Frozen stored sperm samples of two Holstein bulls (A and B) were compared for their abilities to interact with zona-free hamster ova. The percentage of hamster vitelli interacting with sperm from Bull A was significantly higher than that interacting with sperm from Bull B (94.5% vs. 68.2%, P<0.01), and these results were in good agreement with 60 day non-return data for the same months (68.2% for Bull A vs. 64.3% for Bull B). Sperm from Bull A also excelled in average numbers attached to vitelli, and in average numbers of sperm penetrated into the zona-free hamster ova. However, of the penetrated vitelli, sperm of Bull B resulted in more pronuclei. In these experiments the percentage of progressively motile sperm at insemination was highly correlated with the percentage of vitelli interacting with sperm. The percentages and numbers of sperm cells with intact acrosomes were significantly correlated with the average number of sperm attached per vitellus. These observations encourage further development of this test for assessing sperm fertilizing ability.  相似文献

Progress has been made toward developing conditions to enable bull sperm to penetrate and fertilize ova in vitro. Efforts toward understanding oocyte maturation also hold promise. A procedure for bovine in vitro fertilization (IVF) involving recovery of oocytes near the time of ovulation, fertilization with in vitro capacitated sperm, embryo culture and transfer has led to development of normal offspring. Additional research is needed to facilitate recovery of mature oocytes and to improve in vitro conditions to insure development of viable embryos that can continue normal development following non-surgical uterine transfer.Bovine IVF promises a means to overcome certain types of infertility, to produce large numbers of half-siblings simultaneously, to greatly extend valuable semen, a better way to assess functional performance of male and female gametes, to provide synchronously developing pronuclear stage ova for nuclear transfer and/or gene injection, and many possibilities in research and for future applications.  相似文献

In an effort to determine empirically the proper age for separation of pigtailed macaque (Macaca nemestrina ) infants from their dams in a large domestic breeding colony, we conducted a retrospective survey of 1,592 infants. Survivorship was highest for infants not separated from their dams at all and for those separated within the first four months of life. Survivorship was poor for animals separated during or after the fifth month. Thus, not weaning or weaning early are the most acceptable management strategies in terms of mortality risk. This result is paradoxical with regard to nutritional considerations, but appears to be related to a sensitive period in social development. The relative advantages and disadvantages of various weaning ages are discussed.  相似文献

This work represents efforts towards development of the zona-free hamster ovum sperm penetration assay for predicting relative levels of fertility for semen from individual bulls. Results reported here followed insemination of hamster vitelli with bull sperm, after frozen storage, with observations of sperm acrosomes and parallel inseminations of more than 1000 cows with semen from each bull. The average 75-day non-return rate for the four bulls was 74.0% (range 71.6 to 75.6). Laboratory studies indicated the following: the percentage of sperm with intact acrosomes varied from 55 to 73 between bulls, the percentage of motile sperm varied from 41 to 64, the percentage of sperm with progressive motility ranged from 24 to 40, the number of sperm interacting per (zona-free hamster) ovum ranged from 1.6 to 3.8, the number of sperm attached per ovum ranged from 1.4 to 2.9, the number of sperm within each penetrated ovum ranged from 1.5 to 1.8, the percentage of ova interacting with sperm ranged from 76 to 92, the percentage of ova penetrated ranged from 62 to 85, and the percentage of ova with male pronuclei ranged from 33 to 49. Although predictive ranking in the laboratory of these bulls with less than 4% variation in fertility levels was not possible, the zone-free hamster ovum test could be useful in identifying potentially subfertile bulls before they enter a young sire-sampling program.  相似文献

The tissue interaction between the notochord and the somites of the vertebrate embryo establishes the proper shape and constitution of the vertebral cartilage. Soon after somite formation, the somite differentiates into a cartilage-forming part, the sclerotome, and a muscle and skin-forming part, the dermamyotome. These components of the somite were dissected from 312-day-old chick embryos (stage 1812–19 and cultured in vitro in the presence or absence of notochord. It was found that the sclerotome cells respond to the notochord by an increased incidence of hyaline cartilage nodules, greater accumulation of sulfated glycosaminoglycans, synthesis of larger aggregates of proteoglycans, increased DNA accumulation, and accelerated DNA synthesis. The dermamyotome did not show these changes. These results indicate that the notochord enhances cartilage differentiation in the sclerotome. Under these conditions, the notochord did not elicit cartilage formation in the dermamyotome.  相似文献

Previous experiments have shown that when young human diploid cells (HDC) were fused to senescent HDC, neither nucleus synthesized DNA. This paper demonstrates that when young HDC are in S phase at the time of fusion to senescent HDC, they do make DNA in heterodikaryons; therefore, ongoing DNA synthesis is not inhibited by senescent cells. On the other hand, entry into S phase is inhibited: young HDC nuclei in G1 phase do not make DNA in heterodikaryons with senescent HDC.  相似文献

The spleen cells from CFW/D mice injected with dimethylbenzanthracene-induced leukemia virus exhibited a progressive decline in the in vitro response to heterologous erythrocyte antigens in parallel with tumor growth. Cell transfer experiments revealed that this immunodepressed state may involve a B-cell defect rather than extrinsic factors in the cellular environment since: (i) nonresponsiveness could be transferred to irradiated non-tumor-bearing mice with spleen cells, and (ii) T cells from tumorbearing mice cooperated with normal bone marrow cells, but bone marrow from tumorbearing mice did not cooperate with normal T cells. In addition, T cells from the thymic tumor could cooperate with normal bone marrow cells upon transfer to irradiated recipients. TL 485-2 cells, a T-cell line derived from the tumor, could be specifically activated with SRBC thereby indicating that the virus transformed T cells were immunocompetent. Suppressor cells, which appeared in the spleen concomitant with immunodepression and tumor development, may directly raise B-cell thresholds for T-dependent triggering signals since the antibody response of spleen cells from tumor-bearing mice could be restored by adding agents such as LPS, 2 mercaptoethanol, or T cells exogenously preactivated in normal animals. The suppressor cell could be enriched by adherence to plastic and was removed by treatment with carbonyl iron. In addition, it was unlikely that the suppressor cell was a virus-infected cell since transformed, virus-infected cells from the tumor or TL 485-2 cells were not suppressive when added to spleen cells in vitro but rather resulted in a marked, polyclonal enhancement of the PFC response. The interaction of TL 485-2 cells and normal spleen cells resulted in the release of a stimulatory factor which increased DNA synthesis in resting cells as well as increasing PFC. The role of these enhancing factors and suppressor cells in controlling tumor growth remains to be elucidated.  相似文献

Elevation of plasma enteroglucagon has been described in the presence of intestinal hyperplasia in both animals and man. In order to investigate whether enteroglucagon plays a role in the stimulation of small intestinal growth, fasting plasma and small intestinal tissue levels of enteroglucagon immunoreactivity were measured in control rats and in 3 different rat models of intestinal adaptation: 5 weeks after proximal and distal small bowel resection, on the 12th day of lactation and following 5 weeks of cold acclimation induced hyperphagia. Plasma enteroglucagon levels increased significantly from the control value of 89±(SEM) 13.1 fmol/ml to 147±13.6 after proximal resection (p < 0.005) and to 207±32.6 following 5 weeks of hyperphagia (p < 0.001). During lactation ileal tissue enteroglucagon increased by 183% from 58.1±8.6 pmol/g tissue in controls to 163.9±20.4 (p < 0.001). Ileal tissue enteroglucagon levels were also elevated by 60% following proximal resection (p < 0.005) and by 91% following the hyperphagia of cold-acclimation (p < 0.005). In contrast, jejunal tissue enteroglucagon levels of cold-acclimated rats were only increased by 55% when compared to the control value of 27±2.9 pmol/g tissue (p < 0.02). These results confirm that in the rat, ileal enteroglucagon levels are significantly greater than those found in the jejunum (p < 0.005). Enteroglucagon levels were also significantly elevated in the 2 groups of rats with hyperphagia, namely the lactating and cold-acclimated groups. The data lend further support to the proposal that enteroglucagon may play a trophic role in producing intestinal growth.  相似文献

Each of 24 pasture-reared crossbred beef heifers were herded from a large pasture on day 14 of the estrous cycle and assigned randomly to one of four treatment groups as follows: Field Control (FC), Field ACTH (FA), Pen Control (PC) and Pen ACTH (PA). Field groups were maintained in a small field, and pen groups were confined in a pen in a pole barn. ACTH groups received 200 IU of ACTH IM daily for days 17 through 21 of the cycle and control groups received only the gelatin carrier IM on the same cycle days. Average cycle lengths for FA and PA heifers were 25 and 24.3 days with an average period from plasma progesterone decline below 1 ng/ml to estrus of 5.5 days. During the ACTH injection period, follicular growth was suppressed and the proestrus plasma estrogen rise was delayed. Average cycle lengths for FC and PC heifers were 20.8 and 22.8 days respectively. All control group heifers exhibited estrus within 2 days of the plasma progesterone decline below 1 ng/ml. In addition, pen confinement heifers showed a trend for extended luteal function and consequent extended estrous cycle length.  相似文献

The plasma aldosterone radioimmunoassay developed by Ito et al. was found to be non-specific for aldosterone following administration of the spirolactones, spironolactone and canrenoate-K, in rabbits, dogs and humans. The assay interfering principle was identified as a hydroxylated derivative (MB) of canrenone, which itself is a metabolite common to both spironolactone and canrenoate-K. The metabolite MB possessed a high cross-reactivity to the 21-hemisuccinate aldosterone antibody relative to other spirolactones. A modified procedure was developed specific for plasma aldosterone in the presence of MB. Following single doses of spironolactone and canrenoate-K, aldosterone plasma levels were unchanged in humans and in dogs and decreased in rabbits.  相似文献

The involvement of serotonin in mediating the inhibitory effect of immobilization stress on LH secretion in castrated male rats was examined by employing p-chlorophenylalanine (PCPA, 320 mg/kg, ip), an inhibitor of serotonin synthesis, and 5,6-dihydroxytryptamine (5,6-DHT, 50 micrograms, icv), a drug toxic to the indoleaminergic system. Immobilization stress suppressed pulsatile LH release and decreased mean plasma LH levels. Pretreatment with PCPA or 5,6-DHT apparently eliminated the inhibitory effect of immobilization stress on LH release. These results suggest the possible involvement of a serotoninergic mechanism in mediating the suppression of LH release induced by immobilization stress in castrated male rats.  相似文献

This experiment was designed to determine the age at which estradiol-17beta (E(2)) first induces a preovulatory-like surge of luteinizing hormone (LH) in prepubertal heifers. Responses of prepubertal animals 3 to 4 and 5 to 6 months of age were compared with those of postpubertal heifers that received 25 mg prostaglandin F(2)alpha at 0800 hr on day 15 of the estrous cycle. E(2) (500mug) induced surges of LH in 1 5 heifers 3 to 4 months of age, 3 3 heifers 5 to 6 months of age and 5 5 postpubertal heifers. Duration of response and interval between E(2) injection and peak of the response were longer in postpubertal heifers than in those 5 to 6 months old (P<0.10). Peak response and total amount of LH released were greater in animals 5 to 6 months old (P<0.10). Only one prepubertal heifer had elevated concentrations of progesterone following an LH surge. Four of 5 postpubertal heifers receiving E(2) and 3 of 4 postpubertal heifers receiving corn oil had corpora lutea and similar patterns of progesterone concentrations. We conclude that ability to release an LH surge in response to E(2) develops in heifers between 3 and 5 months of age, but that this induced surge does not cause ovulation.  相似文献

A commercially available follicle stimulating hormone preparation (FSH-P) was successfully incorporated into multilamellar vesicles (liposomes). Multilamellar liposomes were found to contain 9.39 +/- 1.14 mg FSH-P (n=4) per 100 mg phospholipid or approximately 19.0% of the original material used to form the liposomes. A 1% solution of Triton X-100 incubated with liposomes containing FSH-P for one-half hour at 37 degrees C released 33% of the entrapped FSH-P; more than 99% of the entrapped FSH-P was released when liposomes were incubated with a 2% solution of Triton X-100. Entrapment of FSH-P increased proportionally to the mole percentage of stearylamine used in liposome formation, suggesting that FSH-P is entrapped in the aqueous interstices of the cationic liposomes. Entrapment of FSH-P in stable liposomes suggests that these multilamellar vesicles may be useful as a FSH-P delivery vehicle used for the superovulation and embryo transfer of food animals.  相似文献

We have examined the effect of adenosine and EHNA, a competitive inhibitor of adenosine deaminase (ADA), upon the ability of human peripheral blood lymphocytes to respond to mitogen. Addition of adenosine at concentrations greater than 10 μm (10?5m) resulted in inhibition of lymphocyte proliferation at 48 hr of culture, provided that the culture medium was relatively free of ADA activity. The actual concentrations of adenosine remaining in inhibited cultures at the time of harvest were considerably lower than those added initially. EHNA alone also inhibited PHA response (and to a lesser extent PWM and Con A responses), but only at high concentrations. Noninhibitory concentrations of EHNA and adenosine together acted synergistically to produce profound inhibition of lymphocyte proliferation. This may provide an in vitro model to explore further the mechanism of the immunodeficiency associated with deficiency of ADA. Adenosine deaminase activity in stimulated cultures did not differ significantly from that found in unstimulated cultures, and the activity per protein or per DNA actually decreased in stimulated versus unstimulated cultures.  相似文献

Treatment with 12-O-tetradecanoylphorbol-13-acetate (TPA) reversibly suppressed myotube formation and expression of acetylcholine receptors in cultures of Day 15 mouse embryo presumptive myoblasts, but was totally ineffective in cultures of adult mouse satellite cells. A subpopulation of TPA-resistant myogenic cells became apparent in cultures prepared from older embryos or newborn mice. Thus, limb presumptive myoblasts are a heterogeneous population, and part of the distinct TPA-resistant subpopulation may represent satellite cell precursors.  相似文献

Serum-free cultures of activated macrophages generate conditioned media containing both potentiating and inhibitory activities for the lectin-induced transformation of syngeneic thymocytes, lymph node cells, or spleen cells, also cultured in serum-free medium. Exhaustive dialysis of macrophage conditioned medium (MCM) eliminates its inhibitory activity. At mitogenic doses of lectin, the dialyzable material enhances the potentiating activity exerted by macro-molecular factors at low and optimal concentration of MCM. The inhibitory effect of intermediate concentrations of nondialyzed MCM on [3H]thymidine uptake can be reversed if the cells are washed and pulsed in fresh medium, and thus is artefactual. On the other hand, high doses have a real inhibitory effect on proliferative response of transformed lymphocytes. Rat MCM is not mitogenic for any of the target lymphocytes tested. Its effect is observed both in primary cultures of lymphocytes and secondary cultures of blast cells.  相似文献

ACTH immunoreactivity (ACTH-IR) in the plasma and cerebrospinal fluid (CSF) collected simultaneously from rhesus monkeys was found to undergo significant diurnal variations. In plasma, the mean peak ACTH-IR was 15.4 +/- 1.95 pg/ml at 0500 h, and the mean minimum concentration was 9.05 +/- 1.80 pg/ml at 1800 h. In CSF, the mean peak ACTH-IR concentration occurred at 1900 h and was 4.64 +/- 0.41 pg/ml. The mean minimum CSF ACTH-IR concentration was 2.93 +/- 0.26 pg/ml, occurring at 0500 h. This is the first report of a diurnal variation in CSF ACTH-IR concentration and is consistent with other work suggesting that plasma ACTH and CSF ACTH originate from different sources.  相似文献
