To verify if non-invasive collection of soft faeces (SF) from rabbits can be used as an index of bacterial biodiversity in caecal contents (CC), 24 weaned rabbits were given diets with low (LI) and high (HI) levels of insoluble fibre (neutral detergent fibre, NDF) and low (LS) and high (HS) levels of soluble fibre (neutral detergent soluble fibre, NDSF). After 21 days, animals were fitted with neck collars for SF collection. Two days later, animals were slaughtered and CC sampled. Total bacterial concentration quantified by real time PCR (log10 ng DNA/mg DM) was higher in SF than CC (2.615 vs. 2.383). Among diets, in CC it was (P = 0.059) lowest in LILS diet, whereas in SF it decreased (P = 0.025) with the NDF level. DGGE profiles showed that structure of bacterial communities of SF was close to that of CC; however, similarity was higher in LI than HI diets (0.82 vs. 0.74). Diversity indexes in CC decreased with NDSF (P < 0.05), whereas the effect of NDF (P < 0.05) was also appreciated in SF. Soft faeces can be an alternative to surgery or slaughter techniques to monitor changes in caecal bacterial community; however, high dietary NDF may decrease similarity between both communities.  相似文献

The ethanolic extract of seeds of S. cumini increased body weight and decreased blood sugar level in alloxan diabetic albino rats. Level of significance for decrease in blood sugar after feeding alcoholic extract of S. cumini seeds in various doses was highly significant. The extract feeding showed definite improvement in the histopathology of islets. The most important finding is that the blood sugar level, which once dropped to normal levels after extract feeding was not elevated when extract feeding was discontinued for 15 days.  相似文献

An improved understanding of the role of forage quality on the processes of particle dynamics and turnover is important for the development of healthier and cost-effective feeding strategies that aim at lowering the proportions of concentrates in the diets of cattle. The aim of this study was to evaluate the effects of feeding hays of different qualities on particle dynamics, digestion kinetics and turnover in the gastrointestinal tract (GIT). Three non-lactating, rumen fistulated Holstein cows were fed diets consisting exclusively of hay with either low quality [Group LH; 605 ± 12.4 g/kg neutral detergent fibre (NDF) and 63 ± 4.7 g/kg crude protein (CP)] or good quality (Group GH; 551 ± 20.1 g/kg NDF and 116 ± 3.6 g/kg CP). Data showed that in situ dry matter (DM) disappearance of the soluble fraction was greater for Group GH (p < 0.05). Feeding good quality hay also lowered the proportion of particles >1.18 mm particularly during the eating process (p < 0.05). Changes in the particle size occurring afterwards were greater for Group GH as well (p < 0.05); approximately 30% in the comminution in the particle size occurred postruminally. Feeding hay of good quality lowered DM content of solid rumen digesta (p < 0.05), accelerated (p < 0.05) the turnover rate of DM and NDF in the GIT and increased DM intake (p < 0.05). In conclusion, feeding forages of better quality significantly promoted degradation processes and kinetics in the GIT with positive effects on turnover rate of digesta and feed intake in Holstein cows.  相似文献

Cannulated beef cattle (four cows: 556 kg initial weight; four steers: 504 kg initial weight) were used in an experiment with two simultaneous Latin squares to determine effects of substituting alfalfa and(or) corn for vegetative bermudagrass (BER; 77% neutral detergent fibre and 5.5% acid detergent lignin) or mature bromegrass (BRO; 70% neutral detergent fibre and 6.6% acid detergent lignin) hay on digestion characteristics. For Controls, BER or BRO was fed at 1.32 or 1.54% body weight of cows and steers, respectively; other treatments entailed substitution for hay DM of alfalfa cubes (17%) or ground corn (33%). A protein supplement was given to all animals. In vitro neutral detergent fibre (NDF) digestion was slightly greater for BER than BRO. Supplement treatments did not affect the concentration of total volatile fatty acids or the molar proportion of propionate in ruminai fluid. True ruminai organic matter (OM) digestion was similar among diets; greater duodenal microbial OM flow and postruminal NDF digestion for BRO than BER diets were responsible for higher (P < 0.05) postruminal OM digestion for BRO diets. Supplement treatment did not affect duodenal microbial nitrogen flow or efficiency of microbial growth. Corn supplementation increased total tract OM digestion (P < 0.05). Alfalfa addition depressed total tract OM and NDF digestibilities more when added to BER than BRO; depressions in total tract NDF digestion with alfalfa and corn substitutions were additive. With constant DM intake, slightly less than ad libitum, alfalfa or corn substituted alone or together for hay did not improve characteristics of digestion by cattle consuming vegetative bermudagrass or mature bromegrass other than increased postruminal and total tract OM digestiblities with corn.  相似文献

A simulation rumen model has been developed to function under non-steady state conditions in order to allow prediction of nutrient availability in dairy cows managed under discontinuous feeding systems. The model simulates availability of glycogenic, aminogenic and lipogenic nutrients to lactating dairy cows fed discontinuously. The model structure considers input of up to three different feeds fed independently at any time during the day. Feeds are described by their nitrogen (N), carbohydrate and fatty acid fractions. The N containing feed fractions include ruminally undegraded crude protein (CP), ruminally insoluble but potentially degradable CP, ruminally soluble CP and ammonia N. The feed carbohydrate fractions include ruminally undegradable neutral detergent fibre (NDF), ruminally degradable NDF, ruminally insoluble starch, ruminally soluble starch and sugars. The fatty acids in the feeds are divided between long chain fatty acids and volatile fatty acids (VFA). Additionally four pools were defined representing absorption of amino acids, glucose, long chain fatty acids and volatile fatty acids. The rumen microbial population is represented as a single pool. Besides a flexible structure, new features to the extant model include adoption of the concept of chewing efficiency (or chewing effectiveness) during eating, variable fractional ruminal absorption rates of VFA and variable fractional ruminal degradation rates of NDF as a function of rumen liquid pH, as well as a variable rumen volume which directly affects rumen concentrations of metabolites. The model continuously (i.e., by minute) predicts release of soluble components from the feeds in the rumen, concentration and absorption of fermentation end products in the rumen, rumen pools of nutrients and microbial biomass dynamics, as well as passage of microbial biomass and non-fermented nutrients from the rumen, in response to various feeding strategies. Model evaluation covered a wide range of feeding strategies that included pasture and housed feeding systems. Overall, the mean square prediction error (MSPE) as a percentage of the observed mean was relatively low (<10%) with a high amount of the total variation explained by random variation (>65%). Deviation from unity varied between 23% (rumen dry matter content) and 25% (NDF), indicating some consistent over and/or under prediction. A more detailed evaluation was done based on studies available that reported diurnal behaviour of key model outputs such as rumen pools, rumen pH, and rumen VFA. The predictions broadly simulated the observed values quantitatively, relative to general diurnal patterns, and relative to differences between treatments in the predicted diurnal patterns. Results show that the model provides a tool to assess potential outcomes of changing feeding strategies which may be particularly valuable in assessing selection of feeds, amounts and times of the day to offer the feeds. The continuous nature of the simulated output also allows determination of the time(s) of the day that ruminal (and/or post-ruminal) delivery of nutrients may limit ruminal output of nutrients (and/or availability of nutrients) to support milk nutrient synthesis.  相似文献

The hypoglycemic effects of Eriobotrya japonica seeds were investigated in type 2 diabetic Otsuka Long-Evans Tokushima fatty (OLETF) rats and KK-A(y) mice. The rats and mice were fed on a diet containing 10% powdered Eriobotrya japonica seeds with the coat intact for 4 months. Although the blood glucose concentration in the OLETF rats fed on the control diet without Eriobotrya japonica seeds was increased with time, the concentration in the OLETF rats fed on the diet with Eriobotrya japonica seeds was consistently low throughout the experimental period and was comparable to the level in Long-Evans Tokushima Otsuka (LETO) rats which are normal non-diabetic rats. Serum insulin was significantly lower in the OLETF rats fed on the Eriobotrya japonica seed diet than in those fed on the control diet at the termination of the experimental period. Eriobotrya japonica seeds suppressed the increment of blood glucose for 4 months and also effectively improved the glucose tolerance in the KK-A(y) mice, these actions being mainly exerted by the ethanol extract of the seeds. These results suggest that Eriobotrya japonica seeds had a hypoglycemic property and the effect is attributable to the components extracted by ethanol.  相似文献

Seeds of four tropical multipurpose trees (Albizia saman, Albizia lebbeck, Albizia rhizonse, Leucaena leucocephala), two shrubs (Tephrosia candida, Tephrosia bracteolata) and two pulse legume (Lablab purpureus, Canavalia ensiformis) were chemically analysed for dry matter (DM), ash, crude protein (CP), neutral detergent fibre (NDF) and ether extract (EE). Qualitative evaluation of secondary metabolites (saponins, phenols, steroids, and alkaloids) was elucidated. The DM, ash, CP, NDF and EE ranged between 88.9-93.6 %, 3.0-5.4 %, 24.8-38.2 %, 22.1-46.9 % and 2.0-17.0 % respectively. All seed species contained at least one group of secondary plant metabolites and steroids were common to all except C. ensiformis that was not implicated for any. A. lebbeck and A. rhizonse showed low saponin content. Indications for water soluble tannins were reported for L. leucocephala while the two species of Tephrosia contained flavonoids or condensed tannins. The study suggested the potentials of the legumes seed species as a feed source for ruminants.  相似文献

A laboratory method for determination of fibres insoluble by cellulase is suggested as a routine analysis for cattle compound feeds with varying proportion and quality of chemically treated straw and other fibrous components, in order to measure whether actual energy content corresponds to that declared.By a two-step analytical procedure, the total fibre is isolated by boiling with neutral detergent solution for 1 h, which removes the readily soluble carbohydrates, proteins and fat. By incubation of the isolated fibre in a solution of buffers and a commercial cellulase reagent for 48 h at 40°C the digestible fibre is removed. The organic matter in the residue, which is termed cellulase insoluble fibre (CIF), is a measure of unavailable organic matter for ruminants.The CIF content, total NDF and proximate constituents were determined in 47 compounds of extreme properties. Compared with digestibility in vitro (IVDOM), more organic matter was dissolved by the CIF-analyses in highly digestible feeds and less in feeds with low digestibility (IVDOM + fat = 87 ? 0.70 CIF).  相似文献

Two experiments were conducted to evaluate the nutritional quality of cattle manure silage for ruminants. In experiment 1 a silage containing 50% cattle manure, 20% cane molasses (containing 0.4% urea) mixed with water in a 1:1.5 ratio, and 30% corn stover replaced 0, 50, 75 and 100% of corn stover in isonitrogenous diets (1.92% N) containing 70 or 80% corn stover (dry matter (DM) basis) in a 4×2 factorial arrangement. Forty lambs (34.5±3.2 kg body weight (BW)) were used to measure apparent digestibility and nitrogen balance in a completely randomized design with factorial arrangement (4×2) with five lambs per treatment. The cattle manure silage was also evaluated in growing Holstein heifers (212±32 kg BW) replacing 0, 25 or 37.5% of corn stover with cattle manure silage in isonitrogenous diets (2.24% N). Apparent digestibility of DM, organic matter (OM), neutral detergent fibre (NDF) and acid detergent fibre (ADF) were not affected. According to these results, digestibility is not affected by the level of cattle manure silage; however, intake was depressed at the highest levels of silage in lambs. Cattle manure silage may be considered as a potential by-product to be included in heifer's diet up to 37.5%, apparently without affecting performance. The Hoatzin Opisthocomus hoazin is the only known bird with a well-developed foregut plant fermentation system; most fermentation takes place in the crop and caudal oesophagus. To test Hoatzin digestive efficiency, balance (total collection) trials with captive Hoatzins were made using two experimental diets of different composition and fibre content. Dry matter (DM) intakes were similar for the diets (mean = 62.8 g DM/kg body mass/ day). Average DM, organic matter and nitrogen digestibilities were not significantly different between diets, with average values of 72.9%, 75.0% and 78.3%, respectively. In vitro organic matter digestibilities by cow ruminal inoculum were very similar to organic matter digestibilities in live Hoatzins for both diets. Fibre digestibility was among the highest recorded for herbivorous birds. Cellulose and acid detergent fibre digestibilities were 58.8% and 52.7%, respectively. Neutral detergent fibre (NDF) digestibility differed among diets—the higher the NDF content of the diet, the higher the NDF digestibility. The NDF digestibilities were 37.9% and 70.9% for the two diets with NDF concentrations of 32.4% and 37.3%, respectively. Differences in NDF digestibility can be attributed to the different concentrations of hemicellulose in the experimental diets. The high overall digestibility by captive Hoatzins is higher than values previously reported for other avian herbivores but similar to those of foregut-fermenting mammals on similar diets. The unique digestive strategy of the Hoatzin maximizes digestion of cell wall and cell contents. The high digestive efficiency in the Hoatzin is not predicted by allometric models of fibre digestion as a function of body mass. Other nutritional benefits, such as detoxification of plant secondary compounds and microbial synthesis of essential amino acids and vitamins, may explain the evolution of foregut fermentation in this avian folivore.  相似文献

Plantain sheath (Musa sapientum) contained 6.4% dry matter and 3.4% crude protein, 31.4% crude fibre, 34.6% cellulose, 15.5% hemicellulose and 6% lignin in dry matter.

Digestibility in vitro studies with fistulated bucks revealed a dry matter disappearance of 53.3% after 24 h incubation. The digestibility of neutral detergent fibre (NDF), acid detergent fibre (ADF) and cellulose after 24 h fermentation was 32.2, 21.5 and 30.3%, respectively. After 48 h fermentation, the dry matter digestibility increased to 63.4%, NDF to 47.0%, ADF to 32.2% and cellulose to 42.3%.

A 45-day feeding trial was conducted with 16 crossbred lambs. Plantain sheath replaced paragrass hay dry matter at 32.6, 54.7 and 76.6% levels. Dried plantain sheath, which contributed 14.1, 25 and 34.5% of total dry matter intake, helped a daily gain of 31.1, 28.9 and 37.8 g when fed mixed with concentrate feed.  相似文献

A study was conducted to determine the effects of stage of maturity on ensiling characteristics and ruminal nutrient degradability of oat silage. Oat was field grown and forage was harvested at the boot or soft dough stage and ensiled in mini-silos for 0, 2, 4, 8, 16 and 45 days. Two lactating Holstein cows fitted with ruminal fistulas were used determine ruminal nutrient degradability. Regardless of the stage of maturity, ensiled forages went through a rapid fermentation with a sharp decline in pH during the first 2 days of ensiling. Extensive proteolysis took place between 0 and 2 days as indicated by a reduction in true protein and neutral detergent insoluble protein (NDICP) and an increase in non-protein nitrogen (NPN). Chemical analysis of the 45 days silage showed that stage of maturity had no effect on neutral detergent fibre (NDF) and acid detergent fibre (ADF) of oat silage. However, oat harvested at the boot stage contained more crude protein (CP) and less starch than that harvested at the soft dough stage. Distribution of protein fractions showed that oat harvested at the boot stage contained lower NPN, NDICP and acid detergent insoluble protein than oat harvested at the soft dough stage. Results of the in situ incubation experiment indicated that oat harvested at the soft dough stage had lower ruminal dry matter (60.6 vs. 66.4%), CP (81.3 vs. 88.7%) and NDF (35.4 vs. 42.2%) degradabilities than oat harvested at the boot stage. It was concluded that chemical composition and ruminal nutrient degradability of oat silage are significantly influenced by stage of maturity.  相似文献

We investigated the food digestibility of East Pacific green turtles, Chelonia mydas agassizii, at tropical coral reefs of Gorgona National Park in the Colombian Pacific and calculated the intake passage time (IPT) of 3 different diets. We collected 150 faecal samples from turtles (mean straight carapace length 61.3 ± 4.12 cm and mean mass 32.3 ± 6.67 kg) to determine digesta composition and for measurement of neutral detergent fibre (NDF), acid detergent fibre (ADF), sulphuric acid lignin and protein. The mean (± S.D) IPT to recover at least 73% of external markers (plastic beads) in the faeces was 23.3 ± 6.6 days. The true NDF digestibility and dry matter digestibility were determined for high protein (fish), plant (fresh leaves of Araceae, Moraceae and Bombaceae) and mixed (combination of both high protein and plant) diets. NDF values obtained for digestibility of the protein, plant and mixed diets were 1%, 63% and 49% respectively. There was a large amount of undigested plant material in the faeces, dominated by fruits of red mangrove (Rhizophora mangle). We considered the relationships between the type of food, IPT and apparent digestibility in the context of the nutritional contribution of an omnivorous diet. Our results suggest that for juvenile green sea turtles in the Eastern Pacific combined diets are an opportunistic strategy responding to habitat features and supply to maximise energy acquisition in transitional habitats such as Isla Gorgona.  相似文献

Specific biochemical marker-based techniques were tested for their ability to distinguish between seeds of Douglas-fir, Pseudotsuga menziesii (Mirbel) Franco, that were filled or unfilled (aborted) at maturity and those that were damaged or emptied by the western conifer seed bug, Leptoglossus occidentalis Heidemann. A polyclonal antibody raised against salivary gland extracts from L. occidentalis successfully identified residual salivary proteins on Western blots containing proteins from Douglas-fir seeds that had sustained various degrees of seed bug feeding damage. In a single blind experiment, the polyclonal antibody correctly identified 100% of undamaged control, 97% of unfilled control (aborted), and 98% of seed bug damaged seeds. Polyclonal antibodies raised against insoluble alfalfa crystalloid storage protein (11S globulin) detected the depletion of 11S globulin and the subsequent appearance of its hydrolyzed fragments in the soluble protein fraction of Douglas-fir seeds that were fed-upon by the seed bug. Feeding by L. occidentalis nymphs caused ca. 98% depletion of insoluble protein, but only ca. 53% reduction in the amount of soluble protein in seeds that appeared empty on radiographs. By comparison, unfilled (aborted) seeds contained significantly less insoluble and soluble protein than empty seeds that were fed-upon by L. occidentalis; moreover, no crystalloid (11S globulin) breakdown products were generated. The biochemical markers described in this study are reliable tools that can be used to identify conifer seeds that have sustained light to severe damage from L. occidentalis feeding.  相似文献

The aqueous extract of S. cumini or Eugenia jambolana seeds and Psidium guajava leaves showed higher inhibition against the porcine pancreatic alpha-amylase among the medicinal plants studied. The alpha-amylase inhibitors from S. cumini seeds were separated from the extract by preparative thin layer chromatography into fractions with different Rf values. The fraction with Rf value between 0.285 and 0.43, which showed maximum inhibitory activity, was eluted and analyzed through LC-MS. The compounds identified from the seed extract ofS. cumini were betulinic acid and 3,5,7,4'-tetrahydroxy flavanone, which were reported earlier from S. formosanum and other plants. Dixon plot showed that the inhibition was noncompetitive in nature.  相似文献

The aim of the present experiment was to determine if a niacin supplementation of 6 g/d to lactating dairy cow diets can compensate negative effects of a rumen nitrogen balance (RNB) deficit. A total of nine ruminally and duodenally fistulated lactating multiparous German Holstein cows were successively assigned to one of three diets consisting of 10 kg maize silage (dry matter [DM] basis) and 7 kg DM concentrate: Diet RNB- (n = 6) with energy and utilisable crude protein at the duodenum (uCP) according to the average requirement of the animals but with a negative RNB (-0.41 g N/MJ metabolisable energy [ME]); Diet RNB0 (n = 7) with energy, uCP and a RNB (0.08 g N/MJ ME) according to the average requirement of the animals and, finally, Diet NA (n = 5), which was the same diet as RNB-, but supplemented with 6 g niacin/d. Samples of milk were taken on two consecutive days, blood samples were taken on one day pre- and post-feeding and faeces and urine were collected completely over five consecutive days. The negative RNB reduced milk and blood urea content and apparent total tract digestibility of DM, organic matter (OM) and neutral detergent fibre (NDF). Also N excretion with urine, the total N excreted with urine and faeces and the N balance were reduced when the RNB was negative. Supplementation of niacin elevated plasma glucose concentration after feeding and the N balance increased. Supplementing the diet with a negative RNB with niacin led to a more efficient use of dietary N thereby avoiding the negative effects of the negative RNB on the digestibility of DM, OM and NDF.  相似文献

Pigs were fed diets containing 40% wheat bran incubated with a water.‐acetic acid mixture (control, C) and a cellulase (Cel‐i) or xylanase (Xyl‐i) preparation or with addition of the cellulase (Cel‐a) or xylanase (Xyl‐a) preparation immediately before feeding. Stomach and ileal samples were analysed for pH, osmolality, soluble saccharides, volatile fatty acids (VFA) and lactic acid. Incubation of wheat bran resulted in a small reduction of NDF and an increase in the amount of soluble starch, ß‐glucans and saccharides (glucose, xylose and arabinose), especially after incubation with the cellulase preparation. Two hours after feeding, significantly higher arabinose and xylose concentrations were present in the stomach for diets Cel‐i, Cel‐a and Xyl‐i. In the ileum xylose and arabinose concentrations were higher 2 to 4 and 6 to 8 hours after feeding the enzyme‐treated diets. In stomach and ileum there were no differences between the diets in pH, osmolality, VFA and lactic acid concentrations, but ileal VFA concentration from 4 h after feeding tended to be higher for diets Cel‐i and Xyl‐i. It can be concluded that the amount of soluble saccharides in stomach and small intestine and the ileal VFA concentration may be increased by cell wall degrading enzyme preparations.  相似文献

In children and adolescents from developed countries, obesity prevalence has strongly increased in the last decades and insulin resistance and impaired glucose tolerance are frequently observed. Some dietary components such as low glycemic index foods and dietary fibre could be used in order to improve glucose homeostasis in these children. Psyllium or ispaghula husk (the husk of the seeds of Plantago ovata) is a mixture of neutral and acid polysaccharides containing galacturonic acid with a ratio of soluble/insoluble fibre of 70/30. Some foods could potentially be enriched with psyllium, like breads, breakfast cereals, pasta and snack foods. The aim of this review was to assess the usefulness of psyllium in the management of obese children and adolescents with abnormalities of carbohydrate and lipid metabolism. After psyllium supplementation, the percentage change in postprandial glucose in type 2 diabetes patients, ranged from -12.2 to -20.2%. In hypercholesterolemic children, the effect of psyllium in LDL-cholesterol serum concentrations ranged from 2.78 to -22.8%; the effect in HDL-cholesterol from -4.16 to 3.05%; and the effect on triglycerides from 8.49 to -19.54%. The reviewed evidence seems to show that psyllium improves glucose homeostasis and the lipid and lipoprotein profile; however, more well controlled trials and further studies are needed to clarify it's effects and the mechanisms involved.  相似文献

Selected quality parameters were measured for forage leaf tissuefrom a spaced-plant nursery. The genotypes used were Ky 31 tallfescue and hybrids of Italian ryegrass (Lolium multiflorum Lam.)x tall fescue (Festuca arundinacea Schreb.) and tall fescuex giant fescue [Fescue gigantea (L ) Vill.]. Hybrid ploidy rangedfrom 2n = 28 to 84 chromosomes. Forage quality was characterizedby neutral detergent fibre (NDF), acid detergent fibre (ADF),total soluble carbohydrates (TSC) nutritive value index (NVI),hemicellulose, and in vitro dry matter disappearance (DMD). Quality of tall fescue, as measured by increased DMD, was improvedby hybridization with giant fescue. Improved DMD and NVI correlatedwith lower NDF and ADF in the hybrids. A few hybrids of Italianryegrass x tall fescue (2n = 28) were higher in some qualityparameters than Ky 31. Tall fescue x giant fescue hybrids (2n= 80 to 84), as a group, had significant quality improvementover Ky 31 in higher DMD and NVI and lower NDF and ADF. Whilesome individual hybrids within each group were significantlyhigher in quality, only the 2n = 80 to 84 chromosome group wasconsistently higher than Ky 31. Prediction equations for DMD,NDF, and ADF were established based on solvent extraction withnear-infrared reflectance spectroscopy (NIRS). Linear correlationcoefficients between chemical measurement and NIRS for eachquality parameter were 0–95 or higher.
