The design and synthesis of a series of C28 amine-based betulinic acid derivatives as HIV-1 maturation inhibitors is described. This series represents a continuation of efforts following on from previous studies of C-3 benzoic acid-substituted betulinic acid derivatives as HIV-1 maturation inhibitors (MIs) that were explored in the context of C-28 amide substituents. Compared to the C-28 amide series, the C-28 amine derivatives exhibited further improvements in HIV-1 inhibitory activity toward polymorphisms in the Gag polyprotein as well as improved activity in the presence of human serum. However, plasma exposure of basic amines following oral administration to rats was generally low, leading to a focus on moderating the basicity of the amine moiety distal from the triterpene core. The thiomorpholine dioxide (TMD) 20 emerged from this study as a compound with the optimal antiviral activity and an acceptable pharmacokinetic profile in the C-28 amine series. Compared to the C-28 amide 3, 20 offers a 2- to 4-fold improvement in potency towards the screening viruses, exhibits low shifts in the EC50 values toward the V370A and ΔV370 viruses in the presence of human serum or human serum albumin, and demonstrates improved potency towards the polymorphic T371A and V362I virus variants.  相似文献

Pastoral farmers in New Zealand have described dramatic demises in populations of the weed Cirsium arvense (L.) Scop., a perennial herb indigenous to Eurasia, following its mowing during rainfall. To test the hypotheses (1) that the mowing of C. arvense during rainfall increases the control of this weed and (2) that the causal organism in this ‘mowing-in-the-rain’ effect is the vascular wilt fungus Verticillium dahliae, two series of field experiments were carried out in C. arvense-infested pastures in New Zealand, one in autumn 2008 (Experiment series 1, 9 farms), and another in spring/summer 2008–2009 (Experiment series 2, 12 farms). The effect of mowing in the rain as compared to mowing in the dry was to reduce the % ground cover of the thistle in the spring following treatment by 21 and 32% in Experiment series 1 and 2, respectively. Correlations of this ‘rain versus dry’ effect with the incidence of V. dahliae in the subterranean parts of C. arvense shoots sampled in each of the two field experiment series provided no statistical evidence that the effect increased with V. dahliae incidence. Thus these experiments provide no support for the hypothesis that V. dahliae is the biological mechanism for the ‘mowing-in-the-rain’ effect. Nevertheless, they do support mowing during rainfall as a simple and effective management tactic for C. arvense.  相似文献

1. Thirteen time series, varying from 17 to 27 years, of the abundance of Vespula germanica and V. vulgaris from lowland England are examined. The time series depend on either spring queens and workers taken in Malaise or suction traps, or collected colonies. 2. During the late 1970s and early 1980s, the abundance of V. germanica declined abruptly but that of V. vulgaris did not. 3. During the early 1980s, the 2‐year cycle of annual abundance of V. vulgaris changed to a nearly perfectly damped pattern of annual abundances. 4. The most likely factor causing these population changes was the increased use of pesticides acting directly by killing the wasps and indirectly by reducing their food resources. 5. The difference in response of the two species to increased pesticide use may be related to a difference in foraging ability.  相似文献

Herein we report the discovery of a novel series of vasopressin 1b (V1b) receptor antagonists, starting from potent but metabolically labile oxindole SSR149415. Masking the proline N,N-dimethyl amide moiety as an oxazole and attaching a benzylic amine moiety to the northern phenyl ring resulted in potent and selective V1b receptor antagonists with improved metabolic stability and improved pharmacokinetic properties in rat. Compound 18c was found to be efficacious in a rat model of anti-depressant activity.  相似文献

Maraviroc, an (HIV-1) entry inhibitor, binds to CCR5 and efficiently prevents R5 human immunodeficiency virus type 1 (HIV-1) from using CCR5 as a coreceptor for entry into CD4+ cells. However, HIV-1 can elude maraviroc by using the drug-bound form of CCR5 as a coreceptor. This property is known as noncompetitive resistance. HIV-1V3-M5 derived from HIV-1JR-FLan is a noncompetitive-resistant virus that contains five mutations (I304V/F312W/T314A/E317D/I318V) in the gp120 V3 loop alone. To obtain genetic and structural insights into maraviroc resistance in HIV-1, we performed here mutagenesis and computer-assisted structural study. A series of site-directed mutagenesis experiments demonstrated that combinations of V3 mutations are required for HIV-1JR-FLan to replicate in the presence of 1 µM maraviroc, and that a T199K mutation in the C2 region increases viral fitness in combination with V3 mutations. Molecular dynamic (MD) simulations of the gp120 outer domain V3 loop with or without the five mutations showed that the V3 mutations induced (i) changes in V3 configuration on the gp120 outer domain, (ii) reduction of an anti-parallel β-sheet in the V3 stem region, (iii) reduction in fluctuations of the V3 tip and stem regions, and (iv) a shift of the fluctuation site at the V3 base region. These results suggest that the HIV-1 gp120 V3 mutations that confer maraviroc resistance alter structure and dynamics of the V3 loop on the gp120 outer domain, and enable interactions between gp120 and the drug-bound form of CCR5.  相似文献

Herein we report the discovery of a novel oxindole-based series of vasopressin 1b (V1b) receptor antagonists. Introducing a substituted piperazine moiety and optimizing the southern and the northern aromatic rings resulted in potent, selective and brain penetrant V1b receptor antagonists. Compound 9c was found to be efficacious in a rat model of anti-depressant activity (3?mg/kg, ip). Interestingly, both moderate terminal half-life and moderate bioavailability could be achieved despite sub-optimal microsomal stability.  相似文献

A series of novel organoantimony(V) complexes have been synthesised by the reactions of the isomers of chlorophenylacetic acids with triphenylantimony(V) dichloride or tetraphenylantimony(V) bromide in 1:2 or 1:1 stoichiometries. All the complexes have been characterized by elemental analysis, IR and NMR (1H, 13C) spectra analyses; furthermore, complexes 1, 2, 3 and 4 have been determined by X-ray single crystal diffraction. The structure of complexes show that the five-coordinated and six-coordinated antimony(V) atoms adopt distorted trigonal bipyramidal geometry and octahedral geometry. And the structural analyses show that complexes 1 and 3 have 2D network structures; complex 2 possesses a 1D polymeric chain structure and complex 4 has a 3D supramolecular framework.  相似文献

The azetidinone LY307174 (1) was identified as a screening lead for the vasopressin V1a receptor (IC50 45 nM at the human V1a receptor) based on molecular similarity to ketoconazole (2), a known antagonist of the luteinizing hormone releasing hormone receptor. Structure-activity relationships for the series were explored to optimize receptor affinity and pharmacokinetic properties, resulting in compounds with Ki values <1nM and brain levels after oral dosing approximately 100-fold higher than receptor affinities.  相似文献

In the course of our investigations of vanadium-containing complexes for use as insulin-enhancing agents, we have generated a series of novel vanadium coordination complexes with bidentate ligands. Specifically we have focused on two ligands: anthranilate (anc), a natural metabolite of tryptophan, and imidizole-4-carboxylate (imc), meant to mimic naturally occurring N-donor ligands. For each ligand, we have generated a series of complexes containing the V(III), V(IV), and V(V) oxidation states. Each complex was investigated using phosphatase inhibition studies of three different phosphatases (acid, alkaline, and tyrosine (PTP1B) phosphatase) as prima facia evidence for potential use as an insulin-enhancing agent. Using p-nitrophenyl phosphate as an artificial phosphatase substrate, the levels of inhibition were determined by measuring the absorbance of the product at 405 nm using UV/vis spectroscopy. Under our experimental conditions, for instance, V(imc)3 appears to be as potent an inhibitor of alkaline phosphatase as sodium orthovanadate when comparing the Kcat/Km term. VO(anc)2 is as potent an inhibitor of acid phosphatase and tyrosine phosphatase as the Na3VO4. Thus, use of these complexes can increase our mechanistic understanding of the effects of vanadium in vivo.  相似文献

The V600E BRAF kinase mutation, which activates the downstream MAPK signaling pathway, commonly occurs in about 8% of all human malignancies and about 50% of all melanomas. In this study, we employed virtual screening and chemical synthesis to identify a series of N-(thiophen-2-yl) benzamide derivatives as potent BRAFV600E inhibitors. Structure–activity relationship studies of these derivatives revealed that compounds b40 and b47 are the two most potent BRAFV600E inhibitors in this series.  相似文献

The effects of breathing depth in attenuating induced bronchoconstriction were studied in 12 healthy subjects. On four separate, randomized occasions, the depth of a series of five breaths taken soon (approximately 1 min) after methacholine (MCh) inhalation was varied from spontaneous tidal volume to lung volumes terminating at approximately 80, approximately 90, and 100% of total lung capacity (TLC). Partial forced expiratory flow at 40% of control forced vital capacity (V(part)) and residual volume (RV) were measured at control and again at 2, 7, and 11 min after MCh. The decrease in V(part) and the increase in RV were significantly less when the depth of the five-breath series was progressively increased (P < 0.001), with a linear relationship. The attenuating effects of deep breaths of any amplitude were significantly greater on RV than V(part) (P < 0.01) and lasted as long as 11 min, despite a slight decrease with time when the end-inspiratory lung volume was 100% of TLC. In conclusion, in healthy subjects exposed to MCh, a series of breaths of different depth up to TLC caused a progressive and sustained attenuation of bronchoconstriction. The effects of the depth of the five-breath series were more evident on the RV than on V(part), likely due to the different mechanisms that regulate airway closure and expiratory flow limitation.  相似文献

Notorious oncogenic BRAF V600E plays a significant role in the signal transduction of the MAPK pathway, which is involved in tumor growth, especially in melanoma. Much effort has been made to suppress BRAF V600E through small molecules like vemurafenib and dabrafenib, but the MAPK pathway remains active through paradoxical activation, where CRAF transmits the signal of the MAPK pathway either alone or along with BRAF V600E. Therefore, we designed and synthesized a new series of N-(3-(3-alkyl-1H-pyrazol-5-yl) phenyl)-aryl amide/urea analogues that showed potent inhibitory activities against BRAF V600E and CRAF. Compound 7c exhibited particularly superior selectivity toward BRAF V600E and CRAF over 30 other protein kinases, implying that this chemotype could be investigated as a BRAF paradox breaker. © 2019 Elsevier Ltd. Background

Mating decreases female receptivity and terminates sex pheromone production in moths. Although significant progress has been made in elucidating the mating-regulated inactivation of pheromone biosynthesis-activating neuropeptide (PBAN) secretion, little is known about the mating induced gene expression profiles in pheromone glands (PGs). In this study, the associated genes involved in Bombyx mori mating were identified through digital gene expression (DGE) profiling and subsequent RNA interference (RNAi) to elucidate the molecular mechanisms underlying the mating-regulated gene expression in PGs.

Results

Eight DGE libraries were constructed from the PGs of mated and virgin females: 1 h mating (M1)/virgin (V1) PGs, 3 h mating (M3)/virgin (V3) PGs, 24 h mating (M24)/virgin (V24) PGs and 48 h mating (M48)/virgin (V48) PGs (M48 and V48). These libraries were used to investigate the gene expression profiles affected by mating. DGE profiling revealed a series of genes showing differential expression in each set of mated and virgin female samples, including immune-associated genes, sex pheromone synthesis-associated genes, juvenile hormone (JH) signal-associated genes, etc. Most interestingly, JH signal was found to be activated by mating. Application of the JH mimics, methoprene to the newly-emerged virgin females leaded to the significant reduction of sex pheromone production. RNAi-mediated knockdown of putative JH receptor gene, Methoprene tolerant 1 (Met1), in female pupa resulted in a significant decrease in sex pheromone production in mature females, suggesting the importance of JH in sex pheromone synthesis.

Conclusion

A series of differentially expressed genes in PGs in response to mating was identified. This study improves our understanding of the role of JH signaling on the mating-elicited termination of sex pheromone production.  相似文献

A series of novel 3,4,5,6-tetrahydro-1H-azepino[4,3,2-cd]indoles was synthesized and tested for vasopressin receptor antagonist activity. We identified compounds with high affinity for the human V2 receptor and good selectivity over the human V1a receptor. Compound 6c bound to V2 receptors with an IC(50) value of 20 nM, had >100-fold selectivity over V1a receptors, and inhibited cAMP formation in a cellular V2 functional assay with an IC(50) value of 70 nM.  相似文献

The action of [Co(X)(NO)2]2 (X = Cl, Br, L) on [V(H)(CO)6?nLn] (L = 1/ndi- and tritertiary phosphine; n = 2, 3) in thf yields [V(CO)5?n(NO)L2] and [V(NO)2(thf)4]X as the two main products. Thf is easilty replaced by other ligands L′, leading to the complexes cis-[V(NO)2(thf)4?nL′n]X, where n = 1 to 4. In the case of L′= CNR (R = Cy, iPr, tBu), the species [VX(NO)2L′3] are formed. The presence of X in the first coordination sphere is established by the normal halogen dependence (Cl < Br < I) of 51V shielding.δ(51V) values have been obtained for the two series of complexes and compared with δ of other nitrosylvanadium species, including [VX(NO)L′4]X. for [V(NO)2L′4]br, 51V shielding increases in the sequence {O} < {S} < NR3 < NCMe < AsEt3 < SbEt3 < PEt2Ph < P(OMe)3 < CNR, reflecting a general increase of shielding as the polarizability of the ligand function increases and its electronegativity decreases. Superimposed effects arising from electronic influences (PEtPh2) < PMe3 < P(OMe)3 and steric conditions (chelate-4 ring < 7 ring < 6 ring < 5 ring) are also discussed. Steric factors are especially pronounced in the [V(CO)3(NO)Ph2P(CH2)mPPh3] series (m = 1–4). The thermo-labile parent compound, [V(CO)5NO], has been characterized by its δ(51V) = ?1489 ppm at 245 K.  相似文献

Summary Synthetic polymeric constructions (SPCs) containing the consensus sequence of the HIV-1 surface envelope gycoprotein gp120 V3 loop (GPGRAF) block the fusion between HIV-1- and HIV-2-infected cells and CD4+-uninfected lymphocytes. By testing the activity of a series of SPC analogs in a cell-to-cell fusion assay, we found that the most active construction is an eight-branched SPC with the hexapeptide motif GPGRAF. This compound is also able to inhibit the infection of human lymphocytes and macrophages by unrelated isolates of HIV-1 and HIV-2. This antiviral activity is specific, since no toxicity was observed at the concentrations that inhibit HIV replication and syncytia formation. These data suggest that V3 SPCs may represent a new class of therapeutic anti-HIV agents, able to neutralize a wide range of viral isolates in infected individuals.  相似文献

1. Two additional octapeptides which stimulate the contractile activity of the cockroach hindgut have been isolated from head extracts of the cockroach, Leucophaea maderae.2. A series of four high performance liquid-chromatographic (HPLC) fractionations yielded sufficient quantities of pure peptides for micro amino acid analysis and primary sequence determinations. The sequence determined for two peptides were: Gly-Ser-Gly-Phe-Ser-Ser-Trp-Gly-NH2 and pGlu-Ser-Ser-Phe-His-Ser-Trp-Gly-NH2. These octapeptides were designated leucokinins V and VI (L-V, L-VI), respectively.3. Minimum concentrations of natural L—V and L-VI required to produce a response from the isolated cockroach hindgut were 4.5 × and 4.9 × 10−11 M, respectively. The capacitance C'(e), presumed to be located across the walls of the transverse tubules of twitch fibers, was identified in earlier impedance measurements by virtue of having a resistance in series with it. When the voltage V(m) across the surface membrane is made to vary, the voltage V(c) across C'(e) will be delayed with respect to V(m), the extent of the delay depending on the location of the series resistance. Model 1 assumes that the resistivity of the lumen of the tubules is negligible; model 2 assumes that the series resistance arises entirely in the tubular lumen; model 3 assumes that the resistivity of the tubular lumen is small, but not negligible and that the bulk of the resistance arises in a structure directly in series with C'(e) and having a similar geometric distribution. If V(m) varies sinusoidally, the relative value of V(c(max)) will fall with increasingly higher powers of the frequency at the center of the fiber if model 2 is applicable, whereas models 1 and 3 predict that V(c(max)) will fall at high frequency only in proportion to the frequency everywhere in the cross-section of the fiber. Equations have been derived for the voltage change V(c) in response to a step change of V(m) and during an action potential. On the assumption that contraction is initiated when V(c) reaches mechanical threshold, the delay between the activation of myofibrils on the axis of the fiber and at the surface would amount to 2.6 msec in model 2 and 0.25 msec in model 3 for frog fibers of about 100 mum diameter during a twitch.  相似文献
