RNA silencing controls endogenous gene expression and drives defensive reactions against invasive nucleic acids like viruses. In plants, it has been demonstrated that RNA silencing can be transmitted through grafting between scions and silenced rootstocks to attenuate virus and viroid accumulation in the scions. This has been obtained mostly using transgenic plants, which may be a drawback in current agriculture. In the present study, we examined the dynamics of infection of a resistance-breaking strain of Tomato spotted wilt virus (RB-TSWV) through the graft between an old Apulian (southern Italy) tomato variety, denoted Sl-Ma, used as a rootstock and commercial tomato varieties used as scions. In tests with non-grafted plants, Sl-Ma showed resistance to the RB-TSWV infection as viral RNA accumulated at low levels and plants recovered from disease symptoms by 21 days post inoculation. The resistance trait was transmitted to the otherwise highly susceptible tomato genotypes grafted onto Sl-Ma. The results from the analysis of small RNAs hallmark genes involved in RNA silencing and virus-induced gene silencing suggest that RNA silencing is involved in the resistance showed by Sl-Ma against RB-TSWV and in scions grafted on this rootstock. The results from self-grafted susceptible tomato varieties suggest also that RNA silencing is enhanced by the graft itself. We can foresee interesting practical implications of the approach described in this paper.  相似文献

Small interfering RNAs (siRNAs) are silencing signals in plants. Virus‐resistant transgenic rootstocks developed through siRNA‐mediated gene silencing may enhance virus resistance of nontransgenic scions via siRNAs transported from the transgenic rootstocks. However, convincing evidence of rootstock‐to‐scion movement of siRNAs of exogenous genes in woody plants is still lacking. To determine whether exogenous siRNAs can be transferred, nontransgenic sweet cherry (scions) was grafted on transgenic cherry rootstocks (TRs), which was transformed with an RNA interference (RNAi) vector expressing short hairpin RNAs of the genomic RNA3 of Prunus necrotic ringspot virus (PNRSV‐hpRNA). Small RNA sequencing was conducted using bud tissues of TRs and those of grafted (rootstock/scion) trees, locating at about 1.2 m above the graft unions. Comparison of the siRNA profiles revealed that the PNRSV‐hpRNA was efficient in producing siRNAs and eliminating PNRSV in the TRs. Furthermore, our study confirmed, for the first time, the long‐distance (1.2 m) transfer of PNRSV‐hpRNA‐derived siRNAs from the transgenic rootstock to the nontransgenic scion in woody plants. Inoculation of nontransgenic scions with PNRSV revealed that the transferred siRNAs enhanced PNRSV resistance of the scions grafted on the TRs. Collectively, these findings provide the foundation for ‘using transgenic rootstocks to produce products of nontransgenic scions in fruit trees'.  相似文献

We examined the transmission of RNA silencing signal in non-transgenic tomato and tobacco scions grafted onto the tobacco Sd1 rootstocks, which is silenced in both NtTOM1 and NtTOM3 required for tobamovirus multiplication. When the non-transgenic tomato scions were grafted onto the Sd1 rootstocks, RT-PCR analysis of the scions showed the reduced level of mRNA compared with that before grafting in both LeTH3 and LeTH1, tomato homologs of NtTOM1 and NtTOM3, respectively. siRNAs from both genes were detected in the scions after grafting but not before grafting. Further tomato scions were inoculated with Tomato mosaic virus (ToMV) and used for virus infection. They showed very low level of virus accumulation. Necrotic responding tobacco to tobamovirus was grafted onto the rootstock of Sdl. RT-PCR analysis showed low level expression of both NtTOM1 and NtTOM3 in the scions but siRNA was detected after grafting. When the leaves of scions were inoculated with ToMV or Tobacco mosaic virus, they produced very few local necrotic lesions (LNLs) while the control scions did many LNLs. These results suggest that RNA silencing was transmitted to non-transgenic tomato and tobacco scions after grafting onto the Sd1 rootstocks and that virus resistance was induced in the scions.  相似文献

Environmental stresses that perturb plant water relations influence abscisic acid (ABA) concentrations, but it is unclear whether long‐distance ABA transport contributes to changes in local ABA levels. To determine the physiological relevance of ABA transport, we made reciprocal‐ and self‐grafts of ABA‐deficient flacca mutant and wild‐type (WT) tomato plants, in which low phosphorus (P) conditions decreased ABA concentrations while salinity increased ABA concentrations. Whereas foliar ABA concentrations in the WT scions were rootstock independent under conditions, salinity resulted in long‐distance transport of ABA: flacca scions had approximately twice as much ABA when grafted on WT rootstocks compared to flacca rootstocks. Root ABA concentrations were scion dependent: both WT and flacca rootstocks had less ABA with the flacca mutant scion than with the WT scion under conditions. In WT scions, whereas rootstock genotype had limited effects on stomatal conductance under conditions, a flacca rootstock decreased leaf area of stressed plants, presumably due to attenuated root‐to‐shoot ABA transport. In flacca scions, a WT rootstock decreased stomatal conductance but increased leaf area of stressed plants, likely due to enhanced root‐to‐shoot ABA transport. Thus, long‐distance ABA transport can affect responses in distal tissues by changing local ABA concentrations.  相似文献

Development, histological process and Grapevine leafroll‐associated virus‐3 localisation were studied in micrografts of three scion/rootstock combinations: healthy/healthy, healthy/infected and infected/healthy. Earlier bud break and faster growth in scions of micrografts were obtained when the healthy shoot segments were used as scions, while earlier bud break in rootstocks and greater fresh weight of roots in micrografts were produced when the healthy shoot segments were used as rootstocks. All histological processes including callus initiation and formation in micrografting conjunctions, and initiation of new cambial cells followed by vascular bundle development connecting scions and rootstocks were similar in micrografts, regardless of the sanitary status of the scions and rootstocks used for micrografting. Virus infection in micrografting conjunctions and systematic infection in micrografts were much more efficient and faster in micrografting combination of the infected scions/healthy rootstocks than in the healthy scions/infected rootstocks. To the best of our knowledge, this is the first report addressing histological process of micrograft development and virus localisation in micrografts. In vitro culture system established in this study facilitates studies on the ‘pure’ impact of the viral infection on micrografting.  相似文献

以体细胞杂种红桔+枳、红桔+粗柠檬和有性杂种Troyer枳橙、Swingle枳柚以及枳作砧木的"国庆4号"温州蜜柑、脐橙(无枳砧)和不嫁接的砧木幼树为试材,通过两年盆栽试验研究接穗对砧木生长、根系活力和根系抗氧化酶活性的影响。结果表明,Swingle枳柚和红桔+粗柠檬的砧粗易受接穗影响,接穗显著地影响砧木根系体积。在一般情况下,同一种砧木嫁接后砧木根系活力、超氧化物歧化酶(SOD)和过氧化物酶(POD)活性低于未嫁接砧木,或与其差异不显著;同时存在接穗促进根系过氧化氢酶(CAT)活性升高的现象。  相似文献

The analysis of the anatomical characteristics of tree rings of the Pinus sibirica scions and Pinus sylvestris rootstocks is presented. The main anatomical features maintain the seasonal dynamics characteristic of the reference (ungrafted) trees of scion and rootstock, pointing to the stability of the genetic control of the xylem differentiation. However, various anatomical characteristics are reduced for rootstocks and increased for scions relative to the reference trees. A mechanism consistent with our results is that the graft union zone creates a barrier for the ascending and descending transport of substances, including phytohormones. The reaction of the scion and rootstock to climatic factors is weakened relative to the reaction of the reference trees. Thus, the presumed shift towards phytohormone disbalance reduces the influence of external factors on xylem differentiation. The interannual variation of the individual characteristics of the scions and rootstocks is increased relative to the reference trees, reflecting either the influence of the graft partners on each other or the non-uniform distribution of the growth regulators across the graft union. Thus, heterografts can be used to evaluate the interaction of the internal and external mechanisms of xylem differentiation.  相似文献

茄子栽培种砧用种质农业生物学性状及嫁接适用性研究   总被引:1,自引:0,他引:1  
青枯病是热带亚热带地区普遍发生的一种土传病害,为选配具有优良抗病性和嫁接适用性的茄子砧木杂交组合,该研究对国内外11份茄子栽培种砧用种质进行农业生物学评价和青枯病抗病性鉴定,研究了不同砧用种质对嫁接苗生长的影响。结果表明:供试砧用种质中,果型为卵圆形5份、倒卵形2份、长卵圆4份;幼果颜色紫色7份、淡紫色1份、绿色1份、淡绿色2份;老熟果实颜色均为黄色;种质J14、J15、J16、J17的主茎紫色,其余均为绿色;J61的花瓣白色,其余均为紫色或淡紫色。供试砧木种质与接穗Rf的嫁接成活率均在80%以上,最高达97%。采用苗期伤根-浸根接种法鉴定砧木种质对青枯病的抗病性,有9份砧用种质自根苗表现高抗(HR),病情指数为3.67~12.33,其中6份砧木种质(BC01、BC03、BC04、BC06、J15、J16)嫁接感病接穗后表现抗病(R),病情指数为16.67~21.00。砧用种质能显著改善接穗的生长,其中,BC01、BC03、BC04、BC06、J15、J16的嫁接苗的生物量增长率显著高于其他种质的嫁接苗。综合试验结果显示茄子栽培种砧用种质农业生物学性状表现多样性,可区分为不同生态类型。抗病砧木种质嫁接高感病接穗的防病效果显著,种质BC01、BC03、BC04、BC06、J15、J16高抗青枯病,嫁接高感病品种表现抗青枯病,可作为配制抗青枯病砧木杂交组合的候选亲本。  相似文献

The objective of this study was to determine the roles of shoot and root in the regulation of premature leaf senescence induced by potassium (K) deficiency in cotton (Gossypium hirsutum L.). Two contrasting cultivars (CCRI41, more sensitive to K deficiency; and SCRC22, a less sensitive cultivar) were selected for self- and reciprocal-grafting, using standard grafting (one scion/one rootstock), Y grafting (two scions/one rootstock) and inverted Y grafting (one scion/two rootstocks) at the seedling stage. Standard grafting was studied in the field in 2007 and 2008. There were no obvious differences in senescence between CCRI41 and SCRC22 scions while supplied with sufficient K. However, SCRC22 scions showed significantly greater K content, SPAD values (chlorophyll content), soluble protein content and net photosynthetic rates than CCRI41 scions while grown in K deficient solution or soil, regardless of rootstock cultivars, grafting types, growth stage and growth conditions. Also, SCRC22 scions had greater yield and less variation in boll weight either between upper- and lower sympodials, or between proximal and distal fruit positions from the main stem in the field under K deficiency, probably owing to reduced leaf senescence. Although the effect of rootstocks on leaf senescence under K deficiency was significant in some cases, the scion cultivars explained the highest percentage of variations within grafting treatments. The shoot-to-root feedback signal(s), rather than high shoot demand for K nutrition, was involved in the shoot regulation of premature senescence in cotton plants, achieved possibly by altering root K uptake.  相似文献

Fruits of citrus cultivars contain bioflavonoids and some other important secondary metabolites in pharmaceutical and nutritional industries. The present experiment was designed to investigate the correlation between the content of flavonoid components like naringin, hesperidin, and neohesperidin of the scions fruits and the same parameters in rootstocks fruits. Six-year-old trees including four citrus cultivars of ‘Moro’, ‘Mars’, ‘Salustiana’, and ‘Italian’ which were previously grafted on the four different rootstocks including ‘Yuzu’, ‘Shelmahalleh’, ‘Citromelo’, and ‘Sour orange’ were selected as experimental trees. The content of the mentioned flavonoids was investigated in the peel and pulp of the fruits of both scions and rootstocks. The results showed that the measured parameters were significantly influenced by scions, rootstocks, and tissues. Based on the obtained results, it can be suggested that the accumulation of chemicals in citrus fruit depends on genetic and inherent abilities of the scion, more than what was previously believed, while the rootstock can also play an important role in the accumulation of these substances.  相似文献

Genetically engineered (GE) rootstocks may offer some advantages for biotechnology applications especially in woody perennial crops such as grape or walnut. Transgrafting combines horticultural grafting practices with modern GE methods for crop improvement. Here, a non-GE conventional scion (upper stem portion) is grafted onto a transgenic GE rootstock. Thus, the scion does not contain the genetic modification present in the rootstock genome. We examined transgene presence in walnut and tomato GE rootstocks and non-GE fruit-bearing scions. Mobilization of transgene DNA, protein, and mRNA across the graft was not detected. Though transgenic siRNA mobilization was not observed in grafted tomatoes or walnut scions, transgenic siRNA signal was detected in walnut kernels. Prospective benefits from transgrafted plants include minimized risk of GE pollen flow (Lev-Yadun and Sederoff, 2001), possible use of more than one scion per approved GE rootstock which could help curb the estimated US$136 million (CropLife International, 2011) cost to bring a GE crop to international markets, as well as potential for improved consumer and market acceptance since the consumable product is not itself GE. Thus, transgrafting provides an alternative option for agricultural industries wishing to expand their biotechnology portfolio.  相似文献

以“托鲁巴姆”茄子为砧木,“西安绿”茄子为接穗,采用盆栽试验研究了自毒物质肉桂酸和香草醛对嫁接、自根和砧木茄子根系生长及抗氧化酶活性、渗透调节物质含量的影响.结果表明:肉桂酸、香草醛对茄子根系生长及生理代谢的化感作用基本表现为“低促高抑”,两物质对自根茄子根系表现促进、抑制效应的临界浓度点分别为0.1和0.5 mmol·kg-1;对嫁接、砧木茄子表现促进、抑制效应的临界浓度点分别为0.5和1 mmo·kg-1.茄子根系对自毒物质耐性大小依次为:砧木茄>嫁接茄>自根茄;在较高浓度肉桂酸(0.5~4 mmol·kg-1)和香草醛(1~4mmol·kg-1)作用下,与自根茄子相比,嫁接茄子根系SOD活性提高了8.50%~24.50%,脯氨酸含量增加了9.39%~27.64%,可溶性糖含量增加了12.77%~81.81%,可溶性蛋白含量、根系鲜、干物质量和根系活力显著高于自根茄.表明嫁接换根使茄子根系具有了砧木抗自毒物质的特性,缓解了自毒物质给茄子根系生长带来的不良影响.  相似文献

The application of grafting in tomato production has substantially improved tomato quality and yields. It has been demonstrated that humidity plays an important role in the graft healing of seedlings. This study focuses on the optimum relative humidity (RH) conditions for scion and rootstock healing of grafted tomato (Solanum lycopersicum L.) seedlings. Two tomato cultivars, ‘Super Sunload’ and ‘Super Dotaerang’, grafted onto ‘B-Blocking’ rootstock were subjected to one of three RH regimens: 70–80, 80–90, or 90–100%. The results showed that the scions of both cultivars showed apparent wilting under the 70–80 and 80–90% RH treatments. On this basis, the 90–100% RH treatment was subdivided into 95–96, 97–98, and 99–100% RH treatments, which were then applied. Among these subdivided RH treatments, the fresh weights of the scions and rootstocks significantly increased in response to the treatments of 97–98 and 99–100% RH, and the graft union connection of both cultivars was also enhanced after two days of healing. Furthermore, lower levels of endogenous H2O2 and less activity of antioxidant enzymes were observed in both cultivars in response to treatment with 95–96 or 97–98% RH, which indicated that less oxidative stress occurred. Overall, it is suggested that 97–98% is the optimal RH level for the graft healing of tomato seedlings.  相似文献

The availability of short stature apple scions that required minimal applications of chemical growth retardants and could be used with a range of rootstocks would be of considerable benefit to fruit growers. We have suppressed the expression of a gene encoding the gibberellin (GA) biosynthetic enzyme GA 20-oxidase to reduce the levels of bioactive GAs in a scion variety, resulting in significant reductions in stem height. Application of GA3 reversed the effect. The scion remained dwarfed after grafting on to normally invigorating rootstocks, whilst control plants of the same cultivar displayed the expected vigour when grafted on to these rootstocks. This approach could be applicable to any perennial crop variety, allowing dwarf trees to be obtained on any available rootstock or on their own roots without the need for chemical growth retardant application. In effect, seedlings that are well suited to local conditions (drought, salinity) could be employed as tree rootstocks, as could existing rootstocks valued for characters other than vigour control, such as pest and disease resistance.  相似文献

Abstract

The effects of mutual grafting on the cadmium (Cd) accumulation characteristics of two ecotypes (farmland and mining) of the potential Cd-hyperaccumulator Solanum photeinocarpum were studied through a pot experiment for one month. Four treatments were used in the experiment: ungrafted farmland ecotype (F-CK), ungrafted mining ecotype (M-CK), the farmland ecotype as the scion grafted onto rootstocks of the mining ecotype (F-Scion), and the mining ecotype as the scion grafted onto rootstocks of the farmland ecotype (M-Scion). Mutual grafting increased the rootstock biomass of both S. photeinocarpum ecotypes. However, mutual grafting decreased the scion biomass of F-Scion compared with F-CK and M-CK, and the scion biomass of M-Scion was higher than that of M-CK and lower than that of F-CK. The Cd content in the rootstock of M-Scion increased compared with F-CK, and the Cd content in the rootstock of F-Scion increased compared with M-CK, but mutual grafting decreased the Cd content in scions of both S. photeinocarpum ecotypes. Mutual grafting increased Cd extraction by rootstocks of both S. photeinocarpum ecotypes, but decreased extraction by scions. Therefore, mutual grafting can increase Cd accumulation in S. photeinocarpum rootstocks but not increase Cd accumulation in S. photeinocarpum scions in a short period.  相似文献

In this work we present the first study of the behaviour of tobacco plants, under saline conditions, grafted to salinity-resistant rootstocks of tomato cultivars. To test the viability and efficiency of this grafting technique in tobacco plants subjected to salinity, we analyse the production of foliar biomass and different quality parameters in this crop. With this aim, Nicotiana tabacum cv. Sevilla (scion) was grafted to two cultivars of Lycopersicum esculentum (rootstocks): cv. Jaguar (Sevilla/Jaguar) and cv. Brillante (Sevilla/Brillante). Furthermore, as controls, tobacco plants of cv. Sevilla were used grafted to themselves (Sevilla/Sevilla) and non-grafted plants of cv. Sevilla. Plants were grafted by needle graft following the procedure described by Rivero RM, Ruiz JM, Romero L (2002) Role of grafting in horticultural plants, pp 229–254. In the present work, we demonstrate that the graft of tobacco scions with tomato rootstocks is an effective agricultural approach to improve production and quality in tobacco leaves under conditions of saline stress. Our results show that the rootstock of the cv. Brillante best induced salt resistance in tobacco cv. Sevilla, registering the lowest foliar concentrations of Na+ and Cl, the lowest lipid peroxidation and the highest proline and sugar concentrations. Overall, this is reflected in better biomass production of the aerial part of the plant. Finally, it is noteworthy that grafting in tobacco plants to tomato rootstocks essentially eliminates foliar nicotine levels (reduced to 1%). These results are of great importance, as this technique implies a rapid, efficient and natural alternative in increasing tobacco-leaf quality and thus reducing harmful effects of this alkaloid on the health of smokers.  相似文献
