The oligosaccharides, sodium (methyl 3-deoxy-7-O-β-d-ribofuranosyl-β-d-manno-2-octulopyranosid)onate, methyl 2-O-β-d-ribofuranosyl-β-d-ribofuranoside, and the anomeric sodium [methyl 3-deoxy-7-O-(2-O-β-d-ribofuranosyl-β-d-ribofuranosyl)-α- and -β-d-manno-2-octulopyranosid]onate were prepared from 1-O-acetyl-2,3,5-tri-O-benzoyl-β-d-ribofuranose and the anomeric methyl (methyl 8-O-benzyl-4,5-O-carbonyl-3-deoxy-α- and -β-d-manno-2-octulopyranosid)onate in high purity and in acceptable over-all yields. They constitute a first series of model compounds for spectroscopic and immunochemical studies of the capsular polysaccharides from Escherichia coli strains LP 1092 and K 23. The essential, interglycosidic linkages [β-d-Ribf-(1→7)-α- or -β-d-dOclA, and β-d-Ribf-(1→2)-β-d-Ribf] were formed by a modification of the silver triflate procedure using appropriate d-ribofuranosyl bromide derivatives. The constitutional and configurational assignments were based on the 250-MHz 1H-n.m.r.-spectra of protected derivatives of the oligosaccharides.  相似文献

A d-galacto-d-mannan ([α]D +72.0 and d-galactose-to-d-mannose ratio 1:1.14) was isolated from the seeds of Melilotus indica All., syn. M. parviflora Desf. The 1H- and 13C-n.m.r., and i.r. spectra indicated the presence of α-d-galactopyranosyl and β-d-mannopyranosyl residues. Methylation of the polysaccharide, followed by hydrolysis, afforded, 2,3,4,6-tetra-, 2,3,6-tri-, 2,3-di-, and 3,4-di-O-methyl-d-mannose, and 2,3,4,6-tetra- and 2,3,6-tri-O-methyl-d-galactose in the molar ratios of 1:2:22:6:27:3. Periodate oxidation of the polysaccharide, followed by reduction and hydrolysis, gave erythritol (1 mol) and glycerol (1.24 mol). Partial acid hydrolysis of the polysaccharide afforded O-β-d-mannopyranosyl-(1→2)-d-mannopyranose, O-β-d-mannopyranosyl-(1→4)-d-mannopyranose, O-α-d-galactopyranosyl-(1→6)-d-mannopyranose, O-α-d-galactopyranosyl-(1→4)-d-galactopyranose, and O-α-d-galactopyranosyl-(1→6)-O-β-d-mannopyranosyl-(1→4)-d-mannopyranose. A highly branched structure having a mannan backbone composed of 36% of (1→4)- and 10% of (1→2)-linked β-d-mannopyranosyl units is proposed for the galactomannan.  相似文献

Partial, acid hydrolysis of the extracellular polysaccharide from Xanthomonas campestris gave products that were identified as cellobiose, 2-O-(β-d-glucopyranosyluronic acid)-d-mannose, O(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-d-glucose, O-(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-[O-β-d-glucopyranosyl-(1→4)]-d-glucose, and O-(β-d-glucopyranosyluronic acid)-(1→2)-O-α-d-mannopyranosyl-(1→3)-[O-β-d-glucopyranosyl-(1→4)-O-β-d-glucopyranosyl-(1→4)-d-glucose. This and other evidence supports the following polysaccharide structure (1) which has been proposed independently by Jansson, Kenne, and Lindberg:

The structure of the antitumor polysaccharide from the actinomycete Microellobosporia grisea has been investigated. By methylation and periodate-oxidation studies, the polysaccharide was shown to consist of (nonreducing)d-mannosyl groups, (1→4)-linkedd-glucosyl residues, and 3,6-branched, (1→4)-linkedd-glucosyl residues in the approximate molar ratios of 2:1:1. Periodate oxidation of the polysaccharide, followed by borohydride reduction and mild hydrolysis with acid yielded glycerol, erythritol, 2-O-β-d-glucopyranosyl-d-erythritol, and 5-O-β-d-glucopyranosyl-2,4-bis(hydroxymethyl)-1,3-dioxane, which were isolated in the molar ratios of 2.0:0.14:0.74:0.35. Partial hydrolysis of the polysaccharide gave α-d-Man p-(1→6)-d-Glcp, β-d-Glcp-(1→4)-d-Glcp, α-d-Man p-(1→3)-d-Glcp, and β-d-Glcp-(1→4)-[α-d-Man p-(1→3)-]-d-Glcp. From these results, it is proposed that the polysaccharide is mainly composed of tetrasaccharide repeating-units having the following structure.  相似文献

Marine sponges are ancient and simple multicellular filter-feeding invertebrates attached to solid substrates in benthic habitats and host a variety of fungi both inside and on their surface because of its unique ingestion and digest system. Investigation on marine sponge-associated fungi mainly focused on the small molecular metabolites, yet little attention had been paid to the extracellular polysaccharides. In this study, a homogeneous extracellular polysaccharide AS2-1 was obtained from the fermented broth of the marine sponge endogenous fungus Alternaria sp. SP-32 using ethanol precipitation, anion-exchange, and size-exclusion chromatography. Results of chemical and spectroscopic analyses showed that AS2-1 was composed of mannose, glucose, and galactose with a molar ratio of 1.00:0.67:0.35, and its molecular weight was 27.4 kDa. AS2-1 consists of a mannan core and a galactoglucan chain. The mannan core is composed of (1→6)-α-Manp substituted at C-2 by (1→2)-α-Manp with different degrees of polymerization. The galactoglucan chain consists of (1→6)-α-Glcp residues with (1→6)-β-Galf residues attached to the last glucopyranose residue at C-6. (1→6)-β-Galf residues have additional branches at C-2 consisting of disaccharide units of (1→2)-β-Galf and (1→2)-α-Glcp residues. The glucopyranose residue of the galactoglucan chain is linked to the mannan core. AS2-1 possessed a high antioxidant activity as evaluated by scavenging of 1,1-diphenyl-2-picrylhydrazyl and hydroxyl radicals in vitro. AS2-1 was also evaluated for cytotoxic activity on Hela, HL-60, and K562 cell lines by the MTT and SRB methods. The investigation demonstrated that AS2-1 was a novel extracellular polysaccharide with different characterization from extracellular polysaccharides produced by other marine microorganisms.  相似文献

The major structural component of the mycobacterial cell wall, the mycolyl–arabinogalactan–peptidoglycan complex, possesses a galactan core composed of approximately 30 galactofuranosyl (Galf) resides attached via alternating β-(1→6) and β-(1→5) linkages. Recent studies have shown that the entire galactan is synthesized by two bifunctional galactofuranosyltransferases, GlfT1 and GlfT2. We report here saturation transfer difference (STD) NMR studies GlfT2 using two trisaccharide acceptor substrates, β-d-Galf-(1→6)-β-d-Galf-(1→5)-β-d-Galf-O(CH2)7CH3 (2) and β-d-Galf-(1→5)-β-d-Galf-(1→6)-β-d-Galf-O(CH2)7CH3 (3), as well as the donor substrate for the enzyme, UDP-Galf. Epitope mapping demonstrated a greater enhancement toward the ‘reducing’ ends of both trisaccharides, and that UDP-galactofuranose (UDP-Galf) made more intimate contacts through its nucleotide moiety. This observation is consistent with the greater flexibility required within the active site of the reaction between the growing polymer acceptor and the UDP-Galf donor. The addition of UDP-Galf to either 2 or 3 in the presence of GlfT2 generated a tetrasaccharide product, indicating that the enzyme was catalytically active.  相似文献

Partial hydrolysis with acid, methylation analysis (including uronic acid degradation), Smith degradation, and p.m.r. spectroscopy have been used to determine the primary structure of the capsular polysaccharide of Klebsiella serotype k64. The hexasaccharide repeating-unit, which also contains one O-acetyl substituent, comprises a 4)-α-d-GlcpA-(1 → 3)-α-d-Manp-(1 → 3)-β-d-Glcp-(1 → 4)-α-d-Manp-(1 → chain with a 4,6-O-(l-carboxyethylidene)-β-d-glucopyranosyl and an l-rhamnosyl group attached to the 4-linked d-mannosyl residue at O-2 and O-3, respectively.  相似文献

BackgoundXylan is the second most abundant plant cell wall polysaccharide after cellulose with α-L-arabinofuranose (L-Araf) as one of the major side substituents. Capacity to degrade xylan is characteristic of many plant pathogens; and corresponding enzymes that debranch arabinoxylan provide tools to tailor xylan functionality or permit its full hydrolysis.MethodThree GH62_2 family α-arabinofuranosidases (Abfs) from plant pathogenic fungi, NhaAbf62A from Nectria haematococca, SreAbf62A from Sporisorium reilianum and GzeAbf62A from Gibberella zeae, were recombinantly produced in Escherichia coli. Their biochemical properties and substrate specificities were characterized in detail. Particularly with 1H NMR, the regioselectivity and debranching preference of the three Abfs were directly compared.ResultsThe activities of selected Abfs towards arabinoxylan were all optimal at pH 6.5. Their preferred substrates were wheat arabinoxylan, followed by soluble oat spelt xylan. The Abfs displayed selectivity towards either α-(1 → 2) or α-(1 → 3)-L- Araf mono-substituents in arabinoxylan. Specifically, SreAbf62A and GzeAbf62A removed m-α-(1 → 3)-L-Araf and m-α-(1 → 2)-L-Araf substituents with a similar rates, whereas NhaAbf62A released m-α-(1 → 3)-L-Araf 1.9 times faster than m-α-(1 → 2)-L-Araf.Major conclusionsBuilding upon the known selectivity of GH62 family α-arabinofuranosidases towards L-Araf mono-substituents in xylans, the current study uncovers enzyme-dependent preferences towards m-α-(1 → 3)-L-Araf and m-α-(1 → 2)-L-Araf substitutions. Comparative sequence-structure analyses of Abfs identified an arginine residue in the xylose binding +2R subsite that was correlated to the observed enzyme-dependent L-Araf debranching preferences.General significanceThis study expands the limited pool of characterized GH62 Abfs particularly those from plant pathogenic fungi, and provides biochemical details and methodology to evaluate regioselectivity within this glycoside hydrolase family.  相似文献

The structure of the capsular polysaccharide elaborated by Haemophilus influenzae type d has been investigated, methylation analysis and n.m.r. spectrometry being the principal methods used. It is concluded that the polysaccharide is composed of repeating units having the structure: →4)-β-d-GlcpNAc-(1→3)-β-d-ManpNAcA-(1→. In addition, single residues of l-alanine, l-serine, or l-threonine, in the proportions 2:2:1, are linked, through their amino groups, to C-6 of the 2-acetamido-2-deoxy-β-d-mannopyranosyluronic acid residues. Human antithrombin III contains four asparagine-linked sugar chains in one molecule. The sugar chains were quantitatively released as radioactive oligosaccharides from the polypeptide portion by hydrazinolysis followed by N-acetylation and NaB3H4 reduction. All of the oligosaccharides, thus obtained, contain N-acetylneuraminic acid. A same neutral nonaitol was released from all acidic oligosaccharides by sialidase treatment. A unique, alkali-soluble polysaccharide has been isolated from the cell walls of the basidiomycete Coprinus macrorhizus microsporus. The polysaccharide, which is primarily a glucan, contains a large proportion of α-(1→4)-linked d-glucose residues and a smaller amount of β-(1→3) and (1→6) linkages, as suggested by methylation, partial acid hydrolysis, periodate oxidation, and enzymic studies. Hydrolysis of the methylated polysaccharide gave equimolar amounts of 2,4-di- and 2,3-di-O-methyl-d-glucose; no 2,6-di-O-methyl-d-glucose was identified, indicating the absence of branch points joined through O-1, O-3, and O-4. The isolation and identification of 2-O-α- glucopyranosylerythritol from the periodate-oxidized polysaccharide suggests that segments of the a-(1→4)-linked d-glucose residues are joined by single (1→3)-linkages. An extracellular enzyme-preparation from Sporotrichum dimorphosporum (QM 806) containing both β-(1→3)- and α-(1→4)-d-glucanohydrolase activity released 76% of the reducing groups from the polysaccharide. The polysaccharide also contains minor proportions of xylose, mannose, 2-amino-2-deoxyglucose, and amino acids.  相似文献

An arabinoxylan isolated from the bark of Cinnamomum zeylanicum was composed of l-arabinose and d-xylose in the molar ratio 1.6:1.0. Partial hydrolysis furnished oligosaccharides which were characterised as α-d-Xylp-(1→3)-d-Ara, β-dXylp-(1→4)-d-Xyl, β-d-Xylp-(1→4)-β-d-Xylp-(1→4)-d-Xyl, β-d-Xylp-(1→4)-β-d-Xylp-(1→4)-β-d-Xylp-Xylp-(1→4)-d-Xyl, xylopentaose, and xylohexaose. Mild acid hydrolysis of the arabinoxylan gave a degraded polysaccharide consisting of l-arabinose (8%) and d-xyolse (92%). Methylation analysis indicated the degraded polysaccharide to be a linear (1→4)-linked d-xlan in which some xylopyranosyl residues were substituted at O-2 or O-3 with l-arabinofuranosyl groups. These data together with the results of methylation analysis and periodate oxidation of the arabinoxylan suggested that it contained a (1→4)-linked β-d-xylan backbone in which each xylopyranosyl residue was substituted both at O-2 and O-3 with l-arabinofuranosyl, 3-O-α-d-xylopyranosyl-l-arabinofuranosyl, and 3-O-l-arabinofuranosyl-l-arabinofuranosyl groups.  相似文献

After partial, acid hydrolysis of the extracellular, acid polysaccharide from Rh. trifolii Bart A, the following products were isolated and characterized: 3,4-O-(1-carboxyethylidene)-d-galactose, 4,6-O-(1-carboxyethylidene)-d-galactose, 3-O-[3,4-O-(1-carboxyethylidene)-β-d)-galactopyranosyl]-d-glucose, 3-O-[4,6-O-(1-carboxyethylidene)-β-d-galactopyranosyl]-d-glucose, O-[3,4-O-(1-carboxyethylidene)-β-d-galactopyranosyl ]-(1→3)-O-d-glucopyranosyl-(1→4)-d-glucose, and O-[4,6-O-(1- carboxyethylidene)-β-d-galactopyranosyl]-(1→3)-O-β-d-glucopyranosyl-(1→4)-d-glucose. The presence of pyruvic acid linked either to O-3 and O-4 or to O-4 and O-6 of the d-galactopyranosyl group of these saccharides indicates that both structures may be present in the original polysaccharide.  相似文献

Haemophilus influenzae type b capsular polysaccharide [repeating unit, →3)-β- d-Ribf-(1 → 1)- d-Ribol-5-(PO 2H→] was partially hydrolyzed with HCl to give oligosaccharides that were isolated by size-exclusion chromatography, and then characterized by 31P- and 13C-n.m.r.-spectral and chemical methods, in order to determine the end-group composition and, hence, the number-average chain-length がL. The ratio (~17:8) of monophosphate end-groups to d-ribofuranose end-groups revealed the relative rates of hydrolysis of the phosphoric diester linkage and the glycosidic linkage in the repeating-unit structure. Cleavage of the phosphoric diester linkage was ~92% regioselective, as indicated by the ~ 12:1 ratio of d-ribofuranose monophosphate end-groups to d-ribitol monophosphate end-groups. The n.m.r. spectra of the oligosaccharide repeating-unit provided evidence for partial stereomutation (~3–8%) that involved rearrangement of the d-ribofuranose phosphoric diester linkage and anomerization at C-1 of d-ribofuranose. Variously sized oligosaccharides (がL = 4, 7, and 12) that had d-ribofuranose end-groups reacted with bovine serum albumin that had an average of ~9 adipyl hydrazide functionalities, to give, within experimental error, quantitative yields of the corresponding, hydrazone-linked, oligosaccharide-protein conjugates.  相似文献

Mixed-linkage (1→3),(1→4)-β-d-glucan (MLG) is a biologically and technologically important hemicellulose, known to occur in three widely separated lineages: the Poales (including grasses and cereals), Equisetum (fern-allies), and some lichens e.g. Iceland moss (Cetraria islandica). Lichenase (E.C. 3.2.1.73) is widely assumed to hydrolyse all (1→4) bonds that immediately follow (1→3) bonds in MLG, generating predominantly the tetrasaccharide β-d-Glcp-(1→4)-β-d-Glcp-(1→4)-β-d-Glcp-(1→3)-d-Glc (G4G4G3G; MLG4), the corresponding trisaccharide (G4G3G; MLG3), and sometimes also laminaribiose (G3G; MLG2). The ratio of the oligosaccharides produced characterises each polysaccharide. We report here that digestion of MLG from barley (Hordeum vulgare), Equisetum arvense and C. islandica by Bacillus subtilis lichenase also yields the unexpectedly stable hexasaccharide, β-d-Glcp-(1→3)-β-d-Glcp-(1→4)-β-d-Glcp-(1→4)-β-d-Glcp-(1→4)-β-d-Glcp-(1→3)-d-Glc (G3G4G4G4G3G, i.e. MLG2–MLG4), identified by thin-layer chromatography, gel-permeation chromatography, HPLC (HPAEC), β-glucosidase digestion, 1H/13C-NMR spectroscopy and mass spectrometry. On HPLC, G3G4G4G4G3G is the major constituent of a peak previously ascribed solely to the nonasaccharide G4G4G4G4G4G4G4G3G. Because it was widely presumed that lichenase would cleave G3G4G4G4G3G to MLG2 + MLG4, our data both redefine the substrate specificity of Bacillus lichenase and show previous attempts to characterise MLGs by HPLC of lichenase-digests to be flawed. MLG2 subunits are particularly underestimated; often reported as negligible, they are here shown to be an appreciable constituent of MLGs from all three lineages. We also show that there is no appreciable yield of water-soluble lichenase products with DP > 9; potential identities of products previously labelled DP > 9 are suggested. Finally, this discovery also provides a opportunity to investigate the spatial distribution of subunits along the MLG chain. We show that MLG2 subunits in barley and Cetraria MLG are not randomly distributed, but predominantly found at the non-reducing end of MLG4 subunits.  相似文献

The linkage pattern of the K6-antigen was investigated using material from the urinary pathogen, Escherichia coli LP 1092. The polysaccharide consists of ribose and 3-deoxy-D-manno-2-octulosonate (KDO) in a ratio of 2:1. Colorimetric procedures, Smith degradation, methylation analysis, and nuclear magnetic resonance spectroscopy were applied to the whole polysaccharide and to a trisaccharide “repeating unit” obtained by mild-acid catalyzed hydrolysis. The capsular polysaccharide from Klebsiella K44 has been investigated by the techniques of methylation, base-catalyzed elimination, Smith degradation, and partial hydrolysis. The last-named yielded an oligosaccharide corresponding to one repeating unit. The anomeric configutations of the sugar residueswere determined by 1H- and 13C-n.m.r. spectroscopy.
