胚珠作为胚囊的携带者,在植物的生殖过程中起重要作用。胚珠是种子的前身,它在受精后发育成种子。近年来通过诱变已创造出一些胚珠和胚囊发育异常的突变体,如 sin1, bell, ovm2, ovm3。这几个突变体的表现型不但是珠被发育异常,而且胚囊不能形成或发育异常,最终结果是雌性不育。同时,已分别从蝶兰和矮牵牛的胚珠中分离出一批胚珠发育特异的基因,其中有关MADS Box基因在胚珠形成和发育中的作用研究得比较清楚,基因转化工作证实胚珠的分化和形成受一类新的MADS Box基因控制。  相似文献

拟南芥温度诱导脂质运载蛋白TIL1参与雌配子体发育   总被引:1,自引:0,他引:1  
雌配子体的正常发育是种子形成的前提条件之一,拟南芥温度诱导的脂质运载蛋白编码基因TIL1突变使胚珠败育,结实率下降明显。基因表达分析表明T-DNA插入使得TIL1基因敲除,突变体TIL1基因功能缺失;互交实验、Alexander染色、花粉离体培养和胚珠透明实验结果表明till-1突变体雄配子体发育正常、雌配子体胚囊发育有缺陷;通过遗传互补实验证明外源克隆的TIL1基因能恢复突变体的败育表型,并确定了TIL1基因主要在胚珠的胚囊中表达。实验结果表明TIL1基因参与了植物雌配子体发育这一重要的生理过程。  相似文献

应用陆地棉徐-142及其种子无短绒无长绒突变体(fl),对棉纤维的发育作了初步研究.扫描电子显微镜观察发现,fl突变体开花当天的胚珠基本没有突起,导致种子无毛.RT-PCR分析表明,在纤维起始期和延长期,野生型胚珠中E6和Expansin基因转录水平高,开花15d(DPA)前后达峰值,而在突变体胚珠中两个基因在整个发育期只有极低的表达.Cotmyb A是棉花Myb家族的一个成员,该基因在野生型胚珠发育过程中明显表达,但在fl胚珠中表达异常.在BT培养基中加入GA3,或IAA和GA3,可使离体fl胚珠部分地恢复纤维发生和生长的能力.此外,外源激素对培养的WT和fl胚珠中E6和Expansin基因表达的影响相似,而对Cotmyb A基因的转录几乎没有影响(这一点WT和fl胚珠也相似).结果提示,fl种子中E6,Expansin和CotmybA基因异常或极低水平的表达与种子无毛有关,产生这些异常的原因有待深入研究.  相似文献

对水稻HDAR胚胎发育过程的进一步研究表明,水稻HDAR中有2.72%(17/434)的不定胚发生和发育,其不定胚起源于胚珠内的球心细胞,不定胚起始细胞启动分裂时,胚囊发育至2核或4核时期,8核胚囊时期胚珠内已形成多细胞不定胚结构,随后不定胚细胞不断分裂并逐渐挤进胚囊,开花传粉后,不定胚利用胚乳提供的营养可以继续发育和分化,不定胚可以和合子胚一起发育,有时合子胚败育,不定胚继续发育并分化。讨论了水  相似文献

动脉平滑肌细胞(SMC)的增殖在动脉粥样硬化(AS)的形成过程中极其重要。我们在建立人主动脉SMC体外培养方法的基础上,观察了LDL,VLDL及HDL和相应的氧化修饰型脂蛋白对培养人SMCsis,jun,H-ras原癌基因及Rb抗癌基因转录表达的影响。结果表明:(1)HDL对SMCsis,jun,ras基因表达无影响;(2)LDL和VLDL有使这些基因表达增加的趋势;(3)ox-LDL,ox-VLDL和ox-HDL具有使SMCsis,jun,和ras基因表达显著增强的作用(P<0.01),且其作用较相应的天然脂蛋白大(P<0.01);(4)天然和氧化修饰型脂蛋白对Rb基因表达均无影响。据上述结果推测:LDL,VLDL,ox-LDL,ox-VLDL和ox-HDL的致AS作用可能与刺激SMCsis,jun和ras原癌基因表达增加有关。  相似文献

黄瓜花发育的早期均有雌蕊和雄蕊原基的分化,但在发育过程中,由于雌蕊或雄蕊的发育受到阻滞,导致雄花和雌花的形成。近年来在拟南芥和金鱼草等植物中遗传学的研究表明,花器官的特征是由同源异形基因决定的。在拟南芥中,由于AG在决定雄蕊和雌蕊特征方面起重要作用,本研究利用 RT-PCR技术,从黄瓜的雌、雄蕊中分离出 AG的同源基因,并对其在花发育过程中的表达和可能作用进行了分析。首先,根据AG同源基因的保守区域设计5’简并引物5’-GA(A/G)AT(T/C/A)AA(T/C/A)AA(G/A)(A/C)G(G/T/C)ATCGA(C/A)AAC-3’,然后进行3’,RA CE PCR,扩增出约1kb大小的片段,序列分析表明该片段含有非常保守的MADS box。进而,利用5’ RACE PCR得到全长度的cDNA。该cDNA的核苷酸序列与CUM1(黄瓜 MADS box gene1)同源性高达97%。 CUM1在接牵牛中过量表达可引起花萼变为心皮状和花瓣变为雄蕊,说明CUM1为AG的同源基因。基于该基因与CUM1序列上的高度同源,我们认为其为黄瓜的AG同源基因。该基因命名为CMB1,基因银行登记号为AF286649。Souther  相似文献

选用互为近等基因系的陆地棉品种徐州142和无絮棉突变体,比较研究开花前5d至开花后6d胚珠内可溶性蛋白质含量,POD和IAA氧化酶活性的变化,结果表明,开花前3d至1d蛋白质大量合成可能与纤维发育相关基因的表达有联系,而POD和IAA氧化酶阻碍纤维的初始发育。  相似文献

本文从质粒M13mp18-hBDNF中酶切回收人脑源性神经营养因子(hBDNF)全长基因,构建真核表达载体pCMV4-hBDNF。利用脂质体的方法转染COS7细胞,对转染后的COS7细胞提取RNA进行狭缝杂交分析和免疫细胞化学反应,分别从转录及翻译水平上检测BDNF基因在COS7细胞中的表达。实验还证实在COS7细胞中表达的hBDNF蛋白可分泌至胞外并可促进中脑黑质细胞的发育和生长,具有良好的生物学活性。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. 采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
