Thermodynamics of the B to Z transition in poly(dGdC)   总被引:1,自引:0,他引:1  
The thermodynamics of the B to Z transition in poly(dGdC) was examined by differential scanning calorimetry, temperature-dependent absorbance spectroscopy, and CD spectroscopy. In a buffer containing 1 mM Na cacodylate, 1 mM MgCl2, pH 6.3, the B to Z transition is centered at 76.4°C, and is characterized by ΔHcal = 2.02 kcal (mol base pair)?1 and a cooperative unit of 150 base pairs (bp). The tm of this transition is independent of both polynucleotide and Mg2+ concentrations. A second transition, with ΔHcal = 2.90 cal (mol bp)?1, follows the B to Z conversion, the tm of which is dependent upon both the polynucleotide and the Mg2+ concentrations. Turbidity changes are concomitant with the second transition, indicative of DNA aggregation. CD spectra recorded at a temperature above the second transition are similar to those reported for ψ(–)-DNA. Both the B to Z transition and the aggregation reaction are fully and rapidly reversible in calorimetric experiments. The helix to coil transition under these solution conditions is centered at 126°C, and is characterized by ΔHcal = 12.4 kcal (mol bp)?1 and a cooperative unit of 290 bp. In 5 mM MgCl2, a single transition is seen centered at 75.5°C, characterized by ΔHcal = 2.82 kcal (mol bp)?1 and a cooperative unit of 430 bp. This transition is not readily reversible in calorimetric experiments. Changes in turbidity are coincident with the transition, and CD spectra at a temperature just above the transition are characteristic of ψ(–)-DNA. A transition at 124.9°C is seen under these solution conditions, with ΔHcal = 10.0 kcal (mol bp)?1 and which requires a complex three-step reaction mechanism to approximate the experimental excess heat capacity curve. Our results provide a direct measure of the thermodynamics of the B to Z transition, and indicate that Z-DNA is an intermediate in the formation of the ψ-(–) aggregate under these solution conditions.  相似文献

Two juvenile, female African elephants (Loxodonta africana) were used in summer and winter trials to determine the apparent digestibility of timothy (Phleum pratense) hay. After 12–14 days of dietary adjustment, dry matter intake and fecal excretion were quantitatively measured for 7 days. Dry matter of timothy hay contained 8.6 and 7.7% crude protein, 57.3 and 44.0% acid detergent fiber, and 6.5 and 6.4% ash during the summer and winter trials, respectively. Estimates of apparent digestibility during summer and winter, respectively, were 39 and 35% for dry matter, 43 and 32% for gross energy (GE), 45 and 30% for crude protein (CP), and 36 and 24% for acid detergent fiber (ADF). While GE and CP digestibility estimates tended (P < .09) to be greater in the summer trial, only the digestibility of ADF was different (P < .05) between summer and winter. Dry matter intake was 1.4–1.6% of body weight (BW), providing an average of 144 kcal of digestible energy per kg BW0.75. This value is similar to that (155 kcal per kg BW0.75) used for estimating digestible energy requirements for maintenance of light-breed horses.  相似文献

Synopsis The capture of recently inseminated or pregnant specimens ofCarcharias taurus, Isurus paucus, I. oxyrinchus, Alopias superciliosus andA. vulpinus has allowed new information to be obtained on the reproductive biology of these species. Oophagy and embryonic cannibalism (adelphophagy) have been documented inC. taurus, but only oophagy in other lamnoid species. The occurrence of up to nine embryos of similar size per uterus inIsurus and no indication of functional erect teeth in embryos leaves considerable doubt that embryophagy occurs in this genus. Considerable data has been collected onCarcharias taurus which allows a lamnoid reproductive model to be developed and tested, in spite of the obvious differences between the reproductive biology of this species and other lamnoids. Gonad structure, ovarian development, fertilization, early embryonic differentiation, embryonic nutrition and parturition, inC. taurus and other lamnoids differs significantly from other elasmobranchs.  相似文献

Livestock grazing plays a significant role in maintaining grasslands and promoting animal production globally. To understand the livestock performance in sown pasture (SP) vs native pasture (NP) is important to ensure more effective grassland-livestock interactions with minimal environmental impact. A 2 (treatment) * 2 (period) Latin Square design experiment was conducted with 10 growing Hu sheep ♂ × thin-tailed Han sheep ♀ rams grazed perennially SP vs NP in an inland arid region of China. The objectives were to evaluate the effects of grazing management on nutrient digestibility, nitrogen (N) and energy utilisation and methane (CH4) emission. The N intake, N retained and energy intake (gross energy (GE), and digestible and metabolisable energy) of sheep grazing in SP were significantly increased compared with those grazing in NP. There were significant linear relationships between DM intake (DMI) (g/kg BW or g/kg BW0.75) or CH4 (g/kg BW or g/kg BW0.75) emissions and forage nutrient and GE concentrations within each grassland type. The linear regression analysis indicated that forage CP or ether extract concentration was a good predictor for DMI (g/kg BW or g/kg BW0.75) (R2 = 0.756 or 0.752), and CH4 emission could be predicted using forage nutrient and GE concentrations (R2 = 0.381–0.503). These results suggest that DMI and CH4 emissions per unit metabolic BW were accurately predicted by multiple-factor combinations of forage nutrients, including ether extract and CP paired with GE. The present output could provide useful information for the development of sustainable sheep grazing systems in the inland arid regions of the world.  相似文献

Spectroscopic and calorimetric melting studies of 28 DNA hairpins were performed. These hairpins form by intramolecular folding of 16 base self‐complementary DNA oligomer sequences. Sequence design dictated that the hairpin structures have a six base pair duplex linked by a four base loop and that the first five base pairs in the stem are the same in every molecule. Only loop sequence and identity of the duplex base pair closing the loop vary for the set of hairpins. For these DNA samples, melting studies were carried out to investigate effects of the variables on hairpin stability. Stability of the 28 oligomers was ascertained from their temperature‐induced melting transitions in buffered 115 mM Na+ solvent, monitored by ultraviolet absorbance and differential scanning calorimetry (DSC). Experiments revealed the melting temperatures of these molecules range from 32.4 to 60.5°C and are concentration independent over strand concentrations of 0.5 to 260 μM; thus, as expected for hairpins, the melting transitions are apparently unimolecular. Model independent thermodynamic transition parameters, ΔHcal, ΔScal, and ΔGcal, were determined from DSC measurements. Model dependent transition parameters, ΔHvH, ΔSvH, and ΔGvH were estimated from a van't Hoff (two‐state) analysis of optical melting transitions. Results of these studies reveal a significant sequence dependence to DNA hairpin stability. Thermodynamic parameters evaluated by either procedure reveal the transition enthalpy, ΔHcalHvH) can differ by as much as 20 kcal/mol depending on sequence. Similarly, values of the transition entropy ΔScalSvH) can differ by as much as 60 cal/Kmol (eu) for different molecules. Differences in free energies ΔGcalGvH) are as large as 4 kcal/mol for hairpins with different sequences. Comparisons between the model independent calorimetric values and the thermodynamic parameters evaluated assuming a two‐state model reveal that 10 of the 28 hairpins display non‐two‐state melting behavior. The database of sequence‐dependent melting free energies obtained for the hairpins was employed to extract a set of n‐n (nearest‐neighbor) sequence dependent loop parameters that were able to reproduce the input data within error (with only two exceptions). Surprisingly, this suggests that the thermodynamic stability of the DNA hairpins can in large part be reasonably represented in terms of sums of appropriate nearest‐neighbor loop sequence parameters. © 1999 John Wiley & Sons, Inc. In the captive Indian rhinoceros (Rhinoceros unicornis), two disease complexes with a high incidence—chronic foot problems and uterine leiomyomas—may be linked to excess body weight (BW). In this study, intake and digestion trials were conducted (by means of 7‐day weigh‐backs, and 5‐day total fecal collections, respectively) with 11 Indian rhinoceroses at four zoological institutions in Europe and the United States to quantify energy and mineral nutrition on conventional or roughage‐only diets. Diets comprising a variety of forages (grass hay only, a combination of grass hay and grass silage, straw, or a mixture of grass and legume hay) were offered as the roughage source, along with various concentrates, produce, and supplements. Water intake was quantified, and urine samples were obtained opportunistically. The animals consumed 0.5–1.1% of their BW in dry matter (DM) daily, with calculated digestible energy (DE, in megajoules MJ) values ranging from 0.27 to 0.99 MJ DE/kg BW0.75/day compared to an estimated requirement of 0.49–0.66 MJ DE/kg BW0.75/day. Seven of 11 rhinos (64%) fed restricted levels of concentrate plus forage consumed DE in excess of this estimate. Even on roughage‐only diets, some individuals consumed energy well above the presumed metabolic requirements. Hence, restriction of both concentrates and roughage may be important for weight management in this species. Water intake ranged from 30 to 49 mL/kg BW daily (3.4–5.2 L/kg ingested DM), similar to values that have been reported for domestic equids. Excretion amounts and patterns also resembled those found in horses. Endogenous fecal losses measured for Ca, P, Cu, Fe, and Zn indicate that the maintenance requirements of these minerals should be met in Indian rhinoceroses by diets that meet recommendations for domestic horses. It is particularly important to evaluate dietary adequacy in mineral nutrition in this species in concert with the need for restricted energy intake, especially with regard to the hypothetical involvement of a low Zn supply in chronic foot problems. ABSTRACT

The objective of this study was to estimate the sulphur amino acid (methionine + cystine) requirements and nitrogen endogenous losses in kittens aged 150 to 240 d. Thirty-six cats were distributed in six treatments (six cats per treatment) consisting of different concentrations of methionine + cystine (M + C): T1, 6.5 g/kg; T2, 8.8 g/kg; T3, 11.3 g/kg; T4, 13.6 g/kg; T5, 16.0 g/kg; and control, 6.5 g/kg. Diets were formulated by serial dilution of T5 (a diet relatively deficient in M + C but containing high protein concentrations) with a minimal nitrogen diet (MND). Thus, crude protein and amino acid concentrations in diets T1–T5 decreased by the same factor. The control diet was the T1 diet supplemented with adequate concentrations of M + C (6.5 g/kg; 8.8 g/kg; 11.3 g/kg; 13.6 g/kg and 16.0 g/kg). All diets were based on ingredients commonly used in extruded cat diets. Digestibility assays were performed for the determination of nitrogen balance. Nitrogen intake (NI) and nitrogen excretion (NEX) results data were fitted with an exponential equation to estimate nitrogen maintenance requirement (NMR), theoretical maximum for daily nitrogen retention (NRmaxT), and protein quality (b). M + C requirements were calculated from the limiting amino acid intake (LAAI) equation assuming a nitrogen retention of 45 to 65% NRmaxT. The NMR of kittens aged 150, 195, and 240 d was estimated at 595, 559, and 455 mg/kg body weight (BW)0.67 per day, respectively, and M + C requirements were estimated at 517, 664, and 301 mg/kg BW0.67 per day, respectively.  相似文献

Utilization of long-chain alcohols (LCOH) as diet composition markers in horses and cattle was assessed in a study conducted with 12 mature crossbreed mares (385±47 kg BW) and six adult non-lactating cows (499±36 kg BW) of Asturiana de los Valles breed. The LCOH data were combined with alkane and long-chain fatty acid (LCFA) data to test the applicability of combining these markers to estimate diet composition. Animals were randomly divided into groups of three animals and received a daily total amount of 1.0 kg dry matter/100 kg BW of diets composed of different proportions of ryegrass (Lolium perenne) and woody species (Ulex gallii and heather). Diet composition was estimated from even-chain LCOH (C20-OH to C30-OH) combined or not with alkane (C25-C31 and C33) and/or LCFA (C22-FA to C28-FA, C30-FA, C32-FA and C34-FA) concentrations in diet components and faeces by least-squares procedures, using marker faecal concentrations uncorrected for incomplete faecal recovery (FR0) or corrected using mean recoveries across diets within animal species (FR1). Results showed large differences between plant species in their LCOH profiles, and that these markers offered additional discriminatory information to that provided by alkanes and LCFA. The LCOH markers were incompletely recovered in the faeces of both animal species. In cattle, LCOH FR tended to increase with carbon-chain length in a linear manner in both diets (P<0.001), whereas in horses overall data showed a curvilinear relationship between these variables. Combination of LCOH, LCFA and alkanes resulted in more accurate diet estimates. Correction of faecal LCOH concentrations to incomplete FR led to more accurate diet composition estimates in both animal species. Results obtained in this study suggest the usefulness of LCOH markers combined with alkanes and LCFA to estimate diet composition of horses and cattle grazing mixed grassy–woody plant communities.  相似文献

BackgroundUrinary creatinine can be used to adjust urinary iodine to evaluate iodine nutritional status during pregnancy. However, the reference intervals and impact factors of urinary creatinine are unknown.Methods24 h urine creatinine concentration (24 hUCr) and spot UCr at four different time periods of the day of pregnant women from Part 1 (n = 743) were measured. Linear regression analysis was performed to identify the impact factors of 24 h urinary creatinine excretion (24 hUCrE) and obtain the estimated 24 h urinary creatinine excretion (24 hUCrEest). Then measured urinary iodine concentration (UIC) of 24 h and at fasting of pregnant women from Part 2 (n = 325), used spot urinary iodine to creatinine concentration ratio (UIC/UCr) and 24 hUCrEest to calculate the estimated 24 h urinary iodine excretion (24 hUIEest), finally checked the consistency and correlation of 24 hUIEest and 24 h urinary iodine excretion (24 hUIE).ResultsIn Part 1, the median 24 hUCrE was 1.24(IQR0.98–1.76)g, and the reference interval was 0.61−2.93 g. The median 24 hUCr was 0.76 (IQR0.57−1.01)g/L, and the reference interval was 0.36−1.88 g/L. Multiple linear regression results showed that pregnancy weight was an influencing factor to 24 hUCrE after adjusting by gestational weeks, age, pre-pregnancy BMI, and percentage of body fat (F = 45.029, p<0.001). In Part 2, there was no statistically significant difference between 24 hUIEest and 24 hUIE (Z =−0.767, p = 0.443). Using 24hUIE as the gold standard, the relative average difference in 24hUIEest was 4.2 %, the relative average differences for UIC and UIC/UCr were 32.4 % and 37.2 %. The reference interval of 24 hUIE and 24 hUIEest were 88.43–585.90 μg and 50.97–700.39 μg, respectively.ConclusionsThe reference intervals of 24 hUCrE, spot UCr, 24 hUIE, and 24 hUIEest during pregnancy were established. 24 hUCrE has important application value in iodine nutrition evaluation to gain more lead time for pregnant women with iodine nutrition-related diseases.  相似文献

We report on the isolation of eight microsatellites from the sand tiger shark, Carcharias taurus, using an enrichment protocol. All loci, with the exception of Cta45–183, were in Hardy–Weinberg equilibrium. Loci exhibited three to 15 alleles, and observed and expected heterozygosities of 0.095–1.000 and 0.284–0.924, respectively. An additional marker (Iox‐12) developed from a shortfin mako library was variable in sand tigers. These markers will be used to examine population genetics and mating patterns of this imperilled species.  相似文献

Abstract

An experiment was conducted in young pigs (initial BW 10.1 kg) to estimate the maintenance requirement for histidine and its efficiency of utilization for protein accretion using a comparative slaughter technique. Three groups of six pigs each were fed a purified diet supplying 0, 14 or 56 mg histidine per kg BW0.75. Following 21 d of feeding, pigs were killed for whole body compositional analysis. A representative group of six pigs was killed at the beginning of the experiment. Retention of histidine and total N were the main criteria of response. Histidine retention (R2 = 0.73) and N retention (R2 = 0.78) were linear functions of histidine intake (p < 0.001). Histidine requirement for zero histidine retention was 15.5 mg/kg BW0.75, whereas histidine required for zero N retention was 4.1 mg/kg BW0.75. At zero histidine retention, the pigs retained daily 82 mg N/kg BW0.75, presumably due to the degradation of histidine-rich compounds such as haemoglobin and/or carnosine. The slope of the regression line relating histidine retention to N retention indicated that 105 mg of histidine was deposited per gram of total N which was considerably less than the estimated histidine concentration in body protein (179 mg/g N). Based on the slopes of regression equations for histidine and N retention, marginal efficiency of histidine utilization was calculated to be 0.94 and 1.34, respectively.  相似文献

In the nutrition of browsing ruminants in captivity, adequate nutrient digestibility and energy content of diet is debated. Problems related to energy‐provision and low forage intake have been reported for the okapi and other browsers like the giraffe, particularly during winter. High‐fiber concentrates like unmolassed beet pulp have some potential to improve the nutritional management of these species. Using a total of six okapis in captivity, seven feeding trials were carried out at two facilities (A+B) on a structured but opportunistic base. Three trials (A1, A2, B1) were conducted when animals were fed their regular diet including grain based energy concentrates, fruits and vegetables, and alfalfa (Medicago sativa) hay. Two trials (A5, B2) examined the effect of unmolassed beet pulp, and two (A3,4) examined the effect of unmolassed beet pulp+fresh browse. Daily intake and feces production were quantified over 8–12 days. Samples were analyzed for dry matter, crude ash, neutral detergent fiber (NDF)/acid detergent fiber (ADF)/acid detergent lignin (ADL), crude protein, and gross energy. Metabolizable energy content of diets was estimated via a factor (0.83) from digestible energy. The proportion of beet pulp in diets was 13% (A3), 24% (A4), 20% (A5), and 21% (B2). Browse proportion was 11% (A3) and 32% (A4). Daily feed intake ranged between 1.5–1.7% of body weight (BW), digestibility of organic matter between 61–74%. Digestibility of fiber (NDF) was higher in beet pulp diets (A3=39%, A4=60%, A5=54%, B2=61%) than in the others (A1=48%, A2=33%, B1=48%). Supply of metabolizable energy (ME) ranged between 0.50–0.70 MJ ME/(kg BW0.75*day), meeting energy requirements of okapis of 0.50–0.53 MJ ME/(kg BW0.75*day) in general. Diets with beet pulp+browse were not found to be highest, but in the upper level of the range of forage proportions of this study. Palatable browse species were preferred over all other feedstuff offered. The use of unmolassed beet pulp as energy‐concentrate for browsing ruminants like the okapi can be recommended because diets high in this high‐fibre feedstuff resulted in adequate energy intakes. Serial ultrasound examination of four mature female sevengill sharks (Notorynchus cepedianus) was carried out over 18 months. Monitoring the reproductive cycle and development of follicles and fetuses in sharks in a noninvasive manner using this technique has not been reported previously. Sharks were caught out of the “Oceanarium” tank by divers using a specially made catch‐out bag, and brought to a holding area for examination. A behavior scoring system was used to monitor the impact of regular handling on the well‐being of the animals. Ultrasound showed the growth and regression of follicles in sevengill ovaries, and allowed an approximation of the reproductive stage of these sharks. Monitoring behavior at five time points during the procedure showed that regular handling of sharks for clinical studies could be done with minimal impact on animal welfare. The ability to follow reproductive events in elasmobranches using ultrasonography is an important step in the application of assisted reproductive technology in these species. Assisted reproductive technology, such as monitoring female reproductive cycles and artificial insemination, could potentially be used to maintain genetic diversity and compliment aquaria‐based breeding programs for endangered species such as the gray nurse shark (Carcharias taurus). Fifty non-pregnant Surti buffalo heifers aged between 17 and 42 months (n=24, <24 months;n=26, >24 months) were randomly assigned to groups subject to either natural daylight +4h supplemental light (n=25) or natural day light (n=25), to study changes in growth, serum prolactin (Prl), progesterone (P4) and luteinizing hormone (LH) to supplemental lighting. Ambient temperatures (T) and relative humidity (RH) generally were >27° C and <70% during the day-time, respectively. Light-supplemented heifers had 16.2 kg net body weight (BW) gain at 9 weeks compared to 20.8 kg for controls, but higher mean Prl after 6.5 weeks (P<0.01), and higher P4 (0.41 vs 0.19 ng/ml;P<0.06) than control heifers. Older heifers had 39.7% greater BW (P<0.01), but a net 4.3% BW gain compared to a 10.1% gain for younger heifers at 10 weeks. Older, light-supplemented heifers had higher mean P4 (0.63 vs 0.19 ng/ml;P<0.07) than the other groups. These weight and hormonal changes suggest that 4 h supplemental light can alter growth and endocrine function in buffaloes under similar planes of nutrition. While light supplementation did not have a positive effect on body wieght during the 10 week study, body weight and endocrine changes due to supplemental light may be important factors for initiation of reproductive cyclicity.  相似文献

Ten diets (196?g CP and 13.0 MJ MEN/kg) with graded levels of tryptophan (Trp) between 0.9 and 2.7?g/kg were offered ad libitum to Ross broilers from day 8?–?21 post-hatch. Each diet was allocated to three pens of ten birds each. In addition to growth and feed conversion, the accretions of protein, Trp, fat, and energy were determined by comparative whole body analyses. A sigmoidal function was fitted to the data. Responses to Trp supply were nonlinear and attained plateau values (ymax) within the studied range of Trp supply. The ymax values estimated for BW gain, whole body protein gain, and Trp gain during the 14 days under study were 615, 104, and 0.87?g/bird, respectively. Based on the response in whole body protein gain, the estimated requirement was 1.9?g Trp/kg of diet or 0.15?g Trp/MJ of MEN. Estimates made from the other response criteria were similar to this value. While the protein concentration in gained BW was unaffected by Trp intake (169?g of protein per kg of gained BW), fat and energy concentrations of gained BW increased with increasing Trp supply, approaching plateau values of 146?g fat and 9.8 MJ energy per kg of gained BW. This is supposed to result from feed intake increasing with Trp supplementation. The Trp concentration in gained whole body protein (0.84?g Trp/16?g of gained N) was not significantly affected by Trp intake. As long as Trp intake was the limiting factor, 59% of supplemented Trp was transferred to gained whole body protein. During the 14 days of the study, broilers inevitably lost 87?mg Trp. This is equivalent to a daily loss of 30?mg/kg BW or a maintenance requirement of 52?mg/kg BW per day. Data determined for the individual factors may be used for future modelling of broilers' Trp requirement.  相似文献

An accurate value for metabolizable energy (ME) requirement for maintenance (MEm) is essential to enable sheep husbandry practice to reach its potential. The objectives of the study were to use calorimetry chamber data of dry ewes (Hu × thin-tail Han F1 crossbred) to develop updated MEm, examine effects of substituting concentrate feed with lucerne hay on energy partitioning, and explore the relationships between energy utilization and fasting heat production (FHP). Data were collected from three experiments. In Exps. 1, 2a and 2b, lucerne hay was used to replace concentrates in three levels (0:40%, 15:25% and 30:10%), with diets containing 60% maize stover (Exp. 1), fresh rye forage (Exp. 2a) or dry rye forage (Exp. 2b). Within each experiment, diets were isoenergetic (digestible energy, DE) and isonitrogenous. Exp. 3 aimed at evaluating effects of three BW levels on nutrient utilization of dry ewes offered diets containing 60% maize stover, 15% lucerne hay and 25% concentrates. Energy metabolism data were measured using the respiration calorimeter chamber technique in all three experiments, followed by the measurement of FHP in Exps. 1, 2b and 3. The MEm derived from the linear regression between energy balance (EB) and ME intake was 0.440 MJ/kg BW0.75. The average FHP was 0.326 MJ/kg BW0.75. The fasting metabolism, net energy requirement for maintenance (NEm) and MEm were estimated to be 0.336, 0.359 and 0.511 MJ/kg BW0.75, respectively, through adjustment of FHP using fasting urinary energy output, activity allowance and efficiency of ME use for maintenance. The FHP was negatively correlated to EB/metabolic BW, ME/gross energy (GE), ME/DE, EB/GE intake and EB/ME intake, while positively correlated to HP/GE intake, HP/ME intake and CH4-E/GE intake. Compared to zero lucerne hay diet, the 15% lucerne hay intake decreased HP (MJ/d), and had no negative effects on EB (MJ/d) or energy utilization efficiencies. The results indicate that nutrient requirement standards currently used across the world are likely to underestimate MEm for dry ewes, and the selection of low FHP ewes for breeding has the potential to improve sheep production efficiency.  相似文献

We evaluated the extent of shrinkage in body size of juvenile marbled sole Pseudopleuronectes yokohamae after preservation in 70% ethanol for 6, 12, 24, or 48 h, or 4 weeks. Standard length (SL, range of the analyzed specimens: 22.82–53.01 mm) decreased by 5.6% after 12 h and body weight (BW 0.174–2.964 g) decreased 27.8% after 24 h preservation in 70% ethanol. There was no further decrease in SL or BW after 12 or 24 h of preservation, respectively. The original body size could be estimated based on the size after preservation in 70% ethanol for 4 weeks using the following equations: SL original = 1.05 SL preserved + 0.37 and BW original = 1.36 BW preserved + 0.04. The condition factor calculated using the body size of preserved individuals was 16.0 and 15.8% lower than that calculated from the original body size and the body size back-calculated using the linear regression equations described above, respectively. Thus, our data suggest that the shrinkage of body size due to preservation in ethanol can cause errors in estimation of the condition factor. Our results can be used to improve the accuracy of estimating size-related biological parameters based on juvenile marbled sole that are preserved in ethanol.  相似文献

Increasing the concentration of dietary lipid is a promising strategy for reducing methane (CH4) emissions from ruminants. This study investigated the effect of replacing grass silage with brewers’ grains on CH4 emissions of pregnant, non-lactating beef cows of two breeds. The experiment was a two×two factorial design comprising two breeds (LIMx, crossbred Limousin; and LUI, purebred Luing) and two diets consisting of (g/kg diet dry matter (DM)) barley straw (687) and grass silage (301, GS), or barley straw (763) and brewers’ grains (226, BG), which were offered ad libitum. Replacing GS with BG increased the acid-hydrolysed ether extract concentration from 21 to 37 g/kg diet DM. Cows (n=48) were group-housed in equal numbers of each breed across two pens and each diet was allocated to one pen. Before measurements of CH4, individual dry matter intake (DMI), weekly BW and weekly body condition score were measured for a minimum of 3 weeks, following a 4-week period to acclimatise to the diets. CH4 emissions were subsequently measured on one occasion from each cow using individual respiration chambers. Due to occasional equipment failures, CH4 measurements were run over 9 weeks giving 10 observations for each breed×treatment combination (total n=40). There were no differences between diets for daily DMI measured in the chambers (9.92 v. 9.86 kg/day for BG and GS, respectively; P>0.05). Cows offered the BG diet produced less daily CH4 than GS-fed cows (131 v. 156 g/day: P<0.01). When expressed either as g/kg DMI or kJ/MJ gross energy intake (GEI), BG-fed cows produced less CH4 than GS-fed cows (13.5 v. 16.4 g/kg DMI, P<0.05; 39.2 v. 48.6 kJ/MJ GEI, P<0.01). Breed did not affect daily DMI or CH4 expressed as g/day, g/kg DMI or kJ/MJ GEI (P>0.05). However, when expressed as a proportion of metabolic BW (BW0.75), LUI cows had greater DMI than LIMx cows (84.5 v. 75.7 g DMI/kg BW0.75, P<0.05) and produced more CH4 per kg BW0.75 than LIMx cows (1.30 v. 1.05 g CH4/kg BW0.75; P<0.01). Molar proportions of acetate were higher (P<0.001) and propionate and butyrate lower (P<0.01) in rumen fluid samples from BG-fed compared with GS-fed cows. This study demonstrated that replacing GS with BG in barley straw-based diets can effectively reduce CH4 emissions from beef cows, with no suppression of DMI.  相似文献

Accurate prediction of energy requirement is important in formulating diets, but an energy model for Yellow Broiler breeder hens is publicly unavailable. The objective of this study was to establish energy prediction models for the nitrogen-corrected apparent metabolisable energy (AMEn) requirement of different categories of Yellow Broiler breeder hens during the egg-laying period. Data for modelling were collected from research papers, public databases and production data from companies. Breeder hens were generally categorised into three BW types: heavy, medium and light (HBWT, MBWT and LBWT). Published articles were cited for providing coefficients of AMEn maintenance requirement (AMEnm, 101 kcal/kg BW0.75, 423 KJ/kg BW0.75) and growth requirement (AMEng, 5.33 kcal/g, 22.3 KJ/g), respectively. Models of AMEn for egg production (AMEnp) were established from the known daily intake of AMEn (AMEni) and those of maintenance and growth by the factorial approach: AMEnp = AMEni ? AMEnm ? AMEng. For the three types of hens, AMEnp HBWT (kcal, KJ) = 2.55 kcal (10.7 KJ) × egg mass (EM, g); AMEnp MBWT (kcal, KJ) = 2.70 kcal (11.3 KJ) × EM (g), and AMEnp LBWT (kcal, KJ) = 2.94 kcal (12.3 KJ) × EM (g) were determined. The total AMEni requirements, depending on Gompertz models, were HBWT: BW (g) = 3 144 × e?EXP(?0.162×(week of age (wk)?15.6)); MBWT: BW (g) = 2 526 × e?EXP(?0.333×(wk?19.1)); LBWT: BW (g) = 1 612 × e?EXP(?0.242×(wk?16.5)). Models of egg production, HBWT: egg production (%) = 124 × e?0.017×wk/(1 + e?0.870×(wk?26.2)); MBWT: egg production (%) = 144 × e?0.020×wk/(1 + e?0.751×(wk?24.9)); LBWT: egg production (%) = 163 × e?0.024×wk/(1 + e?0.476×(wk?26.5))) and egg weight for each wk of the three types of hens during the egg-laying period were all established.
