Vasopressin (AVP) levels were measured in plasma and cerebrospinal fluid (CSF) of rats during acquisition and retention of a passive avoidance response. Only 5 min after the onset of the retention session a significantly higher level of AVP was found in plasma of animals which displayed a long latency, as compared with the levels of animals which showed a weak passive avoidance response (short latencies), or no passive avoidance behavior at all (controls). Moreover no changes in plasma AVP levels were found in plasma of rats submitted to acquisition or extinction of an active avoidance response. It is suggested that, although an elevated plasma AVP level is associated with strong retention of a passive avoidance response the peripheral circulation as well as the CSF are of minor importance for the transport of this neuropeptide to its site of behavioral action.  相似文献

Male Wistar rats sustaining prefrontal cortex aspiration or sham operation at 6 days or 30 days of age were submitted to the following behavioural tests: open-field, acquisition and retention of two-way active as well as passive avoidance tasks. In the open-field the locomotor activity proved enhanced in all the aspirated animals and this enhancement lasted for 30 days. In the two-day active avoidance task, an acquisition deficit was observed in both aspirated groups; but when retrained one month later, they were able to acquire the avoidance task like sham-operated rats and no difference appeared between the groups aspirated at 6 or at 30 days of age. Concerning the passive avoidance task, no difference could be detected between aspirated and sham-operated animals of both groups except that the rats aspirated at an early age (6 days) seemed to display a better avoidance ability in the retention test. These behavioural alterations (hyperactivity and impairment of the acquisition of the 2-way active avoidance) resulted from the prefrontal cortex aspiration, at whatever age this aspiration was performed (6 days or 30 days). They disappeared after a postoperative recovery period of about one month, as evidenced by this longitudinal study.  相似文献

The effect of a lesion of the dorsal septum on active and passive type of avoidance reactions of adult male Wistar (W) and Long-Evans (LE) rats was studied. The rate of acquisition and extinction of the reaction was studied by 3 different testing methods. The animals were operated on when juvenile (30 days) or adult (90 days). The experiments were always started 50 days after the operation, when the "rage syndrome" was no longer present. In the three different tests we found a single common variable--the strain factor. It was this that determined whether the operation was effective, as well as the direction of deviations and the operation age which led to manifest changes. In W males the lesion did not impair either the acquisition or the extinction of an active avoidance reaction (AAR); the passive avoidance reaction (PAR) was acquired and extinguished more slowly after a lesion in adulthood. The spontaneous passive avoidance reaction (i.e. preference of a small, dark space) was likewise negatively affected by the operation. In intact LE animals the AAR was extinguished more slowly than in intact W males; after a lesion produced in juvenile or adult age extinction was speeded up, so that there were no differences compared either with intact or with septal W animals. Intact LE rats also acquired a PAR more slowly than W rats; a septal lesion led to faster acquisition, irrespective of the age at which the operation was performed, so that the rate for septal LE rats were the same as for intact W animals. The extinction of this reaction took longer after an operation at juvenile age and the rate for these septal LE rats were the same as for those of septal W individuals operated on at 90 days. The spontaneous PAR was qualitatively poorer in intact LE animals than in intact W rats, since only 40% of them preferred the small, dark space. A lesion was followed by improvement, which was especially marked after operation on the 30th day, when all the animals preferred this space, and in a very short time, so that they were equal to intact W males. Correlation of the acquisition and the extinction rate showed that there was imbalance of excitation and inhibition processes in the AAR of the W controls, that they were balanced in the PAR and that the lesion reversed these relationships. Both processes were balanced in the LE control and a septal lesion did not alter the situation.  相似文献

Musculo-articular stiffness of the triceps surae (TS) increases with age in prepubescent children, under both passive and active conditions. This study investigates whether these changes in muscle stiffness influence the amplitude of the reflex response to muscle stretch. TS stiffness and reflex activities were measured in 46 children (7-11 yr old) and in 9 adults. The TS Hoffmann reflex (H reflex) and T reflex (tendon jerk) in response to taping the Achilles tendon were evaluated at rest and normalized to the maximal motor response (Mmax). Sinusoidal perturbations of passive or activated muscles were used to evoke stretch reflexes and to measure passive and active musculoarticular stiffness. The children's Hmax-to-Mmax ratio did not change with age and did not differ from adult values. The T-to-Mmax ratio increased with age but remained significantly lower than in adults. Passive stiffness also increased with age and was correlated with the T-to-Mmax ratio. Similarly, the children's stretch reflex and active musculoarticular stiffness were significantly correlated and increased with age. We conclude that prepubescent children have smaller T reflexes and stretch reflexes than adults, and the lower musculoarticular stiffness is mainly responsible for these smaller reflexes, as indicated by the parallel increases in reflex and stiffness.  相似文献

The acquisition of both active and passive avoidance response, the extinction of the former and the retention of the long-term memory trace of the latter were studied in 30- and 90-day-old male rats of the Wistar strain. The rats were in 3 groups which had had a different history between 15 and 30 days of age: (1) normally weaned rats lived from birth in a cage together with mother and siblings, i.e. under usual laboratory breeding conditions; (2) prematurely weaned rats lived under the same conditions for the first 15 days; after this period, their mother was removed from the cage; (3) community-reared rats had the same history up to 15 days of age; then they began to live in a community (5 connected cages) in contact with both young and adult rats from other cages. Ninety- day-old male rats acquired an active avoidance response at the same rate irrespective of their history in youth but 30-day-old rats were relatively slower if they had been prematurely weaned. Among both normally and prematurely weaned 30-day-old rats, the extinction was slower than in community-reared rats of the same age. Passive avoidance response was acquired by all rats at the same rate irrespective of their history and age. The long- term memory trace was always more stable in adult rats than in young ones.  相似文献

Young rats, 13, 16, and 20 days of age, underwent discrete bilateral electrolytic lesions of the posteroventral hippocampo-subiculo-entorhinal area, and were trained on a cool-draft-stimulus passive avoidance task 20 min later. Significant deficits in passive avoidance learning were observed at all ages studied following either small or more extended damage as compared to performance of sham-lesioned animals. The impairment was dependent upon the size of the lesion. Extended bilateral lesions of the parietal cortex overlying hippocampus induced no deficit. These results confirm that this part of the hippocampal complex plays a role in passive avoidance learning in the rat. They also show that this control of behavior is already established by the second week of life, thus supporting our previous experiments that demonstrate a cholinergic nicotinic involvement of this region in acquisition of passive avoidance as early as the 11th day of age.  相似文献

A study was made of the influence of stress (swinging) applied on the first to the fifth day of life of newborn rats, on acquistion of active unilateral avoidance, passive avoidance of a dark "dangerous" compartment, the relative weight of the brain and absolute increase in the weight of the body at an early age. It was shown that in four-week old rats the stress significantly worsens active avoidance acquisition. In two-month old animals avoidance of a dark "dangerous" compartment, i.e. "passive" avoidance, tested immediately after application of electric current is more pronounced in stressed rats. No difference was found in the relative weight of the brain and the absolute increase in the weight of the body between control and stress groups of the animals.  相似文献

Behavioral and histopathological characteristics were studied in mice treated repeatedly with hypertonic saline. In passive avoidance response using a step-through-type shuttle box, the mice treated with hypertonic saline showed shorter latency than control mice. No changes were observed in active avoidance response using a two-way-type shuttle box, spontaneous motor activity or motor function. Histopathological examination revealed marked and frequent degeneration and loss of neurons in the hippocampus as compared with animals after single treatment. The animals with severe hippocampal lesions showed impairment of passive avoidance response. The present brain lesions resulting from repeated treatment with hypertonic saline in mice are considered to be a possible model for memory disorders caused by hippocampal lesions in humans.  相似文献

Three experiments indicated the effects of an early bilateral stimulation of the lateral hypothalamus on later learning behaviour of male rats. The animals were stimulated at 15 days of age and tested during the sixth week of life. Stimulated rats showed an improvement of performances in acquisition of a food-reinforced operant conditioning, but their performance was impaired in two avoidance tests, an inhibitory avoidance response and a two-way avoidance test. These results cannot be interpreted in terms of handling or early experience. An hypothesis of a modified synaptic competition favouring circuits which assure the regulation of approach behaviours is formulated.  相似文献

The effects of moderate (150 +/- 2 ppm) prenatal carbon monoxide (CO) exposure (maternal HbCO concentrations of 15.6 +/- 1.1%) on learning and memory were assessed in young and aged adult rats using a two-way active avoidance paradigm. In experiment 1, the prenatal CO-exposed rats at 120 days of age acquired a conditioned avoidance response equally well as control animals in a 100-trial session. However, following a 24-hr interval the CO-exposed rats failed to demonstrate significant retention of the task as indicated by the absence of significant improvement in performance over the indicated by the absence of significant improvement in performance over the previous day; control subjects did show significant retention. In experiment 2, in which 120-day-old animals received 50 training trials per day until a criterion of ten consecutive avoidance responses was met, the prenatal CO-exposed subjects again acquired the task as well as control animals. When tested for retention 28 days later, a significant memory impairment was again observed in terms of trials required to reattain the avoidance criterion as well as in total percent avoidance responding. In neither experiment did an analysis of initial or average latency to escape the footshock stimulus reveal any significant alterations. These latter results suggest that the observed performance impairment reflected a memory deficit and not a disruption of sensory, motor, or motivational factors. In experiment 3, prenatal CO-exposed rats approximately 1 year of age (300-360 days of age) showed impairment relative to air-exposed controls in both the original learning and retention of the two-way avoidance response. Again, however, there was no evidence for alterations in performance factors per se. Collectively these data indicate that while young adult rats prenatally exposed to 150 ppm CO demonstrate an associative deficit restricted to memory impairment, aged adults similarly exposed during the prenatal period display a more pronounced deficit similar to that recently reported for animals tested as juveniles. The importance of parametric manipulations in uncovering long-term toxicity is also discussed.  相似文献

The role of the pineal gland and its hormone-melatonin-as to the impact of vasopressin (VP) and/or oxytocin (OT) on the regulation of behavior was studied, the passive avoidance task being chosen as an experimental model. The results showed that VP facilitated the avoidance latency during the first retention trial; after pinealectomy, however, VP was ineffective in this regard. Intraperitoneal application of OT was ineffective in modifying the passive avoidance latency when compared with respective saline-treated animals. Melatonin alone, when injected to shamoperated animals 30 min before behavioral experiment, did not affect the passive avoidance response in SA- or OT-treated rats, but blocked the VP-induced lengthening of the passive avoidance latency in the first retention trial. In pinealectomized and OT-treated animals the passive avoidance latency during the second retention trial was severely diminished by melatonin when compared to respective control. We conclude that: a) VP needs a regulated pineal function for developing short-term effects on the passive avoidance response and b) the effect of OT on the avoidance latency in pinealectomized rats develops after melatonin treatment as a long-term effect.  相似文献

Chlorpromazine injected into the amygdala, septum, or caudate delayed the acquisition of a one-way active avoidance response. Injections into nine other brain areas were inactive. Following a standard dose of chlorpromazine at its ED50 for delaying avoidance acquisition, tissue levels of chlorpromazine from those animals displaying reduced acquisition were significantly higher in the caudate and amygdala than from animals not demonstrating a drug effect.  相似文献

Social ranks were determined in rats with different degrees of activity-passivity in coping style. The dominance status of rats was assessed by their behavior during social interaction and during competition for food and water. Coping styles were determined during initial trials of acquisition of active avoidance in a two-way shuttle box. Animals with passive behavior in a shuttle box had average scores of social ranks. Both dominant and subordinate rats displayed significantly more active behavior than subdominant animals. In additional experiment, the rats were caged as a one male-one female pairs. Two weeks later, the social ranks of males were determined during agonistic interactions. Males were caged in triads, and corticosterone levels were assayed in blood samples taken from the tail veins during the first five days. The corticosterone level was significantly higher in subdominants than in dominants. There was no significant difference between the dominants and subordinates in corticosterone levels. The obtained evidence suggests the passive coping in subdominant rats, whereas the dominant and subordinate animals were more "active" under aversive stimulation.  相似文献

Male Wistar rats were exposed to social deprivation in the period from the 22nd to 70th days of postnatal development. The characteristics of the learning of these rats at the age 110-120 days in the passive avoidance box and during training for active avoidance of electroshock were significant lower than those of control animals. Features of motor activities of the "isolants" in learning and testing, and in the "open field" may be related to their higher anxiety level. Morfometric studies of neurons and glial cells and neocortex thickness showed that significantly lower density of neural elements may be the result of the afferent sensory input limitation produced by the social deprivation.  相似文献

DGAVP facilitates consolidation and retrieval of active and passive avoidance behavior. In this study it was tested whether the long-term behavioral effects of DGAVP are the consequence of an initial increase in behavioral arousal during the learning phase. Animals that were preestimated in an open field test to be low active showed a lower number of conditioned avoidance responses (CAR's) during acquisition and extinction of a shuttle-box task than high active rats. DGAVP was administered 40 min prior to the 1st acquisition session. The immediate effect of DGAVP was a shift in the bell-shaped curve of the relation between arousal and performance (13); an increase in acquisition performance was observed with a low dose of DGAVP (0.1 microgram), while a decrease was found with a high dose of DGAVP (1 microgram). A dose-dependent inhibition of extinction was found in both low and high active animals. These results suggest an immediate effect of DGAVP on the rate of acquisition behavior, which may be a direct consequence of its arousing properties, and a long-term effect on extinction, indicating the formation of memory traces specific for vasopressin-related neuropeptides.  相似文献

Twelve non-implanted crossbred bull calves served as controls and 30 crossbred bull calves (10/treatment) were implanted for 82 days, beginning at 34 days of age, to determine the influence of testosterone propionate (TP), dihydrotestosterone propionate (DHTP) and oestradiol-17 beta (E2) on prepubertal and pubertal pituitary-testicular function and on postpubertal social and sexual behaviour. Compared with control bulls, concentrations of serum luteinizing hormone (LH), follicle-stimulating hormone (FSH) and inhibin concentrations were suppressed (P less than 0.01) in all implanted bulls. Testosterone (T) concentration increased (P less than 0.001) in TP-implanted, but decreased (P less than 0.01) in DHTP and E2 bulls during the implant period. LH response to gonadotrophin-releasing hormone (GnRH) challenge during the implant period (2.5 months of age) was less (P less than 0.01) in TP, E2 and DHTP bulls than in controls. A small but significant T response to GnRH occurred in control bulls at 2.5 months of age. LH and T responses to GnRH challenge at 7 months of age (100 days after implant removal) was similar (P greater than 0.20) in control and implanted bulls. Steroid implants administered prepubertally had no effect (P greater than 0.10) on postpubertal social and sexual behaviours, including number of flehmen responses, abortive mounts, services and competitive order score. Body weight did not differ (P greater than 0.10) between treatment groups, but testis size was reduced (P less than 0.01) during the implant period and up to 10 months of age in treated bulls compared with controls. Testes remained smaller in E2-treated bulls up to the end of the study (23 months of age), but daily sperm production and epididymal weight did not differ (P greater than 0.10) between treatment groups at slaughter. Control bulls reached puberty earlier (P less than 0.01; 270 +/- 11 days of age) than did TP (302 +/- 11 days), DHTP (309 +/- 11 days) or E2 (327 +/- 11 days) bulls. Although puberty was delayed in all implant groups, there was no difference in scrotal circumference at puberty (average 28.4 +/- 0.4 cm) between treatment groups. Our findings indicate that TP, DHTP and E2 implants administered prepubertally result in acute suppression of serum LH, FSH and inhibin during the implant period and in post-implant suppression of testis size and delayed puberty in bulls. The lack of treatment effect on behaviour suggests that steroidal programming of sexual behaviour occurs before 1 month of age in bulls.  相似文献

The hypothesis, partly based on findings in social interactions, that aggressive mice generally adopt an active behavioural strategy (cf. fight-flight) in threatening situations, while non-aggressive ones generally assume a passive strategy (cf. conservation-withdrawal) was tested using a two-way active shock avoidance paradigm. Overall, aggressive mice were found to be better active shock avoiders than non-aggressive animals; a finding that is consistent with our hypothesis. However, within the non-aggressive mice a clear dichotomy in high and low avoidance individuals was found. The high intertrial activity in the superior avoidance groups and the low activity in the poor avoidance group was interpreted as another indication of an active versus passive strategy respectively. Accordingly, it was concluded that not all non-aggressive mice assume a passive strategy, but that some mice adopt an active strategy, like all aggressive males.  相似文献

Alcohol administration in drinking water to mother rats during gestation resulted in a permanent learning deficit of the offspring when behavioural reactions were tested in active avoidance conditioned reflex situation in adult age. A similar deficit of learning capacity was observed in adult rats following alcohol administration for two weeks; the acquisition of active avoidance response was tested later. Administration of arginine-vasopressin and ACTH 4-10 during behavioural test led to a significant improvement of learning ability of the animals pretreated with alcohol. The observations indicate that the ethanol-induced deficit of learning capacity involves peptidergic mechanisms and the behavioural manifestations following chronic alcohol treatment are not irreversible processes.  相似文献

The purpose of this work is to study the participation of growth hormone (GH) and growth hormone releasing hormone (GHRH) in the modulation of long-term memory and the extinction response of a passive avoidance task in rats. However, the effect on memory vary according to the age of the animals due to plasma levels of either hormone being modified during the aging process. Male Wistar rats were divided according to age into two experimental blocks (young rats 3 months old and aged rats 24 months old at the start of the experiment) where each block received the same treatment. Each experimental block was then divided randomly into three groups where two were experimental and the other served as control. The animals were then submitted to a one-trial passive avoidance conditioning and tested for memory retention 24 hrs after as well as twice a week until the extinction response occurred. The control group received an isotonic saline solution and the other two groups received 0.8 U.I. Of GH or 4 mcg of GHRH respectively. All substances were in a 0.08 ml volume and applied 24 hrs before training as well as 24 hrs before each retention session. The results indicate that GH and GHRH modulate longterm memory as well as the extinction response and in either case the response seems to vary with age. GH and GHRH facilitates long-term memory in young rats but not in aged rats. Finally, whereas GH delays the extinction response in both groups, GHRH retards the extinction only in aged rats.  相似文献

Experimentally naive male rats were sequentially tested for an exploratory (open-field) and a one-trial learning passive avoidance behavior. Subsequently, α-MPT-induced disappearance of noradrenaline (NA) and dopamine (DA) was determined in microdissected brain regions. The animals were classified as good or poor avoiders on the basis of their performance in passive avoidance retention test. Trained controls were subjected to the same training except of electric foot-shock during the learning trial. The rate constant of NA disappearance was higher in the hippocampal dentate gyrus of the good vs. poor avoiders. In the good avoiders, the rate constant of DA disappearance was significantly higher in the central nucleus of the amygdala. The different turnover of catecholamines in the dorsal hippocampus and the amygdala in relation to passive avoidance performance suggests that individual differences in memory and/or learning may correlate with the catecholamine turnover of certain limbic structures.  相似文献
