In Laminaria japonica Aresch breeding practice, two quantitative traits, frond length (FL) and frond width (FW), are the most important phenotypic selection index. In order to increase the breeding efficiency by integrating phenotypic selection and marker-assisted selection, the first set of QTL controlling the two traits were determined in F2 family using amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers. Two prominent L. japonicas inbred lines, one with “broad and thin blade” characteristics and another with “long and narrow blade” characteristics, were applied in the hybridization to yield the F2 mapping population with 92 individuals. A total of 287 AFLP markers and 11 SSR markers were used to construct a L. japonica genetic map. The yielded map was consisted of 28 linkage groups (LG) named LG1 to LG28, spanning 1,811.1 cM with an average interval of 6.7 cM and covering the 82.8% of the estimated genome 2,186.7 cM. While three genome-wide significant QTL were detected on LG1 (two QTL) and LG4 for “FL,” explaining in total 42.36% of the phenotypic variance, two QTL were identified on LG3 and LG5 for the trait “FW,” accounting for the total of 36.39% of the phenotypic variance. The gene action of these QTL was additive and partially dominant. The yielded linkage map and the detected QTL can provide a tool for further genetic analysis of two traits and be potential for maker-assisted selection in L. japonica breeding.  相似文献

The amplified fragment length polymorphism (AFLP) technique is a DNA technology that generates the so-called AFLP markers. These markers are genomic restriction fragments detected after two rounds of polymerase chain reaction (PCR) without prior knowledge of nucleotide sequence. Here we describe the first application of the AFLP technique in the rabbit. We have tested two primer combinations. The results obtained with the DNA from rabbits of different breeds justify the conclusion that AFLP analysis is an effective tool for genetic studies in the rabbit. In addition, we contribute to the linkage map of the rabbit by localizing two AFLP markers on rabbit linkage group VI (LG VI). For this purpose the progeny of a IIIVO/JU x [IIIVO/JU x AX/JU]F(1) backcross were genotyped for 12 AFLP markers and 3 LG VI classical markers [one coat color marker (e) and two biochemical markers (Es-1 and Est-2)]. AX/JU is a dietary cholesterol-susceptible (hyperresponding) inbred strain and IIIVO/JU is a dietary cholesterol resistant (hyporesponding) inbred strain. Moreover, it is possible to evoke dietary cholesterol-induced aorta atherosclerosis in a relatively short time period in AX/JU rabbits, in contrast to IIIVO/JU rabbits. A significant cosegregation was found between basal serum HDL cholesterol level (i.e., the level on a low-cholesterol, control diet) and an AFLP marker on LG VI. It is concluded that one or more genes of LG VI are regulating the basal serum HDL cholesterol level in rabbits. Thus the present study with rabbits clearly illustrates the value of AFLP markers for the construction of linkage maps and mapping of quantitative trait loci (QTL).  相似文献

To identify genetic factors underlying phosphorus (P) uptake and use efficiency under low-P stress in rice (Oryza sativa L.), 84 selected genotypes (recombinant inbred lines) and their parents (which differed in tolerance for low-P stress) “IR20” and IR55178-3B-9-3, were cultured in liquid medium supplemented with adequate and low P levels in a greenhouse. Plants were sampled after 6 weeks in culture for measurements of plant dry weight, P concentration, P uptake and P use efficiency under both P sufficient and stress conditions. A total of 179 molecular markers, including 26 RFLPs and 153 AFLPs, mapped on all 12 chromosomes of rice based on the 84 selected genotypes were used to detect the quantitative trait loci (QTLs) underlying tolerance for low-P stress. Three QTLs were detected on chromosomes 6, 7 and 12, respectively, for relative plant dry weight (RPDW) and relative P uptake (RPUP). One of the QTLs flanked by RG9 and RG241 on chromosome 12 had a major effect which explained about 50% of the variations in the two parameters across the population. The results coincided with the QTLs for low-P stress based on relative tillering ability from the same population from a cross between Nipponbare and Kasalath under soil condition. The identical major QTL for P uptake and plant growth under low-P stress in both liquid medium and soil strongly suggests that the ability of P uptake mainly controls rice tolerance for low-P stress.  相似文献

对RIL-8群体纹枯病抗性进行QTL检测,检测到一个加性QTL,位于1A染色体上,贡献率为21.57%;检测到4对QTL间互作位点,涉及7条染色体,互作贡献率分别为11.63%、6.54%、14.04%、20.01%,互作总贡献率为52.23%.通过对RIL-SES群体118个系检测,发现1个SSR标记Xgwm526,位于2B染色体上,与纹枯病抗病基因距离为27.9cM;一个ISSR标记IS840,与纹枯病抗病基因距离为16.9cM.  相似文献

利用重组自交系和SSR标记进行陆地棉株型QTL的鉴定和定位   总被引:6,自引:1,他引:6  
通过中棉所12与8891的杂交及多代自交,获得由180个家系构成的重组自交系F8、F9群体。重组自交系群体、两亲本及F1于2002、2003两年种植;对株型性状进行了研究,两年共调查了10个株型形状。利用该重组自交系群体,采用SSR为主体的分子标记构建了遗传连锁图,并对株型性状进行了单位点和双位点水平的QTL定位。结果表明,QTL加性效应和上位性互作效应作为棉花重组自交系株型性状的遗传基础起着重要作用;中棉所12与8891间多态性位点偏少,而表型差异较大且其杂交种湘杂棉二号有很强的杂种优势,QTL互作可部分解释这一现象:结合对产量品质性状的研究结果,认为上位性可能是湘杂棉二号杂种优势的重要遗传基础。  相似文献

Xiangzamian 2 (XZM2) is the most widely cultivated cotton hybrid in China. By crossing two parents Zhongmiansuo 12 and 8891 and upon subsequent selfings, we got F8 and F9 populations having 180 recombinant inbred lines. Ten plant architecture traits were investigated in two years with this population. A genetic map was constructed mainly with SSR markers. Quantitative trait loci (QTL) conditioning plant architecture traits were determined at the single-locus and double-locus levels. The results showed that epistastic effects as well as additive effects of QTL played an important role as the genetic basis of cotton plant architecture. The QTL detected in our research might provide new information on improving plant architecture traits. The polymorphism of molecular markers between ZMS12 and 8891 were quite limited, while significant differences between their phenotypes were found and the hybrid XZM2 expressed high heterosis in yield. All these could be partly explained by the effect of epistatic QTL.  相似文献

以小麦DH群体(旱选10号×鲁麦14)为研究材料,采用复合区间作图法,对小麦幼苗在水分胁迫及非胁迫条件下的水分利用效率(WUE)及其相关性状的QTL进行定位,并对比分析QTL的加性效应.两种水分条件下共检测到14个具显著加性效应的QTL,分布在2A、3A、4A、5A、6A、7A、1B、3B、3D染色体上,可解释表型变异的范围在6.36%~19.73%.其中,非胁迫(对照)条件下检测到10个QTL,包括2个单株WUE的QTL,5个地上部WUE的QTL,1个根系WUE的QTL及2个总耗水量的QTL;水分胁迫条件下上述性状各检测到1个QTL.对于同一性状没有检测到在两种水分条件下均位于同一标记区间的QTL,表明不同水分环境条件下同一性状的QTL表达模式是不同的.论文也讨论了可能用于标记辅助选择的QTL及其分子标记.  相似文献

The Use of Morphological and AFLP Markers in Diversity Analysis of Linseed   总被引:3,自引:0,他引:3  
Different methods of genetic diversity measures could give better judgment of differentiating important accessions for growers, germplasm curators and plant breeders. Data of 60 accessions of linseed, mainly from Ethiopia, were used to assess their genetic diversity, employing morphological and amplified fragment length polymorphism (AFLP) methods. Analyses of genetic distance, principal components and clusters showed the presence of a wide range of diversity among the studied accessions. The mean for genetic distance estimates of the entire 1770 pairwise accessions was 0.6684 for morphology, while that of AFLP was 0.5734. These genetic distances varied from zero to one for morphology in contrast to 0.29 to 0.71 of the AFLP. Morphological and AFLP based clusters and their accompanying analyses showed different hierarchical patterns of genetic diversity among the accessions. Despite their disparity, the two diversity measures were found independently useful for assessing the degree of relatedness and the overall patterns of genetic variation among the analysed linseed accessions.  相似文献

单核苷酸多态性是指DNA序列上的单个碱基变异,它具有分布广、多态信息含量大、易于检测和统计分析等优点,能较好用于基因图谱构建和数量性状QTL定位研究,被称为继RFLP和微卫星标记之后的第3代基因遗传标记。本文综述了单核苷酸多态性的性质及检测技术、利用候选基因SNP进行鸡QTL定位研究的现状,并对未来SNP的应用前景进行了展望。Abstract:Single nucleotide polymorphism (SNP) refers to the change of single nucleotide in DNA sequence.Because of its high density in genomes and easy in detection and analysis statistically,SNP can be used in genetic linkage map construction and QTL mapping.Here,the characters and detecting technology of SNP,as well as the status and foreground of the use of candidate gene SNP in chicken QTL mapping are introduced.  相似文献

One hundred and seventy-four rat loci which contain short tandem repeat sequences were extracted from the GenBank or EMBL data bases and used to define primers for amplification by the polymerase chain reaction (PCR) of the microsatellite regions, creating PCR-formatted sequence-tagged microsatellite sites (STMSs). One hundred and thirty-four STMSs for 118 loci, including 6 randomly cloned STMSs, were characterized: (i) PCR-analyzed loci were assigned to specific chromosomes using a panel of rat x mouse somatic cell hybrid clones. (ii) Length variation of the STMSs among 8 inbred rat strains could be visualized at 85 of 107 loci examined (79.4%). (iii) A genetic map, integrating biochemical, coat color, mutant and restriction fragment length polymorphism loci, was constructed based on the segregation of 125 polymorphic markers in seven rat backcrosses and in two F2 crosses. Twenty four linkage groups were identified, all of which were assigned to a defined chromosome. As a reflection of the bias for coding sequences in the public data bases, the STMSs described herein are often associated with genes. Hence, the genetic map we report coincides with a gene map. The corresponding map locations of the homologous mouse and human genes are also listed for comparative mapping purposes.  相似文献

以UBC 269484和GSLP1569的序列为支点,设计合成了包括UBC 269和GSLP1在内的9条引物,以葡萄 有核亲本红地球和无核亲本红光无核的DNA为模板,对这9个引物进行筛选,结果GSLP1、39970524 5号引物和 39970524 6号引物在无核亲本红光无核上扩增出了特异标记GSLP1569、39970524 5 564、39970524 6 1538和 39970524 6 1200。用这3个特异引物在红地球、红光无核、无核白和红地球×红光无核杂交组合F1代163株杂 种的DNA样上进行PCR扩增,结果4个特异标记在F1群体中与无核主效基因共分离。4个特异标记也出现于所 用组合中无核基因原始供给者无核白上。这些标记和葡萄无核主效基因相连锁。用QTXb17遗传作图软件,对葡 萄无核主效基因S定位与作图,当P=0.01时,LOD值在32.7~46.4之间,置信界限在0.2~9.9之间。这4个 特异标记和无核主效基因S处于在同一连锁群,位于无核主效基因S的两侧,覆盖基因组12.3cM。特异标记 39970524 5 564、GSLP1569、39970524 6 1538、39970524 6 1200距S基因的遗传距离分别为0.6cM、1.2cM、 4.9cM和11.1cM。  相似文献

维生素E (VE)是稻米营养品质的重要指标。水稻(Oryza sativa)是我国种植最广泛的粮食作物, 增加其籽粒的VE含量是实现国民营养强化的一条便捷有效的途径。该研究以籼稻华占(HZ)为父本, 粳稻热研2号(Nekken2)为母本, 构建120个重组自交系(RILs)群体。采用高效液相色谱法(HPLC)对RILs群体的VE各组分含量进行测定, 并基于构建的高密度分子遗传图谱进行QTL定位, 谱系分析后挖掘到122个VE总量和分量相关QTLs, 分布在12条染色体上。其中qT3α/to2-1的LOD值高达10.32, qT3α2-1的LOD值高达9.91, 另有多个控制各异构体含量的主效QTLs, 且区间内包含OsGGR1OsGGR2OsTCOsγTMT等VE生物合成基因。通过qRT-PCR检测亲本中VE合成基因的表达量, 发现在华占中候选基因的表达量均极显著高于热研2号, 推测这些基因的高表达是华占生育酚及生育三烯酚含量高于热研2号的原因。研究挖掘到的QTL数目较多, LOD值也较大, 为进一步筛选和培育高VE含量的水稻新品种奠定了分子基础, 同时为揭示水稻VE生物合成的分子调控机制提供了重要基因资源。  相似文献

用番茄高抗青枯病品种“T51A”与高感青枯病品种“T9230”配制杂交组合,接种鉴定其正反交F1代及F2代分离群体的青枯病发生情况。结果表明,T51A对青枯病的抗性属于细胞质遗传,受1对杂合基因加性控制。用64个EcoRI/seI引物组合对“T51A”、“T9230”两个亲本及其F2代抗病和感病基因池进行AFLP分析,共扩增出约4200条可分辨的带,其中2条为稳定的差异。用“T51A”和“T9230”杂交产生的F2代分离群体对2个特异条带与目的基因的遗传连锁性进行分析,发现特异条带AAG/CAT与暂定名为RRS-342的抗青枯病基因紧密连锁,二者之间的遗传距离为6.7 cM。将AAG/CAT片段回收、克隆和测序,成功地将其转化为SCAR标记,可以更加方便地用于对番茄青枯病基因的标记辅助选择。   相似文献

Three interspecific crosses were developed using Cicer arietinum (ICC 4918) as the female parent and wild Cicer species [C. reticulatum - JM 2100, JM 2106 and C. echinospermum - ICCW 44] as the male parent. Cicer arietinum (ICC 4918) × C. reticulatum (JM 2100) cross produced the largest number of F2 plants and was chosen for linkage mapping using Random Amplified Polymorphic DNA (RAPD) primers. A partial linkage map was constructed based upon the segregation of 36 RAPD markers obtained by amplification using 35 primers. The linkage map consists of two linkage groups with 17 linked markers covering a total of 464.9 cM. Analyses also revealed association of three morphological traits with linked RAPD markers. Out of seven morphological traits tested for association with linked markers in the segregating plants, four Quantitative trait loci (QTL) were detected for the trait leaf length and three QTLs each for the traits leaf width and erect plant habit.  相似文献

Identification of the sex-determining genes of the Nile tilapia (Oreochromis niloticus) has important implications for commercial aquaculture. We previously identified an XX/XY sex-determining locus in this species within a 10-cM interval between markers GM201 and UNH995 on linkage group one (LG1). In order to refine this region, we developed new AFLP markers using bulked segregant analysis of the mapping families. We identified three AFLP markers that showed a sex-specific pattern of segregation. All three mapped near, but just outside, the previously identified sex-determining region on LG1. Hybridization of BAC clones containing these markers to chromosome spreads confirmed that the XX/XY sex-determining locus is on one of the small chromosomes in O. niloticus.  相似文献

Two rabbit (Oryctolagus cuniculus) inbred strains (AX/JU and IIIVO/JU) have been used for genetic analysis of quantitative traits related to dietary cholesterol susceptibility. Application of the AFLP (amplified fragment length polymorphism) technique with 15 primer combinations revealed 226 polymorphisms between the 2 inbred strains. A total of 57 animals from a backcross progeny (IIIVO/JU x [IIIVO/JU x AX/JU]F1) were available for the genetic analysis. These backcross animals were fed a commercial pelleted diet fortified with 0.3% w/w cholesterol during a test period that lasted five weeks. A male genetic map could be constructed, consisting of 12 linkage groups and 103 AFLP markers. Linkage analysis between the cholesterol-related traits and marker loci revealed a significant LOD score for the relative weight of adrenal glands in males (LOD score = 3.83), whereas suggestive linkages were found for basal serum total cholesterol levels in females (LOD score = 2.69), for serum total cholesterol response (area under the curve) in males (LOD score = 2.21), and for hematocrit in males (LOD score = 3.24).  相似文献
