Summary As a foundation for studies on directional intercellular communication and its regulation in apical development, the network of plasmodesmata inArabidopsis root apical meristems was characterized by quantitative electron microscopy and dye-coupling analysis, using symplasmic probes, and real-time imaging in confocal laser scanning microscopy. A tissue-specific plasmodesmatal network, which interconnected the cells in the root apical meristem, was characterized by the following features, (a) Plasmodesmatal distribution and density were found to be tissue-specific, (b) Primary and secondary plasmodesmata were differentially grouped and regulated. Primary plasmodesmata were formed in large numbers in the transverse walls of each tissue, and were subject to deletion during cell differentiation. Secondary plasmodesmata were mostly distributed in longitudinal walls between cell files and common walls between neighboring tissues; they also provided a symplasmic path between different initial tiers in the meristem. Small fluorescent tracers moved through the plasmodesmatal network of the root apical meristem in two distinct phases. At low concentrations molecules trafficked in a non-tissue-specific manner, whereas at higher concentrations, their distribution reflected the presence of tissue-specific movement consistent with plasmodesmatal distribution. These findings are discussed in terms of the role of tissue-specific plasmodesmatal domains in the control of root development.  相似文献

The patterns of resource translocation among interconnected ramets within a clone may show diversity. In order to understand the pattern of inter-ramet water translocation in natural clones of the rhizomatous shrub Hedysarum laeve (Leguminosae), which frequently dominates inland dunes in semi-arid areas of China, acid fuchsin dye application combined with spectrometric analysis was employed. Dye solutions were fed to the base of plants (near the parent ramet) and to the apical portions (near one of the youngest shoots) and the amount of dye in shoots and rhizomes was measured spectrometrically after 24 h. The total amount of dye translocated from the base in the acropetal direction was approximately four times as high as that of basipetal translocation from cut sections near the apex. Most of the dye was detected in the shoots with very little found in the rhizomes. The average percentage allocation of dye from donor ramet transferred to recipient ramets in the ramet hierarchy closer, due to acropetal and/or basipetal translocation, was significantly greater than those in the ramet hierarchy less close to it. The ramet-hierarchy order had a great influence on dye translocation in either the acropetal or basipetal direction. It is inferred that the donor ramet would transport water towards its recipient ramets in different ramet hierarchies in unequal proportions in this species.  相似文献

 The spatial pattern of acropetal and basipetal cytoplasmic streaming velocities has been studied by laser-Doppler-velocimetry (LDV) in the positively gravitropic (downward growing) rhizoids of Chara globularis Thuill. and for the first time in the negatively gravitropic (upward growing) protonemata. The LDV method proved to be precise and yielded reproducible results even when tiny differences in velocities were measured. In the apical parts of the streaming regions of both cell types, acropetal streaming was faster than basipetal streaming. Starting at the apical reversal point of streaming, the velocity increased basipetally with the distance from that point and became fairly constant close to the basal reversal point; subsequently, the velocity decreased slightly acropetally as the apical reversal point was again approached. There was no change in velocity at the basal reversal point. However, at the apical reversal point there was an abrupt decrease in velocity. The pattern of the ratio of acropetal to basipetal streaming velocity (VR) was a function of the relative distance of the site of measurement from the apical reversal point rather than a function of the absolute distance. Upon inversion of the rhizoids, the VR decreased on average by 3.8% (±0.4%), indicating that the effect of gravity on the streaming velocity was merely physical and without a physiological amplification. Rhizoids that had developed on the slowly rotating horizontal axis of a clinostat, and had never experienced a constant gravity vector, were similar to normally grown rhizoids with respect to VR pattern. In protonemata, the VR pattern was not significantly different from that in rhizoids although the direction of growth was inverse. In rhizoids, oryzalin caused the polar organization of the cell to disappear and nullified the differences in streaming velocities, and cytochalasin D decreased the velocity of basipetal streaming slightly more than that of acropetal streaming. Cyclopiazonic acid, known as an inhibitor of the Ca2+-ATPase of the endoplasmic reticulum, also reduced the streaming velocities in rhizoids, but had slightly more effect on the acropetal stream. It is possible that the endogenous difference in streaming velocities in both rhizoids and protonemata is caused by differences in the cytoskeletal organization of the opposing streams and/or loading of inhibitors (like Ca2+) from the apical/subapical zone into the basipetally streaming endoplasm. Plasmodesmatal gating in epidermal cells of Nicotianatabacum was examined in expanding infection sites of tobacco mosaic virus (TMV) expressing a fusion between the viral movement protein and the green fluorescent protein (MP-GFP). The infection sites were circular in profile and within 3 days post-inoculation had developed a brightly fluorescent leading edge, giving them a characteristic ‘halo’ shape. Co-localization of MP-GFP with callose demonstrated that nearly all epidermal cell plasmodesmata were targeted with MP-GFP. The fusion protein was located in the centre of the plasmodesmal pore, between paired callose platelets. Increase in plasmodesmatal size exclusion limit, as determined by the passage of microinjected 10 kDa Texas Red dextran, was restricted predominantly to cells within the fluorescent halo, and was virtually absent from cells in the centre of the expanding infection site. The plasmodesmata of these cells, however, remained fluorescently labelled with MP-GFP. Injections outside the fluorescent infection site failed to show movement of dextran, while dextran injected into cells at the leading edge moved inwards towards the centre of the lesion but not outwards into cells lacking GFP. Leaf incisions through cells ahead of the infection front halted the advance of the virus, indicating that virus replication was absent in non-fluorescent cells outside the infection site. The data provide the first demonstration that within an expanding infection site plasmodesmatal gating is under temporal control.  相似文献

Plasmodesmata were counted in the longitudinal and transverse walls in developmental sequences of merophytes in roots of Azolla pinnata R.Br. The differences between certain categories of longitudinal wall were traced to factors that govern the surface area of the cell plates, the density of plasmodesmata (number per unit area of cell plate), and the amount by which each type of plate expands. No evidence for secondary augmentation of plasmodesmatal numbers after the cell-plate stage of development was found, but plasmodesmata are lost from the walls of sieve and xylem elements during their differentiation. Losses caused by cell separation occur in other tissues. The relatively high density of plasmodesmata in transverse walls is based not so much on a high density in the cell plates as on the relatively low expansion that these walls undergo. There appears to be a compensatory mechanism that relates initial plasmodesmatal density to the future expansion of the cell plate. The root shows determinate growth, the apical cell dividing about 55 times. Beginning at about the 35th division there is a progressive failure to maintain the plasmodesmatal frequencies that were developed in earlier cell divisions in the apical cell. The divisions that occur within the later-produced merophytes also show progressive diminution of plasmodesmatal numbers. The result is that the apex of the root, and particularly the apical cell, becomes more and more isolated symplastically, a phenomenon which could account for its limited lifespan and the determinate growth pattern of the root.  相似文献

Summary Cell development in the root apical meristem is thought to be regulated by position-dependent information, but as yet, the underlying mechanism for this remains unknown. In order to examine the potential involvement of the symplasmic transmission of positional signals, plasmodesmatal frequency and distribution was quantitatively analyzed in root apical meristem cell walls ofArabidopsis thaliana during root development. A consistent distribution pattern of plasmodesmata was observed in the root apex over four weeks. While cells within initial tiers were uniformly interconnected, more symplasmic connections between the initial tiers and their immature-cell (primary-meristem) derivatives were observed than within the initial tiers. Immature cells were connected across transverse walls by primary plasmodesmata according to a tissue-specific pattern. Cells of the immature vascular tissue and cortex had the highest plasmodesmatal frequencies, followed by the immature epidermis and root cap. Although the numbers of plasmodesmata in transverse walls (primary plasmodesmata) was reduced in all tissues as the root aged, the tissue-specific distribution remained constant. The extent of symplasmic coupling across the boundaries of each tissue appeared to be limited by fewer secondary plasmodesmata in longitudinal walls. The frequency of all plasmodesmata decreased as the root aged. The primary plasmodesmata within each tissue increased at one week and then dramatically decreased with root age; the frequency of secondary plasmodesmata in longitudinal walls also decreased, but more gradually. These findings are discussed with respect to the roles likely played by plasmodesmata in facilitating transport of position-dependent information during root development.  相似文献

Summary Movement of both [3H]GA1 and [14C]GA3 through root segments from P. coccineus seedlings was basipetally polarised. The basipetal/acropetal ratio of radioactivity from [3H]GA1 in agar receiver blocks was 9.2 for apical, elongating segments, and 4.0 for more basal, non-elongating segments. Polarity of gibberellin transport was restricted to the stele, and absent from cortical tissues. Transport of [14C]IAA through root segments to agar receivers was preferentially acropetal, particularly so in the stele. Despite the existence of basipetal polarity of gibberellin transport in the root, [3H]GA1 injected into cotyledons moved into and acropetally along the seedling root.  相似文献

In stem sections of lentil seedlings, there is a typical polar movement of IAA labelled with 14C. The degree of polarity, expressed as the ratio of basipetal to acropetal transport, was (25°C) 7.6. A decrease (from 25° to 15°C) and an increase (from 25° to 30°C) of temperature cause a reduction of the IAA uptake by the sections and a decrease of both the basipetal and the acropetal translocation of IAA. Results suggest that the basipetal as well as the acropetal movement of auxin, are dependent of a metabolical component which is discussed.  相似文献

 The frequency of plasmodesmata increases in the shoot apical meristem of plants of Sinapis alba L. induced to flower by exposure to a single long day. This increase is observed within all cell layers (L1, L2, L3) as well as at the interfaces between these layers, and it occurs in both the central and peripheral zones of the shoot apical meristem. The extra plasmodesmata are formed only transiently, from 28 to 48 h after the start of the long day, and acropetally since they are detectable in L3 4 h before they are seen in L1 and L2. These observations indicate that (i) in the Sinapis shoot apical meristem at floral transition, there is an unfolding of a single field with increased plasmodesmatal connectivity, and (ii) this event is an early effect of the arrival at this meristem of the floral stimulus of leaf origin. Since (i) the wave of increased frequency of plasmodesmata is 12 h later than the wave of increased mitotic frequency (A. Jacqmard et al. 1998, Plant cell proliferation and its regulation in growth and development, pp. 67–78; Wiley), and (ii) the increase in frequency of plasmodesmata is observed in all cell walls, including in walls not deriving from recent divisions (periclinal walls separating the cell layers), it is concluded that the extra plasmodesmata seen at floral transition do not arise in the forming cell plate during mitosis and are thus of secondary origin. Fern (Onoclea sensibilis) gametophytes when grown in the dark form a linear file of cells (one-dimensional) called a protonema. In the light two-dimensional growth occurs which results in a heart-shaped prothallus one cell thick. The objective of this paper is to relate the most common pattern of cell division observed in developing gametophytes to the formation of the plasmodesmatal network. Since the prothalli are only two dimensional, we can easily determine from thin sections the total number and the density (number per unit surface area) of plasmodesmata at each developmental stage. As the prothallus grows the number of plasmodesmata increases 50-fold in the apical or meristematic cell. This number eventually reaches a plateau even though the density continues to increase with each new cell division. What is particularly striking is that both the number and density of plasmodesmata between adjacent cells is precisely determined. Furthermore, the pattern of plasmodesmata distribution is predictable so that (1) we can identify the apical meristematic cells by their plasmodesmata number, or density, as well as by their size, shape and location, (2) we can predict, again from plasmodesmata number, the location of a future wall of the apical cell prior to its actual formation, (3) we can show that the density of plasmodesmata in the triangular apical cell of the prothallus (14 plasmodesmata microns-2) is comparable to those reported for secretory glands which are known to have high rates of plasmodesmatal transport and (4) we can show that once the plasmodesmata have been formed during division, no subsequent change in the number of plasmodesmata occurs following cell plate formation.  相似文献

Summary The actin cytoskeleton is involved in the positioning of statoliths in tip growingChara rhizoids. The balance between the acropetally acting gravity force and the basipetally acting net out-come of cytoskeletal force results in the dynamically stable position of the statoliths 10–30 m above the cell tip. A change of the direction and/or the amount of one of these forces in a vertically growing rhizoid results in a dislocation of statoliths. Centrifugation was used as a tool to study the characteristics of the interaction between statoliths and microfilaments (MFs). Acropetal and basipetal accelerations up to 6.5 g were applied with the newly constructed slow-rotating-centrifuge-microscope (NIZEMI). Higher accelerations were applied by means of a conventional centrifuge, namely acropetally 10–200 g and basipetally 10–70 g. During acropetal accelerations (1.4–6 g), statoliths were displaced to a new stable position nearer to the cell vertex (12–6.5 m distance to the apical cell wall, respectively), but they did not sediment on the apical cell wall. The original position of the statoliths was reestablished within 30 s after centrifugation. Sedimentation of statoliths and reduction of the growth rates of the rhizoids were observed during acropetal accelerations higher than 50 g. When not only the amount but also the direction of the acceleration were changed in comparison to the natural condition, i.e., during basipetal accelerations (1.0–6.5 g), statoliths were displaced into the subapical zone (up to 90 m distance to the apical cell wall); after 15–20 min the retransport of statoliths to the apex against the direction of acceleration started. Finally, the natural position in the tip was reestablished against the direction of continuous centrifugation. Retransport was observed during accelerations up to 70 g. Under the 1 g condition that followed the retransported statoliths showed an up to 5-fold increase in sedimentation time onto the lateral cell wall when placed horizontally. During basipetal centrifugations 70 g all statoliths entered the basal vacuolar part of the rhizoid where they were cotransported in the streaming cytoplasm. Summary Using radioactive phenylalanine as a tracer, bidirectional translocation in the phloem of a single stem bundle of Vicia faba L. was detected by analyses of microautoradiographs. Bidirectional translocation occurred in lateral leaf traces subtending a leaf of a certain developmental stage. Electron microscopic observation revealed that poplar (Populus deltoides Marsh.) and winter wheat (Triticum aestivum L. cv. Seward 80004) plasmodesmatal structures significantly changed under short day (SD, 8 h light) and in winter period, and such changes differed also noticeably between these two woody and herbaceous plants. Under long day (LD, 16 h light), many plasmodesmata with strong stain appeared in the cell wall of both poplar apical buds and winter wheat young leaf tissues, and connections of cytoplasmic endoplasmic reticulum (ER) with the ER in some plasmodesmata were observed. In addition, the typical “neck type” plasmodesmata were observed in winter wheat young leaf tissues, and their central desmotubules (appressed-ER) could be clearly identified. Under SD, many poplar plasmodesmata showed only a partial structure in the cell wall and appeared to be discontinued; some plasmodesmata swelled in the mid-wall, forming the cavity, and no appressed-ER appeared. In winter wheat, however, no noticeable alterations of plasmodesmata occurred, and the plasmodesmatal structure essentially remained same as it was under LD. In winter period, poplar plasmodesmata had a similar morphology as those observed under SD, however, winter wheat manifested at least two types of significant plasmodesmatal alterations: one plugged by electron-dense materials and the other of reduced neck region compared to those under LD. The above dynamic difference of the two species plasmodesmata under SD and winter period revealed the difference of their dormancy development under those environmental conditions.  相似文献

Summary There is a predictable and well defined variation in numbers of plasmodesmata in roots ofAzolla. As the apical cell of the root ages, it lays down walls with progressively fewer plasmodesmata, thereby gradually cutting itself off from the rest of the root (Gunning 1978). Electrical coupling was examined between the apical cell and an adjacent merophyte in roots of various lengths. The apical cell becomes increasingly electrically isolated from the rest of the root as it ages. Electrical coupling is strongly correlated with the number of the plasmodesmata between the coupled cells. The resistance of a plasmodesma, as estimated from equivalent electrical circuits, was 150–600 times more resistive than a value based on theoretical considerations. No evidence was found for a change in the physiology of plasmodesmata as the root ages. Coupling experiments, both on root hairs and at the apex, gave some suggestion that plasmodesmata may be less resistive towards the apical cell than away from it.  相似文献

A technique for the preparation of plasmodesmata within a cell wall fraction of Solanum nigrum tissue homogenates has been developed, featuring good ultrastructural preservation of plasmodesmatal structure. SDS-PAGE of protein extracts of this plasmodesmata-containing cell wall fraction revealed the distinct enrichment of two bands, with molecular weights of 28 and 43 kD.  相似文献

Abstract

Exposure of Zea mays seedlings to a continuous electromagnetic field (EMF) for 30 h induced a 30% stimulation in the rate of root elongation compared with the controls. It also resulted in a significant increase of cell expansion, in both the acropetal (metaxylem cell lineage) and basipetal (root cap cells) direction. In addition, in EMF-exposed roots a precocious structural disorder was observed both in differentiating metaxylem cells and root cap cells. All these features may be consistent with an advanced differentiation of root cells that are programmed to die. EMF treatment also resulted in a significant reduction in the size of the quiescent centre in the root apical meristem. The extent to which these responses are causally linked is discussed.  相似文献

The role of actin filaments in regulating plasmodesmal transport has been studied by microinjection experiments in mesophyll cells of tobacco (Nicotiana tabacum L. cv. Samsun). When fluorescent dextrans of various molecular sizes were each co-injected with specific actin filament perturbants cytochalasin D (CD) or profilin into these cells, dextrans up to 20 kilodalton (kDa) moved from the injected cell into surrounding cells within 3–5 min. In contrast, when such dextrans were injected alone or co-injected with phalloidin into the mesophyll cells, they remained in the injected cells. Phalloidin co-injection slowed down or even inhibited CD- or profilin-elicited dextran cell-to-cell movement. Dextrans of 40 kDa or larger were unable to move out of the injected cell in the presence of CD or profilin. These data suggest that actin filaments may participate in the regulation of plasmodesmal transport by controlling the permeability of plasmodesmata.  相似文献

Summary Velocities of cytoplasmic streaming were measured in internodal cells ofNitella flexilis L. andChara corallina Klein ex Willd. by laser-Doppler-velocimetry to investigate the possibility of non-statolith-based perception of gravity. This was recently proposed, based on a report of gravity-dependent polarity of cytoplasmic streaming. Our measurements revealed large spatial and temporal variation in streaming velocity within a cell, independent of the position of the cell with respect to the direction of gravity. In 58% of the horizontally positioned cells the velocities of acropetal and basipetal streaming, measured at opposite locations in the cell, differed significantly. In 45% of these, basipetal streaming was faster than acropetal streaming. In 60% of the vertically positioned cells however the difference was significant, downward streaming was faster in only 61% of these. When cell positions were changed from vertical to horizontal and vice versa the cells reacted variably. A significant difference between velocities in one direction, before and after the change, was observed in approx. 70% of the measurements, but the velocity was faster in the downward direction, as the second position, in only 70% of the significantly different. The ratio of basipetal to acropetal streaming velocities at opposite locations of a cell was quite variable within groups of cells with a particular orientation (horizontal, normal vertical, inverted vertical). On average, however, the ratio was close to 1.00 in the horizontal position and approx. 1.03 in the normal vertical position (basipetal streaming directed downwards), which indicates a small direct effect of gravity on streaming velocity. Individual cells, however, showed an increased, as well as a decreased, ratio when moved from the horizontal to the vertical position. No discernible effect of media (either Ca2 +-buffered medium or 1.2% agar in distilled water) on the streaming velocities was observed. Summary The basipetal movement of IAA in 5-mm Zea coleoptile segments is drastically reduced under anaerobic conditions, but it remains greater than acropetal movement which is closely similar in the presence and absence of oxygen. The polarity of IAA movement has thus been confirmed in Zea coleoptile segments which have been deprived of oxygen. This net polar flux is dependent upon anaerobic metabolism since it is abolished in the presence of the metabolic inhibitiors sodium fluoride and iodoacetic acid.Acropetal movement of IAA is unaffected by the presence of sodium fluoride in air or anaerobic conditions. Uptake of IAA from a basal donor is not affected by sodium fluoride in air, but under anaerobic conditions the inhibitor decreased uptake by approximately 13%.Under anaerobic conditions both inhibitors reduce basipetal movement of IAA to the level of acropetal movement, and both decrease the total uptake of IAA from an apical donor by up to 30–45%. Under aerobic conditions sodium fluoride has no marked effect upon either the uptake of IAA from an apical donor or the basipetal movement of IAA by the segments. On the other hand, iodoacetic acid greatly decreased the uptake of IAA by the segments in air, but the same fraction of the total IAA taken up was recovered in the receiving block in the presence and absence of the inhibitor.This research was supported by Grant Number 83/6 to Professor M. Zusammenfassung Natürliche und synthetische Abscisinsäure wird in Explantaten, Blattstiel- und Internodialsegmenten von Coleus rheneltianus transportiert.In jüngeren Pflanzenteilen erfolgt der Transport vorwiegend oder ausschließlich basipetal, während in älteren Organteilen auch eine erhebliche akropetale Wanderung stattfindet. Eine Bestimmung der Transportgeschwindigkeit ergab für synthetische Abscisinsäure in älteren Blattstielsegmenten ungefähr 24–36 mm/h.
Transport of abscisic acid in explants, petiole and internode segments of Coleus rheneltianus
Summary Synthetic or natural abscisic acid isolated from tomato fruits is transported in explants, petiole and internode segments from Coleus rheneltianus.Transport in segments and explants from younger parts (2nd node or internode) is mainly basipetal. In older parts (segments and explants from 5th nodes or internodes) translocation in acropetal direction is nearly of the same order of magnitude as in basipetal direction.The velocity of transport of synthetic absisic acid was investigated in petiole segments from 5th nodes and was found to be approximately 24 to 36 mm/h.
