The responses of the larvae of the cirripede barnacle Peltogasterella gracilis (Crustacea: Cirripedia: Rhizocephala) that parasitizes the hermit crab Pagurus pectinatus to different combinations of seawater temperature (25, 22, 20, 16, and 12°C) and salinity (from 34 to 8) were studied in a laboratory. The nauplii of P. gracilis completed the entire cycle of development at 22 to 12°C in a narrow range of salinity (from 34 to 28), which agrees well with the environmental conditions of the crab hosts' habitat. At favorable temperatures (22–20°C) and salinity (34–28), the nauplii reached the cypris stage in 88 ± 2 h, while at 12°C and 34–30, the naupliar development took 156 ± 5 h. The cypris larvae appeared more resistant compared with the nauplii, in terms of changes in both the temperature and salinity of seawater. They actively swam at all experimental temperatures and in the salinity range of 34–18. At temperatures (22–16°C) and salinities (34–24) favorable for the cyprids, their longevity in plankton equaled 6–10 days. Thus, the nauplii of P. gracilis is the more vulnerable stage of development in the life cycle of this parasitic barnacle. The tolerance against changes in environmental factors is due to the adaptive capabilities of parasitic larvae and the environmental conditions in the habitats of its host, a typical marine crustacean. The insignificant parasitization rate of the hermit crab by its rhizocephalan parasite may be explained by the death of the nauplii of P. gracilis, which occurs when they enter to the surface water layer.  相似文献

The production of a highly thermostable mannanase by Rhodothermus marinus was increased 16.5-fold by optimising the concentrations of locust bean gum and yeast extract using central composite designs. The optimised medium and culture conditions yielded mannanase activity at 495 nkat ml–1 (248 nkat mg–1 protein). In addition, -L-arabinofuranosidase, -xylanase, -xylosidase, -glucosidase, -mannosidase, -galactosidase, -galactosidase and endoglucanase activities were detected at 32 nkat ml–1, 30 nkat ml–1, 16 nkat ml–1, 15 nkat ml–1, 0.1 nkat ml–1, 1 nkat ml–1, 0.5 nkat ml–1 and 8 nkat ml–1, respectively. No filter paper cellulase activity could be detected. The optimum pH of the mannanase was 5.0–6.5 and it showed high stability from pH 5 to 10 after 16 h incubation at 50 °C. The enzyme activity was maximum at 85 °C, with half lives of 45.3 h at 85 °C and 4.2 h at 90 °C. The endemic grasshopper Cryptobothrus chrysophorus is widely distributed throughout S.E. Australia and its populations display an extensive and spectacular pattern of autosomal variation. While the standard telocentric complement of three long (L1–3), six medium (M4–9) and two short (S10–11) autosome pairs is present throughout most of its range, two quite distinct chromosome races can be defined within this species. Populations in the northern part of its distribution (northern N.S.W. and southern Queensland-northern race) are differentiated from the remainder (southern race) by fixed blocks of distal heterochromatin on autosomes M4, 5, 6, 8 and 9 and by differences in the character of the megameric M7 chromosome. Additionally, while many populations in both races show a polymorphic system of supernumerary segments on the two smallest autosomes (S10–11), that found in the northern race is both more variable and more complex. On the other hand all the populations of the southern race we have examined are polymorphic for a series of centric shifts which convert telocentrics into acro- or meta-centrics. These occur more commonly in the megameric M7 and the two smallest autosomes (S10–11) although in one population (Forbes Creek, N.S.W.) at least 12 different shifts involving 8 of the autosomes (L3, M4, 5, 6, 7, 8, 9 and S10) are known. By contrast, in the northern erace only the small autosomes (S10–11) show centric shifts. These several floating and fixed variants thus involve all chromosomes of the standard set other than the two largest autosomes (L1–2) and the X-chromosome, which appear to be invariate. Finally, morphologically distinct supernumerary (B) chromosomes, intermediate in size between the standard S10 and the M9 elements, are found in both races but are especially common in Tasmania, the most southerly point of the species range. These B-chromosomes are partly heterochromatic and partly euchromatic so that they too add to the considerable heterochromatin variation in this species.  相似文献

Diatom assemblages were analysed in the surface sediments of 44 alkaline lakes in south-western Ontario, Canada, and combined with a pre-existing 58 south-eastern Ontario lake set: (1) to determine if shallow, polymictic Ontario lakes contain different diatom assemblages from deeper, dimictic lakes, and if so, which environmental variables most influence assemblages; (2) to improve the existing transfer functions; (3) to construct and compare transfer functions separately for dimictic, deep lakes and for polymictic, shallow lakes. Polymictic and dimictic lakes covered a similar nutrient range (spring total phosphorus (TP)=4–54 g/l, spring total nitrogen (TN)=200–927 g/l; n=101) and spring pH levels (7.6–9; n=101). However, polymictic lakes were shallower (median mean depth = 2.9 m vs. 7.3 m in dimictic lakes). Benthic diatoms (average 60% relative abundance) dominated the polymictic lakes, whereas planktonic diatoms (average 60%) dominated dimictic lakes. A Canonical Correspondence Analyses with forward selection (p < 0.05, 999 Monte Carlo permutations) identified TP, alkalinity, watershed to volume ratios and lake depth as the most important measured environmental variables influencing diatom distribution in both polymictic and dimictic lakes. Additionally, pH was identified as an important variable in polymictic lakes, whereas TN was also forward selected in the dimictic lakes. Adding more lakes to the original southern Ontario calibration set improved the TN transfer function (r2 jack=0.42, root mean squared error of prediction (RMSEP)jack=0.11 [log g TN/l]), although there was a high systematic error in the revised model (r2 residual = 0.48). However, the strongest TP model was derived from the polymictic lakes (r2 boot =0.44, RMSEPboot=0.20 [log g TP/l]), which was the smallest lake set (n=30) with the lowest number of diatom species. The stronger TP model from the polymictic lakes may be partly due to the relatively low macrophyte cover in our polymictic lakes, which may lead to stronger benthic–pelagic coupling than in lakes with large macrophyte populations. Additionally, our study suggests that the Chrysophyceae cyst:diatom frustule ratio may be useful for indicating trends in TP levels of 35 g/l in alkaline lakes that are dimictic, but is not necessarily indicative of trophic state changes in shallow, polymictic lakes. Our study demonstrates that it may be important to construct separate diatom-based nutrient transfer functions for polymictic and dimictic lakes.  相似文献

Anaerobic digestion of wastewater from a petrochemical plant, manufacturing Nylon-6 was studied in continuously fed up-flow fixed-film column reactors using different biomass support materials such as bonechar, charcoal, bricks, plastic beads and polyurethane foam under varying hydraulic and organic loading rates. Experimental results showed bonechar as the best support material with high biomass-retaining capacity because of its high specific surface area (53.35m2g–1 of bedding material) and pore specific volume (0.244cm3g–1 of bedding material). This system could treat waste water at hydraulic retention times (HRT) as low as 2.5 days with organic loading rates as high as 21.76kg COD m–3 day–1 using acidic feed of pH 2.5 resulting in a 95% COD reduction with biogas production of 11.76m3 m–3 of reactor volume day–1. Total alkalinity of 1700mg CaCO3 l–1 and pH of 7.5 of the treated wastewater were observed at 2.5 days HRT, indicating that methanogenesis appear to be alkalizing step and wastewater with pH as low as 2.5 can be treated as such without neutralization with retention of methanogenic biomass on bonechar.  相似文献

Oocysts of Isospora ernsti n. sp. and Isospora blagburni n. sp. are described from the black-capped bulbul Pycnonotus xanthopygos from Lincoln Park Zoo, Chicago, Illinois. The bird came from southwestern Africa seven years earlier. I. ernsti oocysts are ellipsoidal to bluntly ovoid, 28–38 × 23–31m (mean 34 × 28 m) and have a single-layered oocyst wall. Micropyle, oocyst residuum and polar granules are absent. Sporocysts are elongate ovoid, 24–30 × 11–16 m (mean 27×13 m). Stieda and substiedal bodies and sporocyst residuum are present. I. blagburni oocysts are spherical to subspherical. 21–28 × 19–26 m (mean 25 × 23 m) and have a single oocyst wall. Sporocysts are ovoid and 17–23 × 10–13 m (mean 20 × 12 m). Whilst reliable estimates of chiasma frequencies can usually not be obtained, the probability (b) of a chromosome arm to be bound by at least one chiasma can often be determined. In the absence of interference this probability equals (1–e –2), where 2 is the average chiasma frequency of the chromosome arm and the average crossover frequency or map length. In the presence of interference is shown to retain its genetic meaning as an additive metric that may describe the chromosome arm or other distinctive chromosome segment in terms of genetic recombination. It is a form of potential map length, comparable to, but numerically different from the regular map length. It is termed provisionally crossing-over potential.A chromosome with armsm andn with crossing-over potentials and will form ring bivalents with a frequency (1–e –2).(1–e –2); open bivalents with a frequency (1–e –2).e –2+(1–e –2).e –2; univalent pairs with a frequencye –2.e –2. Estimates of these frequencies yield equations from which and may be solved. Synthetic thioglycoside-analogs of gangliosides such as Neu5Ac)2-S-6)Glc-(1-1)Ceramide (1) and the GM3 analog Neu5Ac(2-S-6)Gal-(1–4)Glc(1-1)Ceramide (2), competitively inhibited GM3 hydrolysis by the sialidase of different subtypes of human and animal influenza viruses with an apparent Ki value of 2.8×10–6 and 1.5×10–5 M, respectively. The inhibitory activity of the ganglioside GM4 analog [Neu5Ac-(2-S-6)Gal-(1-1)Ceramide (3)], in which the glucose of 1 was substituted by galactose, was lower than that of 1 (Ki =1.0×10–4 M). The thioglycoside-analogs (1, 2, 3) of the gangliosides were nonhydrolyzable substrates for influenza virus sialidase. We determined for Vicia faba L the influence of nitrogen uptake and accumulation on the values of photon saturated net photosynthetic rate (P Nmax), quantum yield efficiency (), intercellular CO2 concentration (C i), and carboxylation efficiency (C e). As leaf nitrogen content (NL) increased, the converged onto a maximum asymptotic value of 0.0664±0.0049 mol(CO2) mol(quantum)–1. Also, as NL increased the C i value fell to an asymptotic minimum of 115.80±1.59 mol mol–1, and C e converged onto a maximum asymptotic value of 1.645±0.054 mol(CO2) m–2 s–1 Pa–1 and declined to zero at a NL-intercept equal to 0.596±0.096 g(N) m–2. fell to zero for an NL-intercept of 0.660±0.052 g(N) m–2. As NL increased, the value of P Nmax converged onto a maximum asymptotic value of 33.400±2.563 mol(CO2) m–2 s–1. P N fell to zero for an NL-intercept of 0.710±0.035 g(N) m–2. Under variable daily meteorological conditions the values for NL, specific leaf area (L), root mass fraction (Rf), P Nmax, and remained constant for a given N supply. A monotonic decline in the steady-state value of Rf occurred with increasing N supply. L increased with increasing N supply or with increasing NL.  相似文献

(1–3) glucan and chitin synthases were studied in spontaneously produced protoplasts and in the mycelium (hyphal body) of the entomopathogenic Entomophthorale species Entomophaga aulicae, Conidiobolus obscurus and Entomophthora muscae. The absence of wall in protoplasts was correlated to an absence of chitin synthase and to a very low (1–3) glucan synthase activity, whereas these two polysaccharide synthases were present and active in the walled hyphal bodies. Physicochemical properties of chitin and (1–3) glucan synthases such as localization, optimum pH and temperature, activation by disaccharides and proteases were similar to those found in other fungi unable to spontaneously produce protoplasts and could not be related to the ability for protoplastic Entomophthorale species to produce and proliferate under a protoplast form. The absence or the low chitin and glucan synthase activites in Entomophthorale protoplasts was not due to an absence of proteolytic activation of the enzyme. However, all protoplast fractions contained inhibitory substances of glucan and chitin synthase activities. These inhibitors were stable and specific of the protoplast stage. They were not glucanase nor chitinase. Parasitological examination of European otter originating from Extremadura, Spain revealed the presence of a new isosporan species. Oöcysts of Isospora lutrae n. sp. are spherical to subspherical, 31.2 (27.5–32) × 29.6 (28–31) m and have a smooth wall c. 1 m thick. Sporocysts are ellipsoidal, 18.2 (17–19) × 14.4 (14–16) m and lack Stieda and substieda bodies. A spherical sporocyst residuum is present, consisting of granules scattered among the sporozoites. Sporozoites are spindle-shaped, 12.4 × 2.5 m and have anterior and posterior refractile bodies. Based on its unique morphologic structure and host, I. lutrae is considered to be new.  相似文献

Short-term (up to 5 h) transfers of shade-adapted (100 mol · m–2 · s–1) clonal tissue of the marine macroalga Ulva rotundata Blid. (Chlorophyta) to higher irradiances (1700, 850, and 350 mol · m–2 · s–1) led to photoinhibition of room-temperature chlorophyll fluorescence and O2 evolution. The ratio of variable to maximum (Fv/Fm) and variable (Fv) fluorescence, and quantum yield () declined with increasing irradiance and duration of exposure. This decline could be resolved into two components, consistent with the separation of photoinhibition into energy-dissipative processes (photoprotection) and damage to photosystem II (PSII) by excess excitation. The first component, a rapid decrease in Fv/Fm and in Fv, corresponds to an increase in initial (Fo) fluorescence and is highly sensitive to 1 mM chloramphenicol. This component is rapidly reversible under dim (40 mol · m–2 · s–1) light, but is less reversible with increasing duration of exposure, and may reflect damage to PSII. The second (after 1 h exposure) component, a slower decline in Fv/Fm and Fv with declining Fo, appears to be associated with the photoprotective interconversion of violaxanthin to zeaxanthin and is sensitive to dithiothreitol. Structures of mitochondrial bc 1 complex have been reported based on four different crystalforms by three different groups. In these structures, the extrinsic domain of the Rieske [2Fe–2S]protein, surprisingly, appeared at three different positions: the c 1 position, where the [2Fe–2S]cluster exists in close proximity to the heme c 1; the b position, where the [2Fe–2S] clusterexist in close proximity to the cytochrome b; and the intermediate position where the[2Fe–2S] cluster exists in between c 1 and b positions. The conformational changes betweenthese three positions can be explained by a combination of two rotations; (1) a rotation of theentire extrinsic domain and (2) a relative rotation between the cluster-binding fold and thebase fold within the extrinsic domain. The hydroquinone oxidation and the electron bifurcationmechanism at the QP binding pocket of the bc 1 complex is well explained using theseconformational changes of the Rieske [2Fe–2S] protein.  相似文献

Three new species of Isospora Schneider, 1881 are described from agamid lizards, Isospora cannoni n. sp. in Diporiphora australis from northern Queensland, Australia, I. choochotei n. sp. in Calotes mystaceus from northern Thailand, and I. deserti n. sp. in Agama pallida from Israel. I. cannoni oöcysts are subspherical, 20–25 × 22.5–27.5 m with two ovoid sporocysts, 14–15.5 × 10–11.5 m. I. choochotei oöcysts are spherical to subspherical, 24–32 × 28–32.5 m with two ovoid sporocysts, 11 × 15.5–18 m. I. deserti oöcysts are spherical, 25–28 m in diameter with two ovoid sporocysts, 10–11 × 14–17.5 m. All species had sporocysts with Stieda bodies and underwent endogenous development in the nucleus of the host gut epithelial cells. At completion of merogony and gamogony, the host nucleus was reduced to a thin envelope. The significance of endogenous stage characteristics in Isospora taxonomy is discussed.  相似文献

In vivo studies have shown potent protection by volatile anesthetic agents against cerebral ischemic insults. Volatile agents have also been shown to antagonize glutamatergic neurotransmission at the N-methyl-D-aspartate (NMDA) receptor. This study examined the potential for halothane to reduce neuronal excitotoxic lesions caused by NMDA. Fetal rat cortical cell cultures were allowed to mature 13–16 d. Culture wells (n = 13–16) were treated with 0 mM – 3.96 mM halothane in the presence/absence of 30 M NMDA. Additional cultures were exposed to 30 M NMDA in the presence/absence of 10 M MK-801 or 10 ACEA 1021. Cellular lethality was assessed by measurement of lactate dehydrogenase (LDH) 24 hrs later. A maximal effect of halothane was observed at 0.70 mM (2.1 vol%) wherein a 36% reduction in NMDA-stimulated LDH release occurred relative to untreated controls. Both MK-801 and ACEA 1021 caused complete inhibition of NMDA-stimulated LDH release. These data confirm that halothane has modulatory effects at the NMDA receptor but potency of this drug is less than that of specific antagonists of either glutamate or glycine. These findings suggest that halothane protection in vivo can be partially explained by anti-excitotoxic properties although other mechanisms of action are probably also important.  相似文献

The properties and effects of two plant resistance suppressors (1,3--1,6--glucan and a pentasaccharide of xyloglucan origin) involved in the pathosystem of potato (Solanum tuberosum) and the causal agent of blight (Phytophthora infestans(Mont) de Bary) were compared. The microbial 1,3--1,6--glucan suppressed the defense response over a narrow concentration range (10–2M), whereas the plant pentasaccharide had a broad range of effective concentrations (10–12to 10–6M). In the pathosystem of potato and the causal agent of late blight, the -glucan caused a local and race-specific suppressor effect on the plant host defense response. In contrast, the pentasaccharide caused both local and systemic suppression of potato resistance and the presence of terminal fucosyl residue in the xyloglucan oligosaccharine played a decisive role in its effect. The recognition of both suppressors by potato cell membrane sites is discussed.  相似文献

A Bacillus brevis gene coding for an endo-(1,3–1,4)--glucanase was cloned in Escherichia coli and sequenced. The open reading frame contains a sequence of 759 nucleotides encoding a polypeptide of 252 amino acid residues. The amino acid sequence of the -glucanase gene showed only a 50% similarity to previously published data for Bacillus endo-(1,3–1,4)--glucanases. The optimum temperature and pH for enzyme activity were 65–70°C and 8–10, respectively. When held at 75°C for 1 h, 75% residual activity was measured. The molecular mass was estimated to be about 29 kDa on sodium dodecyl sulphate (SDS)-polyacrylamide gel electrophoresis and the enzyme was found to be resistant to SDS. Correspondence to: T. G. Isozyme patterns of carrot (Daucus carota L.) zygotic embryos between the torpedo stage up to 5-day-old seedlings have been compared with those of the similar stages from the embryogenic cell suspension culture to the late somatic plantlet. Somatic embryos blocked at the torpedo stage by -cyclodextrine have also been analyzed. All these stages have been analyzed with respect to seven different enzyme systems: arylesterase, glucosephosphate isomerase, phosphogluconate dehydrogenase, alcohol dehydrogenase, isocitrate dehydrogenase, aspartate aminotransferase and phosphoglucomutase (EC 2.7.5.1, PGM). The relationships between the different stages of both types of embryogenesis have been visualized using an unrooted tree. Generally, profiles of somatic embryos were different from those of zygotic embryos. Interestingly however, a typical zygotic embryo pattern was found in the cyclodextrine-blocked somatic embryos. Only aspartate aminotransferase patterns revealed a similarity between zygotic and somatic torpedo embryos. Both plantlet types showed close patterns with common isozymes. Moreover, similarities were evident between somatic plantlets and cell suspensions. A few isozymes appeared to be stage specific markers: esterase 10–11 were specific to achenes and early germination, phosphogluconate dehydrogenase 8 was specific to 4–5 day-old seedlings and phosphoglucomutase 1 and 7 and alcohol dehydrogenase 4 were markers for zygotic embryos. No somatic embryogenesis specific isozyme could be found. Semper's larvae were obtained from <300 out of 1800 plankton tows taken in the world's oceans (1964–1993). Zoanthellae (larvae of Sphenopidae) occurred at 217 stations and zoanthinae (larvae of Zoanthidae) at 86, the two larval types showing distributions clearly delimited by a minimum sea temperature (22 °C for zoanthellae, 18 °C for zoanthinae; a statistically significant difference, P<0.001). Length of formalin-fixed zoanthellae was 2–8.6 mm and of zoanthinae 1.5–5.9 mm. Endodermal zooxanthellae were present in 9/24 zoanthinae but in no zoanthellae (of 19). Three larvae contained an endo-commensal/parasitic amphipod. Septa were externally visible in larger zoanthinae and were counted in transverse sections of other larvae, a majority of which (both kinds) had 12 septa, the normal maximum. The pattern was brachycnemic in 40/43 larvae and anomalous (but non-macrocnemic) in three. If macrocnemic genera reproduce by Semper's larvae, they should have been represented in such a large sample. The distribution of adult Epizoanthus was examined: many species are deep sea (recorded down to 5000 m) but shallow-water species are relatively plentiful in, for example, the Adriatic and North Seas. No Semper's larva has ever been recorded from either. Some Parazoanthus species also occur in shallow water, especially associated with western Atlantic reef sponges. If they produce Semper's larvae, these have never been found. It is probable that macrocnemic zoanthids settle from planulae that do not develop into recognizable zoanthellae or zoanthinae.  相似文献

Seasonal changes in alkalinephosphatase activity of bacteria and microalgae werestudied in the dimictic Lake Pavin (Massif Central,France), to test whetherthis activity is primarily algal or bacterial andwhether the APA presents seasonal variations coupledwith abiotic and biotic variables. Samples werecollected at different depths from May 1992 to May1993. The specific phosphatase activities wereanalysed spectrophotometrically with p-NitrophenylPhosphate (p-NPP) as substrate and were related to theprotein concentrations. No correlation was foundbetween alkaline phosphatase activity and solublereactive phosphorus (SRP) concentrations across anannual cycle. The specific activities of the smallclass (0.2–1.2 m) were the highest and thecontribution of this picoplanktonic size class(0.2–1.2 m) increased with depth. In addition, thelinear correlations between alkaline phosphataseactivity and protein concentration seemed to indicatethat most of these enzymes are constitutive. However,it cannot be excluded that the high phosphorusconcentrations repress APA.Finally, the measure of APA does not seem to be avalid quantitative test of the deficiency ofphosphorus for aquatic microorganisms.  相似文献
