Five different karyotypes were present in germinating statoblasts obtained from a sampling of a predominately or exclusively asexually reproducing population of Pectinatella gelatinosa from Lake Tatara-numa, Japan. Karyotypes were consistent in having 18 chromosomes, a unique telocentric, a pair of minute metacentrics, and at least one prominently secondarily constricted element. In a number of cases, a sampling of statoblasts from a single large colony yielded more than one karyotype, perhaps because there had been fusion of smaller colonies with different karyotypes. Some individual statoblasts may have been chimeric, but this was not conclusively demonstrated. The possibility of asexually-maintained permanent chimerism in phylactolacmates involving karyotypically distinct ectodermal and peritoneal cell lines is discussed. Karyotypically, P. gelatinosa is more similar to Lophopodella carteri than to Pectinatella magnifica.  相似文献

Silver-stained chromosome spreads from the phylactolaemate ectoprocts Fredericella indica, Fredericella browni, Plumatella repens, Pectinatella magnifica and Asajirella gelatinosa show that each of these species has a single pair of NORs. In the first four species each NOR is immediately adjacent to the centromere of a large metacentric chromosome. Giemsa-stained preparations from Cristatella mucedo show centromeric gapping in a similar large metacentric, indicating the same type of NOR-chromosome. In Asajirella gelatinosa the two NORs are on a pair of smaller chromosomes, and are not immediately adjacent to a centromere. A close NOR-centromere association would inhibit meiotic crossing over in the NOR region. Since in phylactolaemates reproduction is primarily asexual, often by formation of statoblasts, there is potential intraclonal selection for favorable ribosomal DNA (rDNA) mutations, including from simultaneous germination of otherwise genetically identical statoblasts within a limited area and competition between cell lines within statoblasts. Inhibition of meiotic crossing over within the NOR would maintain its integrity during periods when intraclonal selection would not be occurring. This suggests that a more rapid rate of rDNA evolution is possible in phylactolaemates than in most other organisms, and is consistent with a derivation of this group from the morphologically similar phoronids, despite phylogenetic interpretations of rDNA sequence data that suggest the two groups are not closely related.  相似文献

The course of the statoblast formation in Pectinatella gelatinosa was divided into four stages and studied histologically and histochemically. The bottom of the cystigenous cup is a center of cystigenous cell differentiation and the peripheral zone of the inner cystigenous layer turns to the outer cystigenous layer as the cystigenous cup grows. The annulus is formed by migration and transformation of the outer cystigenous cells. During early stages, the yolk cells have an intensely pyroninophilic or RNA-rich cytoplasm. The cytoplasmic pyroninophilia then diminishes as the amount of yolk granules increases. Several kinds of yolk substances occur in the mature statoblast. During statoblast formation glycogen appears first, then glycoprotein and finally neutral unsaturated lipid. Acid phosphatase activity is associated with granular structures in the cytoplasm. In the cystigenous vesicle, acid phosphatase activity is low and confined to the apical extremity of the cell. Histochemically detectable alkaline phosphatase activity is not involved in the formation of the statoblast.  相似文献

The formation of protein-carbohydrate yolk in the statoblast of a fresh-water bryozoan, Pectinatella gelatinosa, was studied by electron microscopy. Two types (I and II) of yolk cells were distinguished. The type I yolk cells are mononucleate and comprise a large majority of the yolk cells. The type II yolk cells are small in number; they become multinucleate by fusion of cells at an early stage of vitellogenesis. In both types of yolk cells, electron-dense granules (dense bodies) are formed in Golgi or condensing vacuoles, which are then called yolk granules. For the formation of yolk granules, the following processes are considered: 1. Yolk protein is synthesized in the rough-surfaced endoplasmic reticulum (RER) of the yolk cells. 2. The synthesized protein condenses in the cisternal space of the RER and is packaged into small oval swellings, which are then released from the RER as small vesicles (Golgi vesicles, 300-600 A in diameter). 3. The small vesicles fuse with one another to form condensing vacuoles, or with pre-existing growing yolk granules. 4. In the matrix of the condensing vacuoles or growing yolk granules, electron-dense fibers are fabricated and then arranged in a paracrystalline pattern to form the dense body. 5. After the dense body reaches its full size, excess membrane is removed and eventually the yolk granules come to mature. Toward the end of vitellogenesis of the yolk cells, the cytoplasmic organelles are ingested by autophagosomes derived from multivesicular bodies and disappear.  相似文献

With the critical point drying method a scanning electron microscopic study of freshwater Ectoproct dormant structures was made. The suture zones of major phylactolaemate genera were described suggesting methods of their classification. The new structures revealed, throw light on the binding mechanism and early kinetics of germination of these dormant bodies. Median ribs and lateral ribs were described in the suture zones of Plumatella fruticosa, P. casmiana and Pectinatella magnifica. Dormant bodies of Fredericella sultana lack a medial or lateral rib in their suture zone. The fine structure of the polymorphic statoblasts of P. casmiana revealed different binding mechanisms as suited to different seasons. During germination, the internal pressure increases within the statoblast and the well cemented medial rib unfolds, letting its two valves separate. The mechanics of valve separations and subsequent germination were related to their respective environmental conditions favouring their domination.  相似文献

Upon germination of a statoblast, the shell is split into two valves; a mucous pad which represents the basal portion of the body wall of the incipient zooid or ancestrula then appears from between the valves; lastly, a tiny polypide evaginates at the opposite site. When two or more contiguously located statoblasts (floatoblasts or sessoblasts) of the same species germinate simultaneously, their mucous pads often come into contact with each other. The walls of the mucous pads then disappear in the contact areas, thus uniting the coeloms of the ancestrulae. This type of fusion between mucous pads of statoblast-derived ancestrulae was ascertained in Plumatella emarginata, P. repens, P. casmiana, and Hyalinella punctata. The fusion is clearly species specific, and shows no clone specificity or allogeneic recognition. The fusibility test reported here seems to be a useful method for the examination of conspecificity in plumatellid bryozoans.  相似文献

1. The hypothesis that nutrient enrichment will affect bryozoan abundance was tested using two complementary investigations; a field‐based method determining bryozoan abundance in 20 rivers of different nutrient concentrations by deploying statoblast (dormant propagule) traps and an experimental laboratory microcosm study measuring bryozoan growth and mortality. These two methods confirmed independently that increased nutrient concentrations in water promote increases in the biomass of freshwater bryozoans. 2. Statoblasts of the genus Plumatella were recorded in all rivers, regardless of nutrient concentrations, demonstrating that freshwater bryozoans are widespread. Concentrations of Plumatella statoblasts were high in rivers with high nutrient concentrations relative to those with low to moderate nutrient concentrations. Regression analyses indicated that phosphorus concentrations, in particular, significantly influenced statoblast concentrations. 3. Concentrations of Lophopus crystallinus statoblasts were also higher in sites characterised by high nutrient concentrations. Logistic regression analysis revealed that the presence of L. crystallinus statoblasts was significantly associated with decreasing altitude and increasing phosphorus concentrations. This apparently rare species was found in nine rivers (out of 20), seven of which were new sites for L. crystallinus. 4. Growth rates of Fredericella sultana in laboratory microcosms increased with increasing nutrient concentration and high mortality rates were associated with low nutrient concentrations. 5. Our results indicate that bryozoans respond to increasing nutrient concentrations by increased growth, resulting in higher biomasses in enriched waters. We also found that an important component of bryozoan diets can derive from food items lacking chlorophyll a. Finally, bryozoans may be used as independent proxies for inferring trophic conditions, a feature that may be especially valuable in reconstructing historical environments by assessing the abundance of statoblasts in sediment cores.  相似文献

The freshwater bryozoan Pectinatella magnifica (Leidy, 1851) is an invasive species in many countries all over the world. Although native to North America, it has been found in many countries of Europe and Asia. Pectinatella magnifica forms the largest colonial masses from all recently known bryozoan species. Culturing this organism in an aquarium has never been achieved for more than few days so far. Colonies from laboratory culture are important for various studies on its biology and life cycle of this species in experimental conditions. Young colonies successfully hatched from germinating statoblasts of P. magnifica in the laboratory and were maintained over eight weeks. Moreover, this was the first time when the compound colonies of this species were carried from its natural habitat to the laboratory, into a special aquarium system, and kept alive for more than three weeks. In both experiments the physicochemical parameters of the water (temperature, concentration of dissolved oxygen, electrolytic conductivity and pH) and changes in weight of compound colonies of P. magnifica in laboratory conditions were checked. The results found in this study are essential for understanding the invasiveness of this species and identifying methods for elimination of its ecological risks because these are closely resembling those of other invasive species.  相似文献

Abstract

Several different species of freshwater Bryozoa, belonging to the genera Plumatella, Rumarcanella and Fredericella, were detected within the Northern Mallee Pipeline (NMP) system in Victoria, Australia, that required definitive identification. These organisms produce asexual buds called statoblasts, with valves composed of sclerotised chitin that bear minute micro-ornamentations of considerable taxonomical significance. Imaging and analysis of these distinctive micro-ornamentations using scanning electron microscopy (SEM) is often employed for species identification. Meticulous preparation of statoblast samples is therefore required that necessitates the removal of adhering debris, dehydration and drying—whilst mitigating specimen damage and distortion. This technical note describes an approach whereby each of these three steps have been individually designed to be as benign as possible, using mild detergent/sonication to remove debris, a gradual and gentle dehydration procedure using ethanol, and critical point drying. For the overall process, these methods are chosen to optimise control and to minimise the use of harsh and hazardous chemicals.  相似文献

Most species of freshwater bryozoans (Ectoprocta: Phylactolaemata) have few morphological distinctions, and within phylactolaemates, the morphology of the statoblast has been used to determine evolutionary relationships. Here, two controversial phylogenies have been proposed for phylactolaemates with regard to the relationship of Lophopodidae to other families. Two plumatellid genera, Gelatinella and Hyalinella , are candidates for the ancestral type of lophopodids. In addition, the coexistence of spines on the surfaces of the statoblast has led to the suggestion that lophopodids are closely related to the family Cristatellidae. In this study, we analysed mitochondrial DNA sequences of the 12S and 16S rDNA genes of 10 phylactolaemate species. Our results suggest that plumatellids may not be a direct ancestral group of lophopodids and that cristatellids are not a sister group of lophopodids. Fredericella , which was previously thought to be an ancestral group, was revealed to be derived. In addition, our results suggest that Stephanella is the most basal phylactolaemate. Mapping morphological characteristics onto the sequence-based phylogenetic tree revealed convergent evolution of statoblast characters.  相似文献

Valves of S. niagarae var. niagarae Ehr. and S. niagarae var. magnifica Fricke from geographically dispersed sediment and plankton collections were observed by light microscopy and scanning electron microscopy (LM and SEM). Measurements made by LM can be arranged so that means and ranges of diameter, areolar density, or strial density intergrade from one population into the next. Mean diameter is negatively related to increasing areolar and strial densities. No unique features observable by SEM distinguish the two described varieties. Thus, S. niagarae var. magnifica, having large diameter valves with low areolar and strial densities, may represent one end of a trend in overall variation in S. niagarae. At the opposite extreme are the populations with small valves and high areolar densities which are often erroneously referred to as S. astraea. Type material of S. niagarae lies intermediate to these forms. Three populations considered in this study have distinct morphological characteristics. Valves from Yellowstone Lake sediments have spine placements distinctly different from valves of all other populations. Specimens from Lake Superior have nearly flat central areas. Ribs of valves from Grand Traverse Bay (Lake Michigan) are covered with granules.  相似文献

Morphological characters of statoblasts (including floatoblasts and sessoblasts) in freshwater bryozoans have been important in phylactolaemate systematics and identification in that older phylogenetic hypotheses relied heavily on statoblast morphology. To assess the reliability of statoblast characters in drawing conclusions about phylogeny, we examined the phylogenetic distribution of metric and proportional floatoblast characters, floatoblast symmetry, and floatoblast and sessoblast microsculpture in Plumatellidae, the largest family of phylactolaemates, in the context of molecular phylogenetic reconstructions based on nucleotide sequences of parts of the mitochondrial 12S rRNA, 16S rRNA, and cytochrome b (cytb) genes. Adding cytb sequences to a previous phylogeny based on 12S and 16S increased support only for a node including Plumatella vaihiriae in the P. repens clade. Characters of gross floatoblast morphology were generally not phylogenetically informative individually, but collectively discriminated among members of three relatively well-supported clades that were analyzed as pre-defined groups in a discriminant analysis. Two characters of floatoblast microsculpture (reticulation and hypertubercles) were restricted to particular clades; other characters (e.g., villi) were clearly convergent. In nine of 11 cases, fenestral microsculpture was identical or partly correlated between the floatoblast and sessoblast of a species. Overall, our results indicate that statoblast morphology is not highly phylogenetically constrained.  相似文献

We explored the suitability of nuclear and mitochondrial ribosomal markers [small subunit nuclear ribosomal RNA gene, large subunit nuclear ribosomal RNA gene, and a region spanning partial small mitochondrial ribosomal RNA subunit, four transfer RNA genes, and partial large mitochondrial ribosomal RNA subunit (referred to as rrnS‐rrnL)] for resolving patterns of diversification of 27 freshwater bryozoan species (class: Phylactolaemata) and evaluated the utility of statoblast ultrastructural features and molecular phylogenies for species discrimination in the Fredericellidae and Plumatellidae. Molecular data identified Plumatella fruticosa as distinct from the rest of the plumatellids, rendering the latter polyphyletic. rrnS‐rrnL was the most suitable marker for species discrimination and identified two undescribed species of Plumatella and at least two undescribed species of Fredericella. Lack of wide dispersal by fredericellid statoblasts may underlie the observed propensity for cryptic speciation and phylogeographical structure in Fredericella. Conversely, the strong dispersal potential of plumatellid statoblasts may mediate efficient gene flow between distant populations and explain the relatively low intraspecific divergence and lack of evidence for cryptic speciation. We show that species identification based on external features of statoblasts can be problematic in both genera, including for a putatively highly invasive, biofouling species, Plumatella vaihiriae, thereby highlighting the utility of rrnS‐rrnL sequences for species barcoding. © 2013 The Linnean Society of London  相似文献

Summary Comparative histochemical studies on the fish (Channa maruleus) and amphibian (Bufo stomaticus) oogenesis demonstrate a great similarity in the growth and differentiation of their egg follicle. The ooplasm, germinal vesicle and egg-membranes show distinct morphological and cytochemical changes during previtellogenesis and vitellogenesis.During previtellogenesis the various components of the follicle are engaged in the synthesis of protoplasm as shown by the proliferation of yolk nucleus substance, mitochondria and some lipid bodies in the ooplasm and of nucleoli in the germinal vesicle. The substance of the yolk nucleus consisting of proteins, lipoproteins and RNA first appears adjacent to the nuclear membrane. Numerous mitochondria of lipoprotein composition, and some lipid bodies consisting of unsaturated phospholipids lie in association with the yolk nucleus which forms substratum for the former. The lipid bodies, present inside the germinal vesicle, follicular epithelium, and adjacent to the plasma membrane in association with some pinocytotic vacuoles, have been considered to play a significant role in the active transport of some substances from the environment into the ooplasm and from the latter into the germinal vesicle. The follicular epithelium itself is very poorly developed, negating its appreciable role in the contribution of specific substances into the oocyte, which seem to be contributed by the germinal vesicle showing a considerable development of nuclear sap, basophilic granules and nucleoli consisting of RNA and proteins; many large nucleoli bodily pass into the cytoplasm during the previtellogenesis of Channa, where their substance is gradually dissolved. The intense, diffuse, basophilic substance of the cytoplasm is believed due to free ribosomes described in many previous ultrastructural studies.During vitellogenesis, the various deutoplasmic inclusions, namely carbohydrate yolk, proteid yolk and fatty yolk, are deposited in the ooplasm. The carbohydrate yolk bodies rich in carbohydrates originate in association with the plasma membrane and correspond to vesicles and cortical granules of previous studies. The proteid yolk consisting of proteins and some lipoproteins, and fatty yolk containing first phospholipids and some triglycerides and then triglycerides only are deposited under the influence of yolk nucleus substance, mitochondria and cytoplasm. The mitochondria and yolk nucleus substance foreshadow in some way the pattern of these two deutoplasmic inclusions and persist at the animal pole of mature egg while the other inclusions of previtellogenesis disappear from view. The pigment granules, which also show a gradient from the animal to vegetal pole in Bufo, are also formed in association with yolk nucleus substance and mitochondria. Some glycogen also appears in both the species. The nuclear membrane becomes irregular due to the formation of lobes. The lipid bodies of the germinal vesicle come to lie outside the nuclear membrane, suggesting active transport of some substances into the ooplasm; many nucleoli bodily pass into the ooplasm of Bufo, where they are gradually absorbed. The amount of basophilic granules is considerably increased in the germinal vesicle during vitellogenesis. Various egg-membranes such as outer epithelium, thin theca, single-layered follicular epithelium, zona pellucida or vitelline membrane surround the vitellogenic oocytes. The zona pellucida formed between the oocyte and follicle cells consists of a carbohydrate-protein complex. The follicle cells show lipid droplets, mitochondria and basophilic substance in their cytoplasm. Aim The transport of organisms in ships’ ballast tanks is a dominant vector for aquatic invasions worldwide. Until recently, efforts to manage this vector have overlooked the potential transport of invertebrate resting stages in the residual waters and sediments within emptied ballast tanks, i.e. NOBOB (‘No Ballast On Board’) tanks. The resting stages (statoblasts) of freshwater bryozoans are often buoyant and locally abundant and thus can be taken up easily during ballasting operations. They are also resistant to extreme environmental conditions and can generate new colonies after being dormant for decades; as such, they would likely remain viable propagules after lengthy transport in ship ballast tanks. This study quantified the occurrence of freshwater bryozoan statoblasts in ballast tank sediments of transoceanic ships. Location North American Great Lakes. Methods We quantified the frequency of occurrence, abundance and diversity of bryozoans (as statoblasts) in residual sediment samples taken from 51 NOBOB tanks of 33 transoceanic ships visiting the Great Lakes from 2000 to 2002. Results Our study identified 11 species, comprising nearly 12% of the total number of freshwater bryozoans known worldwide. These include two exotic species unrecorded in the Great Lakes (Fredericella sultana and Lophopus crystallinus), an exotic species already established in the region (Lophopodella carteri) and three cosmopolitan species (Plumatella casmiana, P. fungosa and P. repens). Our estimates suggest that a ship with NOBOB tanks may carry up to 106 statoblasts. Main conclusions The discovery of species unrecorded in the Great Lakes and the potentially large numbers of statoblasts being transported in ship ballast tanks indicate a significant risk of new species introductions. Furthermore, the presence of cosmopolitan species and an exotic species already established in the Great Lakes suggests the strong possibility of cryptic invasions via the introduction of exotic genotypes.  相似文献

The distributions of most cosmopolitan invertebrate species are assumed to result from natural processes. Cryptic invertebrates with obscure biogeographic origins are often considered native by default, resulting in potentially severe underestimation of the extent of human-assisted invasions. This problem is exemplified by freshwater Bryozoa (Ectoprocta) and Entoprocta—small and widely distributed invertebrates commonly found in lakes and rivers. A benthic survey of a thermally modified section of the St. Lawrence River revealed the presence of two nonindigenous bryozoans: Carter’s moss animal Lophopodella carteri (Hyatt) and the crystal moss animal Lophopus crystallinus Pallas. Also discovered was a cryptogenic entoproct, the goblet worm Urnatella gracilis Leidy. These species were collected as statoblasts and (in the case of U. gracilis) colonial fragments downstream of the Gentilly-2 nuclear power plant at Bécancoeur, Quebec. Local densities of both U. gracilis and L. carteri increased by an order of magnitude at sites closer to the power plant. The occurrence of Lophopus crystallinus statoblasts in St. Lawrence River sediments is the first documented physical evidence of the species in North America. Contrary to the presumed natural Holarctic distribution of L. crystallinus, our literature review found that published historical records of L. crystallinus in the United States are erroneous or unsubstantiated. We propose that L. crystallinus is a western Palearctic species recently introduced to the St. Lawrence River, most likely as statoblasts discharged with ballast water from transoceanic ships.  相似文献

In the period from 2012 to 2014, twenty localities with a varying density level of the invasive bryozoan Pectinatella magnifica were investigated in the T?eboň region. These localities included water bodies ranging from eutrophic-hypertrophic fishponds to mesotrophic-oligotrophic flooded sandpits. The aim of the study was to investigate and compare the water bodies’ physical, hydrochemical and hydrobiological parameters. Control localities (localities with absence of P. magnifica) were found to be significantly different from localities with occurrence of P. magnifica in most of the measured parameters. Also shown was that P. magnifica tends to form colonies in localities showing above-average qualitative parameters within the T?eboň region: balanced oxygen and pH regime, low concentration of suspended solids (Secchi depth over 1 m) and nitrogen forms (mean TN 1.5 mg L?1), chlorophyll-a mean concentration 54 µg L?1, and zooplankton mean density 117 ind L?1 and biomass 2 mg of wet weight L?1. Furthermore, P. magnifica was also found in brown humic waters. While the sites with P. magnifica occurrence are often mesotrophic flooded sandpits and fishponds under nature protection, fishponds for recreational use, and those with low intensity of fishery management (without formation of massive cyanobacterial water blooms, oxygen regime fluctuations, etc.), localities unoccupied by invasive bryozoans are mostly strictly eutrophic-hypertrophic, semi-intensified, carp fishponds.  相似文献

Differentiating oocytes and associated follicle cells of two species of amphineurans (Mollusca) Mopalia muscosa and Chaetopleura apiculata have been studied by techniques of light and electron microscopy. In addition to the regularly occurring organelles, the ooplasm of young oocytes contains large, randomly situated, basophilic regions. These regions are not demonstrable in mature eggs. As oocytes differentiate, lipid, pigment and protein-carbohydrate yolk bodies accumulate within the ooplasm. Concomitant with the appearance of pigment and the protein carbohydrate containing yolk bodies, the saccules of the Golgi complex become filled with a dense material. Associated with the Golgi complex are cisternae of the rough endoplasmic reticulum which are filled with an electron opaque substance which is thought to be composed of protein synthesized by this organelle. That portion of the cisternae of the endoplasmic reticulum facing the Golgi complex shows evaginations. These evaginations are thought to finalize into protein containing vesicles that subsequently fuse with the Golgi complex. Thus, the Golgi complex in these oocytes might serve as a center for packaging and concentrating the protein used in the construction of the protein containing pigment or protein-carbohydrate yolk bodies. The suggestion is made that the Golgi complex may also synthesize the carbohydrate portion of the formentioned yolk bodies. In an adnuclear position in young oocytes are some acid mucopolysaccharide containing vacuolar bodies. In mature eggs, these structures are found within the peripheral ooplasm and we have referred to them as cortical granules. There is no alteration of these cortical granules during sperm activation.  相似文献

Summary Histological and histochemical characteristics were studied inHalobatrachus didactylus (Schneider, 1801) during oogenesis. Three phases could be differentiated: previtellogenesis (oogonia and basophilic oocytes), vitellogenesis (yolk synthesis) and maturation-spawning. Glycogen, glycoproteins and proteins rich in certain amino acids were present in the previtellogenic as well as in the vitellogenic cytoplasm oocytes. No acid mucosubstances were detected. Three types of yolk (vesicles, vacuoles and granules) contained different types of organic reserves; granules were essentially proteic whereas globules were lipidic. The freshwater bryozoan species Pectinatella magnifica was found in 6 sandpits and in 19 mostly extensively managed ponds in the Protected Landscape Area and Biosphere Reserve T?eboňsko (Czech Republic) from its first record (in 2003) to 2012. Mean fresh biomass and abundance of P. magnifica colonies were 0.6 ± 1.5 kg m?2 and 0.7 ± 1.1 colony m?2 (± SD), respectively, in the shoreline zone during the growing season 2006–2011. The maximum biomass was mostly recorded during the first half of August in all basins. Colonization of further localities was recorded rather than increasing of P. magnifica biomass or abundance in 2012. There were no correlations between water temperature or water transparency and biomass/abundance of P. magnifica during the growing season. P. magnifica colonies preferred to grow on the branches or roots (especially of Salix sp.) to aquatic macrophytes and stones. Most of the water bodies, where this bryozoan species occurred, had lower concentration of total phosphorus in the water when compared with the typical fishponds in the Czech Republic. Inner space of colonies of P. magnifica provided suitable higher trophic level substrate when compared with the water of the sandpits/fishponds especially for green coccal algae. A massive algal colonization was indentified in decomposing colonies at the end of the growing season.  相似文献
