DNA克隆技术是分子生物学研究中一项重要的技术手段。自第一个质粒载体pSC1 0 1作为克隆载体以来 ,随着分子生物学技术的发展 ,克隆载体的整体结构、容载能力和转化效率都有了很大的改善。尤其是人类基因组计划的实施 ,产生了YAC和BAC克隆体系。随着植物基因组计划的进行 ,又产生了既能够克隆大片段DNA又能够将候选克隆直接通过农杆菌介导进行功能互补实验的载体。综述了几种常用大片段克隆载体YAC、BAC、BIBAC、PAC和TAC的特点及其应用 ,并对克隆载体的发展作了展望。 相似文献

A cDNA corresponding to the nitrate reductase (NR) gene from Dunaliella salina was isolated by RT-PCR and (5′/3′)-RACE techniques. The full-length cDNA sequence of 3,694 bp contained an open reading frame
of 2,703 bp encoding 900 amino acids, a 5′-untranslated region of 151 bp and a 3′-untranslated sequence of 840 bp with a poly
(A) tail. The putative gene product exhibited 78%, 65%, 59% and 50% identity in amino acid sequence to the corresponding genes
of Dunaliella tertiolecta, Volvox carteri, Chlamydomonas reinhardtii, and Chlorella vulgaris, respectively. Phylogenetic analysis showed that D. salina NR clusters together with known NR proteins of the green algae. The molecular mass of the encoded protein was predicted to
be 99.5 kDa, with an isoelectric point of 8.31. This protein shares common structural features with NRs from higher plants
and green algae. The full-length cDNA was heterologously expressed in Escherichia coli as a fusion protein, and accumulated to up to 21% of total bacteria protein. Recombinant NR protein was active in an enzyme
assay, confirming that the cloned gene from D. salina is indeed NR. 相似文献

systems. Following optimisation, one of the reporter genes can be replaced with a gene of interest; the second can be used
as a marker to confirm transgenic lines, and to estimate locus number and determine zygosity. This allows simple, efficient

Summary The Dutch potato cultivar Bintje has been transformed by Agrobacterium strain LBA1060KG, which contains two plasmids carrying three different DNAs (TL- and TR-DNA on the Agrobacterium rhizogenes plasmid and TKG-DNA on the pBI121 plasmid). Several transformed root clones were obtained after transformation of leaf, stem, and tuber segments, and plants were then regenerated from these root clones. The expression of the various marker genes [rol, opine, -glucuronidase (GUS), and neomycin phosphotransferase (NPTII)] was determined in several root clones and in regenerated plants. The selection of vigorously growing root clones was as efficient as selection for kanamycin resistance. In spite of the location of NPTII and GUS genes on the same T-DNA, 17% of the root clones did not show GUS activity. Nevertheless, Southern blot analysis showed that these root clones contained at least three copies of the GUS gene. Sixty-four per cent of the root clones contained opines. The expression of these genes, however, was negatively correlated with plant regeneration capacity and normal plant development. The differential expression of the marker genes in the transgenic potato tissues is discussed. 相似文献

In the present study, an efficient Agrobacterium-mediated gene transformation system was developed for ramie [Boehmeria nivea (L.) Gaud.] based on the examinations of several factors affecting plant transformation efficiency. The effects of Agrobacterium cell density, acetosyringone, co-cultivation temperature, co-cultivation duration, co-cultivation photoperiod and pH on stable
transformation were evaluated. Agrobacterium at a concentration of OD = 0.5–0.8 improved the efficiency of transformation. Concentration of acetosyringone at 50 mg/L
during co-cultivation significantly increased transformation efficiency. Co-cultivation at 20°C, in comparison to 15, 25 and
28°C, consistently resulted in higher transformation frequencies. A relatively short co-cultivation duration (3 days) was
optimal for ramie transformation. Co-cultivation medium at pH 5.9 and co-cultivation in darkness both improved the transformation
efficiencies of ramie. An overall scheme for producing transgenic ramie is presented, through which an average transformation
rate from 10.5 to 24.7% in five ramie varieties was obtained. Stable expression and integration of the transgenes were confirmed
by histochemical GUS assay, kanamycin painting assay, PCR and Southern blotting. This optimized transformation system should be employed for efficient
Agrobacterium-mediated transformation of ramie.

水稻高叶绿素含量基因DET1 cDNA的克隆、生物信息学分析及超表达载体的构建 总被引:1,自引:0,他引:1
提高水稻叶片中叶绿素含量可以提高光合作用效率。以水稻高叶绿素含量突变体为材料,在前期精细定位的基础上,克隆了高叶绿素含量基因DET1(DE-ETIOLATED 1)cDNA。测序结果显示:DET1基因全长cDNA为1536 bp,由11个外显子组成,编码512个氨基酸。与野生型珍汕97B相比,DET1基因在第7个外显子上有T-C的转换,该转换引入了一个EcoR I酶切位点,并导致第328位的亮氨酸(L)改变为丝氨酸(S),且突变位点处于高度保守区域内。DET1基因的氨基酸序列与玉米等其它高等植物的氨基酸序列同源性高达62%以上。利用生物信息学方法对DET1基因的序列、进化树、理化性质及亲疏水性等进行分析,并构建了水稻超表达载体pCUPHN-DET1,为水稻高叶绿素含量突变基因DET1的功能验证奠定基础。 相似文献

基因枪和农杆菌介导的遗传转化是目前常用的两种单子叶植物遗传转化方法。载体的发展和改良是提高植物遗传转化效率的重要基础,RNA干扰载体和过表达载体是目前通过遗传转化研究植物基因功能的主要工具。Gateway克隆技术是一种基于lambda噬菌体特异位点重组特性的通用克隆技术,该技术可以将大批目的基因方便、快捷地连接到受体载体上。本文利用Gateway技术结合传统酶切、连接方法,构建了适用于单子叶植物基因枪和农杆菌转化的RNA干扰Gateway载体pAHC-PSK-RNAi、pClean-G185-RNAi和过表达Gateway载体pAHC-PSK-OE和pClean-G185-OE,为利用基因枪和农杆菌介导的遗传转化,在小麦和水稻等单子叶植物中进行规模化基因功能研究奠定了基础。 相似文献

High-frequency transformation of maize (Zea mays L.) using standard binary vectors is advantageous for functional genomics and other genetic engineering studies. Recent advances
in Agrobacterium tumefaciens-mediated transformation of maize have made it possible for the public to transform maize using standard binary vectors without
a need of the superbinary vector. While maize Hi-II has been a preferred maize genotype to use in various maize transformation
efforts, there is still potential and need in further improving its transformation frequency. Here we report the enhanced
Agrobacterium-mediated transformation of immature zygotic embryos of maize Hi-II using standard binary vectors. This improved transformation
process employs low-salt media in combined use with antioxidant l-cysteine alone or l-cysteine and dithiothreitol (DTT) during the Agrobacterium infection stage. Three levels of N6 medium salts, 10, 50, and 100%, were tested. Both 10 and 50% salts were found to enhance
the T-DNA transfer in Hi-II. Addition of DTT to the cocultivation medium also improves the T-DNA transformation. About 12%
overall and the highest average of 18% transformation frequencies were achieved from a large number of experiments using immature
embryos grown in various seasons. The enhanced transformation protocol established here will be advantageous for maize genetic
engineering studies including transformation-based functional genomics. 相似文献

为了分离水稻的基因及其启动子,该实验室构建了T-DNA(GUS)结构的水稻启动子捕获系统,对其中编号为113#、T-DNA单拷贝插入、GUS报告基因为组成型表达的阳性捕获系进行了进一步分析。潮霉素筛选结合GUS组织化学染色获得了T-DNA插入的纯合株(113#-22和113#-26);Inverse法分离得到T-DNA插入位点水稻基因组DNA旁邻序列,测序和BLAST结果表明,T-DNA反方向插在水稻基因组4号染色体预测基因的内含子中;扩增T-DNA插入位点上游2kb左右DNA片段,构建启动子分析质粒转化水稻‘中花11’胚性愈伤组织,获得转基因植株,GUS组织化学染色模式与113#阳性株系一致。结果表明,该预测基因及其启动子是利用启动子捕获系统所捕获到的候选基因。 相似文献

To establish the overexpression and one-step purification system of Bacillus subtilis elongation factor-Tu (EF-Tu), the EF-Tu gene was amplified with or without own ribosome binding site (rbs) by PCR and the only PCR product without rbs was subcloned successfully. For the expression of the EF-Tu gene cloned after PCR amplification, a constitutive expression system and inducible expression system with His6 tag at N-terminus or C-terminus, or glutathione-S-transferase (GST) fusion system were examined in E. coli and B. subtilis. Except GST fusion system in E. coli, however, all other trials were unsuccessful at the step of plasmid construction for the EF-Tu expression. The GST/EF-Tu fusion proteins were highly expressed by IPTG induction and obtained as both soluble and insoluble form. From the soluble GST/EF-Tu fusion protein, EF-Tu was obtained to near homogeneity by one-step purification with glutathione-sepharose affinity column chromatography followed by factor Xa treatment. The purified EF-Tu showed high GDP binding activity. These results indicate that the GST/EF-Tu fusion system is favorable to overexpression and purification of B. subtilis EF-Tu. 相似文献

相邻的反向重复DNA片段有形成单链内二级结构的倾向,属于一种测序困难的DNA模板。解决RNAi载体插入的反向重复片段的测序问题,为该类载体正确性的测序验证奠定基础。采用常规分子克隆方法构建表达小麦TaATG2串联反向重复片段的RNAi载体,设计2种策略对经菌落PCR初步鉴定的载体进行测序验证:一种是以完整的载体质粒为模板进行测序;另一种是先对载体进行酶切处理,切除反向重复片段中的一个后对保留另一个片段的线性载体进行测序。结果表明,第一种测序策略受到串联反向重复片段形成的单链内部二级结构的影响,测序信号在反向重复片段处出现衰减或乱峰,无法读取序列。第二种测序策略排除了2个反向重复片段之间的干扰,保留在载体上的片段测序信号清晰,序列准确。采用酶切切除一个片段后进行测序的方法,经过2次酶切和2次测序可以有效地对载体上的2个反向重复片段分别进行序列测定,进而确认构建载体的正确性。 相似文献

treated with silicon carbide whiskers for 2 min in order to deliver pGreen0029 encoding GUS gene and pRG229 AVP1 gene, encoding Arabidopsis vacuolar pyrophosphatase, having neomycin phosphotransferaseII (nptII) genes as plant-selectable markers. Three crucial transformation parameters, i.e., callus type, days after subculture and
selection marker concentration for transformation of cotton calli were evaluated for optimum efficiency of cotton embryogenic
callus transformation giving upto 94% transformation efficiency. Within six weeks, emergence of kanamycin-resistant (kmr) callus colonies was noted on selection medium. GUS and Southern blot analysis showed expression of intact and multiple transgene
copies in the transformed tissues. Kanamycin wiping of leaves from T1, T2, and T3 progeny plants revealed that transgenes were inherited in a Mendelian fashion. Salt treatment of T1
AVP1 transgenic cotton plants showed significant enhancement in salt tolerance as compared to control plants. Thus far, this is

A genomic library of Zymomonas mobilis DNA was constructed in Escherichia coli using cosmid vector pHC79. Immunological screening of 483 individual E. coli strains revealed two clones expressing pyruvate decarboxylase, the key enzyme for efficient ethanol production of Z. mobilis. The two plasmids, pZM1 and pZM2, isolated from both E. coli strains were found to be related and to exhibit a common 4.6 kb SphI fragment on which the gene coding for pyruvate decarboxylase, pdc, was located.The pdc gene was similarily well expressed in both aerobically and anaerobically grown E. coli cells, and exerted a considerable effect on the amount of fermentation products formed. During fermentative growth on 25 mM glucose, plasmid-free E. coli lacking a pdc gene produced 6.5 mM ethanol, 8.2 mM acetate, 6.5 mM lactate, 0.5 mM succinate, and about 1 mM formate leaving 10.4 mM residual glucose. In contrast, recombinant E. coli harbouring a cloned pdc gene from Z. mobilis completely converted 25 mM glucose to up to 41.5 mM ethanol while almost no acids were formed. 相似文献

A thermophilic lipase of Bacillus thermoleovorans ID-1 was cloned and sequenced. The lipase gene codes 416 amino acid residues and contains the conserved pentapeptide Ala-X-Ser-X-Gly as other Bacillus lipase genes. The optimum temperature of the lipase is 75 degrees C, which is higher than other known Bacillus lipases. For expression in Escherichia coli, the lipase gene was subcloned in pET-22b(+) vector with a strong T7 promoter. London plane tree (Platanus acerifolia Willd.) is an important tree in urban landscaping but it suffers from a number of negative traits which genetic engineering
could be used to address. As with many woody species, P. acerifolia has appeared recalcitrant to genetic transformation. However, the recent development of a method for regenerating shoots
from P. acerifolia leaf explants suggests that such material could be a target for gene-transfer. Using an Agrobacterium tumefaciens strain in which the T-DNA carries the histochemically detected reporter gene β-glucuronidase (GUS), we have followed the
transfer of genes from Agrobacterium to leaf explants of Platanus acerifolia. Using this system, we have identified a set of inoculation and co-cultivation conditions (notably: the pre-treatment of
leaf explants with 0.4 M mannitol, an inoculation period of 10 min, a bacterial OD600 of 0.8–1.0 and a co-cultivation period of 5 days) that permit a good frequency and reliability of transient gene-transfer.
Optimum levels of antibiotics for bacterial elimination and kanamycin-resistant shoot regeneration were also established.
By applying these parameters, we recovered eight independent stably transformed shoots that were kanamycin-resistant and contained
the nptII T-DNA gene, as confirmed by PCR analysis. Furthermore, Southern blot analysis confirmed that, in at least five of these lines,
the transgene was associated with high molecular weight DNA, so indicating integration into the plant genome. 相似文献

Summary The rfb gene cluster of Salmonella typhimurium encodes the enzymes required for the biosynthesis of the O-Antigen. A part of it has been cloned in plasmid vectors pBR322 and pUC9 using an adjacent, previously cloned, part of the his operon (Barnes 1981) as a molecular probe for the first clone. A detailed restriction enzyme map of 7.57 kb of rfb DNA is presented and the approximate locations of two of the genes, rfbK and rfbM have been defined. 相似文献
