The establishment of epitypes (together with the emended diagnoses) for three species of Euglenaria Karnkowska, E. W. Linton et Kwiatowski [Eu. anabaena (Mainx) Karnkowska et E. W. Linton; Eu. caudata (Hübner) Karnkowska et E. W. Linton; and Eu. clavata (Skuja) Karnkowska et E. W. Linton] and two species of Euglena Ehrenberg [E. granulata (Klebs) Schmitz and E. velata Klebs] was achieved due to literature studies, verification of morphological diagnostic features (cell size, cell shape, number of chloroplasts, the presence of mucocysts), as well as molecular characters (SSU rDNA). Now all these species are easy to identify and distinguish, despite their high morphological similarity, that is, spindle‐shaped (or cylindrically spindle‐shaped) cells and parietal, lobed chloroplasts with a single pyrenoid, accompanied by bilateral paramylon caps located on both sides of the chloroplast. E. granulata is the only species in this group that has spherical mucocysts. E. velata is distinguished by the largest cells (90–150 μm) and has the highest number of chloroplasts (>30). Eu. anabaena has the fewest chloroplasts (usually 3–6), and its cells are always (whether the organism is swimming or not) spindle‐shaped or cylindrically spindle‐shaped, in contrast to the cells of Eu. clavata, which are club‐shaped (clavate) while swimming and only after stopping change to resemble the shape of a spindle or a cylindrical spindle; Eu. clavata has numerous chloroplasts (15–20). Eu. caudata is characterized by asymmetrical spindle‐shaped (fusiform) cells, that is, with an elongated rear section and a shorter front section; the number of chloroplasts normally ranges from 7 to 15.  相似文献

The phylogeny of ligulate and sulfuric‐acid containing species of Desmarestia, occurring worldwide from polar to temperate regions, was revised using a multigenic and polyphasic approach. Sequence data, gametophyte characteristics, and sporophyte morphology support reducing a total of 16 taxa to four different species. (1) D. herbacea, containing broad‐bladed and highly branched forms, has dioecious gametophytes. The three other species have monoecious gametophytes: (2) D. ligulata which is profusely branched and, except for one subspecies, narrow‐bladed, (3) Japanese ligulate Desmarestia, here described as D. japonica sp. nov., which is morphologically similar to D. ligulata but genetically distant from all other ligulate taxa. This species may have conserved the morphology of original ligulate Desmarestia. (4) D. dudresnayi, including unbranched or little branched broad‐bladed taxa. A figure of the holotype of D. dudresnayi, which was lost for decades, was relocated. The taxonomy is complemented by a comparison of internal transcribed spacer and cytochrome c oxidase subunit I (cox1) as potential barcode loci, with cox1 offering good resolution, reflecting species delimitations within the genus Desmarestia.  相似文献

The diatom genus Chaetoceros is one of the most abundant and diverse phytoplankton in marine and brackish waters worldwide. Within this genus, Chaetoceros socialis has been cited as one of the most common species. However, recent studies from different geographic areas have shown the presence of pseudo‐cryptic diversity within the C. socialis complex. Members of this complex are characterized by curved chains (primary colonies) aggregating into globular clusters, where one of the four setae of each cell curves toward the center of the cluster and the other three orient outwards. New light and electron microscopy observations as well as molecular data on marine planktonic diatoms from the coastal waters off Chile revealed the presence of two new species, Chaetoceros sporotruncatus sp. nov. and C. dichatoensis. sp. nov. belonging to the C. socialis complex. The two new species are similar to other members of the complex (i.e., C. socialis and C. gelidus) in the primary and secondary structure of the colony, the orientation pattern of the setae, and the valve ultrastructure. The only morphological characters that can be used to differentiate the species of this complex are aspects related to resting spore morphology. The two newly described species are closely related to each other and form a sister clade to C. gelidus in molecular phylogenies. We also provide a phylogenetic status along with the morphological characterization of C. radicans and C. cintus, which are genetically related to the C. socialis complex.  相似文献

The present study provides length–weight relationship (LWR) of three fish species, Cynoglossus puncticeps (Richardson, 1846), Cynoglossus lingua Hamilton, 1822 and Cynoglossus lida (Bleeker, 1851) of family Cynoglossidae from Chilika lagoon (19°28′–19°54′N; 85°05′–85°38′E), India. A total of 147 specimens were sampled during March, July and October of 2017 from screen barrier nets (mesh 14 mm to 26 mm) locally called khonda jal operated by local fishermen. The estimated b values derived from the data sets as follows: 3.12 for C. puncticeps, 3.09 for C. lida, and 2.88 for C. lingua.  相似文献

We use an integrative taxonomic approach to revise the genus Anisopteromalus. In particular, we apply multivariate ratio analysis (MRA), a rather new statistical method based on principal component analysis (PCA) and linear discriminant analysis (LDA), to numerous body measurements and combine the data with those from our molecular analysis of Cytb and ITS2 genetic markers (on a subset of species) and all available published data on morphology, karyology, behaviour, host associations and geographic distribution. We demonstrate that the analysis of quantitative characters using MRA plays a major role for the integration of name‐bearing types and thus for the association of taxa with names. Six species are recognized, of which two are new: A. cornis Baur sp.n. and A. quinarius Gokhman & Baur sp.n. For Anisopteromalus calandrae (Howard), a well‐known, cosmopolitan parasitoid of stored‐product pests, we have selected a neotype to foster continuity and stability in the application of this important name. The species was sometimes confused with the related A. quinarius sp.n. , another cosmopolitan species that is frequently encountered in similar environments. We also show that several species originally described or later put under Anisopteromalus actually belong to different genera: Cyrtoptyx camerunus (Risbec) comb.n. ; Meraporus glaber (Szelényi) comb.n. ; Dinarmus schwenkei (Roomi, Khan & Khan) comb.n. Neocatolaccus indicus Ayyar & Mani is confirmed as a junior synonym of Oxysychus sphenopterae (Ferrière) syn.n. and Anisopteromalus calandrae brasiliensis (Domenichini) stat.rev. must be considered as a valid but doubtful taxon. Asteriscus otolith shapes as well as their morphometry and shape contours were investigated in order to identify four allopatric Alburnus species: A. chalcoides (Güldenstädt, 1772) (Ordu), A. escherichii Steindachner, 1897 (Eski?ehir), Amossulensis Heckel, 1843 (Tunceli), and A. tarichi (Güldenstädt, 1814) (Van) in Turkish inland waters. These were compared using the shape indices (form factor, roundness, circularity, ellipticity, rectangularity and aspect ratio), and the morphological characters [otolith weight (OWE), otolith length (OL), otolith width (OW), otolith perimeter (OP), and otolith area (OA)]. The overall canonical discriminant analysis (CDA) classification score was 93.8%, with the lowest score for A. escherichii (82.5%) and the highest for A. chalcoides (100%). The otolith shapes, morphology and shape contours of all sampled fish were a clear species differentiator, thereby demonstrating that the otolith shape is species‐specific. The current study presents for the first time comprehensive variation information on interspecific left‐right asteriscus otoliths in males and females of each Alburnus species: A. chalcoides from Ordu, A. escherichii from Eski?ehir, A. mossulensis from Tunceli and A. tarichi from Van, based on a total of 307 individuals. Scanning electron microscopy (SEM) images, shape contours and other otolith characters vary within the same genus; these differences should be investigated not only in other freshwater fish species or genera but also in the same species living in different habitats. In addition, further investigation is required not only with respect to the morphometry, biometry, shape, geometry, and shape contours of the otoliths, but also regarding the genetic methods for robust identification of various sympatric and allopatric fish populations.  相似文献

Miscanthus is a rhizomatous C4 grass of great interest as a biofuel crop because it has the potential to produce high yields over a wide geographical area with low agricultural inputs on marginal land less suitable for food production. At the moment, a clonal interspecific hybrid Miscanthus × giganteus is the most widely cultivated and studied in Europe and the United States, but breeding programmes are developing newer more productive varieties. Here, we quantified the physiological processes relating to whole season yield in a replicated plot trial in Wales, UK. Light capture and conversion efficiency were parameterized for four carefully selected genotypes (M. sinensis, M. sacchariflorus and Miscanthus × giganteus). Differences in the canopy architecture in mature stands as measured by the extinction coefficient (k) were small (0.55–0.65). Sensitivity analysis on a mathematical model of Miscanthus was performed to quantify the accumulative intercepted photosynthetically active radiation (iPAR) in the growing season using (i) k, (ii) variation in the thermal responses of leaf expansion rate, (iii) base temperature for degree days and (iv) date start of canopy expansion. A 10% increase in k or leaf area per degree day both had a minimal effect on iPAR (3%). Decreasing base temperature from 10 to 9 °C gave an 8% increase in iPAR. If the starting date for canopy expansion was the same as shoot emergence date, then the iPAR increases by 12.5%. In M. × giganteus, the whole season above ground and total (including below ground) radiation‐use efficiency (RUE) ranged from 45% to 37% higher than the noninterspecific hybrid genotypes. The greater yields in the interspecific hybrid M. × giganteus are explained by the higher RUE and not by differences in iPAR or partitioning effects. Studying the mechanisms underlying this complex trait could have wide benefits for both fuel and food production.  相似文献

Progress in NGS technologies has opened up new opportunities for characterizing biodiversity, both for individual specimen identification and for environmental barcoding. Although the amount of data available to biologist is increasing, user‐friendly tools to facilitate data analysis have yet to be developed. Our aim, with |SE|S|AM|E| Barcode, is to provide such support through a unified platform. The sequences are analysed through a pipeline that (i) processes NGS amplicon runs, filtering markers and samples, (ii) builds reference libraries and finally (iii) identifies (barcodes) the sequences in each amplicon from the reference library. We use a simulated data set for specimen identification and a recently published data set for environmental barcoding to validate the method. In this article, we modify bioenergy model MiscanFor investigating global and UK potentials for Miscanthus × giganteus as a bioenergy resource for carbon capture in the 21st century under the RCP 2.6 climate scenario using SSP2 land use projections. UK bioenergy land projections begin in the 2040s, 60 year average is 0.47 Mega ha rising to 1.9 Mega ha (2090s). Our projections estimate UK energy generation of 0.09 EJ/year (60 year average) and 0.37 EJ/year (2090s), under stable miscanthus yields of 12 t ha?1 year?1. We estimate aggregated UK soil carbon (C) increases of 0.09 Mt C/year (60 year average) and 0.14 Mt C/year (2090s) with C capture plus sequestration rate of 2.8 Mt C/year (60 year average) and 10.49 Mt C/year (2090s). Global bioenergy land use begins in 2010, 90 year average is 0.13 Gha rising to 0.19 Gha by the 2090s, miscanthus projections give a 90 year average energy generation of 16 EJ/year, rising to 26.7 EJ/year by the 2090s. The largest national capabilities for yield, energy and C increase are projected to be Brazil and China. Ninety year average global miscanthus yield of 1 Gt/year will be 1.7 Gt/year by the 2090s. Global soil C sequestration increases less with time, from a century average of 73.6 Mt C/year to 42.9 Mt C/year by the 2090s with C capture plus sequestration rate of 0.54 Gt C/year (60 year average) and 0.81 Gt C/year (2090s). M. giganteus could provide just over 5% of the bioenergy requirement by the 2090s to satisfy the RCP 2.6 SSP2 climate scenario. The choice of global land use data introduces a potential source of error. In reality, multiple bioenergy sources will be used, best suited to local conditions, but results highlight global requirements for development in bioenergy crops, infrastructure and support.  相似文献

The genus Ptilophora currently includes 16 species occurring mostly in subtidal habitats of the Indo‐Pacific Ocean, but its global diversity and biogeography are poorly understood. We analyzed mitochondrial cox1, plastid rbcL and plastid psbA sequences from specimens collected in southern Madagascar during the 2010 Atimo Vatae expedition and studied their morphologies. Both morphological and molecular data sets demonstrated the presence of five species in southern Madagascar: Ptilophora hildebrandtii, P. pterocladioides, and three new species described here, P. aureolusa, P. malagasya, and P. spongiophila. Ptilophora aureolusa is distinguished by its compound pinnae with uniformly spaced pinnules. Ptilophora malagasya has an indistinct midrib and irregularly spaced pinnules. Ptilophora spongiophila, heavily coated with sponges, has cylindrical to flattened main axes, lateral and surface proliferations, and spatulate tetrasporangial sori. The species of Ptilophora found in Madagascar are endemic, except P. hildebrandtii, which also occurs in eastern Africa. Ptilophora comprises four phylogenetic groups that map to eastern Australia, Japan, western Australia/Southeast Asia/Madagascar/eastern Africa, and Madagascar/eastern Africa/Aegean Sea. Biogeographical analysis revealed that the ancestor of Ptilophora originated in Australia, but most of the species radiated from Madagascar.  相似文献

Melting summer snow in the Austrian Alps exhibited a yellowish bloom that was mainly comprised of an unidentified unicellular chrysophyte. Molecular data (18S rRNA and rbcL genes) showed a close relationship to published sequences from an American pond alga formerly identified as Kremastochrysis sp. The genera Kremastochrysis and Kremastochrysopsis are morphologically distinguished by the number of flagella observed with the light microscope, and therefore we assigned the Austrian snow alga and an American pond alga to the genus Kremastochrysopsis. Transmission and scanning electron microscopy revealed that swimming cells had two flagella oriented in opposite directions, typical for the Hibberdiales. Molecular phylogenetic analyses showed that both new species were closely related to Hibberdia. Kremastochrysopsis ocellata, the type species and only known species, has two chloroplasts per cell and the zoospores have red eyespots. Our two organisms had only a single chloroplast and no zoospore eyespot, but their gene sequences differed substantially. Therefore, we described two new species, Kremastochrysopsis austriaca sp. nov and Kremstochrysopsis americana sp. nov. When grown in culture, both taxa showed a characteristic hyponeustonic growth (hanging below the water surface), whereas older immotile cells grew at the bottom of the culture vessel. Ecologically, Kremastochrysopsis austriaca sp. nov., which caused snow discolorations, had no close phylogenetic relationships to other psychrophilic chrysophytes, for example, Chromulina chionophilia, Hydrurus sp., and Ochromonas-like flagellates.  相似文献

Miscanthus × giganteus is a C4 perennial grass that shows great potential as a high‐yielding biomass crop. Scant research has been published that reports M. × giganteus growth and biomass yields in different environments in the United States. This study investigated the establishment success, plant growth, and dry biomass yield of M. × giganteus during its first three seasons at four locations (Urbana, IL; Lexington, KY; Mead, NE; Adelphia, NJ) in the United States. Three nitrogen rates (0, 60, and 120 kg ha?1) were applied at each location each year. Good survival of M. × giganteus during its first winter was observed at KY, NE, and NJ (79–100%), and poor survival at IL (25%), due to late planting and cold winter temperatures. Site soil conditions, and growing‐season precipitation and temperature had the greatest impact on dry biomass yield between season 2 (2009) and season 3 (2010). Ideal 2010 weather conditions at NE resulted in significant yield increases (< 0.0001) of 15.6–27.4 Mg ha?1 from 2009 to 2010. Small yield increases in KY of 17.1 Mg ha?1 in 2009 to 19.0 Mg ha?1 in 2010 could be attributed to excessive spring rain and hot dry conditions late in the growing season. Average M. ×giganteus biomass yields in NJ decreased from 16.9 to 9.7 Mg ha?1 between 2009 and 2010 and were related to hot dry weather, and poor soil conditions. Season 3 yields were positively correlated with end‐of‐season plant height () and tiller density (). Nitrogen fertilization had no significant effect on plant height, tiller density, or dry biomass yield at any of the sites during 2009 or 2010.  相似文献

Understanding the roles of metal ions in restriction enzymes has been complicated by both the presence of two metal ions in many active sites and their homodimeric structure. Using a single-chain form of the wild-type restriction enzyme PvuII (scWT) in which subunits are fused with a short polypeptide linker (Simoncsits et al. in J. Mol. Biol. 309:89–97, 2001), we have characterized metal ion and DNA binding behavior in one subunit and examined the effects of the linker on dimer behavior. scWT exhibits heteronuclear single quantum coherence NMR spectra similar to those of native wild-type PvuII (WT). For scWT, isothermal titration calorimetry data fit to two Ca(II) sites per subunit with low-millimolar K ds. The variant scWT|E68A, in which metal ion binding in one subunit is abolished by mutation, also binds two Ca(II) ions in the WT subunit with low-millimolar K ds. When there are no added metal ions, DNA binding affinity for scWT is tenfold stronger than that of the native WT, but tenfold weaker at saturating Ca(II) concentration. In the presence of Ca(II), scWT|E68A binds target DNA similarly to scWT, indicating that high-affinity substrate binding can be carried energetically by one metal-ion-binding subunit. Global analysis of DNA binding data for scWT|E68A suggests that the metal-ion-dependent behaviors observed for WT are reflective of independent subunit behavior. This characterization provides an understanding of subunit contributions in a homodimeric context.  相似文献

Extensive habitat modifications within the Mississippi and Missouri rivers have presumably interfered with the reproductive isolating mechanisms between the endangered pallid sturgeon, Scaphirhynchus albus, and the sympatric shovelnose sturgeon, Scaphirhynchus platorynchus, causing hybridization between these two species. Several character indices were developed to assist fisheries biologists in identifying specimens of S. albus, S. platorynchus, and their putative hybrids. The indices have numerous assumptions, including that pure strains of both parental species are within the sample analyzed and that hybrids are morphologically intermediate relative to their parents. If these indices have produced inaccurate identifications, then previous work on Scaphirhynchus studies in the Mississippi and Missouri rivers are questionable, including status surveys, captive propagation efforts, or the harvesting of tissues for genetic studies. In this study we tested indices by examining progeny of ‘known’ pallid, shovelnose, and hybrid sturgeon propagated, raised, and preserved at hatcheries. These 60 specimens [78–600 mm standard length (SL)] were propagated with breeding stock from the upper Missouri River drainage, where hybridization between these two species presumably does not occur. Existing indices did not correctly identify small (<250 mm SL) or a combination of small and large (>250 mm SL) sizes of S. albus, S. platorynchus, and their hybrids. Indices worked fairly well in identifying large S. platorynchus, but not in differentiating large S. albus from hybrids. We used principal components analysis (PCA) as an alternative approach to character indices. No a priori knowledge of the identity of the specimen is required with this multivariate technique, which avoids potential circular reasoning present in indices. We were able to completely or almost completely separate both sturgeon species and their hybrids by extracting principal components from a correlation matrix of 13 meristic characters in a standard PCA and extracting size‐corrected principal components from a covariance matrix of 51 morphometric variables using a sheared PCA. Additionally, we demonstrated that first generation hybrids were intermediate with respect to their parental species. Multivariate analyses with a reduced character set of six meristic and 12 morphometric variables also led to accurate and reliable specimen identification. Head spines and numerous qualitative characters are also extremely useful in differentiating between Scaphirhynchus species and their hybrids. In addition to all morphometric characters, some meristic characters and the degree of head spine fusion vary significantly with SL of sturgeons. Recording appropriate data from released specimens, including photovouchers, and making this information available is essential for researchers to have any scientific or legal basis for genetic or any other studies involving these sturgeons.  相似文献
