Summary Lignocellulosic biomass materials were pretreated with dilute sulfuric acid, followed by analysis of partially and extensively washed pretreated solids. It is demonstrated that it is of crucial importance to perform extensive washing for accurate carbohydrate analyses to be obtained. The effects of different thermo-chemical pre-treatment methods were determined on the biodegradability and hydrolysis rate of lignocellulosic biomass. Three plant species, hay, straw and bracken were thermo-chemically pre-treated with calcium hydroxide, ammonium carbonate and maleic acid. After pre-treatment, the plant material was anaerobically digested in batch bottles under mesophilic conditions for 40 days. From the pre-treatment and subsequent anaerobic digestion experiments, it was concluded that when the lignin content of the plant material is high, thermo-chemical pre-treatments have a positive effect on the biodegradability of the substrate. Calcium hydroxide pre-treatment improves the biodegradability of lignocellulosic biomass, especially for high lignin content substrates, like bracken. Maleic acid generates the highest percentage of dissolved COD during pre-treatment. Ammonium pre-treatment only showed a clear effect on biodegradability for straw. 相似文献

Some properties of the cellulolytic complex obtained from Trichoderma reesei QM 9414 grown on Solka floc as carbon source and its ability to hydrolyze the lignocellulosic biomass of Onopordum nervosum Boiss were studied. The optimum enzyme activity was found at temperatures between 50 and 55 degrees C and pH ranging from 4.3 to 4.8. Hydrolysis of 4-nitropnenyl-beta-D-glucopyranoside (4-NPG) and cellobiose by the beta-glucosidase of the complex, showed competitive inhibition by glucose with a K(i) value of 0.8 mM for 4-NPG and 2. 56 mM for cellobiose. Enzymatic hydrolysis yield of Onopordum nervosum, evaluated as glucose production after 48 h, showed a threefold increase by pretreating the lignocellulosic substrate with alkali. When the loss of glucose incurred by de pretreatment was taken into account, a 160% increase in the final cellulose to glucose conversion was found to be due to the pretreatment. 相似文献

An epidemic based model was developed to describe the enzymatic hydrolysis of a lignocellulosic biomass, dilute sulfuric acid pretreated corn stover. The process of substrate getting adsorbed and digested by enzyme was simulated as susceptibles getting infected by viruses and becoming removed and recovered. This model simplified the dynamic enzyme “infection” process and the catalysis of cellulose into a two‐parameter controlled, enzyme behavior guided mechanism. Furthermore, the model incorporates the adsorption block by lignin and inhibition effects on cellulose catalysis. The model satisfactorily predicted the enzyme adsorption and hydrolysis, negative role of lignin, and inhibition effects over hydrolysis for a broad range of substrate and enzyme loadings. Sensitivity analysis was performed to evaluate the incorporation of lignin and other inhibition effects. Our model will be a useful tool for evaluating the effects of parameters during hydrolysis and guide a design strategy for continuous hydrolysis and the associated process control. © 2014 American Institute of Chemical Engineers Biotechnol. The high cost of lignocellulolytic enzymes is one of the main barriers towards the development of economically competitive biorefineries. Enzyme engineering can be used to significantly increase the production rate as well as specific activity of enzymes. However, the success of enzyme optimization efforts is currently limited by a lack of robust high-throughput (HTP) cellulase screening platforms for insoluble pretreated lignocellulosic substrates. We have developed a cost-effective microplate based HTP enzyme-screening platform for ionic liquid (IL) pretreated lignocellulose. By performing in-situ biomass regeneration in micro-volumes, we can volumetrically meter biomass (sub-mg loading) and also precisely control the amount of residual IL for engineering novel IL-tolerant cellulases. Our platform only requires straightforward liquid-handling steps and allows the integration of biomass regeneration, washing, saccharification, and imaging steps in a single microtiter plate. The proposed method can be used to screen individual cellulases as well as to develop novel cellulase cocktails. 相似文献

Catalytic hydrolysis of lignocellulosic biomass into 5-hydroxymethylfurfural in ionic liquid 总被引:1,自引:0,他引:1
Production of 5-hydroxymethylfurfural (HMF) from cellulose catalyzed by solid acids and metal chlorides was studied in the 1-butyl-3-methylimidazolium chloride ([BMIM]Cl) under microwave irradiation. Among the applied catalysts, the use of CrCl3/LiCl resulted in the highest yield of HMF. The effects of catalyst dosage (mole ratio of catalyst to glucose units in the feedstock) and reaction temperature on HMF yields were investigated to obtain optimal process conditions. With the 1:1 mol ratio of catalyst to glucose unit, the HMF yield reached 62.3% at 160 °C for 10 min. Untreated wheat straw was also investigated as feedstock to produce HMF for the practical use of raw biomass, in which the HMF yield was comparable to that from pure cellulose. After the extraction of HMF, [BMIM]Cl and CrCl3/LiCl could be reused and exhibited no activity loss after three successive runs. 相似文献

Calculation of true sugar yields in high solids enzymatic hydrolysis of biomass is challenging due to the varying liquid density and liquid volume resulting from solid solubilization. Ignoring these changes in yield calculations can lead to significant errors. In this paper, a mathematical method was developed for the estimation of liquid volume change and thereafter the sugar yield. The information needed in the calculations include the compositions of the substrate, initial solids loading, initial liquid density, and sugar concentrations before and after hydrolysis. All of these variables are measurable with conventional laboratory procedures. This method was validated experimentally for enzymatic hydrolysis of dilute sulfuric acid pretreated corn stover at solid loadings up to 23% (w/w). The maximum relative error of predicted glucose yield from the true value was less than 4%. Compared to other methods reported in the literature, this method is relatively easy to use and provides good accuracy. 相似文献

This study aims to evaluate the activity of the cellulase enzyme forward the use of ultrasound technology in different conditions of temperature, pH and exposure time, as well, to match the steps of pretreatment and enzymatic hydrolysis in one step. A central composite design (CCRD) and response surface analysis were used to evaluate the effect of ultrasound power, temperature and pH on enzyme activity. Optimum condition in the studied range was 30% for ultrasound power, pH 4.6 and 50?°C, yielding an enzyme activity of 15.5 UPF/mL. From this, we carried out kinetics of enzymatic hydrolysis on filter paper and bagasse malt, in optimized conditions. Total reducing sugars (TRS) were 3.85 and 0.46?mg/mL when the filter paper and bagasse malt were used as substrate, respectively. Ultrasound showed to be a good technology to increase the enzyme activity aiming to intensify enzymatic processes. 相似文献

Pretreatment of lignocellulosic biomass has been taken up as a global challenge as it comprises a large renewable source of fermentable sugars. In this study, effect of electron beam irradiation (EBI) on a hybrid grass variety investigated as a biomass pretreatment method. Dry biomass samples after characterization were exposed to EBI doses of 0, 75, 150 and 250kGy. The pretreated biomass samples were enzymatically hydrolyzed using Trichoderma reesei ATCC 26921 cellulase for 144h. The enzyme loadings were 15 and 30FPU/g of biomass. The structural changes and degree of crystallinity of the pretreated biomass were studied by FTIR, XRD and SEM analyses. The lignocellulosic biomass sample showed 12.0% extractives, 36.9% cellulose, 28.4% hemicellulose, 11.9% lignin and 8.6% ash. Significant improvements in the reducing sugar and glucose yields were observed in the hydrolysate of EBI pretreated biomass compared to the control. In 250kGy exposed samples 79% of the final reducing sugar yield was released within 48h of hydrolysis at an enzyme loading rate of 30FPU/g of biomass. The IR crystallinity index calculated from the FTIR data and degree of crystallinity (XRD) decreased in the EBI treated samples. A significant negative correlation was observed between degree of crystallinity and the glucose yield from enzymatic hydrolysis. 相似文献

Agricultural and herbaceous feedstocks may contain appreciable levels of sucrose. The goal of this study was to evaluate the survivability of sucrose and its hydrolysis products, fructose and glucose, during dilute sulfuric acid processing at conditions typically used to pretreat lignocellulose biomass. Solutions containing 25g/l sucrose with 0.1-2.0% (w/w) sulfuric acid concentrations were treated at temperatures of 160-200 degrees C for 3-12min. Sucrose was observed to completely hydrolyze at all treatment conditions. However, appreciable concentrations of fructose and glucose were detected and glucose was found to be significantly more stable than fructose. Different mathematical approaches were used to fit the kinetic parameters for acid-catalyzed thermal degradation of these sugars. Since both sugars may survive dilute acid pretreatment, they could provide an additional carbon source for production of ethanol and other bio-based products. 相似文献

Pretreatment is a crucial step in the conversion of lignocellulosic biomass to fermentable sugars and biofuels. Compared to thermal/chemical pretreatment, fungal pretreatment reduces the recalcitrance of lignocellulosic biomass by lignin-degrading microorganisms and thus potentially provides an environmentally-friendly and energy-efficient pretreatment technology for biofuel production. This paper provides an overview of the current state of fungal pretreatment by white rot fungi for biofuel production. The specific topics discussed are: 1) enzymes involved in biodegradation during the fungal pretreatment; 2) operating parameters governing performance of the fungal pretreatment; 3) the effect of fungal pretreatment on enzymatic hydrolysis and ethanol production; 4) efforts for improving enzymatic hydrolysis and ethanol production through combinations of fungal pretreatment and physical/chemical pretreatment; 5) the treatment of lignocellulosic biomass with lignin-degrading enzymes isolated from fungal pretreatment, with a comparison to fungal pretreatment; 6) modeling, reactor design, and scale-up of solid state fungal pretreatment; and 7) the limitations and future perspective of this technology. 相似文献

Dicarboxylic organic acids have properties that differ from those of sulfuric acid during hydrolysis of lignocellulose. To investigate the effects of different acid catalysts on the hydrolysis and degradation of biomass compounds over a range of thermochemical pretreatments, maleic, oxalic and sulfuric acids were each used at the same combined severity factor (CSF) values during hydrolysis. Xylose and glucose concentrations in hydrolysates were highest with maleic acid. Oxalic acid gave the next highest followed by sulfuric acid. This ranking was particularly true at low CSF values. The concentrations of glucose and xylose increased with oxalic and sulfuric acid pretreatments as the CSF increased, but they never attained the levels observed with maleic acid. Among sulfuric, oxalic and maleic acid treatments, the amount of xylose released as xylooligosaccharide was highest with sulfuric acid. The fraction of xylooligosaccharide was lowest with the maleic acid and the oligosaccharide fraction with oxalic acid fell in between. Furfural and hydroxymethyl furfural levels were also highest with maleic acid. In subsequent fermentations with pretreated biomass, the ethanol concentration was maximal at 19.2 g/l at CSF 1.9 when maleic acid was used as the pretreatment catalyst. This corresponded to an ethanol volumetric production rate of 0.27 g ethanol/l per h. This was the same condition showing the highest xylose production in following pretreatment with various acid catalysts. These findings suggest that maleic and oxalic dicarboxylic acids degrade hemicelluloses more efficiently than does sulfuric acid. 相似文献

Differences in the relative toxicity of xylose-rich prehydrolysates derived from woody and herbaceous feedstocks are likely due to the relative abundance of a variety of inhibitory compounds. Acetate, as well as several aromatic monomers, has been shown to be an inhibitor of the xylose-fermenting yeast, Pichia stipitis. Comparative information on the concentration of known and likely inhibitors, other than acetate, is lacking. The present study provides data on the aromatic monomer composition of representative herbaceous and woody prehydrolysates. Dilute-acid prehydrolysates were prepared from three feedstocks; two herbaceous, corn stover and switchgrass (Panicum virgatum L.), and one woody (poplar). The prehydrolysates were neutralized with Ca(OH)2 extracted with ethyl acetate, trimethylsilylated, and analyzed by GC-MS. Fourteen aromatic monomers were tentatively identified by comparison with published mass spectra. The concentrations of the aromatic monomers totalled 112, 141 and 247 mg L−1 for corn stover, switchgrass and poplar prehydrolysates, respectively. This is also the order of increasing inhibition of growth and ethanol productivity observed for Pichia fermentations. The woody prehydrolysate contained approximately four-fold more syringyl-based monomers than did the herbaceous prehydrolysates, while guaiacyl-containing compounds were more evenly distributed. AbstractMyceliophthora thermophila encodes for large number of carbohydrate-active enzymes (CAZymes) involved in lignocellulosic biomass degradation. The mould was grown on rice straw in solid state fermentation at pH 5.0 and 45?°C that produced high levels of cellulolytic and xylanolytic enzymes i.e. 2218.12, 515.23, 478.23, 13.34?U/g DMR for xylanase, CMCase, FPase and β-glucosidase, respectively. The secretome analysis of M. thermophila BJAMDU5 by mass spectroscopy, described 124 different proteins with majority of CAZymes consisting of glycosyl hydrolases (GH), lytic polysaccharide mono-oxygenases (LPMO), carbohydrate esterases (CE) and polysaccharide lyases (PL). Furthermore, the enzyme cocktail of the mould was evaluated for hydrolysis of steam treated rice straw that produced 184.59?mg/g substrate reducing sugars after 24?h, which was used for production of bioethanol by using fast fermenting yeast Saccharomyces cerevisiae resulting in high production of bioethanol. 相似文献

Increasing interest is being shown in the cyanobacterium Spirulina platensis because of its nutritional properties when used as food supplement and possible therapeutic effects. One of the most important areas being studied is the development of alternative nutrient sources which can be used to decrease the production costs of commercially produced S. platensis and obtain high productivity. Water from Mangueira Lagoon (Rio Grande do Sul State, Brazil) has high levels of carbonates and a high pH and has the potential to be used as a culture medium for S. platensis, although some nutrient supplementation may be required. We tested the effect of unsupplemented Mangueira Lagoon water (MLW) or MLW supplemented with 1.125 or 2.250 mg/l of urea and/or 21 or 42 mg/l of sodium bicarbonate on the growth of S. platensis in fed-batch culture using a 3(2) factorial design and found that there the addition of 1.125 mg/l of urea resulted in a 2.67 fold increase times in the final biomass concentration of S. platensis. 相似文献

The dimorphic fungus Mucor indicus was grown in different forms classified as purely filamentous, mostly filamentous, mostly yeast-like and purely yeast-like, and the relationship between morphology and metabolite production, inhibitor tolerance and the cell wall composition was investigated. Low concentrations of spores in the inoculum with subsequent aeration promoted filamentous growth, whereas higher spore concentrations and anaerobic conditions promoted yeast-like growth. Ethanol was the main metabolite with glycerol next under all conditions tested. The yields of ethanol from glucose were between 0.39 and 0.42 g g−1 with productivities of 3.2–5.0 g l−1 h−1. The ethanol productivity of mostly filamentous cells was increased from 3.9 to 5.0 g l−1 h−1 by the presence of oxygen, whereas aeration of purely yeast-like cells showed no such effect. All growth forms were able to tolerate 4.6 g l−1 furfural and 10 g l−1 acetic acid and assimilate the sugars, although with different consumption rates. The cell wall content of the fungus measured as alkali insoluble materials (AIM) of the purely yeast-like cells was 26% of the biomass, compared to 8% of the pure filaments. However, the chitosan concentration of the filaments was 29% of the AIM, compared to 6% of the yeast-like cells. 相似文献

Design considerations for enzymatic hydrolysis of lignocellulosic biomass in two and three continuous stirred tank reactors (CSTRs) in series with distributed feeding of substrate and enzyme, followed by a series of CSTRs, are discussed. A previously developed, fitted, and validated kinetic model is extended to accommodate distributed feeding and used along with the micromixing limiting situations of macrofluid and microfluid to describe the reaction system. The capabilities of the reaction system proposed are explored for a range of cumulative substrate concentration from 5 to 20% w/w (dry basis). Continuous distributed feeding does not show advantages in terms of cellulose conversion when compared with the operation where an equivalent mass of substrate is added at the first reactor of the series, but the potential to increase substrate concentration beyond the concentrations that can be handled in conventional CSTRs, and therefore, the volumetric productivity of reactors, is evident. 相似文献

Ethanol produced from lignocellulosic biomass is a renewable alternative to diminishing petroleum based liquid fuels. The release of many new sugarcane varieties by the United States Department of Agriculture to be used as energy crops is a promising feedstock alternative. Energy cane produces large amounts of biomass that can be easily transported, and production does not compete with food supply and prices because energy cane can be grown on marginal land instead of land for food crops. The purpose of this study was to evaluate energy cane for lignocellulosic ethanol production. Energy cane variety L 79-1002 was pretreated with weak sulfuric acid to remove lignin. In this study, 1.4 M sulfuric acid pretreated type II energy cane had a higher ethanol yield after fermentation by Klebsiella oxytoca without enzymatic saccharification than 0.8 M and 1.6 M sulfuric acid pretreated type II energy cane. Pretreated biomass was inoculated with K. oxytoca for cellulose fermentation and Pichia stipitis for hemicellulose fermentation under simultaneous saccahrification and fermentation (SSF) and separate hydrolysis and fermentation (SHF) conditions. For enzymatic saccharification of cellulose, the cellulase and ??-glucanase cocktail significantly increased ethanol production compared to the ethanol production of fermented acid pretreated energy cane without enzymatic saccharification. The results revealed that energy cane variety L 79-1002 produced maximum cellulosic ethanol under SHF (6995 mg/L) and produced 3624 mg/L ethanol from fermentation of hemicellulosic sugars. 相似文献
