Activity of the enzyme glutamine synthetase (GS, EC 6.3.1.2) was determined in vitro for roots of the marine angiosperm Zostera marina L. (eelgrass) collected from a population in Great Harbor, Woods Hole, Massachusetts, U.S.A. The GS synthetase activity was lowest in roots of plants collected from the shallow region of the eelgrass bed (12.0 μmol·g−1 (fresh wt)· h−1) and increased in the mid (3.0 m, 40.3 μmol·g−1 (fresh wt)·h−1) and deep (5.0 m, 72.3 μmol·g−1 (fresh wt)·h−1) plant collection depths. GS transferase activity increased with collection depth in a similar manner: shallow, 28.6 μmol·g−1 (fresh wt)·h−1; mid, 52.0 μmol·g−1 (fresh wt)·h−1; deep, 92.8 μmol·g−1 (fresh wt)·h−1. When sediment-embedded plants were held in continuous darkness for 2 days to create extended root anoxia, root GS activities nearly doubled. In contrast, in vivo incorporation of 14C-glutamate into glutamine and protein residue remained constant or declined under short-term hypoxia and anoxia. During aerobic recovery from anoxia, root labelling of glutamine and protein increased markedly. Free amino acid patterns of eelgrass roots growing in situ were determined over a diurnal cycle. Total free amino acid content was maximal at dawn and decreased 50% by noon. In contrast, the proportion of glutamine was lowest at dawn and maximal at noon for both shallow and deep-growing plants. Despite differences in depth-specific plant sizes, root/rhizome/shoot ratios, and relative growth rates, the daily whole plant nitrogen demand of shallow and deep growing plants were equivalent. When corrected for assay temperature response, the enzyme synthetase activities measured in vitro suggest that all of the plant nitrogen assimilation requirements can be met within daylight hours during the period of peak summer biomass.  相似文献

Interaction between D-glucuronic acid and Zn(II), Cd(II), and Hg(II) metal ion salts has been studied in solution and solid complexes of the type M(D-glucuronate)X · nH2O and M(D-glucuronate)2·nH2O, where M = Zn(II), Cd(II), and Hg(II), X = Cl or Br, and n = 0–2 were isolated and characterized. Spectroscopic and other evidence indicated that in the metal-halide-sugar complexes the Zn(II) and Cd(II) ions bind to two D-glucuronate moieties via 06, 05 of the carboxyl oxygen atoms of the first and 04, 06' of hydroxyl and carbonyl groups of the second as well as to two H2O molecules, whereas in the corresponding M(D-glucuronate)2 · nH2O salts, the metal ions are bonded to two sugar anions through 06 and 06' of the ionized carboxyl groups and two water molecules, resulting in a six-coordination around each metal cation. The Hg(II) ion binds to 06 and 05 oxygen atoms of a sugar anion and to a halide anion or water molecule, in the Hg(D-glucuronate)X·nH2O compounds, while in the corresponding metal-glucuronate salt mercury is bonded to 06 and 06' of the two glucuronate anions with four-coordination around the Hg(II) ion. The β-anomer sugar conformation is predominant in the free acid and in these series of metal-sugar complexes.  相似文献

All 20.000 different fish species vary greatly in their ability to tolerate and survive fluctuating oxygen concentrations in the water. Especially fish of the genus Carassius, e.g. the crucian carp and the goldfish, exhibit a remarkable tolerance to limited/absent oxygen concentrations. The metabolic changes of anoxia-tolerant crucian carp were recently studied and published. Contrary to crucian carp, the hypoxia-tolerant common carp cannot survive a complete lack of oxygen (anoxia). Therefore, we studied the 1H-NMR-based metabolomics of brain, heart, liver and white muscle extracts of common carp, subjected to anoxia (0 mg O2 l?1) and hypoxia (0.9 mg O2 l?1) at 5 °C. Specifically, fish were exposed to normoxia (i.e. 9 mg O2 l?1; controls 24 h, 1 week and 2 weeks), acute hypoxia (24 h), chronic hypoxia (1 week) and chronic hypoxia (1 week) with normoxic reoxygenation (1 week). Additionally, we also investigated the metabolic responses of fish to anoxia for 2 h. Both anoxia and hypoxia significantly changed the tissue levels of standard energy metabolites as lactate, glycogen, ATP/ADP and phosphocreatine. Remarkably, anoxia induced increased lactate levels in all tissues except for the heart whereas hypoxia resulted in decreased lactate concentrations in all tissues except for brains. Furthermore, hypoxia and anoxia influenced amino acids (alanine, valine/(iso)leucine) and neurotransmitters levels (GABA, glutamate). Lastly, we also detected ‘other’ i.e. previously not reported compounds to play a role in the present context. Scyllo-inositol levels changed significantly in heart, liver and muscle, providing novel insights into the anoxia/hypoxic responses of the common carp.  相似文献

The kinetics of malonate replacement in bis- (malonato)oxovanadate(IV), [VO(mal)2H2O]2−(hereafter water molecule will be omitted), by oxalate has been studied by the stopped-flow method. The reaction was found to consist of two consecutive steps (k1 and k2: first-order rate constants) passing through a mixed ligand complex, [VO(mal)(ox)]2−. The rates for each step depended linearly on the concentrations of free oxalate species, Hox and ox2−. The second-order rate constants for the replacement by ox2− were much larger in the k1 step than in the k2 step and the activation parameters were determined as follows: ΔH= 43.5 ± 5.6 kJ mol−1, ΔS±-53 ± 19 J K−1 mol−1 and ΔH≠= 43.6 ± 0.5 kJ mol−1, δS≠ = -62 ± 2 J K−l mol−1 for the k1 and k2 steps, respectively. The volume of activation was determined to be -0.65 ± 0.75 cm3 mol−1 at 20.2 °C by the high-pressure stopped-flow method for the apparent rate constants.  相似文献

Pulse radiolytic studies of α-tocopherol (αTH) oxidation-reduction processes were carried out with low doses (5 Gy) of high-energy electrons in O2−, N2−, and air-saturated ethanolic solutions. Depending on the concentration of oxygen in solution, two different radicals, A· and B·, were observed. The first, A·, was obtained under N2 and results from aTH reaction with solvated electron (kaTH+csolv = 3.4 × 108 mol−1 liter s−1) and with H3C-ĊH-OH, (R·) (kaTH + R· = 5 × 105 mol−1 liter s−1). B·, observed under O2, is produced by αTH reaction with RO2 peroxyl radicals (kaTH + RO2. = 9.5 × 104 mol−1 liter s−1).  相似文献

The extraction equilibrium of the hydronium-uranium(VI)-dicyclohexano-24-crown-8 complex was carried out in the crown ether1,2-dichloroethaneHCl aqueous solution system at different temperatures. The extraction complex has the overall composition (L)2·(H3O+·χH2O)2·UO2Cl42− (L = dicyclohexano-24-crown-8). The values of the extraction equilibrium constants (Kex) increase steadily with a decrease in temperature: 13.5 (298 K), 7.96 (301 K), 4.20 (303 K) and 2.07 (305 K). A plot of log Kex against 1/T shows a straight line. The value of the enthalpy change, ΔH°, was calculated from the slope and equals −212 kJ mol−1. The value of the entropy change, ΔS°, was calculated from ΔH° and Kex and equals −690 J K−1 mol−1, whereas ΔG° = −6.45 kJ mol−1. Comparing these thermodynamic parameters with those of the dicyclohexano-18-crown-6 isomer A [1] (ΔS° = −314 J K−1 mol−1, ΔH° = −101 kJ mol−1 and ΔG° = −8.37 kJ mol−1), it can be seen that ΔH° and ΔS° are more negative for the former than for the latter, and both are enthalpy-stabilized complexes. The molecular structure of the complex has the feature that there are two H5O2+ ions in it, in contrast to the H3O+ ions in the dicyclohexano-18-crown-6 isomer A complex [1]. Each of the H5O2+ ions is held in the crown ether cavity by four hydrogen bonds. The H5O2+ ion has a central bond. The uranium atom forms UO2Cl42− as a counterion away from the crown ether. The formation of this complex is in good agreement with more negative entropy change and less negative free energy change, as mentioned above.  相似文献

Growth and oxygen consumption was measured in developing herring Clupea harengus (L.) embryos. By considering the variations in oxygen consumption with embryonic size and growth rate, an attempt was made to partition oxygen consumption between growth related and growth unrelated (i.e., “maintenance”) processes. The metabolic cost of growth was estimated as ≈ 150 ng O2 · μg dry wt tissue formed−1. This estimate compares favourably with the biochemical estimate of the costs of transport and net biosynthesis. The “maintenance” component was proportional to embryonic mass (77 ng O2 · μg−1· d−1). Over the entire embryonic period, growth processes were responsible for ≈ 25% of the cumulated oxygen consumption.  相似文献

A field experiment was conducted at the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT) Center, Patancheru, India to study photosynthetically active radiation (PAR) interception and dry matter production relationships in pearl millet (Pennisetum americanum (L.) Leeke). Two pearl millet genotypes, BJ 104 (G1) and ICH 226 (G2) were sown at three planting geometries obtained by using combinations of row and plant spacings (S1: 37·5 cm × 26·6 cm; S2: 75·0 cm × 13·3 cm; S3: 150·0 cm × 6·6 cm) such that plant population was constant at 100 000 ha−1 in all treatments. Cumulative intercepted PAR was maximum (330 MJ m−2) in G2S2 and minimum (268 MJ m−2) in G1S3. Conversion efficiency values ranged from 1·87 g MJ−1 in G1S2 to 2·32 g MJ−1 in G2S3. Final above-ground dry matter followed the pattern of cumulative intercepted PAR and maximum dry matter (7·22 Mg ha−1) was produced by G2S2 while G1S3 produced minimum dry matter (4·97 Mg ha−1).  相似文献

An experiment was conducted in the growth chamber to quantify the biomass production, N removal and N2 fixation from a synthetic medium by Chlamydomonas reinhardtii and Anabaena flos-aquae. Nitrogen was supplied at a concentration of 100 mg liter−1 of NO315N and NH4+15 (3·5 atom %), respectively. After 21 days Chlamydomonas reinhardtii removed an average of 83·8 and 78·7 mg N liter−1 as NO3 and NH4+, respectively. Averages of 0·89 and 0·71 g liter−1 (first batch), 1·63 and 0·95 g liter (second batch) algal biomass were collected from NO3 and NH4+ media, respectively. Uptake rates of 0·11 mg 15N g−1 algae day−1 from NO3 medium and 0·10 mg 15N g−1 algae day−1 from NH4+ medium were calculated. Algal cells grown in NO3 and NH4+ medium contained 71 and 65 g N kg−1 (first batch), 39 and 58 g N kg−1 (second batch), respectively. Anabaena flos-aquae produced averages of 0·58 and 0·46 g liter−1 (first batch), 0·55 and 0·48 g liter−1 (second batch) after 14 days of growth from NO3 and NH4+ media, respectively. Blue-green algal biomass contained higher N (81–98 g kg−1) than green algae. Isotope dilution method for determining N2 fixation indicated that 55% and 77% of total N of blue-green algae grown in NO3 and NH4+ media, respectively, was derived from the atmosphere.  相似文献

Using the CO2 flux data measured by the eddy covariance method in the northeast of Qinghai-Tibetan Plateau in 2005, we analyzed the carbon flux dynamics in relation to meteorological and biotic factors. The results showed that the alpine wetland ecosystem was the carbon source, and it emitted 316.02 gCO2 · m−2 to atmosphere in 2005 with 230.16 gCO2 · m−2 absorbed in the growing season from May to September and 546.18 gCO2 · m−2 released in the non-growing season from January to April and from October to December. The maximum of the averaged daily CO2 uptake rates and release rates was (0.45 ± 0.0012) mgCO2 · m−2 · s−1 (Mean ± SE) in July and (0.22 ± 0.0090) mgCO2 · m−2 · s−1 in August, respectively. The averaged diurnal variation showed a single-peaked pattern in the growing season, but exhibited very small fluctuation in the non-growing season. Net ecosystem exchange (NEE) and gross primary production (GPP) were all correlated with some meteorological factors, and they showed a negatively linear correlation with aboveground biomass, while a positive correlation existed between the ecosystem respiration (Res) and those factors.  相似文献

To determine the most favorable conditions for the production of ethanol by Pachysolen tannophilus, this yeast was grown in batch cultures with various initial concentrations of two of the constituents of the culture medium: d-xylose (so), ranging from 1 g·l−1 to 200 g·l−1, and yeast extract (lo), ranging from 0 g·l−1 to 8 g·l−1. The most favorable conditions proved to be initial concentrations of So=25 g·l−1 and lo=4 g·l−1, which gave a maximum specific growth rate of 0.26 h−1, biomass productivity of 0.023 g·l−1·h−1, overall biomass yield of 0.094 g·g−1, specific xylose-uptake rate (qs) of 0.3 g·g−1·h−1 (for t=50 h), specific ethanol-production rate (qE) of 0.065 g·g−1·h−1 and overall ethanol yield of 0.34 g·g−1; qs values decreased after the exponential growth phase while qE remained practically constant.  相似文献

Embryos of Austrofundulus limnaeus are exceptional in their ability to tolerate prolonged bouts of complete anoxia. Hypoxia and anoxia are a normal part of their developmental environment. Here, we exposed embryos to a range of PO2 levels at two different temperatures (25 and 30 °C) to study the combined effects of reduced oxygen and increased temperature on developmental rate, heart rate, and metabolic enzyme capacity. Hypoxia decreased overall developmental rate and caused a stage-specific decline in heart rate. However, the rate of early development prior to the onset of organogenesis is insensitive to PO2. Increased incubation temperature caused an increase in the developmental rate at high PO2s, but hindered developmental progression under severe hypoxia. Embryonic DNA content in pre-hatching embryos was positively correlated with PO2. Citrate synthase, lactate dehydrogenase, and phosphoenolpyruvate carboxykinase capacity were all reduced in embryos developing under hypoxic conditions. Embryos of A. limnaeus are able to develop normally across a wide range of PO2s and contrary to most other vertebrates severe hypoxia is not a teratogen. Embryos of A. limnaeus do not respond to hypoxia through an increase in the capacity for enzymatic activity of the metabolic enzymes lactate dehydrogenase, citrate synthase, or phosphoenolpyruvate carboxykinase. Instead they appear to adjust whole-embryo metabolic capacity to match oxygen availability. However, decreased DNA content in hypoxia-reared embryos suggests that cellular enzymatic capacity may remain unchanged in response to hypoxia, and the reduced capacity may rather indicate reduced cell number in hypoxic embryos.  相似文献

Photosynthesis, transpiration, and leaf area distribution were sampled in mature Quercus virginiana and Juniperus ashei trees to determine the impact of leaf position on canopy-level gas exchange, and how gas exchange patterns may affect the successful invasion of Quercus communities by J. ashei. Sampling was conducted monthly over a 2-yr period in 12 canopy locations (three canopy layers and four cardinal directions). Photosynthetic and transpiration rates of both species were greatest in the upper canopy and decreased with canopy depth. Leaf photosynthetic and transpiration rates were significantly higher for Q. virginiana (4.1–6.7 μmol CO2·m−2·s−1 and 1.1–2.1 mmol H2O·m−2·s−1) than for J. ashei (2.1–2.8 μmol CO2·m−2·s−1 and 0.7–1.0 mmol H2O·m−2·s−1) in every canopy level and direction. Leaves on the south and east sides of both species had higher gas exchange rates than leaves on the north and west sides. Although Quercus had a greater mean canopy diameter than Juniperus (31.3 vs. 27.7 m2), J. ashei had significantly greater leaf area (142 vs. 58 m2/tree). A simple model combining leaf area and gas exchange rates for different leaf positions demonstrated a significantly greater total canopy carbon dioxide uptake for J. ashei compared to Q. virginiana (831 vs. 612 g CO2·tree−1·d−1, respectively). Total daily water loss was also greater for Juniperus (125 vs. 73 Ltree−1·d−1). Differences in leaf gas exchange rates were poor predictors of the relationship between the invasive J. ashei and the codominant Q. virginiana. Leaf area and leaf area distribution coupled with leaf gas exchange rates were necessary to demonstrate the higher overall competitive potential of J. ashei.  相似文献

The critical thermal maximum (CTMAX) is the temperature at which animals exhibit loss of motor response because of a temperature-induced collapse of vital physiological systems. A central mechanism hypothesised to underlie the CTMAX of water-breathing ectotherms is insufficient tissue oxygen supply for vital maintenance functions because of a temperature-induced collapse of the cardiorespiratory system. The CTMAX of species conforming to this hypothesis should decrease with declining water oxygen tension (PO2) because they have oxygen-dependent upper thermal limits. However, recent studies have identified a number of fishes and crustaceans with oxygen-independent upper thermal limits, their CTMAX unchanged in progressive aquatic hypoxia. The previous studies, which were performed separately on cold-water, temperate and tropical species, suggest the oxygen-dependence of upper thermal limits and the acute thermal sensitivity of the cardiorespiratory system increases with decreasing habitat temperature. Here we directly test this hypothesis by assessing the oxygen-dependence of CTMAX in the polar Antarctic krill (Euphausia superba), as well as the temperate Baltic prawn (Palaemon adspersus) and brown shrimp (Crangon crangon). We found that P. adspersus and C. crangon maintain CTMAX in progressive hypoxia down to 40 mmHg, and that only E. superba have oxygen-dependent upper thermal limits at normoxia. In E. superba, the observed decline in CTMAX with water PO2 is further supported by heart-rate measurements showing a plateauing, and subsequent decline and collapse of heart performance at CTMAX. Our results support the hypothesis that the oxygen-dependence of upper thermal limits in water-breathing ectotherms and the acute thermal sensitivity of their cardiorespiratory system increases with decreasing habitat temperature.  相似文献

The productivity and biomass parameters of the symbiotic anemone Aiptasia pulchella (Carlgren, 1943) from a shaded mangrove lagoon (maximum summer irradiance of 100 μE m−2 · s−1) and a sunlit reef flat (maximum summer irradiance of 1400 μE · m−2 · s−1) were examined in Hawaii. Light-shade adaptation was evident in the summer populations (1981) but not observed during the fall (1982). In the summer, zooxanthellae from the lagoon A. pulchella (shade anemones) contained 2.97 pg Chl a cell −1 and those from the reef flat (sun anemones) contained 1.70 pg Chl a · cell−1; but Chl a : c2 ratios were 2.5 in zooxanthellae from both shade and sun anemones. During the fall, there were no significant differences in Chl a and c2 of zooxanthellae (2.25 pg Chl a · cell−1) in shade and sun anemones, but Chl a : c2 ratios averaged 3.9. During both seasons, shade anemones were larger and contained higher densities of zooxanthellae than sun anemones. In addition to differences between shade and sun habitats, there was localized photoadaptation of zooxanthellae within individual anemones due to microhabitat variations in ambient irradiance. Growth rates of zooxanthellae in A. pulchella differed in shade and sun anemones. Specific growth rates for zooxanthellae in situ were the same for shade populations in both summer and fall (0.016 day−1). However, zooxanthellae in sun anemones grew four times faster in the fall (0.033 day−1) than during the summer (0.008 day−1). These results suggest that growth of zooxanthellae in these anemones was independent of ambient irradiance. Photosynthesis-irradiance (P-I) responses of shade and sun anemones during the summer showed that shade anemones had greater photosynthetic efficiencies (α) but lower photosynthetic capacities (Pmax) than sun anemones. Dark-respiration rates of sun anemones were twice those obtained with shade anemones. In the fall, these populations of anemones did not exhibit P-I responses characteristic of light-shade adaptation. Both α and Pmax of shade and sun anemones were higher in the fall, indicating that zooxanthellae in A. pulchella adapted to seasonal reduction in irradiance.  相似文献

The benthic oxygen consumption and carbon dioxide production of undisturbed and sieved sediment cores with various values for the biomass of polychaetes collected from the intertidal mud-flat of Nanakita River estuary of Japan were measured simultaneously. The benthic oxygen consumption and carbon dioxide production increased in proportion to the biomass of a dominant polychaete species Neanthes japonica (Izuka). This increase was not explained by the respiration of the animals alone. The residual increase in benthic O2 and CO2 fluxes may be due to mineralization processes in the burrow wall and enhanced diffusion caused by the pumping activity of the worms. From the average biomass of polychaetes at the study site, total benthic O2 and CO2 fluxes were estimated to be 5.2 mmol·m−2·h−1 and 7.3 mmol·m−2·h−1, respectively, at 20 ° C. The worms were responsible for 79% of the total O2 flux and 73% of the total CO2 flux but the respiration of the worms accounted for only 53% of the total O2 flux and 36% of the total CO2 flux. The residual enhanced fluxes were 26% and 37% for the total O2 and CO2 fluxes, respectively.  相似文献

Photosynthesis and respiration rates of blades from a selected, fast growing strain of the marine red alga. Gigartina exasperata Harvey and Bailey, a carrageenan producer, were measured with an oxygen electrode and compared with rates similarly obtained from wild material of the same species. The measurements, expressed as μl O2 · mg chl a?1, min?1. were made over a light intensity range from 5 to 800 μE · m?2 · sec?1 and a temperature range of 6 to 16°C. The photosynthesis light intensity data are best described by hyperbolic functions.  相似文献

Nuclear magnetic resonance line-widths data have been used to determine the rate of solvent exchange from the first coordination sphere of ferro-and ferriprotoporphyrin(IX) dimethylester (Fe-PPD) in pyridine/chloroform. The average values of kinetic parameters for pyridine (PY) exchange indicate an SN2 mechanism tor Fe(III)-PPD(ΔH&;#; = 36 kJ · mol−1 ; ΔS&;#; = −53 J·mol−1K−1; TM(298 K) = 0.07 msec) and an SNI mechanism for Fe(II)-PPD (ΔH&;#; = 67 kJ·mol−1; ΔS&;#; = 42 J · mol−1K−1; TM(298 K) = 0.06 msec). Parallel to the accelerated ligand exchange rate at rising temperatures a redistribution of the electrons causing a transition of the metal porphyrin from the low-spin state to the high-spin state is observed. Enthalpy and entropy of the thermodynamic equilibrium between low- and high-spin Fe-PPD have been determined from experimental values of the average magnetic moment. A mean lifetime of low-spin Fe(III)-PPD was estimated from line. widths changes (TL→H(298 K)≈ 20 msec) and the corresponding activation parameters have been obtained (ΔH&;#;L→H(298 K) = 26 kJ · mol−1; ΔS&;#;L→H(298K) = −125 J · mol−1K−1).  相似文献
