The teguments of developing and mature cercariae, recently transformed, and 1-wk-old schistosomula and adult worms were examined for the ultrastructural location of macromolecular carbohydrates and polyelectrolytes. The surface of mature cercariae within sporocysts and cercariae released from the snail is covered by a filamentous coat which reacts with cytochemical reagents for the demonstration of vicinal glycols, but neither the coat nor the surface of the tegument plasmalemma binds cationic colloidal iron at low pH.Upon penetrating mammalian skin, the cercaria sheds its surface coat; the tegument surface of newly transformed schistosomula, older schistosomula and adult worms stains en bloc with acidic colloidal iron, as does the tegument plasmalemma of mature cercariae if the overlying filamentous coat is first removed by physicochemical means. The cercarial coat thus serves to mask anionic groups at the surface of the tegument plasmalemma which become functionally exposed after penetration of the mammalian host. The distribution of colloidal iron binding sites coincides with those for the carbohydrate-complexing phytohemmagglutnin, concanavalin A, which suggests that these membrane-fixed anions are acid mucopolysaccharides, glycoproteins or glycolipids. Carbohydrate-containing material was also localized within membrane-bound vesicles of the tegument matrix and perikarya of developing cercariae and postcercarial schistosomes, suggesting that surface mucosubstances contributing to the tegument glycocalyx of these worms are elaborated, at least in part, by the tegument itself.  相似文献

A hallmark feature of parasitic platyhelminths is a cytoarchitecturally unusual syncytial epidermis composed of a peripheral layer of continuous cytoplasm (the ectocytoplasm) connected to underlying nucleated cell bodies by small cytoplasmic bridges. The helminth epidermis, or tegument, plays important roles in protection and nutrient acquisition; cestodes, in fact, completely lack a gastrointestinal tract and absorb all nutritive material through the tegument. Perhaps not surprisingly, the cestode tegument bears certain resemblances to the mucosal epithelium of the vertebrate small intestine, including the possession of a microvillous brush border upon the surface of the ectocytoplasm. In contrast to the intestinal epithelial cell, however, very little is known concerning the nature and organization of the cytoskeleton within the helminth epidermis. Therefore, a number of different microscopical preparative techniques were used to examine the tegument of the tapeworm Hymenolepis diminuta for the presence and distribution of microfilaments, intermediate filaments, and microtubules. It was found that both actin-containing microfilaments and intermediate-sized filaments are present but are restricted to specific locations along the plasmalemmae of the ectocytoplasm. In contrast, microtubules are found throughout the tegument, and are concentrated in the supranuclear regions of the perikarya and in the cytoplasmic bridges interconnecting the perikarya and ectocytoplasm. Unlike brush borders of most other epithelia, the cestode epidermal brush border lacks a filamentous terminal web and is instead associated with microtubules. A network of fine filaments, 5-8 nm in diameter but distinct from actin-containing microfilaments, runs throughout the ectocytoplasm and appears to interlink tegumental vesicles. These fine filaments may represent the primary "skeletal" system responsible for maintaining the structure of the tegumental cytoplasm.  相似文献

A new type of cell has been identified in cercariae of Schistosoma mansoni. The perikarya (cell bodies) of these cells were located in the body (midsegment), in an area oral to the acetabulum (ventral sucker). Cytoplasmic processes extending from the perikarya ramified throughout the parenchyma of the anterior organ (oral sucker), body, and tail segments by following the path of the nerve processes from the neuropile. The perikarya of these cells had heterochromatic nuclei and a predominance of particulate material and granules (240-360 nm) in their cytoplasm. Aggregates of granules (240-360 nm) and associated vesicles (34 nm) were scattered throughout the cytoplasmic processes of the cells and formed distinct varicosed areas. These processes often connected to the tegument in the midsegment (body) of the cercariae. The granules and associated vesicles reacted (became electron dense) with fixatives reported to be detectors of biogenic amines: The glutaraldehyde/osmium tetroxide fixation procedure rendered the granules electron dense while the glutaraldehyde/chromate/osmium tetroxide fixation procedure rendered the granules and the associated vesicles electron dense. The chromate solution of the latter procedure was responsible for the electron density of the associated vesicles. The morphology of these cells (their long ramifying cytoplasmic processes) and their reaction to chromium suggests that they are probably biogenic aminergic sensory cells.  相似文献

The possibility of pinocytosis occurring in the tegument of the plerocercoid of Schistocephalus solidus has been investigated by morphological and experimental methods. Electronmicroscopic study showed that the outer syncytial tegument contained numerous electronlucid vesicles. These vesicles had two gradients, the number of vesicles decreasing from the outer canopy region to the inner canopy and from the apical to the basal plasma membrane for any particular region of tegument. A variety of morphological modifications of the apical plasma membrane very similar to those which have been accepted as evidence of pinocytosis in other tissues were present. In vitro studies using horseradish peroxidase, ruthenium red, and lanthanum nitrate showed that all three tracers are taken up by the tegument into membrane-limited vesicles. Vesicles which contained ruthenium red occurred at the base of the tegument within a 5-min incubation period, and their contents appeared to be released into the underlying interstitial material by exocytosis.  相似文献

Ultrastructural observations using transmission and scanning electron microscopy reveal the tegument to be basically similar to that of other cestodes. The syncytial distal cytoplasm is devoid of organelles except for rod-shaped bodies, believed to be secretory vesicles, and lamellated bodies which probably contribute the raw material for new microtriches. There is evidence that these vesicles originate from the Golgi found in the sub-cuticular cells.Three types of microtriches are described: typical ones with well-developed spines, ones with short filaments instead of spines, and ones with no spines. Microtriches with spines are found only on the anterior part of the worm and may serve to anchor the worm. Microtriches on the posterior have no spines and are believed to be primarily involved in the absorption of nutrients. Between these two regions there is a transitional zone where all three types of microtriches can be found. In general the microtriches are quite uniformly distributed throughout the surface of the worm. The presence of cestodarian-like microtriches raises interesting evolutionary questions.Histochemical tests localized acid and alkaline phosphatase activity on various parts of the tegument, as well as on host intestine, while acian blue tests showed that acid mucopolysaccharide levels correspond with the concentration of the tegument vesicles.  相似文献

寄生鳜鳃部的河鲈锚首吸虫的体壁由表皮合胞体、基板、环肌、纵肌和表皮细胞核周体所组成。合胞体顶部质膜起伏形成表皮的嵴纹,基部质膜折叠形成指状突起伸入到合胞体中。合胞体表面覆盖着一层糖萼。河鲈锚首吸虫的表皮中含有四类分泌体,即电子致密的分泌颗粒、中等电子致密的分泌颗粒、有膜包围的电子稀疏的分泌体和多囊体.可见分泌体和合胞体基质通过胞吐作用排到体外,未见吞饮小泡,推测表皮的主要功能在于分泌和渗透压调节而非营养吸收。在外侧头瓣的乳突状结构所在处,合胞层较薄,基板平滑,在实质组织中的一腔体样结构中可见囊状体、电子致密度各异的颗粒体、泡状体和电子致密的基质团,神经突起分布于腔体周围,这类乳突可能代表一类新的非纤毛感受器类型。  相似文献

The ultrastructure of the tegument and tegument-associated microorganisms of the gyliauchenid digenean Gyliauchen nahaensis is described by transmission and scanning electron microscopy. The tegument is devoid of surface spines and is characterized by a moderately folded apical membrane, abundant vesicles, basal mitochondria, a folded basal plasma membrane, and a thick basal matrix. Microorganisms form a dense biofilm on the tegument of the posterodorsal surface and the excretory papilla. At least 7 microbial morphotypes were identified, including eubacteria, spirochaetes, and nanobacteria.  相似文献

The tegument, or body wall, of schistosomes is the primary tissue for host interaction and site targeted schistosome vaccination. However, many aspects of the cell biology, particularly differentiation and maintenance, remain uncharacterised. A leading vaccine candidate, Schistosoma mansoni tetraspanin 2 has proven efficacy in experimental models, but its function, precise subcellular location in the tegument and role in tegument biology is not well understood. A primary question is whether this molecule is a true surface molecule, that is, whether it appears within the apical membrane of the tegument. Hitherto, the target sequence for antibody localisation studies had not been available for advanced subcellular localisation studies, such as immuno-electron microscopy, due to aldehyde sensitivity. To circumvent this problem, we adapted the methods of high pressure freezing and cryosubstitution with uranyl acetate for immuno-electron microscopy. The tri-dimensional structure of tegument membranes was resolved using electron tomography. Immunolocalisation of Schistosoma mansoni tetraspanin 2 demonstrates that the molecule is localised to tegument membrane compartments, but predominantly within internal structures associated with surface invaginations and internal vesicles. Surprisingly, no label was found at the virtual surface of the parasite. The significance of this localisation pattern is discussed.  相似文献

The multilaminate vesicles present in the tegument cytoplasm appear to fuse with side channels projecting out into the cytoplasm from the base of the surface pits. Their lamellate contents then unroll and spread out to form a trilaminate membranocalyx lining the pits and covering the tegument surface. The plant lectin concanavalin A appears to stabilize the process of vesicle fusion leading to an aggregation of multilaminate vesicles trapped in the lumen of the surface pits. The membranocalyx can be labelled with cationized ferritin. Chase incubations in normal medium indicate that by 4 h most of the label and the membranocalyx to which it is bound have been lost to the medium. This suggests that under the conditions of these experiments the membranocalyx has a half-life of 2-3 h.  相似文献

The morphology of the tegument in the redia of Paramphistomum epiclitum (Digenea: Paramphistomidae) resembles that shown by most larval and adult digeneans; an outer surface syncytium is in continuity with the cytoplasm of in-sunken, nucleated cytons. Although tegumental cytons usually contain a single nucleus, some display up to six nuclei. The tegumental syncytium lining the pharynx of P. epiclitum rediae lack underlying cytons. The apical membrane of the tegument is elaborated by folds and microvilli, which presumably facilitate uptake of nutrients and/or exchange of ions involved in osmoregulation. A single type of secretory body, resulting from the fusion of smaller vesicles produced at Golgi complexes in the cytons, occurs throughout the tegument. Uniciliate sensory receptors occur in the surface syncytium particularly around the oral opening.  相似文献

In the pulmonate mollusc Helix, neurosecretory cells have perikarya that form neurohaemal complexes peripherally beneath the inner surface of the neural lamella and give rise to axons with varicosities in the neuropile. Two categories of secretory inclusions are present throughout the cytoplasm and these accumulate adjacent to sites of release. Secretory granules invariably have electron-dense contents, whereas smaller vesicles have fairly lucent contents following fixation in OsO4, but are dense-cored in material fixed initially with aldehyde. Vesicles (‘synaptic vesicles’) at central sites appear identical to those (‘synaptoid vesicles’) at peripheral, neurohaemal locations. At both neurohaemal and central sites, both granules and vesicles discharge their contents by exocytosis, this process being most clearly visualized in tissues treated with tannic acid.  相似文献

利用透射电镜观察了头槽绦虫 (Bothriocephalusacheilognathi)原尾蚴皮层的超微结构。头槽绦虫原尾蚴的皮层为典型的合胞体结构。皮层表面有浓密的微毛与少量的突起。与成虫相比 ,原尾蚴的微毛长而且粗 ,呈现单态性 ,推测在进入终末宿主的过程中有脱落和再生现象。突起分布在头部与体侧 ,含有较少的电子致密颗粒。在原尾蚴皮层细胞质中观察到三种腺细胞的分泌过程 ,即顶分泌、外分泌和微分泌过程。原尾蚴身体后部观察到尾钩和穿刺腺管道的开口。在核周区和纤维层下可见发育较好的环肌与纵肌。本文还对原尾蚴皮层突起的功能进行了探讨。  相似文献

Summary In the nervous system of the obligatory endoparasite Diphyllobothrium dendriticum immunoreactivity (IR) to growth hormone-releasing factor (GRF), peptide histidine isoleucine (PHI), bovine pancreatic polypeptide (BPP), gastrin, gastrin-releasing peptide (GRP), oxytocin, FMRF-amide (FMRF) and serotonin (5HT) was demonstrated by immunocytochemical methods. A very strong GRF-IR was observed in the CNS and PNS of larvae and of the constantly growing adult worms. GRF-IR axon terminals occur beneath the basal lamina of the tegument along the inside of the bothridia, the holdfast organ of the worm. GRF-IR fibres surround the yolk producing vitelline glands and occur in the wall of the vagina. PHI-IR was observed in the CNS and PNS of larvae and adult worms. PHI-IR terminals occur beneath the basal lamina of the tegument along the strobila, the nutrient absorbing surface of the worm. PHI-IR fibres seem to innervate the testicular follicles. FMRF-IR fibres and perikarya occur close to the vitelline glands and the uterine pore and in the male copulatory organ. Numerous large 5HT-IR perikarya with long varicose fibres were observed in the nervous system of the worm. 5HT-IR perikarya occur close to the genital atrium. D. dendriticum is the phylogenetically lowest organism in which IR to PHI has been demonstrated.  相似文献

Summary The present immunocytochemical study concerns the distribution of four neuropeptides, FMRF-amide, vasotocin, leu-enkephalin and neurotensin, and of the bioamine serotonin in the plerocercoid larva of Diphyllobothrium dendriticum. Anti-FMRF-amide and vasotocin-reactivity occurs in perikarya and nerve fibres in the CNS and PNS of this worm. The peptide-containing fibres surround and seem to innervate the musculature and to terminate beneath the basal lamina of the tegument at the inner surface of the bothridia, suggesting a neurotransmitter function. Antileu-enkephalin reaction occurs in perikarya and fibres in the main nerve cords and in the PNS. Anti-neurotensin reactive fibres were observed in the neuropile of the nerve cords. Serotonin immunoreactivity was found in neurons in the ganglionic commissure of the brain and along the main nerve cords. This study is the first immunocytochemical identification of neuropeptides and serotonin in a parasitic flatworm and the information gained may be of importance for the development of new antihelminthics.  相似文献

Light and electron microscopic studies of the 'buccal suckers' of Gotocotyla secunda showed that they are complex structures with muscular, glandular, (resorptive?), and probably sensory components. The name 'buccal organ' is therefore proposed for them. Ducts run from the lumen of the buccal organs to their dorsal, ventral and lateral surface and communicate with lacunae surrounding them. The surface of the ducts and of the muscular walls of the buccal organs are lined by tegument with short microvilli, tegument with dense, long surface lamellae, and tegument with thick, long microvilli which have an electron-dense core. The latter type of tegument contains many vesicles and electrondense ovoid bodies, and is traversed by gland ducts containing large secretory droplets. Electron microscopic examination showed that the prebuccal convoluted structures in Heteromicrocotyloides mirabilis are a modified tegument connected to subtegumental cells. Light microscopic studies of Gotocotyla bivaginalis. Pseudothoracocotyla indica, Pricea multae and Heteromicrocotyla australiensis showed that the first three species have buccal organs similar to those in G. secunda , whereas the last species resembles H. mirabilis. It is tentatively assumed that the buccal organs play a role in finding suitable sites for bloodfeeding.  相似文献

Changes in the ultrastructure of the tegument and subtegumental cells of 4-day-old Hymenolepis diminuta were studied in vitro in 50% fresh normal rat serum over a 5-h period and compared with heat-inactivated serum and saline controls. First, membrane-bound vesicles accumulate above the microthrix-border. After 30–40 min large vacuoles, which may contain membranous elements, appear in the tegument at a time when the surface of the young strobila is virtually denuded of the microthrix-border. With prolonged incubations there are subtegumental secretory inclusions with dark, enveloping cytoplasm in the tegument and finally the apical plasma membrane, together with the majority of the matrix, is lost. The disrupted portion of the worm is abruptly demarcated from the comparatively intact scolex/anterior neck region by a constriction. Even after 5 h incubation there is no evidence of loss of tegumental matrix components from regions anterior to the constriction but the neck region shows a significant denudation of the microthrix layer and the tegument contains numerous inclusions. The scolex tegument only showed little evidence of loss of membrane from the surface. Possible mechanisms for the avoidance of complement-mediated lysis in the anterior region are discussed.  相似文献

The tegument of the polyopisthocotylean monogenean Atriaster heterodus Lebedev & Parukhin, 1969 was studied using transmission electron microscopy. The outer syncytial layer of the tegument is connected to the internal cell bodies by cytoplasmic extensions which interweave between the muscular fibres. The free surface of the syncytium has projections of the external membrane which are similar to microvilli. The undulating basal membrane, with numerous narrow elongate projections, is associated with the basal lamina situated between the syncytial and muscular layers. The cell bodies and syncytial layer of the tegument exhibit two types of vesicles, one with fibrous contents and one with electron-dense contents; these were analysed using two cytochemical tests, the E-PTA and alcian blue methods, used for the first time on monogeneans.  相似文献

Pricea multae is a gill parasite of Scomberomorus spp. In the present study, the surface specialization and ultrastructure of the tegument and haptor were examined in detail. The buccal cavity and the preoral pit bear uniciliated sensory endings. The genital atrium opens on the ventral surface whereas the vaginal pore opens on the dorsal surface of the anterior region. The general body surface is elevated by pits and folds together with papillae-like uniciliated sensory endings. Microvilli are uncommon.

The syncytium of the tegument is bounded by apical and basal plasma membranes. Some electron-dense granules and electron-lucent vesicles were observed exocytosing at the apical surface. The tegumental perikarya lie amongst the muscle blocks underneath the basal lamina. The body wall muscle fibres are non-striated consisting of thick and thin myofilaments. Each muscle fibre is limited by a sarcolemma and separated from neighbouring fibres by interstitial material.

The haptor possesses two rows of pincer-type clamps. The clamps may be withdrawn by muscles into cavity-like depressions. Electron-dense fibrils are embedded within the clamp sclerite matrix. The intrinsic muscles of the clamp include radial muscles enclosed within the basal lamina. The muscle fibres enclosed within the basal lamina of the clamp appear to differ from the body wall muscles by the absence of interstitial material, the possession of well defined gap junctions between the sarcolemma of adjacent muscle fibres, abundant sarcoplasmic reticulum and numerous mitochondria. The extrinsic muscles of the clamp presumably arise from the dorso-lateral surface of the haptor and are apparently connected to the tendon of the clamp. A cluster of gland cells occurs in the haptor beneath each clamp. The haptor is richly supplied with nerve fibres which are associated with groups of neurones. Each axon is non-myelinated and contains presumed neurosecretory granules. The peripheries of some of the large axons are subdivided by invaginations of the axon wall.  相似文献

A diverse afferent synaptic input to immunostained oxytocin magnocellular neurons of the paraventricular nucleus of the rat hypothalamus is described. By electron microscopy, immunoreactive material is present within cell bodies and neuronal processes and it is associated primarily with neurosecretory granules and granular endoplasmic reticulum. Afferent axon terminals synapse on perikarya, dendritic processes, and possibly axonal processes of oxytocin-containing neurons. The presynaptic elements of the synaptic complexes contain clear spherical vesicles, a mixture of clear spherical and ellipsoidal vesicles, or a mixture of clear and dense-centered vesicles. The postsynaptic membranes of oxytocinergic cells frequently show a prominent coating of dense material on the cytoplasmic face which gives the synaptic complex a marked asymmetry.  相似文献

The tegumental ultrastructure of the intestinal fluke Bucephalus anguillae was studied with the use of scanning and transmission electron microscopy. The surface of the tegument is covered by transverse ridges from which protrude numerous closely packed, digitated, and claw-shaped spines. Cobblestone-like units of the tegument were observed on the crescent-shaped formation of the rhynchus and at the posterior part of the body. Three types of sensory structures were examined, i.e., 2 uniciliated receptors and 1 without cilia. As anterior-posterior differences were observed, particular attention was given to spines and sensory receptors. Spine insertion zones and average cilia length are variable between anterior and posterior tegument areas. Ultrastructural study revealed that the tegument of B. anguillae has a typical syncytial organization with a distal cytoplasm lying over a basal matrix and cytons below. Cytoplasmic bridges allowed transit of secretory vesicles and granules. Diagrams of spines and sensory receptors were made to help in understanding the nature of these structures.  相似文献
