Sulfated polysaccharides (SP) isolated from freshwater green algae, Spirogyra neglecta (Hassall) Kützing, and fractionated SPs were examined to investigate their molecular characteristics and immunomodulatory activity. The crude and fractionated SPs (F1, F2, and F3) consisted mostly of carbohydrates (68.5–85.3%), uronic acids (3.2–4.9%), and sulfates (2.2–12.2%) with various amounts of proteins (2.6–17.1%). d-galactose (23.5–27.3%), d-glucose (11.5–24.8%), l-fucose (19.0–26.7%), and l-rhamnose (16.4–18.3%) were the major monosaccharide units of these SPs with different levels of l-arabinose (3.0–9.4%), d-xylose (4.6–9.8%), and d-mannose (0.4–2.3%). The SPs contained two sub-fractions with molecular weights (Mw) ranging from 164 × 103 to 1460 × 103 g/mol. The crude and fractionated SPs strongly stimulated murine macrophages, producing considerable amounts of nitric oxide and various cytokines via up-regulation of their mRNA expression by activation of nuclear factor-kappa B and mitogen-activated protein kinases pathways. The main backbone of the most immunoenhancing SP was (1→3)-l-Fucopyranoside, (1→4,6)-d-Glucopyranoside, and (1→4)-d-Galactopyranoside.  相似文献

Thymine-requiring mutants of Micrococcus radiodurans have been isolated by selection on solid medium containing trimethoprim. Strains requiring either high concentrations of thymine (50 μg/ml) or low concentrations (2 μg/ml) for normal growth were obtained. The Thy mutant requiring low thymine concentrations has been characterized. It was shown to retain the high ultraviolet light (UV) resistance typical of wild-type M. radiodurans, but it was not resistant to thymineless death. Preliminary exposure of the cells to thymineless conditions resulted in enhanced UV sensitivity, and this interaction occurred under conditions where “unbalanced growth” was inhibited by the addition of chloramphenicol. Upon addition of thymine to deprived cells, UV resistance was gradually restored, and this recovery took place in the absence of protein synthesis. A model is proposed to account for the similarity of thymineless death in bacteria whose deoxyribonucleic acid repair efficiencies differ widely.  相似文献

Six uvr mutants of Saccharomyces cerevisiae with hypersensitivity to ultraviolet (UV) light were isolated after mutagen treatment with ethylmethanesulfonate. UV sensitivity ranges from moderate to extreme, and four of the mutants are also sensitive to nitrous acid. Ranking in terms of UV sensitivity does not parallel ranking in terms of nitrous acid sensitivity. Homozygous diploid mutant strains are somewhat less sensitive than the corresponding haploids. All mutations are recessive. None of the mutants is sensitive to gamma rays, and each shows photoreactivation after UV radiation. Complementation tests and tetrad analysis indicate that each strain represents mutation in a different gene. Two of the uvr genes are linked, and two others are centromere-linked.  相似文献

An ecological study of Micrococcus radiodurans indicated that microorganisms possessing the same morphological and radiation-resistance characteristics as that organism could be isolated from ground beef and from pork sausage. Further studies showed that such organisms also could be isolated from beef hides and from water from a creek adjacent to the packing plant from which the meat samples were obtained. Similar microorganisms were not isolated, however, from a limited number of samples of soil, hay, and fecal material. The use of high levels of gamma-radiation in the initial isolation procedures proved to be advantageous in inactivating most of the other microflora and facilitating the isolation of M. radiodurans. Control experiments indicated that M. radiodurans did not compete well with the microflora present in ground meat, soil, and beef hides. Preincubation before irradiation of meat and soil samples or enrichment culture techniques did not enhance the isolation of M. radiodurans. The presence of M. radiodurans in creek water suggested one possible source of this organism.  相似文献

The modern conceptions on the molecular mechanisms of Micrococcus radiodurans viability under the action of ionized radiation have been considered. Factors providing a high level of the bacterium radioresistance-the peculiarities of the cell wall structure, membranes, DNA, the redundancy of genetic information, the multiplicity of sites of DNA attachment to the membrane, a high level of antioxidant and antiradical systems-have been analysed. It has been shown that the efficiency of accurate, error-free, well balanced DNA repair system in connection with M. radiodurans properties mentioned provides a high radiation resistance of this microorganism.  相似文献

A wild-type strain of Micrococcus radiodurans and its nonpigmented mutant W(1) were tested for sensitivity to 10 antibiotics selected from the standpoint of their mechanism of action. Representatives of groups of antibiotics inhibiting deoxyribonucleic acid (DNA) synthesis, DNA-dependent ribonucleic acid synthesis, protein synthesis, and cell wall synthesis were selected. M. radiodurans and its mutant exhibited full susceptibility to all antibiotics tested (mitomycin C, actinomycin D, chloramphenicol, dihydrostreptomycin, erythromycin, neomycin, kanamycin, benzylpenicillin, bacitracin, and vancomycin), the degree of susceptibility being of the same order as that of a standard strain of Staphylococcus aureus 209 P, with the exception of dihydrostreptomycin.  相似文献

Strand scissions in DNA of M, radiodurans after in vivo irradiation with 60Co gamma rays were investigated by the sedimentation analysis using neutral sucrose gradients. Double-strand scission in DNA was estimated to occur at the rate of one double cut per 800 eV. This rate is in a good agreement of the value reported for mammalian cells. The rejoining of these double-strand scissions was observed during the repair process of the post-irradiation incubation and the mean rejoining time, i.e., the time reducing the remaining fraction of the double-strand scission to 0.37, was found to be 52 min. This rejoining repair was inhibited by adding chloramphenicol, tetracycline or actinomycin D to the postirradiation incubation medium. It is suggested that the high resistance character of M. radiodurans to gamma rays may be due to the efficient capacity of this rejoining repair.  相似文献

The deoxyribonucleic acid (DNA) synthesized following ultraviolet (UV) irradiation of wild-type (Rd) and recombination-defective strains of Haemophilus influenzae has been analyzed by alkaline sucrose gradient sedimentation. Strain Rd and a UV-resistant, recombination-defective strain Rd(DB117) (rec-) are able to carry out postreplication repair, i.e., close the single-strand gaps in the newly synthesized DNA; in the UV-sensitive, recombination-defective strain DB117, the gaps remain open. The lack of postreplication repair in this strain may be the result of degradation of the newly synthesized DNA.  相似文献

The DNA of Micrococcus radiodurans was prepared by three methods. Although the recovery of DNA varied considerably, the percentage molar base ratios of the DNA from the three preparations were essentially the same: guanine, 33±2; adenine, 18±1; cytosine, 33±2; thymine, 17±1. Base compositions calculated from Tm values and from density in caesium chloride gradients also yielded guanine+cytosine contents of 66 and 68% of total bases respectively. No unusual bases were observed. The S20,w values were characteristic of high-molecular-weight DNA. Electron microscopy showed the purified DNA in long strands; occasionally these were coiled.  相似文献

The survival of biological activity in irradiated transforming deoxyribonucleic acid (DNA) has been assayed in the wild type and a radiation-sensitive mutant of Micrococcus radiodurans. The frequency of transformation with unirradiated DNA was lower in the mutant to about the same extent as the mutant's increased sensitivity to radiation. However, in both the wild type and the mutant, the irradiated DNA that was incorporated into the bacterial genome was repaired to the same extent as determined by the loss of transforming activity with increasing radiation dose. This applied to DNA irradiated either with ionizing or ultraviolet (UV) radiation. The rate of inactivation of biological activity after UV radiation was the same in any of the DNA preparations tested. For ionizing radiation, the rate of inactivation varied up to 40-fold, depending on the DNA preparation used, but for any one preparation was the same whether assayed in the wild type or the radiation-sensitive mutant. When recipient bacteria were irradiated with ionizing or UV radiation immediately before transformation, the frequency of transformation with unirradiated DNA fell, rapidly and exponentially in the case of the sensitive mutant but in a more complicated fashion in the wild type. The repair of DNA irradiated with ionizing radiation was approximately the same whether assayed in unirradiated or irradiated hosts. Thus, irradiation of the host reduced the integration of DNA but not its repair.  相似文献

The conversion of prochaetoglobosins as plausible precursors into mycotoxin chaetoglobosin A (1) in a cell-free system of Chaetomium subaffine was unsuccessful. However, reductase activity of the 20-keto-analogues (1), and prochaetoglobosins II (5) and III (6) were found in a microsomal fraction of this fungi. Two new metabolites of chaetoglobosins, named chaetoglobosin Fex (2) and 20-dihydro-chaetoglobosin A (3), were also isolated from the same micro-organisms. Their structures were elucidated by spectroscopic data and chemical transformation.  相似文献

Nitrosoguanidine (NG) mutagenesis of Staphylococcus aureus resulted in the isolation of eight mutants exhibiting 3 to 28 times greater sensitivity to ultraviolet (UV) radiation. These mutants were further characterized by their ability to repair UV-irradiated bacteriophage, to act as recipients in the transduction of antibiotic resistance, and their sensitivity to NG. Based on the available data, six of these mutants are reduced in their ability to perform host-cell reactivation. One of the remaining two mutants may be deficient in post-replication repair.  相似文献

The nature of the patterns of elimimation of chromosomal aberrations in both root and shoot has been studied in both the species of P. canariensis Linn (2n = 12) and P. minor Retz (2n = 28) after irradiating their dry seeds with filtered and unfiltered X-rays.Doses used are 10 kR and 30 kR filtered X-rays and 10 kR and 20 kR unfiltered X-rays.The elimination curves in both root and shoot have been fitted to the equation N1 = N0e?KT for both the species. The pattern of elimination for each type of aberrations was found to be exponential in both root and shoot.  相似文献
