Superoxide anion radical, a radical whose toxicity is well documented, is present in the uterus of albino rat. The cyclic changes that it undergoes during the reproductive cycle are also illustrated. An inverse correlation is seen between the levels of superoxide radical and the enzyme dismutating it, i.e., superoxide dismutase (EC 1.15.1.1), which is also reported to be present in the uterus of albino rat and is shown to exhibit cyclic changes during its oestrous cycle. The high levels of superoxide radical at pro-oestrus enable us to hypothesize that superoxide may be involved in regulating the oedema and cell proliferation of the uterus during proestrus.  相似文献

NADPH dependent O2- production in the ovary and uterus of cycling and pregnant mice was estimated employing a novel technique of time-lapse electron spin resonance enhancement of a superoxide-trapping spin trap. The NADPH dependent O2- generation in the mouse ovary increased during the early pre-ovulatory phase in cycling females and during extended luteal phase in pregnant animals. A peak of uterine NADPH dependent O2- production at proestrous in the cycling animals at pre-implantation phase in pregnant animals suggested a contribution of this enzyme towards generating high levels of superoxide anion radical during the respective stages. Both ovarian and uterine NADPH dependent O2- production appeared to be LH-inducible.  相似文献

Superoxide anion radical plays a significant role in inflammation, like rheumatoid arthritis. Superoxide dismutase enzyme known to dismutate superoxide anion radical does not play any significant role in this multisystem disease. This paper reports that very low levels of circulating superoxide dismutase levels are observed in patients with rheumatoid arthritis and these levels significantly improve with NSAID therapy. The possible mechanism of the action is discussed.  相似文献

Conceptus (embryo and associated extraembryonic membranes) implantation and development require a reciprocal biochemical and physical interactions between the extraembryonic membranes and the endometrium. However, the enzymatic antioxidative pathways controlling reactive oxygen species production at the endometrial-extraembryonic membrane interface early in pregnancy are not known. We aimed therefore to determine the content of malondialdehyde, as biomarkers of lipid peroxidation, and the activities of the major antioxidant enzymes, copper-zinc containing and manganese containing superoxide dismutases, catalase and glutathione peroxidase, in sheep extraembryonic membranes, caruncular and intercaruncular endometrium zones sampled at specific stages of pregnancy corresponding to the conceptus implantation (day 16) and the early post-implantation period (day 21). Malondialdehyde content in caruncular, intercaruncular and extraembryonic tissues was not different between stages of the pregnancy. Extraembryonic membranes demonstrated increased manganese containing superoxide dismutase and glutathione peroxidase activities, whereas catalase activity in these tissues decreased from day 16 to day 21. Caruncular tissues demonstrated increased manganese containing superoxide dismutase activity from day 16 to day 21. Intercaruncular tissues demonstrated increased copper-zinc containing superoxide dismutase, manganese containing superoxide dismutase and catalase activities from day 16 to day 21. The ovine extraembryonic membranes exhibit dynamic changes in enzymatic antioxidative pathways different from those of endometrial tissues during the transition from implantation to post-implantation period. This biochemical data provides novel insights into the developmental changes in antioxidative pathways of extraembryonic membranes and endometrium during early conceptus development.  相似文献

We have recently shown that Bloom syndrome fibroblasts have elevated levels of superoxide dismutase activity compared to those of normal fibroblasts. Based on this observation we decided to test whether an increased rate of superoxide radical production could be responsible for the induction of superoxide dismutase and of chromosomal aberrations and sister-chromatid exchanges characteristic of Bloom syndrome. Utilizing the superoxide-generating herbicide paraquat in Chinese hamster fibroblasts, we assayed the cells for dismutase activity, chromosomal aberrations and sister-chromatid exchanges. All 3 parameters investigated demonstrated a dose-dependent increase with paraquat and, consequently, with the superoxide produced. Since the induction of the enzyme is mediated by its substrate, the superoxide anion radical, we concluded that the increased dismutase activity (in Bloom syndrome and paraquat-treated cells) may be a secondary manifestation of an overall imbalance in oxygen metabolism and that this elevated enzymatic activity is insufficient to detoxify the high superoxide levels, which results in elevated levels of chromosomal damage.  相似文献

Superoxide dismutase, which has been shown to be present in a number of tissues, exhibits cyclic changes during the reproductive cycle of rats. An inverse correlation is seen between the levels of superoxide dismutase and superoxide radical. In immature, pseudopregnant rats, primed with human Chorionic Gonadotropin, lutropin seemed to induce ovarian superoxide dismutase, which could be blocked significantly by the introduction of anti-LH serum. These results point out the specific induction of superoxide dismutase by lutropin. It is reasonable to postulate that during luteal functioning, luteinizing hormone induces superoxide dismutase which in turn seems to play a central role generating hydrogen peroxide from superoxide anion radicals. Hydrogen peroxide, thus formed, drives the peroxidase-ascorbate system, responsible for production of progesterone.  相似文献

In the guinea pig, for which implantation is supposedly progesterone-dependent, actual hormonal requirements were assessed by measuring the levels of circulating estradiol and progesterone and correlating them with their content in the ovaries and uterus, and uterine concentrations of their receptors prior to, during, and immediately after implantation. Ovarian and uterine content and plasma levels of estradiol and progesterone, as well as uterine cytosolic receptors of these two hormones, were high at proestrus. Up to day 3 of pregnancy, estradiol remained high in peripheral plasma, ovarian and uterine tissues, but reached low levels at the time of implantation. The levels of progesterone showed a gradual increase in plasma and ovaries till the time of implantation, with the embryonic site of the uterus accumulating more of progesterone compared to estradiol. As pregnancy progressed, a gradual translocation of cytosolic to nuclear receptors occurred, both with estradiol and progesterone receptors. Comparing the receptor values for estradiol at each uterine site showed no significant alterations between embryonic and interembryonic cytosolic receptors. While significantly high levels of nuclear estradiol receptor were found at the inter-embryonic site on day 9 of pregnancy, the cytosolic and nuclear progesterone receptor concentrations were greater at the embryonic site on the same day. These findings demonstrated that the uterus is adequately exposed to estradiol and progesterone prior to ovulation and again in early pregnancy (day 1-3), thus facilitating implantation in the guinea pig (on days 7-8).  相似文献

Prolactin (PRL) exerts pleiotropic physiological effects in various cells and tissues, although it is mainly considered as a regulator of reproduction and cell growth. Null mutation of the prolactin receptor (PRLR) gene leads to female sterility due to a failure of embryo implantation. Using this mouse model and the method of mRNA differential display, we identified PRL target genes that are regulated during the peri-implantation period. We characterized 1 among the 45 isolated genes, UA-3, which is regulated in the uterus as well as in the ovary during early pregnancy. This gene corresponds to a P311 mouse cDNA that was originally identified for its high expression in late-stage embryonic brain and adult cerebellum. We report here that UA-3 is present in numerous tissues as well as in ovary and uterus at the site of blastocyst apposition, and that its expression is hormonally regulated. Moreover, in situ hybridization reveals high expression in ovarian granulosa cells and in uterine epithelium. Recently, it has been suggested that P311 expression is tightly regulated at several levels by mechanisms that control cellular growth, transformation, motility, or a combination of these. Taken together, these results suggest that P311 could be involved in these processes during pregnancy, although its function remains to be clearly established.  相似文献

Estrogens secreted by the ovary during the pre/periimplantation period and/or by the blastocyst and acting locally on the endometrium are involved in the initiation of implantation. Estrogens induce a cascade of metabolic changes in the uterus and blastocyst prior to and soon after the attachment and implantation of the blastocysts. Antiestrogens either administered intraluminally into the uterus prior to implantation or washing free blastocysts with antiestrogens prior to transfer to uteri of progesterone treated hamsters leads to failure of implantation. A number of antiestrogens which inhibit fertility in the rats do not interfere with implantation in the hamster and monkey when administered post-coitally. However, Zuclomiphene administered during days 5–11 of the menstrual cycle inhibits implantation in the rhesus monkey. Antiestrogens are being evaluated in other non-human primates to confirm the above results and to determine the time in the menstrual cycle susceptible to modification and inhibition of implantation. Tamoxifen administered from days 18–30 of the cycle to mated bonnet monkeys inhibited implantation despite maintenance of high levels of circulating progesterone. Neutralization of the vitamin carrier proteins (by active immunization against these proteins) interferes with established pregnancy in the rat and perhaps in the bonnet monkey. Whether antiestrogens can reduce the levels of vitamin carrier proteins to a level which is not adequate for maintenance of early pregnancy is not clear. Compounds which show antiestrogenic and antiprogestational properties may have advantages in inhibiting implantation or disruption of early pregnancy. Critical experiments need to be carried out in non-human primates to delineate the effectiveness of antiestrogens, with particular emphasis on time, dose, duration and route of administration in inhibition of implantation. Centchroman, an antiestrogen with antiprogestational properties, has been found to provide pregnancy protection with minimal side effects. However, several concerns relating to safety in toxicological studies in monkeys and a dose which would provide acceptable rate of contraceptive efficacy without major effects on the menstrual cycle need to be clarified before considering the potential of centchroman as a possible oral contraceptive administered either post-coitally or once a week. Inhibition of implantation by administration of tamoxifen opens up new possibilities of use of antiestrogens for fertility regulation.  相似文献

Nitrofurantoin, misonidazole, and metronidazole were reduced to their corresponding nitro anion radicals by ascorbate in anaerobic solutions at high pH. The nitrofurantoin anion radical could be detected at neutral pH. In neutral solutions, the nitro anion radicals of misonidazole and metronidazole were too unstable to be observed by electron spin resonance spectroscopy. At neutral pH, solutions containing ascorbate, nitrofurantoin, or misonidazole consumed oxygen. The addition of superoxide dismutase, catalase, or both superoxide dismutase and catalase decreased the rate of oxygen consumption. These results show that nitro anion radicals are formed by reduction with ascorbate, and superoxide anion radical and hydrogen peroxide are produced by reactions of these radicals with oxygen.  相似文献

Countercurrent transfer in the ovarian vascular pedicle elevates the concentration of steroid hormones in blood supplying the oviduct and periovarian part of the uterus during the estrous cycle in the pig. This study was conducted to determine whether during early pregnancy the arterial blood supply to the oviduct and uterus carries greater concentration of steroid hormone than systemic blood. The concentration of ovarian steroid hormones (progesterone, estradiol-17 beta, estrone, androstenedione and testosterone) was measured in 40 gilts on Days 12, 18, 25 or 35 of pregnancy. Silastic catheters were inserted: a) into the jugular vein, b) into the branch of uterine artery close to the ovary (proximal to the ovary) and c) into the branch of the uterine artery close to the cervix (distal to the ovary). On the day following surgery simultaneous blood samples from cannulated vessels were collected every 20 min for 3 hours. The concentration of steroid hormones was determined by radioimmunoassay. The mean concentrations of studied hormones in branches of the uterine artery proximal and distal to the ovary were significantly greater than in the jugular vein (P < 0.001) by 18 to 69% and 7 to 31%, respectively. The concentrations of hormones in proximal and distal to the ovary branch of the uterine artery were also significantly different (P < 0.001). The increase in concentrations of the measured hormones did not differ considerably between investigated days of pregnancy. It is concluded that during maternal recognition of pregnancy, formation of the corpus luteum of pregnancy, implantation of the embryo and the placenta elongation the oviduct and uterus are supplied with locally elevated concentration of steroid hormones compared to systemic blood.  相似文献

The endometrial phospholipid bilayer is shown to be in a highly fluid and polar state at the time of implantation in Mus musculus as evidenced from a spin label study using the stearic acid spin label, 5-doxyl stearate. The positive correlation observed between the superoxide anion radical levels and the fluidity and polarity state of the endometrial cells points towards the possibility of the mediation of this condition of unsaturation of fatty acids to be an act of the superoxide anion radical.  相似文献

Implantation requires reciprocal interaction between blastocysts and a receptive uterus. In mice, one important player in this dialogue involves endocannabinoid signaling via cannabinoid receptor CB1. Anandamide is an endogenous cannabinoid ligand, and its levels are spatiotemporally regulated in the uterus during early pregnancy, showing lower levels in the receptive uterus and at the implantation site. However, the mechanism by which differential uterine anandamide gradients are established under different pregnancy status is not clearly understood. Using multiple approaches, we show here that uterine anandamide levels conducive to implantation are primarily regulated by spatiotemporal expression of Nape-Pld, the gene encoding N-acylphosphatidylethanolamine-hydrolyzing phospholipase D that generates anandamide. The expression is well correlated with its activity and anandamide levels. This study is clinically relevant, since elevated anandamide levels in peripheral circulation are associated with spontaneous pregnancy failure in women.  相似文献

大鼠动情周期以及胚胎着床过程中,子宫内膜会发生结缔组织的降解与重构。胶原酶3(MMP-13)是降解纤维类胶原的主要蛋白水解酶类之一。其活性在这些过程中的变化值得研究。采用液体闪烁计数测定~3H标记胶原的方法,对大鼠动情周期及早期妊娠子宫中胶原酶-3(MMP-13)的活性进行了测定。结果表明:在动情周期中;激活型MMP-13在间情期最低,酶原型及激活型的MMP-13在动前期达高峰,动情后期酶原型和激活型MMP也明显高于间情期(P<0.05)(Fig.1)。妊娠第1、2天酶原型的MMP-13的活性显著高于第3~7天,第3、4天酶原型和激活型MMP-13的活性均低于妊娠第1、2天(P<0.05);而第5天酶原型MMP-13的活性却显著高于第4、6两天(P<0.05);激活型MMP-13的活性也高于第4天(P<0.05)(Fig.2)。着床部位酶原型MMP-13的活性明显高于非着床部位(P<0.05),而激活型MMP-13的活性则无明显差异(P>0.05)(Fig.3)。大鼠假孕早期子宫中MMP-13的活性变化与正常早期妊娠相似,但其活性却明显低于正常早期妊娠(Fig.4)。结果提示:大鼠子宫中MMP-13参与大鼠动情周期及早期妊娠过程,尤其是在胚胎着床过程中可能起着重要作用。  相似文献

Treatment of bovine pulmonary artery smooth muscle microsomes with the superoxide radical generating system hypoxanthine plus xanthine oxidase stimulated iron release, hydroxyl radical production and lipid peroxidation. Pretreatment of the microsomes with deferoxamine or dime thy lthiourea markedly inhibited lipid peroxidation, and prevented hydroxyl radical production without appreciably altering iron release. The superoxide radical generating system did not alter the ambient superoxide dismutase activity. However,addition of exogenous superoxide dismutase prevented superoxide radical induced iron release,hydroxyl radical production and lipid peroxidation. Simultaneous treatment of the microsomes with deferoxamine, dimethylthiourea or superoxide dismutase prevented hydroxyl radical production and liqid peroxidation. While deferoxamine or dimethylthiourea did not appreciably alter iron release, superoxide dismutase prevented iron release. However, addition of deferoxamine, dimethylthiourea or superoxide dismutase even 2 min after treatment did not significantly inhibit lipid peroxidation, hydroxyl radical production and iron release. Pretreatment of microsomes with the anion channel blocker 4,4’- dithiocyano 2,′- disulphonic acid stilbine did not cause any discernible change in chemiluminiscence induced by the superoxide radical generating system but markedly inhibited lipid peroxidation without appreciably altering iron release and hydroxial radical production.  相似文献

Superoxide dismutase, an enzyme which causes the dismutation of superoxide free radical anions to generate hydrogen peroxide, has been localized in the rat ovary. The negative-staining method was used to provide photo-induced reduction of nitro-blue tetrazolium in cryostat sections of rat ovaries for histochemical localization of superoxide dismutase. Superoxide dismutase was found in growing follicles, the membrane granulosa of Graafian follicles, ovulated follicles and blood vessels. It is suggested that superoxide dismutase may play a role in regulating follicular development, ovulation and luteal functions.  相似文献

以生长在开顶箱内的油松为试材,对高浓度臭氧(80 nmol·mol-1)条件下油松(Pinus tabulaeformis)针叶中超氧阴离子自由基(O2·)产生速率、过氧化氢(H2O2)含量、超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶、脱氢抗坏血酸还原酶、单脱氢抗坏血酸还原酶、谷胱甘肽还原酶活性与抗坏血酸(ASA)含量进行测定.结果表明:高浓度臭氧使O2·产生速率提高,H2O2 和MDA含量增加.ASA含量与SOD、抗坏血酸过氧化物酶、脱氢抗坏血酸还原酶、单脱氢抗坏血酸还原酶、谷胱甘肽还原酶活性在高浓度臭氧熏蒸的前期升高,随后下降并低于对照.说明生长季前期,油松抗氧化系统对高浓度臭氧存在适应性反应,但不能抵抗长期臭氧胁迫带来的氧化伤害.  相似文献

Many nitroreductases are strongly inhibited by oxygen. The first intermediate of nitroreductase activity, the nitroaromatic anion free radical, cannot be detected in aerobic microsomal incubations. Even though the nitro compounds are unchanged, both nitrofurantoin and p-nitrobenzoate profoundly increase the NADPH-supported oxygen uptake. This catalytic oxygen consumption is partially reversed by superoxide dismutase, suggesting that superoxide anion free radical is being formed by the rapid air oxidation of the nitroaromatic anion radical.  相似文献

Accompanying the autoxidation of hydroxylamine at pH 10.2, nitroblue tetrazolium was reduced and nitrite was produced in the presence of EDTA. The rate of autoxidation was negligible below pH 8.0, but sharply increased with increasing pH. The reduction of nitroblue tetrazolium was inhibited by superoxide dismutase, indicating the participation of superoxide anion radical in the autoxidation. Hydrogen peroxide stimulated the autoxidation and superoxide dismutase inhibited the hydrogen peroxide-induced oxidation, results which suggest the participation of hydrogen peroxide in autoxidation and in the generation of superoxide radical. An assay for superoxide dismutase using autoxidation of hydroxylamine is described.  相似文献
