Landscape composition and physiognomy affect community structure and species distribution across space and time. The pine processionary moth (PPM) (Thaumetopoea pityocampa Den. & Schiff., Lepidoptera, Notodontidae) is a common pine defoliator throughout southern Europe and Mediterranean countries. We surveyed the spatiotemporal distribution of the PPM in a pine plantation forest in southwestern France and used the density of the winter nests as a proxy for population density. The study spanned 4 years (2005–2008) and showed a high temporal variability in nest density. We found a strong edge effect with nest densities at stand edges more than twice as large as within-stand densities. At the landscape scale, the spatial distribution of the moth exhibited a significant spatial autocorrelation in 3 out of 4 years of our study. The spatial scales of the autocorrelation ranged from ca. 2 km to more than 22 km. We found a positive correlation between spatial distributions corresponding to certain sampling years, but the relationship was not systematic. Landscape configuration appeared to be an important driver of the PPM spatial pattern. Bivariate Moran’s I correlograms showed that patch richness density as well as the percentage of local landscape covered by various land uses were correlated with population density. The study showed that accounting for landscape characteristics may be important in order to understand forest insect pest distribution, even in cases where the host species is abundant and homogeneously distributed throughout the study area, e.g., pure plantation forests.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the blue crab Callinectes danae were analyzed using the substrate p-nitrophenylphosphate. The (Na+,K+)-ATPase hydrolyzed PNPP obeying cooperative kinetics (n=1.5) at a rate of V=125.4±7.5 U mg−1 with K0.5=1.2±0.1 mmol l−1; stimulation by potassium (V=121.0±6.1 U mg−1; K0.5=2.1±0.1 mmol l−1) and magnesium ions (V=125.3±6.3 U mg−1; K0.5=1.0±0.1 mmol l−1) was cooperative. Ammonium ions also stimulated the enzyme through site–site interactions (nH=2.7) to a rate of V=126.1±4.8 U mg−1 with K0.5=13.7±0.5 mmol l−1. However, K+-phosphatase activity was not stimulated further by K+ plus NH4+ ions. Sodium ions (KI=36.7±1.7 mmol l−1), ouabain (KI=830.3±42.5 μmol l−1) and orthovanadate (KI=34.0±1.4 nmol l−1) completely inhibited K+-phosphatase activity. The competitive inhibition by ATP (KI=57.2±2.6 μmol l−1) of PNPPase activity suggests that both substrates are hydrolyzed at the same site on the enzyme. These data reveal that the K+-phosphatase activity corresponds strictly to a (Na+,K+)-ATPase in C. danae gill tissue. This is the first known kinetic characterization of K+-phosphatase activity in the portunid crab C. danae and should provide a useful tool for comparative studies.  相似文献

Many studies have reported the role of COMT Val158Met with breast cancer risk, but the results remained controversial. In addition, previous meta-analysis on COMT Val158Met showed conflicting results. Hence, we performed a meta-analysis to investigate the association between breast cancer and COMT Val158Met (30,199 cases and 38,922 controls) in different inheritance models. When all the eligible studies were pooled into this meta-analysis, there was no evidence of significant association between breast cancer risk and COMT Val158Met polymorphism in any genetic model (dominant model: odds ratio [OR] = 0.99, 95% confidence interval [CI] = 0.94–1.04, P value of heterogeneity test [P h] = 0.009, I 2 = 36.9%; recessive model: OR = 0.97, 95% CI = 0.92–1.02, P h = 0.044, I 2 = 28.6%; additive model: OR = 0.98, 95% CI = 0.91–1.05, P h = 0.004, I 2 = 40.4%). However, significant between-study heterogeneity was detected in any genetic model. Hence, we performed the stratified analysis according to ethnicity, source of controls, menopausal status, and family history. In the stratified analysis by ethnicity significantly decreased breast cancer risk was observed in Caucasian population (recessive model: OR = 0.96, 95% CI = 0.92–1.00, P h = 0.419, I 2 = 3.1%). In conclusion, this meta-analysis indicates that COMT Val158Met polymorphism may be associated with decreased breast cancer risk in Caucasian population. However, a study with the larger sample size is needed to further evaluated gene-environment interaction on COMT Val158Met polymorphisms and breast cancer risk.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the freshwater shrimp, Macrobrachium olfersii, acclimated to 21‰ salinity for 10 days were investigated using the substrate p-nitrophenylphosphate. The enzyme hydrolyzed this substrate obeying cooperative kinetics at a rate of 123.6 ± 4.9 U mg− 1 and K0.5 = 1.31 ± 0.05 mmol L− 1. Stimulation of K+-phosphatase activity by magnesium (Vmax = 125.3 ± 7.5 U mg− 1; K0.5 = 2.09 ± 0.06 mmol L− 1), potassium (Vmax = 134.2 ± 6.7 U mg− 1; K0.5 = 1.33 ± 0.06 mmol L− 1) and ammonium ions (Vmax = 130.1 ± 5.9 U mg− 1; K0.5 = 11.4 ± 0.5 mmol L− 1) was also cooperative. While orthovanadate abolished p-nitrophenylphosphatase activity, ouabain inhibition reached 80% (KI = 304.9 ± 18.3 μmol L− 1). The kinetic parameters estimated differ significantly from those for freshwater-acclimated shrimps, suggesting expression of different isoenzymes during salinity adaptation. Despite the ≈2-fold reduction in K+-phosphatase specific activity, Western blotting analysis revealed similar α-subunit expression in gill tissue from shrimps acclimated to 21‰ salinity or fresh water, although expression of phosphate-hydrolyzing enzymes other than (Na+,K+)-ATPase was stimulated by high salinity acclimation.  相似文献

Plant species in fragmented populations are affected by landscape structure because persistence within and migration among inhabited patches may be influenced by the identity and configuration of surrounding habitat elements. This may also be true for species of the semi-natural vegetation in agricultural landscapes. To determine the effect of landscape elements we analyzed Wood Avens (Geum urbanum L.) populations within three 4×4 km2 agricultural landscapes in Germany, Switzerland and Estonia, which differ in levels of land use intensity and habitat fragmentation. Genetic variation was determined in 15 randomly selected populations in each landscape using 10 microsatellite loci. The landscape structure was assessed at two circles around each population, with radii defined by the range limits of spatial genetic autocorrelation. Multiple regression analysis was used to determine the influence of landscape structure variables for inter- and intrapopulation genetic diversity. Gene diversity was equally high in Germany (He=0.27) and Switzerland (He=0.26) but lower in Estonia (He=0.16). A high overall inbreeding coefficient (FIS=0.89) was found, as expected for a selfing breeding system in G. urbanum. Genetic differentiation among populations was high (overall FST=0.43, 0.48, and 0.45 in Estonia, Switzerland and Germany, respectively), and did not differ among the three landscapes. Only a moderate influence of individual land use types on genetic diversity within and among populations was found with some idiosyncratic relationships. Genetic variation within populations was correlated to the amount of hedgerows positively in Estonia but negatively in Switzerland. The study demonstrates that the distribution of individual land use types affects the genetic pattern of a common plant species. However, different variables were identified to influence the genetic structure in three different landscapes. This indicates a major influence of landscape-specific land use history and stochastic processes determining gene flow and plant population structure.  相似文献

The aim of this study was to characterize the role of the late Na+ current (INa,L) as a mechanism for induction of both tachy and bradyarrhythmias in murine heart and sino-atrial node tissue. The sea anemone toxin ATX-II and ranolazine were used to increase and inhibit, respectively, INa,L. In sixteen hearts studied, exposure to 1–10 nM ATX-II caused a slowing of intrinsic heart rate and prolongations of the P–R and QT intervals, the duration of the monophasic action potential, and the sinus node recovery time, accompanied by frequent occurrences of early afterdepolarisations, delayed afterdepolarisations and rapid, repetitive ventricular tachy and sino-atrial bradyarrhythmias. ATX-II also slowed sinus node pacemaking, and induced bradycardic arrhythmias in isolated sino-atrial preparations (n = 5). The ATX-II-induced alteration of electrophysiological properties and occurrence of arrhythmic events were significantly attenuated by 10 μM ranolazine in intact hearts (n = 11) and isolated sino-atrial preparations (n = 5). In conclusion, the INa,L enhancer ATX-II causes both tachy and bradyarrhythmias in the murine heart, and these arrhythmias are markedly attenuated by the INa,L blocker, ranolazine (10 μM). The results suggest that INa,L blockade may be the mechanism underlying the reductions of both brady and tachyarrhythmias by ranolazine that were observed during the MERLIN-TIMI clinical outcomes trial.  相似文献

The contribution of cationic conductances in shaping the rod photovoltage was studied in light adapted cells recorded under whole-cell voltage- or current-clamp conditions. Depolarising current steps (of size comparable to the light-regulated current) produced monotonic responses when the prepulse holding potential (V h) was −40 mV (i.e. corresponding to the membrane potential in the dark). At V h = −60 mV (simulating the steady-state response to an intense background of light) current injections <35 pA (mimicking a light decrement) produced instead an initial depolarisation that declined to a plateau, and voltage transiently overshot V h at the stimulus offset. Current steps >40 pA produced a steady depolarisation to ≈−16 mV at both V h. The difference between the responses at the two V h was primarily generated by the slow delayed-rectifier-like K+ current (I Kx), which therefore strongly affects both the photoresponse rising and falling phase. The steady voltage observed at both V h in response to large current injections was instead generated by Ca-activated K+ channels (I KCa), as previously found. Both I Kx and I KCa oppose the cation influx, occurring at the light stimulus offset through the cGMP-gated channels and the voltage-activated Ca2+ channels (I Ca). This avoids that the cation influx could erratically depolarise the rod past its normal resting value, thus allowing a reliable dim stimuli detection, without slowing down the photovoltage recovery kinetics. The latter kinetics was instead accelerated by the hyperpolarisation-activated, non-selective current (I h) and I Ca. Blockade of all K+ currents with external TEA unmasked a I Ca-dependent regenerative behaviour.  相似文献

The effect of relative meal size (0.5–24% body mass) on specific dynamic action (SDA) was assessed in Chinese catfish (Silurus asotus Linnaeus) (30.90±1.30 g) at 25.0°C; the cutlets of freshly killed loach without viscera, head and tail were used as a test meal. There was no significant difference in either SDA duration or peak oxygen consumption (VO2) among low meal size ranges. But both increased linearly as meal size increased from 2 to 24% without reaching a plateau. Factorial metabolic scope was 5.92 in fish fed with 24% body mass, the highest documented feeding metabolic scope value in fish till now. The Peak VO2 of satiated meal size groups (175.85±10.55 mg O2 h−1) was above 80% of maximum metabolic rate during locomotion recovery process (215.48±7.07 mg O2 h−1). The relationship between energy expended on SDA (E) and energy ingested (I) was described as: E=0.0000432I 2+0.140I+2.12. The lowest value of SDA coefficient appeared at 2% body mass group.  相似文献

The effects of temperature, salinity and irradiance on the growth of the red tide dinoflagellate Gyrodinium instriatum Freudenthal et Lee were examined in the laboratory. Exposed to 45 different combinations of temperature (10–30 °C) and salinity (0–40) under saturating irradiance, G. instriatum exhibited its maximum growth rate of 0.7 divisions/day at a combination of 25 °C and a salinity of 30. Optimum growth rates (>0.5 divisions/day) were observed at temperatures ranging from 20 to 30 °C and at salinities from 10 to 35. The organism could not grow at ≤10 °C. In addition, G. instriatum burst at a salinity of 0 at all temperatures, but grew at a salinity of 5 at temperatures between 20 and 25 °C. It is noteworthy that G. instriatum is a euryhaline organism that can live under extremely low salinity. Factorial analysis revealed that the contributions of temperature and salinity to its growth of the organism were almost equal. The irradiance at the light compensation point (I0) was 10.6 μmol/(m2 s) and the saturated irradiance for growth (Is) was 70 μmol/(m2 s), which was lower than Is for several other harmful dinoflagellates (90–110 μmol/(m2 s)).  相似文献

Canna indica L. is an upright perennial rhizomatous herb, and Schoenoplectus validus (Vahl) A. Löve and D. Löve is a tall, perennial, herbaceous sedge. The nutrient uptake kinetics of C. indica and S. validus were investigated using the modified depletion method after plants were grown for 4 weeks in simulated secondary-treated wastewater. The maximum uptake rate (Imax) and Michaelis–Menten constant (Km) were estimated by iterative curve fitting. The Imax for NH4N (623 μmol g−1 dry root weight h−1) was significantly higher than that for NO3N (338 μmol g−1 dry root weight h−1) in S. validus. In contrast, no difference was observed in C. indica. The Imax values for NO3N and NH4N were higher in S. validus than in C. indica. A significantly lower Km was detected for NO3N uptake in C. indica (385 μmol L−1) compared to that in S. validus (1908 μmol L−1). The Imax for PO4P did not differ between the plant species. The Km for PO4P was significantly higher in C. indica (157 μmol L−1) than in S. validus (60 μmol L−1). In conclusion, we found that S. validus preferred NH4N over NO3N, had greater capacity for N uptake and higher affinity for PO4P, but C. indica had greater affinity for NO3N. Nutrient uptake capacity is likely related to habitat preference, and is influenced by the structure of roots and rhizomes.  相似文献

Sex allocation theories predict equal offspring number of both sexes unless differential investment is required or some competition exists. Left undisturbed, elephants reproduce well and in approximately even numbers in the wild. We report an excess of males are born and substantial juvenile mortality occurs, perinatally, in captivity. Studbook data on captive births (CB, n = 487) and premature deaths (PD, <5 years of age; n = 164) in Asian and African elephants in Europe and North America were compared with data on Myanmar timber (Asian) elephants (CB, n = 3070; PD, n = 738). Growth in CB was found in three of the captive populations. A significant excess of male births occurred in European Asian elephants (ratio: 0.61, P = 0.044) and in births following artificial insemination (0.83, P = 0.003), and a numerical inclination in North American African elephants (0.6). While juvenile mortality in European African and Myanmar populations was 21–23%, it was almost double (40–45%) in all other captive populations. In zoo populations, 68–91% of PD were within 1 month of birth with stillbirth and infanticide being major causes. In Myanmar, 62% of juvenile deaths were at >6 months with maternal insufficient milk production, natural hazards and accidents being the main causes. European Asian and Myanmar elephants PD was biased towards males (0.71, P = 0.024 and 0.56, P < 0.001, respectively). The skewed birth sex ratio and high juvenile mortality hinder efforts to help captive populations become self-sustaining. Efforts should be invested to identify the mechanism behind these trends and seek solutions for them.  相似文献

Trametes versicolor 1 was shown to grow on phenol as its sole carbon and energy source. The culture growth and degradation ability dependence on culture medium pH value was observed. The optimal pH value of a liquid Czapek salt medium was 6.5. The investigated strain utilized completely 0.5 g/l phenol in 6 days. The dynamics of the phenol degradation process was investigated. The process was characterized by specific growth rate μmax 0.33 h−1, metabolic coefficient k = 4.4, yield coefficient Y x/s  = 0.23 and rate of degradation Q = 0.506 h−1. The intracellular activities of phenol hydroxylase (0.333 U/mg protein) and cis,cis-muconate lactonizing enzyme (0.41 U/mg protein) were demonstrated for the first time in this fungus. In an attempt to estimate the occurrence of gene sequences in T. versicolor 1 related to phenol degradation pathway a dot blot analysis with total DNA isolated from this strain was performed. Two synthetic oligonucleotides were used as hybridizing probes. One of the probes was homologous to the 5′end of phyA gene coding for phenol hydroxylase in Trichosporon cutaneum ATCC 46490. The other probe was created on the basis of cis,cis-muconate lactonizing enzyme coding gene in T. cutaneum ATCC 58094. The results of these investigations showed that T. versicolor 1 may carry genes similar to those of Trichosporon cutaneum capable to degrade phenol.  相似文献

Recent experimental and theoretical studies have found that active dendritic ionic currents can compensate for the effects of electrotonic attenuation. In particular, temporal summation, the percentage increase in peak somatic voltage responses invoked by a synaptic input train, is independent of location of the synaptic input in hippocampal CA1 pyramidal neurons under normal conditions. This independence, known as normalization of temporal summation, is destroyed when the hyperpolarization-activated current, I h, is blocked [Magee JC (1999a), Nature Neurosci. 2: 508–514]. Using a compartmental model derived from morphological recordings of hippocampal CA1 pyramidal neurons, we examined the hypothesis that I h was primarily responsible for normalization of temporal summation. We concluded that this hypothesis was incomplete. With a model that included I h, the persistent Na+ current (I NaP), and the transient A-type K+ current (I A), however, we observed normalization of temporal summation across a wide range of synaptic input frequencies, in keeping with experimental observations.  相似文献

Quasi steady state growth of Lactococcus lactis IL 1403 was studied in glucose-limited A-stat cultivation experiments with acceleration rates (a) from 0.003 to 0.06 h−2 after initial stabilization of the cultures in chemostat at D = 0.2–0.3 h−1. It was shown that the high limit of quasi steady state growth rate depended on the acceleration rate used—at an acceleration rate 0.003 h−2 the quasi steady state growth was observed until μ crit = 0.59 h−1, which is also the μ max value for the culture. Lower values of μ crit were observed at higher acceleration rates. The steady state growth of bacteria stabilized at dilution rate 0.2 h−1 was immediately disrupted after initiating acceleration at the highest acceleration rate studied—0.06 h−2. Observation was made that differences [Δ(μ − D)] of the specific growth rates from pre-programmed dilution rates were the lowest using an acceleration rate of 0.003 h−2 (< 4% of preset changing growth rate). The adaptability of cells to follow preprogrammed growth rate was found to decrease with increasing dilution rate—it was shown that lower acceleration rates should be applied at higher growth rates to maintain the culture in the quasi steady state. The critical specific growth rate and the biomass yields based on glucose consumption were higher if the medium contained S 0 = 5 g L−1 glucose instead of S 0 = 10 g L−1. It was assumed that this was due to the inhibitory effect of lactate accumulating at higher concentrations in the latter cultures. Parallel A-stat experiments at the same acceleration and dilution rates showed good reproducibility—Δ(μ − D) was less than 5%, standard deviations of biomass yields per ATP produced (Y ATP), and biomass yields per glucose consumed (Y XS) were less than 15%.  相似文献

活血丹(Glechoma lonituba)是唇形科活血丹属的多年生克隆草本植物。采用简单重复序列区(ISSR)分子标记技术,比较分析了3个不同斑块活血丹的遗传多样性、克隆多样性以及小尺度空间遗传结构,并探讨其与生境异质性、繁殖体传播和人为干扰的相关性。结果表明:1)活血丹物种水平的遗传多样性很低,各斑块的遗传多样性较低,以水渠边斑块最高,平葛村斑块次之,竹林下斑块最低。2)活血丹物种水平的克隆多样性较高,各斑块活血丹的克隆多样性以水渠边斑块最大,平葛村斑块次之,竹林下斑块最低。3)遗传分化系数Gst为0.7129,表明活血丹的遗传变异大部分存在于斑块间;斑块间的基因流较小,仅为0.2004。4)空间自相关分析表明活血丹一定的空间距离下存在显著的空间遗传结构,竹林下斑块在100cm时存在显著性正相关,其X轴截矩为205.994cm;平葛村斑块在200cm时存在显著性正相关,其X轴截矩为235.388cm;水渠边斑块在150cm时存在显著性正相关,其X轴截矩为240.336cm。应用软件SPAGeDi 1.2软件对各斑块的空间遗传结构进行量化,表明平葛村斑块具有最强的空间遗传结构,水渠边和竹林下斑块的空间遗传结构较弱。活血丹的遗传结构、克隆结构及空间分布格局受到其繁殖体传播特征、人为干扰和繁殖权衡的影响,是其对生境异质性的适应结果。  相似文献

Fourteen metronidazole derivatives (compounds 3af and 4bh) have been synthesized by coupling of metronidazole and salicylic acid derivatives. All of them are reported for the first time. Their chemical structures are characterized by 1H NMR, MS, and elemental analysis. The inhibitory activities against Helicobacter pylori urease have been investigated in vitro and many compounds have showed promising potential inhibitory activities of H. pylori urease. The effect of compounds 4b (IC50 = 26 μM) and 4g (IC50 = 12 μM) was comparable with that of acetohydroxamic acid, a well known H. pylori urease inhibitor used as a positive control. The experimental values of IC50 showed that inhibitor was potent urease inhibitor. A docking analysis using the autodock 4.0 program could explain the inhibitory activities of compound 4g against H. pylori urease.  相似文献

A family of 10 competing, unstructured models has been developed to model cell growth, substrate consumption, and product formation of the pyruvate producing strain Escherichia coli YYC202 ldhA::Kan strain used in fed-batch processes. The strain is completely blocked in its ability to convert pyruvate into acetyl-CoA or acetate (using glucose as the carbon source) resulting in an acetate auxotrophy during growth in glucose minimal medium. Parameter estimation was carried out using data from fed-batch fermentation performed at constant glucose feed rates of qVG=10 mL h–1. Acetate was fed according to the previously developed feeding strategy. While the model identification was realized by least-square fit, the model discrimination was based on the model selection criterion (MSC). The validation of model parameters was performed applying data from two different fed-batch experiments with glucose feed rate qVG=20 and 30 mL h–1, respectively. Medium-chain-length polyhydroxyalkanoates (MCL-PHAs) were produced in carbon-limited, single-stage, fed-batch fermentations of Pseudomonas putida KT2440 by co-feeding nonanoic acid (NA) and glucose (G) to enhance the yield of PHA from NA. An exponential (μ = 0.25 h−1) followed by a linear feeding strategy at a NA:G ratio of 1:1 (w/w) achieved 71 g l−1 biomass containing 56% PHA. Although the same overall PHA productivity (1.44 g l−1 h−1) was obtained when NA alone was fed at the same specific growth rate, the overall yield of PHA from NA increased by 25% (0.66 g PHA g NA−1 versus 0.53 g g−1) with glucose co-feeding. Caragana microphylla is the most dominant and constructive shrub species in the Horqin sandy land in the northeast of China. We evaluated it's the level of genetic variation within and among populations sampled from two different populations types in Horqin sandy land by using inter-simple sequence repeat polymorphism (ISSR) molecular markers. The results showed that eight ISSR primers generated 106 bands, of which 87 were polymorphic. At the species level, genetic diversity was relatively high (P = 82.08%, h = 0.2831, I = 0.4233). Genetic variation in natural populations (h = 0.2152, I = 0.3169) was more than that in plantation populations (h = 0.2021, I = 0.3040). Based on Nei's GST value, more genetic differentiation among plantation populations was detected (GST = 0.7787). Six populations of C. microphylla clustered into two clades. These results have important implications for restoring and managing the degraded ecosystem in arid and semi-arid areas.  相似文献

The nitrogen uptake and growth capabilities of the potentially harmful, raphidophycean flagellate Heterosigma akashiwo (Hada) Sournia were examined in unialgal batch cultures (strain CCMP 1912). Growth rates as a function of three nitrogen substrates (ammonium, nitrate and urea) were determined at saturating and sub-saturating photosynthetic photon flux densities (PPFDs). At saturating PPFD (110 μE m−2 s−1), the growth rate of H. akashiwo was slightly greater for cells grown on NH4+ (0.89 d−1) compared to cells grown on NO3 or urea, which had identical growth rates (0.82 d−1). At sub-saturating PPFD (40 μE m−2 s−1), both urea- and NH4+-grown cells grew faster than NO3-grown cells (0.61, 0.57 and 0.46 d−1, respectively). The N uptake kinetic parameters were investigated using exponentially growing batch cultures of H. akashiwo and the 15N-tracer technique. Maximum specific uptake rates (Vmax) for unialgal cultures grown at 15 °C and saturating PPFD (110 μE m−2 s−1) were 28.0, 18.0 and 2.89 × 10−3 h−1 for NH4+, NO3 and urea, respectively. The traditional measure of nutrient affinity—the half saturation constants (Ks) were similar for NH4+ and NO3 (1.44 and 1.47 μg-at N L−1), but substantially lower for urea (0.42 μg-at N L−1). Whereas the α parameter (α = Vmax/Ks), which is considered a more robust indicator for substrate affinity when substrate concentrations are low (<Ks), were 19.4, 12.2 and 6.88 × 10−3 h−1/(μg-at N L−1) for NH4+, NO3 and urea, respectively. These laboratory results demonstrate that at both saturating and sub-saturating N concentrations, N uptake preference follows the order: NH4+ > NO3 > urea, and suggests that natural blooms of H. akashiwo may be initiated or maintained by any of the three nitrogen substrates examined.  相似文献
