In this study, zinc status and urinary zinc excretion with and without desferrioxamine (DFO) infusion and the relationship between urinary zinc excretion and renal tubular dysfunction in thalassemia major (TM) patients were investigated. Forty TM patients were given four DFO infusions on alternate days over a 1-wk period prior to the transfusion. On each day that DFO was given, a 24-h urine collection initiated. DFO was omitted for 1-wk before the following transfusion and during the period four 24-h urine collections were performed. Twenty healthy children provided 24-h urine collection as controls. Blood samples were taken on each of two consecutive transfusion days of the patients and from the controls. Urinary zinc excretion was measured and plasma and red blood cell (RBC) zinc analysis were performed by inductively coupled plasma-atomic emission spectrophotometry. UrinaryN-acetyl-Β-D-glucosaminidase (NAG) activity and creatinine were determined in morning urine specimens. The mean plasma zinc concentration was significantly lower in the patients not given DFO compared to the values of the patients given DFO and the control group. The mean RBC zinc concentration (Μmol/g Hb) in the patients (with and without DFO) and the control group were similar. Urinary zinc excretion was significantly higher in the patients receiving DFO compared to the control group, whereas urinary zinc excretion in the patients not given DFO was not different from the controls. Urinary NAG indices (U/g Cr) were significantly higher in the patients compared to controls. Urinary zinc excretion was correlated with the urinary NAG indices.  相似文献

Patients admitted for major vascular reconstruction operations received an i.v. dose of 65Zn. From the increased whole-body retention of 65Zn and the findings on other parameters it was concluded that the post-operative zinc metabolism of these patients differed from that of the control subjects. Unexpectedly, this difference persisted for several months. These patients can probably serve as controls in studies of patients who have undergone surgery of the intestinal tract. Increase of whole-body retention of 65Zn may be a sensitive indicator for subclinical zinc deficiency, but application in practice is hampered by the long duration of the period required for the measurement. From the present results it seems likely that measurement of the retention of 65Zn in the forearm as a function of time yields the same information but in a considerably shorter time.  相似文献

Intercropping of companion plants (CPs) that release distinct volatile compounds with horticultural crops has been proposed to improve pest management. Although CP extracts or essential oils have been reported to be efficient in disturbing the settling of insects such as aphids, the effect of using actual CPs remains relatively unknown. Our aim was to screen odorous CP species for their effect on the performance of green peach aphid, Myzus persicae Sulzer (Hemiptera: Aphididae), when released on adjacent pepper plants. Intercropping was tested in growth chambers using potted plants to avoid root interactions and homogenise the abiotic environment of CPs. Our results indicated that eight CP species—rosemary, African marigold, French marigold, geranium, lavender, basil, chives, and pot marigold—affected aphid performance by reducing the reproductive performance and/or settlement of females. Rosemary followed by African and French marigold was associated with the largest reduction in adult female and nymph number. Chemical analyses indicated that rosemary and French marigold emit specific volatiles. In contrast, four CP species (false yellowhead, thyme, savoury, and peppermint) were not associated with a significant change in aphid performance and emitted a lower diversity of volatiles. This screening of CPs under controlled conditions may be considered as a first step towards the identification of volatiles emitted by actual plants that can affect aphid performance and help the choice of CPs to optimise intercropping strategies.  相似文献

Autopsy records (1936–66) show esophageal cancer to be the most common tumor for both men and women among the locally-born negroid population of Curaçao. Crude incidence rate of 20.9 per 100,000, virtually constant over 30 years, is exceeded only by that of certain parts of Russia and the Transkei region of South Africa. The island of Aruba (42 miles distant) has a very low incidence. Drought conditions and food and water supply are virtually identical in both islands, also reverse smoking by women only. But there are striking contrasts in the use of plants for remedies. In Curaçao, more than 100 species of local plants are employed for self-medication and for “tea.” Interrogation of living esophageal cancer victims and survivors of deceased has revealed consistent use of these plants. Species cited most frequently in interviews are being collected, lyophilized, and supplied to the Laboratory of Pathology, National Cancer Institute, for animal-testing.  相似文献

An enzyme system which metabolizes α-tocopherol has been identified in homogenates of etiolated pea shoots. Enzyme activity is considerably increased by the presence of 20% ethanol in the incubation mixture. The enzyme has an absolute requirement for phospholipid. The reaction utilizes molecular oxygen and it is proposed that the enzyme be called α-tocopherol oxidase.  相似文献

Because of regulations at the whole-plant level, cells of droughted plants do not necessarily experience dehydration. In fact, recent data suggest that they usually do not in the range of water deficits compatible with agriculture. In this range, leaf water potential and cell turgor are frequently maintained at high values in spite of decreasing soil water status. As a consequence, water stress cannot be defined by plant water status, except when very rapid and severe water deficits cause catastrophic events such as xylem embolism or severe cell dehydration. In water deficits compatible with agricultural situations, plant water status is in many cases tightly controlled by the plant. Water stress should then be defined by water statuses at plant boundaries, i.e. soil and air. Partial maintenance of plant water status under water deficit is allowed by controls of stomatal conductance, root and leaf expansions and leaf senescence. These processes involve both chemical and hydraulic signallings from roots. All these controls tend to reduce transpiration (stomatal closure, reduced leaf growth or leaf senescence) or to increase water uptake (maintenance of root growth or increase in root/shoot ratio). During relatively mild stresses, the role of abscisic acid, frequently considered as a stress hormone, in fact appears to be to avoid dehydration at the cellular level. In the cases described here, selecting plants for better resistance to cell dehydration may not be the best strategy for improving drought resistance of crops.  相似文献

The report in 1971 by Comuet and Astier‐Manifacier that Chinese cabbage contains an active RNA‐dependent RNA polymerase has been extended to all plants studied. This has met with much opposition because the central dogma of molecular biology requires no replication mechanism for RNA. Only upon RNA virus infection are such enzymes needed, and it was generally believed that these were always and only virus‐coded. The purification and characterization of several of these plant viruses will be reviewed, with particular reference to the fact that while their amount in plant tissue is variably increased by various RNA virus infections their nature is unaffected by the viral genome and is strictly host‐specific. It will be noted, however, that in a specific instance viral infection has been shown to affect an important property of the enzyme. Also, it has become evident that certain plant viruses resemble animal picorna viruses (e.g., polio virus) and that these viruses carry an RNA polymerase gene. The same may be true, but has not been proven, for a small group of plant viruses that shows resemblances to the prokaryotic RNA phages in which a viral gene product together with host proteins form the RNA polymerase. An important question that remains to be solved in future work is the role of RNA polymerases in normal plant cell biology. Also, the mechanism by which viral infection causes the enzyme to become largely membrane or organelle bound and possibly conformationally changed in the process remains to be elucidated.  相似文献

A Zn–salophen complex has been incorporated into POPC large unilamellar liposomes (LUV) obtained in phosphate buffer at pH 7.4. Fluorescence optical microscopy and anisotropy measurements show that the complex is located at the liposomal surface, close to the polar headgroups. The interaction of the POPC phosphate group with Zn2 + slowly leads to demetallation of the complex. The process follows first order kinetics and rate constants have been measured fluorimetrically in pure water and in buffered aqueous solution.The coordination of the phosphate group of monomeric POPC with salophen zinc also occurs in chloroform as detected by ESI-MS measurements.The effect of the Zn–salophen complex on the stability of POPC LUV has been evaluated at 25 °C by measuring the rate of release of entrapped 5(6)-carboxyfluorescein (CF) in the presence and in the absence of Triton X-100 as the perturbing agent. It turns out that the inclusion of the complex significantly increases the stability of POPC LUV.  相似文献

γ-Butyrobetaine hydroxylase (BBOX) is a 2-oxoglutarate and Fe(II) dependent oxygenase that catalyses an essential step during carnitine biosynthesis in animals. BBOX is inhibited by ejection of structural zinc by a set of selenium containing analogues. Previous structural analyses indicated that an undisrupted N-terminal zinc binding domain of BBOX is required for catalysis. Ebselen is a relatively potent BBOX inhibitor, an observation which may in part reflect its cardioprotective properties.  相似文献

This study reports on the dieback and recovery of red-osier dogwood, Cornus sericea L. plants from near-lethal (NL, sublethal) stress after varying lengths of post-stress environment (PSE). Intact dormant stems were subjected to 47° C for one hour during either October, November or December, and then placed into either constant 0° C or 23° C (dark condition) or kept under natural conditions at Corvallis, OR. Plants exposed to NL-heat stress in October died prior to 9 weeks of 0° C PSE, while none of the plants from other PSE treatments showed signs of injury. For plants exposed to NL-heat stress during November and December, stemdieback occurred at 0° C after 12 and 15 weeks, respectively. None of the plants from the other PSE treatments were injured. Post-stress temperatures of 0° or 5° C following NL-heat in October were lethal while temperatures above 10° C allowed recovery. Post-stress exposure to 0° C injured excised stems within 48 h, whereas irreversible damage to whole plants occurred by two weeks. Dormant plants exposed in October to other stresses, e.g., freezing temperature and hydrogen cyanamide, at NL dosages showed that these stresses also caused plant dieback at 0° C and little or no dieback at 23° C PSE.Abbreviations NL Near-Lethal - PSE Post-Stress Environment  相似文献

Rensch's rule refers to a pattern in sexual size dimorphism (SSD) in which SSD decreases with body size when females are the larger sex and increases with body size when males are the larger sex. Many animal taxa conform to Rensch's rule, but it has yet to be investigated in plants. Using herbarium collections from New Zealand, we characterized the size of leaves and stems of 297 individuals from 38 dioecious plant species belonging to three distantly related phylogenetic lineages. Statistical comparisons of leaf sizes between males and females showed evidence for Rensch's rule in two of the three lineages, indicating SSD decreases with leaf size when females produce larger leaves and increases with leaf size when males produce larger leaves. A similar pattern in SSD was observed for stem sizes. However, in this instance, females of small-stemmed species produced much larger stems than did males, but as stem sizes increased, SSD often disappeared. We hypothesize that sexual dimorphism in stem sizes results from selection for larger stems in females, which must provide mechanical support for seeds, fruits, and dispersal vectors, and that scaling relationships in leaf sizes result from correlated evolution with stem sizes. The overall results suggest that selection for larger female stem sizes to support the weight of offspring can give rise to Rensch's rule in dioecious plants.  相似文献

In the present work, we provide compelling evidence for the expression of a ghrelin-like peptide hormone that has only been associated with animals, in various plant tissues. Ghrelin, the appetite stimulating hormone, has been identified from a number of different species including humans, rat, pig, mouse, gerbil, eel, goldfish, bullfrog and chicken. The study here was conducted using an immunohistochemistry assay to screen whether plants have any ghrelin immunoreactivity. In this respect, Prunus x domestica L. and Marus alba were examined. Immunohistochemistry results showed that there is a strong human ghrelin immunoreactivity substance in the parenchyma cells of these plants. This was entirely unexpected since this hormone was considered to be present solely in animals. Thus, this study is the first to report the presence of a peptide with ghrelin-like activity in plants, a finding that has only been observed in the animal kingdom. RIA analysis confirmed that these plants contain significant amounts of this substance. Furthermore, reverse-phase HPLC analyses of plant extracts showed an elution characteristic of the peptide identical to that of human ghrelin. In general, fruit from both plants had higher levels of the peptide than the vegetative parts.  相似文献

Soybean (Glycine max (L.) Merr.) seeds contain the storage protein -conglycinin, encoded by a multigene family. -Conglycinin consists of three subunits; , , and . A genomic clone for a -subunit of -conglycinin has been characterized by restriction-enzyme mapping and hybrid selected in-vitro translation followed by immunoprecipitation. In order to determine the developmental regulation of this -subunit gene, its expression was studied in seeds of transgenic petunia (Petunia hybrida) and tobacco (Nicotiana tabacum L.) plants. The -subunit expressed in seeds of petunia and tobacco was recognized by anti--conglycinin serum at a relative molecular mass of 53 000, equivalent to that of the native protein. Separation of the petunia-seed proteins by isoelectric focusing followed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblot analysis showed that multiple isoelectric forms of the -subunit were produced. There was approximately a twofold variation in the accumulation of the -subunit protein in the mature seeds of transgenic petunia plants, each containing a single -subunit gene. However, the level of protein accumulation in mature seeds and the amount of -subunit mRNA in developing seeds was not correlated. Accumulation of the -subunit protein in transgenic seeds was less than the -subunit protein that accumulated in transgenic petunia seeds containing a single -subunit gene and less than the amount of the -subunit in mature soybean seeds which contain 8–13 -subunit genes. Cultivation of genetically modified crops is presently based largely on four crops containing few transgenes and grown in four countries. This will soon change and pose new challenges for risk assessment. A more structured approach that is as generic as possible is advocated to study consequences of gene flow. Hazards should be precisely defined and prioritized, with emphasis on quantifying elements of exposure. This requires coordinated effort between large, multidisciplinary research teams.  相似文献
