Recent recognition of widespread polyandry in insects has generated considerable interest in understanding why females mate multiple times and in identifying factors that affect mating rate and inhibit female remating. However, little attention has been paid to understanding the question from both a female and male perspective, particularly with respect to factors that may simultaneously influence female remating rates. Here, we report on a study aimed at ascertaining the possible interactive effects that male and female size and diet, and female access to a host could have on mating latency, probability, and duration and female refractory period using two tropical fruit fly species with contrasting life histories. Of all factors tested, adult diet played the most significant role. Both Anastrepha ludens and Anastrepha obliqua males which had constant access to protein and sucrose mated more often, had shorter copulations and induced longer refractory periods in females than males fed a low quality diet (sucrose offered every third day). Female size and the interaction with male diet determined how quickly female A. ludens mated for the first time. Smaller females mated sooner with low quality fed males than with high quality fed males while there was no difference for large females, suggesting that male choice may be at play if high quality fed males discriminate against smaller females. Copulation duration also depended on both male and female nutritional condition, and the interaction between male diet and female size and diet. Large and high quality fed females had shorter copulations regardless of male condition. Importantly, for A. ludens, female refractory period depended on male size and the nutritional condition of both males and females, which could indicate that for this species, female receptivity does not depend only on the condition of the male ejaculate. For A. obliqua refractory period was associated with the interaction between male size and diet and male diet and host presence. We discuss our results in terms of male ability to inhibit female remating and the relative contribution of female condition to this behavior. In field-cage studies, we investigated how the foraging behavior of tephritid fruit flies is modified by experience immediately prior to release on host plants. We observed females of a relatively monophagous species,Rhagoletis mendax (blueberry maggot fly), an oligophagous species,Rhagoletis pomomella (apple maggot fly), and a polyphagous species,Ceratitis capitata (Mediterranean fruit fly). Just prior to release on a host plant, the following kinds of stimuli were supplied: (1) single oviposition in a host fruit, (2) contact with 20% sucrose, (3) contact with a mixture of protein food (bird feces and sucrose), (4) contact with water, and (5) a walk over a host-plant leaf. When flies foraged on host plants without resources, search was most intensive (as measured by number of leaves visited) following a single oviposition in fruit, but residence time generally was the same following exposure to sugar, protein, and fruit stimuli.Rhagoletis mendax andC. capitata females visited the fewest leaves following exposure to water or host leaves, whereasR. pomonella foraged equally intensively following exposure to food stimuli, water, or leaves. On host plants containing resources (fruit and protein food), a single oviposition dramatically increased the number of females of all three species that found fruit compared to females that received experience with food, water or foliar stimuli. We found no significant effect of recent brief experience with any of the stimuli on subsequent attraction to protein food. Overall,C. capitata exhibited a higher propensity to abandon host plants than eitherR. mendax orR. pomonella. We suggest that this may reflect adaptations to differences in distribution of host plants in nature, strategies of dispersal, and host range.  相似文献

Insects display a whole spectrum of morphological diversity, which is especially noticeable in the organization of their appendages. A recent study in a hemipteran, Oncopeltus fasciatus (milkweed bug), showed that nubbin (nub) affects antenna morphogenesis, labial patterning, the length of the femoral segment in legs, and the formation of a limbless abdomen. To further determine the role of this gene in the evolution of insect morphology, we analyzed its functions in two additional hemimetabolous species, Acheta domesticus (house cricket) and Periplaneta americana (cockroach), and re-examined its role in Drosophila melanogaster (fruit fly). While both Acheta and Periplaneta nub-RNAi first nymphs develop crooked antennae, no visible changes are observed in the morphologies of their mouthparts and abdomen. Instead, the main effect is seen in legs. The joint between the tibia and first tarsomere (Ta-1) is lost in Acheta, which in turn, causes a fusion of these two segments and creates a chimeric nub-RNAi tibia–tarsus that retains a tibial identity in its proximal half and acquires a Ta-1 identity in its distal half. Similarly, our re-analysis of nub function in Drosophila reveals that legs lack all true joints and the fly tibia also exhibits a fused tibia and tarsus. Finally, we observe a similar phenotype in Periplaneta except that it encompasses different joints (coxa–trochanter and femur–tibia), and in this species we also show that nub expression in the legs is regulated by Notch signaling, as had previously been reported in flies and spiders. Overall, we propose that nub acts downstream of Notch on the distal part of insect leg segments to promote their development and growth, which in turn is required for joint formation. Our data represent the first functional evidence defining a role for nub in leg segmentation and highlight the varying degrees of its involvement in this process across insects.  相似文献

The pesticide paraquat (PQ) was found to be a suitable xenobiotic to model Parkinson’s disease. The reactive oxygen species (ROS) production was suggested to be the main cause of PQ toxicity but very few evidences were found for its generation in the brain in vivo after ip administration. We compared the effects of PQ-induced ROS generation between the brain structures and the peripheral tissues using two different hydroxyl radical generation markers. Repeated but not single ip PQ administration increased the levels of ROS in the striatal homogenates but, when measured in the extracellular microdialysis filtrate, no change was observed. The increased dopamine release was detected in the striatum after the fourth PQ administration and its basal levels were decreased. A single treatment with the pesticide did not influence ROS production in the lungs or kidneys but repeated intoxication decreased its levels. These results suggest that repeated, systemic administration of a low dose of PQ triggers intracellular ROS formation in the brain and can cause slowly progressing degenerative processes, without the toxic effects in the peripheral tissues.  相似文献

The Arabidopsis GIGANTEA (GI) gene has been shown to be involved in the regulation of the oxidative stress response; however, little is known about the mechanism by which GI gene regulates the oxidative stress response. We show here that enhanced tolerance of the gi-3 mutant to oxidative stress is associated, at least in part, with constitutive activation of superoxide dismutase (SOD) and ascorbate peroxidase (APX) genes. The gi-3 plants were more tolerant to parquart (PQ) or hydrogen peroxide (H2O2)-mediated oxidative stress than wild-type plants. Analyses of concentrations of endogenous H2O2 and superoxide anion radicals as well as lipid peroxidation revealed that enhanced tolerance of gi-3 plants to oxidative stress was not due to defects in the uptake of PQ or the sequestration of PQ from its site of action, and that the gi-3 mutation alleviated oxidative damage of plant cells from PQ stress. Moreover, the gi-3 mutant showed constitutive activation of cytosolic Cu/ZnSOD and plastidic FeSOD as well as cytosolic APX1 and stromal APX genes, which at least in part contributed to constitutive increases in activities of anti-oxidative enzymes SOD and APX, respectively. To our knowledge, we demonstrate, for the first time, that GI gene regulates the oxidative stress response, at least in part, through modulation of SOD and APX genes.  相似文献

The effect of insect age on the neural responsiveness of gustatory sensilla was investigated. Electrophysiological recordings were obtained from type A and type D sensilla on the pro- and meso-thoracic tarsi, and from sensilla on the labellum of the turnip root fly,Delia floralis (Fallen) in response to potassium chloride, sucrose and sinigrin. The age of the fly did have an effect on the numbers of sensilla responding to the test stimuli and on the magnitude of the response, but there was no consistent pattern in these effects among sensilla. The labellar sensilla were more responsive to sucrose than the tarsal sensilla and the proportion of flies whose labellar sensilla responded to sucrose was initially low, but increased after day 2 of adult life. In contrast, the number of flies whose tarsal sensilla responded to stimulation with sucrose was initially high and decreased as the flies aged. There was a similar decline in the proportion of tarsal sensilla responding to potassium chloride. Neither the proportion of flies whose tarsal sensilla responded to sinigrin nor the magnitude of the response was influenced by the age of the fly. These finding are discussed in relation to the feeding and host selection behaviour of the fly.  相似文献

Artificial diets have been developed to sustain the mass rearing of a wide range of arthropod natural enemies, with varying success. In some cases, such diets can be optimized using insect‐derived materials, such as haemolymph. In this study, we examined the effect of supplementing haemolymph of the black soldier fly, Hermetia illucens, to a basic artificial diet for the phytoseiid mite Amblyseius swirskii. The survival, development and reproduction of the predatory mite were assessed when fed on artificial diets composed of honey, sucrose, tryptone, yeast extract and egg yolk, supplemented with 5%, 10%, or 20% of H. illucens pre‐pupal haemolymph. Developmental time from larva to adult was shorter for males and females offered artificial diets supplemented with 20% haemolymph vs. the basic diet. The oviposition rate and total fecundity of females reared on the basic diet were substantially lower than those of females supplied with the enriched diets. The intrinsic rate of increase was highest on the diet containing 20% haemolymph, followed by those containing 10% and 5% haemolymph. In a subsequent diet‐switching experiment, mites fed on the basic diet in their juvenile stages were switched upon adulthood to diet enriched with different concentrations of H. illucens haemolymph. The females that were fed with the enriched diets from the adult stage on had higher oviposition rates and fecundities than those maintained on the basic diet, but their reproductive parameters were not significantly affected by the concentration of the haemolymph in the artificial diet. In conclusion, supplementing artificial diets with black soldier fly haemolymph significantly improved their nutritional value for A. swirskii. Our findings indicate the potential of using H. illucens as a cheap source for haemolymph in artificial diets, as the fly can be cost‐effectively produced at a large scale on organic waste materials.  相似文献

SPINDLY (SPY) gene encodes a putative O-linked N-acetyl-glucosamine transferase, and yeast two-hybrid assay identified GIGANTEA (GI) as a SPY-interacting partner in Arabidopsis. GIGANTEA gene was previously shown to be involved in the regulation of oxidative stress response; however, it is unclear whether SPY gene is also involved in oxidative stress response. Here we showed that SPY plays a role in the regulation of the oxidative stress response. The spy-1 mutant was more tolerant to paraquat (PQ)-or hydrogen peroxide (H2O2)-mediated oxidative stress than wild-type plants. Analyses of endogenous H2O2 and superoxide anion radicals as well as lipid peroxidation revealed that enhanced tolerance of the spy-1 mutant to PQ-stress was not due to defects in the PQ uptake or the PQ sequestration from its site of action but rather the spy-1 mutation alleviated oxidative damage of plant cells upon PQ stress. Higher constitutive activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in spy-1 are more likely to be due to activation of both CSD2 gene encoding chloroplast Cu/Zn SOD and APX1 gene. Taken together, these results suggest that enhanced tolerance of the spy-1 mutant to oxidative stress is associated, at least in part, with constitutive activation of CSD2 and APX1. A range of biologically active secondary metabolites with pharmacological application has been reported to occur in marine sponges. The present study was undertaken to provide a set of data on the safety of a hydro-alcoholic extract (ALE) and an aqueous fraction (AQE) from Aplysina fulva Pallas, 1766 (Aplysinidae, Verongida, Porifera). Salmonella typhimurium strains TA97, TA98, TA100 and TA102, Escherichia coli strains PQ65, OG40, OG100, PQ35 and PQ37 and Balb/c 3T3 mouse fibroblasts were used to detect induction of DNA lesions by ALE and AQE. Assays used for these analyses were a bacterial (reverse) mutation assay (Ames test), the SOS-chromotest and the comet assay. Both extracts presented identical infrared 2-oxazolidone spectra. ALE treatment induced a higher frequency of type-4 comets, indicative of increasing DNA migration, in the alkaline comet assay. ALE also induced a weak genotoxic effect, as expressed by the induction factor (IF) values in the test with E. coli strain PQ35 (IF = 1.5) and by cytotoxic effects in strains PQ35, PQ65 and PQ37. Positive SOS induction (IF = 1.7) was detected in strain PQ37 treated with diluted AQE. No genotoxic effects were observed in strains PQ35, PQ65, OG40 and OG 100 after treatment with AQE dilutions. Using the bacterial (reverse) mutation test and survival assays with or without S9 mix, after 60 min of pre-incubation, we observed for strain TA97 treated with ALE a weak mutagenic response (MI = 2.2), while cytotoxic effects were seen for strains TA98, TA100 and TA102. AQE did not show mutagenic activity in any of the strains tested, but a weak cytotoxic effect was noted in strain TA102. Our data suggest that both ALE and AQE from A. fulva induce DNA breaks leading to cytotoxicity and mutagenicity under the conditions used.  相似文献

Two entomopathogenic fungi in theEntomophthora muscaespecies complex that infect house flies were used in this study:E. muscaeFresenius (16–17 nuclei/conidium) occurred naturally at four southern California dairies, whileEntomophthora schizophoraeKeller and Wilding (4–8 nuclei/conidium) did not. During the first year of the study, onset of measurableE. muscaeinfections occurred between September and November but varied among sites. At least 20% of the flies at all four dairies were infected by November, and infection at one site exceeded 70%. During the fall epizootic period, infection levels were inversely related to temperature. Average weekly temperatures higher than 17–20°C and maximum daily temperatures higher than 26–28°C were statistically correlated with low infection levels. In the second year,E. schizophoraewas introduced by releasing diseased flies at two dairies (four times at one dairy and three times at the other).E. schizophoraewas recovered for a brief time in the house fly population after the first two releases at one site but not at the second site.  相似文献

A guild of 20 late spring- and early summer-flowering species ofIridaceae, Geraniaceae andOrchidaceae is pollinated partly or exclusively by the long-proboscid flyMoegistorhynchus longirostris (Nemestrinidae). This large-bodied fly, active in late spring and early summer, has mouthparts 40–70 (90) mm long and forages for nectar from a variety of species. These plants share a suite of convergent floral features including a straight or weakly curved floral tube usually 50–70 mm long but sometimes to 90 mm, relatively short petals or tepal lobes coloured white, cream or salmon with reddish nectar guides, and often violet or red anthers and pollen. Flowers of most species with these characteristics are zygomorphic with the stamens either arcuate (mostIridaceae) or declinate (Geraniaceae and someIridaceae). The flowers are odourless and typically secrete large amounts of nectar of relatively constant sugar concentration, mostly 24–29%, with a high sucrose:hexose ratio. Guild members utilize five separate sites of pollen deposition on the body of the fly, typically utilizing different deposition sites when two or more co-occur, indicating strong selection to aviod pollen contamination.M. longirostris is restricted to the west coast of southern Africa and at least 8 species appear to depend exclusively on the insect for pollination. The remaining species in the guild are pollinated by one or both of the long-proboscid fliesPhiloliche gulosa andP. rostrata (Tabanidae) over other parts of their range. Species and races pollinated entirely byM. longirostris have longer floral tubes which makes nectar unavailable to other insects, including other species of long-proboscid fly. The only insect with mouthparts long enough to forage effectively on these long-tubed flowers isM. longirostris and this fly must be considered a keystone species in the ecosystems in which it occurs.Dedicated to emer. Chlorophyll a fluorescence rise kinetics (from 50 μs to 1 s) were used to investigate the non-photochemical reduction of the plastoquinone (PQ) pool in osmotically broken spinach chloroplasts (Spinacia oleracea L.). Incubation of the chloroplasts in the presence of exogenous NADPH or NADH resulted in significant changes in the shape of the fluorescence transient reflecting an NAD(P)H-dependent accumulation of reduced PQ in the dark, with an extent depending on the concentration of NAD(P)H and the availability of oxygen; the dark reduction of the PQ pool was saturated at lower NAD(P)H concentrations and reached a higher level when the incubation took place under anaerobic conditions than when it occurred under aerobic conditions. Under both conditions NADPH was more effective than NADH in reducing PQ, however only at sub-saturating concentrations. Neither antimycin A nor rotenone were found to alter the effect of NAD(P)H. The addition of mercury chloride to the chloroplast suspension decreased the NAD(P)H-dependent dark reduction of the PQ pool, with the full inhibition requiring higher mercury concentrations under anaerobic than under aerobic conditions. This is the first time that this inhibitory role of mercury is reported for higher plants. The results demonstrate that in the dark the redox state of the PQ pool is regulated by the reduction of PQ via a mercury-sensitive NAD(P)H-PQ oxidoreductase and the reoxidation of reduced PQ by an O2-dependent pathway, thus providing additional evidence for the existence of a chlororespiratory electron transport chain in higher plant chloroplasts. A chemically defined diet is a useful tool for the study of nutritional physiology of organisms. We have developed such a diet for Camponotus carpenter ants to facilitate experiments on nutritional requirements of these ants. Worker colonies of Camponotus floridanus were fed with a chemically defined diet, containing all essential minerals, amino acids, vitamins, growth factors and sucrose in an agar matrix. After 13 weeks, neither the number of raised pupae, their dry weight, nor the mortality of workers in subcolonies fed with this diet differed significantly from control colonies fed with Bhatkar-Whitcomb-agar, in addition to cockroaches and diluted honey. Therefore, this diet is adequate for a normal brood production and a maximal growth rate of C. floridanus larvae, at least for a period of three months. Fruit flies (Diptera: Tephritidae) have been used previously as a model to explore the effect of nutrition on male reproduction. Here we tested the hypothesis that adult feeding history influences male sexual performance in four species of neotropical fruit flies of the genus Anastrepha (A. ludens, A. obliqua, A. serpentina, and A. striata), an approach that allowed us to consider species-specific ecological factors independent of phylogenetic constraints. We fed, from the moment of emergence, four experimental groups of males of each species with either (1) dry sucrose, (2) a mixture of dry sucrose and hydrolyzed protein (enzymatic yeast hydrolyzate), (3) a mixture of dry sucrose and bird feces, or (4) an open fruit [Citrus sinensis (orange), Mangifera indica (mango), Manilkara zapota (chico zapote), and Psidium guajava (guava) for A. ludens, A. obliqua, A. serpentina, and A. striata, respectively]. Cohorts of 12-day-old individuals (28 males [7 of each diet treatment] and 14 females) were released simultaneously into species-specific field cages and the following parameters recorded over a 4-day period (procedure repeated with five different cohorts for each species): number of males involved in calling activities every hour (i.e., pheromone emissions, wing fanning, wing motions, or extensions of the mouthparts), identity of copulating males, and copula duration. In the case of A. obliqua, we also observed separately cohorts of 20-day-old individuals. In A. obliqua, A. serpentina, and A. striata, males fed on a protein-supplemented diet had higher copulatory success when competing against males fed on other diets. Anastrepha serpentina and A. striata were sensitive to diet on all but two of the parameters analyzed (i.e., males with consecutive copulas and copula duration in the case of A. serpentina and males with multiple and consecutive copulas in the case of A. striata). In sharp contrast to this, the male mating performance of A. ludens was not affected by diet. Anastrepha obliqua was sensitive to different parameters depending on age. Among the host fruits of the Caribbean fruit fly there are a variety of sizes and shapes. These morphological differences may influence the vulnerability of the larvae to parasites. In the laboratory, Caribbean fruit fly larvae placed in the smaller of 2 different sizes of artificial ‘fruit’ (cloth spheres filled with a diet material) were parasitized at a higher rate by the braconid,Diachasmimorpha longicaudata (Ashmead) when spheres were presented separately. However, when parasites were simultaneously presented with 6 different sizes of ‘fruit’ there was no significant relationship between size and parasitization rate. This may be due to the parasites preference to search for larvae in larger ‘fruit’. In field collections of different species of host fruit, a significant inverse correlation exists between fruit radius and rate of parasitization. However, host fruit size accounts for only about 5% of the variance in yearly parasitization rates.   相似文献

—The effect of the para-(PQ) and the ortho-(OQ) quinones of 6-hydroxydopamine (6-OH-DA) on transmitter uptake-storage mechanisms of catecholamine neurons in mouse and rat has been investigated. After the administration of PQ and OQ there was a dose-dependent and long-lasting disappearance of noradrenaline (NA) nerve terminals as demonstrated by fluorescence histochemistry and a reduction of the in vitro uptake of [3H]NA in mouse atrium and iris. These effects could be completely counteracted by blockade of the ‘membrane pump’ transport mechanism with desipramine, while monoamine oxidase inhibition, by nialamide and administration of ascorbic acid potentiated the effects produced by the two quinones. The results obtained after PQ and OQ were largely identical with those seen after administration of 6-OH-DA, well-known for its neurotoxic action on catecholamine neurons. It is therefore concluded that PQ and OQ are able to produce an acute and selective degeneration of NA nerve terminals similar to that of 6-OH-DA. The results obtained after intraventricular injection of the quinones into rat brain were also in agreement with this view. Neonatal administration of PQ or OQ to mice caused a permanent and marked decrease in [3H]NA uptake in the cerebral cortex and the spinal cord, whereas the uptake was markedly increased in the pons-medulla, similar to that seen after 6-OH-DA. The PQ and the OQ were equally potent in most experiments although clearly less potent than 6-OH-DA itself. The quinones were also found to be equally or slightly less potent than 6-OH-DA in affecting [3H]NA uptake and retention in vitro in atrium and cerebral cortex from untreated mice. It may be concluded that PQ and OQ exert their neurotoxic action on NA neurons after transition to 6-OH-DA, after a rapid extraneuronal equilibration. 6-OH-DA thus formed can thereafter be taken up and accumulated intraneuronally by use of the ‘membrane pump’ and the specific degenerative action is elicited. The lower neurotoxic potency of the quinones may be attributed to their known ability to undergo covalent binding with proteins and/or formation of 5,6-dihydroxyindole.  相似文献

Honeydew droplets freshly excreted by two aphid species, Myzus persicae (Sulz.) and Acyrthosiphon pisum (Harr.), feeding on nutritionally adequate, artificial diets containing 10–40% w/v sucrose, were collected in oil. The volume, frequency of excretion, and carbohydrate content of the droplets were determined by measuring the diameter of the droplets in the oil, by recording the number of droplets excreted per aphid per hour, and by using a quantitative colorimetric method for estimating carbohydrates. Rates for the intake or excretion of fluid, the intake and absorption of sucrose, and the excretion of carbohydrate were consequently calculated for the two species on each dietary sucrose concentration. The percentage of dietary sucrose absorbed by the aphids ranged from 36% on diet with 40% sucrose to 97% on diet with 10% sucrose. Such honeydew/diet relationships may allow one to use honeydew carbohydrate concentrations to estimate the sucrose concentrations of the phloem sap ingested by aphids feeding on host plants.
Résumé Des gouttelettes de miellat fraîchement excrétées par deux espèces de pucerons, Myzus persicae (Sulz.) et Acyrthosiphon pisum (Harr.), pucerons nourris sur des régimes nutritifs synthétiques contenant 10–40% de saccharose, ont été récupérées dans de l'huile. Le volume des gouttelettes a été déterminé en mesurant leur diamètre individuel dans l'huile, la fréquence d'excrétion établie par le nombre de gouttelettes excrétées par puceron par heure, et la teneur en glucides par une méthode d'analyse colorimétrique quantitative. Les taux d'ingestion et d'excrétion de liquide, d'ingestion et d'absorption de saccharose ainsi que les taux d'excrétion de glucides ont été par la suite calculés pour les deux espèces d'aphides nourris sur les différentes concentrations de saccharose. Le pourcentage de saccharose alimentaire absorbé par les aphides a été de 36% sur le régime contenant 40% de ce glucide, mais s'est élevé à 97% sur le régime ne contenant que 10% de saccharose. D'après ces relations miellat/régime nutritif, et en connaissant la teneur du miellat en glucides, on peut faire une approximation de la concentration du saccharose dans la sève élaborée du phloème ingérée par les aphides.

One possible control strategy against the olive fly, Bactrocera oleae, the most serious olive crop pest, is the Sterile Insect Technique (SIT) application. However, a number of problems associated with this method remain that decrease the effectiveness of SIT, including the quality of reared insects. Taking the importance of the relationship between the olive fly and bacteria into consideration, the effects of probiotic diets enriched with Pseudomonas putida on B. oleae longevity and fecundity were evaluated. First, we found that the probiotic bacterium, P. putida, is conveyed from diets to the oesophageal bulb as well as to the fly midgut after feeding on the probiotic diet. Subsequently, B. oleae adults fed on either: (a) a standard full protein and sugar diet; (b) a sugar only diet; (c) a probiotic standard full protein and sugar diet; or (d) a probiotic sugar diet. Flies fed on probiotic diets were supplied with an inoculated gel containing P. putida; non‐inoculated gel was provided to the flies fed on non‐probiotic diets. B. oleae males and females that fed on sugar diets did not survive as long as those that fed on protein diets. A comparison of the longevity of adults fed on full diet and sugar with their respective probiotic diets revealed no significant difference. Males fed on the sugar only diet survived longer than males fed on probiotic sugar diet, and females fed on the full protein and sugar diet survived much longer than females fed on the full probiotic diet. As regarding fecundity, both full diets resulted in a higher number of eggs laid per female. Females fed on the probiotic sugar diet laid a higher number of eggs than females that fed on sugar only. The inoculated gel of the probiotic sugar diet contained a significantly higher quantity of leucine, isoleucine and proline than the non‐inoculated gel of the sugar only diet. The possible role of dietary bacteria in relation to functional aspects of olive fly physiology is discussed.  相似文献
