During incubation of potato tuber discs ( Solanum tuberosum L. cv. Bintje) on a callus-inducing medium at 28°C, a high capacity of in vivo alternative pathway respiration develops (75% of uninhibited respiration is azide-resistant). When callus induction takes place at 8°C, only 45% of respiration is resistant to azide. In the lag phase of growth the activity of alternative pathway is low. during the exponential growth phase the activity reaches its maximal rate. This in vivo activity is of the same size at both culture temperatures. As a consequence a greater part of alternative pathway capacity is operating in uninhibited respiration during growth at low temperatures.  相似文献

A postulated role of the CN-resistant alternative respiratory pathway in plants is the maintenance of mitochondrial electron transport at low temperatures that would otherwise inhibit the main phosphorylating pathway and prevent the formation of toxic reactive oxygen species. This role is supported by the observation that alternative oxidase protein levels often increase when plants are subjected to growth at low temperatures. We used oxygen isotope fractionation to measure the distribution of electrons between the main and alternative pathways in mung bean (Vigna radiata) and soybean (Glycine max) following growth at low temperature. The amount of alternative oxidase protein in mung bean grown at 19°C increased over 2-fold in both hypocotyls and leaves compared with plants grown at 28°C but was unchanged in soybean cotyledons grown at 14°C compared with plants grown at 28°C. When the short-term response of tissue respiration was measured over the temperature range of 35°C to 9°C, decreases in the activities of both main and alternative pathway respiration were observed regardless of the growth temperature, and the relative partitioning of electrons to the alternative pathway generally decreased as the temperature was lowered. However, cold-grown mung bean plants that up-regulated the level of alternative oxidase protein maintained a greater electron partitioning to the alternative oxidase when measured at temperatures below 19°C supporting a role for the alternative pathway in response to low temperatures in mung bean. This response was not observed in soybean cotyledons, in which high levels of alternative pathway activity were seen at both high and low temperatures.  相似文献

Alternative oxidase activity in potato tuber (Solanum tuberosum L. cv Bintje) callus mitochondria with exogenous NAD(P)H as substrate is inhibited by low concentrations of the detergent Triton X-100. Alternative oxidase activity with succinate or malate as substrate is not affected by these low concentrations of Triton X-100. Cytochrome pathway activity was not influenced under these conditions, neither with endogenous nor with exogenous substrate. Washing of Triton X-100-treated mitochondria did partially restore both uninhibited and CN-resistant NADH oxidation, indicating that under these conditions Triton X-100 does not permanently remove major components from the mitochondrial membrane. Apparently, it is possible to manipulate mitochondria in such a way that the access of exogenous NADH to the alternative pathway is blocked while access to the cytochrome pathway is uninhibited. It is suggested that membrane conditions have a regulatory function (possibly via influencing the diffusion path) in the oxidation of exogenous NADH via the alternative pathway.  相似文献

Suspension cells of NT1 tobacco (Nicotiana tabacum L. cv bright yellow) have been used to study the effect of growth temperature on the CN-resistant, salicylhydroxamic acid-sensitive alternative pathway of respiration. Mitochondria isolated from cells maintained at 30°C had a low capacity to oxidize succinate via the alternative pathway, whereas mitochondria isolated from cells 24 h after transfer to 18°C displayed, on average, a 5-fold increase in this capacity (from 7 to 32 nanoatoms oxygen per milligram protein per minute). This represented an increase in alternative pathway capacity from 18 to 45% of the total capacity of electron transport. This increased capacity was lost upon transfer of cells back to 30°C. A monoclonal antibody to the terminal oxidase of the alternative pathway (the alternative oxidase) from Sauromatum guttatum (T.E. Elthon, R.L. Nickels, L. McIntosh [1989] Plant Physiology 89: 1311-1317) recognized a 35-kilodalton mitochondrial protein in tobacco. There was an excellent correlation between the capacity of the alternative path in isolated tobacco mitochondria and the levels of this 35-kilodalton alternative oxidase protein. Cycloheximide could inhibit both the increased level of the 35-kilodalton alternative oxidase protein and the increased alternative pathway capacity normally seen upon transfer to 18°C. We conclude that transfer of tobacco cells to the lower temperature increases the capacity of the alternative pathway due, at least in part, to de novo synthesis of the 35-kilodalton alternative oxidase protein.  相似文献

Ethanol, when added to the incubation medium of callus-forming potato tuber discs, inhibits callus growth and causes an increase of the mitochondrial antimycin-A resistant respiration, expressed as a percentage of state III-respiration. This increase in resistance to antimycin-A is the result of a poor development of the cytochrome pathway in tissue discs treated with ethanol. The development of the antimycin-A resistant alternative oxidase sensitive to chelator is about the same for treated and untreated discs. The respiratory control (RC) ratio of the mitochondrial respiration increases after addition of a chelator, which inhibits the alternative pathway. The RC ratio of the uninhibited mitochondrial respiration appears to be inversely related to the capacity of the alternative pathway, when mitochondrial preparations with different capacities to transfer electrons via the alternative path are compared. From the experimentally observed relation between RC-ratio and alternative oxidase capacity, it was concluded that at least half of the capacity of the alternative path is used in uninhibited state IV respiration.  相似文献

Respiration rates of Zea mays L. seedling tissues grown at 30 and 14°C were measured at 25°C at different stages of seedling growth. Accumulation of heat units was used to define the developmental stages to compare respiration between the two temperatures. At both temperatures, respiration rates of most tissues were highest at the youngest stages, then declined with age. Respiration rates of mesocotyl tissue were the most responsive to temperature, being nearly twofold higher when grown at 14 compared to 30°C. Alternative pathway respiration increased concomitantly with respiration and was higher in mesocotyls grown in the cold. When seedlings were started at 30 then transferred to 14°C, the increase in alternative pathway respiration due to cold was not observed unless the seedlings were transferred before 2 days of growth. Seedlings transferred to 14°C after growth at 30°C for 2 days had the same alternative oxidase capacity as seedlings grown at 30°C. Seedlings grown at 14°C for 10 to 12 days, then transferred to 30°C, lost alternative pathway respiratory capacity over a period of 2 to 3 days. Western blots of mitochondrial proteins indicated that this loss of capacity was due to a loss of the alternative oxidase protein. Some in vitro characteristics of mitochondria were determined. The temperature optimum for measurement of alternative oxidase capacity was 15 to 20°C. At 41°C, very little alternative oxidase was measured, i.e., the mitochondrial oxygen uptake was almost completely sensitive to cyanide. This inactivation at 41°C was reversible. After incubation at 41°C, the alternative oxidase capacity measured at 25°C was the similar to when it was measured at that temperature directly. Isolated mitochondria lost alternative oxidase capacity at the same rate when incubated at 41°C as they did when incubated at 25°C. Increasing the supply of electrons to isolated mitochondria increased the degree of engagement of the alternative pathway, whereas lower temperature decreased the degree of engagement. Lower temperatures did not increase the degree of engagement of the pathway in intact tissues. We interpret these observations to indicate that the greater capacity of alternative oxidase in cold-grown seedlings is a consequence of development at these low temperatures which results in elevated respiration rates. Low temperature itself does not cause greater capacity or engagement of the alternative oxidase in mitochondria that have developed under warm temperatures. Our hypothesis would be that the low growth temperatures require the seedlings to have a higher respiration rate for some reason, e.g., to prevent the accumulation of a toxic metabolite, and that the alternative pathway functions in that respiration.  相似文献

Incubation of potato tuber tissue discs on B5 medium supplemented with 1-naphtyl-acetic acid (NAA) led to callus formation, irrespective of the presence of kinetin; without NAA no callus formation occurred. Incubation in the presence of abscisic acid (ABA) reduced the increases in fresh weight and dry weight both in callus-forming and in non-callus-forming tissue. Mitochondrial respiration was lowered by ABA as well. The induction of the alternative, CN-resistant pathway was inhibited by the presence of ABA, especially in mitochondria from non-callus-forming tissue.The in vivo respiration of the callus-forming tissue was higher than that of the non-callus-forming tissue. Total respiration, cytochrome pathway activity and the capacity of the alternative pathway were all lowered in callus-forming tissue by treatment with ABA. The in vivo activity of the alternative pathway was low in all tissue types, especially after ABA-treatment. The slight stimulation by hydroxamates of the oxygen uptake of callus-forming tissue incubated on medium with NAA and ABA indicates the involvement of a hydroxamate-activated peroxidase in the oxygen uptake of this tissue; this peroxidase seemed not to participate in the oxygen uptake of the other tissues types.In non-callus-forming tissue the oxygen uptake of ABA-treated tissue was very low and almost completely resistant to the combined addition of inhibitors of both the cytochrome and the alternative pathway, indicating that the in vivo activity of the mitochondria in the oxygen uptake of the tissue was very low. The possible causes for this ABA-effect are discussed. In non-callus-forming tissue the treatment with ABA creates a situation which is comparable with that observed in intact potato tubers. This situation is characterized by a tissue respiration lower than that of the isolated mitochondria and an alternative pathway capacity that is low or absent.  相似文献

Chloramphenicol (CAP), an inhibitor of the mitochondrial proteinsynthesis inhibits callus induction and subsequent growth ofpotato tuber tissue discs. Tissue respiration increase did notoccur in the presence of CAP. Both with and without CAP theinitially CN-sensitive tissue becomes totally CN-resistantin 1–2 weeks. CAP blocks the development of mitcohondrial cytochrome oxidase.A gradual decrease in the activities of cytochrome oxidase andof cytochrome pathway-mediated mitochondrial respiration isfound in CAP-tissue. The mitochondrial alternative pathway whichis absent in mitochondria from freshly sliced tissue developsduring incubation both in the absence and presence of CAP. Thealternative pathway is only operative in uninhibited state IIIrespiration in mitochondria from CAP-tissue. Cycloheximide, an inhibitor of the cytoplasmic protein synthesisinhibits the developments of the alternative pathway and ofthe cytochrome pathway. Alcohol dehydrogenase activity increasestenfold in the tissue during two weeks of incubation on mediawith and without CAP. The growth (fresh and dry weight increase) of potato tuber ( Solanum tuberosum L. cv. Bintje) callus discs was stimulated by incubation in air with 500 ppm 2,5-norbornadiene (NBD, a competitive inhibitor of ethylene action) and inhibited by incubation in air with 4 000 ppm NBD. Ethylene formation by the callus was stimulated by NBD. The development of the alternative pathway, measured in isolated mitochondria was inhibited by NBD in a concentration-dependent way. The alternative pathway capacity, measured in vivo, was inhibited by 4 000 ppm NBD, but not by 500 ppm. Uninhibited in vivo respiration, which consists of cytochrome path activity and alternative path activity, was stimulated by the treatment with 500 ppm NBD. The main contribution to this stimulation was made by the cytochrome pathway. In 4 000 ppm NBD-treated callus, uninhibited respiration seemed to be unaffected as a consequence of an inhibited cytochrome path activity, which was compensated by a stimulated alternative path activity. Both in 500 and 4 OIK) ppm NBD-treated callus the alternative path activity in vivo was stimulated.

The CN-resistant alternative oxidase pathway was examined inrelation to the formation of callus and adventitious roots ofJerusalem artichoke tuber tissues. During the early stage ofincubation at 28?C, respiration was significantly activatedand the O2-uptake rate via the Cyt pathway increased 2.5–2.6times by the second day of culture regardless of the presenceof 2,4-D. However, further increases and preservation of highlevels of Cyt pathway activities were observed only in the callusformingtissues. The capacity of the alternative pathway also increased,but the actually operating fraction was very small (0–4%of total respiration). On the other hand, during the later stagesof culture in which adventitious roots emerged from the callus,activation of the alternative pathway was observed. These resultssuggested that the alternative pathway was not involved in callusgrowth but was related to root formation in callus tissues ofJerusalem artichoke tubers. The capacity of the alternative respiratory pathway increased in the flavedo tissue of `Marsh' grapefruit (Citrus paradisi Macf.) stored at 5°C for 2 weeks or longer. Elevated O2 levels during respiratory assays enhanced respiration by the tissue at 20°C but not at 5°C. At 20°C, salicylhydroxamic acid alone was inhibitory to O2 uptake only in elevated O2. In conventional Warburg studies, alternative pathway respiration may be limited by the low solubility and/or slow rate of O2 diffusion into plant tissues, such as grapefruit flavedo, and may be responsible for the apparent low utilization of the alternative pathway potential observed in other studies.  相似文献

Four maize (Zea mays L.) inbreds representing genetic differences in seedling cold tolerance were used to determine the effect of growth temperatures on dry weight accumulation and mitochondrial properties, especially the alternative oxidase capacity. Seedlings were grown in darkness at 30°C (constant), 14°C (constant), and 15°C for 16 hours and 8°C for 8 hours. Inbreds B73 and B49 were characterized as cold tolerant while G50 and G84 were cold sensitive. Shoot growth rate of cold-sensitive inbreds in the lower temperatures was slower relative to the tolerant inbreds. Mesocotyl tissue was particularly sensitive to low temperatures during growth after germination. There were no significant differences in relative rates of mitochondrial respiration in the cold-tolerant compared to cold-sensitive inbreds measured at 25°C. Mitochondria from all seedlings grown at all temperatures had the ability to phosphorylate as indicated by the observation of respiratory control. This result indicated that differences in low temperature growth were probably not related to mitochondrial function at low temperatures. Alternative oxidase capacity was higher in mitochondria from seedlings of all inbreds grown at 14°C compared to 30°C. Capacities in seedlings of 14°C-grown B73 and G50 were higher than in B49 and G84. Capacities in seedlings grown for 16 hours at 15°C and 8 hours at 8°C were similar to those from 14°C-grown except in G50 which was lower and similar to those grown at 30°C. Mesocotyl tissue was the most responsive tissue to low growth temperature. Coleoptile plus leaf tissue responded similarly but contained lower capacities. Antibody probing of western blots of mitochondrial proteins confirmed that differences in alternative oxidase capacities were due to differences in levels of the alternative oxidase protein. Male sterile lines of B73 were also grown under the three different temperature regimes. These lines grew equally as well as the normal B73 at all temperatures and the response of alternative oxidase capacity and protein to low growth temperature was similar to normal B73.  相似文献

The effects of temperature upon the respiratory pathways of Arum maculatum mitochondria have been studied. The alternate oxidase sustained a greater proportion of the total respiration at low temperatures than at higher temperatures. Arrhenius plots of respiratory activities show two discontinuities, one at 14°C and one at 21°C. The lower temperature discontinuity was associated with electron transport from succinate dehydrogenase to the alternative oxidase, enzymes that face the inner side of the membrane while the higher temperature discontinuity was associated with electron transport from the external NADH dehydrogenase to cytochrome c oxidase, which face the outer side of the membrane. Both discontinuities resulted in a decrease in the activation energy for electron transport on one side of the membrane. Arrhenius plots of transmembrane electron transport showed discontinuities at both 14° and 21°C but the upper discontinuity resulted in an increase in the activation energy. Activation energies determined for the respiratory activities show that above 21°C the exogenous NADH-cytochrome pathway and the succinate-alternative oxidase pathway were lower than those for the NADH-alternative pathway or the succinate cytochrome pathway.  相似文献

The relationships of potato (Solanum tuberosum L.) tuber membrane permeability and membrane lipid composition to sugar accumulation were examined. Tubers from four potato cultivars were stored for 40 weeks at 3°C and 9°C. Rates of tuber membrane electrolyte leakage, total fatty acid composition, free fatty acid composition, and sugar content were measured throughout the storage period. Storage of tubers at 3°C caused dramatic increases in total fatty acid unsaturation, membrane permeability, and sugar content compared to tubers stored at 9°C. Cultivars with higher levels of fatty acid unsaturation had lower rates of membrane electrolyte leakage and lower sugar contents. We propose that high initial levels or high induced levels of membrane lipid unsaturation mitigate increases in tuber membrane permeability during storage, thus positively influencing the processing quality of stored potato tubers.  相似文献

Oxygen uptake and tetrazolium reduction occurred at higher rates in discs from potato tubers (Solanum tuberosum L.) stored at 0° than in discs from tubers stored at 12.8°. Tetrazolium reduction was at a higher rate in mitochondrial fractions from tubers stored at 0° than in mitochondrial fractions from tubers stored at 12.8°. These physiological activities were more resistant to hypertonic KCl treatments in tissue and mitochondrial fractions from tubers stored at 0° than in tissue and mitochondrial fractions from tubers stored at 12.8°. Inhibition of O2 uptake and tetrazolium reduction progressively increased with increasing concentrations of KCl for tissue and mitochondrial fractions from tubers stored at 0 and 12.8°, but inhibition was more severe and occurred at lower concentrations of KCl for the material from tubers stored at 12.8°. Tissue from tubers stored at 0° was at the same time more sensitive to hypotonic solutions and more resistant to hypertonic solutions than corresponding tissue from tubers stored at 12.8°. Adaptive changes brought on in the tubers by the stress of cold storage were demonstrated in the discs and mitochondrial fractions prepared from cold-stored tubers.  相似文献

The effect of temperature on suberization of potato tuber tissue was measured by diffusive resistance and quantitative chemical procedures. The optimum temperature for formation of aliphatic suberin monomers and development of resistance to water vapor conduction was 26.4°C whereas alkane synthesis was optimal at 18.6°C. Low temperatures (<16.6°C) reduced suberin monomer production more than alkane synthesis.  相似文献

A sharp temperature optimum is observed at 21.5°C when the incorporation of [14C]sucrose into starch is measured with discs cut from developing tubers of potato (Solanum tuberosum L. cv Desirée). By contrast, increasing temperatures over the range 9 to 31°C only enhance release of 14C to respiratory CO2 and incorporation of 14C into the ethanolsoluble fraction. By comparison, starch synthesis in discs from developing corms of cocoyam (Colocasia esculenta L. Schott) is increased by raising the temperature from 15 to 35°C. The significance of a relatively low temperature optimum for starch synthesis in potato is discussed in relation to the yield limitations imposed by continuously high soil temperatures. Amyloplasts isolated from protoplasts prepared from developing potato tubers contain activities of alkaline pyrophosphatase, NAD-dependent glyceraldehyde-3-phosphate dehydrogenase, fructose-1,6-bisphosphatase, and phosphoglucomutase in addition to ADP-glucose-pyrophosphorylase, starch phosphorylase and starch synthase. Cell-free amyloplasts released by thinly slicing developing potato tubers synthesize starch from [14C]triose-phosphate generated from [14C]fructose-1,6-bisphosphate in the reaction medium. This starch synthesis is inhibited by addition of 10 millimolar inorganic phosphate and requires amyloplast integrity, suggesting the operation of a triose-phosphate/inorganic phosphate exchange carrier at the amyloplast membrane. The temperature optimum at 21.5°C observed with tissue discs is not observed with amyloplasts.  相似文献

Electron microscopy revealed that subjecting `Russet Burbank' potato (Solanum tuberosum L.) plants to 2 days of cool temperature growing conditions (18°C days/10°C nights) did not produce visible damage or changes in tuber pith tissue when compared to warm-grown tubers (23°C days/18°C nights). However, damage to some tuber pith cells was observed after 5 days of cool treatment. Eight days of cool treatment produced extensive alterations in cell structure. The cytoplasm of the cool-treated tuber pith cells had become highly vesiculated and there was evidence of complete destruction of amyloplast membranes and tonoplasts. In many cases the starch grains appeared to be undergoing hydrolysis suggesting total disruption of normal cell function. Sixteen days of cool treatment were sufficient to produce visible brown center development in all cool-grown tubers examined. Electron microscopy of these tissues revealed that, although some organelles were still present, the cytoplasm had become extremely vesiculated and lacked any resemblance to that of tissue from warm-grown tubers. Gross, irregular thickening of cell walls was also detected.  相似文献

Capacity of alternative pathway mediated, CN-resistant respirationwas measured in mitochondria isolated from three plant species:Iris bulbs, potato tuber callus and Petunia cells, grown insuspension culture. Succinate, NADH or a combination of succinateand NADH were used as respiratory substrates. In all three plantspecies electrons from exogenous NADH appeared to have no accessto the alternative pathway connected with succinate mediatedrespiration and vice versa. Therefore, besides a (functional)compartmentation of Q-pools a compartmentation of alternativepathways in plant mitochondria is proposed as well. Electron transport, using succinate as a substrate, was measured polarographically in mitochondria isolated from Phaseolus vulgaris and P. acutifolius plants at 25°C and 32°C. Mitochondria isolated from P. vulgaris plants grown at 32°C had reduced electron transport and were substantially uncoupled. Growth at 32°C had no effect on electron transport or oxidative phosphorylation in P. acutifolius compared to 25°C grown plants. Mitochondria isolated from 25°C grown P. vulgaris plants measured at 42°C were completely uncoupled. Similarly treated P. acutifolius mitochondria remained coupled. The uncoupling of P. vulgaris was due to increased proton permeability of inner mitochondrial membrane. The alternative pathway was more sensitive to heat than the regular cytochrome pathway. At 42°C, no alternative pathway activity was detected. The substantially greater heat tolerance of P. acutifollus compared to P. vulgaris mitochondrial electron transport suggests that mitochondrial sensitivity to elevated temperatures is a major limitation to growth of P. vulgaris at high temperatures and is an important characteristic conveying tolerance in P. acutifolius.  相似文献
