A total of 748 Salmonella strains (97 serovars) isolated from human (291), animal (119), environmental (141), food (102) and animal feed (95) sources were examined for resistance to 9 antimicrobial agents. Most of the human isolates were from hospitalized patients (282). An overall resistance rate of 98.8% was determined with 100% for human and environmental isolates. Resistance to sulfadiazine (87.7%) was most common, followed by streptomycin (61.2%), ampicillin (39%) and trimethoprim-sulphamethoxazole (37.9%). Fifty one different resistance patterns were identified with Su (164 strains), Su-Sm (122) and Su-Sm-Tc-Cm-Km-Ap-Nx-Gm-Tm (95) predominating, the latter occurring only in human isolates. Multiple resistance was most frequently found among human isolates, particularly in S. derby and S. typhimurium strains. The relationship between antibiotic resistance, serovar and source of isolation of the Salmonella strains is discussed.  相似文献

Thirty seven strains of the Aspergillus flavus group isolated from animal mixed feeds have been screened for their ability to produce aflatoxins in yeast extract and sucrose (YES), aflatoxin producing ability (APA), and coconut agar medium (CAM) media. The concentration and detection of the aflatoxins by different methods is compared. Five known aflatoxin-positive and one aflatoxin-negative strains have been used as controls. Only 5 out of the 37 strains (13.5%) were aflatoxin-producers in YES medium. Of these five strains and the five known aflatoxin-positive strains, only three showed blue fluorescence in APA medium and four in CAM medium. Generally, the aflatoxin concentration in CAM medium was higher than in YES and APA media. Using the agar-plug method and by direct spotting of the YES broth on TLC plates, some aflatoxin-producing strains were not detected.  相似文献

利用 DIG 末端标记技术和免疫组化技术分析了小鼠精子体外结合内化外源DNA的效率。试验结果表明,不同小鼠个体的精子结合外源DNA的阳性率有明显差异(P<0.01),平均为13%。利用考马斯亮蓝染色评价了小鼠精子顶体反应发生的情况,筛选出TYH培养液为较合适的体外受精液。利用小鼠体外受精技术,将体外转染GFP基因并获能的小鼠精子与成熟卵母细胞进行体外受精,受精卵进行体外培养,表达GFP胎的阳性率为4.7%。验证了精子介导制备转基因小鼠胚胎的可行性,并建立了利用精子载体法制备转基因小鼠胚胎的平台。  相似文献

单核细胞增生李斯特氏菌(Listeria monocytogenes)是重要的食源性致病菌,能引发人类的李斯特菌病,是全球公共卫生问题之一。该菌易感染孕妇,引起胎儿和新生儿的侵袭性李斯特菌病,严重威胁母婴健康。因此,建立有效的单增李斯特菌感染胎盘体内外模型,解析和探究单增李斯特菌经胎盘感染机制,是预防和控制单增李斯特菌感染母婴的关键所在。本文综述了可用于研究单增李斯特菌母婴感染的体内外胎盘模型,总结和讨论了各类模型的优势和局限性;并着重分析了体外三维胎盘屏障模型在单增李斯特菌感染方面的研究进展和未来研究方向。以期为深入解析该菌经胎盘感染的途径、发病机制提供支持,并为预防和控制母婴李斯特菌病提供科学参考。  相似文献

结核病(tuberculosis,TB)由结核分枝杆菌(Mycobacterium tuberculosis,MTB)感染所致,是严重危害患者身心健康的慢性传染病。目前TB的发病机制尚未完全阐明。因此建立恰当的动物模型有助于进一步探索TB发病机制、研发全新高效疫苗,优化治疗方案提供可靠的实验和理论依据。兔TB模型是应用较为广泛的动物模型之一。综述了兔TB模型构建特点及应用进展,从免疫机制和可行性角度深入分析兔TB模型的优势及其局限性,同时梳理了近年来国内外通过构建兔TB模型用于药物研究、疫苗研发和TB治疗方案评价等方面发挥的作用,为临床研究提供参考。  相似文献

Penicilliosis marneffei has emerged as an endemic systemic mycosis in Southeast Asia among humans and wild bamboo rats. To gain an insight into the epidemiology of this life-threatening disease, a survey of bamboo rats for natural infections byPenicillium marneffei was carried out in the central plains of Thailand during June-September, 1987. Thirty-one lesser bamboo rats (Cannomys badius) and eight hoary bamboo rats (Rhizomys pruinosus) were trapped. Portions of their internal organs were cultured to determine if they had been infected byP. marneffei. Six each ofC. badius (19.4%) andR. pruinosus (75%) yielded cultures of this unique, dimorphicPenicillium species. All of the isolates were readily converted to their unicellular form that multiplies by the process of schizogony by incubating them at 37 °C on plates of brain heart infusion agar. Their identity was further confirmed by a specific immunological test. Among the internal organs of the positive rats, the lungs had the highest positivity (83.3%), next in decreased order of frequency were the liver (33.3%) and the pancreas (33.3%). The use and value of domestic and wild animals in locating and demarcating endemic areas of geophilic fungal pathogens are discussed. Penicilliosis marneffei is considered to be a zooanthroponosis — a disease that occurs in lower animals, as well as, humans.  相似文献

A quantitative in vitro method for phototoxic evaluation of chemicals has been developed and validated. The assay uses Saccharomyces cerevisiae, seeded in an agar overlay on top of a plate count agar base. 8-Methoxy psoralen is used as a reference standard against which materials are measured. Activity is quantified by cytotoxicity measured as zones of inhibition. Several known phototoxins (heliotropine, lyral, phantolid, and bergamot oil) and photoallergens (6-methyl coumarin and musk ambrette) are used to validate the assay. An excellent correlation is observed between in vivo studies employing Hartley albino guinea pigs and the in vitro assay for several fragrance raw materials and other chemicals. The in vitro assay exhibits a greater sensitivity from 2–500 fold. For three fragrance oils, the in vitro assay detects low levels of photobiological activity while the in vivo assay is negative. Wild chimpanzees form temporary parties that vary in size and composition. Previous studies have revealed considerable intraspecific variation in party compositions. We examined patterns of association among age, sex, and reproductive classes of chimpanzees at Ngogo in the Kibale National Park, Uganda. We employed a class-based association index and a randomization procedure to control for confounding factors and to test for differences between classes. Results indicate that males associated with other males significantly more than expected if all classes behaved equivalently, while females generally associated with individuals of the same sex less than expected. To interpret these patterns we used two additional indices that separate associations into two components: general gregariousness and preference for particular classes of associates. Males and estrous females were more gregarious than other classes, while anestrous females were less so. After controlling for general gregariousness, adult males as a class showed no specific preference for associating with each other. Anestrous females preferred each other as party members, and estrous females avoided each other. These results are consistent with previous findings that adult males are more gregarious than females. They diverge from the standard picture of chimpanzee society, however, by suggesting a mutual affinity among anestrous females, but not among adult males as a class.  相似文献

Twelve of 30 species examined in the ant genus Polyrhachis carry single nucleotide insertions at one or two positions within the mitochondrial cytochrome b (cytb) gene. Two of the sites are present in more than one species. Nucleotide substitutions in taxa carrying insertions show the strong codon position bias expected of functional protein coding genes, with substitutions concentrated in the third positions of the original reading frame. This pattern of evolution of the sequences strongly suggests that they are functional cytb sequences. This result is not the first report of +1 frameshift insertions in animal mitochondrial genes. A similar site was discovered in vertebrates, where single nucleotide frameshift insertions in many birds and a turtle were reported by Mindell et al. (Mol Biol Evol 15:1568, 1998). They hypothesized that the genes are correctly decoded by a programmed frameshift during translation. The discovery of four additional sites gives us the opportunity to look for common features that may explain how programmed frameshifts can arise. The common feature appears to be the presence of two consecutive rare codons at the insertion site. We hypothesize that the second of these codons is not efficiently translated, causing a pause in the translation process. During the stall the weak wobble pairing of the tRNA bound in the peptidyl site of the ribosome, together with an exact Watson–Crick codon–anticodon pairing in the +1 position, allows translation to continue in the +1 reading frame. The result of these events is an adequate level of translation of a full-length and fully functional protein. A model is presented for decoding of these mitochondrial genes, consistent with known features of programmed translational frameshifting in the yeast TY1 and TY3 retrotransposons.Reviewing Editor: Dr. W. Interactions between herbivores and seed predators may have long-term consequences for plant populations that rely on persistent seed banks for recovery after unpredictable fires. We assessed the effects of browsing by deer and seed predation by rodents, ants and birds on the densities of seeds entering the seed bank of Ceanothus cuneatus var. rigidus, a maritime chaparral shrub in coastal California. Ceanothus produced many more seeds when protected from browsers in long-term experimental exclosures than did browsed plants, but the seed densities in the soil beneath browsed and unbrowsed Ceanothus were the same at the start of an intensive one-year study. The density of seeds in the soil initially increased in both treatments following summer seed drop: while densities returned to pre-drop levels within a few weeks under browsed plants, soil seed densities remained high for 5–8 months beneath unbrowsed plants. Rodent abundance (especially deer mice) was higher near unbrowsed plants than >30 m away, and rodents removed Ceanothus seeds from dishes in the experimental plots. At least in the short term, rodent density and rates of seed removal were inversely related to the intensity of browsing. 土壤动物是土壤生态系统中十分活跃的生物类群之一,也是城市草坪生态系统的重要组成部分。由于外来植物白三叶入侵,草坪土壤动物可能受到直接或间接的影响,进而改变城市草坪生态系统功能及过程。研究白三叶入侵对城市草坪生态系统土壤动物的影响,可为进一步了解外来植物入侵机制及城市草坪的建植养护提供理论依据。采用野外调查的试验方法对4种不同入侵程度下城市草坪的土壤动物群落特征和土壤理化性质进行了研究。结果如下:4种不同入侵程度的草坪样地共捕获土壤动物30099只,隶属于19目43科,其中小杆科、线蚓科和跳虫科为优势类群;土壤动物个体数量和类群数量整体表现为中度入侵 > 轻度入侵 > 对照 > 重度入侵,符合中度干扰假说;土壤动物个体数量和类群数量的季节动态主要表现为夏秋较高,冬春较低。CCA分析显示,土壤理化因子中铵态N对土壤动物影响相对较小,可能与白三叶改善了草坪土壤的供N水平并使其不成为土壤动物分布的限制因子有关;同时,土壤理化因子对土壤动物优势类群影响较小,而主要影响常见和稀有类群。总体而言,一定程度的白三叶入侵增加了土壤动物群落的多样性,但当达到重度入侵时,白三叶形成单优群落,减少了植物多样性,进而使土壤动物可获得的生活资源减少,土壤动物群落多样性呈下降趋势;白三叶入侵后通过改变土壤理化性质,影响到土壤动物常见和稀有类群,并最终改变草坪土壤动物的群落结构。  相似文献

Soil samples from seventeen animal herds (camels, goats, sheep, cows) were surveyed for the occurrence of keratinophilic fungi. Twenty four species related to eleven genera were recovered on defated wool baits. Eleven Chrysosporium species were reported in the following order of dominance: C. keratinophilum, C. tropicum, C. indicum, C. queenslandicum, C. pannicola, C. carmichaelii, C. state of Arthroderma curreyi, C. zonatum, C. state of A. cuniculi and Chrysosporium state of Renispora flavissima. The Aphanoascus teleomorph of C. keratinophilum, C. indicum and C. tropicum were frequently reported in soils from cow and sheep folds. Scopulariopsis, Cephaliophora and Sepedonium, although not keratinolytic, were also recorded on wool baits of soils from cow herds which are slightly acidic. The frequency and distribution of these fungi are discussed in relation to animal species and the general ecological conditions of desert soils.  相似文献

During the 1989 corn harvest season, numerous reports of equine leukoencephalomalacia (ELEM) outbreaks and a pulmonary edema (PPE) syndrome in swine from several regions of the United States were received by the National Veterinary Services Laboratories (NVSL), Ames, Iowa. Previous and concurrent research linked Fusarium moniliforme and fumonisin-contaminated feeds to both diseases. Chemical and mycological investigations revealed fumonisin B1 (FB1) concentrations of 20 to 360 ppm in suspect swine feeds and 8 to 117 ppm in suspect equine feeds. Nonproblem feeds contained concentrations below 8 ppm. Fusarium moniliforme and Fusarium proliferatum were isolated from both problem and nonproblem equine and swine feeds. When cultured on autoclaved corn, the F. moniliforme and F. proliferatum isolates produced respective FB1 and fumonisin B2 (FB2) that range from less than 5 to more than 2450 ppm and less than 5 to more than 1000 ppm, respectively. Isolates from both problem and nonproblem feeds produced high levels (greater than 500 ppm) in culture. Reported here is a review of chemical and mycological data resulting from the study of several cases of PPE and ELEM.  相似文献

Despite years of investigation, pathogenesis of necrotizing enterocolitis (NEC) remains elusive. Bacterial metabolites were implicated by several authors but their roles remain controversial. The aim of our study was to investigate the role of SCFAs and polyamines through a kinetic study of histological and macroscopical digestive lesions in monobiotic quails. Germ-free quails, inoculated with a Clostridium butyricum strain involved in a NEC case, were fed or not with a diet including lactose (7%). Quails were sacrificed at various times between D7 and D24 after bacterial inoculation. NEC-like lesions, i.e. thickening, pneumatosis, and hemorrhages, occurred only in lactose-fed quails and increased with time. The main histological characteristics were infiltrates of mononuclear cells, then heterophilic cells, then gas cyst and necrosis. The first event observed, before histological and macroscopical lesions, is a high production of butyric acid, which precedes an increase of iNOS gene expression. No difference in polyamines contents depending on the diet was observed. These results show the major role of butyric acid produced by commensal bacteria in the onset of the digestive lesions.  相似文献

大别山五针松(Pinus dabeshanensis)为我国特有树种,其天然植株较少且分布范围狭窄,种群更新困难,已被列为国家二级重点保护野生植物。分别于2015年和2016年的10—12月,在目前已知最大天然种群所在地安徽岳西县河图镇大王沟,研究了大别山五针松球果与种子特征、种子雨与土壤种子库、动物对种子的取食和搬运,幼苗分布格局及其与鼠穴分布的关系,以期探明动物在其天然更新中的作用,分析其天然更新不力的原因。结果显示:大别山五针松种子败育率较高,阳坡个体球果发育情况好于阴坡;种子成熟期间没有明显的种子雨,土壤种子库也未调查到完整种子。共记录到母树与球果的访问动物6目11科16种,其中7种动物确定取食种子;地面种子摆放实验显示超过95%以上的种子被啮齿动物捕食或搬运至他处取食或贮藏,不同的摆放处理对啮齿动物的捕食、搬运没有显著影响;小林姬鼠(Apodemus sylvaticus)盗食现象严重,埋藏实验中人工贮点当夜被发现的概率在90%左右,岩松鼠(Sciurotamias davidianus)和小林姬鼠是其种子主要捕食者。研究地的大别山五针松种群于2015年和2016年分别新增一年生幼苗5株和7株,这表明大别山五针松在当地存在天然更新。幼苗多单独生长在母树周围,点格局分析显示在0—0.6 m的尺度范围内呈随机分布;在0.6 m尺度呈聚集分布。大别山五针松幼苗在小林姬鼠巢穴周边分布,其更新格局受到小林姬鼠贮点位置分布情况影响,小林姬鼠极有很可能为大别山五针松的传播者,啮齿动物对大别山五针松种子的捕食与搬运影响了大别山五针松的天然种群更新。  相似文献

The influence of selected steroids on the in vitro generative development of Arabidopsis thalianawas investigated. The activity of the animal steroids androsterone, androstenedione, progesterone, estrone, estriol, and 17-estradiol was compared to 24-epibrassinolide, a member of the regulatory family of brassinosteroids. A. thaliana plants were cultured in vitro in media containing these steroids. The stimulatory effect of the tested substances was evaluated by measurement of the percentage of generative plants versus vegetative plants in the experimental group. It was established that androstenedione, the main testosterone precursor, and androsterone, a typical male hormone, were more effective in stimulating flowering in A. thaliana than the female hormones, estrogens and progesterone. Androsterone at a concentration of 0.1 M increased the percentage of generative plants up to 96% (control 41%). Estrogens at the same concentration decreased the number of generative plants and 24-epibrassinolide did not stimulate A. thalianagenerative development.  相似文献
