The importance of the gut microorganisms in the termites Nasutitermes exitiosus and Coptotermes lacteus was investigated by feeding them with antibiotics. With N. exitiosus, antibiotics which killed both the bacteria and the spirochaetes (ampicillin, kanamycin, chloramphenicol, erythromycin, cephaloridine, tetracycline) reduced the life span of the termite from 250 days to about 13 days, whereas antibiotics which had little effect on the flora (penicillin, methicillin) did not greatly reduce the life span of the termite. The essential role of the spirochaetes in N. exitiosus was shown by feeding metronidazole, or exposing the termites to pure oxygen. Both treatments killed the spirochaetes, but not the bacteria, resulting in a life span for the termite of 13–22 days. Acid fuchsin did not kill the spirochaetes. Fungi were not essential for N. exitiosus. In C. lacteus all treatments, except that with acid fuchsin, killed the protozoa, thereby reducing the life span of the termite from 69 days to 6–29 days.  相似文献

The redox potential and pH of the gut of the termites Nasutitermes exitiosus and Coptotermes lacteus was investigated by feeding the insects with redox dyes and pH indicators. For N. exitiosus the E′0 (pH 7.0) of the foregut was above +200 mV; the midgut was about +100 to +150 mV and the hindgut was in the region of ?20 to +30 mV. For C. lacteus the fore- and midgut were about +30 to +50 mV and the hindgut about ?20 to +20 mV. The colours of the ingested dyes indicated that the gut was aerobic in both termites. The pH of the whole gut ranged from 6.5 to 7.5 for N. exitiosus and 6.0 to 7.0 for C. lacteus.  相似文献

The hindgut of the lower termites, Mastotermes darwiniensis and Coptotermes lacteus and the higher termite Nasutitermes exitiosus were made aerobic by exposure of the termites to pure oxygen, a procedure which killed their spirochaetes and their protozoa (lower termites only). The time taken for the hindgut to become anaerobic after the termites were restored to normal atmospheric conditions ranged from 2 to 4.5 hr. After oxygen treatment the number of gut bacteria increased some six- to ten-fold in all termite species, indicating that the bacteria are poised to use oxygen entering the gut. Removal of all the hindgut microbiota by feeding tetracycline caused the hindgut to become aerobic in M. darwiniensis and N. exitiosus. The transferring of M. darwiniensis to fresh wood, free of antibiotic, resulted in the return of the normal flora and the eventual establishment of anaerobic conditions in the hindgut. Thus the bacteria appear to be important in maintaining anaerobic conditions in the gut. Attempts to determine whether the protozoa (in the lower termites) played any part in maintaining the Eh of the hindgut were unsuccessful. Serratia marcescens failed to colonise the gut of normal C. lacteus and transiently colonized (for 5 days) the gut of normal N. exitiosus. Transient colonization by S. marcescens (from 6 to 10 days) occurred in N. exitiosus when its hindgut spirochaetes were killed and in C. lacteus when its spirochaetes and protozoa were killed, indicating a possible role for the spirochaetes and/or protozoa in influencing the bacteria allowed to reside in the hindgut. Exposure of normal termites to Serratia provoked an increase in the numbers of the normal gut bacteria.  相似文献

The redox potential of the gut of nine species of termites was investigated by feeding the insects with redox dyes. The fore- and mid-gut of all species was aerobic with an E′o probably in excess of + 100 mV. whereas the paunch and colon were anaerobic with an E′o of about ?230 to ?270 mV, except in Coptotermes lacteus and Nasutitermes exitiosus whose colons were at a E′o of ?50 to ?125 mV. In four species (Incisitermes barretti, Glyptotermes brevicornis, Stolotermes victoriensis, Coptotermes lacteus) the rectum was aerobic (E′o about +60 mV), whereas the rectum of the other species was anaerobic (E′o from about ?125 to ?270 mV).  相似文献

The cellulase of the higher termite Nasutitermes exitiosus was located in the foregut (19%), the midgut (59%), the mixed segment (14%) and in the hindgut (8%). Removal of the gut flora by feeding tetracycline or by starving the termite did not affect the activity of the enzymes indicating that the termite secretes its own cellulase and is not dependent on its gut flora for the digestion of cellulose. The cellulase of the lower termite Coptotermes lacteus was distributed through the foregut (19%), midgut (32%) and hindgut (49%). Removal of the gut flora and fauna, left the specific activity of the cellulase of the fore- and mid-gut largely unaffected, but led to a 20% decrease in the specific activity of the cellulase in the hindgut. In starved C. lacteus the distribution of cellulase activity was foregut, 44%; midgut, 30% and hindgut, 26%. These results indicate that C. lacteus synthesises its own cellulase in addition to using the gut protozoa for cellulose digestion.  相似文献

The major gut bacteria of the worker caste of nine species of Australian termites, belonging to four families, were isolated and identified to generic level. All species were either facultative anaerobes or strict aerobes. A correlation appears to exist between the major gut bacterium and the family to which the termite belongs. The major bacterium from the two lowest termites, Mastotermes darwiniensis (family Mastotermitidae) and Cryptotermes primus (family Kalotermitidae), was Streptococcus; from four species belonging to the Rhinotermitidae (Heterotermes ferox, Coptotermes acinaciformis, C. lacteus, Schedorhinotermes intermedius intermedius) it was Enterobacter; and from three species of the Termitidae (Nasutitermes exitiosus, N. graveolus, N. walkeri) it was Staphylococcus. Enterobacter was a minor symbiont of M. darwiniensis, C. primus, and N. graveolus; Streptococcus was a minor symbiont of H. ferox, C. lacteus, S. intermedius intermedius, and N. exitiosus; and Bacillus was a minor symbiont of C. acinaciformis and S. intermedius intermedius. M. darwiniensis possessed another minor symbiont tentatively identified as Flavobacterium. C. acinaciformis from three widely separated locations possessed a similar microbiota, indicating some form of control on the composition of the gut bacteria. Bacteria, capable of growth on N-free medium in the presence of nitrogen gas, were isolated from all termites, except N. exitiosus and N. walkeri, and were identified as Enterobacter. No cellulose-degrading bacteria were isolated.  相似文献

Some parameters of nitrogen utilization between the ambrosia beetle Xyleborus dispar in mutualistic association with its symbiotic fungus Ambrosiella hartigii, were examined. Qualitative and quantitative analyses of the major nitrogenous excretory products were made on the various life stages of X. dispar. The main nitrogenous product found in excreta and hindguts of beetles, larvae, and pupae, was uric acid (range 7·6–14·8 μg uric acid/beetle). No ninhydrin-positive compounds were located in excreta of the beetles. The concentration of ammonia-nitrogen in the various life stages averaged between 0·70 and 1·13 μg NH3-N/beetle.Total nitrogen determinations were made on sapwood samples of Malus sylvestris (0·34 ± 0·005% N by dry weight), attacked wood, ‘pre-brood’ (0·31 ± 0·005% N by dry weight), and attacked wood ‘post-brood’ (0·17 ± 0·02% N). Similar determinations of the artificial medium (l-asparagine medium) indicated that a nitrogen requirement of about 0·08–0·1% N by dry weight was necessary before oviposition could occur.Fixation of atmospheric nitrogen by individual X. dispar beetles in vitro was not indicated using the acetylene ethylene reductase method. Proteolytic enzyme activity was not found on examination of diapause beetles, their excreta, larval and pupal excreta, and the ambrosial and mycelial forms of A. hartigii.Comparative concentrations of soluble proteins and free amino acids suggested that fungus in the mycangia was built up from free amino acids of the insects. At the period of emergence, flight, and attack of new hosts, the females were found to have a concentration of soluble proteins more than double that found in the beetles during the remainder of the year. Extracts of tissues of the lower termites, Reticulitermes flavipes and Coptotermes lacteus, and the higher termite, Nasutitermes exitiosus, possess acetyl-CoA synthetase and all the enzymes of the tricarboxylic acid cycle and are thus able to oxidize acetate to CO2. The specific activities of these enzymes in R. flavipes are sufficient to cope with the rate of acetogenesis by the gut microbiota. The presence of the malic enzyme and malate dehydrogenase, but not pyruvate carboxylase or phosphoenolpyruvate carboxykinase, indicates that they may be important as anaplerotic enzymes for the conversion of pyruvate to oxalacetate. An apparent absence of pyruvate dehydrogenase in all termites suggests that they do not convert pyruvate to acetyl-CoA, but rather convert acetate (transported from the hindgut) to this compound. All the enzymes of glycolysis were present in termite extracts. Thus any glucose absorbed from the midgut, and originating from hydrolysis of cellulose by salivary or midgut enzymes, can be metabolized by termites as an energy source.  相似文献

Summary Experiments were conducted to determine the effect on biological dinitrogen fixation byCasuarina of available nitrogen (N) in the substrate and competition by interplantedEucalyptus. In these experiments, combined N was applied to the plants after nodules were developed and functioning. Both environmental factors, nitrate and competition, were observed to influence biological dinitrogen fixation byCasuarina, but not yield (total dry weight). In one experiment, the proportion of nitrogen derived byCasuarina from atmospheric fixation (pNdfa) was observed to be inhibited by potassium nitrate in a linear fashion. However, substrate N did not significantly affect the weight of root nodules. Thus nodule dry weight was not highly correlated with the proportion of nitrogen fixed. In a second experiment, the presence of a non-fixing interplanted species,Eucalyptus, increased dinitrogen fixation inCasuarina.Casuarina interplanted withEucalyptus obtained a greater proportion of its nitrogen (94.75%) from fixation than didCasuarina grown alone (86.68%) suggesting that competition for substrate N influences the proportion of nitrogen fixed by this actinorhizal plant.Dedicated to the memory of Professor John G. Ninety-three isolates ofMetarhizium anisopliae,mostly derived from a survey of termite material, were screened for activity againstNasutitermes exitiosusandCoptotermes frenchiorC. acinaciformisusing a grooming assay technique. Twenty-six of the most promising isolates were further evaluated by bioassay againstN. exitiosusandC. acinaciformis.All isolates were pathogenic withCoptotermesspp. being more susceptible thanN. exitiosus.A group of nine isolates, chosen for their level of pathogenicity for one or other genus of termites and to represent a genetically diverse group, was finally compared in a minicolony test using termite colonies in 1 liter jars. The isolate, code-named FI-610 (derived from nest-mound material ofC. lacteusin SE New South Wales), was one of the most effective isolates against termites from both of the two colonies tested. This isolate also grew relatively well on agar plates at 36°C. FI-610 was thus selected for field trials and was found to be effective in killing colonies ofC. acinaciformiswhen 10 g (=3 × 1011conidia) or more of conidial powder was blown into the center of the large mound colonies.  相似文献

[14C](lignin)-Acer rubrum L. was produced by infusing stems of A. rubrum with [14C](3′-side chain)-cinnamic acid. Groups (1g) of Nasutitermes exitiosus (Hill) were placed in sealed flasks which were aspirated over a 2-week period. These released up to 8.3% of the [14C](lignin)-A. rubrum as 14CO2. Termites maintained under standard conditions as 50 g non-breeding groups for 4 months or more showed diminished ability to degrade lignin. Optimal lignin degradation and survival of N. exitiosus was at 90 and 96% relative humidity (r.h.). At 75, 80 and 85% r.h., fungal growth in bioassay flasks was seen, but lignin degradation did not increase. At 100% r.h. where bacterial growth in faeces may have been encouraged due to the development of free water, termite survival was poor and lignin degradation decreased. Starved termites contained much more radioactivity (21.8 and 30.8%) than fed termites (1.6% radioactivity), probably due to greater coprophagy on deprivation of food. However, lignin degradation was only marginally higher in starved termites, suggesting lignin becomes progressively more resistant to termite degradation after passage through the gut.  相似文献

The aerobic hydrogen-oxidizing bacterium Alcaligenes latus represented by three strains was found to be able to grow with dinitrogen as the sole nitrogen source: The doubling time of total (Kjeldahl) nitrogen during growth on glucose at 30°C under an atmosphere containing 2% (v/v) oxygen in dinitrogen amounted to 39 h, while that in the presence of ammonium was 3 h. Nitrogen fixation did apparently not occur under air. During diazotrophic growth the cells accumulated up to 75% (w/dry weight) poly--hydroxybutyric acid. The efficiency of nitrogen fixation varied between 10 and 15 mg N per g glucose utilized. The specific nitrogenase activity measured in the acetylene reduction assay amounted to 5–17 nmol C2H4 formed per min and mg protein.  相似文献

Chlamydospores of Phytophthora cinnamomi Rands have been shown to survive in the intestinal tracts of termites (Nasutitermes exitiosus) and two species of forest birds indigenous to West Australian jarrah forests. Viable chlamydospores were recovered from bird feces within the normal rate of passage time for food through the gut. The unusual imino acid, N-methyl-trans-4-hydroxy-Lproline has been isolated from leaves of five species of the leguminous tropical tree Copaifera, and for the first time characterized by 1H and 13C NMR and mass spectrometry. This imino acid can constitute up to 3% of the mature leaf dry weight and 10% of the nitrogen; it also constitutes 2–3 % of the dry weight of the seed. Preliminary feeding trials have shown it to be a very effective inhibitor of larval development of the seed-feeding bruchid beetle Callosobruchus maculatus and to have significant feeding deterrence of the leaf-feeding lepidopteran Spodoptera littoralis. Phenolic compounds, also known to affect herbivores adversely, comprise 6–10% leaf dry weight. However, the imino acid displayed a mean of 50% reactivity compared to standards commonly used in analysis of total phenolics by the Folin-Denis (F-D) assay [gallic acid, tannic acid and (+) catechin], thus resulting in a significant overestimate of phenolics by this assay. It is concluded that assessment of leaf food quality for herbivores of Copaifera by assays widely used for this purpose in ecological studies, such as F-D for phenolics and Kjeldahl for total nitrogen content, give some misleading results.  相似文献

  • 1.1. The main chemical components of Meganyctiphanes norvegica (M. Sars), Thysanoessa inermis (Krøyer) and T. raschii (M. Sars) have been examined.
  • 2.2. Protein accounted for 42–47% of the dry weight of M. norvegica and 32–50% of the dry weight of the Thysanoessa species. On a wet weight basis, the protein content was relatively constant and independent of season.
  • 3.3. The dominating amino acids in the bulk protein of the krill were glutamic acid/glutamine, aspartic acid/asparagine, glycine, alanine, lysine and leucine.
  • 4.4. Lipids were present in amounts of 13–29% of the dry weight in M. norvegica, 15–50% in T. inermis and 12–44% in T. raschii, and the lipid content varied with season.
  • 5.5. The main nitrogen extractives in krill, expressed on a dry weight basis, were free amino acids (5–10%), trimethylamine oxide (about 4%), peptides (about 1%) and nucleotides (0.4–1.3%). Trimethylamine and ammonia were present in very low concentrations in living krill.
  • 6.6. The amino acids taurine, glycine, proline, arginine, sarcosine and alanine made up 89–93 mol% of the free amino acid pool.
  • 7.7. The ash content of krill was in the order of 10–13% of the dry weight, and fluoride represented 1040 and 3200 ppm in the Thysanoessa species and M. norvegioca, respectively.

Since fat accumulation takes place in many algae as a response to exhaustion of the nitrogen supply, it has been suggested that this may provide a means of enhancing the potential food value of algae. To test this possibility, chemostatic continuous cultures of Chlorella sorokiniana and Oocystis polymorpha were subjected to successive reductions in influent nitrogen. As cellular nitrogen content decreased from about 10 to 4%, oxygen evolution, carbon dioxide uptake, chlorophyll content, and tissue production were drastically reduced, but total lipid content was essentially unchanged. Caloric values and C, H, and N analyses suggested a moderate increase in carbohydrate content, but gas chromatographic analyses revealed no significant qualitative or quantitative changes in the fatty acid fraction. In batch-cultured cells, nitrogen could be reduced to 3% of dry weight, causing a concomitant increase in total fatty acids and pronounced changes in the composition of the fatty acid fraction. These results suggest that cellular nitrogen must fall to approximately 3% of dry weight before appreciable increases in lipid synthesis can occur. Cell nitrogen is then apparently completely bound in essential cell constituents, and carbon subsequently fixed is converted into lipid products. The findings indicate that nitrogen limitation may be useful in increasing the food quality of batch-cultured cells, but the technique has little value for continuous culture systems per se.  相似文献

The process of ensiling was studied in fresh maize (15% dry matter (DM)), wilted maize (18 and 24% DM) and maize mixed with 5–20% of wheat straw (18, 25 and 29% DM). Silages with 24% DM were preserved better than those with lower dry matter content. There was a significant change, with time, in pH, titrable acidity, volatile fatty acids, lactic acid, number of lactic acid bacteria, volatile nitrogen and soluble sugars in all the treatments. There was a significant decline in volatile fatty acids (P<0.05) and ammonia (P<0.01) production, and a significant increase in soluble sugar (P<0.01) in silages made after wilting. A significant decline in titrable acidity (P<0.01), volatile fatty acid production (p<0.05) and ammonia nitrogen (P<0.01), and a significant increase in pH (P<0.01) were found in silages of maize mixed with wheat straw. The overall rate of fermentation decreased during the first few days of fermentation in wilted and wheat straw silages, but the final products had characteristics of a good silage. In the second experiment the effect of urea and molasses was studied on wheat straw plus maize (15:85) silage with an initial DM content of 31–34%. Three levels of molasses (0, 3 and 6% of fresh weight) and two levels of urea (0 and 0.5% of fresh weight) were studied. Urea treatment with 3% molasses was found to be the best on the basis of silage characteristics.  相似文献

对基质育苗后水培的菘蓝进行缺氮与复氮处理,分析其生长情况及氮代谢产物含量的变化,探讨缺氮和复氮对菘蓝幼苗生长及氮代谢的影响,以提高菘蓝产量和品质以及栽培过程中的氮素利用效率。结果显示:(1)正常供氮条件下,菘蓝幼苗的叶绿素含量、谷氨酰胺合成酶(GS)活性、硝态氮含量、靛玉红含量为最高,而其株高、主根直径、根的鲜重与干重、叶的鲜重与干重、根系活力均最小。(2)缺氮处理增加了菘蓝幼苗的主根直径和根干重,提高其根系活力和硝酸还原酶(NR)活性,促进游离氨基酸在叶中的积累;但降低了GS的活性,也降低了叶中硝态氮、可溶性蛋白、靛玉红及根中游离氨基酸的含量;缺氮对叶中靛蓝的含量无明显影响。(3)复氮处理增加了菘蓝幼苗的株高、主根长、根鲜重、叶鲜重、叶干重,提高了其根系活力,降低了NR和GS的活性;与对照相比,复氮降低了叶中硝态氮含量,提高了叶中可溶性蛋白、靛蓝及根中游离氨基酸的含量,但对叶中游离氨基酸和靛玉红含量影响较小。研究表明,缺氮后再复氮有利于菘蓝幼苗叶的生长,同时有利于增加其叶内靛蓝含量,从而提高其产量和品质。  相似文献

Two green algae (Chlorella vulgaris and Scenedesmus obliquus) and four blue-green algae (Anacystis nidulans, Microcystis aeruginosa, Oscillatoria rubescens and Spirulina platensis) were grown in 81 batch cultures at different nitrogen levels. In all the algae increasing N levels led to an increase in the biomass (from 8 to 450 mg/l), in protein content (from 8 to 54 %) and in chlorophyll. At low N levels, the green algae contained a high percentage of total lipids (45 % of the biomass). More than 70 % of these were neutral lipids such as triacylglycerols (containing mainly 16:0 and 18:1 fatty acids) and trace amounts of hydrocarbons. At high N levels, the percentage of total lipids dropped to about 20 % of the dry weight. In the latter case the predominant lipids were polar lipids containing polyunsaturated C16 and C18 fatty acids. The blue-green algae, however, did not show any significant changes in their fatty acid and lipid compositions, when the nitrogen concentrations in the nutrient medium were varied. Thus the green but not the blue-green algae can be manipulated in mass cultures to yield a biomass with desired fatty acid and lipid compositions. The data may indicate a hitherto unrecognized distinction between prokaryotic and eukaryotic organisms.  相似文献

The characteristic coating of frass of last-instar tobacco hornworms, Manduca sexta, reared on artificial diet, proved to be uric acid. Results indicated that uric acid is the major nitrogenous excretory product; during most of the larval feeding stage, 5–7% of the excreta (dry weight basis) was uric acid; only minute quantities of allantoic acid were present. The rate of uric acid excretion was linear for the periods when coated pellets were observed. Abrupt increases in uric acid resulted from delays in pellet expulsion associated with delays in feeding activity. A distinctive coating was not generated by penultimate instar larvae, but abrupt changes in uric acid content did occur, which suggests that the phenomenon of coated frass is directly related to a differential in uric acid concentration. The source of uric acid in the frass was the Malpighian tubule system. The transition period between feeding and the wandering stage was a time of rapid decrease in uric acid excretion; there were only low levels in the last fecal pellets and none in Malpighian ampullae of wandering-stage larvae. Since the first appearance of coated fecal pellets preceded the release of ecdysone by about 24 hr, the involvement of this hormone was not indicated.  相似文献
