火红熊蜂Bombus pyrosoma是中国特有的熊蜂资源, 在华北地区分布十分丰富, 是众多野生植物和农作物的重要传粉者。为了探究火红熊蜂的遗传结构及其进化关系, 首次开展了该种熊蜂卫星标记的筛选及其种特异性研究。本研究采用生物素-磁珠吸附分离法从火红熊蜂基因组中筛选微卫星标记, 并选取黑熊蜂亚属Melanobombus其他7种熊蜂进行了这些标记的种特异性验证。结果表明: 不同探针与磁珠杂交产生的微卫星标记阳性克隆率存在很大差异, TC探针的微卫星阳性克隆率最高为82%; TG探针次之, 为28%; AT和GC探针分离未获得微卫星标记克隆。根据获得的阳性克隆的序列设计引物进行筛选验证, 共获得31对熊蜂微卫星序列特异性引物。微卫星标记分析发现, 完美型(perfect)18个, 占58.1%; 非完美型(imperfect)10个, 占32.3%; 混合完美型(compound perfect)1个, 占3.2%; 混合非完美型(compound imperfect)2个, 占6.4%。不同探针的微卫星核心序列重复数不同, TC探针和TG探针的微卫星核心序列最高重复数分别为28和15个, 最低重复数分别为7和11个。利用所获得的31对微卫星引物对黑熊蜂亚属其他7种熊蜂进行检测, 有26对引物可以扩增所测试的7种熊蜂, 其他5对引物只能扩增部分熊蜂种类, 其中BPM5是火红熊蜂种特异性引物。本研究从火红熊蜂基因组中筛选的微卫星标记, 不仅可用于火红熊蜂的遗传结构、 分子进化和资源保护等方面的研究, 而且可进一步用于黑熊蜂亚属其他种群的遗传特性分析。  相似文献

种间转移扩增法筛选长爪沙鼠微卫星位点   总被引:1,自引:0,他引:1  
目的筛选长爪沙鼠新的微卫星位点,为长爪沙鼠遗传分析提供遗传标记物。方法从GenBank中随机选取小鼠微卫星位点引物536对,用这些引物对长爪沙鼠基因组DNA扩增,将阳性目的条带进行序列分析,找出符合微卫星序列特征的短串联重复序列。结果 536对小鼠微卫星引物在长爪沙鼠基因组中扩增出了313个阳性条带,经序列分析,确定130个长爪沙鼠微卫星位点;其中完美型位点占80.77%(105/130),不完美型位点占19.23%(25/130),与小鼠同源性为24.3%(130/536)。将筛选出的微卫星位点在GenBank中注册,注册号从GU562694到GU562823。结论小鼠和沙鼠的微卫星位点具有较高的同源性,用小鼠的微卫星位点引物直接扩增长爪沙鼠基因组DNA可有效地筛选出长爪沙鼠微卫星位点。  相似文献

采用磁珠富集法对已建立的生长较快的西江段野生浅色鲮群体的极大、极小群体各31尾基因组DNA进行了微卫星序列的筛选,128个阳性克隆成功测序93个,其中80个为的微卫星序列,微卫星序列完美型占85%,非完美型占6.25%,混合型占8.75%。利用微卫星序列合成了23对微卫星引物,扩增结果表明,等位基因数为1~10个,扩增片段大小为92~283bp,其中14对引物扩增条带清晰,为中度或高度多态位点,极大、极小群体的平均有效等位基因数和平均多态信息含量分别为3.0784、3.2079和0.5675、0.5974,反映出2个群体遗传多样性均较丰富;高度多态性引物4的位点b,片段大小为119bp,在极大群体中出现频率为61.3%,在极小群体占22.6%,出现频率极大群体高于极小群体近3倍,初步可作为候选差异性分子标记。  相似文献

采用生物素-磁珠吸附微卫星与质粒二次检测相结合的方法克隆牙鲆微卫星,并利用所得到的引物分析野生群体遗传结构.共获得2805个阳性克隆,测序得到3120个含微卫星的序列.其中完美型、非完美型和混合型分别占57.97%、7.25%和34.78%.用Primer 3.0软件设计牙鲆微卫星引物,从中筛选出具稳定多态性的30对,分析中国黄海、渤海海域4个不同海区牙鲆野生群体(大连、北戴河、丹东、青岛)遗传结构.结果显示:有效等位基因数为3.93~9.94,平均为6.95;观测杂合度为0.532~0.895,平均为0.753;期望杂合度为0.635~0.902,平均为0.820;Hardy-Weinberg平衡指数(d)的变化范围为-0.247~0.512,其中7个位点表现为杂合子过剩(d>0),其余位点表现为杂合子缺失(d>0).聚类分析显示:4个群体聚为两类,大连与丹东聚为一类,北戴河与青岛聚为一类.  相似文献

牙鲆CA/GT微卫星标记的筛选   总被引:5,自引:0,他引:5  
采用生物素选择杂交法与放射性同位素杂交法相结合的技术,成功地从牙鲆(Paralichthys olivaceus)基因组中分离出含有CA/GT重复类型的微卫星序列。通过两轮淘选,共获得526个阳性菌落。测序其中的119个菌落,结果获得133个含有微卫星座位的序列。除了两个复合型微卫星外(1.5%),完美型63个(47.37%),非完美型68个(51.13%)。设计并合成22对微卫星引物,对8个人工雌核发育家系的亲本进行遗传背景分析。PCR结果表明,4对引物无扩增带或者扩增带不是目的条带,1对引物表现为单态,其余17对引物均呈多态性,平均每个座位产生5.2个复等位基因,杂合度为0.375~0.846,多态信息含量为0.305~0.823。结果表明,所筛选的大部分微卫星标记能够用于牙鲆群体遗传学研究。  相似文献

长江江豚微卫星DNA分离的初步研究   总被引:1,自引:0,他引:1  
为了开发物种特异性微卫星标记,本文采用一种改良的快速微卫星分离法(FIASCO)从长江江豚(Neophocaena phocaenoides asiaeorientalis)的基因组中筛选得到72条微卫星DNA序列。根据重复单元的排列特点,完美型、非完美型及复合型序列所占的比例分别为58.3%、22.2%和19.5%。选择其中30条序列设计PCR扩增引物,并用12个随机选择的长江江豚样品进行多态性筛选。初步结果表明其中14对引物的扩增产物稳定并且具有多态性;在每个座位上获得2-13个等位基因,平均等位基因数为5.87个;14个微卫星座位的平均观察杂合度(Ho)和期望杂合度(He)分别为0.560和0.709。本研究获得了长江江豚的第一批物种特异性微卫星座位及其扩增引物,这些微卫星标记将在后续的保护遗传学研究中发挥重要作用。    相似文献

为了获得适于三叶木通遗传多样性和遗传结构研究的微卫星分子标记,该研究采用磁珠富集法构建了三叶木通微卫星富集文库。结果表明:在150个阳性克隆中发现了70个微卫星位点,富集效率为46.67%,其中含双碱基重复单元的序列占比为79.37%,三碱基和四碱基重复含有量较少。共设计引物63对,其中筛选出16对高多态引物,对1个三叶木通自然居群48个个体进行了遗传分析,结果显示位点的等位基因数为10~22个,观察杂合度和期望杂合度分别为0.370~0.792和0.724~0.936,多态性信息指数为0.725~0.919,表明以上引物均为高多态性引物。其中,12个位点偏离哈迪-温伯格平衡,呈现出纯合子过剩状态,这可能与哑等位基因和其它因素有关。综上结果表明,该研究所开发的16对引物能够用于三叶木通遗传多样性和遗传结构评价工作。  相似文献

【目的】本研究旨在分离黑头酸臭蚁Tapinoma melanocephalum基因组微卫星标记,确定这些微卫星位点的多态性。【方法】使用454 GS-FLX焦磷酸测序技术开发来自中国华南陆地和岛屿的11个黑头酸臭蚁地理种群基因组微卫星位点。从随机设计的100对微卫星引物中筛选出10对引物,用于确定黑头酸臭蚁4个地理种群[东澳岛(DAD)、荷包岛(HBD)、梅州(MZ)和山咀(SJ)]10个微卫星位点的多态性,分析种群遗传多样性和种群分化。【结果】从11个黑头酸臭蚁地理种群基因组中成功开发和分离10对微卫星引物。在DAD, HBD, MZ和SJ 4个地理种群中,10个微卫星位点中7个有高多态性,这10个位点均显著偏离Hardy-Weinberg平衡;每个位点的等位基因数量(A)是3.50~9.00个,每个地理种群每个位点等位基因丰富度(AR)在1.992~12.938之间。岛屿地理种群(DAD和HBD)的AR和预期杂合度(HE)与大陆地理种群(MZ和SJ)的相比差异不显著。4个地理种群均显示高水平遗传分化(FST=0.15969);HBD和MZ种群与其他配对地理种群相比,遗传分化较高(FST=0.185),基因流较低,说明这两个种群基因流被限制。此外,遗传变异来自种群内个体之间。【结论】筛选新的微卫星位点能够为研究黑头酸臭蚁种群结构和繁殖结构提供有效工具,以深入了解其传播机制。  相似文献

结合已公布的大熊猫Ailuropoda melanoleuca基因组和本实验室所测6只大熊猫的转录组数据,筛选多态性微卫星位点并分析其组成及特征。结果显示:共获得326个多态性微卫星位点,其中二碱基多态性微卫星最多,共228个,占69.93%;三、四、五、六碱基所占比例分别为9.51%、14.11%、5.21%、1.22%。根据分析结果中缺失率与标准差2项指标以及位点序列长度,选取20个多态性二碱基微卫星位点,用于25只大熊猫个体血液DNA进行PCR验证并做后续分析。结果表明:不同位点的等位基因数为2~8,平均等位基因数为3.70,观测杂合度、期望杂合度分别为0~1.000、0.280~0.784,平均值分别为0.472和0.532。在Bonferroni校正后,证实4个位点显著偏离哈迪-温伯格平衡,所有位点未观察到显著连锁不平衡(P>0.01)。20个位点多态信息含量(PIC)在0.246~0.734,其中具有高度多态性的位点9个(PIC>0.50),11个位点呈中度多态性(0.25相似文献

蜈蚣草(Pteris vittata L.)是目前用于砷污染土壤修复最好的超富集植物,但其分子水平上的研究数据较少。为了开发蜈蚣草特异性SSR遗传标记,本文采用改良的FIASCO方法从蜈蚣草AG和AC微卫星富集文库中随机挑选100个克隆,分离得到51个微卫星位点,其中60%为完美型(Perfect)SSR。根据这些位点设计、合成了25对引物,并对江西庐山及湖北恩施两地蜈蚣草种群各20个个体进行了遗传多样性检测,结果发现:其中8个完美型及1个间断型(intermittent)SSR位点的引物能够扩增出清晰、稳定且具有多态性的条带。9对引物共扩增出41个等位基因,各位点等位基因数在2~7之间,平均等位基因数为4.56个;期望杂合度在0.0494~0.8169之间;没有连锁不平衡现象发生。采用大叶井栏边草(Pteris multifida Poir.)进行跨种扩增,结果发现其中6对引物能够进行种间扩增。这些SSR分子标记的开发有助于蜈蚣草生态适应性进化分析、揭示蜈蚣草地理分布格局以及探讨蜈蚣草遗传多样性,还可用于品种鉴定及选育等。  相似文献

Markers for eight new microsatellite DNA or simple sequence repeat (SSR) loci were developed and characterized in trembling aspen (Populus tremuloides) from a partial genomic library. Informativeness of these microsatellite DNA markers was examined by determining polymorphisms in 38 P. tremuloides individuals. Inheritance of selected markers was tested in progenies of controlled crosses. Six characterized SSR loci were of dinucleotide repeats (two perfect and four imperfect), and one each of trinucleotide and tetranucleotide repeats. The monomorphic SSR locus (PTR15) was of a compound imperfect dinucleotide repeat. The primers of one highly polymorphic SSR locus (PTR7) amplified two loci, and alleles could not be assigned to a specific locus. At the other six polymorphic loci, 25 alleles were detected in 38 P. tremuloides individuals; the number of alleles ranged from 2 to 7, with an average of 4.2 alleles per locus, and the observed heterozygosity ranged from 0.05 to 0.61, with an average of 0.36 per locus. The two perfect dinucleotide and one trinucleotide microsatellite DNA loci were the most informative. Microsatellite DNA variants of four SSR loci characterized previously followed a single-locus Mendelian inheritance pattern, whereas those of PTR7 from the present study showed a two-locus Mendelian inheritance pattern in controlled crosses. The microsatellite DNA markers developed and reported here could be used for assisting various genetic, breeding, biotechnology, genome mapping, conservation, and sustainable forest management programs in poplars.  相似文献

An enriched microsatellite library of the mangrove species Avicennia marina was constructed, in which 85.8% of the clones contained microsatellite sequences. Of the microsatellite repeat sequences isolated, 55.0% were di-nucleotides, 34.2% were tri-nucleotides, 50.0% were perfect, 24.2% were imperfect, and 15.0% were compound. Four different di-nucleotide repeats were isolated with repeat lengths ranging from 5 to 33; ten different tri-nucleotide repeats were isolated with repeat lengths ranging from 3 to 25. The most common di-nucleotide was the AC/TG repeat; the most common tri-nucleotide was the CCG/GGC repeat. Sixteen microsatellite sequences were selected for primer design, and 6 primers were selected to investigate the polymorphism detected among 15 individuals of A. marina from three natural populations in Australia. A total of 40 alleles were detected at 6 microsatellite loci. The number of alleles per microsatellite locus ranged from 5 to 13. On average, 7 alleles were detected per locus. All microsatellite loci showed high levels of gene diversity (heterozygosity), with values ranging from 0.53 to 0.88; the mean value of gene diversity was 0.70. Microsatellite loci were also tested for conservation across Avicennia species. There was a decline in amplification success with increasing divergence between Avicennia species. The results indicate that microsatellites are abundant in the Avicennia genome and can be valuable genetic markers for assessing the effects of deforestation and forest fragmentation in mangrove communities, which is an important issue for mangrove conservation and afforestation schemes. This paper reports the development of microsatellite primers for Nelumbo nucifera Gaerten. By screening genomic libraries enriched with 10 kinds of probes, Seventeen polymorphic loci were isolated and primers were designed. Polymorphism of these 17 loci was assessed in 24 individuals. All the 17 loci are polymorphic and the number of alleles ranged from two to seven. Observed heterozygosity and expected heterozygosity ranged from 0.0000 to 0.9176 and from 0.2837 to 0.7917 respectively. These microsatellite loci should be useful for studying the genetic diversity of N. nucifera.  相似文献

? Premise of the study: Microsatellite primers were developed for Paeonia delavayi and P. ludlowii (Paeoniaceae) to study their population genetics and phytogeography. ? Methods and Results: Nine polymorphic nuclear microsatellite loci were isolated from an enriched library of P. delavayi and primers were designed. The number of alleles per locus ranged from two to 16; the observed and expected heterozygosities ranged from 0.014 to 0.687 and 0.042 to 0.875, respectively. Six polymorphic chloroplast microsatellite loci were identified in P. delavayi and primers were provided. The number of alleles per locus ranged from two to six and the polymorphic information content ranged from 0.08 to 0.716. Both nuclear and chloroplast primers were successfully applicable to P. ludlowii. ? Conclusions: The markers developed here will facilitate analyses of genetic diversity, population genetic structure, phytogeographical patterns, and conservation for P. delavayi and P. ludlowii.  相似文献

The water rat (Hydromys chrysogaster) is well adapted to a semiaquatic life and is endemic to dispersed regions of Australia and New Guinea. To analyse the genetic diversity of water rat populations, polymorphic microsatellite markers were developed. A partial genomic library was screened for microsatellite sequences. Following isolation of the microsatellite sequences, primers were designed to amplify seven loci and of these loci, five were polymorphic. The sample tested for polymorphisms came from areas across Australia and New Guinea. Between three and 13 alleles were detected for each locus. In addition the primers amplified two loci in Mus musculus and Rattus rattus.  相似文献

利用磁珠富集法, 以生物素标记的(CA)15为探针, 构建了蛇鳄龟(Chelydra serpentina L.)微卫星富集文库。通过PCR法从富集文库中共筛选出70条微卫星序列, 一共设计了48对微卫星引物, 采用PCR扩增的方法从中筛选出36对引物, 对一个蛇鳄龟养殖群体进行遗传多样性分析。通过分析, 36个位点获得的等位基因数从29不等, 平均为4.361, 有效等位基因为1.4617.767, 平均为3.498。等位基因片段大小为56342 bp, 观测杂合度为0.0671.000, 平均为0.725; 期望杂合度为0.3160.850, 平均0.600; 多态信息含量为0.26550.8359, 平均为0.5573; 结果表明此蛇鳄龟养殖群体存在较高的遗传多样性水平。群体内固定系数-0.6880.856, 平均为-0.214, 说明蛇鳄龟群体中杂合子过剩。    相似文献

血水草(Eomecon chionantha Hance)是中国亚热带地区特有的单种属植物。本研究采用(AG)15、(AC)15两种生物素探针及磁珠富集法从血水草基因组中开发并筛选出13个微卫星位点,且在该物种中能进行稳定的PCR扩增并表现出多态性。利用筛选出的13个SSR多态性标记对血水草4个野生居群24个个体进行PCR扩增,结果显示每个微卫星位点的等位基因数为2~7个,观测杂合度(HO)和期望杂合度(HE)分别为0.125~0.792和0.299~0.691。说明这些SSR标记可用于血水草遗传多样性的后续研究,并揭示该物种地理分布格局形成的原因,有助于保护和利用中国亚热带地区的血水草野生资源。  相似文献

? Premise of the study: A new set of microsatellite primers was developed for Avena sativa and characterized to assess the level of genetic diversity among cultivars and wild genotypes. ? Methods and Results: Using an enrichment genomic library, 14 simple sequence repeat markers were identified. The loci of these markers were characterized and found to be polymorphic in size among 48 genotypes of oat from diverse geographical locations. The number of alleles per locus ranged from two to eight, while the observed heterozygosity ranged from 0.031 to 0.75. ? Conclusions: These newly identified microsatellite markers will facilitate genetic diversity studies, fingerprinting, and genetic mapping of oat. Moreover, these new primers for A. sativa will aid future studies of polyploidy and hybridization in other species in this genus.  相似文献

The objectives of the present study were to develop microsatellite markers for the wild strawberry, Fragaria virginiana, to evaluate segregation patterns of microsatellite alleles in this octoploid species, and assess genetic variability at microsatellite loci in a wild population. A genomic library was screened for microsatellite repeats and several PCR primers were designed and tested. We also tested the use of heterologous primers and found that F. virginiana primers amplified products in cultivated strawberry, Fragaria × ananassa Duch. and Fragaria chiloensis. Similarly, microsatellite loci developed from cultivated strawberry also successfully amplified F. virginiana loci. We investigated four microsatellite loci in detail, three developed from F. virginiana and one from cultivated strawberry. A survey of 100 individuals from a population of F. virginiana in Pennsylvania demonstrated high heterozygosities (He or gene diversity ranged from 0.80 to 0.88 per locus) and allelic diversity (12–17 alleles per locus), but individual plants had no more than two alleles per locus. Segregation patterns in parents and progeny of two controlled crosses at these four loci were consistent with disomic Mendelian inheritance. Together these findings suggest that the genome of F. virginiana is "highly diploidized" and at least a subset of microsatellite loci can be treated as codominant, diploid markers. Significant heterozygote deficiencies were found at three of the four loci for hermaphroditic individuals but for only one locus among females in this gynodioecious species.Communicated by J.
