Three experiments were conducted to examine the effects of spray applications of a mycoinsecticide against the rice grasshopper, Hieroglyphus daganensis, in northern Benin. The experiments were designed to expand on previous evaluation studies and contribute to a better understanding of the overall impacts of mycoinsecticide applications. The first experiment showed that acquisition of spores from the spray residue had a significant effect on mortality rate and a greater single influence on mortality than direct contact with spores from the spray aerosol. The second and third experiments revealed the mycoinsecticide to have sub- and pre-lethal effects, causing a significant reduction in per capita feeding rate and increasing susceptibility to predation. The significance of these results for the overall impact of spray applications and for development of comprehensive evaluation methods are discussed.  相似文献

Abstract. The effects of three plant species, Calotropis procera , Zygophyllum gaetulum and Peganum harmala on survival, feeding behaviour and reproduction in the desert locust ( Schistocerca gregaria ) were studied under laboratory conditions. The species originate in Moroccan arid areas and were seen to be avoided by locusts during the 1995 upsurge in Morocco. Alkaloids were extracted from leaves of the plants and applied topically to lettuce for leaves which were then offered with no other food to young adults of the desert locust for 3 days. The locust were then fed on untreated lettuce for 15 days. Controls were fed with lettuce leaves treated with ethanol. The results indicate that the alkaloids extracted from each of the three plant species cause a significant mortality compared to untreated controls and reduced food intake, as well as causing weight loss. Alkaloids extracted from C. procera and Z. gaetulum prevent sexual maturity both in males and females. However, alkaloids extracted from P. harmala merely delay sexual maturity by at least 8 days and engender a reduction in female fecundity and hatching rate compared to untreated controls. The presence and the concentration of alkaloids and other secondary substances in the investigated plants may explain the avoidance behaviour manifested by locusts under natural conditions.  相似文献

Metarhizium flavoviride, a fungal pathogen of grasshoppers and locusts, appears to be an effective, non-chemical insecticide (mycoinsecticide) for control of grasshoppers and locusts. This study, conducted during June and July, 1997, examined the pathogenic potential of this entomopathogenic fungus to non-target avian species that encounter infected insect prey items or contaminated food sources. Ring-necked pheasant (Phasianus colchicus) chicks were exposed to one of three diets, (spore-coated feed, infected insects, or untreated feed), either from 4 to 9 days of age, or, from 35 to 40 days of age. Necropsies were conducted on birds 10 days and 46 days old, respectively. Neither consumption of infected insects, nor of spore-coated feed, resulted in pathological changes, or significant changes in weight, growth rate, behavior, or mortality rate. Histological examination of organs indicated either no changes related to treatment, or normal tissue responses to antigenic challenge.  相似文献

The effectiveness of tebufenozide applied against the adult, egg, and larvae of codling moth, Cydia pomonella (L.), was evaluated. Significant reductions in fecundity and egg hatch occurred after 1-h and 24-h exposures of females and 24-h exposures of males-only to residues in plastic plates. A significant reduction in egg hatch was also found after a 1-h exposure of males. The ovicidal effects of tebufenozide in field trials did not significantly differ for eggs laid on residues or treated topically. Corrected egg mortality exceeded 95% for cohorts laid <130 degree-days after sprays were applied. Fecundity and egg hatch were measured after either a 24-h exposure of moths or a 10-d exposure of moths and eggs on apple trees. Significant reductions in both fecundity and fertility occurred compared with an untreated control up to 7 d in the 24-h assays and accounted for 60-70% fewer larvae produced per mated female. The mean numbers of larvae produced per mated female after the 10-d exposure were reduced 100-75% in assays started on day 0-21 after the spray application compared with the untreated control. Fecundity was significantly reduced for 7 d and egg hatch was reduced for the entire 21-d test period in these trials. Residues had a 14-d half-life on apple foliage. Residues applied to foliage or to foliage and fruit did not significantly increase the proportion of uninjured fruit compared with the untreated control in bioassays where neonates were placed on foliage 10 cm from fruit. However, the proportion of injured fruits with shallow stings versus deep entries was higher on the treated versus the untreated plants. Field applications of tebufenozide reduced fruit injury >90% when applied early during emergence. Plots treated after the beginning of egg hatch had a larger proportion of injured fruits with shallow larval feeding.  相似文献

Field and laboratory studies demonstrated that Paranosema (Nosema) locustae had significant effects on the morphological phase transformation of Locusta migratoria manilensis (Meyen 1835). In the field, spraying P. locustae on gregarious locusts caused a substantial population reduction by 16 days after treatment, with most of the surviving locusts being phase solitaria. However, the effects of P. locustae on locust phase transformation began before direct mortality had caused a substantial reduction in locust density: locust numbers were still high at day 10, but locusts had already transformed to phase transiens. Laboratory assays showed that while a low dose of P. locustae had no effect on phase transformation, at a higher dose of 1×105 spores/mL, locusts had F/C ratios that were significantly (P<0.05) more solitaria than untreated locusts, with locusts having ratios that were either phase solitaria or on the solitaria side of phase transiens. In a second laboratory experiment that analysed the effects of locust density on phase transformation by P. locustae, there was no obvious effect of density on female locusts 10 days later as all were solitaria at all locust densities. At day 16, female locusts were transiens at higher densities, but were solitaria at 4/cage. With males there were lesser effects. These results provide new evidence for P. locustae having sub-lethal effects on locust phase transformation at a wide range of locust densities.  相似文献

A series of investigations was carried out at ICARDA during April-June 2004 and May-June 2005 to investigate the use of the entomopathogenic fungus Beauveria bassiana to control Eurygaster integriceps and to determine aspects of the insect's biology that could be relevant to control with the fungus. Application in 2004 of an oil-based formulation of B. bassiana showed distribution of the spray decreasing from the top of the wheat to the bottom, although the proportion of tillers with coverage at the base was still 80%. Seventy percent of Sunn Pest had received direct hits, with the highest amount on the dorsal surface, followed by the ventral surface, then the legs and antennae. Mortality of field-treated, then laboratory-maintained insects, ranged from 47.6% for insects exposed only to the B. bassiana spray, 74.2% for unsprayed insects exposed to treated wheat and 86.4% for insects receiving direct spray and being exposed to treated wheat. Some field results demonstrated mortality. Mortality assessments following the 2005 treatments were conducted solely in the field, there was no apparent effect upon Sunn Pest numbers. The majority of studies on Sunn Pest biology were carried out on untreated insects outside the trial plots. Different assessment methods resulted in varying estimates of adults and juveniles in the field. The numbers of Sunn Pest found and the proportion of these at the top of the wheat crop, declined after early morning and increased again in the early evening, suggesting sheltering deeper in the crop or in the soil during the middle part of the day. Mobility and location of marked insects was followed in micro-plots and by mark and recapture studies, demonstrating a range of activities, from movement around the crop for a number of metres, including contact with numerous tillers to almost total immobility; Sunn Pest generally avoided movement on the soil. The potential for Sunn Pest control with a mycoinsecticide and how Sunn Pest behaviour is relevant to a mycoinsecticide application are discussed.  相似文献

The spinosad bait spray GF-120 Naturalyte F® (Dow AgroSciences LLC, Indianapolis, IN, USA) is widely used in commercial olives in California. Because of concern about its non-target effects on beneficial insects, we studied its impact on feeding behavior, mortality, and reproductive parameters of adult green lacewings, Chrysoperla carnea (Stephens), under laboratory conditions. Male and female feeding rate and quantity, and subsequent mortality, were compared over a 5-day period following a 24-h exposure to honey, GF-120, or the bait component (without spinosad) in two-choice and no-choice assays. All treatments were ingested equally when offered alone. Spinosad did not impart repellency or preference to GF-120 relative to the bait. Honey was preferred in choice tests with GF-120 or bait. Significant mortality occurred when GF-120 was offered alone, but was not significant when it was offered in choice with honey or bait. In a separate assay of female longevity and reproduction, mortality rate as a function of time was similar in all treatments, except for some significantly earlier mortality due to GF-120 compared with honey. GF-120 ingestion significantly reduced fecundity compared with bait during the first week after treatment. Egg hatch was not significantly affected. The preference of adult C. carnea for food should ameliorate the potential negative effect of GF-120 on adult C. carnea mortality in the field. The laboratory study finds that GF-120 is a potentially safer choice for C. carnea than an aqueous suspension of spinosad, and provides no new evidence for definitive risk to field populations.  相似文献

The bioactivity of an ethyl acetate extract of ginger (Zingiber officinale) towards Schistosoma mansoni adult pairs, both cultured in vitro and in vivo in laboratory mice, was investigated by monitoring worm mortality and fecundity. In vitro, a concentration of 200 mg l(-1) of extract killed almost all worms within 24 h. Male worms seemed more susceptible than female under these conditions. Cumulative egg output of surviving worm pairs in vitro was considerably reduced when exposed to the extract. For example, after 4 days of exposure to 50 mg l(-1), cumulative egg output was only 0.38 eggs per worm pair compared with 36.35 for untreated worms. In vivo efficacy of the extract was tested by oral and subcutaneous delivery of 150 mg kg(-1) followed by assessment of worm survival and fecundity. Neither delivery route produced any significant reduction in worm numbers compared with untreated controls. Worm fecundity was assessed in vivo by cumulative egg counts per liver at 55 days post infection with mice treated subcutaneously. Such infections showed egg levels in the liver of about 2000 eggs per worm pair in 55 days, in both treated and control mice, with no significant difference between the two groups. To ensure that density-dependent effects did not confound this analysis, a separate experiment demonstrated no such influence on egg output per worm pair, at intensities between 1 and 23 worms per mouse.  相似文献

In laboratory and field cage experiments, the horizontal transmission of the entomopathogenic fungus Beauveria bassiana (Boverol®) between adults of the spruce bark beetle Ips typographus was examined. In a laboratory experiment, conidia transfer between treated and untreated beetles at different ratios resulted in 96% mortality (MST 4.3 days) at a ratio of 1:1 and 90% mortality (MST 4.5 days) at a ratio of 1:2. At ratios of 1:5, 1:10 and 1:20, the mortalities at 7 days were 83, 77 and 75%, respectively. In a second laboratory experiment, the efficacy of B. bassiana was investigated after transmission from contaminated to healthy beetles over a period of 5 days. Two weeks later, the mortality of treated and untreated beetles was 99% (control mortality was 44%), while the rate of mycosis was 79% in the treatment compared to 10% in the control beetles. A single contact between one treated male (2.0×105 conidia/beetle) and an untreated female was found to be sufficient to transmit a lethal dose of B. bassiana of about 1.2×104 conidia. Two experiments in field cages in a spruce stand demonstrated horizontal transmission of B. bassiana (Boverol®) between beetles. In the first experiment the transmission was investigated following the introduction of B. bassiana-inoculated beetles into a population of untreated beetles. Significant reductions were observed in the length of maternal galleries, and the number of larvae and pupae. In the second field cage experiment, natural populations of beetles were lured into a flight cage using a pheromone trap, and treated with B. bassiana or left untreated on alternate days. In this experiment, significant reductions were observed in the number of bore holes and the length of maternal galleries, while no larvae, pupae or juveniles were found under the bark of spruce trunks in the Boverol® transmission treatment. It is concluded that B. bassiana has potential for control of I. typographus, and further field experiments are warranted to investigate its efficacy under more practical conditions.  相似文献

Laboratory and so-called extended laboratory bioassays were conducted in Benin, West Africa, to investigate the pathogenicity and virulence of the entomopathogenic fungus Metarhizium anisopliae var. acridum , a biocontrol agent against locusts and grasshoppers, to two hymenopteran parasitoids, Apoanagyrus ( = Epidinocarsis ) lopezi and Phanerotoma sp. Treatments were carried out under simulated field conditions at standard field dose rates of 2.5 and 5.0 ×10 12 conidia ha -1 . Test organisms were continuously (3 weeks) exposed to spray residues in artificial or simulated natural environments. The standard strain IMI 330 189 of the mycopesticide Green Muscle caused a significant reduction of 24% in the longevity ( = average survival time, AST) of A. lopezi , relative to the untreated control. Mycosis was confirmed in 16% of all cadavers. AST was shorter under low relative humidity (RH) conditions, and these conditions seemed to enhance susceptibility to fungal infection. However, this effect was only marginally significant. In contrast, average longevity of untreated A. lopezi was slightly yet significantly shorter at low RH (50-60%) than at high RH (80-90%). In the extended laboratory assay, the same isolate had no significant effect on mortality, parasitoid emergence ( = beneficial capacity) and sex ratio. In a further screening test with isolates IIBC I91 609, IIBC I93 833 and IMI 330 189 (reference), no infection of A. lopezi was confirmed. Similarly, Phanerotoma sp. was not susceptible to IMI 330 189. It is concluded that mycopesticides based on the three strains tested pose a low risk to parasitic hymenopterans under field conditions.  相似文献

Adverse conditions, including low humidity, UV irradiation, and high temperature, appreciably affect the efficacy of mycoinsecticides. Oil formulation increased the virulence of Metarhizium anisopliae var. acridum (Ascomycota: Hypocreales) against locusts and grasshoppers by reducing the dependence on saturated water. A mycoinsecticide diluent (a water-in-oil emulsion) has been widely used to dilute the oil formulation of M. anisopliae in China. The aim of our study was to elucidate the mechanism by which the mycoinsecticide diluent improves the virulence of M. anisopliae. We investigated the effects of the mycoinsecticide diluent on the virulence, invasion speed, and viability of the conidia under various adverse conditions. The results demonstrated that the mycoinsecticide diluent significantly improved the virulence of conidia at low humidity (68, 75, and 84%). In particular, at an RH of 68%, the LT50 for locusts treated with the emulsion was 5.4 days and was 31.6% lower than the value for locusts treated with an oil formulation. In addition, the concentration of the hyphal bodies found in the haemolymph of locusts treated with emulsion was about 27-fold higher than that in locusts treated with oil formulation four days after inoculation. This result was further confirmed by determining the concentration of M. anisopliae var. acridum DNA in locust haemolymph using quantitative PCR. The percentage germination of conidia in the emulsion was also significantly higher than that in oil at 68% RH. There was no significant difference in percentage germination between conidia treated with the emulsion and oil when exposed to irradiation with ultraviolet-B (UV-B) or high temperature. These results demonstrate that the mycoinsecticide diluent enhances the virulence of M. anisopliae formulated in oil at low humidity by providing adequate water for germination without interfering with the UV tolerance and thermotolerance of the conidia.  相似文献

The effect of infection by the fungal entomopathogen , Metarhizium flavoviride, on feeding by the tropical grasshopper pest , Zonocerus variegatus, was investigated in field - cage studies . A significant reduction in feeding , as indicated by faecal production , was recorded 2 - 3 days after inoculation for a range of spore doses (104 , 105 and 5 105 spores per insect) . This was before any mortality was recorded due to infection . All infected individuals died by day 7 . At this time , faecal production of the treated grasshoppers was equivalent to less than 2 days faecal production by grasshoppers untreated with spores . This reduction in feeding (69 , 71 and 74% total reduction by day 7 in the 104 , 105 and 5 105 doses respectively in comparison to controls) is a substantial contribution to the overall effects of the slow acting pathogen . Furthermore , the rapid reduction in feeding indicated that this effect was not simply due to invasion of the host tissues by the pathogen or production of secondary metabolites . The possibility that reduction in feeding is associated with a behavioural response in which there is a trade - off between host defence and feeding during early stages of infection is discussed  相似文献

The efficacy of a mycoinsecticide formulated in vegetable oil was tested in Brazil against the grasshopper Rhammatocerus schistocercoides . A set of experiments was conducted in the Chapada dos Parecis region (Mato Grosso state), a permanent zone of outbreaks for this pest. Experiments were performed in zones of natural vegetation, against grasshopper bands in the third nymphal instar. Three nymphal bands were treated with a mycoinsecticide formulation based on conidia of the entomopathogenic fungus Metarhizium anisopliae var. acridum ( =M. flavoviride ), strain CG 423. Three non-treated bands were used as control. The application was made with the aid of a hand-held ULV sprayer adjusted to deliver 2 l of the formulation ha -1 , each containing 1 ×10 13 conidia. Treatments were limited to the surface of the grasshopper bands and their immediate borders (5-10 m). The efficacy of the mycoinsecticide was evaluated through band survival after treatment (grasshopper numbers, surface, density, behaviour and daily movement of the band), allowing the insects to move freely in their natural environment. Insects were regularly surveyed and maintained in the laboratory, allowing estimates of the infection rate. Field and laboratory studies showed a clear effect of the product 10 days after treatment. At 14 days post-spraying, mortality caused by the mycoinsecticide in the field was approximately 88%.  相似文献

1. This study investigated the effects of strong density dependence on larval growth, development, and survival of the mosquito Culex restuans (Theobald). It also tested the hypothesis that density reduction early in larval development could result in as many or more individuals surviving to adulthood (compensation or over‐compensation, respectively), or increased reproductive performance via rapid development and greater adult size. 2. In a field study of a natural population of C. restuans, the effects of a 75% lower density on percentage survivorship to adulthood, number of adults, development time, adult size, adult longevity, and size dependent fecundity were tested. 3. No evidence was found of compensation or over‐compensation in adult production, or of effects of lower density on percentage survivorship. Low density yielded significant increases in adult size, adult longevity, and size‐dependent fecundity, and a decrease in development time. 4. Estimated per‐capita population growth rate was significantly greater in the low‐density treatment than in the high‐density treatment. It is inferred that this difference was due to greater per‐capita resources, which increased female size and fecundity, and reduced development time. Greater per‐capita population growth could therefore result from early mortality of larvae, meaning that the hydra effect, which predicts greater equilibrium population with, as opposed to without, extrinsic mortality, may be possible for these mosquitoes.  相似文献

Studies were conducted in 1997 to evaluate the effects of the kaolin-based particle film formulation M96-018 on adults, eggs, and larvae of the obliquebanded leafroller, Choristoneura rosaceana (Harris). Particle film treatments significantly reduced female longevity, mating success, and number of egg masses oviposited compared with moths on untreated apple leaves in sleeve-cage and screen-cage tests. No differences in mating success or oviposition were caused by the application rates and coverage density of M96-018 on foliage. Females avoided ovipositing on particle film-treated leaves in choice tests. Larval hatch was not affected by topical application or residual exposure to M96-018. Larval weight gain and pupal weight were significantly reduced and larval mortality increased in no-choice feeding tests with M96-018. In choice tests, larvae preferred to feed on untreated leaf surfaces. The negative effects on larval development and survivorship on M96-018-treated foliage did not differ across a fourfold difference in spray application rate. A significant reduction in the number of infested shoots was found in orchard trials when M96-018 was applied before bud break in late March compared with untreated trees. No reductions in larval densities were found compared with an untreated control following prebloom and postbloom applications.  相似文献

The cold tolerance of overwintering adult Spur-throated locusts, Austracris guttulosa, was examined using measures of supercooling point relative to gender, environmental acclimation and feeding state as well as mortality for a range of sub-zero temperature exposure treatments. Freezing was lethal and supercooling points ranged from -6 to -12.8°C, but were statistically independent of fresh mass, body water content, acclimation, and/or gut content in fed and starved individuals. A significant interaction effect of gender and feeding status showed that the larger bodied females had decreased supercooling capacity with increased food material in the digestive tract. Post-freezing dissections revealed differences in the amount of freshly consumed and retained food material in the digestive tract between fed and starved individuals of each gender, which could explain this effect based on inoculation of ice crystallisation by food particles. Above supercooling temperatures, neither gender nor the rate of cooling had a significant effect on mortality. When cooled from 25°C at 0.1 or 0.5°Cmin(-1) to a range of experimental minimum temperatures held for 3h, survival was ~74% to -7°C, but declined sharply to ~37% when cooled to -8°C or lower. Although the laboratory experiments reported here suggest that A. guttulosa is not freeze tolerant and unable to rapidly cold harden, exposure to typical cold and frosty nights that very rarely reach below -8°C as a night minimum in the field would be unlikely to cause mortality in the vast majority of overwintering aggregations.  相似文献

Commercial formulations and unformulated conidia of Beauveria bassiana strain GHA were applied to field-grown plants and artificially infested with Trichoplusia ni (Hübner) larvae to compare the relative insecticidal activity resulting from direct spray contact with insecticidal activity due to contact with dry spray residue. In general, applications to cabbage, Brassica oleracea L., resulted in nearly equal mortalities when comparing insects exposed to direct spray contact with those exposed by spray residue, suggesting a potential benefit by improving formulations to extend residual activity. For applications to beans, Phaseolus vulgaris L., direct spray contact provided significant insect mortality, but mortality due to residual contact was generally not different than the untreated control. In contrast to the differences observed for larvae exposed in the field, larvae exposed in laboratory bioassays to leaf disks collected from the same treated cabbage and bean plants (residual contact exposure) resulted in nearly identical mortalities. Field applications of Beauveria showed rapid loss of activity, expressed as a loss of conidia viability and loss of insecticidal activity during the first 8 h after application. Evidence of significant mortality by residual contact and the rapid loss of insecticidal activity with field exposure support additional research to improve formulations to extend the residual activity of fungal biopesticides.  相似文献

Computer simulation models were developed to determine which conditions are favorable for the stochastic fixation of chromosomal mutations within small isolated demes. The models incorporated biological parameters of litter size, age-dependent mortality, overlapping generations, potentially varying sex ratios, and fertility reduction due to meiotic problems when the mutation was present in a heterozygous condition. Results of the models indicated that random processes alone may adequately explain the frequency of fixation of chromosomal mutations under the conditions of a) the number of initial founders is small (5 or 10); b) there is relatively little fecundity reduction due to meiotic problems; c) the number of offspring per mating is high. Furthermore, results of our models imply that fixation of a mutation is unlikely when the number of karyotypic rearrangements is low and when there is high survivorship of individuals from one breeding period to the next. However, if bottlenecks involve larger numbers of individuals (20 or more), few offspring per mating, or substantial reductions in fecundity, then it is unlikely that fixation by random processes will be adequate to explain the situations observed in nature. A significant conclusion of these simulations is that when population size is reduced to five or ten individuals, the extinction rate may exceed 40% or 30%, respectively.  相似文献

Phratora vulgatissima adults exhibit feeding preferences among willow varieties, yet little is known of the effects of willow variety on larval and adult performance. The effects of host variety on adult fitness and fecundity as well as on larval mortality and development were studied under laboratory conditions for 35 willow varieties. The host variety significantly affected the number of days that adults survived and the rates of weight change. On average, males lost weight and females gained weight. The total number of eggs, eggs day1, eggs clutch?1 and the length of the oviposition period were also significantly affected by willow variety. Progeny from eggs laid by adults fed on the different willow varieties showed significant differences in days to pupation and pupal weight when subsequently reared on Salix × dasyclados or on the same variety as fed to the adults. However, there was no correlation between these parameters on S. × dasyclados and the variety fed to adults. The willow variety fed to larvae significantly affected larval mortality (four varieties caused 100% mortality), the shape of larval growth curves (as measured by predicted final weight and time to half the final weight), the number of days to pupation and pupal weight. There were significant positive correlations between previously determined adult P. vulgatissima feeding preferences of the 35 willow varieties and the following: number of eggs laid, length of the oviposition period, larval mortality and development and change in adult weight. There was a considerable degree of variation in these correlations and some varieties did not follow the general trend indicated by the size or sign of particular correlations, for example, having a high feeding preference ranking yet few eggs laid, low larval weight and longer time to pupation. The differences found between varieties for adult and larval performance in conjunction with previously established feeding preferences offer great potential for utilising plant resistance to P. vulgatissima as a means of strategic control. Despite the general correlation of feeding preference and performance parameters, the results found here suggest that it appears to be possible for plant breeders to circumvent this trend.  相似文献

Reserpine, at doses of 20–175 μg per g body weight, severely retards oogenesis in newly emerged adult female migratory locusts (Locusta migratoria migratorioides) but does not increase mortality during the first 9 days and only slightly delays somatic growth. Total protein, and hemolymph vitellogenin content particularly, are significantly reduced in reserpine-treated locusts. The synthesis of juvenile hormone III (JH-III) following adult emergence, essential for induction of vitellogenesis and subsequent oogenesis, is dependent on the maturation and activation of the corpora allata (CA). CA of 7- to 8-day-old female locusts, treated with reserpine at day 1 after adult emergence, are only marginally active in vitro and are only slightly stimulated by an allatotropic factor. The basal activity and response of CA from the reserpine-treated locusts resembles that of newly emerged locusts, suggesting that reserpine specifically retards the initial maturation of the locust CA. Recovery of basal CA activity is evident on days 12–13 in reserpine-treated locusts, but responsiveness to the allatotropic factor is not recovered. Starvation of newly emerged females for 3 days and subsequent feeding did not effect ooctye development or CA activity. Cerebral content of the allatotropic factor, assayed on days 7–8, is not reduced by the reserpine treatment.  相似文献
