Fleurat-Lessard, P., Roblin, G., Bonmort, J. and Besse, C. 1988.Effects of colchicine, vinblastine, cytochalasin B and phalloidinon the seismonastic movement of Mimosa pudica leaf and on motorcell ultrastructure.—J. exp. Bot. 39: 209–221. Colchicine at 1 x 10–3 mol dm–3 does not affectthe seismonastic movement of Mimosa pudica leaves but disruptsmicrotubules in motor cells. Vinblastine at 5 x 10–3 moldm–3 does not affect this movement and partly disruptsmicrotubules. Vinblastine at 1 x 10–4 mol dm–3 alwaysdisrupts microtubules, even after a 12 h reversibility whenthe movement is restored. These drugs, applied at the same respectiveconcentrations, do not alter cytoplasmic and vacuolar fibrils.Cytochalasin B and phalloidin alter the seismonastic movementof Mimosa leaves when applied at concentrations of 1.25 x 10–3and 2.4 x 10–4 mol dm–3 respectively. These drugs,used at the same respective concentrations, also affect themotor cell structure and, in particular, modify the arrangementand the structure of the fibrils but they do not destroy themicrotubules. These data suggest that microtubules are not directly involvedin the seismonastic reaction whereas fibrils, formed by thin(3.0 nm wide) filaments, may be implicated in this reaction. The contents of K, Cl and Ca were determined in various partsof Mimosa pudica plant, namely in young internode (YI), agedinternode (AI), upper half (UP1), and lower half (LP1) of theprimary pulvinus, secondary pulvinus (P2), petiole (Pt) andleaflet blade (B1). Potassium is found in the following distributionYI = LP1 > P2 > UP1 > Pt > AI > B1 and is thuslocalized in parts showing a great metabolic activity. Chloridefollows the distribution of K except for the petiole and toa lesser extent for the young internode. Calcium content islow in the parts showing a high plasticity (pulvini and younginternode). A relationship is thus suggested between the amountof Ca and the rate of the movements which can be performed.The capacity for spontaneous and fusicoccin-induced H+ excretionis greater in the plant parts exhibiting the higher K content.This suggests that larger H+ fluxes may be required in tissuesshowing a high metabolic activity. By using a pH micro-electrode, the changes in the pH of theextracellular fluid in the motor tissue of the main pulvinusof Mimosa pudica were examined. During the rapid movement inducedby dropping ice-cold water on the petiole, the extracellularpH in the lower half shifted from 5.1 to 5.7, and then decreasedgradually to the initial pH within the next 30 min. Petiole heat-girdle treatments (followed by a 5 min 14CO2 assimilation)were performed on mature leaves of Vicia faba, in order to assesstheir effect on the partitioning of photo-assimilates to theminor vein phloem. Whole leaf autoradiographic evidence indicateda high leaf-to-leaf variation in the image intensity over theminor veins (relative to the mesophyll/epidermal background)in both control and heat-girdled groups of leaves. The averagedegree of minor vein labelling in heat-girdled leaves, however,was found to be significantly lower than that in controls. Comparativeassessment of vein labelling was based on microscopic densityreadings of silver grains over veinal and interveinal regionsin autoradiographic images. Investigations into the cause ofthis alteration in vein labelling indicated no involvement ofan inhibition of apoplasmic phloem loading, as both heat-girdledand control leaves of Vicia were shown to have comparable minorvein uptake of exogenously supplied 14C-sucrose. Heat-girdlingwas shown, however, to increase significantly the partitioningof recently fixed carbon into the insoluble (mainly starch)fraction relative to the ethanol-soluble fraction, within 12min of the treatment. We suggest that this carbon partitioningchange can primarily account for the change in vein labelling,since an increase in the insoluble fraction would result in(1) more 14C-activity remaining in the leaf mesophyll and (2)less 14C-activity going into the mesophyll export pool, andthus, less 14C-sucrose being transferred to the minor vein region.Additionally, although leaf export was completely halted inheat-girdled leaves, 14C-activity was found within the majorveins as far as the point of petiole heat-girdling (followinga 5 min assimilation and 4 h chase). Apparently, continued (butlimited) solution flow within the sieve elements is maintainedby transport pathway unloading within the treated leaves. Leaflet pairs from detached pinnae of Mimosa pudica opened afterthe pinnae had been irradiated with light (2 W·m–2)of 726 or 403 nm, whereas they remained almost closed with lightof 585 or 656 nm. Light-induced leaflet opening was observedonly in the daytime, from 6:00 to 16:00. Application of IAAat more than 30 µg/ml to the cut end of the pinna rachisesmade the leaflets open even in darkness with almost constantlag times of about 100 min which were independent of the concentration.NAA and 2,4-D also made the leaflet open at lower concentrationsthan IAA. Auxin-induced leaflet opening showed diurnal variation.Application of IAA for 2 to 6 min, depending on the concentration,was enough to open the leaflets. Biologically closed electrical circuits operate over large distances in biological tissues. The activation of such circuits can lead to various physiological and biophysical responses. Here, we analyse the biologically closed electrical circuits of the sensitive plant Mimosa pudica Linn. using electrostimulation of a petiole or pulvinus by the charged capacitor method, and evaluate the equivalent electrical scheme of electrical signal transduction inside the plant. The discharge of a 100 µF capacitor in the pulvinus resulted in the downward fall of the petiole in a few seconds, if the capacitor was charged beforehand by a 1.5 V power supply. Upon disconnection of the capacitor from Ag/AgCl electrodes, the petiole slowly relaxed to the initial position. The electrical properties of the M. pudica were investigated, and an equivalent electrical circuit was proposed that explains the experimental data.  相似文献

A cytochemical and biochemical study of acid phosphatases inroots of Vicia faba using p-nitrophenylphosphate(p-NPP) as substratehas shown the lack of specificity of the substrate which canbe acted upon by a K+-activated acyl phosphatase, nucleotidepyrophosphatase, glucox-6-phosphatase, 5' (3')-ribonucleotidephosphohydrolase, nucleotide phosph-transferase and those acidphosphatases demonstrable with ß-glycerophosphate.It is suggested that care should be taken in the interpretationof both biochemical and cytochemical studies employing sucha non-specific substrate as P-NPP. Mechanism of Photosynthate Efflux from Vicia faba L. Seed Coats   总被引:2,自引:0,他引:2  
In order to develop a tentative model of the mechanism of photosynthateefflux from the vascular region of Vicia faba L. seed coats,wash-out experiments were performed after removal of the embryo. The sulphydryl group modifiers, pCMBS and NEM, reduced 14C-photosynthateefflux by 40% and 50%, respectively. Their inhibitory effectcould be prevented or reduced (in the latter case) by includingDTT in the bathing solution. Maltose competed with sucrose forefflux; a concentration of 300 mol m–3 inhibited 14C-photosynthaterelease by 35%. The cations K+ , Na+ Mg2+ and TPP+ enhancedefflux significantly, whereas the countenon Cl had noeffect. The presence of the protonophore CCCP (0·1 molm–3) led to a reduction of efflux by 50% net proton extrusiondropped by 34%. To a lesser extent, an efflux inhibition wasalso achieved by decreasing the cytoplasmic pH with the weakacid DM0. In contrast, alterations in the external pH causedonly a feeble response. The ATPase inhibitor, EB, decreasedphotosynthate efflux and H+ extrusion. DES reduced efflux slightly,presumably by affecting ATPase activity as well as energy metabolism. Based on these findings, it is proposed that a sucrose/protonantiport mechanism could be responsible for photosynthate effluxfrom Vicia faba seed coats. Growth of the Vicia faba seedling is accompanied by a rapid15-day increase in amine oxidase activity of the apical parts.Cotyledons and roots were found to be devoid of activity. Thepartially purified enzyme from leaves readily oxidized putrescine,cadaverine, agmatine and spermidine, while dopamine (3-hydroxytyramine)and L- and D-lysine were oxidized more slowly. The Km valueswere 1.9?10–3 M for cadaverine, 3.7?10–5 M for putrescine,7.8?10–4 M for spermidine, and 5.9?10–3 M for dopamine.Carbonyl reagents and copper-binding agents were effective inhibitorsof Vicia faba amine oxidase. Propagation in Vicia faba Stem of a Potential Variation Induced by Wounding   总被引:3,自引:0,他引:3  
Heating of leaf blade in Vicia faba triggered bioelectricalevents ("variation potential") spreading throughout the shoot,preferentially in the acropetal direction. The spreading ofa variation potential, previously described in Mimosa and otherplants, is related to a concomitant transport of a stimulatingsubstance in the transpiration stream. This conclusion is supportedby the observation in Vicia faba stem that conduction in thebasipetal direction is promoted when the transpiration streamis inverted. The conduction is promoted in the vascular bundles in directrelation with the burnt lobe of the leaf, but a lateral conductionmay exist, as electrical variations are recorded in isolatedbundles of the orthostichy opposite to the wounded leaf. A local cooling (1C) of the stem has no influence on the conductionrate but it completely inhibits the bioelectrical variationsin the cooled zone. This last result reinforces earlier demonstrationsthat the substance is transported in the vessels, accordingto a physical process, but that the electrical events requiremetabolic integrality in other cellular members. The tetraammonium salt of the K+ binding fluorescent dye benzofuranisophthalate (PBFI) was used to investigate the influence ofpotassium nutrition (0.1–2.1 mol m–3) on apoplasticK+ inVicia faba leaves by means of ratio imaging. As a referencethe infiltration-centrifugation method was used. Both methodsreflected the influence of K+ supply on apoplastic K+ concentration.The abaxial leaf side revealed significantly higher K+ concentrations(20-25 mol m–3) than the adaxial side (5–8 mol m–3).Application of CCCP led to an immediate increase in apoplasticK+ demonstrating the reliability of the PBFI method. Total uptake and incorporation of 3H-thymidine, 3H-thymine,and 3H-deoxyuridine into DNA have been investigated in the apical3 cm of the primary root of Vicia faba. Evidence has been obtainedthat endogenous TdR in these roots may be transported eitherapically or basally; apical movement being greater than movementfrom the apex towards the base of the root. The results havebeen discussed with respect to the possible distribution ofendogenous pools of thymidine, thymine, and deoxyuridine inthe primary root of V. faba.  相似文献

Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Explants of stem, leaves, roots, and cotyledons from etiolatedaxenically grown Vicia faba seedlings were cultured on a rangeof media. Shoot organogenesis was only obtained with nodal stemand cotyledonary node explants when cultured on MS medium with3% sucrose, 2.0 mg 1–1 BAP and 02 mg 1–1 NAA. Callusproliferation accompanied shoot organogenesis from nodal stemexplants. Successive subculture of nodal stem callus resultedin proliferation of regenerative callus which contained severalshoot bud initials. The capacity for shoot regeneration fromthis callus was maintained for 9 months. Histological studiesreveal de novo formation of meristematic centres in callus andtheir further development into bud primordia. High frequencyrooting of these adventitious shoots was obtained on half-strengthMS medium with 1.5% sucrose, 0.1 mg 1–1 NAA and 0.5 mg1–1 kinetin. Broadbean plants (Vicia faba L.) were submitted to three differentlevels of steady state N limitation. Relative addition ratesof N were 0.06, 0.1 and 0.14d-1. Plants were harvested at fiveevenly distributed times over a 2d period. Shoot growth correspondedwell with the imposed treatment. Root growth, relative to shootgrowth, was highest at the 0.06d-1treatment. Diurnal patternsof soluble sugars, amino acids and starch were analysed. Onaverage, soluble sugar levels were highest in the plants ofthe 0.06d-1treatment whereas average free amino acid levelswere highest in the 0.14d-1treatment. Shoot growth increasedas the concentration of shoot amino acids increased. No suchcorrelation however could be found between root growth and freesugar levels in the root. Apoplastic potassium activities (ak) in leaves of Commelinacommunis L., Vicia faba L. and Pisum sativum L. var. argenteumwere recorded with neutral-carrier-based, potassium-sensitivemicro-electrodes. Measurements were carried out in 0.3–1.4nl volumes contiguous with the extracellular space of attachedleaves and were held for periods of 7–68 min. Mean steady-stateaK values recorded from all three species were below 50 µM.Similar potassium activities were attained, regardless of theinitial values obtained after washing with distilled water orpotassium additions, and the activities recorded showed onlyminimal dependence on the relative vapour pressure difference.Tissue capacity for K+ absorption was increased 15–30-foldin the presence of added Ca2+. By contrast, cyanide reducedboth the initial rate of potassium absorption by the tissuesand their apparent capacity for the cation. These observationsindicate that the free potassium pool in the leaf apoplast issignificantly smaller than has previously been assumed. Theresults contradict the notion that high concentrations of potassiumaccumulate locally as a result of transpiration, and may indicatethe presence in the leaf tissues of potassium transport activitysensitive to Ca2+ and dependent on metabolism. Experiments are reported on the spatial distributions of isotopiccarbon within the mesophyll of detached leaves of the C3 plantVicia faba L. fed 14CO2 at different light intensities. Eachleaf was isolated in a cuvette and ten artificial stomata providedspatial continuity between the ambient atmosphere (0.03–0.05%v/v CO2) and the mesophyll from the abaxial leaf side. Paradermalleaf layers exhibited spatial profiles of radioactivity whichvaried with the intensity of incident light in 2 min exposures.At low light, when biochemical kinetics should limit CO2 uptake,sections through palisade cells contained most radioactivity.As the light intensity was increased to approximately 20% offull sunlight, peak radioactivity was observed in the spongycells near the geometric mid-plane of the mesophyll. The resultsindicate that diffusion of carbon dioxide within the mesophyllregulated the relative photosynthetic activity of the palisadeand spongy cells at incident photosynthetically active lightintensities as little as 110 µE m–2 s–1 whenCO2 entered only through the lower leaf surface. In developing seed ofVicia faba L., solutes imported throughthe phloem of the coats move symplastically from the sieve elementsto a specialized set of cells (the thin-walled parenchyma transfercells) for release to the seed apoplast. Potassium (K+) is thepredominant cation released from the seed coats. To elucidatethe mechanisms of K+ efflux from seed coat to seed apoplast,whole-cell currents across the plasma membranes of protoplastsof thin-walled parenchyma transfer cells were measured usingthe whole-cell patch-clamp technique. Membrane depolarizationelicited a time-dependent and an instantaneous outward current.The reversal potential (ER of the time-dependent outward currentwas close to the potassium equilibrium potential (EK and itshifted in the same direction as EK upon changing the externalK+ concentration, indicating that this current was largely carriedby an efflux of K+. The activation of the time-dependent outwardK+ current could be well fitted by two exponential componentsplus a constant. The instantaneous outward current could alsobe carried by K+ efflux as suggested by ion substitution experiments.These K+ outward rectifier currents elicited by membrane depolarizationare probably too small to represent the mechanism for the normalK+ efflux from seed coat cells. Membrane hyperpolarization morenegative than –80 mV activated a time-dependent inwardcurrent. K+ influx was responsible for the inward current asthe current reversed at membrane voltage close to EK and shiftedin the same direction as EK when external [K+] was varied. Activationof this K+inward rectifier current was well fitted with twoexponential components plus a constant. A regulating functionfor this current is suggested. Vicia faba cv. Maris Bead was grown either on fixed nitrogenor on ammonium nitrate. After 4 weeks growth, nutrient solutionswere supplemented with 50, 75 and 100 mol m–3 NaCl for15 d. Five harvests were made at weekly intervals, beginningat 4 weeks. Effects of salinity were directly related to dose,plant growth (fresh and dry weight) being depressed in bothN-fixing and N-fertilized plants. The number of nodules perplant and the proportion of those formed which developed intothe active nitrogen fixing state were depressed by salt stress.Increased size of nodules in salt-stressed plants only partlycompensated for the lower specific nitrogenase activity. Theeffects of salinity on plant nitrogen content were more pronouncedon N-flxing than on N-fertilized plants. Vicia faba DNA was digested with restriction endonucleases andfractionated on 1% agarose gels. The physical map for EcoRI,BamHI and XbaI cleavage sites in V. faba cytosol rDNA was determinedusing the Southern blot hybridization technique. XbaI and BglIIdigestion or partial EcoRI digestion showed that the lengthof a major repeat unit of V. faba rDNA is 6.7 x 106 daltons.EcoRI and BamHI cleaved this unit into two and five DNA fragments,respectively.
