The primary objective of this study was to better understand how root morphological alteration stimulates N uptake in maize plants after root growth restriction, by investigating the changes in length and number of lateral roots, 15NO3 influx, the expression level of the low-affinity Nitrate transporter ZmNrt1.1, and proteomic composition of primary roots. Maize seedlings were hydroponically cultured with three different types of root systems: an intact root system, embryonic roots only, or primary roots only. In spite of sufficient N supply, root growth restriction stimulated compensatory growth of remaining roots, as indicated by the increased lateral root number and root density. On the other hand, there was no significant difference in 15NO3 influx between control and primary root plants; neither in ZmNrt1.1 expression levels in primary roots of different treatments. Our data suggested that increased N uptake by maize seedlings experiencing root growth restriction is attributed to root morphological adaptation, rather than explained by the variation in N uptake activity. Eight proteins were differentially accumulated in embryonic and primary root plants compared to control plants. These differentially accumulated proteins were closely related to signal transduction and increased root growth.  相似文献

Inter-strain variability in pH compensation point (pHc) in the cyanobacterium Microcystis aeruginosa has been investigated. The pHc allows one to discriminate whether the organism is able to take up HCO3 as inorganic carbon (Ci) source in photosynthesis. Eight subgroups were found according to the pHc value, ranging from 10.44 ± 0.22 to 11.67 ± 0.05. The high variability in pHc (and consequently, in the capacity to use HCO3 as Ci source) suggested that different HCO3 use mechanisms could occur in M. aeruginosa and, from an evolutionary point of view, this trait is not under high natural selective pressure.  相似文献

The ability to cope with NH4+-N was studied in the littoral helophytes Phragmites australis and Glyceria maxima, species commonly occupying fertile habitats rich in NH4+ and often used in artificial wetlands. In the present study, Glyceria growth rate was reduced by 16% at 179 μM NH4+-N, and the biomass production was reduced by 47% at 3700 μM NH4+-N compared to NO3-N. Similar responses were not found in Phragmites. The amounts (mg g−1 dry wt) of starch and total non-structural carbohydrates (TNC) in rhizomes were significantly lower in NH4+ (8.9; 12.2 starch; 20.1; 41.9 TNC) compared to NO3 treated plants (28.0; 15.6 starch; 58.5; 56.3 TNC) in Phragmites and Glyceria, respectively. In addition, Glyceria showed lower amounts (mg g−1 dry wt) of soluble sugars, TNC, K+, and Mg2+ in roots under NH4+ (5.6; 14.3; 20.6; 1.9) compared to NO3 nutrition (11.6; 19.9; 37.9; 2.9, for soluble sugars, TNC, K+, and Mg2+, respectively), while root internal levels of NH4+ and Ca2+ (0.29; 4.6 mg g−1 dry wt, mean of both treatments) were only slightly affected. In Phragmites, no changes in soluble sugars, TNC, Ca2+, K+, and Mg2+ contents of roots (7.3; 14.9; 5.1; 17.3; 2.6 mg g−1 dry wt, means of both treatments) were found in response to treatments. The results, therefore, indicate a more pronounced tolerance towards high NH4+ supply in Phragmites compared to Glyceria, although the former may be susceptible to starch exhaustion in NH4+-N nutrition. In contrast, Glyceria's ability to colonize fertile habitats rich in NH4+ is probably related to the avoidance strategy due to shallow rooting or to the previously described ability to cope with high NH4+ levels when P availability is high and NO3 is also provided.  相似文献

IRBIT (also called AHCYL1) was originally identified as a binding protein of the intracellular Ca2 + channel inositol 1,4,5-trisphosphate (IP3) receptor and functions as an inhibitory regulator of this receptor. Unexpectedly, many functions have subsequently been identified for IRBIT including the activation of multiple ion channels and ion transporters, such as the Na+/HCO3 co-transporter NBCe1-B, the Na+/H+ exchanger NHE3, the Cl channel cystic fibrosis transmembrane conductance regulator (CFTR), and the Cl/HCO3 exchanger Slc26a6. The characteristic serine-rich region in IRBIT plays a critical role in the functions of this protein. In this review, we describe the evolution, domain structure, expression pattern, and physiological roles of IRBIT and discuss the potential molecular mechanisms underlying the coordinated regulation of these diverse ion channels/transporters through IRBIT. Lupins appear to be more sensitive than peas to Fe deficiency. However, when grown in nutrient solutions between pH 5–6, little difference existed between them in their ability to acidify the solution or to release FeIII reducing compounds. This experiment was aimed at determining whether differences between species which occurred when Fe deficiency was induced by withholding Fe from an acid solution, are maintained when Fe deficiency is induced by addition of HCO3 -. Lupins and peas were grown in nutrient solutions at 0, 2 and 6 μM of FeIII EDDHA and either with or without HCO3 - (6 mM). Bicarbonate induced symptoms of Fe deficiency (chlorosis) in both lupins and peas, and markedly decreased the growth of shoots. Symptoms appeared sooner and were more severe in lupins than in peas. Growing plants without HCO3 -, but at the lowest Fe level, decreased the growth and Fe concentration of shoots of lupins but did not induce chlorosis. Growing peas in this treatment, decreased Fe concentrations, but to a lesser extent than in lupins, and did not decrease growth. H+-ion extrusion and release of FeIII reducing compounds was greater in lupins than in peas. Bicarbonate also decreased the growth of roots of lupins but increased the growth of roots of peas. Results indicate that when Fe deficiency is induced by HCO3 -, then the response of lupins and peas are similar to their response in acid solution culture. Differences between species therefore could not be explained by their relative abilities to acidify or release FeIII reducing compounds. Greater control of the distribution of Fe within the shoots, the presence of a pool of Fe within the roots, a lower threshold for Fe uptake, or a higher content of seed-Fe, may therefore be the reason for the lower sensitivity of peas than lupins to Fe deficiency.  相似文献

The role of carboxylic residues at the high-affinity, Mn-binding site in the ligation of iron cations blocking the site [Biochemistry 41 (2000) 5854] was studied, using a method developed to extract the iron cations blocking the site. We found that specifically bound Fe(III) cations can be extracted with citrate buffer at pH 3.0. Furthermore, citrate can also prevent the photooxidation of Fe(II) cations by YZ. Participation of a COOH group(s) in the ligation of Fe(III) at the high-affinity site was investigated using 1-ethyl-3-[(3-dimethylamino)propyl] carbodiimide (EDC), a chemical modifier of carboxylic amino acid residues. Modification of the COOH groups inhibits the light-induced oxidation of exogenous Mn(II) cations by Mn-depleted photosystem II (PSII[−Mn]) membranes. The rate of Mn(II) oxidation saturates at ≥10 μM in PSII(−Mn) membranes and ≥500 μM in EDC-treated PSII (−Mn) samples. Intact PSII(−Mn) membranes have only one site for Mn(II) oxidation via YZ (dissociation constant, Kd = 0.64 μM), while EDC-treated PSII(−Mn) samples have two sites (Kd = 1.52 and 22 μM; the latter is the low-affinity site). When PSII(−Mn) membranes were incubated with Fe(II) before modifier treatment (to block the high-affinity site) and the blocking iron cations were extracted with citrate (pH 3.0) after modification, the membranes contained only one site (Kd = 2.3 μM) for exogenous Mn(II) oxidation by YZ radical. In this case, the rate of electron donation via YZ saturated at a Mn(II) concentration ≥15 μM. These results indicate that the carboxylic residue participating in Mn(II) coordination and the binding of oxidized manganese cations at the HAZ site is protected from the action of the modifier by the iron cations blocking the HAZ site. We concluded that the carboxylic residue (D1 Asp-170) participating in the coordination of the manganese cation at the HAZ site (Mn4 in the tetranuclear manganese cluster [Science 303 (2004) 1831]) is also involved in the ligation of the Fe cation(s) blocking the high-affinity Mn-binding site.  相似文献

The luminostat regime has been proposed as a way to maximize light absorption and thus to increase the microalgae photosynthetic efficiency within photobioreactors. In this study, simulated outdoor light conditions were applied to a lab-scale photobioreactor in order to evaluate the luminostat control under varying light conditions. The photon flux density leaving the reactor (PFDout) was varied from 4 to 20 μmol photons m−2 s−1and the productivity and photosynthetic efficiency of Chlorella sorokiniana were assessed.Maximal volumetric productivity (1.22 g kg−1 d−1) and biomass yield on PAR photons (400-700 nm) absorbed (1.27 g mol−1) were found when PFDout was maintained between 4 and 6 μmol photons m−2 s−1. The resultant photosynthetic efficiency was comparable to that already reported in a chemostat-controlled reactor. A strict luminostat regime could not be maintained under varying light conditions. Further modifications to the luminostat control are required before application under outdoor conditions.  相似文献

Background

Identifying QTLs/genes for iron and zinc in rice grains can help in biofortification programs. Genome wide mapping showed 14 QTLs for iron and zinc concentration in unpolished rice grains of F7 RILs derived from Madhukar × Swarna. One line (HL) with high Fe and Zn and one line (LL) with low Fe and Zn in unpolished rice were compared with each other for gene expression using qPCR. 7 day old seedlings were grown in Fe + and Fe − medium for 10 days and RNA extracted from roots and shoots to determine the response of 15 genes in Fe − conditions.

Results

HL showed higher upregulation than LL in shoots but LL showed higher upregulation than HL in roots. YSL2 was upregulated only in HL roots and YSL15 only in HL shoots and both up to 60 fold under Fe − condition. IRT2 and DMAS1 were upregulated 100 fold and NAS2 1000 fold in HL shoot. NAS2, IRT1, IRT2 and DMAS1 were upregulated 40 to 100 fold in LL roots. OsZIP8, OsNAS3, OsYSL1 and OsNRAMP1 which underlie major Fe QTL showed clear allelic differences between HL and LL for markers flanking QTL. The presence of iron increasing QTL allele in HL was clearly correlated with high expression of the underlying gene. OsZIP8 and OsNAS3 which were within major QTL with increasing effect from Madhukar were 8 fold and 4 fold more expressed in HL shoot than in LL shoot. OsNAS1, OsNAS2, OsNAS3, OsYSL2 and OsYSL15 showed 1.5 to 2.5 fold upregulation in flag leaf of HL when compared with flag leaf of Swarna.

Conclusion

HL and LL differed in root length, Fe concentration and expression of several genes under Fe deficiency. The major distinguishing genes were NAS2, IRT2, DMAS1, and YSL15 in shoot and NAS2, IRT1, IRT2, YSL2, and ZIP8 in roots. The presence of iron increasing QTL allele in HL at marker locus close to genes also increased upregulation in HL.  相似文献

Iron is an important nutrient in N2-fixing legume nodules. The demand for this micronutrient increases during the symbiosis establishment, where the metal is utilized for the synthesis of various iron-containing proteins in both the plant and the bacteroid. Unfortunately, in spite of its importance, iron is poorly available to plant uptake since its solubility is very low when in its oxidized form Fe(III). In the present study, the effect of iron deficiency on the activity of some proteins involved in Strategy I response, such as Fe-chelate reductase (FC-R), H+-ATPase, and phosphoenolpyruvate carboxylase (PEPC) and the protein level of iron regulated transporter (IRT1) and H+-ATPase proteins has been investigated in both roots and nodules of a tolerant (Flamingo) and a susceptible (Coco blanc) cultivar of common bean plants. The main results of this study show that the symbiotic tolerance of Flamingo can be ascribed to a greater increase in the FC-R and H+-ATPase activities in both roots and nodules, leading to a more efficient Fe supply to nodulating tissues. The strong increase in PEPC activity and organic acid content, in the Flamingo root nodules, suggests that under iron deficiency nodules can modify their metabolism in order to sustain those activities necessary to acquire Fe directly from the soil solution.  相似文献

In the search for new therapeutic tools against tuberculosis two novel iron complexes, [Fe(L-H)3], with 3-aminoquinoxaline-2-carbonitrile N1,N4-dioxide derivatives (L) as ligands, were synthesized, characterized by a combination of techniques, and in vitro evaluated. Results were compared with those previously reported for two analogous iron complexes of other ligands of the same family of quinoxaline derivatives. In addition, the complexes were studied by cyclic voltammetry and EPR spectroscopy. Cyclic voltammograms of the iron compounds showed several cathodic processes which were attributed to the reduction of the metal center (Fe(III)/Fe(II)) and the coordinated ligand. EPR signals were characteristic of magnetically isolated high-spin Fe(III) in a rhombic environment and arise from transitions between mS = ± 1/2 (geff ~ 9) or mS = ± 3/2 (geff ~ 4.3) states. Mössbauer experiments showed hyperfine parameters that are typical of high-spin Fe(III) ions in a not too distorted environment. The novel complexes showed in vitro growth inhibitory activity on Mycobacterium tuberculosis H37Rv (ATCC 27294), together with very low unspecific cytotoxicity on eukaryotic cells (cultured murine cell line J774). Both complexes showed higher inhibitory effects on M. tuberculosis than the “second-line” therapeutic drugs.  相似文献

Background

Ferritin exhibits complex behavior in the ultracentrifuge due to variability in iron core size among molecules. A comprehensive study was undertaken to develop procedures for obtaining more uniform cores and assessing their homogeneity.

Methods

Analytical ultracentrifugation was used to measure the mineral core size distributions obtained by adding iron under high- and low-flux conditions to horse spleen (apoHoSF) and human H-chain (apoHuHF) apoferritins.

Results

More uniform core sizes are obtained with the homopolymer human H-chain ferritin than with the heteropolymer horse spleen HoSF protein in which subpopulations of HoSF molecules with varying iron content are observed. A binomial probability distribution of H- and L-subunits among protein shells qualitatively accounts for the observed subpopulations. The addition of Fe2+ to apoHuHF produces iron core particle size diameters from 3.8 ± 0.3 to 6.2 ± 0.3 nm. Diameters from 3.4 ± 0.6 to 6.5 ± 0.6 nm are obtained with natural HoSF after sucrose gradient fractionation. The change in the sedimentation coefficient as iron accumulates in ferritin suggests that the protein shell contracts ∼ 10% to a more compact structure, a finding consistent with published electron micrographs. The physicochemical parameters for apoHoSF (15%/85% H/L subunits) are M = 484,120 g/mol, ν? = 0.735 mL/g, s20,w = 17.0 S and D20,w = 3.21 × 107 cm2/s; and for apoHuHF M = 506,266 g/mol, ν? = 0.724 mL/g, s20,w = 18.3 S and D20,w = 3.18 × 107 cm2/s.

Significance

The methods presented here should prove useful in the synthesis of size controlled nanoparticles of other minerals.  相似文献

Plants have developed numerous strategies to cope with phosphorus (P) deficiency resulting from low availability in soils. Evolution of ethylene and up-regulation of root secreted acid phosphatase activity are common for plants in response to P deficiency. To determine the role of ethylene in response of plants to P deficiency, we investigated the effects of ethylene precursor (1-amino cyclopropane-1-carboxylic acid, ACC) and ethylene synthesis antagonists (aminoethoxyvinylglycine AVG, cobalt, Co2+) on P concentrations in roots and shoots of Medicago falcata seedlings grown in P-sufficient (500 μM H2PO4) and P-deficient (5 μM H2PO4) solution. After transferring M. falcata seedlings from P-sufficient to P-deficient solution for 2 days, root P concentration was significantly reduced. The reduction in root P concentration was reversed by AVG and Co2+, and a similar reduction in root P concentration of seedlings exposed to P-sufficient solution was observed by ACC. Expression of high-affinity phosphate transporters (MfPT1, MfPT5) was enhanced by P-deficiency and this process was reversed by AVG and Co2+. There was a marked increase in activity of root acid phosphatase (APase) and expression of gene encoding APase (MfPAP1) under P-deficient conditions, and the increase in APAse activity and expression of MfPAP1 was inhibited by AVG and Co2+. APase activity and expression of MfPAP1 expression in seedlings grown in P-sufficient solution were enhanced by ACC. Root and shoot P concentrations were increased when organic phosphorus was added to the P-deficient solution, and the increase in P concentration was significantly inhibited by AVG and Co2+. These results indicate that ethylene plays an important role in modulation of P acquisition by possibly mobilizing organic P via up-regulating root APase activity and high-affinity phosphate transporters.  相似文献

Oocytes of Xenopus tropicalis elicit a Ca2+-dependent outwardly rectifying, low-activating current (ICl,Ca) that is inhibited by Cl channel blockers. When inactivated, ICl,Ca shows an exponentially decaying tail current that is related to currents generated by TMEM16A ion channels. Accordingly, RT-PCR revealed the expression of five alternatively spliced isoforms of TMEM16A in oocytes, which, after expression in HEK-293 cells, gave rise to fully functional Cl channels. Upon hyperpolarization to −80 mV a transient current was observed only in isoforms that carry the exon 1d, coding for two potentially phosphorylatable Threonine residues. The identified isoforms are differentially expressed in several tissues of the frog. Thus, it appears that X. tropicalis oocytes express TMEM16A that gives rise to a Ca2+-dependent Cl current, which is different from the previously reported voltage-dependent outwardly rectifying Cl current.  相似文献

A novel composite membrane has been developed by doping cesium phosphotungstate salt (CsxH3−xPW12O40 (0 ≤ x ≤3), Csx-PTA) into chitosan (CTS/Csx-PTA) for application in direct methanol fuel cells (DMFCs). Uniform distribution of Csx-PTA nanoparticles has been achieved in the chitosan matrix. The proton conductivity of the composite membrane is significantly affected by the Csx-PTA content in the composite membrane as well as the Cs substitution in PTA. The highest proton conductivity for the CTS/Csx-PTA membranes was obtained with x = 2 and Cs2-PTA content of 5 wt%. The value is 6 × 10−3 S cm−1 and 1.75 × 10−2 S cm−1 at 298 K and 353 K, respectively. The methanol permeability of CTS/Cs2-PTA membrane is about 5.6 × 10−7, 90% lower than that of Nafion-212 membrane. The highest selectivity factor (φ) was obtained on CTS/Cs2-PTA-5 wt% composite membrane, 1.1 × 104/S cm−3 s. The present study indicates the promising potential of CTS/Csx-PTA composite membrane as alternative proton exchange membranes in direct methanol fuel cells.  相似文献

Salinization is one of the most important causes of crop productivity reduction in many areas of the world. Mechanisms that control leaf growth and shoot development under the osmotic phase of salinity are still obscure, and opinions differ regarding the Abscisic acid (ABA) role in regulation of biomass allocation under salt stress. ABA concentration in roots and leaves was analyzed in a genotype of processing tomato under two increasing levels of salinity stress for five weeks: 100 mM NaCl (S10) and 150 mM NaCl (S15), to study the effect of ABA changes on leaf gas exchange and dry matter partitioning of this crop under salinity conditions. In S15, salinization decreased dry matter by 78% and induced significant increases of Na+ and Cl in both leaves and roots. Dry matter allocated in different parts of plant was significantly different in salt-stressed treatments, as salinization increased root/shoot ratio 2-fold in S15 and 3-fold in S15 compared to the control. Total leaf water potential (Ψw) decreased from an average value of approximately −1.0 MPa, measured on control plants and S10, to −1.17 MPa in S15. In S15, photosynthesis was reduced by 23% and stomatal conductance decreased by 61%. Moreover, salinity induced ABA accumulation both in tomato leaves and roots of the more stressed treatment (S15), where ABA level was higher in roots than in leaves (550 and 312 ng g−1 fresh weight, respectively). Our results suggest that the dynamics of ABA and ion accumulation in tomato leaves significantly affected both growth and gas exchange-related parameters in tomato. In particular, ABA appeared to be involved in the tomato salinity response and could play an important role in dry matter partitioning between roots and shoots of tomato plants subjected to salt stress.  相似文献

The aim of this study was to investigate the cellular toxicity of copper-induced injury to the black tiger shrimp Penaeus monodon. The 24 h, 48 h, 72 h and 96 h LC50 (median lethal concentration) of Cu2+ on P. monodon (11.63 ± 1.14 g) were found to be 3.49, 1.54, 0.73 and 0.40 mg L− 1, respectively. Total haemocyte count (THC), phagocytic activity, respiratory burst (RB), cytoplasmic free-Ca2+ (cf-Ca2+) concentration and apoptotic cell ratio of shrimp were determined after exposure to different concentrations of Cu2+ (0, 0.05, 0.5, 1.5 and 3.5 mg L− 1) for 0, 6, 12, 24 and 48 h. There was no significant effect on the analytic indicator of shrimp exposed to 0.05 mg L− 1 Cu2+. THC decreased after Cu-exposure to 0.5 mg L− 1 for 48 h, 1.5 mg L− 1 for 24 h and 3.5 mg L− 1 for 12 h. Phagocytic activity decreased in P. monodon following 48 h exposure to 3.5 mg L− 1 Cu2+. RB was induced after 6 h exposure to 0.5, 1.5 and 3.5 mg L− 1 Cu2+. cf-Ca2+ concentration increased after 48 h exposure to 0.5 mg L− 1 Cu2+, and 12 h exposure to 1.5 and 3.5 mg L− 1 Cu2+. The percentage of apoptotic cells increased to 9.5%, 16.3% and 18.6% respectively following 48 h exposure to 0.5, 1.5 and 3.5 mg L− 1 Cu2+. These results indicate that Cu can induce oxidative stress, elevation of cf-Ca2+ and cell apoptosis, and inhibit phagocytic activity in the shrimp P. monodon, and the lethal injury of Cu2+ to P. monodon may be mainly due to the sharp reduction of THC caused by ROS-induced apoptosis.  相似文献

Lime application to aquatic systems may be an effective means of stressing macrophyte growth and promoting changes in species assemblage by inducing temporary dissolved inorganic carbon (DIC) limitation of productivity. Shoot and root growth response to lime (as Ca(OH)2) application was investigated for three macrophyte species (Elodea canadensis, Stuckenia pectinata, and Vallisneria americana) grown in experimental outdoor mesocosms. Lime was applied to mesocosms at three treatment levels to maintain pH for 1 week at 9.8–10.0 (1.64 mM), 10.3–10.5 (at the bicarbonate–carbonate equivalence point; 3.00 mM), and 10.8–11.0 (4.34 mM). pH recovered to control levels in all treated mesocosms 20 days after lime application. After treatment, HCO3 and DIC declined by 66, 93, and 93% and 60, 89, and 87%, respectively, versus increasing lime application. Concentrations remained lower in treated mesocosms versus the control throughout post-treatment. Differential growth response was observed in the 1.64 and 3.00 mM treatments, suggesting species-specific tolerances to both DIC concentration and form. V. americana was most sensitive to lime as the 1.64 mM treatment resulted in 54% shoot growth suppression versus the control and shoot plus root biomass loss in the 3.00 and 4.34 mM treatments. S. pectinata and E. canadensis exhibited net shoot and root growth (although significantly lower than controls) in both the 1.64 and 3.00 mM treatments and complete growth suppression in the 4.34 mM treatment. Selective control and shifts in species assemblage may be possible by adjusting lime concentration in relation to compensation point and needs to be investigated under field conditions.  相似文献

Absorbance difference spectroscopy and redox titrations have been applied to investigate the properties of photosystem I from the chlorophyll d containing cyanobacterium Acaryochloris marina. At room temperature, the (P740+ − P740) and (FA/B − FA/B) absorbance difference spectra were recorded in the range between 300 and 1000 nm while at cryogenic temperatures, (P740+A1 − P740A1) and (3P740 − P740) absorbance difference spectra have been measured. Spectroscopic and kinetic evidence is presented that the cofactors involved in the electron transfer from the reduced secondary electron acceptor, phylloquinone (A1), to the terminal electron acceptor and their structural arrangement are virtually identical to those of chlorophyll a containing photosystem I. The oxidation potential of the primary electron donor P740 of photosystem I has been reinvestigated. We find a midpoint potential of 450 ± 10 mV in photosystem I-enriched membrane fractions as well as in thylakoids which is very similar to that found for P700 in chlorophyll a dominated organisms. In addition, the extinction difference coefficient for the oxidation of the primary donor has been determined and a value of 45,000 ± 4000 M− 1 cm− 1 at 740 nm was obtained. Microorganisms living in arsenic-rich geothermal environments act on arsenic with different biochemical strategies, but the molecular mechanisms responsible for the resistance to the harmful effects of the metalloid have only partially been examined. In this study, we investigated the mechanisms of arsenic resistance in the thermophilic bacterium Thermus thermophilus HB27. This strain, originally isolated from a Japanese hot spring, exhibited tolerance to concentrations of arsenate and arsenite up to 20 mM and 15 mM, respectively; it owns in its genome a putative chromosomal arsenate reductase (TtarsC) gene encoding a protein homologous to the one well characterized from the plasmid pI258 of the Gram + bacterium Staphylococcus aureus. Differently from the majority of microorganisms, TtarsC is part of an operon including genes not related to arsenic resistance; qRT-PCR showed that its expression was four-fold increased when arsenate was added to the growth medium. The gene cloning and expression in Escherichia coli, followed by purification of the recombinant protein, proved that TtArsC was indeed a thioredoxin-coupled arsenate reductase with a kcat/KM value of 1.2 × 104 M− 1 s− 1. It also exhibited weak phosphatase activity with a kcat/KM value of 2.7 × 10− 4 M− 1 s− 1. The catalytic role of the first cysteine (Cys7) was ascertained by site-directed mutagenesis. These results identify TtArsC as an important component in the arsenic resistance in T. thermophilus giving the first structural–functional characterization of a thermophilic arsenate reductase.  相似文献
