A systematic approach to the taxonomic classification of the tall larkspur complex (Delphinium spp.) has been developed and implemented using molecular genetics, plant morphology, and alkaloid profiles, as shown in other papers in this series. This approach supports the classification of three distinct species (D. glaucum, D. barbeyi and D. occidentale), as the species differ in genetics and toxicity. Toxic alkaloid concentrations over the growing season were integrated with data on diet selection to make management recommendations on a species-specific basis to reduce the risk of poisoning cattle. Alkaloid concentrations in tall larkspurs in excess of 3 mg/g impart moderate or high risk to grazing cattle if sufficient quantities are consumed. D. glaucum is most toxic, with toxic alkaloid concentrations that exceed 3 mg/g throughout the grazing season until late maturity. Cattle should be denied access to dense patches of this species throughout the grazing season until after seed shatter. Concentration of toxic alkaloids in D. barbeyi is highest in vegetative plants, but D. barbeyi is unpalatable to cattle until flowering racemes begin to elongate. We recommend grazing D. barbeyi ranges early in the season when it is not palatable, then removing cattle from early flowering stage through mid-pod stage when cattle are most likely to be poisoned. Cattle can again safely graze D. barbeyi late in the season when the toxic alkaloid concentration typically declines below 3 mg/g. Some populations of D. occidentale and the D. barbeyi×D. occidentale hybrids do not contain toxic alkaloids, and pose little risk of poisoning throughout the year. Toxicity of northern populations of D. occidentale varies from year-to-year for unknown reasons. Cattle losses from D. occidentale are usually less severe than from D. barbeyi, but generally the same recommendations apply as for D. barbeyi. Toxicity sampling is an essential management tool. Tall larkspur populations can be tested for toxicity throughout the growing season. Whenever toxic alkaloid concentrations exceed 3 mg/g in leaves, flowers, or pods, cattle should be removed from the area and not returned until pods begin to shatter and risk of poisoning is lower. In general, tall larkspurs are safe to graze when alkaloid concentrations fall below 3 mg/g because it is difficult for cattle to eat sufficient larkspur to become fatally poisoned.  相似文献

Systematic approaches to taxonomic classifications of the tall larkspur spp. have been developed using traditional chemical methods to profile alkaloids, comparison of relative toxicity of individual alkaloids, plant morphology/taxonomy and molecular genetics. Using these methods (papers published in this series) toxicology of three distinct species of tall larkspurs including Delphinium glaucum, Delphinium barbeyi and Delphinium occidentale is described. Tall larkspurs (Delphinium spp.) continue to be the most serious cause of cattle losses on mountain rangelands in the western US. Over 40 norditerpenoid alkaloids have been reported in species of larkspurs and toxicology data on 25 of these have been reported by the authors. These alkaloids can be classified into three general types based on their structural characteristics and toxicity: the N-(methylsuccinyl) anthranoyl lycoctonine (MSAL)-type, having high toxicity; the lycoctonine-type, with moderate toxicity; and the 7,8-methylenedioxylycoctonine (MDL)-type, of low toxicity. The structural importance of the methylsuccinimido anthranilic acid ester group at the C18 position is evident in the high toxicity of MSAL alkaloids, particularly methyllycaconitine (MLA), Nudicauline (NUD) and 14-deacetylnudicauline (14-DAN). Other structural aspects of these alkaloids such as the C14 functionality are also important, as demonstrated by the reduced toxicity of barbinine. MLA is the alkaloid of most importance in toxicity of larkspurs on mountain rangelands because of its prevalence in most larkspurs and high toxicity. While NUD and 14-DAN also possess high toxicity, they are relatively minor components in few larkspur species (generally the plains and low larkspurs), but when present at concentrations approaching 1 mg/g dry weight they contribute significantly to overall toxicity. Deltaline (DLT) is often found in high concentrations in many larkspurs but because of low toxicity, its contribution to larkspur poisoning in the field is relatively minor and it will probably not cause toxicosis in the absence of the MSAL-type alkaloids.  相似文献

Microorganisms frequently colonize the nectar of angiosperm species. Though capable of altering a suite of traits important for pollinator attraction, few studies exist that test the degree to which they mediate pollinator foraging behavior. The objective of our study was to fill this gap by assessing the abundance and diversity of yeasts associated with the perennial larkspur Delphinium barbeyi (Ranunculaceae) and testing whether their presence affected components of pollinator foraging behavior. Yeasts frequently colonized D. barbeyi nectar, populating 54–77% of flowers examined depending on site. Though common, the yeast community was species-poor, represented by a single species, Metschnikowia reukaufii. Female-phase flowers of D. barbeyi were more likely to have higher densities of yeasts in comparison to male-phase flowers. Pollinators were likely vectors of yeasts, as virgin (unvisited) flowers rarely contained yeasts compared to flowers open to pollinator visitation, which were frequently colonized. Finally, pollinators responded positively to the presence of yeasts. Bombus foragers both visited and probed more flowers inoculated with yeasts in comparison to uninoculated controls. Taken together, our results suggest that variation in the occurrence and density of nectar-inhabiting yeasts have the potential to alter components of pollinator foraging behavior linked to pollen transfer and plant fitness.  相似文献

Summary The morphologically complex flowers of Delphinium nelsonii, D. barbeyi, and Ipomopsis aggregata are visited by a wide variety of animals. Visitors to each species range from small insects, such as worker bumblebees and solitary bees, to hummingbirds, and thus span roughly an order of magnitude in body mass and metabolic rate while flying; they also differ in type of food collected and in their efficacy as pollinators. Despite these differences, all the visitors to a given plant species fly similar, short distances between successively visited flowers and plants. There are no significant relationships between mean flight distance and metabolic rate or body mass among the visitors to any plant species. Thus there is no evidence that flight characteristics depend on anything as straightforward as whether flower visitors have high or low energetic requirements.  相似文献

Plant secondary compounds mediate interactions with insects and other animals. The norditerpene alkaloids are significant secondary compounds in Delphinium (larkspur) species which are divided into two classes: the 7, 8-methylenedioxylycoctonine (MDL-type) and N-(methylsuccinimido) anthranoyllycoctonine (MSAL-type), and are known to be toxic to herbivorous insects and livestock. Alkaloid concentrations were measured in a whole plant context in vegetative and floral tissues as well as rewards (pollen and nectar) in Delphinium barbeyi and Delphinium nuttallianum. Alkaloid concentrations differed between vegetative tissues, floral tissues and floral rewards. Alkaloid concentrations in floral parts were consistent with optimal defense theory, with tissues more closely tied to plant fitness, such as fruits, being more heavily defended than foliage. However, alkaloid concentrations were significantly lower in nectar compared to other tissues. The norditerpene alkaloids influenced the activity of bumble bees, the dominant pollinator of larkspur, but the effects were concentration dependent. Alkaloids in nectar are found at concentrations that have no effect on bee activity; however, if alkaloid concentrations in nectar were similar to those in foliage bee activity would be reduced significantly. These results suggest that nectar with low alkaloid concentrations may be beneficial to plant fitness by limiting adverse effects on pollinator activity.  相似文献

Hordem murinum is a widespread weedy/wild species, growing in different ecological conditions in Iran. Populations of H. murinum subsp. glaucum (2n = 2x = 14) and subsp. leporinum (2n = 4x, 6x = 28, 42) are found in this region. Inter-retroelement amplified polymorphism (IRAP) markers were used to analyse the genetic diversity of 57 accessions of H. murinum from different regions of Iran, and to examine patterns of diversity related to the taxonomy and geography. Eight IRAP primer combinations amplified a total of 241 distinct DNA fragments sized 150-1400 bp, from which 236 (97.9%) were polymorphic. On average, each primer combination amplified about 30.12 fragments (ranged from 23 to 34) in PCR. The patterns of genetic diversity were closely correlated with taxonomic groups, ploidy levels and geographic origin. Along with the high genetic diversity, three geographic sub-genepools were evident, 1: in the North-Northeast region along the Alborz Mountains, 2: in the West-Northwest region along the Zagros Mountains, and 3: in the Central — Southern region. The genetic diversity in diploids was higher than polyploids. Also genetic diversity in W-NW region along the Zagros Mountains was considerably higher than that of the other regions.  相似文献

Hybridization studies are important to advance our understanding of the interspecific gene flow and its evolutionary consequences in closely related species. Hybridization and admixture patterns were assessed in a contact zone and reference populations of European pine species using sequence data from 26 nuclear genes and a species-diagnostic cpDNA marker. Reference populations formed three distinct genetic clusters comprising Pinus sylvestris, Pinus mugo/Pinus uliginosa, and Pinus uncinata. Evidence of population structure was found only in P. uliginosa. Based on phenotypic characteristics and molecular data, we identified five groups of individuals in the contact zone in Poland, comprising forms of the parental species and intermediates that were most probably the result of interspecific crosses. A combination of nuclear gene sequence data and a diagnostic organelle marker were used to show that hybridization is frequent in the contact zone and results in hybrid trees with distinct phenotypic identity. The influence of selection in maintaining hybrid phenotypes in environments unsuited to parental species was inferred from nucleotide polymorphism data. A lack of admixture in reference populations suggests that hybridization has not occurred during post-glacial migration and so the contact zone represents a distinct, active example of ongoing evolution. Pine populations in this zone will be a valuable system for studying the genetic basis of hybrid advantage in environmental conditions untypical of pure parental species.  相似文献

Hybridization may be a major driver in the evolution of plant pathogens. In a high elevation Alpine larch stand in Montana, a novel hybrid fungal pathogen of trees originating from the mating of Heterobasidion irregulare with H. occidentale has been recently discovered. In this study, sequence analyses of one mitochondrial and four nuclear loci from 11 Heterobasidion genotypes collected in the same Alpine larch stand indicated that hybridization has increased allelic diversity by generating novel polymorphisms unreported in either parental species. Sequence data and ploidy analysis through flow cytometry confirmed that heterokaryotic (n + n) genotypes were not first‐generation hybrids, but were the result of multiple backcrosses, indicating hybrids are fertile. Additionally, all admixed genotypes possessed the H. occidentale mitochondrion, indicating that the hybrid progeny may have been backcrossing mostly with H. occidentale. Based on reticulate phylogenetic network analysis by PhyloNet, Bayesian assignment, and ordination tests, alleles can be defined as H. irregulare‐like or H. occidentale‐like. H. irregulare‐like alleles are clearly distinct from all known H. irregulare alleles and are derived from the admixing of both Heterobasidion species. Instead, all but one H. occidentale alleles found in hybrids, although novel, were not clearly distinct from alleles found in the parental H. occidentale population. This discovery demonstrates that Alpine larch can be a universal host favouring the interspecific hybridization between H. irregulare and H. occidentale and the hybridization‐mediated evolution of a nucleus, derived from H. irregulare parental species but clearly distinct from it.  相似文献

Lactate dehydrogenase (LDH) has been shown to play an important role in adaptation of several aquatic species to different habitats. The genomes of Daphnia pulex, a pond species, and Daphnia pulicaria, a lake inhabitant, encode two L-LDH enzymes, LDHA and LDHB. We estimated relative levels of Ldh gene expression in these two closely related species and their hybrids in four environmental settings, each characterized by one of two temperatures (10°C or 20°C), and one of two concentrations of dissolved oxygen (DO; 6.5–7 mg/l or 2–3 mg/l). We found that levels of LdhA expression were 4 to 48 times higher than LdhB expression (p<0.005) in all three groups (the two parental species and hybrids). Moreover, levels of LdhB expression differed significantly (p<0.05) between D. pulex and D. pulicaria, but neither species differed from the hybrid. Consistently higher expression of LdhA relative to LdhB in both species and the hybrid suggests that the two isozymes could be performing different functions. No significant differences in levels of gene expression were observed among the four combinations of temperature and dissolved oxygen (p>0.1). Given that Daphnia dwell in environments characterized by fluctuating conditions with long periods of low dissolved oxygen concentration, we suggest that these species could employ regulated metabolic depression to survive in such environments.  相似文献

Here we report the variation of diterpenes from three populations of the brown alga Dictyota mertensii. The populations were analyzed by 1H NMR and GC-MS techniques. Both chemical GC-MS and 1H NMR profiles of D. mertensii presented diterpenes as major constituents. In all populations, we detected the prenylated guaiane diterpenes, dictyoxide, pachydictyol A, isopachydictyol A, dictyol B and its acetate, and dictyol C. Although dictyol H has been described as a taxonomic marker, this diterpene was found in only two of the three studied populations. This result reinforces the need for studies of temporal and geographic variation for the establishment of the taxonomic limits of the species chemical patterns. It was also possible to observe mass spectra fragmentation and proton chemical shift in the NMR spectra characteristic from xeniane diterpenes, for the first time in this species, which led to the detection of dictyodial, suggesting that D. mertensii belongs to the chemotaxonomic group of species D. ciliolata, D. jamaicensis, and D. menstrualis, all of which are producers of diterpene from chemical groups I and III.  相似文献

Tibouchina urvilleana Cogn. is native to southern Brazil and currently cultivated as an important ornamental shrub in frost free areas around the world. Its rapid vegetative growth and sterility suggests that it might be of hybrid origin. In this study, Internal Transcribed Spacer (ITS) of nuclear ribosomal DNA and chloroplast trnL-trnF spacer from T. urvilleana and three other congeneric species were sequenced to test its hybrid status. Cloning sequencing revealed two distinct types of ITS sequences from T. urvilleana, with one type almost identical with Tibouchina aspera. Genetic distance between the two types was much larger than the average interspecific genetic distances calculated from other Tibouchina species. Sequencing of chloroplast trnL-trnF spacer showed that T. urvilleana has identical sequence with T. aspera, but differed from other congeneric species by one nucleotide substitution and two indels. Molecular data demonstrated clearly that T. urvilleana indeed was a hybrid, with T. aspera or closely related species acting as the maternal parent.  相似文献

Background and Aims

DNA sequence similarities and hybridization patterns in Trifolium (clovers) section Trifoliastrum suggest that rapid radiation from a common ancestral source led to this complex of diverse species distributed across Europe, western Asia and North Africa. Two of the most geographically and ecologically divergent of these species are the rhizomatous T. ambiguum from high altitudes in eastern Europe and western Asia and the stoloniferous T. occidentale from sea level in western Europe. Attempts were made to hybridize these species to ascertain whether, despite this separation, gene flow could be achieved, indicating the retention of the genetic factors necessary for hybridization.

Methods

Three F1 hybrids formed after embryo rescue were described, characterized by conventional and molecular cytogenetics, subjected to fertility tests and progeny generations were developed.

Results and Conclusions

Partially fertile hybrids between Trifolium ambiguum and T. occidentale were obtained for the first time. The F1 hybrids produced seeds after open-pollination, and also produced triploid progeny in backcrosses to T. occidentale from the functioning of unreduced gametes in the hybrids. These plants were fertile and produced progeny with T. occidentale and with T. repens. Meiotic chromosome pairing in the F1 showed six to eight bivalents per pollen mother cell, indicating pairing between the parental genomes. A chromosome-doubled form of one hybrid, produced using colchicine, showed some multivalents, indicative of interspecific chromosome pairing. The hybrid plants were robust and combined phenotypic characteristics of both species, having stolons, thick roots and a few rhizomes. Results show that despite separation by the entire breadth of Europe, the speciation process is incomplete, and these taxa have partially retained most of the genetic compatibilities needed for hybridization (possibly except for endosperm development, which was not tested). The fertile progeny populations could lead to new clover breeding strategies based on new hybrid forms.  相似文献

A small scale attempt to maintain and rear the worm Halla parthenopeia in laboratory conditions was conducted. Five bivalve species (Paphia undulata, Cerastoderma glaucum, Venerupis pullastra, Ruditapes decussata, and Gafrarium pectinatum) were used to investigate preferred food item, feeding rate and growth of the worm. Hallaparthenopeia are capable of using different types of clams, although they grew better with Paphia undulata and C.glaucum as food items. The highest average daily predation rates in case of P. undulata as a prey were 1.73, 2.13 and 2.57 individuals per predator per day for small, medium and large groups of H. parthenopeia, respectively. The daily predation rate on C. glaucum was low with an average: 0.50, 0.63 and 0.73 individuals per predator per day for the small, medium and large worm groups, respectively. The daily growth rate of H. parthenopiea increased when it was fed P. undulata (average: 0.083, 0.071 and 0.038 g/day for small, medium and large worm groups respectively), compared to an average of 0.044, 0.034 and 0.020 g/day for small, medium and large worm groups, respectively, when worms were fed with C. glaucum. The biochemical composition of three different sizes of the worm was also determined. Protein was the highest biochemical constituent with an average of 51% of the dry weight, followed by lipids with an average of 25.88% of the dry weight; meanwhile carbohydrate was present at an average of 20.72%. Our findings indicate that growth of H. parthenopeia can be improved when fed with a suitable prey item and suggest that it is feasible to successfully culture this protein-rich worm in captivity.  相似文献

Convolvulaceous species have been reported to contain several bioactive principles thought to be toxic to livestock including the calystegines, swainsonine, ergot alkaloids, and indole diterpene alkaloids. Swainsonine, ergot alkaloids, and indole diterpene alkaloids are produced by seed transmitted fungal symbionts associated with their respective plant host, while the calystegines are produced by the plant. To date, Ipomoea asarifolia and Ipomoea muelleri represent the only Ipomoea species and members of the Convolvulaceae known to contain indole diterpene alkaloids, however several other Convolvulaceous species are reported to contain ergot alkaloids. To further explore the biodiversity of species that may contain indole diterpenes, we analyzed several Convolvulaceous species (n = 30) for indole diterpene alkaloids, representing four genera, Argyreia, Ipomoea, Stictocardia, and Turbina, that had been previously reported to contain ergot alkaloids. These species were also verified to contain ergot alkaloids and subsequently analyzed for swainsonine. Ergot alkaloids were detected in 18 species representing all four genera screened, indole diterpenes were detected in two Argyreia species and eight Ipomoea species of the 18 that contained ergot alkaloids, and swainsonine was detected in two Ipomoea species. The data suggest a strong association exists between the relationship of the Periglandula species associated with each host and the occurrence of the ergot alkaloids and/or the indole diterpenes reported here. Likewise there appears to be an association between the occurrence of the respective bioactive principle and the genetic relatedness of the respective host plant species.  相似文献

The oegopsid squid family Onychoteuthidae presently comprises six genera (Moroteuthis, Onychoteuthis, Ancistroteuthis, Kondakovia, Onykia and Chaunoteuthis) but the status of some of these is still uncertain. An interdisciplinary study was undertaken to clarify the phylogenetic relationships, which included a morphological approach (morphometric analysis), and a molecular study of a mitochondrial gene portion (l-rRNA gene, 16S). The morphometric analysis identified two groups, one including some Onychoteuthis and some Moroteuthis knipovitchi, and another including the remaining genera or species. At the intrageneric level, M. knipovitchi appears to be separated from the other species of the genus; M. robusta is closely related to M. ingens and M. pacifica. Morphometric analysis confirmed that Kondakovia longimana is different from M. ingens. Likewise, Onychoteuthis was clearly separated from the other genera, but there is neither geographical grouping nor morphometric differentiation of the Onychoteuthis species with the parameters measured. Some specimens were apparently intermediate between the Onychoteuthis and Ancistroteuthis groups. On a molecular basis, the Onychoteuthidae appeared to be monophyletic. Monophyly of the genus Moroteuthis (four species studied) was not strongly supported: M. knipovitchi was distinct from the others. The molecular analysis showed three Hawaiian species, Onychoteuthis compacta, O. sp. B and O. sp. C, to be closely related. The gene sequence for the newly created species of Onykia is clearly different from all the others, indicating that it is a true species.  相似文献

Centromeres are composed of long arrays of satellite repeats in most multicellular eukaryotes investigated to date. The satellite repeat–based centromeres are believed to have evolved from “neocentromeres” that originally contained only single- or low-copy sequences. However, the emergence and evolution of the satellite repeats in centromeres has been elusive. Potato (Solanum tuberosum) provides a model system for studying centromere evolution because each of its 12 centromeres contains distinct DNA sequences, allowing comparative analysis of homoeologous centromeres from related species. We conducted genome-wide analysis of the centromeric sequences in Solanum verrucosum, a wild species closely related to potato. Unambiguous homoeologous centromeric sequences were detected in only a single centromere (Cen9) between the two species. Four centromeres (Cen2, Cen4, Cen7, and Cen10) in S. verrucosum contained distinct satellite repeats that were amplified from retrotransposon-related sequences. Strikingly, the same four centromeres in potato contain either different satellite repeats (Cen2 and Cen7) or exclusively single- and low-copy sequences (Cen4 and Cen10). Our sequence comparison of five homoeologous centromeres in two Solanum species reveals rapid divergence of centromeric sequences among closely related species. We propose that centromeric satellite repeats undergo boom-bust cycles before a favorable repeat is fixed in the population.  相似文献

Apogamous fern species are often difficult to distinguish from related species because of their continuous morphological variations. To clarify the genetic relationships among the members of the Dryopteris varia complex, we analyzed the nucleotide sequences of the plastid gene rbcL and the nuclear gene PgiC. We also analyzed the diploid sexual species D. caudipinna and D. chinensis, which have not been included in the complex, but were recently shown to be closely related to the complex in a molecular phylogenetic study. The PgiC sequences of the diploid sexual species, D. varia, D. saxifraga, D. sp. ‘protobissetiana’ (undescribed diploid sexual species), D. caudipinna, and D. chinensis, were well differentiated and hence designated A, B, C, D, and E, respectively. Thus, the PgiC constitution of apogamous species in the complex was as follows: D. bissetiana, B + C; D. kobayashii, B + C + E); D. pacifica, A + C, A + B + C, or A + C + D; D. sacrosancta, A + C + E; and D. saxifragivaria, B + C. These results suggest that these apogamous species are formed by hybridizations of species including not only the three diploid sexual species of the D. varia complex (A, B, and C) but also the two diploid sexual species D. caudipinna (D) and D. chinensis (E), which do not belong to the complex.  相似文献

Hierarchy theory has provided a valuable conceptual framework for studies of heterogeneity. However, there have been few empirical studies of hierarchical structure and little is known about how hierarchical structure originates or varies among systems. Here, I explore how scale dependency can influence the detection of hierarchical structure. Specifically, I compared how heterogeneity changed with scale in patches of larkspur (Delphinium spp). The distribution of Delphinium nelsonii inflorescences was quite uniform over the range of measured scales (1 m2-2500 m2) and only a single level of patchiness was observed. Aggregations of D. barbeyi inflorescences were much more pronounced and this patchiness was evident at many scales. The number of hierarchical levels and the scales at which patchiness occurred varied both within and between plots of D. barbeyi. Because patchiness was not strongly scale-dependent for either species, discrete patch boundaries and well-defined hierarchical levels were not usually apparent even when multiple scales of patchiness were present. In several cases, the scales of detected patchiness depended on the difference in scale between grain (resolution) and extent. I predict that because of the lack of dominant and strongly scale-dependent processes, microlandscapes such as Delphinium meadows, may be less likely to exhibit well-defined hierarchical structure than larger-scale landscapes, especially those heavily altered by human activities.  相似文献

Investigation of the production of secondary metabolites of Heterobasidion occidentale led to the isolation and identification of six sesquiterpenes (illudolone A and B, illudolactone A and B, deoxyfomannosin A and B) along with the well-known sesquiterpene fomannosin and the previously described benzohydrofuran fomannoxin. The structures and relative configurations of the compounds were determined by 1D and 2D NMR spectroscopic analysis as well as by HRMS. Their absolute configuration and biosynthesis were suggested and discussed in relation to fomannosin. Four compounds showed growth inhibiting activity against several basidiomycetes, Phlebiopsis gigantea, Phanerochaete chrysosporium and H. occidentale, and toxicity towards the moss Physcomitrella patens. In addition, one compound displayed activity against the bacterium Variovorax paradoxus as well as against the ascomycete Fusarium oxysporum.  相似文献
