In contrast to mammalian erythrocytes, chicken erythrocytes contain fructose 2,6-bisphosphate at levels (0.5 nmol/10(9) cells) similar to those of 2,3-bisphosphoglycerate (1.2 nmol/10(9) cells) and slightly lower than those of glucose 1,6-bisphosphate (5.2 nmol/10(9) cells). In chick embryo erythrocytes the levels of both fructose 2,6-bisphosphate and glucose 1,6-bisphosphate are much lower. They begin to increase at hatching and reach the levels in chicken in a few days.  相似文献

Fructose 2,6-bisphosphate and several glycolytic intermediates were measured in two rat muscles, extensor digitorum longus and gastrocnemius, which were electrically stimulated in situ. Both the duration and the frequency of stimulation were varied to obtain different rates of glycolysis. There was no relationship between fructose 2,6-bisphosphate content and the increase in tissue lactate in contracting muscle. However, in gastrocnemius stimulated at low frequencies (less than or equal to 5 Hz), there was a 2-fold increase in fructose 2,6-bisphosphate at 10s, followed by a return to basal values, whereas lactate increased only after 1 min of contraction. The concentrations of hexose 6-phosphates, fructose 1,6-bisphosphate and triose phosphates were all increased during the 3 min stimulation. During tetanus (frequencies greater than or equal to 10 Hz) fructose 2,6-bisphosphate was not increased, whereas glycolysis was maximally stimulated and resulted in an accumulation of tissue lactate, mostly from glycogen. The concentrations of hexose 6-phosphate increased continuously during the 1 min tetanus, whereas fructose 1,6-bisphosphate was increased at 10s and then decreased progressively. It therefore appears that fructose 2,6-bisphosphate does not play a role in the stimulation of glycolysis during tetanus; it may, however, be involved in the control of glycolysis when the muscles are stimulated at low frequencies for short periods of time.  相似文献

Fructose 2,6-bisphosphate is a powerful activator of yeast phosphofructokinase when assayed at pH levels of ≥7.0. Half maximal stimulation of enzyme activity occurs at 10?7 M levels of Fru 2,6-P2 concentration. This stimulating effect by Fru 2,6-P2 can be synergistic to that exerted by AMP in counteracting the inhibition of phosphofructokinase activity by ATP. The affinity (S0.5) of the yeast enzyme to fructose 6-phosphate changes from 1.5 mM in the absence of Fru 2,6-P2 to 40 μM in its presence.  相似文献

The concentration of fructose 2,6-bisphosphate found in freshly isolated erythrocytes was below the limit of detection (20 pmol/ml of packed cells). However, it increased to about 250 pmol/ml of cells when erythrocytes were incubated with glucose at pH 6.9, but not at pH 7.4 or 8.2. This could be explained by variations in the content of glycerate 2,3-bisphosphate, which was found to inhibit 6-phosphofructo-2-kinase, the enzyme responsible for fructose 2,6-bisphosphate synthesis. Glycerate 2,3-bisphosphate was also found to inhibit the potato enzyme (pyrophosphate:fructose-6-phosphate 1-phosphotransferase) used for the measurement of fructose 2,6-bisphosphate.  相似文献

不同温光条件(4d)下,以粳稻(Dryza sativaL.sp.japonica)“9516”和籼稻(O.sativaL.sp.indica)“汕优63”为材料,测定了与PSⅡ光能转化效率(Fv/Fm)和膜脂过氧化作用有关的生理指标。结果表明:适温、中等光强下两品种Fv/Fm和膜脂过氧化作用的产物(MDA)无变化,未见光抑制和光氧化表现;适温、强化下籼稻“汕优63”的Fv/Fm明显下降,MDA未见变化,有光抑制无光氧化表现;低温、强光下两品种有光抑制和光氧化表现。低温、强光下结合抑制剂实验证明,与粳稻相比,籼稻的D1蛋白量和SOD活性下降较多,叶黄素循环和非光化学猝灭(qN)受抑制程度较大,膜脂过氧化产物MDA含量较高,因而光抑制和光氧化现象较明显。实验提出:光能转化效率和膜脂过氧化表现是预测光氧化的关键指标。  相似文献

It is not known why some species accumulate high concentrations of sucrose in leaves during photosynthesis while others do not. To determine the possible basis, we have studied 10 species, known to differ in the accumulation of sucrose, in terms of activities of sucrose hydrolyzing enzymes. In general, acid invertase activity decreased as leaves expanded; however, activities remaining in mature, fully expanded leaves ranged from low (<10 micromoles per gram fresh weight per hour) to very high (>100 micromoles per gram fresh weight per hour). In contrast, sucrose synthase activities were low and relatively similar among the species (4-10 micromoles per gram fresh weight per hour). Importantly, leaf sucrose concentration, measured at midafternoon, was negatively correlated with acid invertase activity. We propose that sucrose accumulation in vacuoles of species such as soybean and tobacco is prevented by acid invertase-mediated hydrolysis. Initial attempts were made to characterize the relatively high activity of acid invertase from mature soybean leaves. Two apparent forms of the enzyme were resolved by Mono Q chromatography. The two forms had similar affinity for substrate (apparent Km [sucrose] = 3 millimolar) and did not interconvert upon rechromatography. It appeared that the loss of whole leaf invertase activity during expansion was largely the result of changes in one of the enzyme forms. Overall, the results provide a mechanism to explain why some species do not accumulate sucrose in their leaves. Some futile cycling between sucrose and hexose sugars is postulated to occur in these species, and thus, the energy cost of sucrose production may be higher than is generally thought.  相似文献

Livers of genetically obese (fa/fa) rats, starved for 24 h, contained more fructose 2,6-bisphosphate, glucose 6-phosphate, fructose 6-phosphate and glycogen, and more pyruvate kinase and phosphofructokinase 2 activities, than livers of control lean rats. These changes may be explained in terms of cyclic AMP concentration, which was decreased in livers of obese starved rats.  相似文献

为探讨红树植物光适应的生理生态策略,对6种真红树植物[无瓣海桑(Sonneratia apetala)、秋茄(Kandelia candel)、木榄(Bruguiera gymnorrhiza)、桐花树(Aegiceras corniculatum)、老鼠簕(Acanthus ilicifolius)、卤蕨(Acrostichum aureum)]和2种半红树植物[银叶树(Heritiera littoralis)、黄槿(Hibiscus tiliaceus)]的1 a生幼苗在不同生长光强(自然光强的100%、45%、30%、10%)下的光合光响应特征进行了研究。结果表明,不同生长光强对红树植物光响应特征的影响因物种而异,遮荫显著提高了秋茄和木榄的最大净光合速率(Pmax),而对其他红树植物的Pmax没有显著影响;秋茄在45%光强下具有较高的Pmax,木榄的Pmax则在45%和30%光强下显著高于其他2个处理。随着生长光强的下降,秋茄幼苗叶片的光饱和点显著上升,木榄、老鼠簕和卤蕨的光补偿点呈下降趋势,木榄和卤蕨的表观量子效率升高的同时暗呼吸速率下降。木榄、老鼠簕和卤蕨具有较强的耐荫性,...  相似文献

Epididymal bovine sperm contain fructose-1,6-bisphosphatase activity which is inhibited by AMP and by fructose 2,6-bisphosphate. Sperm phosphofructokinase displays kinetic characteristics that are typical of the F-type and it is stimulated by fructose 2,6-bisphosphate. The concentration of sperm fructose 2,6-bisphosphate remained unaffected at 1-2 microM when the glycolytic rate was either increased by glucose, caffeine or antimycin, or decreased by alpha-chlorohydrin or 6-chloro-6-deoxyglucose.  相似文献

为研究不同光强对越南抱茎茶光合作用机理的影响,设置3种处理方式,即全光照(L1)、光强度45%(L2)、光强度15%(L3),分别在遮阴3个月后测定其光合荧光参数的变化。结果表明,不同光强处理下,净光合速率均呈单峰曲线,峰值出现在11:00左右;光合速率下降主要是气孔限制因素引起的;叶绿素荧光参数表明,强光条件下电子传递受阻,热耗散增加,PSII反应中心受到破坏,从而降低了茶花的光合效率;过度遮阴条件下受到光抑制,从而导致光合机构损伤;45%光照条件下光合效率较高。相关性分析表明,净光合速率(Pn)与空气温度(Ta)、相对湿度(RH)、气孔导度(Gs)、蒸腾速率(Tr)、水分利用效率(WUE)、表观电子传递速率(ETR)呈极显著相关,与RH呈极显著负相关,与胞间CO2浓度(Ci)呈显著负相关。影响越南抱茎茶净光合速率的主要环境因子是空气温度和相对湿度。  相似文献

The aim of this work was to investigate whether sucrose synthesis in the cytosol of leaf cells is regulated in response to the supply of energy and organic carbon from the chloroplast. Fluxes into sucrose and metabolite levels in wheat (Triticum aestivum var Timmo) leaf protoplasts were compared in a range of light intensities and CO2 concentrations, showing that sucrose-phosphate synthase and the cytosolic fructose-1,6-bisphosphatase are inhibited in situ when the supply of trioseP from the chloroplasts decreases. Such a regulation might aid CO2 fixation in limiting conditions by permitting stromal metabolites to be maintained at higher levels than would otherwise be possible.  相似文献

The role of fructose 2,6 bisphosphate in partitioning of photosynthate between sucrose and starch has been studied in spinach (Spinacia oleracea U.S. hybrid 424). Spinach leaf material was pretreated to alter the sucrose content, so that the rate of starch synthesis could be varied. The level of fructose 2,6-bisphosphate and other metabolites was then related to the accumulation of sucrose and the rate of starch synthesis. The results show that fructose 2,6-bisphosphate is involved in a sequence of events which provide a fine control of sucrose synthesis so that more photosynthate is diverted into starch in conditions when sucrose has accumulated to high levels in the leaf tissue. (a) As sucrose levels in the leaf rise, there is an accumulation of triose phosphates and hexose phosphates, implying an inhibition of sucrose phosphate synthase and cytosolic fructose 1,6-bisphosphatase. (b) In these conditions, fructose 2,6-bisphosphate increases. (c) The increased fructose 2,6-bisphosphate can be accounted for by the increased fructose 6-phosphate in the leaf. (d) Fructose 2,6-bisphosphate inhibits the cytosolic fructose 1,6-bisphosphatase so more photosynthate is retained in the chloroplast, and converted to starch.  相似文献

In rabbit and sheep erythrocytes the concentrations of 2,3-bisphosphoglycerate, fructose 2,6-bisphosphate and glucose 1,6-bisphosphate suffer important changes after birth, which differ in both species. The changes of fructose 2,6-bisphosphate and glucose 1,6-bisphosphate correlate with the changes in the levels of the enzymatic activities involved in their synthesis. The change of 2,3-bisphosphoglycerate levels in rabbit but not in sheep erythrocytes could be explained by the changes of the phosphofructokinase/pyruvate kinase and 2,3-bisphosphoglycerate synthase/2,3-bisphosphoglycerate phosphatase activity ratios.  相似文献

Fructose 2,6-bisphosphate levels in freeze-clamped livers of C57BL6Jobob mice were 6-fold higher than the level in their lean (+/?) littermates. Overnight starvation reduced the hepatic level of this unique sugar diphosphate to 0.2 nmol/g in both the obese and lean mice. The elevated level in the obese mouse is consistent with the hyperinsulinemia of these animals.  相似文献

通过比较不同遮阴处理下棱角山矾幼苗生长及光合特性的差异,以探讨不同光强对棱角山矾生长及光合作用的影响及其适应机制。结果表明:棱角山矾幼苗的生物量分配对光强的反应敏感,在强光处理下,比叶面积减小,总叶面积变小;弱光处理下通过增大叶生物量比、叶面积比,支持结构生物量比,提高捕光能力。但L100、L30、L10处理下总生物量的积累显著小于L50处理,L30、L50、L100光强处理下总叶面积显著高于L10光强处理。并且10%和100%光强下棱角山矾叶片的最大净光合速率(Pmax)、光饱和点(LSP)、表观量子效率(AQY)均显著低于30%和50%光强处理;而光补偿点(LCP)则随光强的增大而升高;总叶绿素和类胡萝卜素含量在10%~100%随着光强增大而显著降低(P<0.05)。说明棱角山矾幼苗在不同光环境下具有一定适应性,但过强或过弱的光环境均会对棱角山矾的生长产生负面影响,其中50%光强是棱角山矾幼苗生长的最佳光强;棱角山矾为中性树种,苗木适宜生长在荫蔽的环境下。  相似文献

The Cyt f and P700 contents in leaves of three Sorghum, varietieswere measured, in relation to their carbon assimilation, underdifferent light intensities during growth. At the maximum irradiationused (1,800 µE m–2 s–1) the ratio of P700to Cyt f was close to unity, whereas under low irradiation (450µE m–2 s–1) the ratio of P700 to Cyt f rangedfrom two to three. A strikingly positive correlation existedbetween the P700 contents of the leaves and their rates of carbondioxide fixation, dry matter production and Cyt f contents,only when the plants were grown under high light intensities.The P700 content of the leaves in plants grown under low irradiationwas unrelated to the contents of Cyt f. Thus, at a high lightintensity there is a close relationship between the Cyt f andP700 levels, but at low intensities the amounts of electroncarriers and the reaction centre are independent.
