作为新兴生物燃料的生物柴油近年来发展迅速,以微藻为代表的第二代生物能源是解决能源危机的长远之计,但如何提高其产量仍是研究的热点问题。以提高产油自养微藻生物量和油脂含量为目的,在气升式光反应器中运用均匀设计实验方法进行了条件优化试验。分别得出了氮原子浓度、通气速率、二氧化碳体积浓度和光照强度4个因素对小球藻C2生物量积累和油脂含量影响的显著回归方程和反应器优化培养条件。以生物量为指标的优化培养条件是:氮原子浓度0.178 g/L,通气速率5 L/min,二氧化碳体积浓度3%(V/V),光照强度6000 lx。该优化条件下,生物量为2.11 g/L,即生产速率为0.352 g/(L.d),比测试实验中检测到的最高生物量[1.88 g/L,即生产速率为0.313 g/(L.d)]提高了12.2%;以油脂含量为指标的优化培养条件是:进气速率0.400 L/min,二氧化碳体积浓度1.94%(V/V),得到油脂含量为22.4%,比测试实验中检测到的最高油脂量(20.7%)提高7.7%。  相似文献

当前国内外致力于发展生物柴油,因其性能优良,成为化石柴油的替代品。由于以植物油脂生产生物柴油原料成本占总成本的70%-85%,所以亟待开发廉价油脂资源。微生物油脂主要是微生物利用碳水化合物合成的甘油脂,其脂肪酸组成和植物油相近。产油微生物具有资源丰富、油脂含量高、碳源利用谱广等特点,开发潜力大。然而,目前微生物油脂生产成本偏高,研究工作仍以富含多不饱和脂肪酸的高附加值菌油为目标。随着现代分子生物学和生物化工技术的发展,对产油微生物菌种筛选、改良、代谢调控和发酵工程的研究日趋深入,将降低微生物油脂生产成本,为未来生物柴油产业提供廉价原料。  相似文献

圆红冬孢酵母两阶段培养法生产微生物油脂   总被引:5,自引:2,他引:5  
为缩短发酵时间,减少原料消耗,采用细胞增殖和油脂积累分离的两阶段模式,培养圆红冬孢酵母Rhodosporidium toruloides AS 2.1389生产微生物油脂。结果表明,细胞增殖阶段获得的R.toruloides AS 2.1389细胞,重悬接种在葡萄糖溶液中,可快速积累油脂,菌体油脂含量超过自身干重的55%。增殖阶段细胞的菌龄越高,产油能力越强。油脂积累阶段使用高浓度葡萄糖溶液或未灭菌的葡萄糖溶液,油脂合成可以有效进行。油脂中脂肪酸成分以含16和18个碳原子的长链脂肪酸为主,可作为制备生物柴油的新型原料。  相似文献

为探索两株链带藻(Desmodesmus sp.T28-1和Desmodesmus sp.NMX451)在室外培养的最优氮源,首先在室内就不同氮源(尿素、硝酸钠、碳酸铵以及尿素和硝酸钠混合氮源)下微藻的生长和油脂积累做了研究,筛选出最优的混合氮源在室外进行了培养的可行性研究。室内研究结果表明两株链带藻在尿素下培养油脂含量最低,在铵氮下培养生物量最低。且NMX451在混合态氮下的油脂产率显著性的高于其他氮源下的油脂产率。对两株链带藻在混合氮源下的脂肪酸组分做进一步分析,结果表明油脂组分适合生物柴油生产要求,估算的生物柴油品质达到国际和国内生产标准。将两株链带藻置于室外140 L柱式反应器中用混合氮源进一步扩大培养,结果表明NMX451比T28-1的油脂含量和油脂产率高,生产成本更低,且脂肪酸组分更适宜生物柴油生产。研究表明用混合氮源在室外培养微藻是非常可行的培养方法,也说明NMX451比T28-1在生物柴油生产方面具有更好的潜力。  相似文献

为了筛选具有产油能力的微藻,从自然界水体中分离出14株微藻,根据形态特点对它们进行了初步鉴定。对其中12株微藻在自养和异养条件下的生长特性和产油性能进行了比较。通过微藻的生长曲线,生物量和油脂含量等指标,从中筛选出高产藻株并对该藻株进行了分子生物学鉴定。结果表明:藻株Y06在12种微藻中的油脂产量和产率最高,经18S rDNA鉴定确定为栅藻(Scenedesmus abundans)。藻株Y06在自养条件下的油脂产率为9.40 mg/(L.d),在异养条件下的油脂产率为201.29 mg/(L.d)。  相似文献

以新近分离的淡水绿藻--尖状栅藻(Scenedesmus acuminatus)为研究对象,将改良的BG-11培养基中的初始NaNO3浓度降低为6.0mmol/L和3.6mmol/L,利用新设计的内置拉筋平板式光生物反应器对尖状栅藻(S. acuminatus)进行大量培养。测定不同时相的生物量、总脂含量、脂组分含量及脂肪酸组成和含量,分析尖状栅藻(S. acuminatus)大量培养时的生长和油脂积累规律。当初始NaNO3浓度为6mmol/L时其最高生物量(6.27g/L)明显高于初始NaNO3浓度为3.6mmol/L时的生物量(5.30g/L);而最高的总脂含量在初始NaNO3浓度为3.6mmol/L时获得为干重的56.6%,高于初始NaNO3浓度为6mmol/L时的总脂含量(51.6%)。总脂经硅胶柱层析分级后得到三种类型的脂组分:中性脂、糖脂和磷脂,随着培养时间的延长中性脂含量逐渐增加,培养至18d后,中性脂的含量分别达到总脂的 90.9%(6 mmol/L NaNO3)和 92.0%(3.6 mmol/L NaNO3)及干重的 47.5%(6.0 mmol/L NaNO3)和 51.4%(3.6 mmol/L NaNO3)。主要脂肪酸组成为棕榈酸、棕榈油酸、硬脂酸、油酸、亚麻油酸和亚麻酸,这六种脂肪酸在不同时相的含量变化范围分别为89.92%~96.18%(占总脂肪酸)和12.5%~50.7%(占细胞干重)。总脂、中性脂及总脂肪酸单位体积产率分别为:0.18 g/L/d,0.16 g/L/d和0.15 g/L/d(6.0 mmol/L NaNO3)及0.16 g/L/d,0.15 g/L/d和0.15 g/L/d(3.6 mmol/L NaNO3)。研究结果表明,尖状栅藻(S. acuminatus)是一株易于规模化培养、脂肪酸组成适合于生物柴油生产的高产油微藻。  相似文献

研究在发酵培养基中添加色醇对斯达氏油脂酵母发酵的影响。结果表明, 在接种后0 h或12 h时添加色醇, 能够明显抑制菌体生长和油脂积累; 而在培养24 h或36 h添加, 能够明显促进菌体生长, 并增强菌体对底物利用率。与对照组比较, 在36 h添加100 mmol/L色醇, 生物量、油脂量和脂肪系数分别增加7.4%、13.9%和14.2%, 发酵时间缩短13.3%, 明显提高了油脂生产效率。气相色谱分析表明, 添加色醇对菌油脂肪酸组成及其相对含量无显著影响。实验结果有助于建立调控油脂发酵的新策略, 具有重要的理论和工程应用意义。  相似文献

从油脂植物南方红豆杉Taxusmairei茎中分离到26株内生真菌,用形态学方法鉴定表明,交链孢霉属Alternaria、无孢菌群MyceliaSterilia和硬内囊霉属Sclerocystis为优势类群;用苏丹黑染液对这26株内生真菌菌丝染色后,在光学显微镜下观察,发现其中14株内生真菌菌丝中有明显的油滴存在,选出其中油滴较大较多的7株在PDA液体培养基中培养6d后提取油脂,结果它们的油脂含量为细胞干重的13.2%~29.5%;这26株内生真菌在以微晶纤维为唯一碳源的液体培养基中培养8d后,测定发酵液的滤纸酶活,发现酶活为6.5~17.8μg葡萄糖/mL·min。研究结果表明南方红豆杉内生真菌中存在大量产油菌株,且它们有以纤维素为碳源生长的潜力。本研究为筛选能以秸秆中纤维素为碳源积累油脂的菌株打下基础。  相似文献

高、低氮浓度对2株真眼点藻的生长和油脂积累的影响   总被引:1,自引:0,他引:1  
【目的】研究氮浓度对真眼点藻纲(Eustigmatophyceae)的2株高产油微藻大真眼点藻(Eustigmatos magnus,EM)和波氏真眼点藻(Eustigmatos polyphem,EP)的细胞形态、生长、总脂含量、脂质组成和脂肪酸组成与含量的时序变化规律。【方法】利用高氮(18.0 mmol/L NO3?-N)和低氮(3.6 mmol/L NO3?-N)浓度培养微藻。【结果】形态观察结果表明,大真眼点藻(E. magnus)和波氏真眼点藻(E. polyphem)营养细胞具有1个周生的裂叶状叶绿体,细胞质中有液泡,内含能够振动的颗粒物,以及一个较为明显的红色色素体;生殖方式通过形成2个D形或4个四角形的似亲孢子;随着培养周期的延伸和营养盐的消耗,细胞中油体逐步形成,其数量不断增加,体积不断增大。实验结果表明,初始氮浓度对2种微藻的总脂积累及生长均有显著影响(P<0.05),低氮浓度下2种微藻的生物质浓度分别为9.0 g/L和8.5 g/L,均低于高氮浓度下的生物质浓度。而低氮浓度下2种微藻的总脂、中性脂和总脂肪酸的含量以及总脂、中性脂与总脂肪酸的单位体积产率均明显高于高氮浓度组,其最高值分别为:59.10%、51.90%、46.95%和0.28、0.24、0.22 g/(L·d) (EM);64.20%、56.80%、50.01%和0.32、0.28、0.25 g/(L·d) (EP)。脂肪酸分析结果表明,两种微藻的脂肪酸主要成分均为棕榈酸(C16:0)、棕榈油酸(C16:1)、油酸(C18:1)和二十碳五烯酸(C20:5,EPA),四者的总含量(占总脂肪酸)分别达到85.83%和85.48%,其中棕榈油酸的含量最高。【结论】低氮浓度胁迫有利于大真眼点藻和波氏真眼点藻细胞内油脂的积累,两种微藻均为适合于生产生物柴油的油脂生产藻株。  相似文献

The new amylolytic oleaginous red yeast, Sporidiobolus pararoseus KX709872, produced both α-amylase (540?±?0.09?mU/mL) and amyloglucosidase (23?±?0.00?mU/mL) and showed good ability to directly convert rice residue from canteen waste to biomass and lipids. Effects of medium composition and cultivation conditions on growth and lipid accumulation for strain KX709872 were investigated under shaking flask and upscaling levels. At C?:?N ratio of 25?:?1, pH 5.45, 22.36°C, and 199.40?rpm for 7 days, volumetric production of biomass and lipids, lipid content, and lipid productivity reached 17.69?±?0.44, 8.35?±?0.19?g/L, 49.48?±?0.41% (w/w), and 1.67?±?0.11?g/L/day, respectively. Production of lipids was also implemented in 5.0-L stirred tank bioreactor with 2.5?L of optimized medium at 300?rpm and 3.0 vvm for 5 days. Volumetric production of biomass and lipids, lipid content, and lipid productivity were 16.33?±?0.49, 8.75?±?0.13?g/L, 56.61?±?0.04% (w/w), and 2.19?±?0.03?g/L/day, respectively. Meanwhile, the fatty acids of lipids from strain KX709872 had high oleic acid content (60?62%) which was similar to those of vegetable oils, indicating that these lipids are promising as an alternative biodiesel feedstock. Moreover, the biodiesel derived from lipids of strain KX709872 had properties satisfying the criteria of ASTM D6751 and EN 14214 standards.  相似文献

本研究主要探讨深黄被孢霉M2菌株对生物质全糖的利用,考察其碳源同化能力、不同碳源下产脂情况以及对玉米皮渣的利用能力。研究结果表明,M2菌株能够利用葡萄糖、木糖、阿拉伯糖和甘露糖进行生长和油脂积累。M2菌株以6%糖浓度的玉米皮渣水解液为底物发酵培养,油脂微生物生物量达18.2g/L,干菌体油脂含量45.7%,单位体积发酵液油脂产量为8.3g/L。  相似文献

Microbial lipids have the potential to displace terrestrial oils for fuel, value chemical, and food production, curbing the growth in tropical oil plantations and helping to reduce deforestation. However, commercialization remains elusive partly due to the lack of suitably robust organisms and their low lipid productivity. Extremely high cell densities in oleaginous cultures are needed to increase reaction rates, reduce reactor volume, and facilitate downstream processing. In this investigation, the oleaginous yeast Metschnikowia pulcherrima, a known antimicrobial producer, was cultured using four different processing strategies to achieve high cell densities and gain suitable lipid productivity. In batch mode, the yeast demonstrated lipid contents more than 40% (w/w) under high osmotic pressure. In fed-batch mode, however, high-lipid titers were prevented through inhibition above 70.0 g L−1 yeast biomass. Highly promising were a semi-continuous and continuous mode with cell recycle where cell densities of up to 122.6 g L−1 and maximum lipid production rates of 0.37 g L−1 h−1 (daily average), a nearly two-fold increase from the batch, were achieved. The findings demonstrate the importance of considering multiple fermentation modes to achieve high-density oleaginous yeast cultures generally and indicate the limitations of processing these organisms under the extreme conditions necessary for economic lipid production.  相似文献

Summary Differentiation in keratinocytes can be experimentally modulated by changing the culture conditions. When cultured under conventional, submerged conditions, the extent of cellular differentiation is reduced in the presence of low calcium medium and is enhanced in medium containing physiologic calcium concentrations. Moreover cultures grown at the air-medium interface or on a dermal substrate, or both, differentiate even further. Herein we report the effect of culture conditions on lipid composition in normal human keratinocytes and three squamous carcinoma cell (SCC) lines that vary in their capacity to differentiate as assessed by cornified envelope formation. Under submerged conditions, the total phospholipid content was lower, triglyceride content higher, and phospholipid: neutral lipid ratio lower in direct correlation to the degree of differentiation in these cultures. When grown at the air-medium interface on the-epidermized dermis, evidence of further morphologic differentiation was found only for well-differentiated SCC cells and normal keratinocytes. Similarly, the phospholipid content remained high in poorly differentiated SCC cells and it, decreased modestly in well-differentiated SCC cells and markedly in normal keratinocytes. In all cell lines the triglyceride content was increased and cholesterol content decreased when compared to parallel submerged cultures, but these differences were most pronounced in well-differentiated cell lines. Acylceramides and acylglucosylceramides were found only in normal keratinocytes and only under the most differentiation-enhancing conditions. These studies demonstrate differentiation-related changes in the lipid content of both normal and neoplastic keratinocytes. 目的用斯达油脂酵母(Lipomyces starkeyi)作为发酵菌株,以纯木糖溶液为油脂发酵原料,对L.starkeyi利用木糖积累油脂进行系统研究。方法 L.starkeyi于斜面培养基中活化后,接种于YPD液体培养基,于30℃、200 r/min摇床培养。在摇瓶中培养一段时间后,测定发酵液细胞浓度,离心发酵液收集细胞。将离心后得到的菌体加入木糖溶液重悬,并转接于含50 mL木糖溶液的250 mL摇瓶中进行发酵生产。结果相比一阶段法,两阶段发酵方法可以在更短的时间内达到较高的油脂含量,油脂含量能够达到细胞自身干重的60%以上。实验发现高菌龄酵母产油速度更快;并且初始木糖浓度高达120 g/L时,酵母细胞仍然能够高效合成油脂。结论 L.starkeyi能够有效利用木糖进行发酵产生油脂,是以木质纤维素为原料生产微生物油脂的优良菌种。  相似文献

The effects of salinity, growth temperature, pH and composition of the medium on the accumulation of intracellular organic solutes, by nuclear magnetic resonance spectroscopy (NMR) in Halomonas pantelleriense were examined. The modulation of lipid pattern in different growth conditions was also reported. H. pantelleriense accumulated glycine betaine, ectoine, hydroxyectoine and glutamate. The type of osmoprotectant and the relative proportion depended on growth conditions. The main lipids identified by NMR studies were 1,2 diacylglycero-3-phosphorylethanolamine (PEA), 1,2 diacylglycero-3-phosphoryl-glycerol (PG) and cardiolipin, (DPG). The predominant fatty acids were C16:0 and C18:1, minor fatty acids were C16:1 and C18:0. The relative percentage of polar lipids and fatty acids were affected by growth conditions.  相似文献
