Good quality of antibiotic discs is a fundamental prerequisite to accurate antibiotic susceptibility tests. Capillary electrophoresis (CE) is a widely used method for quantitative analysis. Here, using ceftazidime as an example, we report an easy-to-perform strategy to determine ceftazidime content in discs. First, a serial of ceftazidime standard solutions was prepared to determine the detection linearity. Utilizing the background buffer containing 50 mmol/L Na2HPO4, 0.045 mmol/L beta-cyclodextrin and 3.15 mmol/L Tris (hydroxymethyl) aminomethane, ceftazidime of concentrations ranging from 1.875 to 60 microg/ml showed a linear response to detected peak areas. Subsequently, four kinds of ceftazidime discs were selected from different manufacturers. The discs were homogenized using 1 ml deionized water, and then detected by CE after filtration. The results showed that one kind of discs were of poor quality as further confirmed by disc diffusion tests using standard strains. This study proved the potential of CE as an easy choice to perform disc quality control under appropriate conditions.  相似文献

The objective of this study was to quantitatively analyze the effect of diurnal loading on the transport of various charged antibiotics into negatively charged human intervertebral disc (IVD). Transport of charged antibiotics into a human lumbar disc was analyzed using a 3D finite element model. The valence (z) of the electrical charge of antibiotics varied from z = +2 (positively charged) to z = −2 (negatively charged). An uncharged antibiotic (z = 0) was used as a control. Cases with transient antibiotic concentration at disc boundaries [to mimic intravenous (IV) infusion] were simulated. Our results showed that diurnal compression increased the concentrations in the nucleus pulposus (NP) region, but degreased the concentrations in the annulus fibrosus (AF) region for all charged or non-charged drugs. The overall concentration (averaged over disc) increased with diurnal compression. The diurnal compression had more effects on negatively charged antibiotics than positively charged ones. For example, at day 5 with diurnal compression, the diurnal compression increased the concentration of negatively charged drug (z = −1) in NP by 18.3%, but only by 6.6% for positively charged one (z = +1). In AF, diurnal compression decreased the concentration by 13.2% for negatively charged drug (z = −1) versus 1.2% for positively charged one (z = +1). Note these percentages are the averaged values over day 5. This study provides quantitative information on understanding the mechanisms of charged drug transport in human IVDs.  相似文献

We have established a primary culture system for Drosophila eye imaginal discs. With this system, we were able to obtain neurite outgrowth from intact eye discs, eye disc fragments, and dissociated eye imaginal disc cells. Immunoreactivity to antibody 24B10 indicates that these extending neurites are photoreceptor axons. Three culture media were tested for their ability to support the survival of and neurite extension from eye disc fragments in vitro at 23°C. These, with supplements, were: five parts of Schneider's Drosophila medium with four parts of basal Eagle's medium (“4+5”); Leibovitz's L-15 medium (L-15); and Shields and Sang's M3 modified medium (MM3). We obtained the best results with MM3 supplemented with 2% fetal bovine serum (FBS). Eye disc fragments survived in this medium for at least 20 days. Pigmentation in the nonphotoreceptor pigment cells in cultures from the prepupa required the presence of 20-hydroxyecdysone (20-HE) (1 μg/ml), whereas neurite outgrowth was seen in the absence of 20-HE. Donor animals had to fall within a range of ages to obtain appropriate eye disc differentiation in vitro. Eye discs from 5-h pupae (P+5) or older commenced ommachrome synthesis in vitro, in a temporal sequence close to that found in vivo, whereas the in vitro synthesis of this pigment was delayed in eye discs from younger flies. Average neurite length was not affected by age among pupae younger than P+5; but neurite outgrowth from P+24 was scarce, probably because by this time photoreceptor axons had already grown in vivo and were severed and unable to regenerate in vitro. Eye discs taken from third instar larvae or white prepupae continued their mitotic activity in vitro. Together with the advance of the morphogenetic furrow at the leading edge of retinal development, this observation is consistent with the evidence that pattern formation continues in vitro. Morphogenetic changes were manifested in cultures. Viability tests with calcein AM and ethidium bromide revealed few dead cells in living cultures. © 1995 John Wiley & Sons, Inc.  相似文献

AIMS: To investigate the influence of the culture medium on antibiotic susceptibility testing of food-associated lactic acid bacteria (LAB) with the agar overlay disc diffusion (DD) method. METHOD: The antibiotic resistance profile of 39 food-associated lactobacilli and enterococci was determined with the agar overlay DD method using a defined medium (i.e. Iso-sensitest agar; ISA) or an undefined medium (i.e. de Man, Rogosa, Sharpe or MRS agar). RESULTS: The study revealed that ampicillin discs and, although to a lesser extent, also tetracycline discs consistently produced larger zones on MRS medium compared to ISA medium. For the antibiotics gentamicin, bacitracin and erythromycin, the radius of the inhibition zones produced on MRS medium was significantly smaller in relation to ISA. For categorizing LAB isolates into resistant, intermediate and susceptible groups, it was demonstrated that major errors can occur in determining bacitracin and gentamicin resistance if MRS medium instead of ISA medium is used. On the other hand, the performance of both media was found to be equivalent for testing tetracycline resistance. CONCLUSIONS, SIGNIFICANCE AND IMPACT OF THE STUDY: Despite the fact that MRS medium generally supports the growth of lactic acid bacteria much better than the nutrient-poor ISA medium, the present study clearly demonstrates that both media are not compatible in susceptibility testing against various classes of antibiotics. These results may stimulate future discussions on a generally recommended DD method for susceptibility testing of food LAB strains.  相似文献

Summary New cell lines, designated as ML-DmDl≈10, were established from dissociated imaginal discs ofDrosophila melanogaster. The culture medium was prepared by mixing in a 1:1 ratio Cross and Sang’s M3(BF) medium, supplemented with 10% heat inactivated fetal bovine serum (FBS), with the supernatant of a primary embryonic cell culture made in the M3(BF) medium and supplementing this mixture with insulin. One cell line was established in the medium containing larval hemolymph instead of the primary culture supernatan, and another was established in fresh M3(BF) medium supplemented with insulin and FBS. In these mediums, imaginal disc cells first formed aggregates and cellular vesicles within a few weeks followed by the proliferation of thin-layered cells around them after about 1 mo. Ten cell lines have so far been established from two kinds of imaginal discs and disc mixtures. The ploidy of these cell lines was predominantly diploid. Population doubling time was about 50 to 70 h at 3 to 10 mo. after initiation of the culture. When the cell aggregates formed in vitro were implanted in metamorphosing larvae, they differentiated at high frequency into adult cuticular strutures in the early phase of the primary culture. This differentiation of aggregates was also observed, though at low frequency, in a culture maintained by dilution-transfer for 6 to 15 mo. in vitro.  相似文献

The aim of the study was to assess prevalence and antibiotic susceptibility profiles ofGram-negative strictly anaerobic bacteria isolated from clinical specimens taken from hospitalized patients in 2005-2006. Biochemical identification and antibiotic susceptibility were done in an automated system ATB Expression (bioMerieux sa). From 12262 specimens examined 867 strains of obligate anaerobes were isolated. Gram-negative strictly anaerobic bacteria were cultured in number of 138 strains (15,9%). All cultures were performed on Columbia agar and Schaedler agar media (bioMerieux sa) supplemented with 5% sheep blood and incubated at 37 degrees C for 48-120 h in 85% N2, 10% H2, 5% CO2. Most frequently isolated was Bacteroides spp. (41,3%). For this group beta-lactamase activity was evaluated by using nitrocefin disc test (Cefinase BBL, Becton Dickinson and Co., Cockeysville, MD, USA). Production of ESBLs was detected with the use of two disc diffusion methods: the double-disc synergy test (DDST) according to Jarlier et al. and the diagnostic disc (DD) test according to Appleton. ESBLs were produced by 5,3% strains of Bacteroides spp. For all Bacteroides spp. strains MIC values were determined by gradient diffusion method Etest (AB BIODISK, Sweden). ESBLs and MIC were performed on Wilkins-Chalgren solid medium supplemented with 5% sheep blood (Difco Lab., USA) and all plates were incubated at 35 degrees C for 48 hours in 85% N2, 10% H2, 5% CO2. Most Gram-negative obligate anaerobes isolated from clinical specimens are still susceptible to imipenem (100%), metronidazole (99,3%) and beta-lactam antibiotics with beta-lactamase inhibitors: piperacillin/tazobactam (99,3%), ticarcillin/clavulanate (99.3%), amoxicillin/clavulanate (97.8%).  相似文献

We are investigating the role of extracellular proteases in imaginal disc eversion to understand the mechanism that controls cell rearrangements within epithelia. We have identified three cation-dependent neutral proteases released by Drosophila leg discs everting in culture. Serine protease inhibitors block disc eversion and inhibit activity of disc proteases. The pattern of extracellular proteases changes when eversion is blocked with added protease inhibitors. Changes in protease activity occur when released disc proteases are treated with trypsin. Trypsin treatment of intact imaginal discs releases protease and inhibitor activities to the medium, indicating their presence on the cell surface before release. Our results suggest that extracellular proteases are required for imaginal disc morphogenesis and are regulated by more than one mechanism.  相似文献

Analysis of tetracycline and erythromycin diagnostic discs manufactured in the USSR showed their conformity with the requirements of the USA Federal Register. Comparison of the antibiotic amounts extracting and diffusing from the discs showed that as an average 92 and 90 per cent of the extracted amounts of erythromycin and tetracycline respectively diffused into the agar. Subsequently, it is desirable that on control testing of the disc quality both the similarity of the antibiotic content in the discs and the real amount of the antibiotic diffusing into the agar should be considered. A statistically reliable correlation between the values of the growth inhibition zones around the discs with definite and constant amounts of erythromycin, tetracycline or oxacillin and different resistance levels for every staphylococcal strain was found. On the basis of such a control system it is possible to divide the staphyloccal strains into the groups with high, low and intermediate resistance levels to the above antibiotics. However, it is not possible to use such data for accurate calculation of the value of the minimum inhibitory concentration of unknown strains because of a high value of the main error.  相似文献

The transport of oxygen and lactate (i.e., lactic acid) in the human intervertebral disc was investigated accounting for the measured coupling between species via the pH level in the tissue. Uncoupled cases were also analyzed to identify the extent of the effect of such coupling on the solute gradients across the disc. Moreover, nonlinear lactic production rate versus lactic concentration and oxygen consumption rate versus oxygen concentration were considered. The nonlinear coupled diffusion equations were solved using an in-house finite element program and an axisymmetric model of the disc with distinct nucleus and anulus regions. A pseudotransient approach with a backward integration scheme was employed to improve convergence. Coupled simulations influenced the oxygen concentration and lactic acid concentration throughout the disc, in particular the gradient of concentrations along the disc mid-height to the nucleus-anulus boundary where the solutes reached their most critical values; minimum for the oxygen tension and maximum for the lactate. Results suggest that for realistic estimates of nutrient and metabolite gradients across the disc, it could be important to take into account the coupling between the rates of synthesis and overall local metabolite/nutrient concentration.  相似文献

The antibiotic resistance profiles of 75 Campylobacter isolates of food and human clinical origin was determined by two agar diffusion susceptibility methods; disc diffusion and epsilometer-test (E-test). The most common therapeutic antimicrobials, erythromycin, ciprofloxacin and tetracycline were studied, along with chloramphenicol, ampicillin and naladixic acid. The resistance observed for each antimicrobial, as determined by both of methods, were statistically compared using Fisher two-tailed analysis.Of the six antimicrobials studied only two were shown to have statistically different patterns when resistance was compared by disc diffusion and E-test. The percentage of isolates resistant to clinically relevant antimicrobials using both techniques ranged from 6.6 to 21.3% for erythromycin, 25.3–26.6% for tetracycline and 33.3–36.0% for ciprofloxacin. The prevalence of multi-drug resistant (MDR) campylobacters (isolates resistant to 2 or more antimicrobials) for both disc diffusion and E-test was 44%. It can be concluded that, for four of the six antimicrobials assessed, antimicrobial resistance prevalences could be equally determined by either of the methods studied.  相似文献

Electrical and tracer techniques were used to investigate the movement of Na+, K+ and Cl? through discs of a range of thicknesses cut from the root tissue of sugar beet, Beta vulgaris L. cv. Amono. At low external concentration the electrical resistance across a discs is less than that of an equivalent volume of solution. This does not appear to be due to a low resistance symplastic pathway but rather, to an enhanced concentration of cation in the apoplast. The resistance is proportional to the thickness of tissue. Although measurement of diffusion potential gives about 25 mV difference across the disc for a ten-fold change in cation concentration, there is little discrimination between K+ and Na+. The observed tracer kinetics of 86Rb through the disc are consistent with those of diffusion, with a coefficient of diffusion, D, of 0.19 × 10?9 m2 s?1 and a tissue partition coefficient, k, of 0.27 (or of 2.7 if referred to the cell wall phase only). 36Cl gives a similar value for D, but has a k reduced by a factor of 3.3, a result that is consistent with the diffusion potential observation. However, a much larger discrimination would have been expected from the chemically measured cation exchange capacity.  相似文献

One hundred and forty isolates of beta-hemolytic streptococcus cultured from patients with clinical pharyngitis were studied by disc diffusion for antibiotic sensitivity to lincomycin, erythromycin, cephalexin and penicillin and by agar dilution to cephalexin and penicillin. All isolates were sensitive to ≤ 0.1 μg./ml. penicillin and ≤ 1.56 μg./ml. cephalexin. The disc-diffusion test was reliable in predicting the sensitivities in vitro. One strain of group A betahemolytic streptococcus was resistant to erythromycin by disc diffusion. When compared to Lancefield grouping 18% of strains were incorrectly identified as group A by the bacitracin-disc test. Cephalexin was uniformly effective in vitro in inhibiting beta-hemolytic streptococci and the 30 μg. cephalexin disc was reliable in predicting these sensitivities.  相似文献

Echinomycin is a peptide antibiotic of the quinoxaline group produced by Streptomyces echinatus. In propitious circumstances it can be determined by ultraviolet spectrophotometry, but for work with analogues and congeners a more specific assay is required. A disc agar diffusion technique has been developed and optimised for this purpose. It yields log dose-response curves which are linear over at least a 20-fold range of antibiotic concentration, and the variation of sensitivity with inoculum size, preincubation time, and temperature of incubation has been investigated. Other quinoxaline antibiotics can be assayed by the same technique; they yield quite different log dose-response curves indicative of lower potency associated with triostins as compared to quinomycins. The application of the technique to follow echinomycin production by S. echinatus A8331 in culture in maltose minimal medium is described.  相似文献

Patterns of polypeptide synthesis in wing, leg and eye-antenna imaginal discs and in whole larvae of wild-type and and mutant Drosophila melanogaster have been examined using two-dimensional polyacrylamide gel electrophoresis and autoradiography. After 2 hr of labeling with 35S during the third larval instar, the synthesis of more than 318 polypeptides has been detected in imaginal discs. Of these, 268 are present in similar amounts in all three disc types. The remaining polypeptides detected in the three imaginal disc types fall into two categories: those unique to a particular disc type, and those specific for a particular pair of disc types. These results are discussed in relation to the spectrum of gene expression in imaginal discs.  相似文献

Ninety clinical isolates of Pseudomonas aeruginosa were examined for susceptibility to carbenicillin by the broth dilution and disc diffusion methods. Inhibition zone diameters varied at given minimal inhibitory concentration levels of the antibiotic. Nevertheless, the results obtained allowed the proposal of the following tentative criteria for the interpretation of inhibition zones. Pseudomonadaceae yielding zones of inhibition measuring at least 10 and 16 mm in diameter around 25-and 100-mug discs, respectively, are sensitive to this antibiotic when examined by the standardized Bauer-Kirby method of disc susceptibility testing. Isolates characterized by zones of less than 100 mm in diameter around 25-mug discs should be tested with 100-mug discs before they are reported as sensitive or resistant to carbenicillin.  相似文献

Leg, wing, and eye-antennal imaginal discs of Drosophila melanogaster were obtained by dissection from late-third instar larvae and cultured in vitro in Schneider's medium containing β-ecdysone. Differentiation of adult structures was obtained in more than 90% of all cultures. Differentiation was somewhat slower than normal in vivo development, but synchronous, repeatable, and reasonably completed structures were obtained. Our initial efforts at analyzing the molecular events of imaginal disc differentiation in culture have been to study the protein synthetic pattern which occurs throughout the culture period. Discs were pulse labeled with [35S]methionine, and the proteins were separated by SDS-gel electrophoresis. Analysis of the synthetic pattern was done by autoradiography of these gels using X-ray film. In all three disc types, pronounced changes in protein synthetic patterns occurred throughout the culture period. These changes appeared to be under strict temporal control. Although disc-specific differences could be seen, a comparison of the three discs types revealed a striking similarity in the changes which occurred in the patterns of protein synthesis during the 5-day culture period. In general, the protein synthetic patterns of different imaginal discs at the same period during differentiation showed greater similarities than the patterns of a single disc type at different periods. These results are consistent with a view of differentiation as a tightly controlled program of gene activation and deactivation operating throughout the differentiation process.  相似文献

Summary The regulative behavior of fragments of the imaginal discs of the wing and first leg was studied when these fragments were combined with fragments of other thoracic imaginal discs. A fragment of the wing disc which does not normally regenerate when cultured could be stimulated to regenerate by combination with certain fragments of the haltere disc. When combined with a haltere disc fragment thought to be homologous by the criteria of morphology and the pattern of homoeotic transformation, such stimulated intercalary regeneration was not observed. Combinations of first and second leg disc fragments showed that a lateral first leg fragment could be stimulated to regenerate medial structures when combined with a medial second leg disc fragment but not when combined with a lateral second leg disc fragment. Combinations of wing and second leg disc fragments showed that one fragment of the second leg disc is capable of stimulating regeneration from a wing disc fragment while another second leg disc fragment fails to stimulate such regeneration. It is suggested that absence of intercalary regeneration in combinations of fragments of different thoracic imaginal discs is a result of homology or identity of the positional information residing in the cells of the fragments. The pattern of correspondence of positional information revealed by this analysis is consistant with the pattern of homology determined by morphological observation and by analysis of the positional specificity of homoeotic transformation among serially homologous appendages. The implications of the existence of homologous positional information in wing and second leg discs which share a common cell lineage early in development are discussed.  相似文献

Despite recent advances in imaging diagnostic technology and additional treatment options our ability to prevent or inhibit discogenic back pain has not drastically improved. The challenge of linking early degenerative patterns to dysfunction and pain remains. Using a novel material testing device designated the tissue diagnostic instrument (TDI) we measured the local stiffness and strain energy absorption in the radial direction of 13 intact intervertebral discs; effectively generating a mechanical profile of each disc. Prior to measuring mechanical properties, an MR image was taken of each spine segment and the discs were radiologically scored according to the Pfirrmann scale. After testing, a sagittal portion of each L1–L2 disc was excised from each of four spines for histology. No significant correlations were found between Pfirrmann grade and mechanical data. However, polarized light microscopy images of disc sections indicated correlations between local tissue modulus measured with the TDI and the clarity and density of lamellar striations.  相似文献

目的了解儿童感染肺炎链球菌和金黄色萄萄球菌耐药情况,为临床合理用药提供理论依据。方法对年龄为1个月~12岁的上呼吸道感染的儿童鼻咽分泌物或痰进行培养,分离肺炎链球菌和金黄色萄萄球菌;采用纸片扩散法及E-test法对分离株进行常用的10种抗生素敏感性检测。结果从2 250例呼吸道感染儿童鼻咽部、痰液分离出肺炎链球菌共120株,金黄色葡萄球菌共151株。120株肺炎链球菌对常用抗菌药物耐药率分别为青霉素41.7%(50/120)、苯唑西林26.7%(32/120)、头孢曲松9.2%(11/120)、红霉素95.8%(115/120)、克林霉素90.0%(108/120)、四环素70.8%(85/120)、氯霉素6.7%(8/120)、利福平0.8%(1/120)和氧氟沙星0.8%(1/120),肺炎链球菌青霉素不敏感株对β-内酰胺酶类耐药率明显高于青霉素敏感株。151株金黄色葡萄球菌中对常用抗菌药物耐药率分别为青霉素97.4%(147/151)、苯唑西林9.9%(15/151)、头孢曲松9.3%(14/151),对红霉素52.3%(79/151)、四环素31.8%(48/151)、克林霉素24.5%(37/151)、氯霉素9.9%(15/151)、氧氟沙星3.3%(5/151)和利福平2.0%(3/151)。未检出耐万古霉素菌株。结论肺炎链球菌对红霉素、克林霉素高度耐药;而金黄色葡萄球菌对青霉素高度耐药,临床应尽量减少此类药物的经验性用药,依据药敏结果选择抗菌药物进行治疗。  相似文献
