio was also found as gas exchange rates declined following excision of a sunflower leaf, which was not initially cycling, in air. These results suggested that change in stomatal aperture was the only controlling factor involved and that it was affecting both processes proportionately. Visible loss of leaf turgur and measurable water stress developed in both pepper and cotton at peak exchange rates, but the gas exchange ratio remained constant. The failure of water stress and increased stomatal aperture to lower the gas exchange ratio suggested an absence of any significant leaf mesophyll resistance (r′m) to inward diffusion of CO2. The possibility that r′m was low is discussed generally, and in relation to the use of chemical antitranspirants to raise the gas exchange ratio. Within the limits of the experiments, water stress apparently had no direct adverse effect on rates of net photosynthesis. The gas exchange ratio did not rise as exchange rates declined. Ultimately, at very low exchange rates, the ratio fell, declining to zero in cotton, but not in pepper. This decline was attributed to the onset of significant gas exchange through the cuticle, which was apparently less permeable to CO2 than to water vapour. Positive net cuticular photosynthesis therefore probably does not occur in cotton. Except at very low exchange rates, the gas exchange ratio was higher in cotton than in pepper; it was similar in sunflower and cotton.

Cotton plants, Gossypium hirsutum L. were grown in a growth room under incident radiation levels of 65, 35, and 17 Langleys per hour to determine the effects of vapor pressure deficits (VPD's) of 2, 9, and 17 mm Hg at high soil water potential, and the effects of decreasing soil water potential and reirrigation on transpiration, leaf temperature, stomatal activity, photosynthesis, and respiration at a VPD of 9 mm Hg.

Transpiration was positively correlated with radiation level, air VPD and soil water potential. Reirrigation following stress led to slow recovery, which may be related to root damage occurring during stress. Leaf water potential decreased with, but not as fast as, soil water potential.

Leaf temperature was usually positively correlated with light intensity and negatively correlated with transpiration, air VPD, and soil water. At high soil water, leaf temperatures ranged from a fraction of 1 to a few degrees above ambient, except at medium and low light and a VPD of 19 mm Hg when they were slightly below ambient, probably because of increased transpirational cooling. During low soil water leaf temperatures as high as 3.4° above ambient were recorded. Reirrigation reduced leaf temperature before appreciably increasing transpiration. The upper leaf surface tended to be warmer than the lower at the beginning of the day and when soil water was adequate; otherwise there was little difference or the lower surface was warmer. This pattern seemed to reflect transpiration cooling and leaf position effects.

Although stomata were more numerous in the lower than the upper epidermis, most of the time a greater percentage of the upper were open. With sufficient soil water present, stomata opened with light and closed with darkness. Fewer stomata opened under low than high light intensity and under even moderate, as compared with high soil water. It required several days following reirrigation for stomata to regain original activity levels.

Apparent photosynthesis of cotton leaves occasionally oscillated with variable amplitude and frequency. When soil water was adequate, photosynthesis was nearly proportional to light intensity, with some indication of higher rates at higher VPD's. As soil water decreased, photosynthesis first increased and then markedly decreased. Following reirrigation, photosynthesis rapidly recovered.

Respiration was slowed moderately by decreasing soil water but increased before watering. Respiration slowed with increasing leaf age only on leaves that were previously under high light intensity.

Photosynthesis, transpiration, stomatal conductance and chlorophyll fluorescence characteristics were examined in kidney bean plants, with developing gradually water stress for several days after watering and then permitted to recover by re-watering. The photosynthetic rate, transpiration rate, and stomatal conductance decreased rapidly by withholding water for 2 days. The Fv/Fm of chlorophyll fluorescence characteristics slightly decreased when the water was withheld for 7 days. After re-watering the rate of recovery of photosynthesis, transpiration, and stomatal conductance decreased gradually as the days without watering became longer. The differences existed in rates of recovery of photosynthesis, transpiration, and stomatal conductance following drought stress. Among the fractional recoveries the highest was photosynthesis, and the lowest was stomatal conductance. Photosynthesis rate following drought stress was rapidly recovered until 2 days after re-watering, then recovered slowly. The critical time for the recovery of photosynthesis was recognized. The results show clearly a close correlation between the leaf water potential and the recovery level and speed of photosynthesis, transpiration, and stomatal conductance.

The effect of soil flooding on photosynthesis, transpiration and stomatal conductance of Jatropha curcas seedlings were studied under natural environmental variables. Soil flooding reduced photosynthesis (P N), transpiration (E) and stomatal conductance (gs) in response to leaf positions of Jatropha curcas plants. Based on the results, we conclude that decrease in stomatal opening and stomatal limitation of photosynthesis, followed by decrease in individual leaf area are the main causes of reductions in carbon uptake of flooded seedlings. A mathematical relationship was successfully developed to describe photosynthesis, transpiration and stomatal response of Jatropha under soil flooding stress.

Background and Aims

Daytime root-zone temperature may be a significant factor regulating water flux through plants. Water flux can also occur during the night but nocturnal stomatal response to environmental drivers such as root-zone temperature remains largely unknown.

Methods

Here nocturnal and daytime leaf gas exchange was quantified in ‘Shiraz’ grapevines (Vitis vinifera) exposed to three root-zone temperatures from budburst to fruit-set, for a total of 8 weeks in spring.

Key Results

Despite lower stomatal density, night-time stomatal conductance and transpiration rates were greater for plants grown in warm root-zones. Elevated root-zone temperature resulted in higher daytime stomatal conductance, transpiration and net assimilation rates across a range of leaf-to-air vapour pressure deficits, air temperatures and light levels. Intrinsic water-use efficiency was, however, lowest in those plants with warm root-zones. CO2 response curves of foliar gas exchange indicated that the maximum rate of electron transport and the maximum rate of Rubisco activity did not differ between the root-zone treatments, and therefore it was likely that the lower photosynthesis in cool root-zones was predominantly the result of a stomatal limitation. One week after discontinuation of the temperature treatments, gas exchange was similar between the plants, indicating a reversible physiological response to soil temperature.

Conclusions

In this anisohydric grapevine variety both night-time and daytime stomatal conductance were responsive to root-zone temperature. Because nocturnal transpiration has implications for overall plant water status, predictive climate change models using stomatal conductance will need to factor in this root-zone variable.

Daily measurement of gas exchanges during a complete growth cycle: photosynthesis, transpiration and night respiration in a soil culture of maize. During a complete growth cycle, photosynthesis (P), night-respiration (R) and transpiration (T) have been measured daily for maize (Zea mays L. cv. INRA F7 × F2) in a self-regulating growth chamber. P and T varied according to three different kinetics. During the growing period there was a fast and concomitant variation of both P and T, which could be filled to a power function of time. During the fructification period we observed a linear decrease of P and T. Finally, during the senescence period P decreased to zero, whereas T kept a value higher than the evaporation of the naked soil. In the first two periods, the P/T ratio decreased as leaves aged. From this we could deduce that a slow increase of stomatal resistance (RS) and mesophyll resistance (RM) took place. R increased until the panicle appeared; it remained constant during fructification and then it decreased with senescence. Each nitrogen supply induced a transient increase of R. Finally, a water stress led to a greater decrease of P than of R, but the after effect of water stress disappeared a day later. These results were compared with those obtained in hydroponic conditions.
