Zapata, A., Carrato, A. 1980. Ultrastructure of elasmobranch and teleost thrombocytes. (Departamento de Morfología Microscópica, Facultad de Biología, Universidad Complutense, Madrid, Spain.) — Acta zool. (Stockh.) 61 (3): 179–182. The ultrastructure of circulating thrombocytes of Raja clavata and Torpedo marmorata (Elasmobranchs) and Rutilus rutilus and Gobio gobio (Teleosts) was described. In elasmobranchs, only one thrombocytic cell type was found, while teleosts presented two types. These cells contained microtubules and cytoplasmic granules, whose functional significance related to clotting processes is analyzed.  相似文献

The rate of thrombopoiesis was measured by platelet incorporation of 35S-sulfate in mice having markedly stimulated or suppressed erythropoiesis, and compared to controls. Twenty-four hr exposure to 0.4 atm in a low pressure chamber caused increased platelet incorporation of 35S, while exposure for 2 to 5 days caused reductions in both 35S uptake and platelet counts at time of sacrifice. These values were progressively reduced with the longer periods of hypoxia. Two units of human urinary erythropoietin (ESF) were injected twice daily for 1 to 5 days and also caused reduced platelet 35S uptake in mice given the ESF for 2, 4, or 5 days. Mice rendered polycythemic by hypertransfusion or previous exposure to low barometric pressure had increased platelet 35S uptake and increased platelet counts. One possible explanation is that the severe hypoxia or the administration of ESF strongly stimulated erythropoiesis, but might have inhibited thrombopoiesis by depleting megakaryocyte precursors in the bone marrow.  相似文献

Jørgensen, J. M. 1980. The morphology of the Lorenzinian ampullae of the sturgeon Acipenser ruthenus (Pisces: Chondrostei). (Zoological Laboratory, University of Aarhus, Denmark.) — Acta zool. (Stockh.) 61 (2): 87–92. The snout of a sturgeon, Acipenser ruthenus (Chondrostei, Osteichthyes) is provided with sensory pores. Light and electron microscopical examination of these reveals that the ampullary organs have a sensory epithelium very similar to what has been found in the Lorenzinian ampullae, which are electroreceptors previously examined at a fine structural level in elasmobranchs and the paddle-fish, Polyodon spathula. The sensory cells are pear-shaped with a very small apical part, in the centre of which there is a short cilium. Basally, the sensory cells make several contacts with button-shaped nerve-endings. The presumed synaptic area in the sensory cell is characterized by a presynaptic sheet surrounded by vesicles. Only one type of nerve ending, an afferent type, has been observed.  相似文献

Carbon and nitrogen stable isotope analyses have improved our understanding of food webs and movement patterns of aquatic organisms. These techniques have recently been applied to diet studies of elasmobranch fishes, but isotope turnover rates and isotope diet–tissue discrimination are still poorly understood for this group. We performed a diet switch experiment on captive sandbar sharks (Carcharhinus plumbeus) as a model shark species to determine tissue turnover rates for liver, whole blood, and white muscle. In a second experiment, we subjected captive coastal skates (Leucoraja spp.) to serial salinity reductions to measure possible impacts of tissue urea content on nitrogen stable isotope values. We extracted urea from spiny dogfish (Squalus acanthias) white muscle to test for effects on nitrogen stable isotopes. Isotope turnover was slow for shark tissues and similar to previously published estimates for stingrays and teleost fishes with low growth rates. Muscle isotope data would likely fail to capture seasonal migrations or diet switches in sharks, while liver and whole blood would more closely reflect shorter term movement or shifts in diet. Nitrogen stable isotope values of skate blood and skate and dogfish white muscle were not affected by tissue urea content, suggesting that available diet–tissue discrimination estimates for teleost fishes with similar physiologies would provide accurate estimates for elasmobranchs.  相似文献

Gill development begins on the sixth day of incubation at 10°C and is complete by 31 days (hatching). Gill arches are formed by fusion and perforation of ectoderm and endoderm across the pharyngeal wall. A primary branchial artery forms within each arch and a second branchial artery forms as a branch from its ventral end. A series of filament loop vessels forms connecting the two arteries and when several are patent a unidirectional blood flow is established via afferent (second) branchial artery, filament loop vessels to efferent (primary) branchial artery. Part of the efferent branchial artery just above its junction with the afferent branchial artery constricts and occludes. It is suggested that this change in the pattern of blood flow is dependent on differences in resistance of the two branchial arteries. A later extension of the gill ventrally is thought not to be homologous with similar regions in elasmobranchs and Acipenser.  相似文献

Intravital visualization of thrombopoiesis revealed that formation of proplatelets, which are cytoplasmic protrusions in bone marrow megakaryocytes (MKs), is dominant in the steady state. However, it was unclear whether this is the only path to platelet biogenesis. We have identified an alternative MK rupture, which entails rapid cytoplasmic fragmentation and release of much larger numbers of platelets, primarily into blood vessels, which is morphologically and temporally different than typical FasL-induced apoptosis. Serum levels of the inflammatory cytokine IL-1α were acutely elevated after platelet loss or administration of an inflammatory stimulus to mice, whereas the MK-regulator thrombopoietin (TPO) was not elevated. Moreover, IL-1α administration rapidly induced MK rupture–dependent thrombopoiesis and increased platelet counts. IL-1α–IL-1R1 signaling activated caspase-3, which reduced plasma membrane stability and appeared to inhibit regulated tubulin expression and proplatelet formation, and ultimately led to MK rupture. Collectively, it appears the balance between TPO and IL-1α determines the MK cellular programming for thrombopoiesis in response to acute and chronic platelet needs.  相似文献

A spin-echo NMR method was applied to study hydrodynamic parameters of cytoplasmic streaming and to examine translational diffusion of cytoplasmic water in stems of elodea (Elodea densa (Planch.) Casp.) treated with antimitotic drugs. Within the first few hours after colchicine treatment, the dynamics of translational displacement followed the curve with a transient maximum. The action of vinblastine was manifested in a monotonic decrease of the translational diffusion. After 24-h treatments with colchicine and vinblastine, the translational diffusion was slowed down. All these changes induced by antimitotic drugs were observed on the background of unaltered rate of cytoplasmic streaming.  相似文献

Gill morphometrics of the three thresher shark species (genus Alopias) were determined to examine how metabolism and habitat correlate with respiratory specialization for increased gas exchange. Thresher sharks have large gill surface areas, short water–blood barrier distances, and thin lamellae. Their large gill areas are derived from long total filament lengths and large lamellae, a morphometric configuration documented for other active elasmobranchs (i.e., lamnid sharks, Lamnidae) that augments respiratory surface area while limiting increases in branchial resistance to ventilatory flow. The bigeye thresher, Alopias superciliosus, which can experience prolonged exposure to hypoxia during diel vertical migrations, has the largest gill surface area documented for any elasmobranch species studied to date. The pelagic thresher shark, A. pelagicus, a warm‐water epi‐pelagic species, has a gill surface area comparable to that of the common thresher shark, A. vulpinus, despite the latter's expected higher aerobic requirements associated with regional endothermy. In addition, A. vulpinus has a significantly longer water–blood barrier distance than A. pelagicus and A. superciliosus, which likely reflects its cold, well‐oxygenated habitat relative to the two other Alopias species. In fast‐swimming fishes (such as A. vulpinus and A. pelagicus) cranial streamlining may impose morphological constraints on gill size. However, such constraints may be relaxed in hypoxia‐dwelling species (such as A. superciliosus) that are likely less dependent on streamlining and can therefore accommodate larger branchial chambers and gills. Mitochondrial and nuclear DNA information was analysed among four subspecies of the African Queen butterfly, Danaus (Anosia) chrysippus sensu lato (s.l.), along with four other Danaus species drawn from all three subgenera (D. (Danaus) plexippus, D. (Salatura) genutia, D. (A.) gilippus, D. (A.) eresimus) and two outgroup species from the same tribe, Tirumala septentrionis and Amauris niavius. A mitochondrial phylogeny derived from the 12S rRNA (347 bp) and COI (537 bp) loci indicates two very distinct haplotypes for subspecies D. (A.) c. dorippus, dorippus‐1 and dorippus‐2. Interestingly, dorippus‐1, on the one hand, and all other D. (A.) chrysippus haplotypes, on the other, are the most distantly related clades within the genus and have different most recent ancestors from different subgenera, though sharing the common ancestor of the monophyletic genus. A phylogeny based on the EF1‐α nuclear locus (400 bp) shows that the two well‐separated mitochondrial lineages of dorippus are identical for this gene and reciprocally monophyletic to the other D. (A.) chrysippus lineages. Thus, nuclear and cytoplasmic phylogenies are not only discordant, but also suggest that both D. (A.) chrysippus s.l. and subspecies dorippus are polyphyletic. Paradoxically, four African subspecies, chrysippus‐orange, chrysippus‐brown, alcippus and dorippus, though substantially vicariant, hybridize extensively in East Africa wherever the ranges of two or more of them overlap. Linkage disequilibrium, and hence sexual isolation, in sympatry between colour (nuclear) genes and unlinked mitochondrial (cytoplasmic) loci is consistent across populations and therefore indicates the operation of positive natural selection. Together with data from previous experimental and field work, our results suggest that extensive hybridization occurs among once allopatric or parapatric lineages, that are now nascent species. We deduce that hybridism among lineages in sympatry is currently enforced, in the face of assortative mate choice, by a bacterial symbiont, Spiroplasma, a male‐killer that forces females in female‐biased populations to pair with heterotypic males. In discussion we emphasize that neither D. It is often assumed that the von Bertalanffy growth model (VBGM) is appropriate to describe growth in length-at-age of elasmobranchs. However, a review of the literature suggests that a two-phase growth model could better describe growth in elasmobranchs. We compare the two-phase growth model (TPGM) with the VBGM for 18 data sets of elasmobranch species, by fitting the models to 36 age-length-at-age data pairs available. The Akaike Information Criteria (AIC) and the difference in AIC between both models revealed that in 23 cases the probability that the TPGM was true ≥50%. The VBGM tends to estimate larger L values than the two-phase growth model, while the k parameter tends to be underestimated. The growth rate in length-at-age appears tends to decrease near the age at first maturity in several species of elasmobranch. The importance of the TPGM lies in that it may better describe this aspect of the life history of many elasmobranchs. In this context, we conclude that the TPGM should be used along with other growth models in order to precisely estimate elasmobranch life history parameters.  相似文献

In elasmobranchs (sharks, rays) the total weight of lymphomyeloid structures isolated by dissection varied between about 0.6 and 1.0% of the body weight. In a holocephalan (Chimaera) the corresponding value was 1.7%. Reciprocal size relations may exist between different types of lymphomyeloid organs. A thymus was found in adult specimens of rays (Raja) and the rabbit-fish (Chimaera). The lymphomyeloid organs supposedly are active in haemopoiesis and the immune responses.  相似文献

The Peruvian sea represents one of the most productive ocean ecosystems and possesses one of the largest elasmobranch fisheries in the Pacific Ocean. Ecosystem-based management of these fisheries will require information on the trophic ecology of elasmobranchs. This study aimed to understand the diet, trophic interactions and the role of nine commercial elasmobranch species in northern Peru through the analysis of stomach contents. A total of 865 non-empty stomachs were analysed. Off northern Peru, elasmobranchs function as upper-trophic-level species consuming 78 prey items, predominantly teleosts and cephalopods. Two distinctive trophic assemblages were identified: (a) sharks (smooth hammerhead shark Sphyrna zygaena, thresher shark Alopias spp. and blue shark Prionace glauca) that feed mainly on cephalopods in the pelagic ecosystem; and (b) sharks and batoids (Chilean eagle ray Myliobatis chilensis, humpback smooth-hound Mustelus whitneyi, spotted houndshark Triakis maculata, Pacific guitarfish Pseudobatos planiceps, copper shark Carcharhinus brachyurus and school shark Galeorhinus galeus) that feed mainly on teleosts and invertebrates in the benthonic and pelagic coastal ecosystem. This study reveals for the first time the diet of T. maculata and the importance of elasmobranchs as predators of abundant and commercial species (i.e., jumbo squid Dosidicus gigas and Peruvian anchovy Engraulis ringens). The results of this study can assist in the design of an ecosystem-based management for the northern Peruvian sea and the conservation of these highly exploited, threatened or poorly understood group of predators in one of the most productive marine ecosystems.  相似文献

Summary The distribution of nerves showing neuropeptide Y (NPY)-like immunoreactivity in the cardiovascular system of elasmobranchs and teleosts has been investigated. Two species of teleosts, the rainbow trout (Salmo gairdneri) and the Atlantic cod (Gadus morhua), and three species of elasmobranchs, the spiny dogfish (Squalus acanthias), the little skate (Raja erinacea) and the starry ray (Raja radiata), were used in this study. An innervation of the cardiovascular system by an NPY-like substance was found only in the two species of Raja. A rich innervation was encountered in these skates, with the highest density of fibres in the wall of the ventricle, the conus arteriosus, the coeliac artery and smaller mesenterial vessels. In the vessels, the fibres formed a plexus at the adventitio-mediol border. Few fibres were found in the walls of the dorsal aorta, the sinus venosus and the atrium, and no fibres were observed in the walls of the ventral aorta. Falck-Hillarp fluorescence histochemistry showed the presence of a rich innervation of arteries and arterioles of the gut by catecholamine-containing nerve fibres.  相似文献

The recent cloning of thrombopoietin has brought many insights into the cellular and molecular mechanisms of megakaryocyte and platelet development. Thrombopoietin was cloned based on its binding to the product of the proto-oncogene c-mpl and was found to affect all aspects of thrombopoiesis. Many of the molecular pathways that mediate thrombopoietin action have been discerned. Upon hormone binding, the megakaryocyte thrombopoietin receptor homodimerizes, activating members of the JAK family of kinases, which, in turn, phosphorylate the receptor, generating docking sites for second messengers that affect multiple signalling pathways. Ultimately, cellular proliferative and anti-apoptotic mechanisms are initiated, increasing megakaryocyte numbers, as are processes that uncouple DNA synthesis from nuclear and cytoplasmic division, generating polyploid cells. As the net result of thrombopoietin action is an expansion of cells that give rise to mature platelets, the availability of the recombinant hormone has provided new opportunities to manipulate blood cell development for therapeutic benefit. The maize (Zea mays L.) CRINKLY4 (CR4) gene encodes a serine/threonine receptor-like kinase that controls an array of developmental processes in the plant and endosperm. The Arabidopsis thaliana (L.) Heynh. genome encodes an ortholog of CR4, ACR4, and four CRINKLY4-RELATED (CRR) proteins: AtCRR1, AtCRR2, AtCRR3 and AtCRK1. The available genome sequence of rice (Oryza sativa L.) encodes a CR4 ortholog, OsCR4, and four CRR proteins: OsCRR1, OsCRR2, OsCRR3 and OsCRR4, not necessarily orthologous to the Arabidopsis CRRs. A phylogenetic study showed that AtCRR1 and AtCRR2 form a clade closest to the CR4 group while all the other CRRs form a separate cluster. The five Arabidopsis genes are differentially expressed in various tissues. A construct formed by fusion of the ACR4 promoter and the GUS reporter, ACR4::GUS, is expressed primarily in developing tissues of the shoot. The ACR4 cytoplasmic domain functions in vitro as a serine/threonine kinase, while the AtCRR1 and AtCRR2 kinases are not active. The ability of ACR4 to phosphorylate AtCRR2 suggests that they might function in the same signal transduction pathway. T-DNA insertions were obtained in ACR4, AtCRR1, AtCRR2, AtCRR3 and AtCRK1. Mutations in acr4 show a phenotype restricted to the integuments and seed coat, suggesting that Arabidopsis might contain a redundant function that is lacking in maize. The lack of obvious mutant phenotypes in the crr mutants indicates they are not required for the hypothetical redundant function.  相似文献

TwoVibrio species identified asV. damsela and a new sucrose-positiveVibrio sp.,V. carchariae sp. nov., were simultaneously isolated from a brown shark which died while being held in captivity at a large aquarium. Pathogenicity studies were subsequently conducted using a variety of elasmobranchs, including smooth dogfish and lemon sharks. Both bacterial strains proved pathogenic, causing death in nearly all of the elasmobranch hosts challenged. Virulence studies revealed that both bacterial strains were cytotoxic for Y-1 mouse adrenal cells. TheV. damsela strain was highly cytotoxic causing Y-1 cellular damage at culture supernatant dilutions up to 1 : 128. Both strains were hemolytic, but neither exhibited the Kanagawa phenomenon. They were both capable of urea hydrolysis, an interesting trait, considering that elasmobranchs retain large (ca 300 milliosmolal) urea concentration in their tissue.  相似文献

Summary S-100 protein-immunoreactive cells were demonstrated by immunocytochemical procedures in the hypophysis and saccus vasculosus of two species of elasmobranchs (Mustelus manazo and Scyliorhinus torazame). In the saccus vasculosus of M. manazo, immunoreactivity was detectable exclusively in the fibrous portions interposed between the epithelial layer and the blood vessels. In the neurohypophysis, tanycytes and astrocytes of the median eminence were immunostained, but only a few labeled cells were found in the neurointermediate lobe. In S. torazame, the neurohypophysis displayed a similar distribution of immunoreactivity, but there were no labeled cells in the saccus vasculosus. In both species, none of the glandular cells of the hypophysis displayed immunoreactivity. Electron-microscopic examination showed that the immunostained cells in the saccus vasculosus correspond to astrocytes.  相似文献

This study aimed to estimate trophic discrimination factors (TDFs) and metabolic turnover rates of nitrogen and carbon stable isotopes in blood and muscle of the smallnose fanskate Sympterygia bonapartii by feeding six adult individuals, maintained in captivity, with a constant diet for 365 days. TDFs were estimated as the difference between δ13C or δ15N values of the food and the tissues of S. bonapartii after they had reached equilibrium with their diet. The duration of the experiment was enough to reach the equilibrium condition in blood for both elements (estimated time to reach 95% of turnover: C t95%blood = 150 days, N t95%blood = 290 days), whilst turnover rates could not be estimated for muscle because of variation among samples. Estimates of Δ13C and Δ15N values in blood and muscle using all individuals were Δ13Cblood = 1·7‰, Δ13Cmuscle = 1·3‰, Δ15Nblood = 2·5‰ and Δ15Nmuscle = 1·5‰, but there was evidence of differences of c.0·4‰ in the Δ13C values between sexes. The present values for TDFs and turnover rates constitute the first evidence for dietary switching in batoids based on long‐term controlled feeding experiments. Overall, the results showed that S. bonapartii has relatively low turnover rates and isotopic measurements would not track seasonal movements adequately. The estimated Δ13C values in S. bonapartii blood and muscle were similar to previous estimations for elasmobranchs and to generally accepted values in bony fishes (Δ13C = 1·5‰). For Δ15N, the results were similar to published reports for blood but smaller than reports for muscle and notably smaller than the typical values used to estimate trophic position (Δ15N c. 3·4‰). Thus, trophic position estimations for elasmobranchs based on typical Δ15N values could lead to underestimates of actual trophic positions. Finally, the evidence of differences in TDFs between sexes reveals a need for more targeted research.  相似文献

A rice (Oryza sativa L.) cDNA clone coding for the cytoplasmic ribosomal protein L5, which associates with 5 S rRNA for ribosome assembly, was cloned and its nucleotide sequence was determined. The primary structure of rice L5, deduced from the nucleotide sequence, contains 294 amino acids and has intriguing features some of which are also conserved in other eucaryotic homologues. These include: four clusters of basic amino acids, one of which may serve as a nucleolar localization signal; three repeated amino acid sequences; the conservation of glycine residues. This protein was identified as the nuclear-encoded cytoplasmic ribosomal protein L5 of rice by sequence similarity to other eucaryotic ribosomal 5 S RNA-binding proteins of rat, chicken, Xenopus laevis, and Saccharomyces cerevisiae. Rice L5 shares 51 to 62% amino acid sequence identity with the homologues. A group of ribosomal proteins from archaebacteria including Methanococcus vanniellii L18 and Halobacterium cutirubrum L13, which are known to be associated with 5 S rRNA, also related to rice L5 and the other eucaryotic counterparts, suggesting an evolutionary relationship in these ribosomal 5 S RNA-binding proteins.  相似文献

The myobiid genus Acanthophthirius Perkins, to date comprising four subgenera, is reviewed and divided into just two subgenera, the nominate subgenus and Myotimyobia Fain. The male genital shield and female opisthogastric sclerites, which are here considered to be part of female genitalia, are adopted as the criteria for dividing the subgenera. These structures are essentially the same in form and position in the subgenus Acanthopthirius in its revised sense, while they are both more heterogeneous in species of the redefined subgenus Myotimyobia. The subgenera Acanthophthirius Fain and Chiromyobia Fain are thought to represent species-groups in the nominate subgenus, named respectively the etheldredae and miniopteri groups. The following one new subspecies and 11 new species are described: A. (A.) womersleyi eptesicus, A. (A.) glauconycteris, A. (A.) mauritaniensis, A. (A.) philetoris, A. (A.) otonycteris, A. (A.) steatocaudatus, A. (A.) nyctophilis, A. (M.) vagus, A. (M.) longus A. (M.) baueris, A. (A.) hesperopteris (female only) and A. (M.) pixonixeos (female only). A. (M.) hanensis is synonymised with A. (M.) namurensis, and A. (M.) capacini is emended to A. (M.) capaccinii and relegated to a subspecies of A. (M.) myotis. All the known and new species are assigned to their respective subgenera and shown in a table. Incongruent host-relationships of some of the mites are clarified, although not completely solved, by the introduction of the new classification for the subgenera.  相似文献
