Seabass (SBS) and seabream (SBM) juveniles were reared with the goal of obtaining three different final densities (SINT = 40 kg m?3; INT = 20 kg m?3; SEM = 0.2 kg m?3) to ascertain the effects thereof on the welfare of the fish. Significant blood metabolites and hepatic glycogen were determined every 3 months and at harvest. Trials lasted 18 months in seabass and 17 months in seabream. At the end of experiment the main biometric productive parameters and quality of body composition were also recorded. Regarding intensively reared seabass (SINT‐SBS, INT‐SBS), the plasma triglycerids, total cholesterol and transaminases (AST, ALT) were always significantly higher than in semi‐intensively maintained fish (SEM‐SBS). At the final sampling in the SINT‐SBS batch, the total protein and glucose were also markedly increased. Conversely, at harvest the liver glycogen content decreased in SINT‐SBS (34 ± 8 mg g?1 liver) with respect to INT‐SBS (57 ± 12 mg g?1 liver) and SEM‐SBS (63 ± 11 mg g?1 liver). No differences among groups were observed for creatine kinase (CK) and lactate dehydrogenase (LDH). With regard to seabream, SINT‐SBM and INT‐SBM constantly showed plasma triglycerids and total cholesterol as being significantly higher when compared to SEM‐SBM. In the final two blood samplings, SINT‐SBM exhibited the most elevated values for LDH. At harvest, AST, ALT, total protein and glucose markedly increased in SINT‐SBM, whereas liver glycogen content was reduced (22.5 ± 9 mg g?1 liver), more than in INT‐SBM (70 ± 16 mg g?1 liver) and SEM‐SBM (75 ± 20 mg g?1 liver). In both seabass and seabream, body composition was very similar in the different stocking densities, except for total cholesterol. Total n‐3 polyunsaturated fatty acid (PUFA) in seabream was significantly different from fish of the semi‐intensive groups; however, nutritional values and fatty acid profiles were equally good. The intermediate final density of seabass and seabream at 20 kg m?3 seemed to give the best results in terms of their well being when compared to fish reared at 40 kg m?3. The absence of differences in blood metabolites and hepatic glycogen levels between the intensive batch and the semi‐intensive groups until harvest was a reference to the positive status of the fish. A density of 20 kg m?3 can be considered acceptable for farm strategy planning for raising healthy on‐growing seabass and seabream juveniles.  相似文献

Background  The potential of Atipamezole (ATI) to reverse Ketamine/Xylazine (KET/XYL) anesthesia in the Olive baboon ( Papio anubis ) was studied.
Methods  Anesthesia was induced with 10 mg/kg KET and 0.5 mg/kg XYL intramuscularly. Mean arousal time (MAT), heart rate (HR), systolic arterial blood pressure (SAP), rectal temperature, respiratory rate (RR), and hemoglobin oxygen saturation (SpO2) were monitored. Baboons were treated with: KET/XYL only, KET/XYL followed by 100 μg/kg ATI or by 200 μg/kg ATI administered 25 minutes after KET/XYL.
Results  Atipamezole rapidly reversed depressed HR and SAP (10 ± 5.2 minutes), RR (5 ± 2 minutes) and SpO2 (3 ± 6 minutes) and significantly decreased MAT (13 ± 2.2 minutes) vs. KET/XYL alone (35 ± 5 minutes). Emesis was absent and salivation was observed after administration of 200 μg/kg ATI only.
Conclusions  Atipamezole at 100 μg/kg is sufficient for rapid and smooth reversal of KET/XYL anesthesia in the Olive baboon with minimal side effects.  相似文献

The main purpose of this study was to investigate the effects of benzo(a)pyrene (BaP) on seabass (Dicentrarchus labrax) juveniles using parameters at different levels of biological organization. Liver antioxidant status, BaP biotransformation and accumulation, growth, and behavior were determined in juveniles after 28 d exposure to BaP (1–16 μ g/l). Liver ethoxyresorufin O-deethylase increased in seabass exposed to 1–8 μ g/l of BaP. Liver glutathione S-transferases and catalase activities were significantly increased at 4 and 8 μ g/l, but a slight decrease was observed at the highest concentrations tested. Bile BaP metabolites were significantly different from the control group at 1 and 16 μ g/l BaP. Liver BaP metabolites and lipid peroxidation significantly increased at 8 and 16 μ g/l BaP. These results suggest that BaP metabolites' accumulation induces oxidative damage in seabass liver. Body weight and length increase were significantly reduced in fish exposed to BaP, with LOECs of 16 and 4 μ g/l, respectively. Food intake and swimming velocity were significantly decreased after exposure to BaP, with LOEC values of 16 and 8 μ g/l, respectively. Results suggest that at concentrations of BaP equal or higher than 8 μ g/l, the detoxification capacity decreases, an accumulation of liver BaP metabolites occurs causing lipid peroxidation, affecting growth and swimming capability of fish.  相似文献

Aims:  To develop solid-state fermentation system (SSF) for hyper production of tylosin from a mutant γ-1 of Streptomyces fradiae NRRL-2702 and its parent strain.
Methods and Results:  Various agro-industrial wastes were screened to study their effect on tylosin production in SSF. Wheat bran as solid substrate gave the highest production of 2500 μg of tylosin g−1 substrate by mutant γ-1 against parent strain (300 μg tylosin g−1 substrate). The tylosin yield was further improved to 4500 μg g−1 substrate [70% moisture, 10% inoculum (v/w), pH 9·2, 30°C, supplemental lactose and sodium glutamate on day 9]. Wild-type strain displayed less production of tylosin (655 μg of tylosin g−1 substrate) in SSF even after optimization of process parameters.
Conclusion:  The study has shown that solid-state fermentation system significantly enhanced the tylosin yield by mutant γ-1.
Significance and Impact of the Study:  This study proved to be very useful and resulted in 6·87 ± 0·30-fold increase in tylosin yield by this mutant when compared to that of wild-type strain.  相似文献

To test whether there are differences between living lineages of domestic guinea pigs Cavia porcellus , we studied 118 specimens from six breeds collected along six Andean countries as well as 15 from the wild cavy species ( Cavia tschudii ). The mean weight and body length of 15 adult wild cavies (295±31 g, 242±8.3 mm) were significantly smaller than 25 creole guinea pigs from Bolivia and Chile (639±157 g, 287±23.7 mm, respectively). Eighteen laboratory/pet guinea pigs (including the English Pirbright breed) were also smaller (900±173 g, 308±21 mm) than 25 improved ones from Peru (Tamborada breed, 1241±75.4 g, 317±12 mm) and Ecuador (Auqui breed, 1138±65.5 g, 307±8 mm). Similar size increases appeared in the first axis of a principal component analysis of six skeletal measurements, recovering 84% of total variation. Phylogenetic and haplotype analyses of complete cytochrome b gene sequences consistently joined all 22 domestic individuals (13 shared unambiguous substitutions, 100% bootstrap in 1000 replicates), probably from a single first ancient domestication in the western Andes. Six laboratory/pet sequences were also joined within a common branch (six shared substitutions, 96% bootstrap), probably from a documented European second phase. By contrast, those from improved Auqui joined a northern creole subgroup (one shared substitution, 84% bootstrap), and those from Nativa and improved Tamborada clustered together and with a southern creole subgroup (four shared substitutions, 86% bootstrap); this suggests at least two independent modern events during a more complex third phase, producing two improved guinea pigs selected for size and meat. Deltamethrin, a sintetic pyrethroid, is the insecticide that has been replacing recently to others like organochlorines, organophosphates and carbamates which are less toxic for birds and mammals, although, unfortunately, all of them are highly toxic to various non-targeted aquatic organisms including fish. In the present study, the consequences of the exposition of gilthead seabream (Sparus aurata L.) specimens to sublethal bath dose of deltamethrin (0.1 ppb) on organo-somatic indexes, immunity, seric metabolic parameters, oxidative stress and liver histology were determined after 1, 3, 7 and 14 days of exposure. Deltamethrin alters gilthead seabream immune status, the hepato-somatic index and various seric metabolic parameters since the first exposure day while important progressive deleterious morphological changes in liver were also observed. However, no statistically significant deviation was detected in the expression of oxidative stress-related genes whilst the expression of cytochrome P450 gene was up-regulated in head-kidney and liver of exposed fish. Overall, the present results indicate severe immunotoxicological and metabolic effects of deltamethrin in gilthead seabream, the species with the highest rate of production in Mediterranean aquaculture. In general, the values obtained for the tested parameters during the trial seem to indicate that specimens try to adapt to this adverse situation although the continuous presence of the toxic impede the hypothetic recovery of homoeostasis. The use of deltamethrin in the proximities of seabream farms should be carefully considered.  相似文献

The study objective was to evaluate the enhanced profitability of monosex all-female stock (MS) culture compared to bisexual stock (BS) of Eurasian perch ( Perca fluviatilis L.) under intensive conditions. Monosex all-female stock was produced by mating masculinized females (neomales) with common females of a local strain; the bisexual stock was obtained from a local strain perch broodstock. Preceded by pond-nursing (60 days) and habituating (14 days) periods, the experiment lasted 126 days. Perch were held in six 50-L aquaria connected to a recirculation system, with each group tested in triplicate. No significant differences (P = 0.07) in total survival were observed between all-female and bisexual stock after 126 days of rearing, but MS gained 20% more (P = 0.04) in total body weight (final weight 25.2 ± 9.7 g; mean ± SD) than BS fish (21.0 ± 7.5 g). It is suggested that the culture of monosex female perch might therefore be of significant economic benefit. Overall feed conversion ratio for tanks with BS (1.43 ± 0.21) was significantly (P = 0.02) higher than in tanks with MS (1.30 ± 0.11) perch.  相似文献

The macro and trace mineral composition of vertebrae and muscle in wild and cultured haddock Melanogrammus aeglefinus was determined. Results indicated that haddock deposit c . 10% higher amounts of mineral than salmonids. Based on distribution between the analysed tissues as well as concentration differences, the minerals in wild and cultured haddock were divided into four categories: (1) As, Mn, Na, S and Zn concentration did not show any significant ( P  > 0·05) variation in both muscle and vertebrae; (2) Cr, Co, Mg, Mo and Si concentrations in wild and culture stocks muscle did not vary but vertebrae concentration showed significant differences ( P  < 0·05); (3) Ca and Cu concentration varied in muscle while remaining the same in vertebrae; (4) Al, Ba, Fe, P, K and Sr concentration showed a significant difference in both muscle and vertebrae. Vertebrae P content of cultured haddock was lower than that of wild fish whereas muscle P content was relatively high in cultured fish. Vertebrae and muscle Fe content of cultured haddock was significantly higher than in wild fish. The Mg, K, Co and Cr concentrations in vertebrae were significantly higher in wild haddock than in cultured fish, whereas Mo and Al levels were low. These differences in trace element composition of wild and cultured haddock were probably related to differences in food source and therefore the mineral composition of the diet. The nutritional significance of these findings is discussed.  相似文献

Heat-, cold- and hydrostatic pressure shocks were applied in order to improve triploidy induction in European catfish ( Silurus glanis L . ). A 41°C heat shock (45 s, starting 9 min after gamete activation) provided 88% triploids and a high percentage of malformation (38.8 ± 4.1%). The superior 6°C cold shock (20 min, starting 9 min after gamete activation) gave 100% triploids and a 33.4 ± 3.8% triploid yield. The earliest hydrostatic treatments (600 kg cm2), lasting 4 min and starting 3 min after gamete activation, gave 97.8 ± 1.8% triploids and a 33.7 ± 16.9% triploid yield. The ploidy level was investigated using four approaches: karyotyping, quantification of Ag-stained nucleoli per cell, flow cytometry, and erythrocyte nuclear sizing by computer-assisted image analysis. Induction of triploidy under mass conditions in three experiments gave triploid percentages of 74%, 83% and 66%. Five months later, the percentage of triploids significantly decreased to 12.4%, 8.2% and 21.4%. The growth performance of yearlings was better in diploids than in triploids. Differences between diploids and triploids were 13.5% (NS), 27.6% (P   < 0.001) and 25.4% (P   < 0.05) in the three experiments. Analysis of variance showed the influence of ploidy (P   < 0.001) on growth rate, and multiple range analysis (LSD) assessed differences between total diploids (12.6 g) and total triploids (9.5 g) at the P   < 0.01 level.  相似文献

The effects of high Zn concentration were investigated in sugar beet ( Beta vulgaris L.) plants grown in a controlled environment in hydroponics. High concentrations of Zn sulphate in the nutrient solution (50, 100 and 300 μ m ) decreased root and shoot fresh and dry mass, and increased root/shoot ratios, when compared to control conditions (1.2 μ m Zn). Plants grown with excess Zn had inward-rolled leaf edges and a damaged and brownish root system, with short lateral roots. High Zn decreased N, Mg, K and Mn concentrations in all plant parts, whereas P and Ca concentrations increased, but only in shoots. Leaves of plants treated with 50 and 100 μ m Zn developed symptoms of Fe deficiency, including decreases in Fe, chlorophyll and carotenoid concentrations, increases in carotenoid/chlorophyll and chlorophyll a / b ratios and de-epoxidation of violaxanthin cycle pigments. Plants grown with 300 μ m Zn had decreased photosystem II efficiency and further growth decreases but did not have leaf Fe deficiency symptoms. Leaf Zn concentrations of plants grown with excess Zn were high but fairly constant (230–260 μg·g−1 dry weight), whereas total Zn uptake per plant decreased markedly with high Zn supply. These data indicate that sugar beet could be a good model to investigate Zn homeostasis mechanisms in plants, but is not an efficient species for Zn phytoremediation.  相似文献

Application of sodium-dikegulac reduced plant height with associated increase in branch and leaf number and root biomass inC. roseus (L.) G. DON. Chlorophyll content reduced significantly after first month of 100 and 250 μg/ml DK application. However, such reduction was replaced by significant rise after forth month in 250 μg/ml DK application and fifth month in 100 μg/ml DK application followed by appreciable decline only in 250 μg/ml DK treatment but 100 μg/ml DK maintained higher level till harvest. Total sugar content was significantly high during forth and fifth month stage of growth after DK application. Amino acid content was higher during third to fifth month in 100 μg/ml DK treatment and during third to forth month in 250 μg/ml DK treatment. Tryptophan, on the other hand showed higher content at the fifth month stage of growth after application of DK in both the concentrations. Leaf and root dry weight as well as total alkaloid content were highest in 100 μg/ml DK application. DK, therefore, appears to be a potential chemical for increasing biomass and alkaloid content inC. roseus.  相似文献

The reproduction of Astyanax scabripinnis captured bimonthly in the Cristais small stream (20o00'03'S and 43o56'46'W), in the Das Velhas River basin, was studied using gross anatomical and histological techniques. Standard length of females (56 ± 8 mm) was longer than males (49 ± 6 mm). The diameter of the nucleus of the primary spermatogonia was 7.5 ± 0.9  μ m and of the spermatozoon head 1.8 ± 0.4  μ m. For the females, the oogonia nuclei measured 10.2 ± 2.2  μ m and the vitellogenic oocyte nuclei measured 675.0 ± 66.7  μ m. The following stages of the reproductive cycle were established: developing, mature, partially spent/spawned and completely spent/spawned. Maximum number of males (61.3%) and females (58.2%) at the mature stage was observed during November/December. The gonadosomatic index of males and females was highest during the mature stage (2.4 ± 0.6; 7.5 ± 1.3, respectively) and decreased in subsequent stages. In females, the hepatosomatic index was highest during the developing stage (2.8 ± 0.3), and slightly decreased in the following stages. The highest stomach repletion index found for males was observed during the partially spent stage (2.0 ± 1.2), while for the females, this index varied discretely between the reproductive stages. The highest values of Fulton's condition factor were registered during the mature stage in both sexes (2.9 ± 0.4 for males; 3.0 ± 0.3 for females). The continuous reproductive period and the presence of partially spawned females during the entire sampling period indicated that the species has fractional spawning.  相似文献

The impact of delayed feeding early in development on late feeding ability and development of spotted sand bass Paralabrax maculatofasciatus larvae was examined. Larvae were sampled from hatching until day 19 after delayed feeding for zero (control), 1, 2, or 3 days. Feeding incidence was evaluated as the percentage of larvae with food in the gut and feeding intensity was measured by direct counting of prey after dissection of the gut. Delayed feeding effects due to starvation were observed early in development. By day 5, significant differences ( P  < 0·05) were observed between controls and larvae submitted to degrees of delayed feeding, including total length ( L T), eye diameter, and the size of the head, liver, gut, muscles and body. Differences were still apparent in L T, and body, liver and muscle heights at the end of the studied period. Larvae under total starvation did not survive beyond day 5. Initial feeding incidence was 35, 60, 90 and 10% for larvae first fed on day 2, 3, 4 or 5, respectively. Mean ±  s . d . feeding intensity was 3·6 ± 0·8, 2·8 ± 1·3, 5·2 ± 0·3, and 10·2 ± 1·5 rotifers per larva depending on whether larvae were initially fed on day 2, 3, 4 or 5, respectively. The point of no return occurred between 4 and 5 days after hatching.  相似文献

117 goosefish plasma from the Mediterranean coast have been analysed. Total plasma proteins gave mean values of 2.8±0.9 for males and 3.3±1.1 g/100 for females. The cellulose acetate and starch-gel electrophoresis showed 7 and 13 fractions respectively. Different albumin patterns were observed but their distribution did not have a genetic basis. In 107 specimens a mean TIBC value of 278 ± 125 μg Fe% was found. The saturation coefficient of the transferrin was from 15–20%. The specimens had a single Tf band and one individual had a slower Tf band. The presence of an haptoglobin polymorphism has been suggested. The plasma haptoglobin concentration by an electrophoretic method was determined and a concentration from 28–31 mg Hp% was obtained.  相似文献

Changes in anatomical organisation of the leaf, photosynthetic performance and wood formation were examined to evaluate the temporal and spatial patterns of acclimatisation of micropropagated slow-growing black mulberry ( Morus nigra L.) plantlets to the ex vitro environment. Leaf structure differentiation, the rates of net photosynthesis (Pn), transpiration (E) and stomatal conductance (gs), and secondary xylem growth were determined in the course of a 56-day acclimatisation. Differentiation of palisade parenchyma was observed 7 days after transfer. At this stage, the rates of Pn, E and gs reached maximum values, after which the rates of all three gas exchange parameters gradually decreased. The highest proportion of woody area occupied by vessels was also observed 7 days after transfer. An important feature of developing woody tissue is the difference in patterns of vessel distribution from the characteristic differentiation patterns of earlywood and latewood vessels in mature wood of ring-porous trees. Vessels with lumen areas over 3000 μm2 were only differentiated in acclimatised plantlets, whereas vessels in stems sampled on days 0 and 7 had very small lumen areas of up to 560 μm2. Full acclimatisation, observed 56 days after transfer to the ex vitro environment, was associated with the rapid growth of new in vivo formed leaves, very low rates of E and gs, and much increased secondary xylem tissue within the stem area.  相似文献

Blood chemistry values were measured from 31 harbor porpoises ( Phocoena phocoena ) released from herring weirs in the Bay of Fundy, Canada. Sodium, phosphorus, calcium, chloride, magnesium, total protein, albumin, globulin, urea, cholesterol, serum osmolality, and alanine aminotransferase levels fell within ranges reported for captive harbor porpoises and other odontocetes. Glucose, potassium, creatine kinase, aspartate aminotransferase, hemoglobin, thyroxine, bilirubin, and alkaline phosphatase levels were generally higher than those reported for captive odontocetes. Cortisol and creatinine levels were significantly higher ( P = 0.0356 and 0.0174, respectively) in porpoises that were handled for longer periods and fitted with electronic tags (mean cortisol value = 314 ± 107 nmol/L, mean creatinine value = 94 ± 14 μmol/L) than those receiving roto-tags (mean cortisol value = 222 ± 82 nmol/L, mean creatinine value = 75 ± 15 μmol/L). A more controlled study of duration of handling, time spent out of water, degree of manipulation, and method of tag attachment is required to identify which factors are responsible for the stress response.  相似文献

Acetate, a glial-specific substrate, is an attractive alternative to glucose for the study of neuronal-glial interactions. The present study investigates the kinetics of acetate uptake and utilization in the rat brain in vivo during infusion of [2-13C]acetate using NMR spectroscopy. When plasma acetate concentration was increased, the rate of brain acetate utilization (CMRace) increased progressively and reached close to saturation for plasma acetate concentration > 2–3 mM, whereas brain acetate concentration continued to increase. The Michaelis–Menten constant for brain acetate utilization (      = 0.01 ± 0.14 mM) was much smaller than for acetate transport through the blood–brain barrier (BBB) (      = 4.18 ± 0.83 mM). The maximum transport capacity of acetate through the BBB (      = 0.96 ± 0.18 μmol/g/min) was nearly twofold higher than the maximum rate of brain acetate utilization (      = 0.50 ± 0.08 μmol/g/min). We conclude that, under our experimental conditions, brain acetate utilization is saturated when plasma acetate concentrations increase above 2–3 mM. At such high plasma acetate concentration, the rate-limiting step for glial acetate metabolism is not the BBB, but occurs after entry of acetate into the brain.  相似文献

The morphogenetic differentiation of synapses in the cerebellum and the optic tectum of darkand light- reared rainbow trout was investigated at critical stages of development. During normal differentiation the cerebellum is characterized by the appearance of 'indented', spinelike synapses. This type of synapses increases with age and prevails from day 60 on. At the same time the number of 'flat' synapses decreases. In the cerebellum the highest synaptic density (123 ± 12 synapses/1,000 μm2) is reached 30 days after hatching when the larvae begin to swim. The optic tectum is characterized by a preponderance of flat synapses in early postnatal and adult life; maximal synaptic density (66 ± 5 synapses/1,000 μm2) is reached 60 days after hatching when the larvae have reached optimal visual acuity.
Light deprivation causes a considerable and significant reduction in the number of synapses per unit area in the cerebellum and the optic tectum. The length of synaptic contacts do not change. If light-deprived, the density of synaptic vesicles decreases significantly in the optic tectum of a 25-day-old trout (74 ± 3 instead of 132 ± 7 vesicles/μm2). The objective of this study was to develop a novel liposomal formulation, containing phosphatidylserine (PS), of buparvaquone (BPQ) and to evaluate its in vivo effectiveness in Leishmania (L.) infantum chagasi-infected hamsters. The activity of BPQ was evaluated against both the promastigote forms of different Leishmania species and the intracellular amastigotes of L. (L.) infantum chagasi. Buparvaquone was entrapped in PS-liposomes (BPQ–PS-LP), and the drug was quantified by ultra-high-performance liquid chromatography. The treatment was quantified by detecting the RNA of the living amastigotes in the spleen and the liver by real-time PCR. In vitro assays with L. (L.) infantum chagasi intracellular amastigotes were performed in peritoneal macrophages for the evaluation of the 50% inhibitory concentration (IC50). BPQ–PS-LP at 0.33 mg/kg/day for eight consecutive days reduced the number of amastigotes by 89.4% (P < 0.05) in the spleen and by 67.2% (P > 0.05) in the liver, compared to 84.3% (P < 0.05) and 99.7% (P < 0.05), respectively, following Glucantime® treatment at 50 mg/kg/day. Free BPQ at 20 mg/kg/day failed to treat the hamsters when compared to the untreated group. BPQ was significantly (P < 0.05) selective against L. (L.) infantum chagasi intracellular amastigotes, with an IC50 value of 1.5 μM; no in vitro mammalian cytotoxicity could be detected. Other cutaneous species were also susceptible to BPQ, with IC50 values in the range 1–4 μM. BPQ–PS-LP caused a significant reduction in the parasite burden at a 60-fold lower dose than did the free BPQ. These results show the potential of PS-liposome formulations for the successful targeted delivery of BPQ in visceral leishmaniasis.  相似文献
