Comparative studies were made of the activities of Cu,Zn-superoxide dismutase, Mn-superoxide dismutase, catalase and glutathione peroxidase in organ homogenates from two herbivorous (grass carp and silver carp) and three omnivorous fish (barbel, crucian carp and common carp). The protein contents and lipid peroxidation of the organ homogenates were also compared. These comparative measurements provided control values for subsequent toxicological examinations. For the herbivorous fish, the highest total superoxide dismutase activity was observed in the kidney, followed in turn by the liver or spleen. In contrast, the highest total superoxide dismutase activity in the barbel was found in the roe, in the crucian carp in the liver, and in the common carp in the liver too. The quantitative distributions of the two peroxide metabolism enzymes glutathione peroxidase and catalase in the organ homogenates present very varied pictures. The results of the lipid peroxidation are similarly difficult to fit into one framework, even for fish following the same diet. It appears that the enzyme activities mentioned in points 4 and 5, together with the lipid peroxidation, depend not on the species, but on the variety.  相似文献

1. Superoxide dismutase (SOD), catalase (C), peroxidase (P) and glutathione peroxidase (GSH-Px) were estimated in the erythrocytes and liver of the carp, the tench and the crucian carp during a two-years period, 1983-1984 (autumn). 2. The lipid peroxidation (LP) of the erythrocytes and liver homogenates was also compared. 3. The activities of the enzymes examined showed significant interspecies variations. 4. Significant differences were observed between enzyme activities (except for SOD in the liver) and lipid peroxidation in erythrocytes and liver of investigated fish species. 5. The very high values of peroxide enzyme activities in fish erythrocytes and SOD in fish liver suggest the predominant role of these enzymes in protection of polyunsaturated acids against uncontrolled oxidative processes.  相似文献

The effects of paraquat on the superoxide dismutase, catalase, glutathionine peroxidase activities and lipid peroxidation at different times of paraquat exposure of Cyprinus carpio morph L. erythrocytes were studied. Typical characteristics were observed in the changes of the enzyme activities of the erythrocytes after exposure to paraquat. The haemoglobin concentration of common carp haemolysates was decreased by exposure to paraquat.  相似文献

Superoxide dismutase, catalase and peroxidase activities were estimated in erythrocytes of the carp, the tench and the crucian carp. The enzymes showed higher activities in spring (1982) than in autumn (1981). The content of lipid peroxidation products--diene conjugates, lipid hydroperoxides, thiobarbituric acid reactive substances (TBARS), vitamins A, E and carotenoids and the activity of antioxidant enzymes--superoxide dismutase, glutathione peroxidase and catalase in the liver of freshwater fishes of different species (silver carp, grass carp and common carp) in different seasons have been studied. It was established the activity of antioxidant defence system in the liver of fish depends significantly on the season and fish species. In particular, the content of lipid peroxidation products in the liver of freshwater fishes at the beginning of winter and spring was significantly higher compared to their content at the beginning of summer and autumn. The superoxide dismutase and glutathione peroxidase activities in the liver of these fish species at the beginning of winter and spring were significantly lower than at the beginning of summer and autumn while the seasonal changes of catalase activity in the liver of fish are expressed insignificantly. The content of vitamins E, A1, A2 and carotenoids in the liver of fishes of different species at the beginning of winter and spring was significantly lower than at the beginning of summer and autumn. The content of lipid peroxidation products and vitamins E, A1 and A2 in the liver of common carp is significantly lower than in the liver of silver carp and grass carp and species differences in antioxidant enzymes activity are insignificant.  相似文献

Contaminant-related changes in antioxidative processes in the freshwater crustacea Daphnia magna exposed to model redox cycling contaminant were assessed. Activities of key antioxidant enzymes including catalase, superoxide dismutase, glutathione peroxidase and glutathione S-transferases and levels of lipid peroxidation measured as thiobarbituric acid-reactive substances (TBARS) and lipofucsin pigment content were determined in D. magna juveniles after being exposed to sublethal levels of menadione, paraquat, endosulfan, cadmium and copper for 48 h. Results denoted different patterns of antioxidant enzyme responses, suggesting that different toxicants may induce different antioxidant/prooxidant responses depending on their ability to produce reactive oxygen species and antioxidant enzymes to detoxify them. Low responses of antioxidant enzyme activities for menadione and endosulfan, associated with increasing levels of lipid peroxidation and enhanced levels of antioxidant enzyme activities for paraquat, seemed to prevent lipid peroxidation, whereas high levels of both antioxidant enzyme activities and lipid peroxidation were found for copper. For cadmium, low antioxidant enzyme responses coupled with negligible increases in lipid peroxidation indicated low potential for cadmium to alter the antioxidant/prooxidant status in Daphnia. Among the studied enzymes, total glutathione peroxidase, catalase and glutathione S-transferase appeared to be the most responsive biomarkers of oxidative stress.  相似文献

The effects of paraquat, the active ingredient of Gramoxone, were studied on frogs (Rana esculenta) kept at 4 degrees C or 20 degrees C, to establish its effects on the survival, the antioxidant enzyme system and the lipid peroxidation of the tissues (liver and lung). The lower temperature was found to increase the survival. The activities of the antioxidant enzymes were decreased or not influenced by the LD50 of paraquat at the lower temperature, whereas the LD100 often resulted in a significant enzymatic activity increase. It was an important finding that the lipid peroxidation of the lipid decreased in response to paraquat at 4 degrees C, but increased at 20 degrees C. The lipid peroxidation of the lung increased at both temperatures.  相似文献

Phylo- and ontogenetic aspects of lipid peroxidation and antioxidative enzyme system in the retina of vertebrates were studied. It was established that both the intensity of lipid peroxidation and the activity of glutathione peroxidase in the retina of different vertebrate animals (carp, frog, tortoise, pigeon, rabbit) considerably diminished with evolution. The differences in the intensity of lipid peroxidation and the activity of glutathione peroxidase between dark- and light-adapted retinas also decreased depending on the level of the development. The activity of glutathione peroxidase in the retina of chick embryos was found only at the end of the incubation period.  相似文献

采用聚丙烯酰胺凝胶垂直板电泳技术,对彭泽鲫、银鲫D系以及野鲫三种鲫鱼品系的心、肝和肾脏组织的乳酸脱氢酶(LDH)、苹果酸脱氢酶(MDH)、苹果酸酶(ME)、酯酶(EST)和超氧化物歧化酶(SOD)同工酶表型进行了比较研究.结果表明彭泽鲫乳酸脱氢酶同工酶比银鲫D系在肝组织多出二条酶带(LDH7'和LDH8');超氧化物歧化酶在心和肾组织中分别多出一条酶带(SOD12'),表明彭泽鲫和银鲫已在生化水平产生明显的分化,推测它们可能起源于不同的地区,由不同的祖先,独立演化而形成.此外,彭泽鲫和银鲫D系的同工酶电泳图谱都包含野鲫的基本酶带,而彭泽鲫和野鲫的酯酶同工酶电泳图谱尤为相似,推测彭泽鲫和银鲫可能起源于野鲫,而彭泽鲫和野鲫的关系较近,银鲫和野鲫的关系较远.  相似文献

设计了5组等氮(粗蛋白约38%)的饲料, 饲料中小球藻的添加量分别为0(对照)、17%、34%、51%和68%, 替代0(对照组)、21.8%、43.6%、65.5%和87.3%的鱼粉, 探讨小球藻替代鱼粉对初始体重为(5.540.08) g的鲫幼鱼生长、体组成、肝脏组织学及脂肪代谢相关酶活性的影响, 实验期为8周。实验结果表明: 随着小球藻替代鱼粉水平的增加, 鲫的增重率呈现先增加后下降的趋势; 与此相反, 饲料系数呈现先下降后增加的趋势。随着小球藻替代比例的增加, 肝体比和脏体比有增大的趋势, 而各个处理组肠脂比差异不显著。小球藻替代鱼粉使鲫肌肉和肝脏的蛋白含量降低, 而肝脏的脂肪含量随着替代比例的增加先上升后下降。当饲料中小球藻替代鱼粉的水平从0增加到65.5%, 肝脏生脂酶如G6PD、ME和FAS酶的活性显著增加, 而当饲料小球藻替代水平更进一步增加到87.3%, 肝脏生脂酶如G6PD、ME和FAS酶的活性下降。各个处理组6PGD酶活性没有显著差异。小球藻替代鱼粉对鲫的肝脏组织结构产生不利影响, 替代组的肝细胞体积有所增大, 部分肝细胞出现细胞核溶解、核消失、肝细胞坏死。以增重率和饲料系数为因变量进行二次线性回归分析, 表明小球藻对鱼粉的最适替代比例分别为47.14%和49.88%。    相似文献

1. Studies were performed regarding the effects of CuSO4 in concentrations of 5, 10, 25 and 50 ppm and ZnSO4 in concentrations of 10 and 100 ppm on the antioxidant enzyme activities, lipid peroxidation and protein contents of tissues of common carp (Cyprinus carpio morpha L.) exposed to these pollutants for 24 hr. 2. The results demonstrated that CuSO4 was more toxic than ZnSO4 and that both treatments brought about significant changes in these parameters in carp hepatopancreas (liver), gill and white muscle. 3. An increase of the CuSO4 concentration led to significant decreases in the antioxidant enzyme activities, except that of glutathione peroxidase, which was increased significantly, and significant increases in the lipid peroxidation and protein contents. 4. An increase of the ZnSO4 concentration led to slight changes in the antioxidant enzyme activities, lipid peroxidation and protein contents of carp tissues.  相似文献

Analyses were made of the phsopholipid fatty acids and the antioxidant enzymes in the carp (Cyprinus carpio morpha) at three different oxygen concentrations, corresponding to hyperoxia, hypoxia and anoxia. Variations of the oxygen concentration were found to influence the quantities of phsopholipid fatty acids, as well as the antioxidant enzyme activities. In hyperoxia and hypoxia the amount of polyunsaturated fatty acids in carp liver was higher than in anoxia, but in other tissues there was no significant differences. As to the antioxidant enzyme system, the glutathione peroxidase activity and the lipid peroxidation value increased significantly with decrease of the oxygen concentration, while the total superoxide dismutase activity decreased on lowering of the oxygen level.  相似文献

The mechanism of Cd2+ on the DNA cleavage and Ce3+ on the DNA repair in the kidney of silver crucian carp (Carassius auratus gibelio) is investigated by agarose gel electrophoresis methods and assaying biochemical indexes. It proves that Cd2+ induces the classical laddering degradation of DNA in vivo, but DNA cleavage is repaired after injecting with a low Ce3+ concentration under various Cd2+ concentrations. The DNA cleavage caused by Cd2+ is the result of the activation of deoxyribonuclease (DNase) and accumulation of reactive oxygen species (ROS), and Cd2+ destroys the antioxidant system, which diminishes the activities of superoxide dismutase, catalase, and peroxidase, and the increase of the lipid peroxidation (LPO) level. However, Ce3+ could inhibit activation of Cd2+ on DNase activity, relieve inhibition of Cd2+ on activities of the antioxidant enzyme, and diminish ROS accumulation. The results show that Ce3+ could relieve the toxicity of Cd2+ to silver crucian carp.  相似文献

为研究饲料脂肪水平对芙蓉鲤鲫(Furong crucian carp)幼鱼[初始体重, (2.040.01) g]血清生化指标、免疫指标及抗氧化能力的影响, 以鱼油为主要脂肪源, 配制五种脂肪水平分别为2.50%、4.39%、6.61%、8.42%和10.81%的等氮实验饲料。实验在室内循环水养殖系统中进行, 每种饲料3个重复, 每个重复随机放养30尾芙蓉鲤鲫, 养殖周期为60d。结果显示: 随着饲料中脂肪水平的升高, 芙蓉鲤鲫增重率和蛋白质效率呈现先显著升高后显著降低的趋势(P0.05), 而饲料系数呈现相反的趋势。饲料脂肪水平升高显著提高了芙蓉鲤鲫血清中总胆固醇(CHO)和球蛋白(GLB)含量, 而显著降低总蛋白(TP)和白蛋白(ALB)含量。饲料脂肪水平未对鱼体血清高密度脂蛋白胆固醇(HDL-C)和甘油三酯(TG)含量造成显著影响(P0.05)。芙蓉鲤鲫血清免疫球蛋白M (IgM)含量、血清和肝胰脏超氧化物歧化酶(SOD)活力、肝胰脏总抗氧化能力(T-AOC)随脂肪水平增加先升高后下降(P0.05)。血清中过氧化氢酶(CAT)活力10.81%组显著高于其他各组(P0.05)。肝胰脏CAT和丙二醛(MDA)活力在各处理组组间差异不显著(P0.05)。研究结果表明, 在饲料中适宜的脂肪添加有助于改善芙蓉鲤鲫健康状况, 但过高的脂肪水平加重了芙蓉鲤鲫的代谢压力和氧化速率。增重率与饲料脂肪水平作二次回归分析显示, 芙蓉鲤鲫获得最大增长所需饲料脂肪最佳水平为6.94%。  相似文献

Glutathione peroxidase (GSHPx), a seleno-enzyme, reduces lipid hydroperoxides while producing oxidized glutathione (GSSG), which can efflux from cells. To study the role of GSHPx in antioxidant defense, isolated lungs from selenium-deficient rats were perfused for 2 h with or without 1 mM paraquat. Perfusate GSSG was measured as an index of GSHPx activity, and malondialdehyde (MDA) as an index of lipid peroxidation. Selenium deficiency decreased lung GSHPx activity 75-80%. During perfusion control lungs showed GSSG efflux of 8.5 +/- 4.5 nmol/h and with paraquat 49.1 +/- 12.1 nmol/h. Selenium-deficient lungs with or without paraquat showed GSSG efflux of 16.4 +/- 5.3 and 13.7 +/- 8.9 nmol/h, respectively. MDA efflux occurred only in paraquat-perfused selenium-deficient lungs (7.8 +/- 2.7 nmol/h). Lung homogenates from this group had lower GSH + GSSG than the other three groups. These results indicate an inverse correlation between GSSG efflux and MDA accumulation from paraquat-perfused lungs and suggest that increased turnover of the GSHPx reaction protects paraquat-perfused lungs from lipid peroxidation.  相似文献

Leaves of 7- and 18-day-old plants of two maize strains, one resistant (LIZA) and one sensitive (LG11) to water stress, were floated in 1 m M paraquat and 1 m M H2O2 for 12 h in light and in darkness. The aim of this work was to analyse the effects of these substances on the activities of enzymes involved in the scavenging of active oxygen species during senescence. Three senescence parameters; chlorophyll loss, lipid peroxidation and conductivity; showed a general cell damage caused by both oxidative treatments and revealed a higher tolerance of LIZA than LG11 to paraquat and H2O2 both in light and in darkness. Activities of antioxidative enzymes increased by the effect of oxidative treatments in young and senescent leaves of the drought-resistant maize strain LIZA. These increases were about 3-to 6-fold in glutathione reductase. 3-to 4-fold in superoxide dismutase and 2-fold in ascorbate peroxidase activities. The possible correlation between water stress resistance. senescence and the potential of antioxidant enzymes was analysed.  相似文献

The in vivo effect of the known herbicide, paraquat, on both hepatic oxidative stress and heme metabolism was studied. A marked increase in lipid peroxidation and a decrease in reduced glutathione (GSH) content were observed 1 h after paraquat administration. The activity of liver antioxidant enzymes, superoxide dismutase, catalase and glutathione peroxidase was decreased 3 h after paraquat injection. Heme oxygenase-1 induction started 9 h after treatment, peaking at 15 h. delta-aminolevulinic acid synthase induction occurred once heme oxygenase had been enhanced, reaching its maximum (1.5-fold of control) at 16 h. delta-aminolevulinic acid dehydratase activity was 40% inhibited at 3 h showing a profile similar to that of GSH, while porphobilinogenase activity was not modified along the whole period of the assay. Administration of alpha-tocopherol (35 mmol/kg body weight) 2 h before paraquat treatment entirely prevented the increase in thiobarbituric acid reactive substances (TBARS) content, the decrease in GSH levels as well as heme oxygenase-1 and delta-aminolevulinic acid synthase induction. This study shows that oxidative stress produced by paraquat leads to an increase in delta-aminolevulinic acid synthase and heme oxygenase-1 activities, indicating that the herbicide affects both heme biosynthesis and degradation.  相似文献

采用质粒克隆测序方法,获得了异源四倍体鲫鲤5个个体、异源四倍体鲫鲤雌核发育二倍体后代2个个体、三倍体湘云鲫2个个体及红鲫、湘江野鲤和日本白鲫各1个个体的线粒体DNA 12S rRNA基因的全序列。经对比发现,异源四倍体5个个体共享2种单元型,异源四倍体鲫鲤雌核发育二倍体后代2个个体、三倍体湘云鲫2个个体以及红鲫、湘江野鲤和日本白鲫各1个个体分别共享1种单元型。用MEGA 1.0 软件分析了它们的碱基组成和核苷酸序列差异,用邻接法构建系统进化树。它们间的序列同源性在95%~99%之间,异源四倍体鲫鲤、三倍体湘云鲫和它们母本(分别为红鲫和日本白鲫)之间的序列同源性大于异源四倍体鲫鲤、三倍体湘云鲫和它们父本(分别为湘江野鲤和异源四倍体鲫鲤)之间的序列同源性,结果表明:异源四倍体鲫鲤和三倍体湘云鲫在线粒体DNA 12S rRNA基因上具有母性遗传特征。本研究另一值得注意地方的是异源四倍体鲫鲤经过9代(F3-F11)繁殖后,在5个个体中发现了2种单元型,说明在四倍体基因库中存在遗传多样性,为四倍体基因库的繁殖、保护和种群复壮提供了一些有价值的信息。  相似文献

The effects of garlic and neem leaf extracts on lipid peroxidation and antioxidant status during administration of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), a carcinogenic nitrosamine were evaluated in male Wistar rats. Extracts of garlic and neem leaf were administered orally for five consecutive days before intraperitoneal injection of MNNG. Enhanced lipid peroxidation in the stomach, liver and circulation of MNNG-treated rats was accompanied by a significant decrease in glutathione (GSH) and the activities of glutathione peroxidase (GPx), glutathione-S-transferase (GST) and gamma glutamyl transpeptidase (GGT). Administration of garlic and neem leaf extracts significantly decreased the formation of lipid peroxides and enhanced the levels of antioxidants and detoxifying enzymes in stomach, the primary target organ for MNNG, as well as in the liver and circulation. The results of the present study suggest that garlic and neem may exert their protective effects by modulating lipid peroxidation and enhancing the levels of GSH and GSH-dependent enzymes.  相似文献
