The basic fragmentation mechanisms occurring in the mass spectra of the phosphatidylcholines have been described previously, and evidence was adduced to show that many of the more abundant ions are related by electron impact-induced processes. A molecular ion was demonstrated for dioleoyl glycerylphosphorylcholine by accurate mass measurement and by metastable scanning. In the present paper, results are reported which further extend the previous work by including a more detailed investigation of "nonapparent" fragment ions for a series of phosphatidylcholines with different acyl side chains and also for a series of phosphatidyl amino alcohols of fixed acyl composition. The results demonstrate the effect of the choline quaternary nitrogen on the stability of the molecular ion, and estimates for the appropriate rate constants are given. Nitrogen-containing fragments have been demonstrated by recording spectra at low electron voltages. The work has also been extended to include natural phosphatidylcholine preparations of mixed acyl composition, and the possibility of detecting particular molecular species has been established. Quantitative estimates may be made using suitable synthetic phosphatidylcholines, and results are presented to show the variation of the molar correction factor with acyl chain length.  相似文献

The fragmentation processes occurring in various 9alpha-hydroxy steroids upon electron ionization have been studied. The mechanisms proposed for the formation of the prominent ions in these spectra have been confirmed with the aid of deuterium labelling, measurements on metastable ion decompositions, low eV spectra and high resolution mass measurements. The fragmentation of the corresponding 9-oxo-9,10-seco steroids, which shows analogy to that of the 9alpha-hydroxy compounds in many respects, has also been discussed.  相似文献

Fully acetylated methyl x-deoxy-x-fluoro-alpha-D-glucopyranosides have been studied using electron impact and ammonia chemical ionisation mass spectrometry. Mass analysed metastable ion kinetic energy spectroscopy (MIKE), collisional activation (CID), and accelerated voltage scanning have been used to evaluate complete fragmentation schemes. Characteristic differences in the fragmentation of positional isomers were noted on analysis of the spectra, and these make it possible to determine the location of fluorine in the molecules studied. Collisionally activated fragmentation of [M-OCH3]+ ions, produced by electron impact, provides an alternative method for localisation of the fluorine atoms. To the contrary, MIKE and CID spectra of [M + NH4]+ cluster ions produced by chemical ionisation did not afford such structural information.  相似文献

The mass spectra of a number of C-4alpha- and C-4beta-alkylated cholestan-3beta, 5alpha-diols have been found to contain an intense ion at m/e 332. the corresponding 6beta-alkylated (R) cholestan-3beta, 5alpha-diols exhibit an abundant ion at m/e 331 + R. The mass spectra of cholestan-5alpha-ol and cholestan-3beta, 5alpha-diol also show an ion at m/e 332 which, however, is of relatively low abundance. The ion in question appears to arise from fragmentation processes which are characteristics of 5alpha-hydroxysteroids. A similar fragmentation has been found to occur in cases of C-4 and C-6beta-alkylated 5alpha-hysroxy-cholestan-3-ones. The results of isotopic labeling, high resolution and metastable ion defocusing studies are discussed in terms of the origin of several of the ions in the spectra of the various compounds.  相似文献

Homologous series of synthetic long-chain monoesters and diesters of 1,2-ethanediol were analyzed by mass spectrometry, and the patterns of fragmentation were studied. Under electron impact saturated ethanediol monoesters yielded prominent ions characteristic of the short-chain diol, such as the rearranged ion formed by 2,3-cleavage (m/e 104) and the ion caused by 3,4-cleavage (m/e 117). Fragments characteristic of the constituent long-chain moieties were the acylium ions [RCO](+), [RCO - 1](+), and the ions [RC(OH)(2)](+). The mass spectra of ethanediol diesters exhibited very intense peaks due to the ions formed by loss of the acyloxy group, [M - RCOO](+), or one carboxylic acid, [M - RCOOH](+). These ions are distinctive for diol diesters. Precise mass measurements by high resolution mass spectrometry verified the composition of the ion fragments. Spectral studies of some monoesters and diesters of 1,3-propanediol, 1,4-butanediol, 2,3-butanediol, and also of some monounsaturated homologues, demonstrated that mass spectrometry is very suitable for the identification, distinction, and analysis of diol lipids.  相似文献

The fragmentation of methyl 2,3,5,6-tetra-O-methyl-d-glucofuranoside under electron impact is described using data for labelled analogues and metastable measurements. Although the fragmentation showed features of that of hexopyranosides and pentofuranosides, there were characteristic differences. Mass-spectral data that are useful for identification of pentoses and hexoses as pyranosides or furanosides are tabulated.  相似文献

A study of small (n = 3 to 6) oligonucleotide and the metastable and collisionally activated decompositions of their (M-H)- species desorbed by using fast atom bombardment (FAB) is reported. Data were obtained for both ribo- and 2'-deoxyribotrinucleotides and for 2'-deoxyribotetra-, penta-, and hexanucleotides. The favored metastable decompositions of all of the oligonucleotides studied are eliminations of neutral CONH and loss of BH, where B is the base moiety. The BH elimination, however, provides little sequence information in the higher oligonucleotides and the process is more indicative of the different bases present in the oligomer. The chemistry observed upon collisional activation changes as one goes from trinucleotides to hexanucleotides. The formation of sequence ions is more facile for processes involving the 3' terminus, allowing the sequence to be determined. As one goes to the higher oligonucleotides, however, several different competitive fragmentation processes become as facile as or more facile than the reactions giving the sequence ions. This hinders proper ion assignments and makes sequence determination difficult.  相似文献

Mass spectrometry has played an integral role in the identification of proteins and their post-translational modifications (PTM). However, analysis of some PTMs, such as phosphorylation, sulfonation, and glycosylation, is difficult with collision-activated dissociation (CAD) since the modification is labile and preferentially lost over peptide backbone fragmentation, resulting in little to no peptide sequence information. The presence of multiple basic residues also makes peptides exceptionally difficult to sequence by conventional CAD mass spectrometry. Here we review the utility of electron transfer dissociation (ETD) mass spectrometry for sequence analysis of post-translationally modified and/or highly basic peptides. Phosphorylated, sulfonated, glycosylated, nitrosylated, disulfide bonded, methylated, acetylated, and highly basic peptides have been analyzed by CAD and ETD mass spectrometry. CAD fragmentation typically produced spectra showing limited peptide backbone fragmentation. However, when these peptides were fragmented using ETD, peptide backbone fragmentation produced a complete or almost complete series of ions and thus extensive peptide sequence information. In addition, labile PTMs remained intact. These examples illustrate the utility of ETD as an advantageous tool in proteomic research by readily identifying peptides resistant to analysis by CAD. A further benefit is the ability to analyze larger, non-tryptic peptides, allowing for the detection of multiple PTMs within the context of one another.  相似文献

Electron impact mass spectra of a series of aldobiouronic and pseudoaldobiouronic acid per-O-methyl derivatives and of the corresponding 4,5-unsaturated analogues, found normally among the products of methylation of uronic acid containing disaccharides as a result of methylation accompanying beta-elimination, have been studied. Using labelling experiments, metastable transition measurements and high resolution mass spectrometry, the fragmentation mechanisms of substances of this class have been deduced. Application of the information to the structure elucidation of this type of compound is discussed. It is concluded that from the mass spectra alone it is possible to determine the molecular weight, the cycle masses as well as the mode of linkage between the monomeric units. The appearance potentials of ions formed by cleavage of the glycosidic linkages have also been determined and the energetic differences encountered in the fission of the glycosidic linkages of various types of uronic acid containing oligosaccharides are discussed.  相似文献

The basic fragmentation mechanism of methyl(methyl 4-deoxy-2,3-di-O-methyl-β-l-threo-hex-4-enopyranosid)uronate has been deduced by deuteromethylation analysis, metastable transition measurements, and by interpreting the spectra of weakly excited foregoing molecules. The differences in fragmentation of partially methylated derivatives of methyl 4-deoxy-β-l-threo-hex-4-enopyranosiduronic acid compared to that of the fully methylated substance are discussed in detail and the criteria are proposed for identification of the compounds concerned by mass spectrometry.  相似文献

A new norsesquiterpene lactone, 1-hydroxyplatyphyllide, was isolated from roots of Senecio gilliesiano together with the previously known platyphyllide and the common pyrrolizidine alkaloids, senecionine and retrorsine. The structures of the products were determinated by spectral and chemical means and by comparison with standards and previously reported data. The genetic pathway of fragmentation by electron impact induced fragmentation of platyphyllide and 1-hydroxyplatyphyllide have been studied with the aid of high resolution measurements and metastable descompositions by the use of linked scanning.  相似文献

The mass spectra of 20 differently substituted dimethylthiophosphinic esters of aromatic hydroxy compounds are presented. Fragmentation routes were investigated using high resolution mass measurements, decoupled metastable determinations and deuterium labelling. All compounds exhibited abundant molecular ions and typical phosphorus-containing ions. Characteristic elimination processes strongly dependent upon the respective type of substitution were observed. Due to their high stability, their great ease of formation and their good gas chromatographic properties these new types of derivatives are of special interest for establishing gas chromatography mass spectrometry profiles of acidic catecholamine metabolites.  相似文献

The electron impact ionization of C-4-alkylated cholest-5-en-3β-hydroxysterols has been investigated. The mass spectra of the C-4-alkylated cholesterols contain a number of ions in the high mass region for which analogous ions are not found in the spectrum of cholesterol. Detailed studies of the composition and origin of these ions have been made by high resolution mass spectrometry and analysis of metastable ions. In addition, a large number of isotopically (deuterium and 18O) substituted C-4-alkylated analogues have been prepared to assist in the interpretation of the spectra. The combined results indicate the occurrence of a number of very complex and unusual electron ionization induced fragmentations. Most notable of the findings reported herein concerns the demonstration of the formation of an ion involving loss of the elements of ring A with an intramolecular shift of the oxygen and hydrogen atoms of the hydroxyl function to the charge-retaining species.  相似文献

Ethonium, an antimicrobial chemotherapeutic agent, was investigated by mass spectrometry (MS) under various ionization conditions: electron impact, field ionization, field desorption (FD) and fast atom ionization. FDMS was found to be the most suitable procedure for ethonium identification. Relation of the ED mass spectra to the distance between the nitrogen atoms in bis-quaternary ammonium compounds is discussed. It was shown that the most intensive ions with m/z 499, 315 in the FD mass spectra corresponded to the ethonium specific fragmentation and their occurrence in the spectra could serve as a sufficient criterion useful in qualitative and quantitative assay of the drug in the sample.  相似文献

iTRAQ reagents allow the simultaneous multiplex identification and quantification of a large number of proteins. Success depends on effective peptide fragmentation in order to generate both peptide sequence ions (higher mass region, 150–2200 m/z) and reporter ions (low mass region, 113–121 m/z) for protein identification and relative quantification, respectively. After collision‐induced dissociation, the key requirements to achieve a good balance between the high and low m/z ions are effective ion transmission and detection across the MS/MS mass range, since the ion transmission of the higher m/z range competes with that of the low m/z range. This study describes an analytical strategy for the implementation of iTRAQ on maXis UHR‐Qq‐ToF instruments, and discusses the impact of adjusting the MS/MS ion transmission parameters on the quality of the overall data sets. A technical discussion highlights a number of maXis‐specific parameters, their impact of quantification and identification, and their cross‐interactions.  相似文献

A Bacillus strain,denoted as PY-1,was isolated from the vascular bundle of cotton.Biochemical,physiological and 16S rDNA sequence analysis proved that it should belong to Bacillus subtilis.The PY-1strain showed strong ability against many common plant fungal pathogens in vitro.The antibiotics producedby this strain were stable in neutral and basic conditions,and not sensitive to high temperature.From theculture broth of PY-1 strain,five antifungal compounds were isolated by acidic precipitation,methanolextraction,gel filtration and reverse-phase HPLC.Advanced identification was performed by mass spec-trometry and nuclear magnetic resonance spectroscopy.These five antifungal compounds were proved to bethe isomers of iturin A:A2,A3,A4,A6 and A7.In fast atom bombardment mass spectrometry/mass spec-trometry collision-induced dissociation spectra,fragmentation ions from two prior linear acylium ions wereobserved,and the prior ion,Tyr-Asn-Gln-Pro-Asn-Ser-βAA-Asn-CO~ ,was first reported.  相似文献

The data obtained with 252Cf plasma desorption (PD) and fast atom bombardment mass spectrometry of eight tri-, tetra- and pentapeptides were compared. Good spectra were obtained with 1-10 nmol of peptide. In both techniques molecular weight information was obtained. The PD mass spectra are often dominated by the cationized molecular ions in contrast to the fast atom bombardment (FAB) mass spectra, where cationization is rarely observed. Amino acid content is reflected in the immonium ions equally well in both techniques. The fragmentation patterns observed with the two techniques are almost identical. However, practical sequencing of peptides based on either FAB or PD mass spectrometry of underivatized peptides alone is difficult. This is due to the unpredictable and sometimes absent cleavage yield at certain peptide bonds. Another difficulty is the many simultaneous fragmentation pathways. However, for many peptides enough information is present to allow sequence determination for at least a major part of the molecule.  相似文献

The negative ion mass spectrometric characteristics of a series of di- and trisaccharides and the tetrasaccharide stachyose have been studied using fast atom bombardment mass spectrometry. The molecular weight of the compounds can easily be derived from their mass spectra, which all show an abundant [M - H]- ion peak. The application of metastable ion and collisional activation techniques to selected pseudomolecular and fragment ions appears to be appropriate for the determination of the position and anomeric type of linkage in the molecules, and provides information concerning the monosaccharide units involved. Important fragmentation reactions have been traced and reaction mechanisms, supported by deuterium labelling experiments, are proposed. An experiment describing the application of the findings of this study to a glycosphingolipid molecule demonstrates its potential value for biological systems.  相似文献

Results of measurements of the absolute density of ArII ions in a mirror trap with a toroidal divertor are presented. The ion density was measured by the method of laser-induced fluorescence with the use of ArII metastable states. A technique is proposed to estimate the plasma electron temperature from the ratio of the fluorescence signals obtained by comparing the intensities of different optical transitions of ArII.  相似文献
