Sections of teeth partly demineralised in 10% formic acid were examined by X-ray diffraction, microradiography and scanning electron microscopy. In the undemineralised circumpulpal dentin, the tubules were empty, lying in a matrix containing hydroxyapatite. In the 'plume' areas of remineralisation, the tubules were filled with mineral deposits. X-ray diffraction revealed the presence of brushite and monetite in these areas. In the outer layers of dentin the tubules were empty, lying in a matrix containing some residual hydroxyapatite. These findings confirmed that the remineralisation process occurred within the dentinal tubules.  相似文献

An examination of roots of hybrid larch from a farm forestry site by scanning electron microscopy has revealed crystalline deposits encrusting mantle hyphae of the associated ectomycorrhizal fungus. Electron probe micro-analysis identified calcium in the crystals and X-ray diffraction showed them to be whewellite, the monohydrate form of calcium oxalate. Abstract

Internal mineral deposits in stem and seta were found in living, healthy-looking specimens of the moss Homalothecium sericeum from a Spanish locality. The deposits consisted of an amorphous crust of smooth texture. After examination with EDS probes coupled to a transmission and a scanning electron microscope, and X-ray diffraction analysis, the minerals were identified as mainly opal, carbonates, and Fe and Al hydroxides, although it is irregular throughout a given mineralized region of the moss. These mineral crusts are considered to be the product of biomineralization of ions taken from the substratum. A comparison is established between these deposits and the mineral crystals found in a previous study of Hookeria lucens.  相似文献

Various chemotypes (Re, Rd2, Rd1P, Rd1, RcP, Rc, Rb3, Rb2, Rb1, and Ra) of R-form lipopolysaccharides (LPSs) of Salmonella spp. were crystallized by treatment with 70% ethanol containing 250 mM MgCl2, and crystals of the LPSs were observed electron microscopically and analyzed by electron diffraction and synchrotron X-ray diffraction. All the LPSs tested formed three-dimensional crystals showing very similar shapes; hexagonal plate, solid column, discoid, square or rectangular plate, lozenge plate and truncated hexangular or rectangular pyramid forms. Electron diffraction patterns from the hexagonal plate crystals of all these LPSs obtained by electron irradiation from the direction perpendicular to the basal plane showed that they consist of hexagonal lattices with the lattice constant of 4.62 Å. The crystals of all the LPSs thus formed gave ring-like X-ray diffraction patterns because of their small sizes. The long-axis values were calculated from the X-ray diffraction patterns from crystals of all the LPSs in the low-angle region and they corresponded roughly to the length of the proposed primary chemical structures of the R cores of the LPSs. The volume occupied by a single molecule of all the LPSs were calculated from the molecular weights based on the proposed structures and the crystallographic data obtained by electron diffraction, X-ray diffraction, and density determination.  相似文献

Mutant human lysozymes (Ile56Thr & Asp67His) have been reported to form amyloid deposits in the viscera. From the standpoint of understanding the mechanism of amyloid formation, we searched for conditions of amyloid formation in vitro using hen egg lysozyme, which has been extensively studied from a physicochemical standpoint. It was found that the circular dichroism spectra in the far-ultraviolet region of the hen egg lysozyme changed to those characteristic of a beta-structure from the native alpha-helix rich spectrum in 90% ethanol solution. When the concentration of protein was increased to 10 mg/mL, the protein solution formed a gel in the presence of 90% ethanol, and precipitated on further addition of 10 mM NaCl. The precipitates were examined by electron microscopy, their ability to bind Congo red, and X-ray diffraction to determine whether amyloid fibrils were formed in the precipitates. Electron micrographs displayed unbranched protofilament with a diameter of approximately 70 A. The peak point of the difference spectrum for the Congo red binding assay was 541 nm, which is characteristic of amyloid fibrils. The X-ray diffraction pattern showed a sharp and intense diffraction ring at 4.7 A, a reflection that arises from the interstrand spacing in beta-sheets. These results indicate that the precipitates of hen egg lysozyme are amyloid protofilament, and that the amyloid protofilament formation of hen egg lysozyme closely follows upon the destruction of the helical and tertiary structures.  相似文献

The cell wall of Staurastrum luetkemuelleri Donnat & Ruttner was examined with scanning electron microscope (SEM) using whole cells, in thin sections with transmission electron microscope (TEM), and in air dried whole cells and unstained thin sections with X-ray microanalysis in the scanning-transmission electron microscope (STEM). The cell wall was ornamented with spines and wartlike structures. Spines were solid structures, consisting of deposits of cell wall material between two main cell wall layers. The wart-like structures were pore organs extending through the cell wall and the mucilaginous layer outside the cell wall. The pore cylinder was surrounded by deposits of cell wall material similar to the ones in the spines. X-ray microanalysis of selected areas on whole cells from a natural population showed iron accumulation in discrete locations on the cell extensions of S. luetkemuelleri. In the unstained thin sections iron was found only in the cell wall deposits in the spines. Cells grown in laboratory cultures failed to show iron accumulation regardless of readdition of iron-EDTA (Fe-EDTA) to the culture medium.  相似文献

Summary In view of the importance of calcium in the induction of long-term potentiation (LTP), experiments were carried out to localize calcium at the electron microscopic level in the CA1 region of guinea pig hippocampal slices, following high-frequency stimulation of the Schaffer collaterals. Apart from the ultrastructural localization, a semiquantitative method was used to count the calcium-containing deposits in electron micrographs. Significantly more calcium-containing deposits were seen in the dendrites of the stratum radiatum in slices with LTP than in those without it. A moderate increase of the extradendritic deposits was observed, too. The calcium content of the deposits was determined by means of EGTA incubation and X-ray analysis. The presented results, together with the relevant literature data, underline the importance of calcium-activated processes in postsynaptic structures probably involved in the generation of LTP.  相似文献

Summary To investigate calcium distribution in cardiac muscle cells, two methods, one using oxalate and another using lead acetate, were used concomitantly to determine the subcellular localization of calcium. Particular attention was paid to the specificity of the methods employed. Chemical and physical analyses of the electron-opaque deposits of the reaction end-products was performed by chelation with EGTA or X-ray and electron diffraction. Results obtained show that the distribution of the calcium deposits in the cardiac muscle cells is more complex than that described for striated muscle fibers. The implications of these findings are discussed and an original viewpoint on the calcium distribution and movement in cardiac muscle cells is presented.  相似文献

Human pancreatic cells of the Capan-1 line form domes in culture during the stationary growth stage. The domes are thought to be a result of the transport of water and electrolytes by the Capan-1 cells. In older Capan-1 cultures, the epithelial sheets formed thickenings from several layers of cells of which the outermost ones were joined by tight type junctions. In the intracellular space, deposits of insoluble calcium salts were observed. Culture of Capan-1 cells in the presence of fibroblasts prolonged survival of the cultures with intact domes for more than 80 days. The Capan-1 cells proliferated forming multilayers and closed cavities which we called super-domes. X-ray spectrometry and electron diffraction analysis showed that the abundant deposits inside these cavities consisted of calcium phosphate in an apatite structure. The number of these deposits increased with time in culture, and they appeared to be formed at the sites of contact with an extracellular matrix consisting of cell debris. Deposits were not observed within the culture medium. Cells from domes were stained cytochemically for ATPases and alkaline phosphatases and examined by light and electron microscopy. The Capan-1 cells surrounding the domes were differentiated, polarized cells containing placental type alkaline phosphatases on their apical membranes and Ca2(+)-ATPases on their basolateral membranes. These enzymes were thought to play a role in the accumulation of phosphate and Ca2+ ions in the dome cavities, which then formed crystals in the presence of organic compounds produced by lysis of cells of the deepest layers of the super-domes. The crystals of hydroxyapatite observed in standard Capan-1 cell cultures and those cocultured with fibroblasts were assumed to be a result of transepithelial transport of Ca2+ and phosphate ions by these cells.  相似文献

The cubic phases of lipid-water systems have been studied by freeze-fracture electron microscopy. The preservation of the sample structure following cryofixation was verified by low temperature X-ray diffraction. Different types of fracture planes were identified; all display highly ordered two-dimensional domains, each subdivided into sub-domains related to each other by displacements and rotations related to the symmetry of the space group. The images were filtered using cross-correlation averaging techniques and the filtered images were compared to the corresponding planar sections of the electron density maps. Several conclusions were drawn: 1) when properly cryofixed, as assessed by low temperature X-ray diffraction, the structure of the sample was well preserved in the replicas; 2) the symmetry of the space group was faithfully reflected in the electron microscope images; 3) the crystallographic orientations of the most frequently identified'fracture planes coincided with those of the most intense X-ray reflections indicating that the fracture propagates, preferentially, in regions where the electron density variations are the largest; 4) when different structural models are compatible with X-ray diffraction data, it is possible to determine the correct model by comparing the filtered images with sections of the corresponding electron density maps; and 5) this approach constitutes a new and powerful tool of general interest for the low resolution study of three-dimensionally ordered specimens.  相似文献

Carnations were treated with a silver thiosulphate complex to prevent wilting of the flowers. The ultrastructural localization of silver and sulphur in the receptacle tissue was investigated by electron microscopy. Electron-dense deposits were present in the receptacle tissue. Coarse-grained deposts (diam. 60–100 nm) were predominantly observed at the inner side of the cell wall, whereas fine-grained deposits (diam. 20–60 nm) were predominantly present inside the cell-wall region and in the intercellular spaces. These particles were analyzed for chemical elements by X-ray analytical electron microscopy (Philips EM 400 plus Edax energy dispersive analyzer, type 711). In both types of deposits, the presence of silver and sulphur was verified. Models for the spatial distribution of protein, lipid and water in gap junction structures have been constructed from the results of the analysis of X-ray diffraction data described here and the electron microscope and chemical data presented in the preceding paper (Caspar, D. L. D., D. A. Goodenough, L. Makowski, and W.C. Phillips. 1977. 74:605-628). The continuous intensity distribution on the meridian of the X-ray diffraction pattern was measured, and corrected for the effects of the partially ordered stacking and partial orientation of the junctions in the X-ray specimens. The electron density distribution in the direction perpendicular to the plane of the junction was calculated from the meridional intensity data. Determination of the interference function for the stacking of the junctions improved the accuracy of the electron density profile. The pair-correlation function, which provides information about the packing of junctions in the specimen, was calculated from the interference function. The intensities of the hexagonal lattice reflections on the equator of the X-ray pattern were used in coordination with the electron microscope data to calculate to the two-dimensional electron density projection onto the plane of the membrane. Differences in the structure of the connexons as seen in the meridional profile and equatorial projections were shown to be correlated to changes in lattice constant. The parts of the junction structure which are variable have been distinguished from the invariant parts by comparison of the X-ray data from different specimens. The combination of these results with electron microscope and chemical data provides low resolution three- dimensional representations of the structures of gap junctions.  相似文献

Envelopes of the mineralizing protist, Trachelomonas hispida var. coronata Lemm., were examined by light and electron microscopy and by electron diffraction analysis. The ellipsoidal hollow envelope is made of mineralized strands of mucilage (muci-strands) that form a compact wall 0.2–4.0 μm thick, interspersed with numerous puncta 0.2–0.3 μm in diameter and, in some instances with tapered spines ca. 0.6 μm long and 0.7 μm wide at the base. The mucilage strands are cylindrical, anastomosing threads 20 nm thick. Electron-dense, needle-shaped mineral deposits form axial cores in the strands. Also, powdery granular mineral deposits are dispersed sparingly throughout the mucilage matrix. Micromicro-electron diffraction analyses verify the crystalline nature of the needle-shaped deposits, which are 4–9 nm thick and vary in length (20–100 nm). The mucilage strands and microcrystallites pervade the whole of the envelope matrix, including the spines, and may be preferentially oriented along the envelope surfaces.  相似文献

A recent model for the structure of microtubules is used to interpret X-ray fiber diffraction patterns from microtubules, obtained under various conditions. The results suggest that tubulin may undergo conformational changes under conditions of reduced water-activity. Such changes could account for some of the differences in the structure of tubulin as determined by electron microscopy and X-ray diffraction.  相似文献

Well-defined X-ray diffraction patterns have been recorded from erythrocyte membranes in the frozen state. At ?40°C, lamellar periodicities range from 19 to 95 nm depending on the glycerol content (0–40%, respectively). Freeze-fracture electron micrographs of samples frozen in two stages to approximate to the diffraction conditions show ice formation external to membrane stacks. The membrane stacks have periodicities of the same order of magnitude as those obtained by X-ray diffraction.  相似文献

This article presents a research study on the deposition process of Ca2+ induced by Synechocystis sp. PCC6803 in BG11 liquid medium with different Ca2+ concentrations and different pH. The changes of Ca2+ concentrations were measured by using atomic absorption method and the corresponding dynamical models were studied. Minerals and cells were analyzed by high resolution transmission electron microscope, selected area electron diffraction, scanning electron microscope, energy dispersive X-ray spectroscope, X-ray diffraction. The selected area electron diffraction patterns were analyzed by Digital Micrograph 3.7 software. The result showed that Ca2+ concentrations decreased faster in the experimental group. The changes of calcium carbonate precipitation were fitting to an exponential model. PH 7 and Ca2+ concentration of 1.5 g/L were most conducive to calcium carbonate precipitation in the corresponding gradient range. The result of high-resolution transmission electron microscopy showed that minerals in the experimental group differed obviously from that of the control group in the surface morphology, but both of them were calcites. It also showed that a certain number of minute calcites adhesion to the outer surfaces of S. PCC6803 cells. The result of scanning electron microscopy displayed that many sunken holes emerged on the surfaces of the prismatic calcium carbonate minerals. The results of X-ray diffraction proved that minerals induced by S. PCC6803 were calcites with preferential orientation. This article discusses the process of carbonate formation and the possible role played by S. PCC6803. It may be useful to further study the mechanism of microbial carbonates deposition in the field of geology.  相似文献

Some species of anuran amphibians possess a calcified dermal layer (the Eberth-Kastschenko layer) located between the "stratum spongiosum" and the "stratum compactum." This layer consists of calcium phosphate deposits, proteoglycans, and glycosaminoglycans. Although regarded as a protective layer against desiccation, a calcium reservoir, or possibly a remnant of a dermal skeleton present in anuran ancestors, very little is known about its origin, structure, and function. Thus, we studied the structure and composition of the mineralized dermal layer of Corythomantis greeningi, a peculiar hylid from the Brazilian semiarid region (caatinga), using conventional and cryosubstitution methods combined with transmission, scanning, and analytical electron microscopy. Results show that the dermal layer consists of dense, closely juxtaposed, globular structures. Although the electron opacity of the globules was variable, depending on the type of preparation, crystal-like inclusions were present in all of them, as confirmed by dark field microscopy. Electron probe X-ray microanalysis showed calcium, phosphorus, and oxygen, and electron diffraction revealed a crystalline structure comparable to that of a hydroxyapatite.  相似文献

The analysis of single crystals of the lipovitellin complex from lamprey has made it possible to compare electron density projections derived from X-ray diffraction and density modification methods with previously published electron micrographs. The close correlation between the images obtained by the two methods demonstrates that the fidelity of images obtained by electron microscopy is excellent despite the loss of specimen order. These correlations also attest to the ability of density modification methods, currently used in macromolecular crystallography, to estimate the phases of small angle X-ray reflections.  相似文献

The X-ray diffraction pattern of myelin of frog sciatic nerve has been investigated, using a Kratky small angle slit camera to obtain the electron density distribution across the membrane. All major reflections observed were related to a fundamental repeat distance of 171 ± 2.8 A. There was no further increase in the number of reflections on varying the experimental conditions (varying pH, applying tension, immersion in various isotonic buffer solutions, etc.) or by varying the camera slit arrangement. The degree of disorder within the myelin sheath was examined by comparing the crystallite size to the half-width of the diffraction peak at half-height. The limiting of the diffraction spectra to five major reflections was determined not to be caused by disorder. It is concluded that the observed X-ray diffraction pattern is a consequence of the particular electron density distribution of the membrane. Therefore, the membrane cannot contain sharply distinct step-function regions of electron density, but approaches a modified cosine distribution.  相似文献
