Indocyanine green (ICG) is a photosensitive reagent with clinically relevant diagnostic and therapeutic applications. Recently, ICG has been investigated for its utility as an exogenous chromophore during laser-induced heating. However, ICG's effectiveness remains hindered by its molecular instability, rapid circulation kinetics, and nonspecific systemic distribution. To overcome these limitations, we have encapsulated ICG within dextran-coated mesocapsules (MCs). Our objective in this study was to explore the ability of MCs to induce thermal damage in response to laser irradiation. To simulate tumorous tissue targeted with MCs, cylindrical phantoms were prepared consisting of gelatin, intralipid emulsion, and various concentrations of MCs. The phantoms were embedded within fresh chicken breast tissue representing surrounding normal tissue. The tissue models were irradiated at lambda = 808 nm for 10 min at constant power (P = 4.2 W). Five hypodermic thermocouples were used to record the temperature at various depths below the tissue surface and transverse distances from the laser beam central axis during irradiation. Temperature profiles were processed to remove the baseline temperature and influence of light absorption by the thermocouple and subsequently used to calculate a damage index based on the Arrhenius damage integral. Tissue models containing MCs experienced a maximum temperature change of 18.5 degrees C. Damage index calculations showed that the heat generation from MCs at these parameters is sufficient to induce thermal damage, while no damage was predicted in the absence of MCs. ICG maintains its heat-generating capabilities in response to NIR laser irradiation when encapsulated within MCs. Such encapsulation provides a potentially useful methodology for laser-induced therapeutic strategies. 相似文献

The presence of indocyanine green during extended traditional electrophoresis and immunoelectrophoresis of serum is associated with bimorphism of albumin. This occurs over a range of dye—albumin molar ratios an order or more greater than was obtained in similar phenomena described previously. The bimorphism seems not to be dose dependent beyond a certain point, and the two albumins so separated show tinctorial differences. The phenomenon has been observed to apparently the same degree in all normal sera tested, and may represent a means of distinguishing ligand-loaded and ligand-light serum albumin. 相似文献

Since 2007, the annual green tide disaster in the Yellow Sea has brought serious economic losses to China. There is no research on the genetic similarities of four constituent species of green tide algae at the genomic level. We previously determined the mitochondrial genomes of Ulva prolifera, Ulva linza and Ulva flexuosa. In the present work, the mitochondrial genome of another green tide (Ulva compressa) was sequenced and analyzed. With the length of 62,311 bp, it contained 29 encoding genes, 26 tRNAs and 10 open reading frames. By comparing these four mitochondrial genomes, we found that U. compressa was quite different from the other three types of Ulva species. However, there were similarities between U. prolifera and U. linza in the number, distribution and homology of open reading frames, evolutionary and codon variation of tRNA, evolutionary relationship and selection pressure of coding genes. Repetitive sequence analysis of simple sequence repeats, tandem repeat and forward repeats further supposed that they have evolved from the same origin. In addition, we directly analyzed gene homologies and translocation of four green tide algae by Mauve alignment. There were gene order rearrangements among them. With fast-evolving genomes, these four green algal mitochondria have both conservatism and variation, thus opening another window for the understanding of origin and evolution of Ulva. 相似文献

The computations of the geometries, electronic structures, dipole moments and polarizabilities for indoline and triphenylamine (TPA) based dye sensitizers, including D102, D131, D149, D205, TPAR1, TPAR2, TPAR4, and TPAR5, were performed using density functional theory, and the electronic absorption properties were investigated via time-dependent density functional theory with polarizable continuum model for solvent effects. The population analysis indicates that the donating electron capability of TPA is better than that of indoline group. The reduction driving forces for the oxidized D131 and TPAR1 are slightly larger than that of other dyes because of their lower highest occupied molecular orbital level. The absorption properties and molecular orbital analysis suggest that the TPA and 4-(2,2diphenylethenyl)phenyl substituent indoline groups are effective chromophores in intramolecular charge transfer (IMCT), and they play an important role in sensitization of dye-sensitized solar cells (DSCs). The better performance of D205 in DSCs results from more IMCT excited states with larger oscillator strength and higher light harvesting efficiency. While for TPA dyes, the longer conjugate bridges generate the larger oscillator strength and light harvesting efficiency, and the TPAR1 and TPAR4 have larger free energy change for electron injection and dye regeneration. 相似文献

The fine structure of the cells was investigated on the ultrathin sections of green sulphur bacteria, two strains of Chlorobium vibrioforme, two strains of Pelodictyon luteolum, and one strain of Pelodictyon phaeum. All strains possess similar photosynthetic structures --"chlorobium-vesicules" underlying the cytoplasmic membrane. Irregularly localized, gaseous vesicules of the rhombic shape were discerned in the cytoplasm of P. luteolum and P. phaeum. The vesicules were surrounded by a unilayer membrane. The cytoplasmic membrane produced invaginations of the mesosomal type. Elementary sulphur as a product of oxidation of hydrogen sulphide, is presumed to be liberated from the cells by means of sacs, or invaginations, formed by the cytoplasmic membrane. The taxonomy of the vibrioid green sulphur bacteria is discussed. 相似文献

The effects of colchicine on plasma elimination and biliary excretion of indocyanine green (ICG) and sulfobromophthalein (BSP) in rats were examined. Elimination of two different doses of ICG (6 mg and 20 mg/kg body weight) from plasma was significantly delayed when rats were treated with colchicine (3 mg/kg body weight) 3 h prior to the administration of the dye. On the other hand, disappearance of BSP (100 mg/kg) from plasma was not influenced by colchicine. The fact that the difference in the ICG elimination from plasma between colchicine-treated and saline-treated rats was minimal in the early period (i.e., 2 min after administration of the dye), but evident after its half-life (i.e., 10 min, when 6 mg/kg body weight of ICG was given), suggested that colchicine mainly affected the hepatocellular transport of ICG rather than the uptake of the dye by hepatocytes. Colchicine also significantly reduced the excretion of ICG (6 mg and 20 mg/kg) into bile but did not alter that of BSP (100 mg and 200 mg/kg). On the other hand, the same amount of lumicolchicine (3 mg/kg) did not have any effect on the biliary excretion of ICG. These results suggested that ICG is transported through hepatocytes into bile with the aid of the cytoplasmic microtubular system, whereas BSP is handled by hepatocytes in a different way. 相似文献

This study has investigated the expression of green fluorescent protein (GFP) variants in the cytosol and the endoplasmic reticulum (ER) of HeLa cells and evaluated the effects of the different cellular environments on the fluorescence properties of these GFP variants. Several GFP variants have been constructed by adding different N- or C-terminal signal sequences. These proteins were expressed and folded in distinct cellular compartments in HeLa cells. The localization of these GFP variants targeted to the endoplasmic recticulum was confirmed by the co-localization of DsRed2-ER as assessed by confocal microscopy. The addition of signal peptides targeting GFP variants to the ER or cytosol did not appear to alter the optical spectra of these GFP variants. However, the fluorescence intensity of these GFP variants in the ER was significantly less than that in the cytosol. Thus, the results clearly suggest that the cellular environment affects the formation and/or maturation of green fluorescence protein in vivo. These findings will be helpful in the future development and application of GFP technology aimed at investigating cellular functions performed in the ER and the cytosol. 相似文献
