Ginkgo biloba exocarp polysaccharide (GBEP) was obtained by hot water extraction, the crude polysaccharide was deproteinized by Sevag method and fractionized by a DEAE Sepharose fast flow anion-exchange column. Five fragments were obtained, including neutral polysaccharide (GBEP-N) and four acidic polysaccharides (GBEP-A1, GBEP-A2, GBEP-A3 and GBEP-A4). GBEP-N and GBEP-A3 were further purified by Superdex 200 gel column chromatography. The resulted two fractions GBEP-NN, and GBEP-AA were characterized by FT-IR, and HPGFC (high pressure gel filtration chromatography). Monosaccharide composition was determined by RP-HPLC method of precolumn derivatization with 1-phenyl-3-5-pyrazolone. GBEP-NN was mainly composed of rhamnose, arabinose, mannose, glucose and galactose, while GBEP-AA was mainly made up of mannose, rhamnose, glucuronic acid, galacturonic acid, galactosamine, glucose, galactose, xylose, arabinose, and fucose. The crude GBEP exhibited certain antioxidant activity. At the concentration of 5 mg/mL, the hydroxyl radical scavenging effect of GBEP was 90.52%, greater than 77.37% for the positive control ascorbic acid. 相似文献

城市大规模的地表硬化改变了城市环境,影响了城市中植物的正常生长。利用土壤水分温度仪ECH2O测定了不同硬化地表下土壤含水率(WCS)和土壤温度(Ts),利用LI-6400光合仪,并配备荧光叶室,测定了银杏生长的环境因子和银杏生理生态参数。研究结果表明,城市地表硬化对银杏生长环境的影响主要表现为空气温度(Ta)和土壤温度(Ts)升高,空气相对湿度(RH)和土壤含水率(WCS)下降,银杏对地表硬化的生理生态响应表现为净光合速率(Pn)、蒸腾速率(Tr)、气孔导度(Gs)降低,叶面饱和水汽压亏缺(VPDL)增大,叶片含水率(LWC)下降;最大光化学效率(Fv/Fm)和PSⅡ实际光合电子传递量子效率(ΦPSⅡ)下降,硬化程度越高,银杏受到的胁迫越重;受城市地表硬化的影响,银杏吸收的光能用于光化学反应的比例减少,而用于热耗散和荧光耗散的比例增加,城市硬化地表上银杏的水分利用效率(WUE)、光能利用效率(LUE)和CO2利用效率(CUE)明显下降。 相似文献

Eight primer combinations that produced clear and a large number of polymorphic bands were screened from 64 EcoR I/Mse I primer combinations (Mse I fluorescent labeled). The genetic relationships of 21 ornamental cultivars of Ginkgo biloba L. from the United States of America, Holland, Japan, France, and China were analyzed. These primer combinations produced a total of 1 119 bands, 229 specific loci (including 54 absent bands, and 175 monomorphic bands). Among them, 983 polymorphic bands (PPB), accounting for 88%, were detected. The percentage of identification per primer combination was as high as 100%. The average PPB of 14 foreign cultivars was 35.86% and the average PPB of seven domestic cultivars was 31.51%. Genetic similarity coefficient (SC) among all cultivars varied from 0.4899 to 0.8499, and all cultivars were divided into the four clusters when SC was set at 0.7300. The cultivars from the same origin did not fall into the same group. The cultivars from France and China were classified into three groups. According to the comprehensive analyses based on specific loci, similarity coefficient, and clustering results, eight cultivars ‘Fastigiata’, ‘Tit’, ‘Tubifolia’, ‘Daeryinxing’, ‘Variegata’, ‘Horizontalis, ‘Pendula’, and ‘Yiyuanyeziyinxing’ were considered to be important germplasms of ornamental cultivars of Ginkgo biloba. 相似文献

Orthosiphon stamineus (Java tea) has been widely used as traditional herb and several bioactive compounds against animal cells have been isolated. However, no bioactive compound against plants has been reported. Therefore, we investigated possible allelopathic properties and substances in O. stamineus. Aqueous methanol extracts of O. stamineus inhibited root and hypocotyl growth of cress (Lepidium sativum) and lettuce (Lactuca sativa) seedlings. Increasing the extract concentration increased the inhibition, which suggests that O. stamineus may have allelopathic properties. When the extract was divided into an ethyl acetate and an aqueous fraction, the ethyl acetate fraction showed the stronger inhibitory effect. Thus, the ethyl acetate phase was further purified, and the main allelopathic substance was isolated and identified as 13-epi-orthosiphol N, a novel compound, by spectral data. 13-epi-Orthosiphol N inhibited root and hypocotyl growth of cress and lettuce at concentrations greater than 10 μmol/L. The concentrations required for 50% inhibition ranged from 41 to 102 μmol/L. These results suggest that 13-epi-orthosiphol N may be an allelochemical and main contributor to the growth inhibitory effect of O. stamineus and may have potential as a template for the development of new plant control substances. 相似文献

Ginkgo biloba extract (GBE) is a popular herbal ingredient used worldwide, but it is reported to induce bleeding as a serious adverse event. In this study we examined whether GBE induced spontaneous bleeding or accelerated warfarin anticoagulation via herb-drug interaction. Mice were given GBE or various active components of GBE orally for 5 days and blood coagulation parameters and hepatic cytochrome P450 enzymes (CYPs) were measured. Mice also received warfarin (racemate, (S)- or (R)-enantiomer) for the last 3 days of the 5-day regimen to examine GBE-warfarin interactions. Neither GBE (up to 1000 mg/kg) nor ginkgolide B (up to 140 mg/kg), a platelet-activating factor antagonist, influenced blood coagulation parameters. In contrast, GBE attenuated the anticoagulant action of warfarin. Bilobalide, a component of GBE that markedly induced hepatic CYPs including (S)-warfarin hydroxylase, showed similar effects. For (S)-warfarin, the anticoagulation action and the interaction with GBE was clear, while the influence on metabolism was greater for (R)-warfarin than for (S)-warfarin, which corresponded to the CYP types induced by GBE. These results suggest that GBE and ginkgolide B have no influence on blood coagulation in vivo, and that GBE attenuates the anticoagulation action of warfarin via induction of hepatic CYPs by bilobalide. 相似文献

An araban type polysaccharide (GBPw) was purified from the leaves of Ginkgo biloba. The present study aimed to investigate the protective effects of GBPw on focal ischemia/reperfusion (I/R) injury in rat brain. The results of this study demonstrated that GBPw had a positive effect on the rat brain when administered 7 days before focal cerebral I/R injury. This effect was evident with the improvements in neurological deficits, reduction in infarct volume, MDA content and the levels of pro-inflammatory cytokines (TNF-α and IL-1β), and elevation in the SOD and MPO activities and the levels of anti-inflammatory cytokine (IL-10). Thus, the beneficial effects of GBPw on cerebral I/R injury may result from the reduction of oxidative stress and the inhibition of NO production and inflammation induced by I/R. The neuroprotective effects of GBPw supplement may have potential implication in the future for prevention/protection against cerebral ischemic stroke. 相似文献

A novel polyhydroxylated C29-sterol, 25ξ-methyl-22-homo-5α-cholest-7,22-diene-3β,6β,9α-triol, designated globosterol (1), together with one known tetrahydroxylated ergosterol (22E, 24R)-ergosta-7,22-diene-3β,5α,6β,9α-tetraol (2) has been isolated from the cultures of an endophytic fungus, Chaetomium globosum ZY-22 originated from the plant Ginkgo biloba. The structures and relative configurations of 1 and 2 were established on the basis of extensive spectroscopic analyses including 1D and 2D NMR (1H-1H COSY, HSQC, HMBC, and NOESY) experiments and comparison with the literature. Monocytes expressing toll-like receptor 4 (TLR4) play a major role in regulating the innate immune response and are involved in systemic inflammation. Previous studies have shown that Ginkgo biloba extract (GBE) may act as a therapeutic agent for some cardiovascular and neurological disorders. The objective of this study was to determine whether GBE could modulate immunity in human cells. The monocytic cell line THP-1 was used. Enzyme-linked immunosorbent assay results showed that lipopolysaccharide (LPS) induces the expression of monocyte chemotactic protein-1 (MIP-1), tumor necrosis factor-α, stromal cell-derived factor-1, and MIP-1α, and this induction may be repressed by GBE treatment due to TLR4 blockade. The Griess reagent assay and western blot analysis showed that GBE-mediated inhibition of TLR4 expression was associated with the activation of mitogen-activated protein kinase and production of nitric oxide (NO). Actinomycin D chase experiments demonstrated that GBE decreased the TLR4 mRNA stability in cells. Confocal microscopy and real-time polymerase chain reaction showed that GBE induced the expression of intracellular tristetraprolin (TTP). Transfection with TTP siRNA reversed the effects of GBE in naïve or TLR4-overexpressing cells. Treatment with SNAP (an NO donor) may increase intracellular TTP expression in cells. Immunoprecipitation analysis showed that GBE mediates TTP activation and increases the interaction of TTP with the 3′ untranslated region (UTR) of TLR4 mRNA by regulating NO production. Our findings indicate that GBE could decrease the sensitivity of monocytes to LPS. Utilizing TTP to control TLR4 expression may be a promising approach for controlling systemic inflammation, and GBE may have potential applications in the clinical treatment of immune diseases. 相似文献

Taxol is a well-known effective anticancer compound. Due to the inability to synthesize sufficient quantities of taxol to satisfy commercial demand, a biotechnological approach for a large-scale cell or cell-free system for its production is highly desirable. Several important genes in taxol biosynthesis are currently still unknown and have been shown to be difficult to isolate directly from Taxus, including the gene encoding taxoid 9α-hydroxylase. Ginkgo biloba suspension cells exhibit taxoid hydroxylation activity and provides an alternate means of identifying genes encoding enzymes with taxoid 9α-hydroxylation activity. Through analysis of high throughput RNA sequencing data from G. biloba, we identified two candidate genes with high similarity to Taxus CYP450s. Using in vitro cell-free protein synthesis assays and LC–MS analysis, we show that one candidate that belongs to the CYP716B, a subfamily whose biochemical functions have not been previously studied, possessed 9α-hydroxylation activity. The residue of peas (Pisum sativum L.) has allelopathic activity and the putative compound causing this inhibitory effect was isolated from a methanol extract of pea shoots. Chemical structure of this compound was determined by high-resolution MS, IR and 1H NMR spectral data as pisatin. Pisatin inhibited growth of cress (Lepidium sativum L.) and lettuce (Lactuca sativa L.) seedlings at concentrations greater than 10 and 30 microM, respectively. The doses required for 50% growth inhibition of roots and hypocotyls of cress were 61 and 91 microM, respectively, and those of lettuce were 78 and 115 microM, respectively. The concentration of pisatin in the pea shoots was 32.7 nmol x g(-1) fresh weight. The effectiveness of pisatin on growth inhibition in cress and lettuce, and its occurrence in pea shoots suggest that it may contribute to the growth inhibitory effect of pea residue, and may play an important role in pea allelopathy. 相似文献

Allelopathic effects of fresh tissue and dry powder of a nonsexual and a sexual strain of the macroalga Ulva pertusa on the growth of the microalgae Heterosigma akashiwo and Alexandium tamarense were evaluated using long-term coexistence culture systems in which several concentrations of macroalga fresh tissue and dry powder were used. The effects of macroalga culture medium filtrate on the two HAB algae were also investigated. Moreover, isolation co-culture systems were built to confirm the existence of allelochemicals and preclude the growth inhibition by direct contact. Short-term algicidal effect assays of macroalgae on H. akashiwo were carried out to measure the rate of algal cell lysis. The results of the coexistence assays showed that the growth of H. akashiwo and A. tamarense was strongly inhibited by fresh tissue and by dry powder of both strains of U. pertusa. The allelochemicals were lethal to H. akashiwo at relatively higher concentrations. The macroalga culture medium filtrate exhibited no apparent growth inhibitory effect on the two HAB algae under initial or semicontinuous filtrate addition, which suggested that continuous release of small quantities of rapidly degradable allelochemicals from the fresh tissue of both strains of U. pertusa was essential to effectively inhibit the growth of H. akashiwo and A. tamarense. 相似文献

The content of linolenic acid and its fat-soluble derivatives in Ginkgo leaves has been determined. By utilization of uniformly 14C-labelled linolenic acid it has been shown that the linolenic acid in Ginkgo leaves is converted into 2-hexenal when the leaves are macerated in the presence of air. The conversion of linolenic acid to 2-hexenal under the conditions of temperature and pH existing in the Ginkgo leaf requires the presence in the leaves of an enzyme or other catalyst. This is not lipoxidase but is a hexane-insoluble, water-soluble substance. A preparation of this substance strongly catalyzes the absorption of oxygen by linolenic acid in water at 20°. 相似文献

Aqueous methanol extracts of Bangladesh rice (Oryza sativa L. cv. BR17) inhibited the growth of roots and shoots of cress (Lepidium sativum), lettuce (Lactuca sativa), alfalfa (Medicago sativa), timothy (Phleum pratense), Digitaria sanguinalis, Echinochloa crus-galli and Echinochloa colonum. Increasing the extract concentration increased the inhibition, suggesting that the BR17 may have growth inhibitory substances
and possess allelopathic potential. The aqueous methanol extract of the BR17 was purified and a main inhibitory substance
was isolated and determined by spectral data as 2,9-dihydroxy-4-megastigmen-3-one. This substance inhibited root and shoot
growth of cress and E. crus-galli seedlings at concentrations greater than 0.03 and 3 μM, respectively. The concentrations required for 50% growth inhibition
on cress roots and shoots were 0.22 and 0.47 μM, respectively, and on E. crus-galli roots and shoots were 36 and 133 μM, respectively. These results suggest that 2,9-dihydroxy-4-megastigmen-3-one may contribute
to the growth inhibitory effect of BR17 and may play an important role in the allelopathy of BR17. Thus, Bangladesh rice BR17
may be potentially useful for weed management in a field setting.

The inhibitory effect of Citrus junos peel on plant growth using lettuce (Lactuca sativa L.) as a bioassay material was investigated, since the powder of the peel had been found to inhibit growth of weeds. Basic, neutral and acidic fractions were separated from the aqueous fraction obtained from the methanol extract of C. junos peel. All fractions inhibited the growth of lettuce seedlings, but by far the greatest inhibition was observed with the neutral fraction. Thus, the latter was further purified and an allelopathically active substance was isolated. The structure of the substance was determined from high-resolution MS and 1H and 13C NMR spectral data as abscisic acid-beta-D-glucopyranosyl ester (ABA-GE). ABA-GE inhibited hypocotyl and root growth of lettuce seedlings at concentrations greater than 0.3 microM, and the concentrations for 50% inhibition of hypocotyl and root growth were 2.3 and 1.4 microM, respectively. The effectiveness of ABA-GE on inhibition of growth and the occurrence of ABA-GE in the peel itself suggested that ABA-GE may play an important role in the allelopathic potential of C. junos peel. 分析了太行山南麓低丘山区降水、泉水、地下水、土壤水以及栓皮栎、扁担杆的氢氧稳定同位素特征,结合Iso Source模型确定了栓皮栎和扁担杆水分来源的季节性差异,并对栓皮栎和扁担杆水分利用策略进行分析。结果表明,同一生态系统中的栓皮栎和扁担杆枝条水的δ18O和δD值差别明显。雨季中栓皮栎和扁担杆水分来源较浅,以0—20 cm土壤水分为主,但旱季中栓皮栎和扁担杆水分主要来源均比雨季明显加深,其中栓皮栎主要利用40—60 cm土壤水分,扁担杆则主要利用20—40 cm土壤水分。此外,旱季后期栓皮栎还利用部分泉水,其比例达到了19.6%。二者水分来源的不同,使得栓皮栎与扁担杆在旱季期间能避开用水冲突。旱季中生长在生态系统上层的栓皮栎中午部分气孔关闭,蒸腾速率下降,生长在生态系统下层的扁担杆日均蒸腾速率、气孔导度则分别比栓皮栎下降了46.94%和30.58%。栓皮栎和扁担杆分别采取了深水源及部分气孔关闭和浅水源及低蒸腾耗散的水分利用策略来利用旱季中有限的水分,因而其组成的生态系统表现出较强地适合太行山南麓脆弱环境的生态适应性。 相似文献

The introduced, invasive species Conyza canadensis L. covers large areas of the sandy levees next to the River Tamiš (Serbia), forming dense microcomplexes and dominating the other herbaceous species in the ruderal phytocoenosis with its aboveground mass and abundance. In addition to this species, a further 28 plant species were found, but the abundance and cover of these was significantly lower. The allelopathic influence of the species C. canadensis was investigated through analyzing the total phenolics and phenolic acids, as the main allelochemicals, in dead and vegetative parts and the soil beneath them. Seed germination and seedling growth of the target plants (Dactylis glomerata L. and Trifolium repens L.), which grow in this community, served as a measure of the inhibitory capacity of this species. It was established that the content of total phenolics and phenolic acids (p-coumaric, ferulic, p-hydroxybenzoic, vanillic and syringic) varies, following the order: vegetative plant parts > dead plant parts > sandy soil under C. canadensis. Water leachate and soils inhibited seed germination and seedling growth of the test plants to varying degrees, following the order: vegetative parts > dead parts > sandy soil, which is directly related to the content of total phenolics and phenolic acids in them. It was concluded that the pioneer species C. canadensis plays a decisive role in the first phases of vegetation succession and the process of soil formation on the barren sandy levees, owing to the synthesis of secondary phenolic metabolites. 相似文献

Two compounds from allelopathic rice accession and their inhibitory activity on weeds and fungal pathogens 总被引:17,自引:0,他引:17
A flavone (5,7,4'-trihydroxy-3',5'-dimethoxyflavone), a cyclohexenone (3-isopropyl-5-acetoxycyclohexene-2-one-1) and a liquid mixture of low polarity, containing long-chain and cyclic hydrocarbons, were isolated from leaves of allelopathic rice accession PI 312777 using column chromatography. Their structures and constituents were identified by means of HR-MS, NMR and GC/MS analyses, respectively. Bioassays showed that both the flavone and cyclohexenone significantly inhibited the growth of weeds Echinochloa crus-galli, Cyperus difformis and Cyperus iris, and the spore germination of fungal pathogens Pyricularia oryzae and Rhizoctonia solani at all tested concentrations. Moreover, the combination of the inactive mixture of low polarity and the active flavone or cyclohexenone significantly enhanced the inhibitory activities on weed growth. In addition, the two compounds and the mixture of low polarity from the leaves of PI312777 did not inhibit the rice growth at the same concentrations. It was also established that both compounds could be released into the soil, and was especially induced by E. crus-galli. The results suggest that 5,7,4'-trihydroxy-3',5'-dimethoxyflavone and 3-isopropyl- 5-acetoxycyclohexene-2-one-1 may act as allelochemicals participating in the defense of rice against weeds and pathogens. 相似文献
