Contact kairomones are essential for host-acceptance behaviour by female parasitoids. In the solitary endoparasitoid wasp, Diadromus pulchellus, this behaviour depends mainly on compound(s) in the cocoon of their host, Acrolepiopsis assectella pupae. Extracts of empty cocoons and polypeptides extracted from cocoons were tested in acceptance behaviour assays using cotton fibre lures bearing extracts. Extractions with solvents of increasing polarity indicated that the active compounds were polar, while SDS-PAGE showed that four glycopolypeptides contained enough information to trigger host-acceptance behaviour in female wasps. This kairomonal activity was found to be due to the protein moieties, and was independent of any glycosylation. These four glycopolypeptides might be two variants of two soluble sericin-like polypeptides differing in their degree of glycosylation.  相似文献

Proteins isolated from the host cocoon of Acrolepiopsis assectella (Lepidoptera: Yponomeutoidea) act as kairomones for host acceptance by the endoparasitoid wasp Diadromus pulchellus Wesmael (Hymenoptera: Ichneumonidae). In this study, morphological, ultrastructural and electrophysiological studies were carried out in order to identify the contact chemoreceptive sensilla on the parasitoid antennae that perceive the protein kairomones. Three types of sensillum on the antennae of the females were found to have a chemosensory function. The receptor cell(s) of one sensillar type were shown to give a positive electrophysiological response to protein kairomones. This sensillar type is apically multiporous and female specific. Consequently, this sensillum could be the one implicated in the perception of the protein kairomone that triggers the host-acceptance behaviour of D. pulchellus females.  相似文献

In braconid species, teratocytes are derived from a serosal cell membrane which envelops the developing parasitoid embryo. On hatching, this membrane dissociates into individual cells, the teratocytes, which then circulate in the haemolymph of the host. We describe herein such a membrane, surrounding the embryo in eggs of the ichneumonid parasitoid wasp, Diadromus pulchellus. This membrane consisted of a single sheet of tightly packed cells with large 12+/-1.4 &mgr;m nuclei. These cells were released after hatching in vitro and cells of the same size were detected in vivo, in the vicinity of the D. pulchellus embryo. The number of nuclei detected suggests that the serosal membrane consists of about 450+/-150 cells. These cells did not grow after hatching of the parasitoid egg in the parasitized host, Acrolepiosis assectella, during the development of the parasitoid wasp larva. Southern blot experiments, using D. pulchellus satellite DNA or the ribosomal genes as probes, showed that free-living floating cells of wasp origin were present in the body of the parasitized host. This is the first time that free-floating teratocyte-like cells have been described in species of the Ichneumonidae.  相似文献

Abstract.  Recent studies on the leek Allium porrum L. demonstrate that a sharp increase in the release of volatile and nonvolatile sulphur compounds occurs in response to attack by the specialist phytophage, the leek moth Acrolepiopsis assectella (Zeller). The consequences of that response on the developmental capacity and behaviour of the leek moth and on the host-searching behaviour of the parasitoid hymenoptera Diadromus pulchellus Wesmael have been studied. No immediate adverse effect on larval development is seen, but the duration of male development is significantly longer. In addition, an almost 20% decrease is observed in the number of oocytes produced by adult females that developed on attack-damaged leek plants. Leek moths do not avoid damaged plants systematically. On attacked plants, larvae tend to consume more and females to lay more eggs, although the latter tendency disappears after intensive attack. Concerning host-searching by D. pulchellus , the data indicate that females make use of the induced leek response. Parasitoids are attracted more strongly to attacked plants than to undamaged plants and mechanically damaged plants. The results of this study in a tritrophic system involving a biannual plant suggest a complex defence system combining direct and indirect mechanisms. Sulphur compounds may act by reducing second-generation pests, thereby preventing the specialist insect population from exploding at the expense of the leek.  相似文献

Abstract. Experiments were carried out to determine the origin of dialkyl disulphides found in the larval frass of Acrolepiopsis assectella Zell. which enable the specialist parasitoid Diadromus pulchellus Wsm. to find its host. These kairomones are not specific: they are also found in the frass of the generalist Cacoecimorpha pronubana Hb. fed a leek-based diet. They are emitted only after the frass emerges from the gut. Bacteria are present in the gut of larvae and in their frass. The addition of antibiotics to the larval diet prevents bacterial development and suppresses disulphide emission. When grown in the presence of sulphur amino acids, precursors of sulphur volatiles, some bacteria metabolize kairomones. The production of volatile kairomones, responsible for host– parasitoid relationships, depends on the presence of additional organisms, i.e. bacteria, and not on enzymes arising from the plant or the phytophage.  相似文献

Mature larvae of the gregarious endoparasitoid Cotesia kariyai construct cocoons for pupation approximately 10 days after parasitization and emerge from their host Pseudaletia separata under a long day photo-regime (16L8D) at 25 +/- 1 degrees C. The parasitoid larvae make capsules in the host hemocoel just prior to their emergence. These capsules function as "anchors," which enable them to press against the host integument from inside the host. It was predicted that this anchor might be composed of silk proteins secreted from the parasitoid larvae, because a previous study showed that the anchor was made up of a glycoprotein and that the silk gland of parasitoid larvae developed from 2nd larval stage. Fibroin-like proteins in C. kariyai larva mainly consist of two proteins with molecular masses of the 300.6 and 46.7 kDa estimated by SDS-PAGE. The fibroin-like proteins with the same molecular mass were detected from the anchor proteins just prior to parasitoid emergence. These results indicate that the anchor was assembled with fibroin-like proteins and was formed just before parasitoid emergence while in the host body cavity. Injection of bovine pancreatic trypsin inhibited the emergence of parasitoid larvae from the host because the anchor was decomposed by trypsin. Trypsin activity in the parasitized host hemolymph increased only after parasitoid emergence.  相似文献

Many parasitoids control the behavior of their hosts to achieve more preferable conditions. Decreasing predation pressure is a main aim of host manipulation. Some parasitoids control host behavior to escape from their enemies, whereas others manipulate hosts into constructing defensive structures as barriers against hyperparasitism. Larvae of the parasitoid wasp Cotesia glomerata form cocoon clusters after egression from the parasitized host caterpillar of the butterfly Pieris brassicae. After the egression of parasitoids, the perforated host caterpillar lives for a short period and constructs a silk web that covers the cocoon cluster. We examined whether these silk webs protect C. glomerata cocoons against the hyperparasitoid wasp Trichomalopsis apanteroctena. In cocoon clusters that were not covered by silk webs (bare clusters), only cocoons hidden beneath others avoided hyperparasitism. In covered cocoon clusters, both cocoons hidden beneath others and those with a space between them and the silk web avoided hyperparasitism, whereas cocoons that contacted the silk webs were parasitized. The frequency of cocoons that were hidden beneath others increased with the increasing number of cocoons in a cluster, but the defensive effect of cluster size was thought to be lower than that of silk webs. However, the rate of hyperparasitism did not differ between covered and bare clusters when we allowed the hyperparasitoids to attack the cocoon clusters in an experimental arena. This result was thought to have been caused by low oviposition frequency by these hyperparasitoids. As a result, silk webs did not guard the cocoons from hyperparasitoids in our experiments, but would protect cocoons under high hyperparasitism pressure by forming a space through which the ovipositors could not reach the cocoons.  相似文献

Silk proteins were isolated from the cocoons of the nonmulberry silkworm, Philosamia ricini. Three polypeptides of 97, 66, and 45 kDa were identified. The 66-kDa molecule represented sericin, whereas the 97-kDa and the 45-kDa polypeptides linked together through a disulfide bond constituted the fibroin protein. Antibodies raised against the 97-kDa P. ricini fibroin heavy chain reacted specifically with this molecule and did not recognize fibroin heavy chain from another nonmulberry silkworm, Antheraea assama or from the mulberry silkworm, Bombyx mori, suggesting the presence of P. ricini species-specific determinants in this heavy chain. Antibodies generated against fibroin light chain of P. ricini also showed similar reactivity pattern. Immunoblot analysis with proteins isolated from the silk glands of P. ricini at different stages of larval development showed that the expression of fibroin heavy chain was developmentally and spatially regulated. The protein was most abundant in the 5th instar larva, and could be detected in the middle and the posterior but not the anterior silk glands. The amino acid composition of the 97-kDa fibroin protein showed abundance of glutamic acid and did not contain (Gly-Ala)(n) motifs, a characteristic feature of B. mori fibroin heavy chain. Our study reveals significant differences between the nonmulberry silkworm P. ricini and the mulberry silkworm B. mori in the biochemical composition and immunochemical characteristics of fibroin heavy chain. These differences might be responsible for the differences seen in the quality of silk produced by these two silkworms.  相似文献

The study of various behavioural criteria of femaleDiadromus pulchellus parasitoids in the presence of theirAcrolepiopsis assectella hosts has shown the essentially chemical nature of the stimulant determining host recognition. The physical stimuli of the cocoon seem not to be implicated. Thus cocoons whose original texture has been completely altered, either mechanically or chemically, as well as the silk excreted by the host caterpillars significantly stimulates the female parasitoids. The cocoon contact kairomones are detected in testing the aqueous extracts ofA. assectella which provoke a positive behavioural response from the females in a threshold concentration of 1 cocoon-equivalent. The comparison of aqueous extracts ofA. assectella host cocoons, and of non-host species:Bombyx mori, Ephestia kuehniella andCacoecimorpha pronubana demonstrate the kairomone specificity of the silk, the extracts of non-host cocoons being ignored by the parasitoid, as were the silk threads left byE. kuehniella caterpillars. Finally the contact kairomones linked to the silk seem to be independent of the host plant and of the nutrient diet of the host caterpillars. The cocoons spun by the caterpillars reared on leeks or on artificial diets with or without powdered leek provoke similar responses in the parasitoids.  相似文献

Rearing and release procedures forCotesia melanoscela (Ratzeburg) (Hymenoptera: Braconidae), an early-season parasitoid ofLymantria dispar (L.), were evaluated in 1995 and 1996 at the Beltsville Agricultural Research Center, Beltsville, Maryland, USA. Mass rearing procedures were developed during the winter/spring of 1995/1996 to produce diapausingC. melanoscela cocoons for experimentation. Program efficiency as measured by percent adult emergence was estimated to be 84% with a measured sex ratio of 48:52 M:F, at an estimated cost per 1 000 cocoons of U.S. $5.26 for materials and a labor requirement of approximately 6 hours. A host larvae:female parasitoid ratio of 100:1 was found to be most efficient in terms of number of parasitoid cocoons produced per female parasitoid, but a ratio of 100:2 was most efficient in terms of number of cocoons per rearing cup. OverwinteringC. melanoscela cocoons under natural conditions for 25 weeks (November placements) before expectedL. dispar egg hatch (mid-April) resulted in excellent synchrony with host egg hatch. Alternatively, holdingC. melanoscela cocoons in cold storage for 23 or more weeks at 5°C resulted in adult parasitoid emergence beginning 10 days after removal from cold storage, and proper timing of removal would result in similar synchrony of adult emergence with host egg hatch. 1 Predation and parasitism on litter‐buried cocoons of the common pine sawfly Diprion pini (L.) were compared in different forest types with endemic sawfly populations by field exposure of laboratory‐reared cocoons during three consecutive years (1993–1995). 2 The impact of cocoon predation was dependent on season and forest type. The highest predation (up to 95%) was found during autumn in forest stands with a dense understory vegetation. 3 Cocoon parasitism varied between year, season and forest type. The highest parasitoid attack was observed in pure pine forests with more or less barren soils, but did not exceed 24% of exposed cocoons. 4 Cocoons were exposed in small patches. Predators tended to exploit all cocoons of a patch, whereas parasitoids only attacked a few cocoons of a patch. Predation was similar on cocoons placed in the litter and those buried more deeply in the soil, whereas parasitism of soil‐buried cocoons was rare. 5 These results indicate that predators can have a remarkable potential for limiting endemic sawfly densities, if habitat conditions in a forest maintain their population and support their foraging behaviour. A notable effect of parasitoids on sawfly cocoons deposited in the litter is obviously restricted to typical pure and barren pine forests, but may play there a similar role as predation.  相似文献

Sex determination in the order Hymenoptera is based on arrhenotoky, hymenopteran males are usually haploid and females diploid. Males of the Ichneumonidae Diadromus pulchellus, solitary endoparasitoid of A Acrolepiopsis assectella pupae, are normally haploid, but diploid males are present in a natural population and can be obtained in an experimental population. The future of an ovocyte laid by mated females of the solitary endoparasitoid D. pulchellus was characterised by six probabilities related to the sex and development of the ovocyte. The probabilities of fertilisation of female ovocyte (k1) or non-fertilisation (k3) showed that an inseminated female functioned as a unmated female for half of the time (since k1 = 0.492 and k3 = 0.455) with the probability of fertilisation of male ovocyte, k2, equal to 0.053. The survival probabilities of each type of ovocyte showed that an ovocyte had a high probability of developing up to the adult stage, although the difference between the calculated sex ratios at laying (males/females = 1.032) and at emergence (0.90) revealed a slight reduction in the number of haploid sons. The probabilities of fertilisation and of viability of all the ovocytes laid by each of the 33 mated females were analysed by an ascending hierarchical classification of Euclidean distances and by an analysis of their principal components. The 33 mothers were distributed into four distinct sub-groups characterised by a sex ratio varying from an exclusive presence of females to an exclusive presence of males. Our hypothesis was that this distribution in four sub-sets could not simply result from the random nature of the sample.  相似文献

In spiders, sex pheromones are often associated with silk produced by females, and function in mate attraction, recognition, and evaluation. Silk-bound pheromones typically elicit courtship behaviour in web-building spiders. Here we (1) describe courtship interactions of Steatoda grossa males with virgin or mated females, and (2) show that silk and methanol extracts of silk produced by virgin females trigger courtship behaviour (silk production) by males, whereas silk of mated females does not. Our results indicate that (1) virgin females produce a silk-bound sex pheromone, (2) males discriminate between virgin and mated females based on silk cues, and (3) male silk likely functions in sexual communication.  相似文献

The biology and ecology of Eriborus terebrans (Grav.), a parasitoid of the poplar clearwing moth, Paranthrene tabaniformis (Rott.), were studied during the period 1987–98. One-year-old poplar ( Populus spp.) shoots infested with P. tabaniformis larvae were collected from poplar seedlings at 11 localities in Bulgaria and examined in both field and laboratory conditions. Eriborus terebrans was recorded in seven localities as a solitary internal larval parasitoid of P. tabaniformis which developed two generations in early and mid-stage host larvae. Eriborus terebrans overwintered as a larva in P. tabaniformis overwintering larvae. In the field adult parasitoids of the overwintering generation appeared between late April/early May, and June or July. The peak activity of E. terebrans adults only coincided with the beginning of host emergence, which resulted in low levels of parasitism, being no more than 6.2%. Parasitoid adults of the summer generation appeared in late June–mid August. In this period enough larvae of the host were suitable for attacking and parasitism reached 24.4–39% in some cases. The average mortality of P. tabaniformis overwintering larvae caused by this parasitoid in Bulgaria during the period of the study was 4.7%. A significant part of the parasitized P. tabaniformis larvae constructed tunnel structures of frass and silk threads over the external openings of the galleries. It is possible that these structures protect the parasitoid cocoons from natural enemies – hyperparasitoids and predators.  相似文献

Spider dragline silk is a unique fibrous protein with a combination of tensile strength and elasticity, but the isolation of large amounts of silk from spiders is not feasible. In this study, we generated germline-transgenic silkworms (Bombyx mori) that spun cocoons containing recombinant spider silk. A piggyBac-based transformation vector was constructed that carried spider dragline silk (MaSp1) cDNA driven by the sericin 1 promoter. Silkworm eggs were injected with the vector, producing transgenic silkworms displaying DsRed fluorescence in their eyes. Genotyping analysis confirmed the integration of the MaSp1 gene into the genome of the transgenic silkworms, and silk protein analysis revealed its expression and secretion in the cocoon. Compared with wild-type silk, the recombinant silk displayed a higher tensile strength and elasticity. The results indicate the potential for producing recombinant spider silk in transgenic B. mori.  相似文献

The use of doped silk fibroin (SF) films and substrates from Bombyx mori cocoons for green nanotechnology and biomedical applications has been recently highlighted. Cocoons from coloured strains of B. mori, such as Golden‐Yellow, contain high levels of pigments that could have a huge potential for the fabrication of SF based biomaterials targeted to photonics, optoelectronics and neuroregenerative medicine. However, the features of extracted and regenerated SF from cocoons of B. mori Golden‐Yellow strain have never been reported. Here we provide a chemophysical characterization of regenerated silk fibroin (RSF) fibers, solution, and films obtained from cocoons of a Golden‐Yellow strain of B. mori, by SEM, 1H‐NMR, HPLC, FT‐IR, Raman and UV‐Vis spectroscopy. We found that the extracted solution and films from B. mori Golden‐Yellow fibroin displayed typical Raman spectroscopic and optical features of carotenoids. HPLC‐analyses revealed that lutein was the carotenoid contained in the fiber and RSF biopolymer from yellow cocoons. Notably, primary neurons cultured on yellow SF displayed a threefold higher neurite length than those grown of white SF films. The results we report pave the way to expand the potential use of yellow SF in the field of neuroregenerative medicine and provide green chemistry approaches in biomedicine. © 2016 Wiley Periodicals, Inc. Silk cocoons provide protection to silkworm from biotic and abiotic hazards during the immobile pupal phase of the lifecycle of silkworms. Protection is particularly important for the wild silk cocoons reared in an open and harsh environment. To understand whether some of the cocoon components resist growth of microorganisms, in vitro studies were performed using gram negative bacteria Escherichia coli (E. coli) to investigate antibacterial properties of silk fiber, silk gum, and calcium oxalate crystals embedded inside some cocoons. The results show that the previously reported antibacterial properties of silk cocoons are actually due to residues of chemicals used to isolate/purify cocoon elements, and properly isolated silk fiber, gum, and embedded crystals free from such residues do not have inherent resistance to E. coli. This study removes the uncertainty created by previous studies over the presence of antibacterial properties of silk cocoons, particularly the silk gum and sericin. © 2013 Wiley Periodicals, Inc. During evolution, certain endoparasitoid wasps have developed mechanisms to suppress the defence systems of their hosts. For this purpose, these species, all of which belong to the families Ichneumonidae and Braconidae, inject various kinds of virus-like particles. The most studied of these particles are classified as polydnaviruses (family Polydnaviridae) which are symbiotic viruses. Over the past decade, it has also been shown that several wasp species harbour reoviruses (family Reoviridae), and that two of these suppress host defence, allowing the development of the parasitoid eggs. In this paper, we summarize the key features of these viruses and their relationships with their wasp hosts. Five reoviruses are known that appear to be non-pathogenic for the wasps. Three of these, McRVLP, HeRV, OpRVLP, use their wasp hosts as vectors, and do not appear to be involved in host defence suppression. The fourth, DpRV-1, is a commensal reovirus detected in most field populations of the wasp, Diadromus pulchellus. This reovirus is always found associated with an ascovirus, DpAV-4a, which is indispensable for host immune suppression. Although DpRV-1 has not been shown to directly increase D. pulchellus parasitic success, it may contribute to this success by retarding DpAV-4a replication in the wasp. The fifth reovirus, DpRV-2, occurs in a specific population of D. pulchellus in which DpRV-1 and DpAV-4 are absent. This virus has a mutualistic relationship with its wasp host, as its injection by females during oviposition is essential for host immunosuppression. Interestingly, these viruses belong to several different reovirus genera.  相似文献

In this study we produced germline transgenic silkworms that spin cocoons containing recombinant human serum albumin (rHSA) in the sericin layer. A piggyBac-based transformation vector was constructed that carried HSA cDNA driven by sericin-1 gene promoter, viral enhancer hr3, and gene encoding viral trans-activator IE1. Isolated silk glands were bombarded with the vector and transplanted into host larvae. Three days later, the transplants were immunohistochemically analyzed, which showed that middle silk gland (MSG) cells expressed rHSA and secreted it into the MSG lumen. Then, silkworm eggs were injected with the vector and developed to larvae. The obtained transgenic silkworms spun silk threads whose sericin layers contained rHSA at 3.0microg/mg of cocoons. Most (83%) of the rHSA in cocoons was extracted with phosphate buffered saline, which was then subjected to ammonium sulfate precipitation and affinity chromatography. Finally, we obtained 2.8mg of 99%-pure rHSA from 2g of cocoons. Measurements of circular dichroism spectra of rHSA, and equilibrium dissociation constants of rHSA to warfarin and naproxen indicated that rHSA was conformationally and functionally identical to natural plasma HSA. Germline transgenic silkworms will be useful for producing various recombinant proteins in the sericin layer of cocoons.  相似文献
