Summary The fine structure, origin, and distribution density of the autonomic nerve endings in the tarsal muscle of the mouse were studied by histochemistry and electron microscopy. With histochemical methods, the fine nerve plexus in the normal muscle shows both catecholamine-positive varicose fibers and acetylcholinesterase-active varicose fibers. The former are distributed more densely than the latter. After superior cervical ganglionectomy, the catecholamine-positive fibers disappear, while after pterygopalatine ganglionectomy, the acetylcholinesterase-active fibers vanish. In electron micrographs, the varicosities appear as expansions containing many synaptic vesicles. The axonal expansions partly lack a Schwann sheath and directly face the pinocytotic vesicle-rich zones of the smooth muscle cells. A relatively wide space, 0.1 to 1.0 m in width, lies between nerve expansion and muscle cell. The expansions can be classified into two types: Type I having small granular synaptic vesicles, and Type II having agranular vesicles instead of small granular synaptic vesicles. Type I undergoes degeneration after superior cervical ganglionectomy, while Type II degenerates after pterygopalatine ganglionectomy. This indicates that Type I corresponds to the synaptic ending of the adrenergic fiber originating from the superior cervical ganglion, and Type II to the synaptic ending of the cholinergic nerve fiber derived from the pterygopalatine ganglion. Ganglion cells and paraganglionic (PG) cells in the developing rat superior cervical ganglion were studied following postnatal treatment with p-chlorophenylalanine (pCPA) for 5 to 8 days. Litter mates, injected with saline solution, served as controls. Ganglion cells of control animals were differentiated ultrastructurally according to L. Er?nk? (1972a) into late sympathicoblasts and young sympathetic nerve cells. In both maturation stages treatment with pCPA caused marked swelling of mitochondria, concomitant with minor changes of other cell organelles. Parallel to the ultrastructural alterations, fluorescence microscopy and cytophotometry revealed a slight diminution of diffuse fluorescence intensity in sympathetic neurons as the expression of a mainly extragranular amine depletion. In distinction from ganglion blocking agents the alterations are regarded as a general toxic effect of pCPA upon maturing sympathetic neurons, which secondarily influences catecholamine storage sites. Following treatment with pCPA, in PG-cells an alteration of mitochondria was scarcely to recognize. Specific granules were distinctly decreased in number, in some cases to an almost complete degree. Concordant to ultrastructural observations a marked diminution of fluorescence intensity was demonstrable in SIF-cells. In addition in these elements the fluorescence spectrum shifted towards the green field. Fluorescence cytophotometric evaluations confirmed the optical impression. Provided, that PG-cells, demonstrated with electron microscopy, are identical with SIF-cells in fluorescence microscopy, the results are discussed on the basis of a specific decrease of primary catecholamines due to an enzyme inhibition involved in catecholamine synthesis.  相似文献

Summary The ultrastructure of the paracervical (Frankenhäuser) ganglion in the rat was studied after immersion or perfusion fixation with glutaraldehyde followed by post-osmification. This ganglion is located at the uterovaginal junction in the vicinity of arteria uterina and contains three neuronal cell types. (1) Principal neurons have a fine structure mainly similar to the ganglion cells of other autonomic ganglia. (2) Small granule-containing cells occur in clusters often close to fenestrated capillaries. They are divided into two subgroups according to the size of their cytoplasmic granules; those containing only small granulated vesicles of 800 to 1400 Å in diameter and those having also large granulated vesicles of 2000 to 3000 Å in diameter. (3) Vacuolated nerve cells are large cells that resemble the principal neurons in their cytoplasmic components, except that they contain one to ten vacuoles with corpuscles of different size and shape. The possible physiological significance of the small, granule-containing cells in the uterine function is discussed.  相似文献

A volume of tissue of lettuce (Lactuca sativa L.) plants extending 2 mm basipetally from the apical meristem and including leaf primordia and young expanding leaves was surveyed using electron-probe microanalysis (EPMA) on both frozen-hydrated and freeze-dried samples. This analysis was carried out either 2 or 5 d following NaCl salinization of the medium from the 10 mol · m–-3 control level up to 80 mol · m–-3. The objective was the investigation of possible changes in the nutritional status of the apical meristem that might account for some aspects of salt-induced growth inhibition. Sodium and chloride increased significantly in tissues basal to the apical meristem, while both phosphorus and potassium decreased in the same region. These changes were evident in specimens collected just 2 d after the commencement of salinization (20 h after completion of the salinization) and were not exacerbated by an additional 3 d of treatment; they were present in tissue as close as 100 m to the meristem and extending down to 500 m. The apical 10–50 m were relatively protected from both the increase in sodium and chloride and the decrease in phosphorus and potassium that occurred in more basal regions. Young leaves (up to 1.5 mm in length) appear to control their own mineral nutrient levels when challenged by salinization of the medium, presumably because of altered growth. A decrease in the concentration of total Ca as a result of salinization was significant in cells 500 m basal to the meristem, but was evident as a tendency in the data even within the first 50 m. Using an improved automatic method for the analysis of calcium by EPMA, it was found that total Ca was reduced by salinization, especially in basal regions (500 m below the apex) and also in young leaves (1–1.5 mm in length). We suggest that the nutrition of the shoot apical meristem may be disturbed soon after salinization and that the shoot meristem might be the source of a signal to expanding leaves, as well as exerting its own direct influence over leaf emergence.Abbreviation EPMA electron-probe microanalysis This work was supported by U.S. Department of Agriculture grant 87-CRCR-1-2462.  相似文献

Summary Changes in the density of nuclear chromatin in the shoot apical meristem ofSinapis alba L. during floral transition (floral evocation) are described using Feulgen-stained 2 m thick semi-thin sections and scanning cytophotometric techniques. In both G1 and G2 nuclei the chromatin becomes less heterogeneous and less dense in evoked meristems compared to vegetative meristems. When chromatin is resolved into two fractions the dispersed fraction increases relative to the condensed fraction at evocation. This decondensation process occurs earlier in G1 than in G 2 nuclei. These chromatin changes are presumably closely related to the dramatic stimulation of biosynthetic activity and cell division during floral transition.  相似文献

Summary The formation and development of synaptic contacts between dissociated chick spinal cord neurons has been investigated. By the 6th day in vitro immature profiles with few vesicles were observed. By 14–18 days mature types with numerous vesicles were found, indistinguishable from those of newly hatched chick spinal cord. After this period degeneration occurred, and was especially marked in the post-synaptic element. Such degeneration could be postponed by the addition of small numbers of somatic muscle cells. The Kanaseki and Kadota (1969) technique was applied to the study of coated vesicles at various stages of synaptic development.  相似文献

Summary In white muscle fibres of a teleost fish T-tubule openings may occur regularly at all Z-disc levels between adjacent peripheral myofibrils, the T-tubule openings thus occurring at a density of ca. 0.9 m-2.In frog white fibres, T-tubule openings are infrequently seen in material fixed like the fish material. In material prepared according to the albumin method of Gray (1975, 1976 a, b) which renders the muscle fibres swollen, straight tubules or sometimes chains of vesicles instead are seen opening at the sarcolemmal surface. Such tubules occur at a higher density than expected from experiments with local activation of contraction. Lability and dynamics within the T-system normally and during fixation are discussed.  相似文献

Summary The sinus gland is a neurohemal organ located in the crayfish eyestalk and represents a storage site for neurohormones prior to their release into the circulation. The sinus gland contains three classes of electron dense, membrane-limited granules. Class 3 granules are the largest and most electron dense of the granules found in the sinus gland. Granules of class 1 are the smallest, while those of class 2 are the most abundant. Generally, all granules undergo similar changes during their release.Release of neurosecretory material may be initiated by a preliminary fragmentation of the parent granules into smaller granules. Following the formation of numerous smaller granules, these move to the plasma membrane and their limiting membrane apparently fuses with it thus releasing its contents into the external lamina which is applied to the sinusoidal surface of the axon terminals. Granule release does not appear to occur along the entire plasma membrane adjacent to the blood sinus but, instead, probably occurs only at specific active sites on the membrane. The active sites are characterized in part by an accumulation of small granules and clear vesicles against the cytoplasmic side of the plasma membrane. At the site of release of the neurohormone, there is often an accumulation of dense homogeneous material beneath the axolemma.Occasionally, axon endings filled with large, electron lucent vesicles are seen. These clear granules vary from 1150–1750 Å in diameter and often exhibit broken limiting membranes. Few small vesicles are seen near the plasma membrane of these endings; however, instances of invaginations of the plasma membrane occur. The predacious watermold Zoophagus insidians traps loricate rotifers on short, lateral branches of the main, hyphal axis. These branches or traps are packed at their distal ends with a number of vesicles filled with an electron-dense matrix. Electron micrographs of the mycelium disclose a two-layered wall; the outer layer is electron dense and the inner, electron transparent. The outer dense layer on the tip of the trap is organized into a number of fine ridges and occasional discontinuities. Thin sections through recently trapped rotifers indicate that the cilia of the animals are stuck to the trap by a glue. This adhesive is derived from secretion of the matrix of the vesicles aggregated in the tip of the trap. The secretion mechanism is triggered by the animal and is accompanied by: 1. The separation of the two layers of the wall, 2. fusion of the vesicles with the cell membrane and 3. extrusion of the glue through pits in the tip of the inner wall of the trap. After snaring a rotifer, the previously arrested branch grows as a haustorium into the body cavity of the animal. The host tissues disintegrate within a few hours and appear to be the main nutrient source of the fungus.  相似文献

Summary The Polian vesicles are tubules of variable length consisting of four layers: an external peritoneal epithelium, a connective tissue layer, a muscular layer, and an inner epithelium. The two simple epithelia produce a polysaccharide in their Golgi complexes. Their cellular junctions consist of an extensive zonula adhaerens, and an apical septate desmosome. Their surfaces possess microvilli and cilia. A basement membrane is lacking. The muscular layer is composed of paramyosin fibers. The connective tissue layer contains abundant collagen fibers that have a period of 640 Å and are embedded in an acid mucopolysaccharide matrix. Amoebocytes containing waste products and an acid mucopolysaccharide enter the Polian vesicles from the coelom, pass through them, and empty into the lumen along with material secreted by the inner epithelium. While establishing close contact with cells of the muscular layer and inner epithelium the amoebocytes seem to transfer part of their contents to these cells.Polian vesicles appear to be a very primitive excretory organ.Research performed under C.N.R. contract.  相似文献

Summary Root tips of radish,Raphanus sativus, were fixed in glutaraldehyde followed by osmium tetroxide. The fine structure of young root hairs, not exceeding about 130, in length, was studied to relate their apical growth pattern to their cytoplasmic organization. The cytoplasm in the terminal 3–5 it of the root hair is characterized by an electron dense matrix in which lie numerous smooth-surfaced vesicles, large irregularly-shaped fibrous inclusions, and clusters of ribosomes. Other organelles are largely or entirely excluded from this region. Farther than about 5, from the tip, the hair cytoplasm is filled with plastids, rough endoplasmic reticulum, mitochondria, and dictyosomes. The latter produce smooth vesicles similar in size and morphology to those present in the apical dome. Vesicles of a different kind appear in the peripheral cytoplasm along the entire length of the hair. These vesicles possess an alveolate or chambered coat about 20 m thick and have a diameter of about 85 m, including coat. They originate by evagination from the large, smooth-surfaced vesicles in the vicinity of dictyosomes. It is suggested that proteins and carbohydrates are concentrated in the dictyosomes and then segregated in the smooth vesicles released from the dictyosome cisternae. The coated vesicles which bud from the smooth vesicles may serve to isolate the proteins and transport them to the hair surface for participation in wall synthesis. The smooth vesicles are believed to convey carbohydrates to the region of active wall extension at the hair apex.This work was supported in part by grant GM-10493 from the National Institutes of Health. Summary In the cerebral (= supraesophageal, suprapharyngeal) ganglion of the earthworm, a number of neurosecretory Gomori-positive perikarya are bipolar; others are unipolar, or multipolar. Some of the neurosecretory cell processes project centrally into a fibrous zone; peripheral processes enter small nerves which leave the dorsocaudal aspect of the ganglion.In the central fibrous zone, the neurosecretory fibers form varicose Gomoripositive terminals. Here, also zinc-iodine-osmium (ZIO)-positive fibers and monoamine fluorescent fibers are found. With the electron microscope, nerve terminals containing synaptic vesicles and either large neurosecretory peptidergic granular vesicles (diameter more than 1500 Å), or smaller granular vesicles (diameter about 1300 Å, or 900 Å) are observed. These axon endings mainly form axo-dendritic synapses. Peptidergic profiles are both pre- and postsynaptic. Some of the extraganglionic peptidergic fibers appear to terminate around vessels, but most of them form terminals on the visceral muscle cells which surround the ganglion.We think that the central neurosecretory processes communicate with the fibers of the synaptic zone of the ganglion. The peripheral neurosecretory peptidergic fibers are supposed to form a primitive neurohemal area and/or to function as vasomotor nerves. The fibers innervating the visceral muscle cells may represent vegetative nerves.  相似文献

A characteristic layer containing numerous fibrils is associated with the basement membrane of the inner enamel epithelium during the early stages of odontogenesis. However, its nature is not well understood. In this study, the layer was examined with high-resolution electron microscopy and immuno-histochemical staining. Tooth germs of monkeys (Macaca fuscata) were studied and each fibril in the layer was found to be a tubular structure, 8–9 nm in width, resembling a basotubule, the tubular structure previously observed in various basement membranes. The space between the fibrils was filled with a network formed by irregular anastomosing strands with an average thickness of 4 nm; these strands resembled the cords forming the network in the lamina densa of basement membranes. After immunoperoxidase staining, fine threads immunoreactive for laminin staining were seen winding along the strands of the network, and 1.5-nm wide filaments, immunoreactive for type IV collagen, took the form of a network arrangement. The 5-nm-wide ribbon-like structures associated with the strands were identified as heparan sulfate proteoglycan by immunostaining. These results are similar to those obtained for the cord network of the lamina densa. The fibrillar layer therefore represents a highly specialized lamina fibroreticularis of the basement membrane of the inner enamel epithelium, and rich in basotubules.  相似文献

Summary Freeze-fracture studies were conducted on the membranes of normal cockroach hemocytes. The plasmalemma is asymmetric with the A fracture face containing 80–100 Å membrane intercalated particles at a concentration of 2500/2. The B fracture face contains 120–150 Å particles with a relatively low density (800/2). The nuclear envelope displays an asymmetry with the A fracture face containing 1500 particles/2 and the B face containing 300/ 2. No significant particle size differences were observed in nuclear envelope fracture faces. Two types of symmetric membranes were also found in these cells. Both A and B fracture faces of the membrane surrounding the numerous cytoplasmic inclusion bodies contain particle sizes and concentrations similar to the B face of the plasmalemma. A second type of symmetry was observed in cells apparently engaged in exocytosis. Vesicles (0.1 D) from this process were completely particle free on both fracture faces. Summary The role and fate of male germ cells in planarian regeneration was studied in a population ofDugesia lugubris s.1. which provided a suitable karyological marker to distinguish diploid male germ cells from triploid embryonic and somatic cells. The nuclear Feulgen-DNA content in non-replicating triploid muscle cells of the pharynx and in non-replicating male gonia of testes from intact animals were measured by the cytophotometric technique. The pharynx was then removed by transection and each anterior regenerant was allowed to completely regenerate this organ. Measurements of the Feulgen-DNA content in muscle cells of the regenerated pharynx showed that most of these cells (95%) have a DNA content typical of triploid cells; however, some muscle cells (5%) with a nuclear DNA content typical of male gonia alone were observed.These results were interpreted in the following way. After transection, young male germ cells move from the testes to the wound where they participate in blastema formation along with reserve and/or somatic dedifferentiated cells. During regeneration some of these cells of male origin differentiate into pharyngeal muscle cells. Our findings are discussed in relation to the occurrence of mataplasia in planarians.  相似文献

Summary Ventral and dorsal aortic blood pressure, heart rate and plasma concentrations of adrenaline and noradrenaline have been measured in Atlantic cod before and after stress. The stress was induced by lowering the water level in the tank, which forced the animals to lie on their side struggling to regain the normal posture. The effects of stress were studied in fish in which the nerve supply to the head kidney was sectioned, using sham-operated animals as controls. In control animals, there was an increase in the ventral aortic blood pressure and plasma levels of both catecholamines as a result of stress while the dorsal aortic blood pressure remains constant and heart rate, if anything, decreases. The effect on the heart rate can be blocked by atropine, indicating a vagal reflex. In fish where catecholamine release from chromaffin tissue was strongly reduced by sectioning the nerve supply to the head kidney, the dorsal aortic blood pressure was lower before stress by comparison to controls, and decreased further following stress. No significant changes in ventral or dorsal aortic blood pressure and heart rate were observed in another group of fish where the sympathetic innervation of the gills had been sectioned, when compared to sham-operated controls before or after stress. It is concluded that circulating catecholamines released from the head kidney play a major role in the control of branchial vascular resistance after stress, counter-acting the effect of a non-adrenergic constrictory innervation of the gills. Mild cerebral anoxic/ischemic/stress insults promote tolerance and thereby protect the brain from subsequent lethal anoxic/ischemic insults. We examined whether specific activation of PKC , , , or isoforms is associated with ischemic preconditioning (IPC) in rat brain. IPC was produced by a 2-minute global cerebral ischemia. Membrane and cytosolic fractions of the hippocampi were immunoblotted using specific antibodies for PKC, , , and . PKC showed a significant translocation to the membrane fraction from 30 min to 4 h and PKC at 4 h following IPC. In contrast, the membrane/cytosol ratio of PKC showed a tendency to decrease at 30 min and 8 h, and the membrane/cytosol ratio of PKC was significantly decreased from 30 min to 24 h following IPC. These findings indicate PKC isoform-specific membrane translocations in the hippocampus after brief global brain ischemia and suggest that activation of PKC and PKC may be associated with IPC-induced tolerance in the rat hippocampus.  相似文献

Cytoplasmic membrane vesicles were prepared by a lysozyme-salt treatment from Arthrobacter P1 grown on methylamine as the carbon and energy source. In the presence of an ascorbate-phenazine methosulphate electron donor system, these vesicles accumulated methylamine in unmodified form by an inducible transport system. This system has a high affinity for methylamine (Kapp=20–25 M). The effect of the ionophores valinomycin and nigericin combined with membrane potential () and pH-gradient (pH) measurements demonstrated that methylamine uptake is electrogenic and driven by the . Optimal activity is observed at pH 6.5 and 30°C. Methylamine uptake was not affected by the presence of ammonium ions but was inhibited by the primary amines ethylamine (competitively), propylamine, butylamine and benzylamine. In addition, formaldehyde and acetate, at a concentration of 1 mM, inhibited methylamine uptake almost completely. These compounds were shown to be non-competitive inhibitors. A strong inhibition observed in the presence of plumbagin could be relieved by addition of dithiothreitol. This indicates that the oxidation-reduction state of, probably, carrier dithiol-disulfide-groups is an important factor in methylamine translocation in Arthrobacter P1.  相似文献

Summary Male ventral and female prostates of Praomys (Mastomys) natalensis were examined with the electron microscope. The findings support and add to information obtained with the light microscope on tissues from normal, castrated and ovariectomised animals.Our results indicate that although the female prostate may be considered a homologue of the male ventral prostate anatomically and histologically, there are differences in sub-cellular morphology and hormone dependence.Cells of the intact ventral prostate of the male are characterised by prominent dilated Golgi vesicles and electron-dense mature secretory granules seen in the apical region of the cell. In the cells of the female prostate these features are absent. These morphological differences reflect the influence of hormones upon the cells, as shown by the reduction of the dilated Golgi vesicles in the castrated male and conversely, their occasional presence in the cells of the oestrous female.Comparison of castrated and ovariectomised animals shows that the male ventral prostate is much more dependent on androgens than the female is on ovarian hormones.There are several modes of secretion in the male ventral and the female prostate. These are by acellular and cellular blebbing, by a variety of secretory vesicles into the acinar lumina, and by a system of double walled vesicles not previously described.We are grateful to Dr. R.C.B. This study reports a novel splice variant form of the voltage-dependent calcium channel 2 subunit (2g). This variant is composed of the conserved amino-terminal sequences of the 2a subunit, but lacks the -subunit interaction domain (BID), which is thought essential for interactions with the 1 subunit. Gene structure analysis revealed that this gene was composed of 13 translated exons spread over 107 kb of the genome. The gene structure of the 2 subunit was similar in exon-intron organization to the murine 3 and human 4 subunits. Electrophysiological evaluation revealed that 2a and 2g affected channel properties in different ways. The 2a subunit increased the peak amplitude, but failed to increase channel inactivation, while 2g had no significant effects on either the peak current amplitude or channel inactivation. Other subunits, such as 3 and 4, significantly increased the peak current and accelerated current inactivation.  相似文献
